Summary DNA-DNA hybridisation experiments show that chloramphenicol induces a burst of initiation from the oriC region of a dnaA46 mutant of Escherichia coli at 36.5° C but not from the isogenic dnaA + strain. Following this stimulation of initiation, DNA replication proceeds normally towards the terminus. The temporal pattern of the extra initiation is in parallel with the induced stimulation of RNA synthesis caused by chloramphenicol in the same strain. This is consistent with the hypothesis that the stimulation of initiation in the dnaA mutant is the result of the stimulation of the synthesis of an RNA species.  相似文献

Summary Two loci,ma-l + andry +, necessary for xanthine dehydrogenase activity inDrosophila melanogaster have been studied for dosage effects utilizing deficiencies and duplications induced for this purpose. Comparisons of one, two and three doses ofma-l + in the female or one and two doses in the male indicate that there is no increase in specific enzyme activity with dose. On the other hand, comparisons of one, two and three doses ofry + in the male and female reveal an increase in enzyme activity that is roughly proportional to dose. Since dosage ofry + is limiting, whereas that ofma-l + is not, the final concentration of xanthine dehydrogenase is shown to depend on the number of doses ofry +.The implications of these findings with respect to the hypothesis of dosage compensation and to the mechanism of control of enzyme and protein concentration are discussed.Operated by Union Carbide Corporation for the U.S. Atomic Energy Commission.  相似文献

Summary A heat resistant mutant of E. coli dnaAts46 was isolated, which grows normally only at temperatures above 39°. After a temperature shift from 42° to 32° the mutant overproduces DNA relative to protein. This is due to overinitiation of rounds of chromosome replication at low temperature, as indicated by hybridization and other experiments. The mutation is cotransduced by Pl with ilv and could not be separated from dnaAts46 by transduction.  相似文献

Summary cur-1, a mutation found in many strains of the Y10 line of Escherichia coli K-12, but not previously described, maps at 27 min, close to galU. cur-1 causes mucoidity or uracil requirement at 29°C depending on the genetic background. The recA + gene of Vibrio cholerae O1 has been cloned, its nucleotide sequence determined and the product characterized. A deletion mutation was constructed in the recA gene and mutants showed the typical sensitivity to UV and to DNA-damaging agents, as well as an inability to mediate homologous DNA recombination. The chromosomal recA deletion mutants in V. cholerae do not show altered virulence in the infant mouse cholera model and are thus ideal strains for use in complementation studies.  相似文献

Summary The sdrA102 mutation confers upon cells the ability to replicate DNA in the absence of protein synthesis. This mutation was combined with the recA200 mutation, which renders the recA protein thermolabile, and had little effect on normal replication. However, the sdrA102 recA200 double mutant exhibited temperature-sensitive stable DNA replication: it replicated DNA continuously in the presence of chloramphenicol at 30°C, whereas at 42°C DNA replication ceased after the DNA content increased only 40–45%. Suppressor mutants (rin; recA-independent) capable of stable DNA replication at 42°C were isolated from the double mutant. The suppressor mutant retained all other recA characteristics, i.e., deficient general recombination, severe UV-sensitivity, and incapability of prophage induction in lysogens. This indicates that the rin mutation specifically suppresses the recA + dependency of stable DNA replication. It is suggested that the recA + protein stabilizes a specific structure, similar to an intermediate in recombination, which may function in the initiation of stable DNA replication.  相似文献

Summary We describe recJ mutants of Salmonella typhimurium. The recJ gene maps between sufD and serA (min 62) and is transcribed counterclockwise. Unlike recJ mutants of Escherichia coli, recJ strains of S. typhimurium are sensitive to irradiation with UV light. This sensitivity is equivalent to or greater that that displayed by recBCD mutant strains. The residual ability of phage P22 abc (anti-recBCD) mutants to form plaques on recBCD + strains is eliminated in recJ hosts. Thus host RecJ function appears to substitute for the anti-RecBCD functions of phage P22 and may serve to limit RecBCD activity.  相似文献

为了提高‘云资粳41’和‘云资粳43’的抗稻瘟病能力,利用农杆菌介导法将二价表达载体pCAMBIA1300-Pi-ta+-Bchi转化到水稻愈伤组织中。经组织培养获得再生苗,再通过氯酚红显色法、PCR检测和抗稻瘟病鉴定法获得抗稻瘟病的转基因植株,为进一步创建持久、广谱抗稻瘟病水稻新材料奠定基础。结果显示:(1)抗性愈伤组织经分化和生根培养后,共获得T0代水稻再生苗137株,其中‘云资粳41’14株,‘中花11’82株,‘云资粳43’41株。(2)经氯酚红显色法和PCR对再生苗检测,‘中花11’、‘云资粳41’、‘云资粳43’的转化苗阳性率分别为66%、43%和55%。证明2个外源基因已经整合到水稻基因组中。(3)对转化阳性植株温室接种稻瘟病病菌66b鉴定结果显示,转基因植株较非转基因植株对稻瘟病的抗性明显增强,而且转Pi-ta+基因和几丁质酶基因双价的水稻植株比转单价Pi-ta+基因或几丁质酶基因的水稻植株抗稻瘟病能力强。(4)氯酚红检测结果存在一定的假阳性,PCR检测结果更真实可靠,但氯酚红显色法方便、快速,结果观察直观,可对大量的转基因植株进行初步筛选。研究表明,转Pi-ta+基因和几丁质酶基因双价基因的水稻植株具有更高的抗稻瘟病能力。  相似文献

When the fission yeastSchizosaccharomyces pombe is starved for nitrogen, the cells are arrested in the G1 phase, enter the G0 phase and initiate sexual development. Theste13 mutant, however, fails to undergo a G1 arrest when starved for nitrogen and since this mutant phenotype is not suppressed by a mutation in adenylyl cyclase (cyr1), it would appear thatste13 + either acts independently of the decrease in the cellular cAMP level induced by starvation for nitrogen, or functions downstream of this controlling event. We have used functional complementation to clone theste13 + gene from anS. pombe genomic library and show that its disruption is not lethal, indicating that, while the gene is required for sexual development, it is not essential for cell growth. Nucleotide sequencing predicts thatste13 + should encode a protein of 485 amino acids in which the consensus motifs of ATP-dependent RNA helicases of the DEAD box family are completely conserved. Point mutations introduced into these consensus motifs abolished theste13 + functions. The predicted Ste13 protein is 72% identical to theDrosophila melanogaster Me31B protein over a stretch of 391 amino acids. ME31B is a developmentally regulated gene that is expressed preferentially in the female germline and may be required for oogenesis. Expression of ME31B cDNA inS. pombe suppresses theste13 mutation. These two evolutionarily conserved genes encoding putative RNA helicases may play a pivotal role in sexual development.  相似文献

Six cultivars of spring barley ( Hordeum vulgare L. cvs Salve, Nümberg II, Bomi, Risø 1508, Mona and Sv 73 608) were grown in water culture for three weeks with various combinations of mineral supply and differential roots/shoot temperatures during the growth period. Most important for growth and accumulation of N, K+, Ca2+ and Mg2+ was the mineral supply, followed by the root temperature and the choice of cultivar. Treatments with low mineral supply or low root temperature induced a uniform reduction in growth and accumulation of the ions studied. The effects of low mineral supply and low root temperature on growth and N accumulation was additive, which indicates that these factors exert their influence independently of each other.
Roots grown at 10°C were smaller and Rb+(86Rb) influx was higher than in roots grown at 20°C. It is suggested that the control of Rb+(86Rb) influx is affected by the root temperature and the age of the plants. The higher 86Rb+ (86Rb) influx into the low temperature roots could not compensate for the smaller root size. However, the lower total mineral accumulation made up for the needs of the smaller plants and cannot explain the reduction in growth.  相似文献

Summary A class of F plasmids, designated Fpoh +, was previously shown to be able to replicate extrachromosomally on Hfr strains by virtue of carrying the specific site or region poh + (permissive on Hfr) of the E. coli chromosome (Hiraga, 1975, 1976a). These plasmids were now found to replicate on E. coli mafA mutants (mafA1 and mafA23) that cannot support vegetative replication of F and some other F-like plasmids. The derivatives of Fpoh + that have lost the poh + site, on the other hand, failed to replicate on mafA mutants. These mutants harboring Fpoh + (but not Poh- derivatives thereof) exhibit abnormal cell division and form elongated cells, presumably due to competition between Fpoh + and the host chromosome for some factor(s) essential for the initiation of DNA replication of the both replicons. It is tentatively concluded that the poh + site is required for F plasmids to replicate on mafA mutants as well as on Hfr strains. In view of the fact that the mechanism of inhibition of autonomous F DNA replication in mafA mutants and in Hfr strains are clearly different, the present data seem to provide strong support to the notion that the poh + region contains the replication origin of the E. coli chromosome.  相似文献

TheSchizosaccharomyces pombe rhp51 + gene encodes a recombinational repair protein that shares significant sequence identities with the bacterial RecA and theSaccharomyces cerevisiae RAD51 protein. Levels ofrhp51 + mRNA increase following several types of DNA damage or inhibition of DNA synthesis. Anrhp51::ura4 fusion gene was used to identify the cis-acting promoter elements involved in regulatingrhp51 + expression in response to DNA damage. Two elements, designated DRE1 and DRE2 (fordamage-responsiveelement), match a decamer consensus URS (upstream repressing sequence) found in the promoters of many other DNA repair and metabolism genes fromS. cerevisiae. However, our results show that DRE1 and DRE2 each function as a UAS (upstream activating sequence) rather than a URS and are also required for DNA-damage inducibility of the gene. A 20-bp fragment located downstream of both DRE1 and DRE2 is responsible for URS function. The DRE1 and DRE2 elements cross-competed for binding to two proteins of 45 and 59 kDa. DNase I footprint analysis suggests that DRE1 and DRE2 bind to the same DNA-binding proteins. These results suggest that the DRE-binding proteins may play an important role in the DNA-damage inducibility ofrhp51 + expression.  相似文献

Summary We have analysed as a function of cell doubling times the control of R6K plasmid replication in rep + and rep strains of Escherichia coli. The rep mutation results in an alteration or loss of an enzyme that unwinds helical DNA. We found in rep + bacteria that R6K relative dosage (plasmids per genome equivalent) remained nearly constant as growth rates increased. From this we concluded that the average plasmid concentration (plasmids per unit cell mass or volume) fell relative to the average concentration of chromosome origins when growth rates increased. In this context, the control of R6K replication is similar to that of other plasmids as seen by different workers. We also found that the relative dosage of R6K in rep mutants is greater than in rep + bacteria when both strains were grown at fast growth rates. This finding was expected since at fast growth rates the number of genome equivalents per unit mass is expected to be lower in rep mutants. Unexpectedly, however, we found the effect of the rep mutation on R6K relative dosage had occurred in a step-like manner at a slow growth rate of about 120 min per generation. This implies that both the relative dosage and concentration of R6K had increased in a step-like manner. We also found that the effect of the rep mutation on R6K concentration was lost at fast growth rates while the effect of the mutation on R6K relative dosage was not lost.  相似文献

We previously identified four nuclear genes (caf1 + to caf4 +) in Schizosaccharomyces pombe, mutations in which can confer caffeine resistance. Here we report the cloning and sequencing of caf1 +, caf2 + and caf4 +. All three genes are allelic to genes (hba1 + , crm1 + and trr1 + , respectively) involved in multidrug resistance mechanisms or in stress response systems. In agreement with this the caffeine-resistant mutants caf1(hba1)-21, caf2(crm1)-3 and caf4(trr1)-83 are also resistant to brefeldin. Disruption of caf1(hba1) + and caf4(trr1) + makes cells sensitive to high pH. The overlapping ranges of pleiotropic effects and the genetic interaction detected between caf1(hba1) + and caf2(crm1) + suggest that the three genes function in interlinked systems. Six cultivars of barley ( Hordeum vulgare L., cvs Salve, Nürnberg II, Bomi, Risø 1508, Mona and Sv 73 608) were exposed for three weeks to combinations of high and low mineral supply and differential root/shoot temperature. For all the parameters tested [fresh and dry weights, contents and levels of N, K+, Ca2+ and Mg2+, and influx of Rb+(86Rb)] the cultivar differences were influenced by the mineral supply, the root temperature and the age of the plants.
The cultivar differences in N nutrition of three-week-old plants could partly be attributed to variation in root size, uptake of N and in use-efficiency of the element. The cultivar variation in root-shoot partitioning of N was small, except when low mineral supply was combined with a low root temperature. Similarly, cultivar differences in contents of K+, Ca2+ and Mg2+ were influenced by variation in uptake, use-efficiency and root/shoot partitioning of the elements. Low root temperature increased cultivar variation in K+, Ca2+ and Mg2+ partitioning.
The modern cultivar Salve was compared with Nürnberg II, which is derived from a German land race. Nürnberg II performed better than Salve when low root temperature and restricted mineral supply were combined. Otherwise Salve grew better, partly due to a more efficient use of N.
Two high-lysine lines, Risø 1508 and Sv 73 608, were compared with their mother lines Bomi and Mona. The differences obtained revealed no general effect of the high-lysine genes on growth and mineral nutrition of up to three-week-old barley plants.  相似文献
