The endocrine pancreas of the grass lizard, Mabuya quinquetaenia-ta, and of the desert lizard, Uromastyx aegyptia, was investigated histologically and immunohistochemically. In both lizard species four cell types were observed in the endocrine pancreas, namely insulin (B), glucagon (A), somatostatin (D) and pancreatic polypeptide (PP) cells. In both species in B, A and D cells could be detected by their cross-reactivity with antisera raised against mammalian insulin, glucagon and somatostatin. However, these cells showed different tinctorial propertis in the two lizard species. In both species the endocrine tissues were concentrated in the splenic lobe of the pancreas. In the grass lizard the endocrine tissue in the splenic lobe of consisted mainly of B, A and D cells and in the ventral lobe the major cell types were PP and D cells. In the desert lizard, on the other hand, the frequency and the pattern of orientation of B, A and D cells were the same in both the splenic and the ventral lobes, but PP cells in the ventral lobe outnumbered those of the splenic lobe. The PP and D cells scattered in the exocrine parenchyma and the long protrusions which they exhibited suggested that these cell exerted paracrine control on the acinar cells. It is speculated that this control by PP cells may be trophic and by D cells inhibitory.  相似文献

Summary Using histological and immunhistochemical techniques, nine endocrine cell types were observed in the mucosa of the gastrointestinal tract of the toad,Bufo regularis, viz. enterochromaffin, somatostatin, glucagon, pancreatic polypeptide (PP), secretin, gastric inhibitory peptide (GIP), gastrin-C-terminal pentapeptide (GTPP), neurotensin and bombesin cells. The enterochromaffin cells were distributed throughout the gastrointestinal tract except the rectum. Somatostatin, glucagon, PP, secretin, GIP and GTPP cells were observed both in the ileum and bombesin cells only in the pyloric and antral parts of the stomach. Immunostaining of consecutive sections did not reveal more than one polypeptide hormone in any of these cell types. It is concluded from the present results that the toad gastrointestinal mucosa contains endocrine cell types that are more or less homologous to those in the mammal alimentary tract, though some of them exhibit a different topographic distribution.  相似文献

The pre- and post-hatching development and differentiation of the endocrine cells in quail gizzard were examined histologically and immunohistochemically. A total of 158 heads (from 5th d of incubation to adult) were used in this study. The formation of gizzard tubular glands began from 11th d of incubation. 8 kinds of endocrine cells, argyrophil cells, and gastrin releasing polypeptide (GRP)-, 5-hydroxytryptamine (5-HT)-, somatostatin-, glucagon-, avian pancreatic polypeptide (APP)-, neurotensin-, and gastrin-immunoreactive cells were detected in the gizzard. These endocrine cells began to appear from 10th d of incubation. Argyrophil cells and GRP-immunoreactive cells in the gizzard were increased with age. Other kinds of immunoreactive cells were found rarely and irregularly, and some of them were found transitory in the embryonic stage.  相似文献

Serotonin-producing pancreatic endocrine tumours are rare neoplasms which in most cases exhibit malignant biological behaviour. These tumours, in the majority of the well-documented cases, are composed of argyrophil- and argentaffin-positive cells which contain large pleomorphic neurosecretory granules. In contrast, argyrophilic non-argentaffin pancreatic endocrine tumours with tumour cells containing round neurosecretory granules are exceptional. In this study we describe such a tumour not associated with clinical evidence of carcinoid syndrome in a 60-year-old woman. Histological examination revealed tumour extension in pancreatic lymphatic vessels and veins but no evidence of locoregional or distant metastases. Ten months after surgery the patient showed no recurrence of the disease. Immunohistochemistry revealed cytoplasmic serotonin production in the tumour cells which were negative for anti-gastrin, insulin, glucagon, somatostatin, pancreatic polypeptide (PP), vasoactive intestinal peptide (VIP) and ACTH. This study emphasizes the usefulness of combined ultrastructural and immunohistochemical investigations in order to identify and characterize the rare pancreatic endocrine tumours with serotonin production.  相似文献

Ten healthy adult Kilis goat mesenteric lymph nodes were used to examine the general structure of lymph nodes, lymphocytes, plasma cells, reticular cells and reticular fibers using histological methods. We also detected T lymphocytes using anti-CD3 [SP7], anti-CD4 [74-12-4], mouse anti-bovine CD4 [CC30] and mouse anti-bovine CD8 [CC63] monoclonal antibodies (mAb); and B lymphocytes using anti-CD79a [HM57] mAb, macrophages using anti-macrophage [MAC387] mAb and follicular dendritic cells using anti-S100 polyclonal antibody (pAb). The distribution of these cells also was studied. Although the primer antibodies we used for CD3, CD8, CD79a, MAC387 and S100 worked well, the primer antibodies for CD4 were ineffective for paraffin embedded goat lymph nodes.  相似文献

The regional distribution and frequency of pancreatic endocrine cells in ddY mice were studied by an immunohistochemical (peroxidase anti-peroxidase; PAP) method using four types of specific antisera against insulin, glucagon, somatostatin and human pancreatic polypeptide (hPP). In the pancreatic islets, most of insulin-immunoreactive (IR) cells were located in the central portion. Most of glucagon- and somatostatin-IR cells were observed in peripheral regions although a somewhat smaller number of cells were also located in the central regions. HPP-IR cells were randomly distributed throughout the entire islets. In the exocrine pancreas, insulin-, glucagon-, somatostatin- and hPP-IR cells were detected; they occurred mainly among the exocrine parenchyma as solitary cells. Cell clusters consisted of only insulin- or only glucagon-IR cells and were distributed in the pancreas parenchyma as small islets. In addition, insulin- and glucagon-IR cells were also demonstrated in the pancreatic duct regions. Insulin-IR cells were located in the epithelium and sub-epithelial connective tissue regions as solitary cells and/or clusters (3-4 cells), and glucagon-IR cells were mainly located in the epithelium as solitary cells. Overall, there were 63.89+/-5.39% insulin-, 26.52+/-3.55% glucagon-, 7.25+/-2.83% somatostatin- and 1.90+/-0.58% hPP-IR cells. In conclusion, some strain-dependent characteristic distributional patterns of pancreatic endocrine cells were found in the ddY mouse.  相似文献

Galanin is a 29 amino acid peptide, initially isolated from the porcine small intestine. The peptide has been shown to occur in intrapancreatic nerves in close association to the islets. Its effects on islet hormone secretion and its possible mechanisms behind these effects are reviewed. Galanin has been shown to inhibit basal and stimulated insulin secretion both in vivo and in vitro under a variety of experimental conditions. The peptide has also been shown to inhibit somatostatin secretion and the secretion of pancreatic polypeptide (PP). With regard to glucagon secretion, however, results in the literature are not consistent since both stimulatory and inhibitory effects have been reported. A direct interaction with the pancreatic beta-cells has been proposed behind its inhibitory action on insulin secretion, since galanin inhibits insulin secretion from isolated beta-cells from obese, hyperglycaemic, mice. Galanin has thereby also been shown to induce repolarization and to reduce the free Ca2+ concentration, [Ca2+]i. The reduction in [Ca2+]i is probably not due to a direct interference with the voltage-activated Ca2+ channels, since there is no effect of galanin when these channels are opened by depolarization induced by high concentrations of K+. Instead, preliminary studies indicate that galanin activates the K+ channels that are regulated by ATP, in turn inducing a repolarization-induced reduction in [Ca2+]i resulting in reduced insulin secretion. However, the possibility that galanin inhibits the insulin secretory mechanism at a step distal to the regulation of cytoplasmic free Ca2+ concentration should not be overlooked.  相似文献

The ontogenesis of the endocrine pancreas has been a subject of controversy. The discussion essentially was about organogenesis and histogenesis of islets of Langerhans. Now, the endodermic origin seems well established by several experimental approaches. The variation of the aspects of the islets and of the number of endocrine cells are re-called as well as the functional activity during fetal life. Numerous neuropeptides have been found in endocrine pancreas, so the content of the different endocrine cell types is reviewed with respect to the classic hormones.  相似文献

Summary With the use of the Falck-Hillarp histochemical technique for the detection of monoamines, nerve fibre fluorescence is observed throughout the tail of the pancreas of the cat and the arrangement and distribution of the nerve fibres can be studied in both the exocrine and endocrine tissue. In the exocrine pancreas, adrenergic nerve fibres innervate arterioles, larger veins and major pancreatic ducts. Adrenergic nerve fibres also appear to terminate on the non-adrenergic nerve cell bodies of the intrapancreatic ganglia. In the islets of Langerhans, adrenergic nerve fibres innervate both the endocrine cells and blood vessels. Some of the islet cells exhibit fluorescence with the Falck-Hillarp technique and these cells have been identified as alpha cells. In animals treated with reserpine, the fluorescence in nerve fibres and in alpha cells is absent.The author wishes to thank ProfessorG. C. Schofield and Dr.G. C. The regional distribution and frequency of pancreatic endocrine cells in the red-bellied frog, Bombina orientalis, were studied by the immunohistochemical peroxidase anti-peroxidase (PAP) method using five types of specific mammalian antisera to insulin, glucagon, somatostatin, bovine pancreatic polypeptide (PP) and secretin. The frequency was calculated as the mean number of each endocrine cell type/1,000 total cells (including exocrine and endocrine cells) using an automated image analysis process. The percentage of each immunoreactive (IR) cell species to the total IR cell population was also calculated. In the pancreas of the red-bellied frog, all five endocrine cell types were demonstrated. Insulin IR cells were located in the pancreas as single cells or islet-like clusters. The latter were localized in central regions. The insulin-IR cells showed a frequency of 65.40 plus/minus 14.56/1,000 cells. Glucagon IR cells were also detected as single cells or as clusters but in the case of clusters, two distributional patterns were detected - a central core type and a marginally distributed type. They showed an abundance of 32.70 plus/minus 7.32/1,000 cells. Somatostatin-IR cells were dispersed throughout the pancreatic parenchyma as single cells, three to four cells, or clusters. The clusters were located in the marginal regions. The somatostatin-IR cell frequency was 19.40 plus/minus 6.52/1000 cells. PP-IR cells were randomly distributed throughout the pancreatic parenchyma as single cells with a frequency of 14.70 plus/minus 4.92/1,000 cells. Secretin-IR cells were demonstrated as clusters or as single cells, and as clusters they occupied the central regions. They showed a frequency of 39.60 plus/minus 10.36/1,000 cells. This is the first report of the presence of secretin-IR cells in amphibian pancreatic endocrine cells. Overall, there were 37.20 plus/minus 6.84% insulin-, 21.90 plus/minus 5.55% glucagon-, 11.60 plus/minus 4.33% somatostatin-, 8.60 plus/minus 2.72% PP- and 23.40 plus/minus 4.45% secretin-IR cells.  相似文献

Hermaphroditism and sex reversal have not previously been ruled out adequately as possible explanations for the existence of unisexual species of reptiles. Therefore, we examined serial histological sections of the complete urogenital systems of nine F2 specimens belonging to two ontogenetic series of Cnemidophorus exsanguis raised in captivity in isolation from males, as well as the urogenital system of the F1 mother of one of these series. No evidence of spermatozoa or testicular tissue was found in any specimen. Comparative material reveals that the histology of the urogenital tract, including the mesonephric kidney in adults, is similar to that of females of bisexual species of Cnemidophorus. We conclude that C. exsanguis is a unisexual, parthenogenetic species whose normal reproduction does not involve sex reversal, self-fertilization, gynogenesis, hybridogenesis, or spermatozoa in any way whatsoever.  相似文献

Since it is generally accepted that a tight metabolic control might prevent the chronic complications of diabetes mellitus, several systems have been developed in which insulin is administered in a manner that mimicks the characteristics of insulin secretion. 1) In the so-called closed-loop systems, insulin delivery is regulated by the concomitant plasma glucose level. These systems, which involve continuous measurement of plasma glucose concentration are not to date implantable and have been used so far only for short-term studies. 2) By contrast, in the "open-loop systems", no glucose sensor is needed and insulin delivery is pre-programmed to achieve a constant basal infusion with peaks of insulin delivery during the meal periods. These systems have been used in man for several months. The present achievement and limitations of both kinds of systems are discussed in this review.  相似文献
