The applicability of “inter-simple sequence repeats” (ISSR)-PCR as a molecular character complex for differentiation of Equisetum taxa is described with a special focus on the detection of hybrids. All Central European Equisetum species and the four most frequent hybrids are considered: E. arvense, E. fluviatile, E. telmateia, E. palustre, E. sylvaticum, E. pratense, E. × litorale, E. ramosissimum, E. hyemale, E. variegatum, E. × moorei, E. × trachyodon and E. × meridionale. The ISSR banding patterns are highly typical for each species. The positions of the clusters of hybrid taxa prove their hybrid origin and enable identification of the parental species. Genetic similarities of populations suggest that in some species vegetative reproduction predominates (e.g. E. arvense), whereas for others (e.g. E. telmateia) sexual reproduction seems to be more frequent. In addition to the molecular analyses, the characteristics of stem cross sections of the four hybrids and their parental species are shown and discussed.  相似文献

1. Selection does not only operate in a genotype (G) × environment (E) context, but can also be modulated by the activities of organisms interacting with their environment (G × G × E). 2. The influences of aphid clonal identity and host plant (Vicia faba) intraspecific genetic variation on the performance of five genotypes of pea aphid (Acyrthosiphon pisum) were investigated – with and without interaction with a competing heterospecific clone of vetch aphid (Megoura viciae) – across three cultivars of V. faba. 3. Pea aphid performance in the presence of a competing vetch aphid clone (G × G × E) compared with the absence of competition (G × E) revealed strong context‐dependent, genotype‐specific shifts in performance, influenced by plant cultivar, competitor presence and their interaction. 4. The performance of vetch aphid in competition with each pea aphid clone was also compared. Here, competitor's genotype and abundance underlay a remarkably varied response by vetch aphid across interactions. 5. The study shows that aphid genotypes exhibit a varying degree of risk spreading, contingent on competitor identity and the patterns of aggregation across three plant cultivars. Owing to feedback loops between species activities and selective forces acting on them, our findings suggest that there are context‐dependent responses by competitors that are shaped via the interplay of the co‐occurring species and their biotic environment. 6. This work highlights the complexity of species interactions and the importance of investigating reciprocity between competition and intraspecific genetic variation. A better understanding of the eco‐evolutionary interactions between phloem‐feeding insects and their host plants can potentially be used to enhance crop protection and pest control.  相似文献

A Potamogeton hybrid recently discovered in Finland was proven to be a new taxon, which has a different parentage than the most similar P.×vepsicus (=P. natans×praelongus) described from Russia. Based on molecular and morphological investigation, the new hybrid was identified as P. alpinus×natans and is here described as P.×exilis nothosp. nov. The hybrid is known only from several nearby localities in a single river system. All discovered colonies almost certainly originate from a single hybridization event that occurred in the upper part of the river system. Other localities downstream have been established through fragmentation and dispersal of stems and rhizomes of this sterile hybrid. A detailed morphological comparison of the new hybrid with similar hybrids and species is provided.  相似文献

Because the name Potamogeton ×babingtonii has been synonymised with P. ×angustifolius, no valid binominal exists for the hybrid P. lucens×praelongus. A morphological and anatomical study presented here demonstrates that specimens regarded by Hagström as Potamogeton ×babingtonii f. danicus really represent the hybrid between P. lucens and P. praelongus. As a consequence, a new binominal for this hybrid is introduced and its basionym Potamogeton ×babingtonii f. danicus is lectotypified. Morphological and anatomical characters of Potamogeton ×jutlandicus nom. nov. (=P. lucens×praelongus) are provided and its taxonomic delimitation is discussed.  相似文献

Miscanthus is a rhizomatous C4 grass of great interest as a biofuel crop because it has the potential to produce high yields over a wide geographical area with low agricultural inputs on marginal land less suitable for food production. At the moment, a clonal interspecific hybrid Miscanthus × giganteus is the most widely cultivated and studied in Europe and the United States, but breeding programmes are developing newer more productive varieties. Here, we quantified the physiological processes relating to whole season yield in a replicated plot trial in Wales, UK. Light capture and conversion efficiency were parameterized for four carefully selected genotypes (M. sinensis, M. sacchariflorus and Miscanthus × giganteus). Differences in the canopy architecture in mature stands as measured by the extinction coefficient (k) were small (0.55–0.65). Sensitivity analysis on a mathematical model of Miscanthus was performed to quantify the accumulative intercepted photosynthetically active radiation (iPAR) in the growing season using (i) k, (ii) variation in the thermal responses of leaf expansion rate, (iii) base temperature for degree days and (iv) date start of canopy expansion. A 10% increase in k or leaf area per degree day both had a minimal effect on iPAR (3%). Decreasing base temperature from 10 to 9 °C gave an 8% increase in iPAR. If the starting date for canopy expansion was the same as shoot emergence date, then the iPAR increases by 12.5%. In M. × giganteus, the whole season above ground and total (including below ground) radiation‐use efficiency (RUE) ranged from 45% to 37% higher than the noninterspecific hybrid genotypes. The greater yields in the interspecific hybrid M. × giganteus are explained by the higher RUE and not by differences in iPAR or partitioning effects. Studying the mechanisms underlying this complex trait could have wide benefits for both fuel and food production.  相似文献

The aim of this study was to further test the viability of the roach Rutilus rutilus × common bream Abramis brama hybrid in terms of reproductive behaviour and sexual production. Egg release, mating and aggressive acts in reproductive behaviour, as well as absolute fecundity and sperm density in milt for sexual production were examined in the first generation of these hybrids at their first sexual maturity. The F2 and backcrosses of hybrids were also studied. The results revealed that these hybrids expressed a normal and typical mating behaviour, producing viable gametes. Under experimental reproduction between hybrids (hybrid reproduction), the number of egg‐release acts (range, 21–66) was nearer (χ2 test, P > 0.05) the number of mating acts (11–65). Moreover, hybrid males exhibited territorial and aggressive behaviours. However, in experimental reproduction of female and male hybrids mixed with parental males (mixed reproduction), the egg‐release act and the mating act were inhibited by the intense territorial and aggressive activities of the common bream male. Absolute fecundity values (median, <2.2 × 103 eggs) and sperm density (<7 × 109 spermatozoa ml?1) of hybrids showed a greater decrease (U test, P < 0.05) than in parental species (median, >6.0 × 103 eggs and >14 × 109 spermatozoa ml?1, respectively). Nevertheless, these hybrids were fertile. F2 and backcross generations were produced, although with a significantly lower viable hatching rate (FEP test, P < 0.05) in F2 individuals from the female and its corresponding hybrid male (<6%), indicating a very low chance of survival in rivers.  相似文献

The corticotropin‐releasing hormone type I receptor (CRHR1) gene has been implicated in the liability for neuropsychiatric disorders, particularly under conditions of stress. On the basis of the hypothesized effects of CRHR1 variation on stress reactivity, measures of adulthood traumatic stress exposure were analyzed for their interaction with CRHR1 haplotypes and single‐nucleotide polymorphisms (SNPs) in predicting the risk for alcoholism. Phenotypic data on 2533 non‐related Caucasian individuals (1167 alcoholics and 1366 controls) were culled from the publically available Study of Addiction: Genetics and Environment genome‐wide association study. Genotypes were available for 19 tag SNPs. Logistic regression models examined the interaction between CRHR1 haplotypes/SNPs and adulthood traumatic stress exposure in predicting alcoholism risk. Two haplotype blocks spanned CRHR1. Haplotype analyses identified one haplotype in the proximal block 1 (P = 0.029) and two haplotypes in the distal block 2 (P = 0.026, 0.042) that showed nominally significant (corrected P < 0.025) genotype × traumatic stress interactive effects on the likelihood of developing alcoholism. The block 1 haplotype effect was driven by SNPs rs110402 (P = 0.019) and rs242924 (P = 0.019). In block 2, rs17689966 (P = 0.018) showed significant and rs173365 (P = 0.026) showed nominally significant, gene × environment (G × E) effects on alcoholism status. This study extends the literature on the interplay between CRHR1 variation and alcoholism, in the context of exposure to traumatic stress. These findings are consistent with the hypothesized role of the extra hypothalamic corticotropin‐releasing factor system dysregulation in the initiation and maintenance of alcoholism. Molecular and experimental studies are needed to more fully understand the mechanisms of risk and protection conferred by genetic variation at the identified loci .  相似文献

Stipa ×brozhiana M. Nobis nothosp. nov. (Poaceae) is described and illustrated. The taxon belongs to sect. Smirnovia Tzvel. and originated from the hybridization of species belonging to sect. Smirnovia and Barbatae Junge. Stipa ×brozhiana is morphologically close to Stipa lipskyi Roshev., but is easily distinguished by its much shorter hairs on the seta, thinner, uni‐ or indistinctly bigeniculate awns, differently shaped callus and longer ligules of vegetative shoots. Characters distinguishing S. ×brozhiana from the parental species and other similar hybrid taxa belonging to sect. Smirnovia and occurring in the Pamir Alai Mts are discussed. In addition, the taxonomical status of S. ×tzvelevii Ikonn. pro sp., another taxon that has originated from hybridization between species belonging to sect. Smirnovia and Barbatae, is discussed. The taxon was originally described as a distinct species, but is now considered to be the hybrid S. caucasica×S. orientalis. The main characters distinguishing S. ×tzvelevii from parental species are given.  相似文献

Fecal prevalence of Escherichia coli O157 in ruminants is highest in the summer decreasing to very low levels in the winter. We hypothesize that this seasonal variation is a result of physiological responses within the host animal to changing day-length. To determine the effects of melatonin (MEL) on fecal shedding of E. coli O157:H7 in cattle, eight crossbred beef steers identified as shedding E. coli O157:H7, were allotted to treatment: control or MEL (0.5 mg/kg body weight (BW); 1×) administered orally daily for 7 days. After a 5-day period of no treatment, a second MEL dose (5.0 mg/kg BW; 10×) was administered daily for 4 days. Fecal samples were collected daily for qualification of E. coli O157:H7. No differences (P > 0.10) were observed in the percentage of E. coli O157:H7 positive fecal samples in steers receiving the 1× MEL dose, however the 10× dose decreased (P = 0.05) the percentage of fecal samples E. coli O157:H7 positive. Serum MEL concentrations were higher in the 1×, but not 10×, treated animals compared to control animals. Although it is difficult to explain, this may be a result of decreasing day-length increasing serum melatonin concentrations that may have masked any treatment effect on serum melatonin. In a second similar experiment, a second group of cattle (heifers and steers) were administered tryptophan (TRP) over a 17-day experimental period (5 g/head/day for 10 days followed by 10 g/head/day for 7 days). Tryptophan had no effect (P > 0.20) on the percentage of fecal samples positive for E. coli O157. Serum TRP (P < 0.05), but not MEL (P > 0.20), concentrations were elevated in TRP-treated animals. The decrease in the number of positive fecal samples observed in the first experiment, may be related to gastrointestinal MEL, affected by the 10×, but not 1× MEL dose.  相似文献

Species of Lasiodiplodia are important pathogens of a wide variety of plants covering a wide geographical distribution. These fungi can be associated with different symptoms such as stem cankers, shoot blights, fruit rots, dieback and gummosis. Diseases caused by Lasiodiplodia were surveyed on Eucalyptus urophylla × grandis, Polyscias balfouriana and Bougainvillea spectabilis in a nursery in southern China. Based on morphology characteristics and phylogenetic analyses of ITS rDNA sequences and translation elongation factor 1‐alpha (TEF‐1α) gene regions, four species of Lasiodiplodia were identified. Lasiodiplodia theobromae was identified from E. urophylla × grandis, P. balfouriana and B. spectabilis. L. hormozganensis, L. iraniensis and L. pseudotheobromae were identified from B. spectabilis. To our knowledge, with the exception of L. theobromae on E. urophylla × grandis, this study represents the first report of these fungi on the host plants. Pathogenicity tests showed that all Lasiodiplodia spp. obtained in this study are virulent to E. urophylla × grandis and B. spectabilis, and L. theobromae was virulent to P. balfouriana.  相似文献

Rosa spinosissima is an endangered species in Norway, found only within a limited area on the southwestern coast. Presumed hybrid forms between R. spinosissima and rose species within R. sect. Caninae have been recorded from this area since the late 19th century. Here, analyses of such hybrid plants and selected populations of the tentative parent species were performed using AFLP markers, nuclear DNA content, pollen viability and seed germination rates, in addition to classic morphometric analysis. It is established that the hybrid rose represents a single hybrid taxon, viz Rosa × sabinii, formed by recurrent asymmetrical hybridization events between R. spinosissima and R. mollis, with the latter being the obligate ovule donor. The evidence presented does not indicate hybridization with other co‐occurring rose species, or of introgression from R. mollis into R. spinosissima through backcrossing with R. ×sabinii.  相似文献

Elymus cylindricus (2= 6= 42) and E. breviaristatus (2= 6= 42) are distributed in grasslands and deserts of northern and north‐western China. Their genomic constitution and taxonomic status are unclear. Elymus cylindricus was crossed with E. wawawaiensis J.R.Carlson & Barkworth ( StH ), Roegneria grandis Keng ( StY ) and Campeiostachys dahurica (Turcz. ex Griseb.) B.R.Baum, J.L. Y ang & C. Y en var. dahurica ( StYH ). Meiotic pairing in the hybrids E. cylindricus × E. wawawaiensis ( StH ), E. cylindricus × R. grandis ( StY ) and E. cylindricus × C. dahurica var. dahurica ( StYH ) showed on average 10.00, 11.30 and 20.92 bivalents per cell, respectively. Elymus breviaristatus was crossed with C. dahurica var. dahurica ( StYH ) and E. cylindricus. Chromosome pairing in the hybrids of E. breviaristatus × C. dahurica var. dahurica and E. breviaristatus × E. cylindricus showed on average 19.60 and 19.27 bivalents, respectively. Genomic in situ hybridization (GI SH ) revealed the presence of St , Y and H genomes in E. cylindricus and E. breviaristatus. An intergenomic rearrangement was observed in E. cylindricus using GI SH . Meiotic pairing data and GI SH indicated that both E. cylindricus and E. breviaristatus are allohexaploids containing the StYH genomes. Elymus cylindricus and E. breviaristatus should be treated as Campeiostachys dahurica var. cylindrica and Campeiostachys breviaristata, respectively.  相似文献

The perennial grass, Miscanthus×giganteus is a sterile triploid, which due to its growth rate and biomass accumulation has significant economic potential as a new bioenergy crop. The sterility associated with the triploid genome of this accession requires labor‐intensive vegetative, instead of seed propagation for potential commercial production. Chromosome doubling was used to produce hexaploid plants in an effort to restore fertility to M×giganteus. Tissue culture derived calli from immature inflorescences were treated with the antimitotic agents, colchicine and oryzalin in liquid and solid media. Calli survival rate decreased with increasing concentrations and durations of colchicine or oryzalin treatments and ranged from 0% to 100%. Nuclear DNA content, as determined by flow cytometry, indicated that the frequency of chromosome‐doubled calli varied between compounds and concentrations with the greatest proportion of callus doubling observed using 2‐day treatments of 15 μm oryzalin (78%) or 939 μm colchicine (67%). Liquid media treatments were more effective than solid gels for chromosome doubling. Although oryzalin was effective at chromosome doubling, it inhibited callus growth and plant regeneration frequency. Seven hexaploid plants with doubled DNA content were generated, which displayed increased stomata size (30.0±0.2 μm) compared with regenerated triploid M. ×giganteus plants (24.3±1.0 μm). Following clonal replication these plants will be evaluated for growth rate, biomass accumulation, and pollen viability. Successful chromosome doubling and plant regeneration of M.×giganteus suggests that ploidy manipulation of this plant and its parental species (Miscanthus sinensis and Miscanthus sacchariflorus) could be a means to access genetic variability for the improvement of Miscanthus as a biofuel/bioenergy crop.  相似文献

Aims: The survival capability of pathogens like Escherichia coli O157:H7 in manure‐amended soil is considered to be an important factor for the likelihood of crop contamination. The aim of this study was to reveal the effects of the diversity and composition of soil bacterial community structure on the survival time (ttd) and stability (irregularity, defined as the intensity of irregular dynamic changes in a population over time) of an introduced E. coli O157:H7 gfp‐strain were investigated for 36 different soils by means of bacterial PCR‐DGGE fingerprints. Methods and Results: Bacterial PCR‐DGGE fingerprints made with DNA extracts from the different soils using bacterial 16S‐rRNA‐gene‐based primers were grouped by cluster analysis into two clusters consisting of six and 29 soils and one single soil at a cross‐correlation level of 16% among samples per cluster. Average irregularity values for E. coli O157:H7 survival in the same soils differed significantly between clusters (P = 0·05), whereas no significant difference was found for the corresponding average ttd values (P = 0·20). The irregularity was higher for cluster 1, which consisted primarily of soils that had received liquid manure and artificial fertilizer and had a significant higher bacterial diversity and evenness values (P < 0·001). Conclusions: Bacterial PCR‐DGGE fingerprints of 36 manure‐amended soils revealed two clusters which differed significantly in the stability (irregularity) of E. coli O157 decline. The cluster with the higher irregularity was characterized by higher bacterial diversity and evenness. Significance and Impact of the Study: The consequence of a high temporal irregularity is a lower accuracy of predictions of population behaviour, which results in higher levels of uncertainty associated with the estimates of model parameters when modelling the behaviour of E. coli O157:H7 in the framework of risk assessments. Soil community structure parameters like species diversity and evenness can be indicative for the reliability of predictive models describing the fate of pathogens in (agricultural) soil ecosystems.  相似文献

This work presents the biocidal (insecticidal, ixodicidal, nematicidal, and phytotoxic) effects and chemical compositions of three essential oils obtained from the industrial steam distillation (IEOs) of hyssop (Hyssopus officinalis L.), lavandin (Lavandula × intermedia or L. × hybrida var. Super ), and cotton lavender (Santolina chamaecyparissus L.). Their chemical composition analyzed by gas chromatography coupled to mass spectrometry showed 1,8‐cineole (53%) and β‐pinene (16%) as the major components of H. officinalis, linalyl acetate (38%) and linalool (29%) of L. × intermedia; and 1,8‐cineole (10%) and 8‐methylene‐3‐oxatricyclo[5.2.0.02,4]nonane (8%) in S. chamaecyparissus. The biocidal tests showed that L. × intermedia IEO was the most active against the insect Spodoptera littoralis and toxic to the tick Hyalomma lusitanicum, IEO of H. officinalis was strongly active against S. littoralis, and finally, S. chamaecyparissus IEO was a strong antifeedant against the aphid Rhopalosiphum padi, toxic to H. lusitanicum and with moderate effects against Leptinotarsa decemlineata, S. littoralis, and Lolium perenne.  相似文献

Interspecific hybridization is a primary cause of extensive morphological and chromosomal variation and plays an important role in plant species diversification. However, the role of interploidal hybridization in the formation of hybrid swarms is less clear. Epidendrum encompasses wide variation in chromosome number and lacks strong premating barriers, making the genus a good model for clarifying the role of chromosomes in postzygotic barriers in interploidal hybrids. In this sense, hybrids from the interploidal sympatric zone between E. fulgens (2n = 2x = 24) and E. puniceoluteum (2n = 4x = 56) were analyzed using cytogenetic techniques to elucidate the formation and establishment of interploidal hybrids. Hybrids were not a uniform group: two chromosome numbers were observed, with the variation being a consequence of severe hybrid meiotic abnormalities and backcrossing with E. puniceoluteum. The hybrids were triploids (2n = 3x = 38 and 40) and despite the occurrence of enormous meiotic problems associated with triploidy, the hybrids were able to backcross, producing successful hybrid individuals with broad ecological distributions. In spite of the nonpolyploidization of the hybrid, its formation is a long‐term evolutionary process rather than a product of a recent disturbance, and considering other sympatric zones in Epidendrum, these events could be recurrent.  相似文献

The perennial rhizomatous grass, Miscanthus×giganteus is an ideal biomass crop due to its rapid vegetative growth and high biomass yield potential. As a naturally occurring sterile hybrid, M. ×giganteus must be propagated vegetatively by mechanically divided rhizomes or from micropropagated plantlets. Plant regeneration through somatic embryogenesis is a viable approach to achieve large‐scale production of plantlets in tissue culture. Effect of the callus types, ages and culture methods on the regeneration competence was studied to improve regeneration efficiency and shorten the period of tissue culture in M. ×giganteus. Shoot‐forming calli having a yellow or white compact callus with light‐green shoot‐like structures showed the highest regeneration frequency. Percentage of shoot‐forming callus induction from immature inflorescence explants was 41% on callus induction medium containing 13.6 μM 2,4‐d and 0.44 μM benzyladenine (BA). The use of a regeneration medium containing 1.3 μM NAA and 22 μM BA was effective at shortening the incubation period required for plantlet regeneration, with 69% of total regenerated plantlets obtained within 1 month of incubation on regeneration medium. Embryogenic‐like callus morphotype could maintain regeneration competency for up to 1 year as suspension cultures. Field grown regenerated plants showed normal phenotypic development with DNA content and plant heights comparable to rhizome propagated plants. Winter survival rates of the regenerated plants planted in 2006 and 2007 at the University of Illinois South Farm, Urbana‐Champaign, Illinois, were 78% and 56%, respectively.  相似文献

Miscanthus × giganteus is a C4 perennial grass that shows great potential as a high‐yielding biomass crop. Scant research has been published that reports M. × giganteus growth and biomass yields in different environments in the United States. This study investigated the establishment success, plant growth, and dry biomass yield of M. × giganteus during its first three seasons at four locations (Urbana, IL; Lexington, KY; Mead, NE; Adelphia, NJ) in the United States. Three nitrogen rates (0, 60, and 120 kg ha?1) were applied at each location each year. Good survival of M. × giganteus during its first winter was observed at KY, NE, and NJ (79–100%), and poor survival at IL (25%), due to late planting and cold winter temperatures. Site soil conditions, and growing‐season precipitation and temperature had the greatest impact on dry biomass yield between season 2 (2009) and season 3 (2010). Ideal 2010 weather conditions at NE resulted in significant yield increases (< 0.0001) of 15.6–27.4 Mg ha?1 from 2009 to 2010. Small yield increases in KY of 17.1 Mg ha?1 in 2009 to 19.0 Mg ha?1 in 2010 could be attributed to excessive spring rain and hot dry conditions late in the growing season. Average M. ×giganteus biomass yields in NJ decreased from 16.9 to 9.7 Mg ha?1 between 2009 and 2010 and were related to hot dry weather, and poor soil conditions. Season 3 yields were positively correlated with end‐of‐season plant height () and tiller density (). Nitrogen fertilization had no significant effect on plant height, tiller density, or dry biomass yield at any of the sites during 2009 or 2010.  相似文献

Terpenes are important compounds in plant trophic interactions. A meta‐analysis of GC‐MS data from a diverse range of apple (Malus × domestica) genotypes revealed that apple fruit produces a range of terpene volatiles, with the predominant terpene being the acyclic branched sesquiterpene (E,E)‐α‐farnesene. Four quantitative trait loci (QTLs) for α‐farnesene production in ripe fruit were identified in a segregating ‘Royal Gala’ (RG) × ‘Granny Smith’ (GS) population with one major QTL on linkage group 10 co‐locating with the MdAFS1 (α‐farnesene synthase‐1) gene. Three of the four QTLs were derived from the GS parent, which was consistent with GC‐MS analysis of headspace and solvent‐extracted terpenes showing that cold‐treated GS apples produced higher levels of (E,E)‐α‐farnesene than RG. Transgenic RG fruit downregulated for MdAFS1 expression produced significantly lower levels of (E,E)‐α‐farnesene. To evaluate the role of (E,E)‐α‐farnesene in fungal pathogenesis, MdAFS1 RNA interference transgenic fruit and RG controls were inoculated with three important apple post‐harvest pathogens [Colletotrichum acutatum, Penicillium expansum and Neofabraea alba (synonym Phlyctema vagabunda)]. From results obtained over four seasons, we demonstrate that reduced (E,E)‐α‐farnesene is associated with decreased disease initiation rates of all three pathogens. In each case, the infection rate was significantly reduced 7 days post‐inoculation, although the size of successful lesions was comparable with infections on control fruit. These results indicate that (E,E)‐α‐farnesene production is likely to be an important factor involved in fungal pathogenesis in apple fruit.  相似文献
