灵芝液体发酵条件的优化研究   总被引:2,自引:0,他引:2  
通过单因素和正交试验,优化灵芝5760深层培养的营养需求与环境条件对灵芝菌丝体生物量及次生代谢产物发酵液多糖的作用。结果表明,适宜灵芝液体菌种培养和液体发酵的培养基配方为麦麸粉5%,黄豆饼粉3%,KH2PO40.10%,MgSO40.09%;适宜的环境条件为初始pH5.5,装液量32%,生物量最大时为第8~9d;适宜接种的种龄为6d,接种量为10%;发酵周期6d,搅拌转速150r/min,消沫剂添加量可为0.20%。从硒对灵芝5760菌丝体生长及深层发酵生物量的影响,灵芝5760富硒培养适宜浓度为100μg/mL;从加硒时间对生物量的作用,在菌龄适宜时期加入即第1d。  相似文献

为探究不同野生灵芝的主要活性成分以及对野生灵芝的开发利用价值,对13种野生灵芝菌株在同一条件下进行液体发酵,采用化学分析的方法,比较菌丝体胞内三萜和多糖的含量差异。结果显示,13种灵芝菌株的三萜和多糖含量有很大差异,其中无柄紫灵芝Ganoderma mastoporum、亮盖灵芝G. lucidum和树舌灵芝G. applanatum的三萜含量较高;树舌灵芝G. applanatum、紫芝G. sinense和褐灵芝G. brownii的多糖含量较高。目前国内广泛栽培灵芝G. lingzhi的野生菌株发酵产物中的三萜和多糖含量并不是最高的,研究结果表明不同种类的野生灵芝还有进一步挖掘的潜在价值。  相似文献

对3个樟芝野生菌株和5个物理诱变菌株进行液体发酵,通过测定发酵液黏度变化情况,观测培养性状、菌球及粗提物产量和多糖、蛋白质及三萜含量,从中筛选出适宜液体发酵的多糖、三萜及蛋白质高产优良菌株。试验结果显示:供试的8个樟芝菌株生长曲线基本一致,其中,菌株327和Gg生长速度较快,培养的第9–10天开始出现菌丝自溶现象;经过筛选,327菌株为多糖和蛋白质高产菌株,多糖和蛋白质的产量分别比出发菌株提高238.20%和10.33%;Gg为三萜高产菌株,三萜产量比出发菌株提高57.86%。野生樟芝菌株经物理诱变效果明显。  相似文献

对灵芝MP-01菌株在三种培养基上的生长性状进行了研究,并对其液体发酵过程中的pH值变化、发酵全液的多糖含量变化、菌丝生物量及发酵培养性状等进行了观测。结果表明,玉米面培养基是灵芝MP-01菌株液体发酵的适宜培养基,其适宜的终止发酵时间在培养的第6 d,此时发酵液多糖含量达到10.32 mg/mL。  相似文献

狭长孢灵芝Ganoderma boninense是最重要的木生真菌之一,主要分布于热带地区,在马来西亚和印度尼西亚是油棕榈树的严重病原菌。本研究以分离自马来西亚雪兰莪州而榄的油棕榈种植园中,生长于油棕榈活树基部狭长孢灵芝子实体的3株菌株为材料,对其抗氧化活性进行了评估。在液体培养的第2、4、6、8、10和12天分别取菌丝体和发酵液,测定菌丝体生物量及发酵上清液中粗多糖、多酚、黄酮和抗坏血酸的含量,并对发酵上清液的相关抗氧化性能指标进行了分析。3个菌株的发酵上清液均表明狭长孢灵芝具有良好的抗氧化活性,但不同菌株在某些次级代谢产物含量和抗氧化指标上表现出强弱程度及出现时间的差异。狭长孢灵芝的多糖和多酚含量以及铁离子还原能力均优于另外两个重要的药用木生真菌,即灵芝Ganoderma lingzhi和栎生桑黄Sanghuangporus quercicola。本研究表明狭长孢灵芝具有开展人工栽培和进行更多药用研究的价值。  相似文献

筛选茯苓高产胞内多糖和胞内三萜的优良液体发酵出发菌株。采用PDA富集固体平板培养与液体发酵培养测定菌丝体生长速率;采用液体发酵策略分析16种茯苓菌株产胞内多糖与胞内三萜的潜能。实验结果表明菌株生长于固体培养基与种子培养基的生长速率之间没有关联性;降低一级种子培养基初始pH值到4.0时能有效缓解茯苓菌株培养物褐化现象;AS5.137胞内多糖含量最高,达377.60±0.10 mg/g,而DB菌株显示出最高的胞内多糖产量,达1.01±0.13 g/L;Y1菌株胞内三萜含量最高,达83.89±4.28 mg/g,而Jingzhou28菌株胞内三萜产量最高,达136.63±26.66 mg/L。就生产茯苓胞内多糖与胞内三萜而言,AS5.137与DB菌株适合作为液体发酵产胞内多糖的出发菌株;Y1,Jingzhou28,Z(z)与Xingpinzhong菌株均较适合作为液体发酵产胞内三萜的出发菌株。  相似文献

Submerged cultures of Ganoderma lucidum are used to produce fungal mycelium, which is used as a functional food and in the production of various triterpenoids, including ganoderic acids (GAs). Specific culture approaches that produce fungal mycelium with high levels of GAs and good biological activity are critical in the functional food industry. In this study, a solid-medium culture approach to producing mycelium was compared to the submerged culture system. Production of GAs, biomass, intracellular polysaccharides, and cytotoxicity of the cultured mycelium were compared as between solid and submerged culture. Growing G. lucidum strains on solid potato dextrose agar medium increased biomass, the production of ganoderic acid 24 (lanosta-7,9(11), 24-trien-3α-o1-26-oic acid), GAs, and total intracellular polysaccharides as compared to fungi grown in submerged culture. Triterpenoid-enriched methanol extracts of mycelium from solid-medium culture showed higher cytotoxicity than those from submerged culture. The IC(50) values of methanol extracts from solid-medium culture were 11.5, 8.6, and 9.9 times less than submerged culture on human lung cancer cells CH27, melanoma cells M21, and oral cancer cells HSC-3 respectively. The squalene synthase and lanosterol synthase coding genes had higher expression on the culture of solid potato dextrose medium. This is the first report that solid-medium culture is able to increase GA production significantly as compared to submerged culture and, in the process, produces much higher biological activity. This indicates that it may be possible to enhance the production of GAs by implementing mycelium culture on solid medium.  相似文献

采用水、3%草酸和0.2mol/L氢氧化钠为溶剂分别提取深层培养的云芝〔Polystictusversicolor(L.)Fr.〕菌丝体蛋白多糖,其得率分别为菌丝体干重的20%、49%和19%。提取的粗多糖经SephadexG100柱层析进一步分离、纯化,纯化的多糖经聚丙烯酰胺凝胶电泳分离后呈现一个斑点,在SephadexG100柱层析中有一个对称峰,紫外及红外光谱分析和蛋白质含量测定表明该多糖为蛋白多糖,其蛋白质和多糖含量在水提多糖中分别为13%和62%、在酸提多糖中分别为5%和91%。在碱提多糖中分别为3%和81%。组成该多糖的主要单糖是葡萄糖和木糖。  相似文献

The basidiomycetes Ganoderma lucidum, Hericium erinaceus, Lentinus edodes and Trametes versicolor were used for preparation of aqueous extracts. A polysaccharide preparation (VPG) was isolated from the G. lucidum aqueous extracts. The mycelium was grown under submerged conditions according to an original procedure. Preliminary exposure of mice with tumors to cyclosphosphamide in a low dose for prolonged elimination of T-suppressors and rapid recovery of T-killers induced an increase in the efficacy of the H. erinaceus and L. edodes extracts. Investigation of the aqueous extracts and VPG on different tumor strain lines for the potential Modifiers of Biological Response (Ca755, s/c P388, s-180) demonstrated antitumor activity and satisfactory tolerabily after oral administration. Inhibition of the tumor growth by the H. erinaceus and T. versicolor extracts and VPG amounted to 88-99% and that of s-180 treated with the L. edodes aqueous extract amounted to 66-75%. Compositions 1, 2, 4 amd 5 were significantly more active by the duration and value of the effect on the animal tumor nodes as compared to the aqueous extracts and VPG included to the compositions and composition 4. Composition 5 (T. versicolor + H. erinaceus + G. Lucidum) proved to be the most efficient by all the criteria. The results of the design of the technologies for cultivation of the mycelum of the medicinal basidiomycetes, investigation of the antumor properties of the extracts and polysaccharide fraction of the mycelium and development of efficient compositions on their basis are summarized. Composition 5 proved to be the most promising for the clinical trials.  相似文献

通过鹅膏菌抑菌活性物质对杨树烂皮病病原菌金黄壳囊孢菌的菌丝生长及其孢子抑制效果的研究,分析鹅膏菌的抗真菌活性。将培养15 d的鹅膏菌液体发酵物分成培养液和菌丝体两部分,然后分别将培养液、菌丝体提取物和发酵液本身对金黄壳囊孢菌菌丝生长和孢子萌发抑制效果进行检测。结果表明:培养液对金黄壳囊孢菌菌丝生长的抑制率最高,为68.90%;三者对孢子萌发都有明显的抑制效果,即在处理后10 d孢子都没有萌发。菌丝体在超声波、加热和浸泡的条件下,利用乙醇,丙酮、乙酸乙酯和正丁醇进行提取,其提取物用于病原菌的抑制效果检测,结果表明:用乙醇和丙酮提取时,超声波、浸泡及加热处理,对金黄壳囊孢菌的抑制效果都与对照有显著差异(a=0.01),而正丁醇的提取物的抑制效果较差,与对照没有显著差异。通过不同提取物对金黄壳囊孢菌菌丝生长和孢子萌发的抑制效果进行分析,并对抑菌活性物质提取工艺进行筛选,得到抑菌活性物质提取的最佳方法为摇床培养,乙醇是最适提取溶剂,处理方式(超声波、浸泡或者加热)相互间差异不显著。  相似文献

对深层发酵培养的姬松茸菌丝体依次采用热水、冷碱、热碱提取,得到了四种主要的多糖组分。其中冷碱提取的水不溶性多糖(CAASP)得率最高,达8.5 8g/ 10 0g干菌,且不同的多糖组分具有不同的单糖组成。其中热水提取的水溶性多糖(HWSP)及冷碱提取的水溶性多糖(CAWSP)在2 0mg/kg·d的剂量下对小鼠S180 移植瘤显示出较高的抗肿瘤活性,抑瘤率分别高达5 9.84 %和6 4 .6 6 %。同时发现在四种多糖在发挥抗肿瘤作用过程中,小鼠的脾脏指数和胸腺指数有不同程度的提高。并且各多糖活性组分在体外均具有显著的刺激脾淋巴细胞增殖的作用  相似文献

响应面设计法优化蕨渣基质的灵芝培养条件   总被引:1,自引:0,他引:1  
探索灵芝在以蕨渣为主要成分的固态基质中的培养条件,可为蕨渣的开发利用提供理论依据。以蕨渣为主要原料,采用响应面法对灵芝培养条件(基质蕨渣比例、基质含水量和培养温度)进行优化。结果表明,基质蕨渣比例、基质含水量和培养温度对灵芝菌丝日平均生长速率均有极显著的影响(p<0.01),且基质含水量与培养温度之间、基质蕨渣比例与基质含水量之间存在交互作用。优化出灵芝培养条件为蕨渣比例85%,基质含水量62.5%,培养温度27℃,在此条件下,灵芝菌丝日平均生长速率为3.48mm/d。多元回归分析结果显示,基质蕨渣比例、基质含水量、培养温度与菌丝日平均生长速率之间回归模型高度显著,可用于实际生产预测。首次报道了利用蕨渣培养灵芝,为蕨渣进一步的开发研究奠定基础。  相似文献

Ganoderma lucidum was grown in submerged culture in shake flasks on a medium containing peptone, yeast extract and glucose. In pre-cultures, inoculated from an agar-grown culture, morphological and metabolic events were linked: the pellets originally produced protuberances when glucose was present in the medium, although glucose was not consumed. The protuberances were then liberated into the medium as second-generation pellets, at which time glucose consumption began and the rate of exopolysaccharide (EPS) production increased. The synchrony between events was repeated in cultures fed with either glucose or peptone and yeast extract. In main cultures, inoculated from a 16-day-old pre-culture, the biomass concentration increased linearly, while glucose consumption and EPS production were initially slow but then accelerated. Protuberances were produced and liberated similarly to the pre-culture, but there was less synchrony amongst the pellets. When glucose was added to such a culture on day 10, an EPS concentration of 5.7 g L(-1) was achieved on day 13, this being the highest reliable EPS concentration yet reported for submerged culture of G. lucidum. We conclude that a greater understanding of the morphological and physiological events during the culture of G. lucidum will allow the proposal of culture strategies to improve EPS production.  相似文献

以10个不同来源的伞形多孔菌(猪苓)菌株为材料,通过ITS序列分析比较菌株间的亲缘关系,基于菌落、菌丝的形态特征、生长速率、菌丝体干重和产糖能力5个方面的14个指标,采用聚类分析方法分析各指标之间、菌株之间的相关性,主成分分析法提取4个主成分进行分析,通过综合分值比较菌株优劣。ITS序列分析表明,10个菌株间的相似性较高。聚类分析结果显示,指标之间胞外多糖、菌丝团、黑色素、菌丝颜色、长势和分泌物的相关性高;菌丝形态、草酸钙方晶、生长速率和无性孢子的相关性高。5#菌株多糖含量和菌丝生长速率最高;2#和9#菌株胞内多糖含量较高;4#、13#和14#菌株多糖含量较低;10#、11#菌株多糖含量及菌丝体干重也较低。主成分分析显示,5#、2#和1#菌株是综合分值最高的3个菌株,其菌丝生长快、长势旺、产量及多糖含量高。综合上述结果,5#、2#和1#菌株是初步筛选出可用于大规模生产及繁殖的优良菌株。  相似文献

Ulocladium atrum and Gliocladium roseum are fungal antagonists capable of suppressing sporulation of Botrytis spp. on dead plant parts. The effect of temperature (3 to 36 °C) on antagonist conidial germination and mycelial growth was assessed on agar. In addition conidial germination of U. atrum was measured on dead lily leaves. The optimum temperature of both antagonists for both conidial germination and mycelial growth was between 27 and 30 °C. U. atrum was less affected by lower temperatures than G. roseum. At optimum temperature, 50% of conidia of U. atrum and G. roseum germinated within 2.6 and 10.0 hrs, respectively. At low sub-optimal temperatures (6 °C), 50% of conidia germinated within 18 and 96 hours, respectively.In bioassays on dead onion leaves, U. atrum suppressed sporulation of B. cinerea and B. aclada at all temperatures tested (6 to 24 °C) by more than 85%. On dead cyclamen leaves, G. roseum was more efficient than U. atrum at 21 and 24 °C but, in contrast to U. atrum, showed no antagonistic activity at temperatures below 21 °C. On dead hydrangea leaves, U. atrum significantly reduced sporulation of B. cinerea at temperatures as low as 3 and 1 °C. Under Dutch growing conditions, the mean air temperature during leaf wetness periods in onion and lily fields was 15 °C with temperatures only occasionally above 20 °C. In greenhouse crops of cyclamen, the mean temperature during high humidity periods was 17 °C. It is therefore concluded that U. atrum is better adapted than G. roseum to temperatures which occur in the field, in greenhouse crops such as cyclamen, or during cold storage of plant stocks.  相似文献
