选用常用的两种报告基因、两种抗性基因、两种启动子序列和两种终止子序列为探针,将其PCR扩增产物用MicroGrid Ⅱ型全自动点样仪按矩阵排列点样于包埋有氨基的载玻片上,制备成转基因作物检测型基因芯片。利用该芯片对4种转基因水稻、木瓜、大豆、玉米进行检测,结果表明,该芯片能对转基因作物做出快速、准确的检测。 Abstract:Some selected available sequences of reporter genes,resistant genes,promoters and terminators are amplified by PCR for the probes of transgenic crop detection gene chip.These probes are arrayed at definite density and printed on the surface of amino-slides by bioRobot MicroGrid Ⅱ.Results showed that gene chip worked quickly and correctly,when transgenic rice,pawpaw,maize and soybean were applied.  相似文献

目前,生物安全性问题已成为转基因技术及其产品能否获得足够的研究空间和良好开发市场的关键所在.然而,在重视安全性的同时,也必须认识到转基因技术及其产品的出现乃是人类社会发展的客观必然.当务之急不应该是简单地反对或者赞成,而是必须对转基因产品进行全面、充分的研究和客观、具体的评价,从而使其在造福于人类的同时,能有效地减少或避免对人类健康及其生存环境可能产生的潜在危害.当然,对社会公众进行科普宣传和指导,从而使其在对转基因产品具有客观了解的基础上培养一种健康的接受心态也是必要的.本文在简述转基因产品安全性及其研究现状的基础上,提出若干对应策略,希望能引起社会各界的共识。 Abstract:Currently,biosafety has become a key to transgenic technique and its products,which gain enough research space and better exploiture market or not.Then,as well as we pay attention to the security,we should realize that transgenic technique and its products are essential result in human society.Today,they would not simply oppose or praise GMOs,but we should carry out thorough studies and estimate without influence by human feelings,so that the technique and the products are used for the benefit of mankind,not doing harm for human health and the environment.Of course,we should provide some essential scientific knowledge and guidance for public people.In the paper,we will put forward some suggestions on the base of analysing transgenic products.  相似文献

转基因抗虫作物的商业化种植带来了显著的经济、生态和社会效益,对转基因抗虫作物的安全性评价也一直是国内外学者研究的热点。对生物非靶标效应的研究是转基因抗虫作物安全性评价的重要组成部分,农田天敌类生物是其中的重点内容之一。瓢虫是农田生态系统重要的捕食性天敌,评价其在转基因抗虫作物田间是否能通过捕食猎物或取食花粉而接触到杀虫蛋白并富集于体内,进而对自身产生一定的非靶标效应,这对捕食性瓢虫的安全性研究具有重要的意义。本文从捕食性天敌瓢虫的生命表参数、行为功能参数、田间群落参数及体内微环境指标等方面综述了转基因抗虫作物对瓢虫的安全性研究进展,并对后期转基因抗虫作物对田间捕食性天敌的研究方向提出了建议,以期为转基因抗虫作物的环境安全性研究提供理论指导和为进一步完善转基因抗虫作物对瓢虫安全性评价的技术体系提供系统的数据资料。  相似文献

植物凝集素是一类具有特异糖结合活性的蛋白,具有一个或多个可以与糖或寡聚糖特异可逆结合的非催化结构域。其糖结合活性是针对外源寡糖,参与植物的防御反应。本文综述了有关植物凝集素分子生物学的研究进展,介绍了植物凝集素的分类、糖结合特性、近年来有关植物凝集素蛋白晶体结构的研究,及其与糖结合能力相关的生物学功能。并对植物凝集素在抗虫植物基因工程中的应用现状及发展前景做了阐述。 Abstract:Plant lectins are proteins possessing at least one non-catalytic domain that binds reversibly to specific mono-or oligosaccharides.They distinguish themselves from other plant proteins by the ability of carbohydrate binding.Most plant lectins are directed to bind foreign polysacchride.Plant lectin is believed to take part in the defense responses against invader.In this paper we presented a review on the classification,characters,functions,crystal structure and,functions related to the carbohydrate binding activity.The status and prospect of plant lectins utilization were also discussed.  相似文献

采用共培养和脂质体介导两种方法将金黄地鼠附睾内的精子与NFκB-luc+质粒共孵育后,再与成熟的卵母细胞进行体外受精,用PCR、Southern blot及荧光原位杂交(FISH)等技术检测外源基因在精子细胞和在受精卵中的存在及存在部位,并对相关数据进行统计学比较分析。结果表明,金黄地鼠附睾内的活精子具有主动捕获外源基因的能力,而且外源基因只存在于精子细胞的头部,而且通过受精将外源基因带入了卵细胞,脂质体的存在增加了外源基因转染精子的效率和转基因效率。Abstract: Incubate golden hamster spermatozoa and NFκB-luc+ in co-culture and liposome mediated methods, and then process the IVF with oocytes, And two groups of samples were detected by PCR, Southern Blot and FISH. Analyzed the relative data by statistics. The results showed that spermatozoa of Golden hamster epididymis can take the foreign DNA voluntarily, and the foreign DNA exist in the head of the spermatozoa can enter into zygotes by IVF. The exist of liposome can improve the foreign gene transfect ratio and transgenic ratio.  相似文献

The aim of resistance management is to slow and ideally reverse the development of resistance in the pest population. Since 1996, million of acres of crops have been planted that are genetically engineered with Bacillus thuringiensis (Bt) for insect resistance. The novelty for resistance management is that with Bt technology it is possible to control the principal force in an agroecosystem microevolutionary process from the outset, i.e., selection pressure. In Bt crops, the toxin can be expressed constitutively at a relatively constant dose or expression of the toxin can be restricted to specific crop stages, tissues, or both. Here we propose that more precise control of selection eases the practical application of resistance management strategies (high-dose–refugia) compared to other resistance strategies. The population genetics and ecological and operational factors related to the high-dose–refugia strategy currently used for Bt crops are also reviewed.  相似文献

Transgenic resistance to insects has been demonstrated in plants expressing insecticidal genes such as δ -endotoxins from Bacillus thuringiensis (Bt), protease inhibitors, enzymes, secondary plant metabolites, and plant lectins. While transgenic plants with introduced Bt genes have been deployed in several crops on a global scale, the alternative genes have received considerably less attention. The protease inhibitor and lectin genes largely affect insect growth and development and, in most instances, do not result in insect mortality. The effective concentrations of these proteins are much greater than the Bt toxin proteins. Therefore, the potential of some of the alternative genes can only be realized by deploying them in combination with conventional host plant resistance and Bt genes. Genes conferring resistance to insects can also be deployed as multilines or synthetic varieties. Initial indications from deployment of transgenics with insect resistance in diverse cropping systems in USA, Canada, Argentina, China, India, Australia, and South Africa suggest that single transgene products in standard cultivar backgrounds are not a recipe for sustainable pest management. Instead, a much more complex approach may be needed, one which may involve deployment of a combination of different transgenes in different backgrounds. Under diverse climatic conditions and cropping systems of tropics, the success in the utilization of transgenics for pest management may involve decentralized national breeding programs and several small-scale seed companies. While several transgenic crops with insecticidal genes have been introduced in the temperate regions, very little has been done to use this technology for improving crop productivity in the harsh environments of the tropics, where the need for increasing food production is most urgent. There is a need to develop appropriate strategies for deployment of transgenics for pest management, keeping in view the pest spectrum involved, and the effects on nontarget organisms in the ecosystem.  相似文献
