Multiple paternity was found in two clutches of Podocnemis expansa using eight microsatellite loci. When loci were analysed separately a minimum of two males was estimated for nest N23, and three for nest C17. When all loci were combined, three patrilines were detected in N23, and six in C17. The distribution of full-sib cluster sizes indicated a disproportionate contribution of one male to clutch C17, consistent with possible sperm competition, or the mixing of leftover and newly acquired sperm. High mutation rates were detected at several loci. Multiple paternity has positive implications for this endangered species as it may slow the loss of genetic variability caused by drift. This is the first report of multiple paternity in the suborder Pleurodira.  相似文献

Giant Amazon river turtles, Podocnemis expansa, are indigenous to the Amazon, Orinoco, and Essequibo River basins, and are distributed across nearly the entire width of the South American continent. Although once common, their large size, high fecundity, and gregarious nesting, made P. expansa especially vulnerable to over-harvesting for eggs and meat. Populations have been severely reduced or extirpated in many areas throughout its range, and the species is now regulated under Appendix II of the Convention on International Trade in Endangered Species. Here, we analyse data from mitochondrial DNA sequence and multiple nuclear microsatellite markers with an array of complementary analytical methods. Results show that concordance from multiple data sets and analyses can provide a strong signal of population genetic structure that can be used to guide management. The general lack of phylogeographic structure but large differences in allele and haplotype frequencies among river basins is consistent with fragmented populations and female natal-river homing. Overall, the DNA data show that P. expansa populations lack a long history of genetic differentiation, but that each major tributary currently forms a semi-isolated reproductive population and should be managed accordingly.  相似文献

Like coloration of the integument, eye color can be a significant but understudied component of communication and reproductive behavior. Eye color can change with sexual maturation and become sexually dimorphic, but in a few birds and fish, eye color can also change rapidly in response to the environment. There are few cases of the latter, and we report here several instances of such change in eye color in the Eastern box turtle (Terrapene carolina carolina), the first non-avian tetrapod in which this capability has been reported. In male turtles, the iris changed from a pale yellow color (often characteristic of juveniles) to a bright red color (characteristic of mature males) in a period of <5 s. The nature of the color change is similar to that observed in some birds and suggests a common mechanism and/or adaptive role, which could be further explored in Eastern box turtles.  相似文献

In recent years many cases of hybridization and introgression became known for chelonians, requiring a better understanding of their speciation mechanisms. Phylogeographic investigations offer basic data for this challenge. We use the sister species Mauremys caspica and M. rivulata, the most abundant terrapins in the Near and Middle East and South-east Europe, as model. Their phylogeographies provide evidence that speciation of chelonians fits the allopatric speciation model, with both species being in the parapatric phase of speciation, and that intrinsic isolation mechanisms are developed during speciation. Hybridization between M. caspica and M. rivulata is very rare, suggesting that the increasing numbers of hybrids in other species are caused by human impact on environment (breakdown of ecological isolation). Genetic differentiation within M. caspica and M. rivulata resembles the paradigm of southern genetic richness and northern purity of European biota. However, in west Asia this pattern is likely to reflect dispersal and vicariance events older than the Holocene. For M. caspica three distinct Pleistocene refuges are postulated (Central Anatolia, south coast of Caspian Sea, Gulf of Persia). Morphologically defined subspecies within M. caspica are not supported by genetic data. This is one of the few studies available about the phylogeography of west and central Asian species.  相似文献

The development and cornification of the ramphoteca (beak) in turtles are not known. The microscopic aspects of beak formation have been analyzed in the pleurodirian turtle Emydura macquarii using histological, immunocytochemical and ultrastructural methods. At embryonic Stage 15 the maxillar beak is originated from discontinuous placodes (one frontal and two oral) formed in the epidermis above and below the mouth that later merge into the epidermis of the beak. The mandibular beak is formed by two lateral placodes. In the placodes, basal keratinocytes in contact with local mesenchymal condensations become columnar, and generate suprabasal cells forming 5–6 layers of embryonic epidermis at Stages 17–20 and a compact shedding alpha‐layer at the base of the embryonic epidermis. These keratinocytes contain irregular or aggregated reticular bodies made of 30–40 nm thick strands of coarse filaments, mixed with tonofilaments and sparse lipid droplets. Beneath the shedding layer are present 3–4 layers of keratinocytes accumulating coarse filaments mixed with beta‐corneous packets, and underneath spindle‐shaped beta‐cells differentiate where beta‐corneous packets completely replace the reticulate bodies. Differently from scales where corneocytes partially merge, beak corneocytes remain separated but they are joined by numerous interlocking spines. The production of beta‐cells in the thick corneous layer of the developing beak, like in claws, occurs before the differentiation of beta‐cells in the body scutes. This indicates that a massive mesenchymal condensation triggers beta‐differentiation before this process is later activated in most of body scutes of the carapace and plastron. J. Exceptional variability in the shell of the pancake tortoise Malacochersus tornieri , both in the keratinous surficial scutes and the underlying bones, in addition to its remarkably fenestrated bony shell are unique among tortoises. Based on 14 individuals of different sizes and ages, the observed variation in M. tornieri was described in detail, with additional notes on the typically testudinid skull, inner ear and brain endocast using microCT‐scan data, as well as the limbs. Similar degrees of variation have not yet been described in any other extant turtle species and therefore seem notable in M. tornieri , and might be related to the species' unique lifestyle. Within the carapace, the peripherals and suprapygals are most variable in number. Furthermore, different combinations of peripherals are participating in the central plastral fontanelle and in some individuals additional bones take part in the formation of the plastron. J. Mating‐induced ovulation is common in mammals, but has been rarely described in other taxa. Observations of several mature female loggerhead sea turtles, Caretta caretta, held in captivity seemed to indicate that ovulation did not occur in the absence of a male. This study was designed to determine whether this was an effect of captivity or an effect of the absence of a male. Two mature female loggerheads were followed over six annual reproductive cycles. Ultrasound exams were performed approximately every 2 weeks to follow the development of follicles in the ovaries. During the first two seasons, no male was present, in the next two seasons, a mature male was present without mating, and in the final two seasons a mature male was present, mating with one or both females. When no male was present or when present without mating, ovarian follicles developed to full size, but ovulation did not occur and the follicles gradually began to decrease in diameter and undergo changes evident on ultrasound. In the fifth season, only one of the females mated, dropping two eggs after 7 days, and continued to oviposit throughout the following months (total 275 eggs). The unmated female did not ovulate, showing the same pattern as earlier seasons. In the final season, both females mated and ovulated, dropping eggs for the next four months (total 539 eggs). The following year, no males were present and neither female ovulated. This study provides clear evidence that ovulation in loggerhead sea turtles is induced by mating. Chelonians are expected to be negatively impacted by climate change due to limited vagility and temperature‐dependent sex determination. However, few studies have examined how freshwater turtle distributions may shift under different climate change scenarios. We used a maximum entropy approach to model the distribution of five widespread North American Kinosternon species (K. baurii, K. flavescens, K. hirtipes, K. sonoriense, and K. subrubrum) under four climate change scenarios. We found that areas with suitable climatic conditions for K. baurii and K. hirtipes are expected to decline substantially during the 21st century. In contrast, the area with suitable climate for K. sonoriense will remain essentially unchanged, while areas suitable for K. flavescens and K. subrubrum are expected to substantially increase. The centroid for the distribution of four of the five species shifted northward, while the centroid for K. sonoriense shifted slightly southward. Overall, centroids shifted at a median rate of 37.5 km per decade across all scenarios. Given the limited dispersal ability of turtles, it appears unlikely that range shifts will occur rapidly enough to keep pace with climate change during the 21st century. The ability of chelonians to modify behavioral and physiological responses in response to unfavorable conditions may allow turtles to persist for a time in areas that have become increasingly unsuitable, but this plasticity will likely only delay local extinctions.  相似文献

The Cofán Indians of Aguarico and Zábalo Rivers in Ecuador in 1990 began a turtle recuperation effort by raising turtle hatchlings (Podocnemis expansa and P. unifilis) in small pools. To evaluate the effectiveness of this program, the Cofán developed and tested three methods to document changes in the populations of turtles. The first method consisted of interviews with people from neighbouring communities about their perceptions of changes in turtle abundance. The second method was a direct count of turtles observed along the rivers from canoe. The third method was an indirect count, involving turtle nest censusing along community beaches. Two of the methods yielded numerical, statistically positive turtle population trends. The turtle program has influenced the community's perception of turtle conservation issues so that, instead of hunting the adults and eggs, they now protect them. The training in record-keeping permits them to make a transparent history of the equitability of access to the turtle egg resource. The Cofán monitoring experience strengthened their successful petition to the Ecuadorian government for the rights to manage their territorial lands within the Cuyabeno Wildlife Reserve, and to receive protected area status and management rights to other ancestral lands. The experience gained by the Zábalo monitors is now serving as a model for the design and implementation of the park monitoring system of the newly-established Cofán Park Ranger Corps, which will be patrolling three Ecuadorian Ecological Reserves which overlap Cofán inhabited areas.  相似文献

Evolutionary morphologists and physiologists have long recognized the phylogenetic significance of the ectothermic sauropsids. Sauropids have been classically considered to bridge between early tetrapods, ectotherms, and the evolution of endotherms. This transition has been associated with many modifications in cardiovascular form and function, which have changed dramatically during the course of vertebrate evolution. Most cardiovascular studies have focused upon adults, leaving the development of this critical system largely unexplored. In this essay, we attempt a synthesis of sauropsid cardiovascular development based on the limited literature and indicate fertile regions for future studies. Early morphological cardiovascular development, i.e., the basic formation of the tube heart and the major pulmonary and systemic vessels, is similar across tetrapods. Subsequent cardiac chamber development, however, varies considerably between developing chelonians, squamates, crocodilians, and birds, reflected in the diversity of adult ventricular structure across these taxa. The details of how these differences in morphology develop, including the molecular regulation of cardiac and vascular growth and differentiation, are still poorly understood. In terms of the functional maturation of the cardiovascular system, reflected in physiological mechanisms for regulating heart rate and cardiac output, recent work has illustrated that changes during ontogeny in parameters such as heart rate and arterial blood pressure are somewhat species‐dependent. However, there are commonalities, such as a β‐adrenergic receptor tone on the embryonic heart appearing prior to 60% of development. Differential gross morphological responses to environmental stressors (oxygen, hydration, temperature) have been investigated interspecifically, revealing that cardiac development is relatively plastic, especially, with respect to change in heart growth. Collectively, the data assembled here reflects the current limited morphological and physiological understanding of cardiovascular development in sauropsids and identifies key areas for future studies of this diverse vertebrate lineage. Despite recognition of the western drywood termite, Incisitermes minor (Hagen), as one of the most economically important and destructive termite species in the USA, both its population and colony breeding structure genetically remain unclear. Here, we present primer sequences and initial characterization for 15 polymorphic microsatellite loci. In a sample of 30 individuals, representing six geographically distinct locations collected in California, USA, three to 15 alleles were detected segregating per locus. Within a single population observed heterozygosity ranged from 0.050 to 0.866.  相似文献

Juvenile growth rate is a critical demographic parameter, as it shortens the time to maturity and often dictates how long individuals remain vulnerable to predation. However, developing a mechanistic understanding of the factors determining growth rates can be difficult for wild populations. The gopher tortoise (Gopherus polyphemus) is an ecosystem engineer threatened by habitat loss and deficient management of pinelands in the southeastern United States. We investigated the factors governing immature gopher tortoise growth and explored the use of drone-based imagery for habitat assessment by comparing predictive models based on ground-based plant surveys versus drone-derived data. From 2021 to 2022, we tracked and measured immature tortoises in native sandhill and human-modified, ruderal habitat in south-central Florida. Using quarterly, high-resolution drone imagery, we quantified plant cover types and vegetation indices at each occupied burrow and measured the frequency of occurrence of forage species by hand. Annual growth rates of immature tortoises in ruderal habitat were higher than those in sandhill and were the highest published for this species. Models based on drone-derived data were able to explain similar proportions of variation in growth as ground-collected measures of forage, especially during the late dry season when both types of models were most predictive. Habitat differences in forage nitrogen content were also more pronounced during this season, when dominant ground cover in ruderal habitat (bahiagrass) had much higher nitrogen content than dominant ground cover in sandhill (wiregrass). Despite concerns about potential growth-survival trade-offs, tortoises in ruderal habitat did not exhibit lower apparent survival. Our findings indicate that habitat dominated by nutritious non-native grass can provide a valuable supplement to native sandhill through the mechanism of increased growth rates due to higher forage quality. Finally, our study demonstrates that drone technology may facilitate management by providing less labor-intensive ways to assess habitat quality for this and other imperiled herbivores.  相似文献

通过对桉树属(Eucalyptus)的10000条EST序列进行分析,在其中的1499条序列上共发现1775个微卫星重复序列。含有微卫星的EST序列约占序列总数的15%。此外,还发现桉树EST序列所含微卫星长度的变异速率与重复单元长度呈负相关;微卫星的丰度与重复单元长度也呈负相关(三碱基重复微卫星除外)。在桉树EST序列中,重复单元长度为三碱基的微卫星最为丰富。三碱基重复单元微卫星的过度富集可能是由于遗传密码选择所致。在微卫星的丰度及长度变异方面,桉树EST序列与杨树(Populus trichocarpa)基因组注释的转录序列随重复单元长度的变化呈现出相同的规律,但桉树EST序列中微卫星频率及三碱基重复微卫星的含量显著偏低,推测含微卫星的基因表达丰度极有可能低于不含微卫星的基因。通过对发现的所有微卫星位点进行引物设计,并对设计的引物进行PCR检测,结果表明所设计的引物具有极高的扩增成功率。  相似文献

Fourteen microsatellite primer pairs were developed from a cDNA library of heat‐treated seedlings of Fragaria vesca cv. yellow wonder. Transferability to 13 species of Fragaria ranged from 71% in diploid species F. gracilis Losinsk., F. iinumae Makino, F. nilgerrensis Schltdl. ex J. Gay and F. nipponica Makino, to 100% in octoploid domestic strawberry and its progenitors. Polymorphism was high in polyploid Fragaria species. However, polymorphism and heterozygosity of eight EST‐SSRs (expressed sequence tag–simple sequence repeats) was low in 14 F. vesca genotypes.  相似文献

We cloned seven microsatellite loci from house wrens (Troglodytes aedon) using a biotin enrichment protocol. Starting with fragments generated using DOP–PCR, fragments containing microsatellite motifs AC and AAC were captured using biotinylated probes and streptavidin coated magnetic particles. Captured fragments were cloned into plasmids; prior to sequencing, the plasmids were screened for microsatellites using a simple PCR approach. Five of the loci showed variation in a sample of nine individuals.  相似文献

We have isolated and developed 180 new polymorphic chicken microsatellite markers. In addition, primers have been developed for 91 microsatellites derived from the GenBank sequence database (isolated by the laboratory of Terry Burke, Leicester University), of which 89 were polymorphic, and six existing polymorphic markers (HUJ) have been modified. The primer sequences were designed to allow optimal performance of the markers, in sets containing multiple microsatellites, on ABI sequencers. The average number of alleles for the 275 polymorphic markers described was 4·0. Of these markers, 93% were polymorphic in the Wageningen resource population whereas 57% of the markers were polymorphic in the East Lansing reference population and only 44% could be mapped in the Compton reference population. The microsatellite markers described in this paper, in combination with the microsatellite markers published previously, are particularly well suited for performing a total genome scan for the detection of quantitative trait loci (QTL).  相似文献
