Heading time in bread wheat ( Triticum aestivum L.) is determined by three characters – vernalization requirement, photoperiodic sensitivity and narrow-sense earliness (earliness per se) – which are involved in the phase transition from vegetative to reproductive growth. The wheat APETALA1 ( AP1 )-like MADS-box gene, wheat AP1 ( WAP1 , identical with VRN1 ), has been identified as an integrator of vernalization and photoperiod flowering promotion pathways. A MADS-box gene, SUPPRESSOR OF OVEREXPRESSION OF CO 1 ( SOC1 ) is an integrator of flowering pathways in Arabidopsis . In this study, we isolated a wheat ortholog of SOC1 , wheat SOC1 ( WSOC1 ), and investigated its relationship to WAP1 in the flowering pathway. WSOC1 is expressed in young spikes but preferentially expressed in leaves. Expression starts before the phase transition and is maintained during the reproductive growth phase. Overexpression of WSOC1 in transgenic Arabidopsis plants caused early flowering under short-day conditions, suggesting that WSOC1 functions as a flowering activator in Arabidopsis . WSOC1 expression is affected neither by vernalization nor photoperiod, whereas it is induced by gibberellin at the seedling stage. Furthermore, WSOC1 is expressed in transgenic wheat plants in which WAP1 expression is cosuppressed. These findings indicate that WSOC1 acts in a pathway different from the WAP1 -related vernalization and photoperiod pathways.  相似文献

We review functional data on MADS-box genes, recent phylogenetic analyses of these coding regions, and their roles in the development and evolution of key morphological innovations in plants. We map the origin of important morphological structures in particular diverse stages of the life cycle in different plant clades onto organismal phylogenies, and present relevant molecular genetic aspects of development related to the MADS-box genes. We focus on reproductive structures of the sporophyte because most functional characterizations have been done of MADS-box genes involved in flower development. We discuss MADS-box evolution in flowering plants, but we also review studies in the other nonflowering vascular plants, gymnosperms (conifers and gnetales), and ferns and preliminary data from the algae. We suggest that floral (e.g. flowering time, inflorescence, and flower meristem identity) MADS-box and nonfloral plant MADS-box genes should be the focus of future comparative research. Cloning and functional analyses of MADS-box genes in bryophytes, particularly in the experimental system Physcomitrella patens (Hedw.) B.S.G., are needed. The ABC model of floral organ specification is an excellent general representation of an important network of genes; however, formal analytical tools are required to integrate data on complex gene interaction in comparative analyses. This and other analytical approaches to constructing gene network models will help to frame homology hypotheses in an evolutionary and developmental framework.  相似文献

During the past decade, MADS-box genes have become known as key regulators in both reproductive and vegetative plant development. Traditional genetics and functional genomics tools are now available to elucidate the expression and function of this complex gene family on a much larger scale. Moreover, comparative analysis of the MADS-box genes in diverse flowering and non-flowering plants, boosted by bioinformatics, contributes to our understanding of how this important gene family has expanded during the evolution of land plants. Therefore, the recent advances in comparative and functional genomics should enable researchers to identify the full range of MADS-box gene functions, which should help us significantly in developing a better understanding of plant development and evolution.  相似文献

A MADS domain gene involved in the transition to flowering in Arabidopsis   总被引:20,自引:0,他引:20  
Flowering time in many plants is triggered by environmental factors that lead to uniform flowering in plant populations, ensuring higher reproductive success. So far, several genes have been identified that are involved in flowering time control. AGL20 (AGAMOUS LIKE 20) is a MADS domain gene from Arabidopsis that is activated in shoot apical meristems during the transition to flowering. By transposon tagging we have identified late flowering agl20 mutants, showing that AGL20 is involved in flowering time control. In previously described late flowering mutants of the long-day and constitutive pathways of floral induction the expression of AGL20 is down-regulated, demonstrating that AGL20 acts downstream to the mutated genes. Moreover, we can show that AGL20 is also regulated by the gibberellin (GA) pathway, indicating that AGL20 integrates signals of different pathways of floral induction and might be a central component for the induction of flowering. In addition, the constitutive expression of AGL20 in Arabidopsis is sufficient for photoperiod independent flowering and the over-expression of the orthologous gene from mustard, MADSA, in the classical short-day tobacco Maryland Mammoth bypasses the strict photoperiodic control of flowering.  相似文献

A late-flowering mutant was isolated from rice T-DNA-tagging lines. T-DNA had been integrated into the K-box region of Oryza sativa MADS50 (OsMADS50), which shares 50.6% amino acid identity with the Arabidopsis MADS-box gene SUPPRESSOR OF OVEREXPRESSION OF CO 1/AGAMOUS-LIKE 20 (SOC1/AGL20). While overexpression of OsMADS50 caused extremely early flowering at the callus stage, OsMADS50 RNAi plants exhibited phenotypes of late flowering and an increase in the number of elongated internodes. This confirmed that the phenotypes observed in the knockout (KO) plants are because of the mutation in OsMADS50. RT-PCR analyses of the OsMADS50 KO and ubiquitin (ubi):OsMADS50 plants showed that OsMADS50 is an upstream regulator of OsMADS1, OsMADS14, OsMADS15, OsMADS18, and Hd (Heading date)3a, but works either parallel with or downstream of Hd1 and O. sativa GIGANTEA (OsGI). These results suggest that OsMADS50 is an important flowering activator that controls various floral regulators in rice.  相似文献

Evolutionary developmental genetics (evodevotics) is a novel scientific endeavor which assumes that changes in developmental control genes are a major aspect of evolutionary changes in morphology. Understanding the phylogeny of developmental control genes may thus help us to understand the evolution of plant and animal form. The principles of evodevotics are exemplified by outlining the role of MADS-box genes in the evolution of plant reproductive structures. In extant eudicotyledonous flowering plants, MADS-box genes act as homeotic selector genes determining floral organ identity and as floral meristem identity genes. By reviewing current knowledge about MADS-box genes in ferns, gymnosperms and different types of angiosperms, we demonstrate that the phylogeny of MADS-box genes was strongly correlated with the origin and evolution of plant reproductive structures such as ovules and flowers. It seems likely, therefore, that changes in MADS-box gene structure, expression and function have been a major cause for innovations in reproductive development during land plant evolution, such as seed, flower and fruit formation.  相似文献

Many plants in temperate regions have a requirement for vernalization in order to initiate the reproductive growth phase. In cereals, this requirement has been linked to the VRN1 locus, which encodes an APETALA1 -like ( AP1 -like) MADS-box gene. In perennial ryegrass ( Lolium perenne L.), we have isolated two MADS-box genes that are regulated by vernalization, LpMADS1 , which co-localize to the VRN1 locus in ryegrass, and LpMADS10 , which is an SVP -like MADS-box gene. In the shoot apex, LpMADS1 is increasingly induced by cold exposure, whereas LpMADS10 is increasingly repressed. Comparison of LpMADS1 promoter regions from several ryegrass varieties, with and without vernalization requirement, suggests that a putative MADS-box protein-binding site (CArG-box) might be important for the vernalization-regulated expression of LpMADS1 . Although the LpMADS10 expression pattern suggests it to be involved in floral repression, ectopic expression of LpMADS10 did neither affect flowering time significantly in Arabidopsis thaliana nor in L. perenne . Interestingly, we found that LpMADS1 interacts with LpMADS10 in a yeast two-hybrid assay. This finding is discussed in regard to the regulation of vernalization response in perennial ryegrass.  相似文献
