Adult stem cells are proposed to have acquired special features to prevent an accumulation of DNA-replication errors. Two such mechanisms, frequently suggested to serve this goal are cellular quiescence, and non-random segregation of DNA strands during stem cell division, a theory designated as the immortal strand hypothesis. To date, it has been difficult to test the in vivo relevance of both mechanisms in stem cell systems. It has been shown that in the flatworm Macrostomum lignano pluripotent stem cells (neoblasts) are present in adult animals. We sought to address by which means M. lignano neoblasts protect themselves against the accumulation of genomic errors, by studying the exact mode of DNA-segregation during their division. In this study, we demonstrated four lines of in vivo evidence in favor of cellular quiescence. Firstly, performing BrdU pulse-chase experiments, we localized 'Label-Retaining Cells' (LRCs). Secondly, EDU pulse-chase combined with Vasa labeling demonstrated the presence of neoblasts among the LRCs, while the majority of LRCs were differentiated cells. We showed that stem cells lose their label at a slow rate, indicating cellular quiescence. Thirdly, CldU/IdU- double labeling studies confirmed that label-retaining stem cells showed low proliferative activity. Finally, the use of the actin inhibitor, cytochalasin D, unequivocally demonstrated random segregation of DNA-strands in LRCs. Altogether, our data unambiguously demonstrated that the majority of neoblasts in M. lignano distribute their DNA randomly during cell division, and that label-retention is a direct result of cellular quiescence, rather than a sign of co-segregation of labeled strands.  相似文献

Most sex allocation theory is based on the relationship between the resource investment into male and female reproduction and the consequent fitness returns (often called fitness-gain curves). Here we investigate the effects of resource availability on the sex allocation of a simultaneously hermaphroditic animal, the free-living flatworm Macrostomum lignano. We kept the worms under different resource levels and determined the size of their testes and ovaries over a period of time. At higher resource levels, worms allocated relatively more into the female function, suggesting a saturating male fitness-gain curve for this species. A large part of the observed effect was due to a correlated increase in body size, showing size-dependent sex allocation in M. lignano. However, a significant part of the overall effect was independent of body size, and therefore likely due to the differences in resource availability. Moreover, in accordance with a saturating male fitness-gain curve, the worms developed the male gonads first. As the group size was kept constant, our results contrast with expectations from sex allocation models that deal with local mate competition alone, and with previous experiments that test these models.  相似文献

Mitotic metaphase plates of a new species of the genus Macrostomum were studied using conventional Giemsa staining, Orcein staining and semi-thin sections stained after Richardson. Because of the ease in culturing it, this species has the potential to become the new model organism for developmental and evolutionary studies among the lower metazoa. The chromosome number was found to be 2 n  = 8. Reaching a relative length of 45.9% of the haploid genome size, the chromosomes of one pair were significantly larger than all other chromosomes. Of the smaller pairs, chromosomes of one pair were slightly larger (relative length of 21.3%) than the chromosomes of the remaining two pairs (each with a relative length of 16.4%). All chromosomes were metacentric (2 m  + 2 m  + 2 m  + 2 m ). For the first time, a diploid chromosome set of four pairs was described for the genus, as compared with previous studies showing predominantly 2 n  = 6 for 20 different Macrostomum species.  相似文献

The free-living flatworm Macrostomum lignano is used as a model in a range of research fields—including aging, bioadhesion, stem cells, and sexual selection—culminating in the establishment of genome assemblies and transgenics. However, the Macrostomum community has run into a roadblock following the discovery of an unusual genome organization in M. lignano, which could now impair the development of additional resources and tools. Briefly, M. lignano has undergone a whole-genome duplication, followed by rediploidization into a 2n = 8 karyotype (distinct from the canonical 2n = 6 karyotype in the genus). Although this karyotype appears visually diploid, it is in fact a hidden tetraploid (with rarer 2n = 9 and 2n = 10 individuals being pentaploid and hexaploid, respectively). Here, we report on a phylogenetically informed search for close relatives of M. lignano, aimed at uncovering alternative Macrostomum models with the canonical karyotype and a simple genome organization. We taxonomically describe three new species: the first, Macrostomum janickei n. sp., is the closest known relative of M. lignano and shares its derived genome organization; the second, Macrostomum mirumnovem n. sp., has an even more unusual genome organization, with a highly variable karyotype based on a 2n = 9 base pattern; and the third, Macrostomum cliftonensis n. sp., does not only show the canonical 2n = 6 karyotype, but also performs well under standard laboratory culture conditions and fulfills many other requirements. M. cliftonensis is a viable candidate for replacing M. lignano as the primary Macrostomum model, being outcrossing and having an estimated haploid genome size of only 231 Mbp.  相似文献

Megophthalmidia Dziedzicki is a small leiine genus (Mycetophilidae) with seven species described from the Neotropics and ten species from the Palearctic region. Two species of Megophthalmidia have been reported for North America. Recent collecting of Mycetophilidae in California and Arizona, however, shows current North American diversity of Megophthalmidia is at least on par to other regions of the world. Eight new species of Megophthalmidia are described here, increasing the number of Nearctic Megophthalmidia species to nine. Included is a particularly atypical member of the genus, M. saskia sp. n., which expands the genus concept of Megophthalmidia. Of the two species previously recorded for North America, only one actually belongs in the genus. Megophthalmidia occidentalis Johannsen, is fully described and illustrated. The other named species, M. marceda (Sherman) is illustrated and transferred to the genus Ectrepesthoneura Enderlein. A lectotype is designated for this species. A key to the species of Megophthalmidia of North America is provided. The biology of these flies is not yet known. Three of the new Megophthalmidia species – M. lenimenta, M. misericordia, and M. radiata – are only known to occur within small protected areas within the California State Park and UC Natural Reserve systems.  相似文献

Marinithermus hydrothermalis Sako et al. 2003 is the type species of the monotypic genus Marinithermus. M. hydrothermalis T1(T) was the first isolate within the phylum "Thermus-Deinococcus" to exhibit optimal growth under a salinity equivalent to that of sea water and to have an absolute requirement for NaCl for growth. M. hydrothermalis T1(T) is of interest because it may provide a new insight into the ecological significance of the aerobic, thermophilic decomposers in the circulation of organic compounds in deep-sea hydrothermal vent ecosystems. This is the first completed genome sequence of a member of the genus Marinithermus and the seventh sequence from the family Thermaceae. Here we describe the features of this organism, together with the complete genome sequence and annotation. The 2,269,167 bp long genome with its 2,251 protein-coding and 59 RNA genes is a part of the Genomic Encyclopedia of Bacteria and Archaea project.  相似文献

In the traditional sense, several families of Chlorococcales sensu lato , such as Golenkiniaceae, Micractiniaceae, and Scenedesmaceae contained taxa with bristle formation, whereas the Chlorellaceae contained only genera without bristles. However, phylogenetic analyses of the small subunit (SSU) and internal transcribed spacer (ITS) rDNA sequences have shown that the genera Micractinium, Diacanthos (formerly Micractiniaceae) and Didymogenes (formerly Scenedesmaceae) are closely related to the genus Chlorella . The bristle formation within the Chlorella -clade is originated independently in four lineages: Micractinium pusillum, Diacanthos belenophorus, Didymogenes anomala , and Micractinium parvulum (also known as Golenkinia minutissima ). The latter species is to exclude from the genus Micractinium . Consequently, we proposed the new genus Hegewaldia and transferred M. parvulum to this genus. In contrast, Diacanthos belenophorus is closely related to Micractinium pusillum . As a result, the new combination ( Micractinium belenophorum comb. nov.) is proposed. Comparisons of the secondary structure of ITS-1 and ITS-2 rDNA sequences among the strains of Didymogenes and Hegewaldia are provided to support the species concept in these genera.  相似文献

Theory predicts that partially asexual organisms may make the “best of both worlds”: for the most part, they avoid the costs of sexual reproduction, while still benefiting from an enhanced efficiency of selection compared to obligately asexual organisms. There is, however, little empirical data on partially asexual organisms to test this prediction. Here we examine patterns of nucleotide diversity at eight nuclear loci in continentwide samples of two species of cyclically parthenogenetic Daphnia to assess the effect of partial asexual reproduction on effective population size and amount of recombination. Both species have high nucleotide diversities and show abundant evidence for recombination, yielding large estimates of effective population sizes (300,000–600,000). This suggests that selection will act efficiently even on mutations with small selection coefficients. Divergence between the two species is less than one-tenth of previous estimates, which were derived using a mitochondrial molecular clock. As the two species investigated are among the most distantly related species of the genus, this suggests that the genus Daphnia may be considerably younger than previously thought. Daphnia has recently received increased attention because it is being developed as a model organism for ecological and evolutionary genomics. Our results confirm the attractiveness of Daphnia as a model organism, because the high nucleotide diversity and low linkage disequilibrium suggest that fine-scale mapping of genes affecting phenotypes through association studies should be feasible.  相似文献

Sex allocation (SA) theory for simultaneous hermaphrodites predicts an influence of group size on SA. Since group size can vary within an individual's lifetime, this can favor the evolution of phenotypically plastic SA. In an emerging comparative context, we here report on SA plasticity in three closely related Macrostomum flatworm species, namely Macrostomum janickei, Macrostomum cliftonensis, and Macrostomum mirumnovem. For each species, we experimentally raised worms in three group sizes (isolated, pairs, and octets) and two enclosure sizes (small and large) in all factorial combinations and studied the effects of these factors on different estimates of SA. In addition, we also evaluated whether isolated worms engage in self‐fertilization. We found that all species have plastic SA, with M. cliftonensis being more plastic than the other two species, as assessed by comparing standardized effect sizes of (a) the presence/absence of mating partners and (b) the strength of sexual competition. Moreover, we found that sperm production rate—but not sperm morphology—is plastic in M. cliftonensis, and that only M. mirumnovem self‐fertilized during our observation period. Our study suggests that both SA and SA plasticity can diverge even between closely related species.  相似文献

Evolutionary theory predicts that males should produce more sperm when sperm competition is high. Because sperm production rate is difficult to measure in most organisms, comparative and experimental studies have typically used testis size instead, while assuming a good correspondence between testis size and sperm production rate. Here we evaluate this common assumption using the marine flatworm Macrostomum lignano, in which we can estimate sperm production rate because the accumulation of produced sperm can be observed in vivo. In earlier studies we have shown that testis size is phenotypically plastic in M. lignano: worms can be induced to make larger testes by raising them in groups instead of pairs, and these larger testes have a higher cell proliferation activity (i.e. they are more energetically costly). Here we demonstrate that worms with such experimentally enlarged testes have a higher sperm production rate. Moreover, although testis size and sperm production rate were related linearly, worms with experimentally enlarged testes had a higher sperm production rate per unit testis size (i.e. a higher spermatogenic efficiency). We thus show that phenotypically plastic adjustment of sperm production rate includes a component that is independent of testis size. We discuss possible reasons for this novel finding, and suggest that the relationship between testis size and sperm production needs to be evaluated in other species as well.  相似文献

Spermiogenesis in Macrostomum lignano (Macrostomorpha, Rhabditophora) is described using light‐ and electron microscopy of the successive stages in sperm development. Ovoid spermatids develop to highly complex, elongated sperm possessing an undulating distal (anterior) process (or “feeler”), bristles, and a proximal (posterior) brush. In particular, we present a detailed account of the morphology and ontogeny of the bristles, describing for the first time the formation of a highly specialized bristle complex consisting of several parts. This complex is ultimately reduced when sperm are mature. The implications of the development of this bristle complex on both sperm maturation and the evolution and function of the bristles are discussed. Fertilization success of free spawning organisms such as Mytilus species depends on gamete interactions. Therefore, gamete traits such as sperm movement are important for determining fertilization success in free spawning organisms. Since little is known about sperm movement pattern in Mytilus species, the purpose of this study was to investigate sperm movement pattern of blue mussel M. edulis, M. trossulus and their hybrids using computer-assisted sperm movement video analysis. Sperm of all genotypes were found to conduct circular movement in a two-dimensional plane. Furthermore, new sperm movement parameters, real time radius (R), angle change rate (θ) and the center of circular track (O(t)) were developed to verify and quantitatively describe the plane circular movement pattern using software (Image-J) that may be widely applied to sperm movement study in other organisms. Angle change rate was positively correlated to fertilization success. However, no correlation between fertilization and real time radius was detected. Although no interspecific differences were found in the radius, the F1 (first generation) hybrid sperm had a lower angle change rate than M. edulis and M. trossulus. Published studies have shown that sperm circular movement is more prevalent in aquatic broadcast spawning species than in species with mating behavior or internal fertilization. Therefore, a two-dimensional circular movement pattern in sperm may represent a trait that increases fertilization success for broadcast spawning species by either increasing gamete interaction rate at a small scale and/or avoiding swimming further away from the eggs before sperm detects the chemoattractant gradient.  相似文献

A novel moderately thermophilic, facultatively anaerobic chemoorganotrophic bacterium strain P3M‐2T was isolated from a microbial mat developing on the wooden surface of a chute under the flow of hot water (46°C) coming out of a 2775‐m‐deep oil exploration well (Tomsk region, Russia). Strain P3M‐2T is a moderate thermophile and facultative anaerobe growing on mono‐, di‐ or polysaccharides by aerobic respiration, fermentation or by reducing diverse electron acceptors [nitrite, Fe(III), As(V)]. Its closest cultivated relative (90.8% rRNA gene sequence identity) is Ignavibacterium album, the only chemoorganotrophic member of the phylum Chlorobi. New genus and species Melioribacter roseus are proposed for isolate P3M‐2T. Together with I. album, the new organism represents the class Ignavibacteria assigned to the phylum Chlorobi. The revealed group includes a variety of uncultured environmental clones, the 16S rRNA gene sequences of some of which have been previously attributed to the candidate division ZB1. Phylogenetic analysis of M. roseus and I. album based on their 23S rRNA and RecA sequences confirmed that these two organisms could represent an even deeper, phylum‐level lineage. Hence, we propose a new phylum Ignavibacteriae within the BacteroidetesChlorobi group with a sole class Ignavibacteria, two families Ignavibacteriaceae and Melioribacteraceae and two species I. album and M. roseus. This proposal correlates with chemotaxonomic data and phenotypic differences of both organisms from other cultured representatives of Chlorobi. The most essential differences, supported by the analyses of complete genomes of both organisms, are motility, facultatively anaerobic and obligately organotrophic mode of life, the absence of chlorosomes and the apparent inability to grow phototrophically.  相似文献

It has long been thought that the genera Mobiluncus and Falcivibrio contain the same organisms. Using a polyphasic approach, it was found that Mobiluncus curtisii and Mobiluncus mulieris were the same as Falcivibrio vaginalis and Falcivibrio grandis, respectively. As the genus name Mobiluncus takes precedence, it is proposed that F. vaginalis and F. grandis be transferred to the genus Mobiluncus. In agreement with previous studies, results from phenotypic tests did not support the separation of M. curtisii strains into its two subspecies, M. curtisii subsp. curtisii and M. curtisii subsp. holmesii. Phenotypic complexity within M. curtisii dictates that the species should be treated as a complex until more in-depth analyses of the species have been performed. Phylogenetic analyses, based on 16S rRNA gene sequences, demonstrated that the genus Mobiluncus was associated with Varibaculum cambriense and the two subspecies of Actinomyces neuii, and that A. neuii is only distantly related to Actinomyces sensu stricto.  相似文献

大口涡虫属所有物种均为雌雄同体,具一套交配器官。作者于2015年在广东省的两处淡水环境,首次发现2个具有双雄性交配器官的大口涡虫(Macrostomum sp.)标本。通过对活体、整装片、连续组织切片的显微镜观测,发现2只标本的两套交配器官呈左右排列;每套交配器官具备完整的假储精囊、储精囊、颗粒囊与交配刺;假储精囊与储精囊内具有精子;雄孔分别为1个与2个;交配刺的结构与中国已经记录的物种都不相同。本研究对其做了较为详细的描述,并初步探讨了大口涡虫多交配器官发生的原因。  相似文献
