The action of thiamine on neuromuscular transmission in the frog sartorius muscle was investigated. It was found that thiamine at a concentration of 1×10–14 to 1×10–4 M increases transmitter secretion at the nerve endings. This is demonstrated by the increased frequency, amplitude, and quantal content of miniature endplate potentials, and is due to the enhanced likelihood of transmitter release. The role of thiamine in regulating synaptic transmission and the mechanism of its interaction with thiamine-sensitive receptors are examined.A. V. We have studied the effect of substance P on the end-plate currents (EPC) and the miniature EPC (MEPC) after acetylcholine esterase (ACE) inhibition in the cut neuromuscular preparation of the frog sartorius muscle using the voltage-clamp technique. At concentrations of 5·10–7–1·10–6 moles/liter substance P had no effect on the amplitude and the time characteristics of single EPC and MEPC but promoted prolongation of EPC decay on repetitive stimulation of the nerve with a frequency of 10/sec, indicating intensification of postsynaptic potentiation. Elevation of the concentration of the given peptide to 5·10–6 moles/liter led to the shortening of the decay of single EPC and a more marked depression of the EPC amplitude in the trains as compared to the control, reflecting a decrease in the sensitivity of the postsynaptic membrane to the mediator, i.e., development of desensitization.S. Thiamine at a concentration of 1×10–14 to 1×10–4 M facilitated neuromuscular transmission at the glutaminergic synapse of the crayfish adapter, manifesting as increased amplitude and quantal content of excitatory postsynaptic potentials and raised frequency of miniature excitatory postsynaptic potentials. Thiamine augmented spontaneous electrical activity and the amplitude of synaptic potentials in the longitudinal muscle of guinea pig taenia coli. It was found from studying the effects of thiamine on the membrane potential of rat brain synaptosomes that its presynaptic action is brought about by depolarization of the nerve terminal membrane. Interaction between thiamine and the nerve endings was described by a Hill coefficient of 0.22–0.30, indicating that it has several binding sites within the structure of the receptor concerned.A. V. The effects of carbachol (CCh) on the frequency (f) of the miniature endplate potentials were tested at temperatures between 5 and 30°C. Higher CCh concentrations, 1 × 10–5 and 5 × 10–6 M, reduced the f to 60% and the temperature dependence was negligible. However, an inverse temperature dependence was found when low concentrations 3 × 10–7 and 6 × 10–7 M were applied. The depression of f was 40–50% in 5–10°C but only 10–20% of the control in the 25 and 30°C. During application of CCh, the new steady of f was reached at temperatures between 5 and 30°C within 17–20 min (Q10 = 1.07). Much greater temperature dependence of recovery was observed during washing out CCh (Q10 = 1.6). The temperature-independence of the steady state effects of CCh, good agreement with Langmuir adsorption-desorption theory and non-steady kinetics indicate that physical rather than receptor-mediated events are responsible for the depression of f.  相似文献

The effects of liquid fluorocarbons as bathing media were determined by use of in vitro neuromuscular preparations. Rat hemidiaphragms were bathed in either oxygenated fluorocarbon (FC) emulsion or standard oxygenated Krebs solution. Contractile force in response to simple supramaximal nerve stimuli as well as to high frequency stimulation was greater, while twitch:tetanus ratio was smaller in FC emulsion. With such medium, post-tetanic potentiation of contraction was also more consistently observed. Indirectly stimulated diaphragms survived longer in FC emulsion. After cessation of oxygenation, oxygen tension (ρO(2)) of the medium declined more rapidly with Krebs than with FC emulsion; ρO(2) directly correlated with force of contraction. Similarly, in the chick biventer cervicis preparation, FC emulsion enhanced nerve-stimulated force of contraction; returning the preparation to standard Krebs solution reversed this phenomenon. Dose-resonse curves of muscle contraction in response to acetycholine and KCl administration were shifted upward during FC emulsion superfusion. Frequency of miniature endplate potentials was lower in FC emulsion than that observed in Krebs solution, measured from the same cell of the rat diaphragm. Resting membrane potentials were also greater in muscle cells sampled from FC emulsion-bathed preparations. These data suggest that FC emulsion is superior to standard Krebs solution as a bathing medium for in vitro neuromuscular preparations by virtue of the high solubility of oxygen in it.  相似文献

Using mathematical modeling of the process of generation of a miniature end-plate current (MEPC), we studied the effect of acetylcholinesterase (AChE) inhibition on the amplitude and frequency parameters of synaptic signals in the neuromuscular junction. The density of acetylcholine receptors on the postsynaptic membrane and the number of acetylcholine molecules in its quantum were varied. AChE inhibition against the background of a decreased receptor density was shown to result in a much higher increase in the amplitude of modeled MEPC than that in control and in the case of the changed transmitter amount released in the synaptic cleft. Endogenous phosphorylation of proteins from rat brain synaptosomal plasma membranes was studied in vitro. Cyclic AMP (cAMP) markedly stimulated32P incorporation in three protein bands with molecular weights of 75,000, 57,000, and 54,000, respectively. The effect of the behaviorally active peptide ACTH1–24 on this endogenous phosphorylation in vitro was studied using peptide concentrations from 10–10 to 10–4 M. In a number of protein bands, a biphasic effect of ACTH1–24 was observed: in concentrations of 10–4–10–5 M, a reduced amount of32P was found; in concentrations of 10–6–10–7 M, hardly any effect could be detected, whereas consistently at concentrations around 10–8 M, a significant decrease was again observed. The phosphoprotein bands affected by in vitro addition of ACTH1–24 were of a smaller molecular weight than those affected by in vitro addition of cAMP.  相似文献

Summary The physiological, pharmacological and morphological characteristics of guinea-pig vas deferens supplied by hypogastric nerves rendered inactive by decentralisation were compared with those of vas deferens in which the nerve supply had been chronically stimulated for 3–9 days using implanted electrodes. No change was seen in decentralised preparations prior to 7 days, but from 8–15 days, increased sensitivity to application of noradrenaline in vitro was observed, which was shown to be related to reduced transmitter uptake by nerve terminals as well as to an increase in postjunctional sensitivity; there was also increased fatigability 7–14 days following decentralisation. Continuous stimulation of hypogastric nerves at 2 Hz for 4–8 h daily for 4–8 days resulted in enhanced transmitter uptake and reduced responses to noradrenaline; this was associated with a slight increase in noradrenaline content and a faster adrenergic neuromuscular response with a shorter latency. No appreciable changes in nerve or muscle structure studied by electron microscopy were observed following decentralisation, but there was an increase of between 12.5 and 29.6% in the number of close (< 100 nm) neuromuscular junctions following chronic stimulation for 8 days.  相似文献

The action of the carbonyl reagent phenylhydrazine hydrochloride (Phen. HCl) on locust excitatory neuromuscular systems was studied by examining the effects of this compound on the mechanical and electrical properties of the retractor unguis and extensor tibiae muscles of the locust Schistocerca gregaria.Low concentrations of Phen. HCl (10?9 w/v to 2·5 × 10?5 w/v) potentiated the muscle contractions and the excitatory post-synaptic potential (EPSP), the optimum concentration being about 10?5 w/v. 10?8 w/v Phen. HCl increased miniature EPSP frequency, but this increase became less pronounced as the concentration was raised, and no increase at all was observed at 10?5 w/v. There was no change in miniature EPSP amplitude at any concentration. Higher concentrations of Phen. HCl (> 2·5 × 10?6 w/v) depressed the neurally evoked contraction, the EPSP, and the response of the muscle to iontophoretically applied l-glutamate. A gradual increase in muscle input conductance was observed on perfusion with these high concentrations of Phen. HCl. The presence of magnesium in the bathing fluid (15 m-moles/l.) reduced the effectiveness of Phen. HCl in potentiating the EPSP and delayed or reduced the increase in input conductance observed on perfusion with high concentrations of Phen. HCl.The results indicate that low concentrations of Phen. HCl act presynaptically, possibly by depolarizing the excitatory nerve terminals. Higher concentrations may act directly on the post-synaptic glutamate receptors.  相似文献

Evoked currents and endplate potentials in frog cutaneopectoral muscle subjected to section or curarization to prevent contraction were investigated in summer and winter conditions using different indirect stimulation rates (of 1 and 100 Hz), with animals maintained at different temperatures prior to trials (of between 5 and 20–25°C). Seasonal changes in amplitude, facilitation, and depression of postsynaptic response discovered in experiments involving curarized and uncurarized dissected muscles differed both quantitatively and qualitatively. These differences may be explained by assuming that sensitivity of the neuromuscular synapse to curare (presumably presynaptic) is liable to seasonal variation.A. A. Amyotrophic Lateral Sclerosis (ALS) is a neurodegenerative disease leading to motor neuron dysfunction resulting in impairment of neuromuscular transmission. A2A adenosine receptors have already been considered as a potential therapeutical target for ALS but their neuromodulatory role at the neuromuscular junction in ALS remains to be clarified. In the present work, we evaluated the effects of A2A receptors on neuromuscular transmission of an animal model of ALS: SOD1(G93A) mice either in the pre-symptomatic (4–6 weeks old) or in the symptomatic (12–14 weeks old) stage. Electrophysiological experiments were performed obtaining intracellular recordings in Mg2+ paralyzed phrenic nerve-hemidiaphragm preparations. Endplate potentials (EPPs), quantal content (q. c.) of EPPs, miniature endplate potentials (MEPPs) and giant miniature endplate potential (GMEPPs) were recorded. In the pre-symptomatic phase of the disease (4–6 weeks old mice), the selective A2A receptor agonist, CGS 21680, significantly enhanced (p<0.05 Unpaired t-test) the mean amplitude and q.c. of EPPs, and the frequency of MEPPs and GMEPPs at SOD1(G93A) neuromuscular junctions, the effect being of higher magnitude (p<0.05, Unpaired t-test) than age-matched control littermates. On the contrary, in symptomatic mice (12–14 weeks old), CGS 21680 was devoid of effect on both the amplitude and q.c. of EPPs and the frequency of MEPPs and GMEPPs (p<0.05 Paired t-test). The results herein reported clearly document that at the neuromuscular junction of SOD1(G93A) mice there is an exacerbation of A2A receptor-mediated excitatory effects at the pre-symptomatic phase, whereas in the symptomatic phase A2A receptor activation is absent. The effects of lanthanum(La) on contractions induced by prostaglandin F(PGF) or isotonic K+ were investigated in the isolated stomach muscle of guinea-pig.Low concentrations of La(0.1–1 μM) inhibited the contraction to PGF 1 μM in a dose-dependent manner, without affecting the tonic contraction to isotonic K+.0.1 and 1 μM La shifted the dose-response curve for PGF(0.001 – 1 μM) to the right and reduced the maximum response.The IC50 of La against PGF and K+ were 0.6 μM and 30 μM, respectively.These results support the suggestion that PGF -induced contraction in the stomach muscle depends mainly on the intracellular release of sequestered Ca, which would be depleted or immobilized by La.  相似文献

The contractions of the dorsal longitudinal muscle of the mollusc Lymnaea stagnalis L., which are evoked by electric stimulation of n. cervicalis inferior were studied. It has been shown that an increase of magnesium ion concentration in saline to 10–15 mM decreases reversibly amplitude of the evoked contractions. Application of serotonin produced a dual effect: at concentrations of 2 × 10–5–10–6 M, it enhanced muscle contractions, whereas at concentrations above 10-5 M, on the contrary, decreased them. The inhibitory effect of the serotonin antagonist mianserin on the evoked contraction amplitude increased with elevation of its concentrations in the studied range (from 10–5 to 10–3 M). The enhancing effect of serotonin on muscle contractions was blocked either by previous mianserin application or its application on the background of the already acting serotonin. A participation of serotoninergic mechanisms in the control of the contractile function of the studied muscle is suggested.  相似文献

The possible effects of the alkaloids vinblastine and colchicine on the postsynaptic membrane of the frog neuromuscular junction were investigated using voltage-clamp techniques. Concentrations of vinblastine and colchicine which had been shown to exert no effect on the amplitude and duration of miniature endplate currents (MEPC) and the current-voltage relationship of low-quantal endplate currents (EPC) together with the coefficient of voltage-dependent EPC decay did produce a considerable rise in the amplitude of response to iontophoretically applied acetylcholine (ACh). In addition, vinblastine and colchicine accelerate MEPC and EPC during acetylcholine esterase inhibition while further depressing the amplitude of multi-quantal EPC succeeding at the rate of 10 Hz as well as response to regular (5–10 Hz) application of ACh from a micropipet. The dosage-frequency effects of vinblastine and colchicine on the postsynaptic membrane (as described) are presumed to be unconnected with the action of these agents on muscle fiber cytoskeleton but the results of accelerated desensitization of cholinoreceptors.S. The effects were studied of ethimizol, a substance activating memory processes, on features of synaptic transmission during experiments on frog cutaneous pectoris muscle. It was found that the presynaptic action of ethimizol consists of raising the frequency of miniature potentials, when used at a concentration of 0.5–10 mM, and modulating quantal content of synaptic transmission due to changes in binomial quantal release parameters p and n when 0.5–2 mM ethimizol was used. This substance facilitated transmission at synapses with a low initial level of transmitter release. This substance facilitated transmission at synapses with a low initial level of transmitter release. Ethimizol was also found to have a postsynaptic action, consisting of reducing amplitude at a concentration of 5–10 mM and prolonging synaptic currents and potentials when concentrations of 0.5–10 mM were used. The latter effect produced a considerable increase in the time integral of endplate potentials. The postsynaptic action of ethimizol is perhaps seen in its effects on features of postsynaptic ionic channels. Summary This paper reviews the evidence for impacts of metals on the growth of selected plants and on the effects of metals on soil microbial activity and soil fertility in the long-term. Less is known about adverse long-term effects of metals on soil microorganisms than on crop yields and metal uptake. This is not surprising, since the effects of metals added to soils in sewage sludge are difficult to assess, and few long-term experiments exist. Controlled field experiments with sewage sludges exist in the UK, Sweden, Germany and the USA and the data presented here are from these long-term field experiments only. Microbial activity and populations of cyanobacteria,Rhizobium leguminosarum bv.trifolii, mycorrhizae and the total microbial biomass have been adversely affected by metal concentrations which, in some cases, are below the European Community's maximum allowable concentration limits for metals in sludge-treated soils. For example, N2-fixation by free living heterotrophic bacteria was found to be inhibited at soil metal concentrations of (mg kg–1): 127 Zn, 37 Cu, 21 Ni, 3.4 Cd, 52 Cr and 71 Pb. N2-fixation by free-living cyanobacteria was reduced by 50% at metal concentrations of (mg kg–1): 114 Zn, 33 Cu, 17 Ni, 2.9 Cd, 80 Cr and 40 Pb.Rhizobium leguminosarum bv.trifolii numbers decreased by several orders of magnitude at soil metal concentrations of (mg kg–1): 130–200 Zn, 27–48 Cu, 11–15 Ni, and 0.8–1.0 Cd. Soil texture and pH were found to influence the concentrations at which toxicity occurred to both microorganisms and plants. Higher pH, and increased contents of clay and organic carbon reduced metal toxicity considerably. The evidence suggests that adverse effects on soil microbial parameters were generally found at surpringly modest concentrations of metals in soils. It is concluded that prevention of adverse effects on soil microbial processes and ultimately soil fertility, should be a factor which influences soil protection legislation.  相似文献

The effects of 0.1% testicular hyaluronidase on miniature endplate potentials and currents (MEPP and MEPC) were investigated in frog pectorocutaneous muscle. The action of hyaluronidase on preparations with armine-induced blockade of acetylcholinesterase was associated with decreased amplitude and duration of MEPP and MEPC half-decay time and rising phase. The correlation between amplitude and half-decay time of MEPP and MEPC declined at the same time, while MEPC decay remained exponential. Treating preparations having intact acetylcholinesterase with hyaluronidase increased the length of MEPC halfdecay, with duration of the rising phase and amplitude remaining constant. It is suggested that enzymatic breakdown of a proportion of the glycocalix of cells forming the neuromuscular junction and a portion of the extracellular matrix at the synaptic cleft leads to attenuation of nonspectific acetylcholine binding, thus facilitating acetylcholine diffusion into the synaptic cleft.A. The effect of exposure to different concentrations of food and suspended silt on filtration, respiration and condition were studied in the freshwater mussel Hyridella menziesi. Using a milk solids-based food and kaolin to simulate silt, mussels were maintained at different combinations of food and silt concentrations for 3 weeks. Between treatments mean filtration rates ranged from 0.97–1.66 l g–1 h–1, and respiration from 0.50–1.35 mg O2 g–1 h–1. Silt (non-volatile suspended solids up to 35 mg l–1) failed to have a significant effect on filtration rate or condition, but with increasing food levels (volatile suspended solids up to 35 mg l–1) filtration rate was reduced, and condition was reduced at the lowest food concentration (<5 mg l–1). Respiration showed a food × silt interaction between treatment blocks. When food was low respiration increased with increasing silt concentrations, and when silt was low (<5 mg l–1) respiration increased with increasing food concentrations. The observed effects of food and silt on filtration, respiration and condition are discussed in terms of their potential for affecting contaminant bioaccumulation. In low-food situations (i.e., <5 mg l–1), if mussels are pumping large volumes of water, contaminant uptake rates could be enhanced, whereas abundant food would result in lower pumping rates and lower uptake rates. Changes in metabolism with food concentration have implications for contaminant elimination, and changes in biochemical composition associated with changing condition could affect the tissue distribution and retention of contaminants.  相似文献

Effects of acetylcholine, octopamine, and their antagonists, as well as of glutamic acid were studied on contractions of dorsal longitudinal muscle of the mollusc Lymnaea stagnalis L., evoked by electrical stimulation of n. cervicalis inferior. Acetylcholine and octopamine increased amplitude of contractions evoked by applications at concentrations about 10–8 M and decreased at concentrations higher than 10–7 M. Glutamic acid produced only inhibitory effect on the contraction amplitude, which appeared at concentrations beginning from 10–9 M and higher. The acetylcholine antagonists atropine and d-tubocurarine also decreased amplitude of evoked contractions. Their blocking effect rose with increase of their concentrations in the range from 10–9 M to 10–5 M. Specificity of the effect of the studied substances was established in experiments with a combined application of the transmitters and their antagonists. Effects of pyrocatechol on neuromuscular transmission were studied both in the frog pectoral-cutaneous muscle and in the mouse phrenic-diaphragmatic preparation by means of extracellular microelectrode recording of synaptic signals. Pyrocatechol applied in a concentration of 0.05 mM increased the frequency of miniature end-plate currents (MEPC) and the amplitude of end-plate current (EPC) by increasing its quantum content. Pyrocatechol also increased the duration of presynaptic response. When voltage-dependent potassium channels had been blocked, pyrocatechol affected neither the EPC quantum content nor the duration of presynaptic response.
