【目的】明确柑桔全爪螨Panonychus citri(Mc Gregor)高致病力菌株芽枝状枝孢霉Cladosporium cladosporioides(CQBBW3)的生物学特性。【方法】通过单因素试验,测试了不同培养基、碳源、氮源、温度及pH对CQBBW3菌株生长和产孢的影响。【结果】CQBBW3菌株的LC50为4.66×108个孢子/m L,LT50为6.2353 d。最佳生长培养基是淀粉琼脂培养基,最适产孢培养基为察氏培养基;菌丝生长最适碳源和氮源为甘露醇和甘氨酸,产孢最适碳源和氮源为乳糖和赖氨酸;最适生长温度为25℃,最适产孢温度为15℃和30℃;最适生长pH值为6.0~8.0,最适产孢pH值7.0。【结论】CQBBW3菌株对柑桔全爪螨有较好的杀虫活性,喜好25℃、中性、富含有机营养的环境。本研究为菌株快速培养及其真菌杀虫制剂的开发与田间应用奠定基础。  相似文献

源自不同寄主的灰葡萄孢生物学特性的比较研究   总被引:2,自引:0,他引:2  
本研究以分离自番茄、辣椒、草莓、葡萄的灰葡萄孢Botrytis cinerea为供试菌株,从生长温度、pH适应性、碳源、氮源营养利用等方面对不同寄主来源的灰葡萄孢菌株的生物学性状进行了比较研究。结果表明,5个不同寄主来源的灰葡萄孢菌株的菌丝生长温度范围相同,均为0-35℃;但它们的最适生长温度和分生孢子致死温度存在差异,来自和县番茄菌株HX12最适生长温度为20℃,分生孢子致死温度为47℃ 10min,其余最适生长温度均为25℃,分生孢子致死温度均为48℃ 10min;不同菌株在相同温度下的生长速率有显著差异。pH对不同寄主来源灰葡萄孢菌株菌丝生长的影响存在差异,来自长丰辣椒的菌株LJ菌丝在pH2-9的范围内均能生长,以在pH3-6.5时生长较快,pH6时最快;其余4个菌株在pH2-12的范围内均能生长,以在pH3-9时生长较快,pH6左右最快。不同碳源、氮源营养对灰葡萄孢菌株菌丝生长和分生孢子产生均有显著影响,不同寄主来源的菌株间在碳源、氮源营养利用差异均极显著。在相同碳源、氮源营养条件下,不同寄主来源的菌株的线性生长、菌丝干重和分生孢子产量均有显著差异。  相似文献

对番茄褐斑病原菌—长蠕孢菌产孢条件进行了研究,结果表明燕麦片琼胶培养基、Czapek培养基和PDA+番茄叶片能促进产孢子,V8汁、PSA和番茄汁培养基抑制产孢;碳源果糖明显促进产孢,甘露醇抑制产孢子;氮源氯化铵促进产孢,蛋白胨和硫酸铵抑制产孢;光照和紫外线照射对长蠕孢菌产孢有明显促进作用,特别是紫外线照射60~80min时产孢量达到最大;偏低温或偏高温以及微碱性环境能促进长蠕孢菌产孢,温度为15℃或30℃,pH8~9时最有利于产孢。  相似文献

对牛筋草离孺孢属(Bipolaris sorokiniana)NJ-08菌株的生物学特性及代谢产物除草活性测定结果表明:此菌株的菌丝体生长速度慢, 但产孢量大, 致病性强。菌丝体生长最适温度25°C, 致死温度是55°C/10 min, 最适pH值为8, 碳源以葡萄糖最佳, 供试氮源则不利于菌丝体生长。产生分生孢子最适温度25°C, 最适pH值为8, 碳源以葡萄糖最好, 氮源以硝酸钠产孢量最高。分生孢子萌发温度在20°C~35°C之间没有差异, 适宜pH值为5~7, 碳源以D-木糖的孢子萌发率最高, 氮源以蛋白胨的孢子萌发率最高, 不同处理间存在明显差异。NJ-08菌株产生的代谢产物作用于牛筋草, 可使叶片黄化或枯死, 代谢产物对指示植物及杂草如柱花草、地桃花、猪屎豆和辣椒的胚根胚芽生长都有明显的抑制作用, 抑制率均超过了90%, 显示出很好的除草活性。  相似文献

对前期筛选得到的具有生防潜力的杀蜱真菌Beauveria bassiana AT17菌株进行相关生物学性状的研究,建立一套规范的实验室培养的技术和方法,同时对其液固双相发酵技术进行优化,在于提供优质高效的生防材料,为其真菌制剂的规模化生产提供实践和理论基础。通过采用单因素筛选方案对其最适基础培养基、温度、pH值、碳源、氮源、微量元素及液固双相发酵配方进行研究,发现该菌株在PDA、PPDA、PDAY、SDAY、SMAY 5种培养基上均能较好生长,在PDA上生长最快,在PDAY上产孢量最大。温度对菌丝的生长和产孢影响显著,25°C菌丝生长最快,且产孢量最大。B.bassiana AT17菌株在pH 4-10范围内均可生长,在偏碱性环境内生长最快,在pH 5-6的偏酸性环境内产孢量最大。综合评价真菌各项指标后,葡萄糖和酵母粉为菌丝生长和产孢的最佳碳源和氮源,固体物料麦麸+玉米粉+米粉与基础培养液3/4 SDAY按2:1均匀混合后,添加0.05%K+可作为菌株固体发酵的最佳物料配方组合。  相似文献

茄链格孢Alternaria solani在体外培养条件下不产生或产生极少量的分生孢子。研究了培养基、菌丝损伤、紫外线照射和温度变化对7个茄链格孢菌株产孢的影响,发现这些处理方法对供试大多数菌株的产孢都有显著影响,确立了茄链格孢大量产孢的最优条件。利用优化后的条件对160株采自不同地区的茄链格孢进行体外诱导产孢,发现120株可大量产孢,占总数的75%,产孢量为1.26×104–5.03×104个/cm2。  相似文献

两步培养法测定真菌产孢营养需求   总被引:2,自引:0,他引:2  
生防真菌产孢条件的测定一般都是通过连续培养方法,即在同一种限定培养基上完成其生长和产孢过程。文中我们提出分离真菌的生长和产孢阶段,测定产孢营养需求的两步培养新方法。6种生防真菌首先在平板上进行营养生长,然后转移至营养成分和浓度确定的新鲜培养基中继续产孢过程来测定菌株产孢阶段实际营养需求。通过与连续培养方法比较,发现只有淡紫拟青霉Paecilomyces lilacinus、金龟子绿僵菌Metarhizium anisopliae二者产孢条件一致,而厚孢轮枝菌Pochonia chlamydosporia、球孢白僵菌Beauveria bassiana、蜡蚧轮枝菌Lecanicillium lecanii、绿色木霉Trichoderma viride菌株产孢的营养条件存在显著的差异。基于这一方法,确定了绿色木霉最佳产孢条件,即起始碳浓度2g C/L,碳氮比10:1,最佳碳氮源组合纤维二糖和酵母浸膏,为真菌生防制剂生产调控提供了依据。  相似文献

玫瑰粟色链霉菌(Streptomyces rosecocastaneus) No. 336变异株对碳源或氮源的利用及其和产葡萄糖异构酶的关系,用半合成培养基进行了摇瓶液体培养试验。测定以15种单精和糖醇、8种寡糖和5种多糖作为碳源,6种含氮化合物作为氮源对菌的生长和产酶的影响。证明N。.336菌株除利用L一阿拉伯糖和D二木糖等五碳糖作为碳源外,也能利用D一葡萄糖等六碳糖作为碳源和能源。糖醇对酶的形成或无明显影响,或有抑制作用。在供试的氮源中,玉米浆和酵母膏对产酶的影响明显优于蛋白胨,该菌几乎不能以无帆氮化合物或乙酸镶作为唯一的氮源。C/N比的试验表明,2:1、1:1或3:1的培养基组成有利于葡萄糖异梅酶的形成,提高氮素含量虽能促进菌的生长,但对产酶不利。以麦麸水解液和玉米浆为主要成分的培养基有利于No.336菌株的生长和产酶,每毫升培养液的酶活力在180单位以上。  相似文献

【目的】明确对蔗根土天牛Dorysthenes granulosus具有良好杀虫活性的金龟子绿僵菌Metarhizium anisopliae JC002菌株生长和产孢所需要的最适营养条件和环境条件。【方法】通过单因素试验,测试了不同培养基、碳源、氮源、温度、pH及紫外线照射不同时间对JC002菌株生长和产孢的影响。【结果】JC002菌株培养基的最佳配方是葡萄糖30 g、蛋白胨15 g、马铃薯200 g、琼脂20 g、水1 000 mL;蔗糖、NaNO3是菌落生长及产孢的最适碳源和氮源;菌株培养的最适温度为25℃,培养基的最适pH值为7.0;紫外光对菌株的生长速率无显著影响,但对产孢量有较大的抑制作用,紫外光照射时间越长,产孢越少。【结论】本研究为JC002菌株的大量培养及其孢子制剂的大量生产和有效利用奠定基础。  相似文献

AIMS: To find a supplemental ingredient that can be added to routinely used growth media to increase conidial production and decrease aflatoxin biosynthesis in small sclerotial (S strain) isolates of Aspergillus flavus. METHODS AND RESULTS: Molasses was added to three commonly used culture media: coconut agar (CAM), potato dextrose agar (PDA), and vegetable juice agar (V8) and production of conidia, sclerotia, and aflatoxins by A. flavus isolate CA43 was determined. The effect of nitrogen sources in molasses medium (MM) on production of conidia, sclerotia and aflatoxins was examined. Water activity and medium pH were also measured. Conidia harvested from agar plates were counted using a haemocytometer. Sclerotia were weighed after drying at 45 degrees C for 5 days. Aflatoxins B(1) and B(2) were quantified by high-performance liquid chromatography. Addition of molasses to the media did not change water activity or the pH significantly. Supplementing CAM and PDA with molasses increased conidial production and decreased aflatoxins. Two-fold increased yield of conidia was found on MM, which, like V8, did not support aflatoxin production. Adding ammonium to MM significantly increased the production of sclerotia and aflatoxins, but slightly decreased conidial production. Adding urea to MM significantly increased the production of conidia, sclerotia and aflatoxins. CONCLUSIONS: Molasses stimulated conidial production and inhibited aflatoxin production. Its effect on sclerotial production was medium-dependent. Water activity and medium pH were not related to changes in conidial, sclerotial or aflatoxin production. Medium containing molasses alone or molasses plus V8 juice were ideal for conidial production by S strain A. flavus. SIGNIFICANCE AND IMPACT OF THE STUDY: Insight into molecular events associated with the utilization of molasses may help to elucidate the mechanism(s) that decreases aflatoxin biosynthesis. Targeting genetic parameters in S strain A. flavus isolates may reduce aflatoxin contamination of crops by reducing the survival and toxigenicity of these strains.  相似文献

对草莓炭疽病具有良好生防效果的菌株MT-06的抑菌谱、生物学特性及其分类地位进行了研究。结果表明:MT-06具有广谱抗菌性,对多种病原菌具有拮抗作用;其营养生长的最适条件是:在PDA或PSA培养基上,28-35℃,初始pH为5,24h全光照;在查氏培养基上,25-28℃,24h全光照条件下孢子产量最高。经形态观察和rDNA-ITS序列及β-tubulin序列分析,将该菌株鉴定为Talaromyces flavus(Klcker)Stolk Samson。  相似文献

Six natural media were examined for growth and sporulation of six isolates of the nematophagous fungus Hirsutella rhossiliensis , using solid and/or liquid culture. Twenty carbohydrates, 19 nitrogen (N) compounds, and nine vitamins were also tested for their effects on growth, sporulation, and spore germination of a further three isolates (ATCC46487, OWVT-1 and JA16-1). Variations in nutritional requirements existed among the fungal isolates. In general, V-8 juice agar (VA), cornmeal agar and potato dextrose agar were good media for growth, and malt extract agar, VA and yeast dextrose agar were good for sporulation of all six isolates. Glycogen was the best and sucrose, inulin, D- ( + ) - trehalose and soluble starch were also good carbon (C) sources for growth and spore germination of the three isolates ATCC46487, OWVT-1 and JA16-1 in both liquid and solid culture. None of the isolates utilized D- ( + )xylose as a C source. L- sorbose, D- ribose, citric acid and D- fructose were poor for growth of all isolates. The best C source for sporulation was D- ( + )-trehalose for ATCC46487, D- sorbitol for OWVT-1 and D- ( + )-cellobiose for JA16-1. Casein was the best N source for growth of ATCC46487 and OWVT-1, while peptone was best for JA16-1. L- asparagine, L- proline, and peptone were also good for growth of all three isolates. L - cystine was not utilized by H. rhossiliensis and DL- methionine inhibited growth of all isolates. Spore germination of all isolates was well supported by most N compounds examined but was inhibited by L- cystine. No significant difference in sporulation of ATCC46487 was observed among the N sources. DL- threonine was the best N source for spore production by OWVT-1 and L- phenylalanine was best for JA16-1. Vitamins generally enhanced fungal growth and sporulation, with thiamine having the greatest influence. Excluding some vitamins individually from the medium containing all other test vitamins sometimes increased growth and/or sporulation of certain isolates.  相似文献

Valdensinia heterodoxa (Sclerotiniacae) is a potential fungal bioherbicide for control of salal (Gaultheria shallon). The effect of culture media, substrates and relative humidity (RH) on growth, sporulation and conidial discharge of V. heterodoxa was determined for two isolates PFC2761 and PFC3027 in vitro. Culture media significantly affected the growth, sporulation, and conidial discharge of V. heterodoxa. Of eight agar media used, colony radial growth was optimal on salal oatmeal agar and salal potato dextrose agar for isolates PFC2761 and PFC3027, respectively; whereas sporulation was at an optimum on salal oatmeal agar for both isolates. Of the eight liquid media tested, mycelial production was highest on wheat bran–salal–potato dextrose broth. Growth on solid substrates greatly stimulated sporulation and conidial discharge of V. heterodoxa. Of the 12 solid substrates used, the greatest numbers of discharged conidia were observed from wheat bran and wheat bran–salal within 14 d of sporulation. Sporulation on solid substrates continued for 42 d. RH significantly affected the sporulation and conidial discharge for both isolates across all solid substrates tested. No conidia were produced or discharged below 93 % RH on wheat bran–salal and millet. With an increase of the RH from 93 to 97 %, sporulation and the number of discharged conidia increased significantly for both isolates on wheat bran–salal, but not on millet.  相似文献

椰心叶甲[Brontispa longissima(Gestro)]是椰子的重要害虫,近年来,该虫在海南岛发生普遍,椰子受害严重。由于椰心叶甲受到自然界中某些致病微生物的侵袭,在受害的椰子树心叶上常可发现椰心叶甲僵虫,并发现大部分僵虫表面长出了霉菌,本研究的目的在于从椰心叶甲僵虫表面的霉菌中分离出绿僵菌,并对分离菌株进行鉴定和致病性测定。从僵虫表面刮下孢子或菌丝体,置于绿僵菌选择性培养基(DOA)上培养,挑出真菌菌落,经纯化后,进行生物学特性、菌落生长速率及产孢量的测定,并从PPDA、OMA、VSA和PDA中筛选菌落生长及产孢最适培养基,同时对所分离的菌株进行对椰心叶甲的致病性测定。结果表明,所有分离菌株均鉴定为金龟子绿僵菌[Metarhizium anisopliae(Metschnikoff)],PPDA是菌落生长及产孢的最适培养基,大多数菌株对椰心叶甲有较强的致病力。选取强毒菌株MA4在田间进行防治效果的初步测定,结果表明,该菌株能显著降低椰心叶甲成虫的虫口密度。这些金龟子绿僵菌菌株是首次从海南的椰心叶甲僵虫中分离到的昆虫病原真菌,该菌对海南的椰心叶甲具有很好的生防潜能。  相似文献

AIMS: To investigate how linoleic acid affects conidial production and sclerotial development in a strictly mitotic Aspergillus parasiticus field isolate as related to improving biocompetitivity of atoxigenic Aspergillus species. METHODS AND RESULTS: We disrupted A. parasiticusDelta12-oleic acid desaturase gene (odeA) responsible for the conversion of oleic acid to linoleic acid. We examined conidiation and sclerotial development of SRRC 2043 and three isogenic mutant strains deleted for the odeA gene (DeltaodeA), either with or without supplementing linoleic acid, on one complex potato dextrose agar (PDA) medium and on two defined media: nitrate-containing Czapek agar (CZ) and Cove's ammonium medium (CVN). The DeltaodeA mutants produced less conidia than the parental strain on all media. Linoleic acid supplementation (as sodium linoleate at 0.3 and 1.2 mg ml(-1)) restored the DeltaodeA conidial production comparable to or exceeding the unsupplemented parental level, and the effect was medium dependent, with the highest increase on CVN and the least on PDA. SRRC 2043 and the DeltaodeA mutants were unable to produce sclerotia on CVN. On unsupplemented PDA and CZ, DeltaodeA sclerotial mass was comparable to that of SRRC 2043, but sclerotial number increased significantly to two- to threefold. Supplementing linoleic acid to media, in general, tended to decrease wild type and DeltaodeA sclerotial mass and sclerotial number. CONCLUSIONS: Linoleic acid stimulates conidial production but has an inhibitory effect on sclerotial development. The relationship between the two processes in A. parasiticus is complex and affected by multiple factors, such as fatty acid composition and nitrogen source. SIGNIFICANCE AND IMPACT OF STUDY: Conditions that promote sclerotial development differ from those required to promote maximum conidial production. Manipulation of content and availability of linoleic acid at different fungal growth phases might optimize conidial and sclerotial production hence increasing the efficacy of biocompetitive Aspergillus species.  相似文献

Time-dependent sporulation, conidial size and germ tube formation in Alternaria tenuissima (Kunze ex Pers.) Wiltshire on different media were studied. The results indicate that maximum number of first spore was formed on potato dextrose agar (PDA) and new leaf extract of pigeon pea followed by old leaf extract of pigeon pea and distilled water (DW). The sporulation was relatively poor on other media. The maximum number of spores in a chain were produced on PDA and albumen +0.9% sodium citrate. Only one spore was seen in DW. According to time-dependent sporulation, the media were divided into three categories: on some media the first spore was formed in 18 h, in another group it took 30–36 h and in the third group the first conidium was formed after 40 h. Four germ tubes were commonly formed on most of the media. The largest conidia were observed on potato dextrose and the smallest ones were seen on DW.  相似文献

Summary This paper gives an account of some cultural and physiological studies of an isolate ofPhytophthora parasitica Dast. var.macrospora Ashby, the causal agent of fruit rot ofAnona squamosa L. Among the various culture media studied, non-synthetic ones like oat meal agar, corn meal agar, lima-bean agar, carrot extract agar, soya-bean extract agar and steamed rice agar were the best, on which the organism showed marked growth and sporulation. Non-synthetic types were poor in this direction. Regarding the effect of various carbon sources, sucrose, lactose, maltose, raffinose, inulin, dextrin, dulcitol, glycogen, rhamnose and xylose supported growth and sporulation of the organism. Sodium nitrate, ammonium nitrate, magnesium nitrate, potassium nitrate, ammonium lactate and asparagin were the best sources of nitrogen. 6.5 was found to be the best pH for the growth and sporulation of the organism.A portion of Senior Author's M.Sc. Aim: To identify media and environmental conditions suitable for rapid mycelial growth and sporulation of Diplocarpon mali. Methods and Results: Liquid shake cultures were used to evaluate effects of media and environmental conditions on mycelial growth and conidial production of D. mali. Carrot sucrose broth (CSB), potato and carrot dextrose broth (PCDB) and potato and carrot sucrose broth (PCSB) were most favourable for rapid mycelial growth. PCDB, PCSB, PCB (potato and carrot broth) and carrot dextrose broth (CDB) were favourable for conidial production. All carbon sources tested and peptone favoured for mycelial growth. Carbon and nitrogen sources tested did not significantly stimulate conidial production. The optimum temperature for mycelial growth and conidial production was 25°C. No mycelial growth occurred at 5 or 30°C, but D. mali survived at these temperatures. Active mycelial growth occurred at pH 5–7, and pH 5–8 was favourable for sporulation. Conclusions: PCDB and PCSB incubated at 25°C for 14 day are recommended for mycelial growth and conidial production of D. mali. Significance and Impact of the Study: The information generated in this study will facilitate mycological and pathological research on D. mali and Marssonina leaf blotch of apple caused by D. mali.  相似文献
