Summary This study investigates the possibility of reusing metal-contaminated equilibrium fluid catalytic cracking (FCC) catalyst after bioleaching. Leaching with Aspergillus niger culture was found to be more effective in the mobilization of nickel from the catalyst particles compared to chemical leaching with citric acid. Bioleaching achieved 32% nickel removal whereas chemical leaching achieved only 21% nickel removal from catalyst particles. The enhanced nickel removal from the catalysts in the presence of A. niger culture was attributed to the biosorption ability of the fungal mycelium and to the higher local concentration of citric acid on the catalyst surface. It was found that 9% of solubilized nickel in the liquid medium was biosorbed to fungal biomass. After nickel leaching with A. niger culture, the hydrogen-to-methane molar ratio and coke yield, which are the measures of dehydrogenation reactions catalysed by nickel during cracking reactions, decreased significantly.  相似文献

Depletion of high-grade ores and presence of significant quantities of metals in low-grade oxide ores has enforced to utilize the overburdens (COB) and wastes (low-grade ores) generated during mining operations. The impact of ore mineralogy and mineral–microbe interaction during bioleaching could not be ignored. Seeking to the need, a systematic study was performed to establish the reaction mechanism involved for recovery of nickel and cobalt from chromite overburden (COB), Sukinda, Orissa using pure culture of Aspergillus niger. Mineralogical analysis reveals a complete conversion of goethite into hematite phase leading to exposure of nickel particles into the micro-pores and cracks developed in the matrix which was initially found to be intertwined in the goethite lattice. As a result, it became more susceptible to attack by the fungal bio acids which in turn accelerate the dissolution rate. Organic acids like oxalic and citric acids were detected in the culture filtrate using HPLC. TEM analysis of the leached samples shows that nickel dissolute into the solution leaving a porous space in the matrix of the hematite by forming nickel oxalate or nickel citrate. Kinetics of the nickel bioleaching was studied to support the mechanism of the reaction. It was observed that the initial rate of reaction follows the chemical control dissolution reaction where as the later part fits to shrinking core model. 18% of nickel and 37.8% of cobalt was recovered from pre-treated COB at 2.5% pulp-density with 10% (v/v) fungal inoculum at 30 °C within 25 days in shake flask while 32.5% of nickel and 86% of cobalt was recovered in bioreactor.  相似文献

Lateritic nickel ore from the Sukinda Mines, Orissa, India, was leached using Thiobacillus ferrooxidans, Bacillus circulans, Bacillus licheniformis and Aspergillus niger at 5% (w/v) solid: liquid ratio for 5–20 days. Maximum leaching of Ni was achieved with B. circulans (85%) and Aspergillus niger (92%) after 20 days. Bacillus circulans showed significantly higher rate of leaching than the other organisms giving 80% Ni extraction after 15 days. The importance and usefulness of heterotrophic organisms in metal extraction are discussed.  相似文献

The leaching process of copper and zinc from copper converter slag with ferric iron in sulfuric acid solutions obtained using the association of acidophilic chemolithotrophic microorganisms was investigated. The best parameters of chemical leaching (temperature 70°C, an initial concentration of ferric iron in the leaching solution of 10.1 g/L, and a solid phase content in the pulp of 10%) were selected. Carrying out the process under these parameters resulted in the recovery of 89.4% of copper and 39.3% of zinc into the solution. The possibility of the bioregeneration of ferric iron in the solution obtained after the chemical leaching of slag by iron-oxidizing acidophilic chemolithotrophic microorganisms without inhibiting their activity was demonstrated.  相似文献

Both the volume and concentration of filtrate ofAspergillus niger andTrichoderma viride influenced the development of the radicles of cocoa seedlings. Radicles placed in Petri dishes containing filter paper moistened with 20 and 30 ml of undiluted filtrate and filtrate dilutions of 1∶1 and 1∶2 failed to develop lateral roots and eventually died. The culture filtrate ofA. niger was more repressive. Radicles in the same volume of filtrate (20 and 30 ml) of higher dilutions (1∶5 and 1∶10) developed lateral roots and survived. Radicles placed in less volume (≤10 ml) survived and produced lateral roots irrespective of concentration of filtrates. Development of the radicles and roots in the control was consistently better (P=0.05) than in filtrate solutions of eitherA. niger orT. viride. The hypocotyls of seedlings under the influence of metabolites ofA niger showed greater cambial activity and formed xylem vessels and tracheids with larger lumina.  相似文献

The present study deals with the production of citric acid from a ram horn peptone (RHP) by Aspergillus niger NRRL 330. A medium from RHP and a control medium (CM) were compared for citric acid production using A. niger in a batch culture. For this purpose, first, RHP was produced. Ram horns were hydrolyzed by treatment with acids (6 N H2SO4, 6 N HCl) and neutralizing solutions. The amounts of protein, nitrogen, ash, some minerals, total sugars, total lipids and amino acids of the RHP were determined. RHP was compared with peptones with a bacto-tryptone from casein and other peptones. The results from RHP were similar to those of standard peptones. The optimal concentration of RHP for the production of citric acid was found to be 4% (w/w). A medium prepared from 4% RHP was termed ram horn peptone medium (RHPM). In comparison with CM, the content of citric acid in RHPM broth (84 g/l) over 6 days was 35% higher than that in CM broth (62 g/l). These results show that citric acid can be produced efficiently by A. niger from ram horn.  相似文献

There are a need for novel, economical and efficient metal processing technologies to improve critical metal sustainability, particularly for cobalt and nickel which have extensive applications in low-carbon energy technologies. Fungal metal biorecovery processes show potential in this regard and the products of recovery are also industrially significant. Here we present a basis for selective biorecovery of Co and Ni oxalates and phosphates using reactive spent Aspergillus niger culture filtrate containing mycogenic oxalate and phosphate solubilized from struvite. Selective precipitation of oxalates was achieved by adjusting phosphate-laden filtrates to pH 2.5 prior to precipitation. Co recovery at pH 2.5 was high with a maximum of ~96% achieved, while ~60% Ni recovery was achieved, yielding microscale polyhedral biominerals. Co and Ni phosphates were precipitated at pH 7.5, following prior oxalate removal, resulting in near-total Co recovery (>99%), while Ni phosphate yields were also high with a recovery maximum of 83.0%.  相似文献

Ochratoxin A is a mycotoxin produced by several Aspergillus and some Penicillium species which may be present in food and feed products. It can be enzymatically hydrolyzed into ochratoxin α and l-β-phenylalanine, thereby decreasing its toxicity. The ochratoxin A degradation capacity of Aspergillus niger is well known and here we report the isolation and purification of a novel enzyme from A. niger that hydrolyzes this mycotoxin. A wheat germ medium supplemented with ochratoxin A was used to produce the enzyme, which was purified from culture filtrate by acetone precipitation and anion exchange chromatography. An overall purification of 2.5-fold with a recovery of 68% and a final specific activity of 36 U/mg was obtained. The enzyme is a metalloenzyme as it was inhibited at 10 mM EDTA, whereas PMSF had no effect. The ochratoxin A hydrolytic enzyme presented a V max of 0.44 μM/min and a K m of 0.5 mM when the reaction was carried out at pH 7.5 and 37°C.  相似文献

Organic acids that are excreted by microorganisms dissolve nickel from lateritic ores. In chemical leaching experiments, fifteen organic acids were tested in the concentration range of 0.05–0.5 M. The most effective were hydroxycarboxylic acids. The leaching of nickel is dependent on the type of mineralization. With completely limonitized ore, no mobilization occurred, while up to 90% of the nickel was extracted from silicate-bearing laterites by 0.5 M citric acid. In biological leaching tests, Penicillium was found to be the most effective microorganism. After improvement of the leaching conditions, up to 70% of the nickel was extracted at considerably lower citric acid concentrations than with the chemical leaching process. Generally, leaching of nickel from lateritic ores with heterotrophic microorganisms is possible and seems to be promising. Possibilities for future investigations are discussed.  相似文献

The continuous production of citric acid from dairy wastewater was investigated using calcium-alginate immobilizedAspergillus niger ATCC 9142. The citric acid productivity and yield were strongly affected by the culture conditions. The optimal pH, temperature, and dilution rate were 3.0, 30°C, and 0.025 h−1, respectively. Under optimal culture conditions, the maximum productivity, concentration, and yield of citric acid produced by the calcium-alginate immobilizedAspergillus niger were 160 mg L−1 h−1, 4.5 g/L, and 70.3% respectively. The culture was continuously perfored for 20 days without any apparent loss in citric acid productivity. Conversely, under the same conditions with a batch shake-flask culture, the maximum productivity, citric acid concentration, and yield were only 63.3 mg L−1 h−1, 4.7 g/L and 51.4%, respectively. Therefore, the results suggest that the bioreactor used in this study could be potentially used for continuous citric acid production from dairy wastewater by applying calcium-alginate immobilizedAspergillus niger.  相似文献

Plasmid pBS501, responsible for the resistance of the wild-type Pseudomonas sp. BS501(pBS501) to cobalt and nickel ions, was conjugatively transferred to the rhizosphere Pseudomonas aureofaciens strain BS1393, which is able to synthesize phenazine antibiotics and to suppress a wide range of phytopathogenic microorganisms. The transconjugant P. aureofaciens BS1393(pBS501) turned out to be resistant to cobalt and nickel with an MIC of 8 mM. When grown in a synthetic medium with 0.25 mM cobalt, the transconjugant accumulated 6 times more cobalt than the wild-type strain BS501(pBS501) (1.2 versus 0.2 g Co/mg protein). Electron microscopic studies showed that cobalt accumulates on the surface of transconjugant cells in the form of electron-opaque granules. In a culture medium with 2 mM cobalt or nickel, strain BS1393 produced phenazine-1-carboxylic acid in trace amounts. The transconjugant P. aureofaciens BS1393(pBS501) produced this antibiotic in still smaller amounts. Unlike the parent strain BS1393, the transconjugant P. aureofaciens BS1393(pBS501) was able to suppress in vitro the growth of the phytopathogenic fungus Gaeumannomyces graminis var. tritici1818 in a medium containing 0.5 mM cobalt or nickel.  相似文献

A process for biological removal of iron from quartz sands, kaolins and clays was developed in which these industrial minerals were leached at 90°C with lixiviant produced as a result of the cultivation of acid-producing heterotrophic microorganisms, mainly strains of Aspergillus niger, at 30°C in a nutrient medium containing molasses as a source of carbon and energy. The lixiviant, i.e. the fermentation fluid, contained oxalic and citric acids as main components and after the cultivation was acidified to a pH of 0.5 by means of hydrochloric acid. The leaching was carried out in mechanically stirred acid-resistant vats for a period of from 1 to 5 hours. The iron content of some sands treated by this method was lowered from 0.035–0.088 to below 0.012% Fe2O3 making them suitable for the preparation of high quality glass. The iron content of different kaolins was lowered from 0.65–1.49 to 0.44–0.75% Fe2O3 and as a result of this their whiteness was increased from 55–87 to 86–92%. The iron content of a clay was lowered from 6.25 to 1.85% Fe2O3 and this increased the fireproofness of the clay from 1 670 to 1 750°C. Similar process was used for leaching of aluminium from aluminosilicates, mainly clays and kaolins. However, after the cultivation the fermentation fluid was acidified either by means of sulfuric or hydrochloric acid or by means of different mixtures of inorganic acids. For enhancing aluminium solubilization the aluminosilicates were heated before leaching at 600–650°C for 1–2 hours. Over 90% of the aluminium present in different clays and kaolins was leached within 3–6 hours in this way. “Silicate” bacteria related to the species Bacillus circulans and B. mucilaginosus were used to leach silicon from low-grade bauxite ores containing aluminosilicates as impurities. The bacterial action was connected with the formation of mucilaginous capsules consisting of expolysaccharides. The solid residues after leaching were characterized by higher values of alumina content and were suitable for processing by means of the BAYER process for recovering aluminium. Heterotrophic bacteria were used to leach manganese from oxide ores using different organic compounds as reducing agents.  相似文献

Ectomycorrhizal fungi can produce antifungal compounds in vitro as well as in symbiosis with the host plant that can reduce root diseases. The objective of this study was to isolate antifungal compounds from culture filtrate of Paxillus sp. 60/92, which can form mycorrhizas with Picea glehnii seedlings. Culture filtrate of Paxillus sp. 60/92 showed antifungal activity against Pythium vexans at pH 3–4 but not at pH 5–10, although sterile MMN-b liquid medium (pH 3–10) did not show antifungal activity. Upon separation of antifungal compounds in the culture filtrate, antifungal activity was detected in the organic acid and water-soluble phenolics fractions adjusted to pH 3. Although antifungal activity of individual fractions was lower than that of the culture filtrate, a mixture of these fractions showed antifungal activity similar to that of the culture filtrate. Furthermore, antifungal activity of oxalic acid, which is known to be produced by Paxillus involutus, was increased by mixing with the water-soluble phenolic fraction. Our findings indicate that Paxillus sp. 60/92 produces organic acids and water-soluble phenolics that together show antifungal activity at pH 3–4 against P. vexans.  相似文献

Copper and cobalt were determined in tissue extracts of five metallophytes from Shaba Province, Zaïre. About 4O% of the cobalt and copper was extractable into deionized water and a further 4O% was extractable in 0.2M hydrochloric acid. It was concluded that copper and cobalt are bound to several different ligands instead of a single ligand as in the case of nickel hyperaccumulators. Proton microprobe studies on the cobalt accumulator Haumaniastrum robertii showed a strong inverse correlation between the spatial distributions of cobalt and potassium, as well as a direct relationship between cobalt and calcium. It is suggested that some of the cobalt may be immobilized with calcium in oxalate crystals.  相似文献

The highest enzymatic extraction of covalent linked chlorogenic (36.1%) and caffeic (CA) (33%) acids from coffee pulp (CP) was achieved by solid‐state fermentation with a mixture of three enzymatic extracts produced by Aspergillus tamarii, Rhizomucor pusillus, and Trametes sp. Enzyme extracts were produced in a practical inexpensive way. Synergistic effects on the extraction yield were observed when more than one enzyme extract was used. In addition, biotransformation of chlorogenic acid (ChA) by Aspergillus niger C23308 was studied. Equimolar transformation of ChA into CA and quinic acids (QA) was observed during the first 36 h in submerged culture. Subsequently, after 36 h, equimolar transformation of CA into protocatechuic acid was observed; this pathway is being reported for the first time for A. niger. QA was used as a carbon source by A. niger C23308. Production of tannase (tannin acyl hydrolase, EC 3.1.1.20) by Aspergillus nigerHA37 on a synthetic culture medium containing tannic acid at different concentrations has been studied. Maximal enzymatic activity increased according to the initial concentration of tannic acid; respectively 0.6, 0.9 and 1.5 enzyme activity units (EU) ml−1 medium in the presence of 0.2%, 0.5% and 1% of tannic acid. Tannase production by A. niger HA37 on fourfold diluted olive mill waste waters (OMWW) as substrate, was between 0.37 and 0.65 EU ml−1. Enzyme production on the diluted OMWW remained globally stable during more than 30 h. Growth of A. niger HA37 on OMWW was correlated with about 70% degradation of phenolic compounds present in the waste. This strain has therefore the capacity to degrade complex wastewaters which cause environmental damage to aquatic streams.  相似文献

Summary The efficiency of a biological bleaching process on a highly contaminated iron oxides kaolin was analyzed. The bio-bleaching method consists of two steps: first, the fermentation of a chemically-defined medium byAspergillus niger leads to a spent liquor which, in a second step, is employed as leaching agent for kaolin. In the leaching process 43% of iron oxides was removed when the kaolin was treated at 60°C for 5 h and the whiteness index was increased to 67%. Extracellular enzymes were not responsible for leaching, which is basically due to organic acids produced during the fermentation.  相似文献

Abstract: Leaching of silicate ores, particularly nickel laterites, with the aid of heterotrophic organisms has been briefly reviewed. Samples of laterite ores from Greece were characterised mineralogically and a number of microorganisms isolated from them. One of these organisms (code FI) was successfully acclimatized to 6400 ppm nickel. Samples of the high-grade Greek Kastoria nickel laterite were leached with sulphuric acid and a number of organic acids. Sulphuric and citric acids extracted over 60 and 40% of the contained nickel, respectively, but the other acids employed were less efficient leachants. Oxalic acid precipitated nickel oxalate. Roughly the same extraction of iron was observed. The main leaching parameter was confirmed to be hydrogen ion concentration, although complexation with organic anions was a contributor. Organism FI (a strain of Penicillium ) was used in comparison with organisms from various culture collections to bioleach nickel from samples of the low-grade Greek Litharakia nickel laterite. The organisms were cultivated in a mixture of a sugar-based nutrient mineral medium and finely ground ore. Several penicillia and aspergilli leached 55–60% of the contained nickel and cobalt, and 25–35% of the iron when sucrose was the carbon source, but FI was not efficient. However, in molasses medium, Fl extracted nearly 40% of the nickel. Biosorption and bioprecipitation reactions were observed. The mechanism of bioleaching or in situ leaching is discussed in terms of close physical and chemical association between the fungal hyphae and mineral phases in the ore. This accounted for the low overall hydrogen ion concentration observed during bioleaching.  相似文献

Summary Copper contained in a solid industrial waste produced in a silicone manufacturing process was leached with spent iron medium from aThiobacillus ferrooxidans culture. Most effective leaching was observed in a continuously fed, dual reactor system. Spent iron medium was generated by growingT. ferrooxidans in 0.9 K iron medium at pH 1.5 in the first reactor, and was transferred to a second reactor in which it leached the copper from the waste. Leaching was effective at a pulp density of the waste material as high as 20%. In experiments run at a pulp density of 2.5%, the spent iron medium was most efficient in leaching copper when it was first diluted 100-fold with a mineral salts solution at pH 1.5. Removal of the copper from the waste appeared to involve its displacement by acid, dissolved mineral salts, and ferric iron. Potentials for practical application of this process are discussed.  相似文献
