介绍了一种适用于丝状真菌染色体DNA大片段的快速提取方法,该方法以(100mM Tris,100mM NaGl,50mM EDTA-Na2 2%SDS,pH值9.0)为提取液,经石英砂研磨破壁.应用该方法成功地提取了粗糙脉胞菌(Neurospora crassa)、米曲霉(Aspergillus oryzae)、产黄青霉(Penicillium chrysogenum)和头孢霉菌(Cep- halosporium sp.)等4种不同丝状真菌的染色体DNA大片段,且所提DNA片段均大于20kb,可直接用于限制性酶切、PCR等分子生物学研究.  相似文献

Improved methods are described for the isolation of pure, high molecular weight DNA from small and large scale cultures of filamentous fungi. The methods depend on the extraction of DNA under conditions which prevent nuclease activity and contamination by carbohydrate. The small scale method depends on enzymatic digestion of the wall whereas the large scale method uses partial damage followed by autolysis. High yields of DNA are obtained by both methods and the DNA is suitable for restriction analysis. Southern Blotting, RFLP analysis, dot blotting and the production of gene libraries. The small scale method can be used for the simultaneous analysis of multiple cultures.  相似文献

A simple and rapid method for the preparation of a cell-free extract with the CCAAT-binding activity was established with Aspergillus nidulans as a model fungus. Proteins were extracted with 6 M guanidine hydrochloride directly from mycelia and renatured by dialysis. This method was found applicable to other filamentous fungi such as Aspergillus oryzae and Trichoderma viride.  相似文献

The biodegradation of phenanthrene and benzo[a]pyrene was assayed in liquid and wet sand cultures in the presence of five filamentous fungi. In the controls, 85% volatilisation of phenanthrene occurred within 28 days in liquid cultures while it was only 50% in wet sand. In the later system, remaining phenanthrene and benzo[a]pyrene amounted to 6–51 and 53–92% of their initial levels, respectively, according to the strains. Then, wet sand used as a screening tool evidenced Trametes versicolor and Cunninghamella elegans as the most efficient polycyclic aromatic hydrocarbons degraders among ten strains. © Rapid Science Ltd. 1998  相似文献

Lipases from psychrotrophic fungal isolates BPF4 and BPF6 identified as Penicilium canesense and Pseudogymnoascus roseus respectively were characterized for their compatibility towards laundry detergent. BPF4 and BPF6 lipases showed maximum activity at pH 11 and 9 respectively and at 40?°C. The residual activities at 20?°C and 4?°C of BPF4 lipase were 35% and 20% and of BPF6 lipase were 70% and 20?°C respectively. Both the enzymes were stable at 4?°C, 20?°C and 40?°C for 2?h losing at the most 20% of activities. Both the enzymes were metalloenzymes with activity enhancement by nearly threefold by Ca2+. Contrary to BPF6 lipase, BPF4 enzyme was not stimulated by EDTA nor inhibited, rather stimulated by SDS and Triton X-100 by 125% and 330% respectively. Both the lipases showed minor to moderate inhibition by NaClO3 and H2O2, and exhibited nearly 90% residual activity after 1?h of incubation in selected detergent brands thus indicating potential for their inclusion in detergent formulation thereby facilitating cold-washing as a step towards mitigation of climate change.  相似文献

从四个不同银耳段木栽培场地采集杂菌样本,经初步鉴定后纯化得到48个菌株。通过对子实体、孢子及菌丝等的形态观察确认从银耳栽培段木上分离的48株杂菌隶属于炭团菌属,在形态上与截头炭团菌Annulohypoxylon annulatum最为相似。为了进一步确认从银耳栽培段木上分离的48株杂菌的分类地位,对其中14个供试菌株进行了ITS系统发育分析,结果表明:14个供试菌株之间的遗传距离为0.036时可以形成两个明显分枝,且分枝与地理来源之间并无明显相关性;供试菌株在分类地位上属于炭团菌属,与形态学鉴定结果一致,并与截头炭团菌的关系最为接近,遗传距离在0.2左右;与银耳伴生菌香灰菌的遗传距离为0.29左右。从75条引物中筛选出13条引物对所有48个供试菌株进行ISSR扩增,扩增共获得103条带,其中多态性条带72条,占条带总数的69.9%。聚类分析结果表明:在相似水平为75%时,48个菌株可以划分为5个类群,且类群的划分与地理来源之间并无明显相关性;多数供试菌株之间的遗传相似性较低,这表明供试菌株在DNA水平上存在比较显著的遗传变异,具有较丰富的遗传多样性。  相似文献

Entomopathogenic fungi have great potential to control agricultural and horticultural insect pests, however optimizing conidial production systems to demonstrate high productivity and stability still needs additional efforts for successful field application and industrialization. Although many virulent entomopathogenic fungal isolates have been viewed as potential candidates in a laboratory environment, very few of the isolates are being used in practice for application in agricultural fields as commercial products. I. javanicus is an entomopathogenic fungus that is parasitic to various diverse coleopteran and lepidopteran insects and thought good candidate as biopesticdes. In this work, the basic characteristics of two entomopathogenic fungi, I. javanica FG340 and Pf04, were investigated in morphological examinations, genetic identification, and virulence against Thrips palmi, and then the feasibility of various grains substrates for conidial production was assessed, particularly focusing on conidial productivity and thermotolerance. Isaria javanica FG340 and Pf04 conidia were solid-cultured on 12 grains for 14?days in a Petri dish. Of the tested Italian millet, perilla seed, millet and barley-based cultures showed high conidial production. The four-grain media yielded >1?×?109 conidia/g of I. javanica FG340 and Pf04. Pf04 strain had enhanced thermotolerance up to 45?°C when cultured on Italian millet. In application, it was easy to make a conidial suspension using the cultured grains, and several surfactants were tested to release the conidia. This work suggests several possible inexpensive grain substrates by which to promote conidial production combined with enhanced stability against exposure to high temperature.  相似文献

The ganglioside II3NeuAc-GgOse4Cer and other glycosphingolipids can be radiolabeled to high specific activity by the galactose oxidase-NaB3H4 procedure, by purifying the oxidized compounds prior to reductive labeling. The oxidized products are separated from nonoxidized compounds and detergents (Triton X-100 and sodium taurocholate) present during the enzymatic oxidation. Since the oxidized derivatives are separated, the final specific activity depends solely upon the specific activity of the NaB3H4 and the reduction conditions.  相似文献

A simple and easy protocol for extracting high-quality DNA from different yeast and filamentous fungal species is described. This method involves two important steps: first, the disruption of cell walls by mechanical means and freezing; and second, the extraction, isolation, and precipitation of genomic DNA. The absorbance ratios (A260/A280) obtained ranged from 1.6 to 2.0. The main objective of this procedure is to extract pure DNA from yeast and filamentous fungi, including those with high contents of proteins, polysaccharides, and other complex compounds in their cell walls. The yield and quality of the DNAs obtained were suitable for micro/minisatellite primer-polymerase chain reaction (MSP-PCR) fingerprinting as well as for the sequence of the D1/D2 domain of the 26S rDNA.  相似文献

Arabinoxylan (`Cellace') corn fiber, containing 28.1% (w/w) as l-arabinose and 32.8% (w/w) as d-xylose, was hydrolyzed by a crude enzyme containing -xylanase, -xylosidase and -l-arabinofuranosidase originating from the extracellular culture broth of Penicillium funiculosum. The resultant hydrolysate contained l-arabinose, d-xylose and small amounts of other mono- and oligosaccharides. The l-arabinose and d-xylose were 21.3% (w/w) and 18.7% (w/w), respectively, based on the initial arabinoxylan. Williopsis saturnus var. saturnus, which metabolizes d-xylose without using l-arabinose, was aerobically cultured in the hydrolysate at 30 °C for 96 h. The sugar solution after removal of yeast cells contained l-arabinose and d-xylose which were 20.3% (w/w) and 0.002% (w/w), respectively, of the initial arabinoxylan. The solution was decolorized with activated carbon, and deionized with cation- and anion-exchange resins. The clear sugar solution thus obtained was composed of l-arabinose and d-xylose which were 19.3% (w/w) and 0.001% (w/w), respectively, of the initial arabinoxylan. After concentration of the sugar solution under reduced pressure, followed by crystallization of l-arabinose, 16% (w/w) l-arabinose (based on the initial arabinoxylan) was obtained as crude crystals. No d-xylose was detected in the final preparation.  相似文献

Summary We have implemented a routine procedure for screening protein sequences for evidence of intragenic duplications. We tested 163 protein sequences representing 116 superfamilies of unrelated proteins. Twenty superfamilies contain proteins with internal gene duplications. The intragenic duplications detected can be divided into two major types. (1) One or more duplications of all or part of a gene produce a protein with two or several detectable regions of sequence homology. Sequences from 18 superfamilies contained this type of duplication. (2) Repeated reduplication of a small DNA segment can produce a protein that is repetitive over most of its length. Three superfamilies contain such repetitive sequences. We also investigated the limits of detection of ancient duplications using sequences derived by random mutation of a model sequence consisting of ten 10-residue repeats. The original repetitive nature of the sequence was usually detected after 250 point mutations even though the ancestral segment could not be accurately reconstructed.  相似文献

Intention, Goal and Scope: Dealing with data gaps, data asymmetries, and inconsistencies in life cycle inventories (LCI) is a general prohlem in Life Cycle Assessment (LCA) studies. An approach to deal with these difficulties is the simplification of LCA. A methodology that lowers the requirements for data quality (accuracy) for process emissions within a simplified LCA is introduced in this article. Background: Simplification is essential for applying LCA in the context of design for environment (DfE). The tool euroMat is a comprehensive DfE software tool that is based on a specific, simplified LCA approach, the Iterative Screening LCA (IS-LCA). Within the scope of the IS-LCA, there is a quantitative assessment of energy-related processes, as well as a semi-quantitative assessment of non-energy related emissions which supplement each other. Objectives: The semi-quantitative assessment, which is in the focus of this article, aims at lowering the requirements for the quality of non-energy related emissions data through combined use of qualitative and quantitative inventory data. Methods: Potential environmental impacts are assessed based on ABC-categories for qualities (harmfulness) of emissions and XYZ-categories for quantities of emitted substances. Employing statistical methods assignment rules for the ABC/XYZ-categories were derived from literature data and databases on emissions to air, water, and soil. Statistical tests as well as a DfE case study (comparing the materials aluminum and carbon fiber reinforced epoxy for a lightweight container to be used in an aerospace application) were conducted in order to evaluate the level of confidence and practicality of the proposed, simplified impact assessment. Results: Statistical and technical consistency checks show that the method bears a high level of confidence. Results obtained by the simplified assessment correlate to those of a detailed quantitative LCA. Conclusions: Therefore, the application of the ABC/XYZ-categories (together with the cumulative energy demand) can be considered a practical and consistent approach for determining the environmental significance of products when only incomplete emission data is available. Future Prospects: The statistical base of the method is expanded continuously since it is an integral part of the DfE software tool euroMat, which is currently being further developed. That should foster the application of the method. Outside DfE, the method should also be capable of facilitating simplified LCAs in general.  相似文献

A simple, sensitive precipitin-type single-cell secretion assay is described and applied to the study of immunoglobulin-secreting cells. Evidence is presented that its efficiency is comparable to that achieved with reverse hemolytic plaque techniques. Also described is a modification of the simplified procedure which possesses substantially increased sensitivity. Enhanced sensitivity is achieved through the use of monoclonal rheumatoid factors which preferentially react with rabbit or human immunoglobulins that are incorporated into immune complexes as a result of interaction with antigen. Addition of rheumatoid factor to the agarose-cell mixture leads to additional crosslinking of immune complexes that form around active cells, thereby increasing the probability of forming a detectable precipitate. The application of this procedure to the detection of cells producing T-cell products is also discussed.  相似文献
