Macduff, J. H., Hopper, M. J. and Wild, A. 1987. The effectof root temperature on growth and uptake of ammonium and nitrateby Brassica napus L. CV. Bien venu in flowing solution culture.II. Uptake from solutions containing NH4NO3.J. exp. Bot.38: 5366 The effects of root temperature on uptake and assimilation ofNH4+ and NO3 by oilseed rape (Brassica napus L. CV. Bienvenu) were examined. Plants were grown for 49 d in flowing nutrientsolution at pH 6?0 with root temperature decrementally reducedfrom 20?C to 5?C; and then exposed to different root temperatures(3, 5, 7, 9, 11, 13, 17 or 25?C) held constant for 14 d. Theair temperature was 20/15?C day/night and nitrogen was suppliedautomatically to maintain 10 mmol m3 NH4NO3 in solution.Total uptake of nitrogen over 14 d increased threefold between313?C but was constant above 13?C. Net uptake of NH4+exceeded that of NO3 at all temperatures except 17?C,and represented 4765% of the total uptake of nitrogen.Unit absorption rates of NH4+ and of 1?52?7 for NO3suggested that NO3 absorption was more sensitive thanNH4+ absorption to temperature. Rates of absorption were relativelystable at 3?C and 5?C compared with those at 17?C and 25?C whichincreased sharply after 10 d. Tissue concentration of N in theshoot, expressed on a fresh weight basis, was independent ofroot temperature throughout, but doubled between 325?Cwhen expressed on a dry weight basis. The apparent proportionof net uptake of NO3 that was assimilated was inverselyrelated to root temperature. The results are used to examinethe relation between unit absorption rate adn shoot:root ratioin the context of short and long term responses to change ofroot temperature Key words: Brassica napus, oilseed rape, root temperature, nitrogen uptake 相似文献

Root Zone Temperature Effects on Growth and Phosphate Absorption in Rape Brassica napus cv. Emerald 总被引:3,自引:0,他引:3
Solution culture experiments with fodder rape (Brassica napuscv. Emerald) show that reduced root temperatures appear to havelittle effect on phosphate inflow over a wide range of P concentration. At a cool root temperature (10 ?C) plant growth rate was reducedbut this was compensated for by a low root: shoot ratio, sothat inflow remained relatively steady. An increased inflowper unit length of root was only achieved at an elevated roottemperature of 35 ?C. The minimum phosphate concentration towhich plants could lower the culture solution (Cmln) rangedfrom 0.15 to 2.5 mmol m3 according to whether roots wereat a low (5 ?C) or high (35 ?C) temperature respectively. Thetotal phosphorus concentration in tissues was affected by rootzone temperature and at low root temperatures this could bea growth limiting factor. The organic (assimilated) fractionof P in shoot tissues was smaller in low temperature plants. These showed visual symptoms of apparent P deficiency. Levelsof inorganic P in roots may also be a factor in feedback ofcontrol of inflow. Key words: Temperature, Roots, Phosphate, Rape (Brassica napus) 相似文献

Barley plants (Hordewn vulgare L. cv. Atem) were grown fromseed for 28 d in flowing solution culture, during which timeroot temperature was lowered decrementally to 5?C. Plants werethen subjected to root temperatures of 3, 5, 7, 9, 11, 13, 17or 25 ?C, with common air temperature of 25/15 ?C (day/night).Changes in growth, plant total N, and NO3 levels, andnet uptake of NH4+ and NO3 from a maintained concentrationof 10 mmol m3 NH4NO3 were measured over 14 d. Dry matterproduction increased 6-fold with increasing root temperaturebetween 325 ?C. The growth response was biphasic followingan increase in root temperature. Phase I, lasting about 5 d,was characterized by high root specific growth rates relativeto those of the shoot, particularly on a fresh weight basis.During Phase I the shoot dry weight specific growth rates wereinversely related to root temperature between 313 ?C.Phase 2, from 514 d, was characterized by the approachtowards, and/or attainment of, balanced exponential growth betweenshoots and roots. Concentrations of total N in plant dry matterincreased with root temperature between 325 ?C, moreso in the shoots than roots and most acutely in the youngestfully expanded leaf (2?l6?9% N). When N contents wereexpressed on a tissue fresh weight basis the variation withtemperature lessened and the highest concentration in the shootwas at 11 ?C. Uptake of N increased with root temperature, andat all temperatures uptake of NH4+, exceeded that of NO3,irrespective of time. The proportions of total N uptake over14 d absorbed in the form of NH4+ were (%): 86, 91, 75, 77,76, 73, 77, and 80, respectively, at 3, 5, 7, 9, Il, 13, 17,and 25 ?C. At all temperatures the preference for NH4+ overNO3 uptake increased with time. An inverse relationshipbetween root temperature (311 ?C) and the uptake of NH4+as a proportion of total N uptake was apparent during PhaseI. The possible mechanisms by which root temperature limitsgrowth and influences N uptake are discussed. Key words: Hordeum vulgare, root temperature, ammonium, nitrate, ion uptake, growth rate 相似文献

Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown as simulated swards for 71 d in flowing nutrientsolutions with roots at 11 C and shoots at 20/15 C, day/night,under natural illumination. Root temperatures were then changedto 3, 5, 7, 11, 13, 17 or 25 C and the total N2, fixation over21 d was measured in the absence of a supply mineral N. Alltreatments were subsequently supplied with 10 mmol m2NO2 in the flowing solutions for 14 d, and the relativeuptake of N by N2, fixation and NO3 uptake was compared.Net uptake of K+ was measured on a daily basis. Root temperature had little effect on root d. wt over the 35-dexperimental period, but shoot d. wt increased by a factor of3.5 between 3 and 25 C, with the sharpest increase occurringat 711 C. Shoot: root d. wt ratios increased from 25to 68 with increasing temperature at 725 C. N2-fixationper plant (in the absence of NO2 ) increased with roottemperature at 313C, but showed little change above13 C. The ratios of N2 fixation: NO2 uptake over 14d (mol N: mol N) were 0.470.77 at 37 C, 092154at 1117 C, and 046 at 25 C, reflecting the dominanceof NO3 uptake over N2 fixation at extremes of high andlow root temperature. The total uptake of N varied only slightlyat 1125 C (095110 mmol N plant1),the decline in N2 fixation as root temperature increased above11 C was compensated for by the increase in NO 3 uptake. The % N in shoot dry matter declined with decreasing root temperature, from 32% at 13 C to 15% at 3 C. In contrast, concentrationsof N expressed on a shoot water content basis showed a modestdecrease with increasing temperature, from 345 mol m3at 3 C to 290 mol m3 at 25 C. Acclimation of NO3 transport fluxes (influx, efflux)in roots of oilseed rape (Brassica napus L. cv. Bien venu) andtheir sensitivity to growth at low root temperature was studiedin relation to external NO3 supply, defined by constantconcentrations ranging from sub- to supra-optimal with respectto plant growth rate. Plants were grown from seed in flowingnutrient solutions containing 250 mmol m3 NO3at 17°C for 20d, and solution temperature in half the cultureunits was then lowered decrementally over 3 d to 7°C. Threedays later plants were supplied with NO3 at 1, 10, 100or 1000 mmol m3 maintained for 18 d. Dry matter productionwas decreased more by low root zone temperature than low [NO3]e. Root specific growth rates were inversely related to [NO3]eand shoot:root ratios increased with time at [NO3]e between101000 mmol m3. Net uptake of NO3 at 17°Cwas twice that at 7°C, and at both temperatures it doubledwith increasing [NO3]e between 110 mmol m3with further small increases at higher [NO3]e. Mean unitabsorption rates of NO3 between 06 d and 614d were linearly related (r2 of 0.790.99) to log10[NO].Steady-state Q10 (717°C) for uptake between 06d were 0.91, 1.62, 1.27, and 1.10, respectively, at [NO3]eof 1, 10, 100, and 1000 mmol m3, compared with correspondingvalues of 0.98, 1.38, 1.68, and 1.89 between 614 d. Thedata indicated that net uptake rates at 7 and 17°C divergedover time at high [NO3]e. Short-term uptake rates from1 mol m3 NO3 measured at 17°C were higherin plants grown with roots at 7°C than at 17°C; for7°C plants there was a strong inverse linear relationship(r2=0.94) between uptake rate and treatment log10 [NO3]ewhilst rates in 17°C plants were independent of prior [NO3]e. Rates of NO3 influx and efflux under different steady-stateconditions of NO3 supply and root temperature were calculatedfrom dilution of 15N added to culture solutions. Efflux wassubstantial relative to net uptake in all treatments, and wasinversely related to [NO3]e at 17°C but not at 7°C.Ratios of influx: efflux ranged from 1.62.9 at 17°Cand 1.31.8 at 7°C, indicating the proportionatelygreater impact of efflux at low root temperature. Ratios ofefflux: net uptake were 0.531.56 at 17°C and 1.213.58at 7°C. The apparent sensitivities of influx and effluxto steady-state root temperature varied with [NO3]e.Both fluxes were higher at 17°C than 7°C in the presenceof 1001000 mmol m3 NO3 but the trend wasreversed at 110 mmol m3 NO. Concentrations oftotal N measured in xylem exudate were at least 2-fold higherat 7°C compared with 17°C, attributable mainly to higherconcentrations of NO3 glutamine and proline. The resultsare discussed in terms of acclimatory and other responses shownby the NO3 transport system under conditions of limitingNO3 supply and low root temperature. Ten-day old kidney bean plants (Phaseolus vulgaris L. cv. Shin-edogawa)were exposed to 2.0 and 40 parts 106 NO2, and0.1, 0.2, and 0.4 parts 106 O3 alone or in combinationfor 2, 4, and 7 d. The effects of these air pollutants wereexamined with respect to the growth, partitioning of assimilates, nitrogen uptake, soluble sugar content, and root respiration. Decreased dry matter production was significant with all treatmentsexcept 2.0 parts 106 NO2 and 0.1 parts 106 O3.Exposure to mixtures of the gases produced more severe suppressionof growth than exposure to the single gases. Root/shoot ratiowas significantly lowered at 7 d by the gas treatments otherthan 2.0 parts 106 NO2 and 0.1 parts 106 O3. Thetotal nitrogen content of plants was increased by all treatments;the higher percent of nitrogen found with O3 exposure will resultfrom the growth retardation which increases the concentrationof nitrogen in the plants because the absorption of nitrogenby roots was unaffected. The combination of O3 with NO2 significantlydecreased the assimilation of NO2 by the plants. The concentration of soluble sugars in roots was decreased bythe gas treatments. There was a strong positive correlationbetween soluble sugar content and dry weight of the roots harvestedat 7 d. Root respiration was relatively unchanged until 5 dand then decreased significantly at 7 d by 2.0 parts 106NO2 and 02 parts 106 O3. Retarded growth of theroots and the decreased root respiration may be due to diminishedtranslocation of sugars from leaves to roots caused by exposureto air pollutants. The uptake of soil nitrogen was not closelyrelated with root respiration in the case of O3 exposure. Key words: NO2, O3, Phaseolus vulgaris, Growth, Sugars, Root respiration 相似文献

Macduff, J. H., Hopper, M. J., Wild, A. and Trim, F. E. 1987.Comparison of the effects of root temperature on nitrate andammonium nutrition of oilseed rape (Brassica napusL.) in flowingsolution culture. II. Cation-anion balance.J. exp. Bot.38: 1589-1602. The effects of root temperature and form of N nutrition (NH4or NOJ) on the mineral composition of the plant, the balanceof inorganic cation-anion uptake and on the apparent net effluxof H +/OHions from the roots were studied with 49-d-oldoilseed rape (Brassica napusL. cv. Bien venu) in flowing solutionculture. Plants were pre-treated for 14 d at a root temperatureof 5 °C prior to constant root temperatures of 3, 7, 11or 17°C for 14 d, with a common shoot temperature of 20/15°Cday/night. Nitrogen was supplied as NH+44 or NO3 at 10mmol m3. Values of Q10 (7-17°C) for mean unit absorptionrates of all the major nutrient ions (K+ , Mg++ , NH+4, SO4,H2PO4, NO3), except Ca++, were > 2.0 over the first 5 d oftreatment but thereafter were < 1.5; the apparent effectof temperature on uptake rates diminished with time. Under NH+4nutrition, inorganic cation uptake (Mg+ + + K++Ca+ + +NH+4)exceeded inorganic anion uptake (SO4 4+ H2PO4) over 14d at all temperatures, with the proportion of cation uptakeas NH4 remaining constant (0.67-0-68) irrespective of root temperature. The net efflux of H + from the roots approximately balancedNH+4 uptake (1:1) over 14 d at each temperature and also balancedthe difference between the total uptake of inorganic cationsand inorganic anions. Under NO3 nutrition, the sum of the netefflux of OH and the change in the carboxylate contents of plantsover 14 d approximately balanced the sum of NO3 and SO 4reduced in the plant. The majority of the negative charge associatedwith the reduction of NO3 and SO 4 was apparently effluxedas OH, but this fraction was lower at low root temperatures. The results are discussed in terms of mechanisms that have beenproposed to regulate the internal pH of plants. Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown for 71 d in flowing nutrient solutions containingN as 10 mmol m3 NH4NO3, under artificial illumination, with shoots at 20/15°C day/night temperatures and root temperaturereduced decrementally from 20 to 5°C. Root temperatureswere then changed to 3, 7, 9, 11, 13, 17 or 25°C, and theacquisition of N by N2 fixation, NH4+ and NO3 uptakewas measured over 14 d. Shoot specific growth rates (d. wt)doubled with increasing temperature between 7 and 17°C,whilst root specific growth rates showed little response; shoot:root ratios increased with root temperature, and over time at11°C. Net uptake of total N per plant (N2 fixation + NH4++ NO3) over 14 d increased three-fold between 3 and 17°C.The proportion contributed by N2 fixation decreased with increasingtemperature from 51% at 5°C to 18% at 25°C. Uptake ofNH4+ as a proportion of NH4+ + NO3 uptake over 14 d variedlittle (5562%) with root temperature between 3 and 25°C,although it increased with time at most temperatures. Mean ratesof total N uptake per unit shoot f. wt over 14 d changed littlebetween 9 and 25°C, but decreased progressively with temperaturebelow 9°C, due to the decline in the rates of NH4+ and NO3uptake, even though N2 fixation increased. The results suggestthat N2 fixation in the presence of sustained low concentrationsof NH4+ and NO4 is less sensitive to low root temperaturethan are either NH4+ or NO3 uptake systems. Root Hydraulic Conductivity of Two Cactus Species in Relation to Root Age, Temperature, and Soil Water Status 总被引:6,自引:1,他引:5
The effects of root age, temperature, and soil water statuson root hydraulic conductivity (LP) were investigated for twocactus species, Ferocactus acanthodes and Opuntia ficus-indica.The volumetric flux density of water was measured for excisedroot segments, either using negative hydrostatic pressures appliedto the proximal end or using reverse flow of water from theroot to the soil. For both species, LP at 20 ?C increased withroot age, average values reaching a maximum of 3.9 ? 107m s1 MPa1 for F. acanthodes and 5.2 ? 107m s1 MPa1 for O.ficus-indica at 11 to 17 weeksof age; LP subsequently declined with increasing root age forboth species. LP was maximal at a temperature of about 10 ?Cfor the youngest roots (13 weeks), this optimum shiftingto 40 ?C for 8-week-old roots of both species. For older roots(up to 1.5-years-old), LP increased with temperature from 0?C to 50 ?C, with a Q10 of 1.3 between 20 ?C and 30 ?C. At asoil water potential (soil) of 0.016 MPa, root LP wasindependent of the direction of water flow for both species.Depending on root age, LP declined 45- to 500-fold for F. acanthodesand 90- to 800-fold for O.ficus-indica as soil was reduced from0.016 to 1.06 MPa, consistent with a rectifier-likebehaviour with respect to water movement between soil and roots.Incorporation of such responses into water uptake models shouldlead to a better understanding of root function. Single, clonal plants of white clover were grown without inorganicnitrogen in four contrasting day/night temperature regimes, with a 12 h photoperiod, in controlled environments. Root andnodule respiration and acetylene reduction activity were measuredin a flow-through system during both day and night for plantsacclimated to day/night regimes of 23/18, 15/10 and 10/5 ?C.Similar measurements were made on plants acclimated to 20/15?C and stepwise at temperatures from 4 to 33 ?C. Peak rate of ethylene production, nitrogenase-linked respirationand basal root + nodule respiration increased approximatelylinearly from 5 to 23 ?C both in temperature-acclimated plantsand in plants exposed to varying measurement temperatures. Themeasured attributes did not vary significantly between day andnight. Temperatures above 2325 ?C did not further enhancethe rate of ethylene production, which remained essentiallythe same up to the maximum measured temperature of 33 ?C. The measurements of nitrogenase-linked respiration between 5and 23 ?C, during both day and night, demonstrated a constantenergetic cost of acetylene reduction of 2.9 µmolCO2 µmol C2H41,. Over the same temperature range, the approximate activation energy of acetylene reduction was60 kJ mol1. The integrated day plus night nitrogenase-linkedrespiration accounted for 13.416% of the plantsnet shoot photosynthesis in a single diurnal period: there wasno significant effect of temperature between 5 and 23 ?C. The effect of root temperature and form of inorganic nitrogensupply on in vitro nitrate reductase activity (NRA) was studiedin oilseed rape (Brassica napus L. cv. bien venu). Plants weregrown initially in flowing nutrient solution containing 10 µMNH4NO3 and then supplied with either nitrate or ammonium for15 d at root temperatures of 3, 7, 11 or 17 °C. Shoot temperatureregime was similar for all plants; 20/15 °C, day/night.Root NRA was highest when roots were grown at 3 and 7 °C.In laminae and petioles NRA was highest when roots were 11 or17 °C. The plants supplied with ammonium had much lowerlevels of NRA in roots after 5 d than the plants supplied onlywith nitrate. NRA in the laminae of plants supplied with ammoniumwas low relative to that in plants supplied with nitrate onlywhen root temperature was 11 or 17 °C. Values of the apparent activation energy (Ea) of NR, calculatedfrom the Arrhenius equation, in laminae and petioles were differentfrom roots suggesting difference in enzyme conformation. Evidencethat the temperature at which roots were growing affected Eawas equivocal. Three cultivars of spring rape (Brassica napus ssp. oleifera),Tower, Willi and Duplo, were used for a study of induction andgrowth of microspore-derived embryos, Buds, 2.0mm in length, containing uninucleate microspores were harvestedand stored for 14 d at 4 ?C in darkness. Anthers were then removedand cultured on a liquid medium based upon that of Murashigeand Skoog and containing 8% sucrose, 0.5 mg l1 naphthylaceticacid and 0.05 mg l1 benzylaminopurine. Cultures werepre-incubated at 35 ?C for 03 d and then incubated at30 ?C. After a total of 42 d incubation, cultures were scoredfor the presence of macroscopic embryos (12 mm in length)and for the presence of anthers containing abortive embryoidswhich had not developed further. The cultivars differed greatly in terms both of the frequencyof anthers showing induced embryoids and of the final yieldof embryos. Tower showed the highest frequency of induction(maximum 38% of cultured anthers with induced embryoids) whereasthe highest yield (equivalent to 1.1 embryo per cultured anther)was obtained from anthers of the cv. Duplo after a 3 d treatmentat 35 ?C. Yields from the other cultivars were much lower andwere relatively unaffected by the 35 ?C treatment. The Partitioning of Nitrate Assimilation Between Root and Shoot of a Range of Temperate Cereals and Pasture Grasses 总被引:2,自引:1,他引:1
Nitrate reductase activity (NRA, in vivo assay) and nitrate(NO-3) content of root and shoot and NO-3 and reduced nitrogencontent of xylem sap were measured in five temperate cerealssupplied with a range of NO-3 concentrations (0·120mol m3) and three temperate pasture grasses suppliedwith 0·5 or 5 0 mol m3 NO-3 For one cereal (Hordeumvulgare L ), in vitro NRA was also determined The effect ofexternal NO-3 concentration on the partitioning of NO-3 assimilationbetween root and shoot was assessed All measurements indicatedthat the root was the major site of NO3 assimilation in Avenasatwa L, Hordeum vulgare L, Secale cereale L, Tnticum aestivumL and x Triticosecale Wittm supplied with 0·1 to 1·0mol m3 NO-3 and that for all cereals, shoot assimilationincreased in importance as applied NO-3 concentration increasedfrom 1.0 to 20 mol m3 At 5.020 mol m3 NO3,the data indicated that the shoot played an important if notmajor role in NO-3 assimilation in all cereals studied Measurementson Lolium multiflorum Lam and L perenne L indicated that theroot was the main site of NO-3 assimilation at 0.5 mol m3NO-3 but shoot assimilation was predominant at 5.0 mol m3NO-3 Both NRA distribution data and xylem sap analysis indicatedthat shoot assimilation was predominant in Dactylis glomerataL supplied with 0.5 or 5.0 mol m3 NO-3 Avena sativa L., oats, Hordeum vulgare L., barley, Secale cereale L., rye, x Triticosecale Wittm., triticale, Triticum aestivum L., wheat, Dactylis glomerata L., cocksfoot, Lolium multiflorum Lam., Italian ryegrass, Lolium perenne L., perennial ryegrass, nitrate, nitrate assimilation, nitrate reductase activity, xylem sap 相似文献

Nitrate assimilation was examined in two cultivars (Banner Winterand Herz Freya) of Vicia faba L. supplied with a range of nitrateconcentrations. The distribution between root and shoot wasassessed. The cultivars showed responses to increased applied nitrateconcentration. Total plant dry weight and carbon content remainedconstant while shoot: root dry weight ratio, total plant nitrogen, total plant leaf area and specific leaf area (SLA) all increased. The proportion of total plant nitrate and nitrate reductase(NR) activity found in the shoot of both cultivars increasedwith applied nitrate concentrations as did NO3: Kjeldahl-Nratios of xylem sap. The cultivars differed in that a greaterproportion of total plant NR activity occurred in the shootof cv. Herz Freya at all applied nitrate concentrations, andits xylem sap NO3: Kjeldahl-N ratio and SLA were consistentlygreater. It is concluded that the distribution of nitrate assimilationbetween root and shoot of V. faba varies both with cultivarand with external nitrate concentration. Plants of Lolium perenne L. cv. S23 were grown in sand culturesupplied with either ammonium (NH4+) or nitrate (NO3)in an otherwise complete nutrient solution at 12°C or 20°C.Three weeks after germination, plants were clipped weekly tosimulate grazing. After 10 weeks growth all nitrogen (N) wassupplied enriched with 15N to quantify the effects of form ofN supply and temperature on the relative ability of currentroot uptake and remobilization to supply N for laminae regrowth. The form of N supply had no effect on the dry matter partitioning, while at 20°C more dry weight was allocated to laminae regrowthand less to the remaining plant material. The current root uptakeof N, which subsequently appeared in the laminae regrowth, wassimilar for plants supplied with NH4+ or NO3, and bothwere equally reduced at the lower temperature of growth. Remobilizationof N to laminae regrowth was greater for plants receiving NH4+than NO3; remobilization with either form of N supplywas reduced at the lower temperature of growth. Remobilizationwas reduced to a lesser extent at 12°C than current rootuptake. It was concluded that remobilization became relativelymore important in supplying N for regrowth of laminae at lowertemperatures. Cytoplasmic pH (pHc) in Chara corallina was measured (from [14C]stribution)as a function of external pH (pH0)and temperature. With pH0near 7, pHc at 25?C is 7.80; pHcincreases by 0.005 pH units?C1 temperature decrease, i.e. pHc at 5 ?C is 7.90. WithpH? near 5.5, the increase in pHc with decreasing temperatureis 0.015 units ?C1 between 25 and 15?C, but 0.005 units?C1 between 15 and 5?C. This implies a more precise regulationof pHc with variations in pHo at 5 or 15 ?C compared with 25?C. The observed dp Hc/dT is generally smaller than the 0.017units ?C1 needed to maintain a constant H+/OH1,or a constant fractional ionization of histidine in protein, with variation in temperature. It is closer to that needed tomaintain the fractional ionization of phosphorylated compoundsor of CO2HCO3 The value of dpHc/dT has importantimplications for several regulatory aspects of cell metabolism. These include (all as a function of temperature) the rates ofenzyme reactions, the H+ at the plasmalemma(and hence the energy available for cotransport processes),and the mechanism for pHc regulation by the control of bidirectionalH+ fluxes at the plasmalemma. 相似文献

Upland grasslands are a major component of natural vegetationwithin the UK. Such grasslands support slow growing relativelystable plant communities. The response of native montane grassspecies to elevated atmospheric carbon dioxide concentrationshas received little attention to date. Of such studies, mosthave only focused on short-term (days to weeks) responses, oftenunder favourable controlled environment conditions. In thisstudy Agrostis caplllaris L.5, Festuca vivipara L. and Poa alpinaL. were grown under semi-natural conditions in outdoor open-topchambers at either ambient (340µmol mol1) or elevated(680µmol mol1) concentrations of atmospheric carbondioxide (CO2 for periods from 79 to 189 d, with a nutrient availabilitysimilar to that of montane Agrostis-Fescue grassland in Snowdonia,N. Wales. Whole plant dry weight was increased for A. capillarisand P. alpina, but decreased for F. vivipara, at elevated CO2.Major components of relative growth rate (RGR) contributingto this change at elevated CO2 were transient changes in specificleaf area (SLA) and leaf area ratio (LAR). Despite changes ingrowth rate at 680 µmol mol1 CO2, partitioningof dry weight between shoot and root in plants of A. capillarisand P. alpina was unaltered. There was a significant decreasein shoot relative to root growth at elevated CO2 in F. viviparawhich also showed marked discoloration of the leaves and increasedsenescence of the foliage. The Interaction of Genotype, Explant and Media on the Regeneration of Shoots from Complex Explants of Brassica napus L. 总被引:7,自引:0,他引:7
The relative importance of explant type, genotype and growthregulator regime in the determination of shoot regenerationfrequencies from complex explants of Brassica napus L. has beenevaluated. Cotyledon, hypocotyl and stem sections taken fromone spring (Westar) and three winter (Ariana, Cobra, Libravo)varieties of B. napus were cultured on three different growthregulator regimes, 0.5 mg dm3 NAA + 2.0mg dm3BAP, 0.5 mg dm3 NAA + 4.0mg dm3 BAP and 1.0mgdm3 NAA + 4.0mg dm3 BAP. The most significanteffects on shoot regeneration were due to explant type and variety. The regeneration from stem segments was not only two to threetimes higher than from hypocotyls or cotyledons, in all varieties, but the response was also more uniform across the varieties. The explant effect accounted for 4495% of the regenerationresponse. In contrast, the contribution of growth regulatorregime was negligible. Although the growth regulator regimeas an independent effect was unimportant, regeneration fromboth Ariana and Libravo was significantly affected by the interactionof genotype with growth regulator regime. The importance ofboth the high shoot regeneration frequency from stem segmentsand the relative uniformity of response across the four testedgenotypes is discussed with respect to the potential benefitsof using this explant source in Agrobacterium-based transformationexperiments. The freezing tolerance of many plants, such as pea (Pisum sativum),is increased by exposure to low temperature or abscisic acidtreatment, although the physiological basis of this phenomenonis poorly understood. The freezing tolerance of pea shoot tips, root tips, and epicotyl tissue was tested after cold acclimationat 2C, dehydration/rehydration, applications of 104M abscisic acid (ABA), and deacclimation at 25C. Tests wereconducted using the cultivar Alaska, an ABA-deficientmutant wil, and its wildtype. Freezinginjury was determined graphically as the temperature that caused50% injury (T50) from electrical conductivity. Endogenous ABAwas measured using an indirect enzyme-linked immunosorbant assay, and novel proteins were detected using 2-dimensional polyacrylamidegel electrophoresis. The maximum decrease in T50 for root tissuewas 1C for all genotypes, regardless of treatment. For Alaskashoot tips and epicotyl tissue, exogenous ABA increased thefreezing tolerance by 1.5 to 4.0C, while coldtreatment increased the freezing tolerance by 7.5 to14.8C. Cold treatment increased the freezing toleranceof shoot tips by 9 and 15C for wiland wild-type, respectively. Cold acclimationincreased endogenous ABA concentrations in Alaskashoot tips and epicotyls 3- to 4-fold. Immunogold labeling increasednoticeably in the nucleus and cytoplasm of the epicotyl after7 d at 2C and was greatest after 30 d at the time of maximumfreezing tolerance and soluble ABA concentration. Cold treatmentinduced the production of seven, three, and two proteins inshoot, epicotyl, and root tissue of Alaska, respectively. In Alaska shoot tissue, five out of seven novelproteins accumulated in response to both ABA and cold treatment. However, only a 24 kDa protein was produced in wiland wild-type shoot and epicotyl tissues aftercold treatment. Abscisic acid and cold treatment additivelyincreased the freezing tolerance of pea epicotyl and shoot tissuesthrough apparently independent mechanisms that both resultedin the production of a 24 kDa protein. Effect of Salinity on Growth, Nodulation and Nitrogen Yield of Chickpea (Cicer arietinum L.) 总被引:2,自引:0,他引:2
Chickpea cultivar ILC 482 was inoculated with salt-tolerantRhizobium strain Ch191 in solution culture with different saltconcentrations added either immediately with inoculation or5 d later. The inhibitory effect of salinity on nodulation ofchickpea occurred at 40 dS m1 (34.2 mol m3 NaCl)and nodulation was completely inhibited at 7 dS m1 (61.6mol m3 NaCl); the plants died at 8 dS m1 (71.8mol m3 NaCl). Chickpea cultivar ILC 482 inoculated with Rhizobium strain Ch191spcstrwas grown in two pot experiments and irrigated with saline water.Salinity (NaCl equivalent to 14 dS m1) significantlydecreased shoot and root dry weight, total nodule number perplant, nodule weight and average nodule weight. The resultsindicate that Rhizobium strain Ch191 forms an infective andeffective symbiosis with chickpea under saline and non-salineconditions; this legume was more salt-sensitive compared tothe rhizobia, the roots were more sensitive than the shoots, and N2 fixation was more sensitive to salinity than plant growth.
