Between October and December of 1996–1999, off eastern Antarctica (60°-150°E), we darted 31 crabeater seals with midazolam and pethidine at estimated dose rates of 0.15–0.4 mg/kg and 1–3 mg/kg, respectively. Maximum sedation was reached at 23 ± 9 min (n = 18) and first signs of recovery were noted at 54 ± 24 min (n = 4). Seals greater than 250 kg body-mass were sedated by administration of approximately 90–100 mg midazolam and 600 mg pethidine, but the degree of sedation was unpredictable and did not permit invasive procedures in some cases. Behavior of the seal and adjacent conspecifics affected the success of procedures and our ability to monitor vital signs. Naloxone and flumazenil reversed sedation, making this combination attractive for use in animals adjacent to water. Additional ketamine was administered to two seals, resulting in improved restraint.  相似文献

Eleven lactating female Weddell seals were immobilised using inhaled isoflurane and oxygen, having initially been sedated using an intramuscular injection of midazolam. The seals were selected from colonies in Long Fjord, East Antarctica. Isoflurane was delivered using a precision, out-of-circle vaporiser in a portable, heated, semi-closed circle system anaesthetic machine. Induction time (time from injection of midazolam to detected maximal effect of midazolam) ranged from 12 min to 29 min. The maximal effect of midazolam was assessed as being either moderate sedation (n=9) or heavy sedation (n=2), and the maximal effect of inhaled isoflurane and oxygen was assessed as being light anaesthesia (n=11). The level of chemical restraint achieved using this combination allowed attachment of heart rate monitoring units and collection of biological samples. Recovery time ranged from 1 min to 11 min. The anaesthetic regime proved a practical, safe and reliable method for the immobilisation of lactating Weddell seals under conditions of low environmental temperature.  相似文献

We immobilized 200–550-kg leopard seals ( Hydrurga leptonyx ) on sea ice in Prydz Bay, Antarctica (68°25'S, 77°10'E) between November 1997 and February 2000. Midazolam (0.18–0.27 mg/kg)/ pethidine (1.0–1.5 mg/kg) was administered by dart to 16 leopard seals. Unpredictable immobilization, poor airway maintenance, and our inability to fully assess the suitability of flumazenil (0.003–0.01 mg/kg), naloxone (0.01–0.013 mg/kg), and naltrexone (0.05–0.12 mg/kg) as reversal agents limited suitability of midazolam/pethidine. Tiletamine/zolazepam 1:1 (0.5–1.5 mg/kg) was, therefore, administered to 19 leopard seals. It produced faster induction (19 ± 3 min), more effective and reliable response to dose (rank correlation: r s= 0.88, n = 18), and better pulmonary ventilation and faster return of cognitive function upon recovery, in comparison to midazolam/pethidine. Best results were achieved with tiletamine/zolazepam (1.2–1.4 mg/kg) which safely immobilized seven of nine seals for 20–30 min. Entry to the water upon darting was minimized, but not eliminated, by the use of lightweight air-pressurized darts and a thorough knowledge of leopard seal behavior.  相似文献

We evaluated the effectiveness and practicality of using isoflurane as an inhalation anesthetic with oxygen as a gas carrier for American martens (Martes americana) in a field setting. Sixty-eight martens were trapped in the Waswanipi Cree Model Forest (Québec, Canada) from October to November 2005 and anesthetized with isoflurane in 100% oxygen (1 l/min) using a face mask. Induction setting of isoflurane was 3% for all animals. Mean (+/-SD) length of induction was 1.8+/-1.2 min. Maintenance isoflurane settings ranged from 1% to 4%. Procedures lasted an average of 16.4+/-7.1 min and were uneventful. Length of recovery, defined as the interval between the end of the procedure and animal release, was short (6.3+/-2.8 min), and well below reported lengths of recovery using injectable anesthetics (>/=70 min). As compared to open drop administration of isoflurane described in previous studies, the use of an anesthesia machine prevents the risk of potential fatal anesthetic overdose. We conclude that among anesthesia techniques currently available, isoflurane with oxygen as a gas carrier is a safe and useful field anesthetic in martens, when issues with equipment portability can be overcome.  相似文献

Isoflurane is a representative inhalant anesthesia used in laboratory animals. However, isoflurane mediates respiratory depression and adverse clinical reactions during induction. In the present study, we established a novel balanced anesthesia method in mice that combined isoflurane anesthesia with midazolam and butorphanol (MB). Thirty-four male C57BL/6J mice received either isoflurane alone or isoflurane with an intra-peritoneal MB premedication (3 mg/kg midazolam and 4 mg/kg butorphanol). The minimum alveolar concentration (MAC) in each group was evaluated. Induction time and adverse clinical reactions were recorded in each group. Core body temperature, heart rate, respiratory rate, and oxygen saturation (SPO2) were assessed before and for 1 h after induction. Premedication with MB achieved a significant reduction in MAC compared with isoflurane monoanesthesia (isoflurane, 1.38 ± 0.15%; isoflurane with MB, 0.78 ± 0.10%; P<0.05). Induction time was significantly shortened with MB premedication, and adverse reactions such as excitement or incontinence were observed less frequently. Furthermore, isoflurane anesthesia with MB premedication caused increase of respiratory rates compared to isoflurane monoanesthesia. No significant decrease of SPO2 was observed in MBI anesthesia, while a decrease in SPO2 was apparent with isoflurane monoanesthesia (baseline, 98.3% ± 1.1; 10 min after induction, 91.8 ± 6.4%; P<0.05). In conclusion, premedication with MB was effective for the mitigation of respiratory depression induced by isoflurane in mice, with rapid induction and fewer adverse clinical reactions.  相似文献

Abstract: We evaluated a chamber and nose cone method of isoflurane delivery for anesthetizing eastern gray squirrels (Sciurus carolinensis; summer n = 43, winter n = 48) and Allegheny woodrats (Neotoma magister; summer n = 24, winter n = 13) for use when pain or stress was possible from sampling procedures. Mean induction time for squirrels (from beginning of isoflurane administration to safe removal from trap), was 4.63 ± 0.58 minutes. Squirrels awoke more quickly in summer (1.40 ± 0.15 min) than in winter (3.62 ± 0.24 min) after removal of the nose cone. We manually restrained woodrats and administered the nose cone for 0.5 minutes to each animal. Woodrats awoke after 4.76 ± 0.58 minutes following the final dose of isoflurane for both seasons. These methods are useful for working with small mammals in the field and provide an appropriate anesthetic when there may be more than slight pain or distress.  相似文献

Summary Stomach contents of crabeater (Lobodon carcinophagus) and leopard (Hydrurga leptonyx) seals collected in the pack ice west of the antarctic Peninsula in August–September 1985 were analyzed. Food remains were found in 7 of 56 crabeater seals and 5 of 29 leopard seals. The primary foods were krill (Euphausia superba) which occurred in all 12 stomachs, and fish (Pleuragramma antarcticum) which occurred in 3. Eleven of the seals with food in their stomachs were collected in the southern portion of Bismark Strait. The incidence of feeding seemed highest in pregnant females. These results, and comparisons with previous collections, suggest that krill were not abundant or widely distributed in the area at the time the seals were collected. The sizes of krill eaten by crabeater and leopard seals were very similar, and were significantly larger than krill found in 2 samples taken by midwater trawls in nearby open water.  相似文献

Information on health parameters, such as antibody prevalences and serum chemistry that can reveal exposure to pathogens, disease, and abnormal physiologic conditions, is scarce for Antarctic seal species. Serum samples from Antarctic fur seals (Arctocephalus gazella, n=88) from Bouvet?ya (2000-2001 and 2001-2002), and from Weddell seals (Leptonychotes weddellii, n=20), Ross seals (Ommatophoca rossii, n=20), and crabeater seals (Lobodon carcinophagus, n=9) from the pack-ice off Queen Maud Land, Antarctica (2001) were analyzed for enzyme activity, and concentrations of protein, metabolites, minerals, and cortisol. Adult Antarctic fur seal males had elevated levels of total protein (range 64-99 g/l) compared to adult females and pups (range 52-79 g/l). Antarctic fur seals had higher enzyme activities of creatine kinase, lactate dehydrogenase, and amylase, compared to Weddell, Ross, and crabeater seals. Antibodies against Brucella spp. were detected in Weddell seals (37%), Ross seals (5%), and crabeater seals (11%), but not in Antarctic fur seals. Antibodies against phocine herpesvirus 1 were detected in all species examined (Antarctic fur seals, 58%; Weddell seals, 100%; Ross seals, 15%; and crabeater seals, 44%). No antibodies against Trichinella spp., Toxoplasma, or phocine distemper virus (PDV) were detected (Antarctic fur seals were not tested for PDV antibodies). Antarctic seals are challenged by reduced sea ice and increasing temperatures due to climate change, and increased anthropogenic activity can introduce new pathogens to these vulnerable ecosystems and represent a threat for these animals. Our data provide a baseline for future monitoring of health parameters of these Antarctic seal species, for tracking the impact of environmental, climatic, and anthropogenic changes in Antarctica over time.  相似文献

We sequenced a portion ( c . 475 bp) of the mitochondrial control region of three species of Antarctic phocid carnivores (Weddell seal, Leptonychotes weddellii , N  = 181; crabeater seal, Lobodon carcinophaga , N  = 143; and Ross seal, Ommatophoca rossii , N  = 41) that live seasonally or permanently in the fast ice and seasonal pack ice of the western Amundsen and Ross seas of western Antarctica. We resolved 251 haplotypes with a haplotype diversity of 0.98 to 0.99. Bayesian estimates of Θ from the program LAMARC ranged from 0.075 for Weddell seals to 0.576 for crabeater seals. We used the values of theta to estimate female effective population sizes ( NEF ), which were 40 700 to 63 000 for Weddell seals, 44 400 to 97 800 for Ross seals, and 358 500 to 531 900 for crabeater seals. We used mismatch distributions to test for historical population size expansions. Weddell seals and crabeater seals had significant, unimodal mean pairwise difference distributions ( P  = 0.56 and 0.36, respectively), suggesting that their populations expanded suddenly around 731 000 years ago (Weddell seals) and around 1.6 million years ago (crabeater seals). Both of these expansions occurred during times of intensified glaciations and may have been fostered by expanding pack ice habitat.  相似文献

The morphology of the principal sections of the gastrointestinal system of two Antarctic seals with different dietary habits, namely, the Weddell seal (Leptonychotes weddellii) and the crabeater seal (Lobodon carcinophagus), has been investigated. Histologically examined by light microscopy, the tissue layers of the gastrointestinal tract of both seals are almost identical to those observed in most other mammals and no major differences in principle organization could be found between the two seal species. The ultrastructure of the gastric and intestinal epithelial cells has been examined and is also closely comparable to that of these cells in other mammals; however, Paneth cells have not been found in our material. In general, therefore, adaptations of the gastrointestinal tract to the aquatic environment or the diet are not obvious at the morphological levels of organization studied. Histochemical differences are found between the two closely related species; mucins of the surface epithelium in the stomach of Weddell seals are highly sulfated, while those in the crabeater seal are not. Mucous neck cells in Weddell seals contain acid mucosubstances, while those of crabeater seals contain neutral ones. Goblet cells in the small and large intestine in Weddell seals contain both neutral and acid mucosubstances. Both mucin types are detected in the crabeater seal; however, the mucins of the colon in the crabeater seal are more highly sulfated than those in the Weddell seal. The ratio of globet cells to enterocytes in the large intestine of crabeater seals is higher than that in Weddell seals. © 1995 Wiley-Liss, Inc.  相似文献

Twenty‐four microsatellite loci were isolated from three species of Antarctic seals (Subfamily Monachinae, Tribe Lobodontini). Eleven loci were cloned from Weddell seal, Leptonychotes weddellii, seven from leopard seal, Hydrurga leptonyx, and six from crabeater seal, Lobodon carcinophagus. Variability was assessed in Weddell seals collected in McMurdo Sound, leopard seals from Bird Island, South Georgia, and crabeater seals sampled in the eastern Ross Sea. All loci were variable in the three species used for cloning and 22 of these loci amplified variable products in the Ross seal, Ommatophoca rossii. Cross‐species amplification was largely successful, with an average of 19 loci amplifying products in other phocids.  相似文献

Harbor seals ( Phoca vitulina ) are small pinnipeds that are widely distributed throughout the temperate coastal regions of the Atlantic and Pacific oceans. We determined birth mass, neonatal growth rates, weaning age, and weaning mass of NE Pacific harbor seals ( P. v. richardsi ) during a capture-recapture study that spanned the nursing period (Sidney Island, British Columbia, Canada). Of 46 harbor seal pups initially captured, 28 were classified as newborns ( i. e. , < 24 h old). Mean body mass of newborns was 11.2 ± SE 0.31 kg. Pups were individually tagged and recaptured throughout the nursing period. Average daily mass gain during the nursing period was 394 ± 26 g. Mean birth mass of males did not differ significantly from females, although pups found with fetal pelage (lanugo) (21.4% of all newborns) were smaller at birth (9.8 ± 0.44 kg) than non-lanugo pups (11.6 ± 0.33 kg). Mean weaning mass was estimated at 23.6 ± 1.2 kg at a mean weaning age of 32 d ± 1.5 d. While birth and weaning masses differed little from the published data for offshore Sable Island harbor seals ( P. v. concolor ), British Columbia harbor seals are characterized by half the daily mass gain and a longer nursing period.  相似文献

Spores of Bacillus cereus ATCC 7004, 4342 and 9818 were obtained in nutrient agar at several pH from 5·9 to 8·3. The optimum pH for sporulation was around 7, but good production of spores was obtained in the range 6·5–8·3. With all three strains, D -values clearly dropped with sporulation pH, decreasing by about 65% per pH unit. z -Values were not significantly modified ( P > 0·05) by this factor. Mean z -values of 7·13 °C ± 0·16 for strain 7004, 7·67 °C ± 0·04 for 4342 and 8·80 °C ± 0·64 for 9818 were obtained.  相似文献

Northern elephant seals, Mirounga angustirostris , undergo an annual moult during which they shed all of their pelage and underlying epidermis. Moulting takes place on land and lasts a mean of 32.0±6.6 days. During this time the mean mass loss of adult females was 24.7±6.1%. Mean body composition at arrival (25.6±4.8%fat) did not differ significantly from that at departure (24.9±3.2%fat). Fat catabolism accounted for 93.6%of derived energy and 41%of mass lost. Approximately 3.5%of total mass loss was associated with the shedding of the pelage and epidermis. Moulting female northern elephant seals express an average daily metabolic rate of 2.0±0.6 times that predicted for adult terrestrial mammals. This energy demand was met by losing 3.0 kg d-1 of total body mass. These energy expenditures suggest that, similar to data for harbour seals, the moult period is a time of relatively low energy expenditure.  相似文献

Selecting the appropriate anesthetic protocol for the individual animal is an essential part of laboratory animal experimentation. The present study compared the characteristics of four anesthetic protocols in mice, focusing on the vital signs. Thirty-two male ddY mice were divided into four groups and administered anesthesia as follows: pentobarbital sodium monoanaesthesia; ketamine and xylazine combined (K/X); medetomidine, midazolam, and butorphanol combined (M/M/B); and isoflurane. In each group, rectal temperature, heart rate, respiratory rate, and O2 saturation (SPO2) were measured, and the changes over time and instability in these signs were compared. The anesthetic depth was also evaluated in each mouse, and the percentage of mice achieving surgical anesthesia was calculated. K/X anesthesia caused remarkable bradycardia, while the respiratory rate and SPO2 were higher than with the others, suggesting a relatively strong cardiac influence and less respiratory depression. The M/M/B group showed a relatively lower heart rate and SPO2, but these abnormalities were rapidly reversed by atipamezole administration. The pentobarbital group showed a lower SPO2, and 62.5% of mice did not reach a surgical anesthetic depth. The isoflurane group showed a marked decrease in respiratory rate compared with the injectable anesthetic groups. However, it had the most stable SPO2 among the groups, suggesting a higher tidal volume. The isoflurane group also showed the highest heart rate during anesthesia. In conclusion, the present study showed the cardiorespiratory characteristics of various anesthetic protocols, providing basic information for selecting an appropriate anesthetic for individual animals during experimentation.  相似文献

Observations on five groups of crabeater seals were conducted between 29 October and 17 November 1985 in the Antarctic pack ice near 66°S 50°E, off Enderby Land. The pups studied were born in the last half of October. Two of them increased in weight at a rate of approximately 4.2kg per day. Pups were weaned in the first half of November at 80–110 kg. The lactation period was 2–3 wk and thus is one of the briefest among pinnipeds. Pups decreased in weight after weaning. The only pup visited after it left the natal floe must have been feeding, as it had lost only 2 kg in a 10–day period during which it moved 13 km. Molt of lanugo appeared to be influenced more by a pup's weight than by whether or not it was weaned.  相似文献

We compared the hemodynamic and respiratory effects, recovery time, and cost of two gas inhalants (isoflurane and sevoflurane) for anesthetic induction and maintenance of beaver (Castor canadensis) during surgery to implant radio transmitters in the peritoneal cavity. Heart rate, respiratory rate, relative hemoglobin saturation with oxygen (SpO2), and body temperature were measured every 5 min for the first 45 min, and arterial blood gas was measured once, 25 min into the anesthetic procedure. Induction for either agent was smooth and rapid. Heart rate and respiratory rate both decreased during the procedure though neither was lower than baseline values reported in the literature for beaver. Relative hemoglobin saturation with oxygen, body temperature, and blood gas variables did not differ between each anesthetic regime. Both inhalants caused slight respiratory acidosis. Recovery time from anesthesia was highly variable (1-178 min) but did not differ statistically between drugs. Sevoflurane costs ($22.30/60 min) were much higher than isoflurane costs ($3.50/60 min). We recommend isoflurane or sevoflurane for anesthetic induction and maintenance of beaver because of the lack of physiologic differences.  相似文献

Summary Faecal material of leopard, crabeater and elephant seals was collected from the vicinity of Davis station, Antarctica. Very few identifiable remains were found in elephant seal droppings. Fish remains, mainly of Pleuragramma antarcticum, were found in both leopard and crabeater seal droppings. The mysid Antarctomysis maxima was also found in crabeater seal droppings and amphipods and decapod crustaceans in leopard seal droppings.  相似文献

Diving physiology and at-sea behavior of a juvenile leopard seal (Hydrurga leptonyx) were opportunistically measured in the Antarctic Peninsula during winter 2002. Total body oxygen stores were estimated from measures of hematocrit, hemoglobin, myoglobin, and total blood volume and were used to calculate an aerobic dive limit (ADL). Movement patterns and diving behavior were measured by equipping the seal with a Satellite Relay Data Logger that transmitted data from 8–31 August 2002. The seal remained in a focal area, in contrast to crabeater seals tracked simultaneously. The seal displayed short, shallow dives (mean 2.0±1.4 min, 44±48 m) and spent 99.9% of its time within the estimated ADL of 7.4 min. The shallow diving behavior contradicts previous diet research suggesting Antarctic krill (Euphausia superba) is the primary prey of leopard seals during the winter months as krill were found at deeper depths during this period. These measurements of diving and movement of a leopard seal provide valuable preliminary data necessary to develop future research on the at-sea behavior of an apex predator in the Antarctic ecosystem.  相似文献
