The immunological function of the thymus was first documented 50 years ago by using neonatally thymectomized mice, while studying its role in virus-induced leukaemia. Since then, an enormous wealth of reports has helped to define the importance of this primary lymphoid organ. In this article, I summarize the key advances that have led to our current knowledge of the functions of the thymus and its T cells in immunity.  相似文献

Cell fractions isolated from lacunae of palatine tonsils with different functional activities were taken from 40 children 6-15 years of age. The cell fractions were examined using vital phase-contrast microscopy, light microscopy of Pappenheim-stained smears and the Feulgen cytophotometry. Lymphocytes with signs of immunological involvement were shown to constitute 95 per cent of the whole cell lacunar population of the active functional tonsils. 35 per cent of lymphocytes, being in different stages of blast transformation, were found. The most intense blast transformation was revealed in patients with tonsil inflammation. 5-13 per cent of lacunar nuclei with DNA content above the diploid level were found in these patients. Some multicellular islets accumulating small lymphocytes, lymphocytes at different stages of blast transformation, and macrophages were revealed to imitate the cooperation of immunocompetent cells. The data obtained can be helpful in the practical surgery of palatine tonsils.  相似文献

The development of splenic B cells that can be induced to proliferate by soluble anti-immunoglobulin (anti-Ig) reagents requires 7 to 9 months in normal mice. We have found that this age-associated response is enhanced by thymic deprivation. Both neonatally thymectomized LAF1 mice and thymectomized, lethally irradiated, and bone marrow-restored Balb/c mice respond earlier and more strongly to anti-Ig than their sham controls. Nevertheless, at least 3–4 months are still required after thymectomy before a response can be measured. The earlier and enhanced response to anti-Ig seen in thymectomized animals is not due simply to an increase in the total number of Ig-positive spleen cells. The age-associated response of splenic B cells to anti-Ig we have observed in normal mice may be explained by the “natural” loss of thymic influence that occurs with age.  相似文献

In studies of the mouse thymus, lymphocyte mitoses are seen to be most frequent in the thymus cortex. There is evidence from thymic grafts that a hypothetical factor, thymopoietin, may stimulate mitosis of thymic lymphocytes. It is a factor which is postulated to act in conjunction with the PAS-positive mesenchymal reticular cells and epithelial reticular cells of the cortex. The thymus medulla is necessary for the integrity of thymic grafts, and may also elaborate a secretion for maintaining the cellular functions of the gland. Thymectomy has been used as a gauge for judging normal thymic function and results, in the mouse, in lymphopenia, degeneration of spleen and lymph nodes, delayed rejection of skin allografts, reduced ability of spleen cells to mount the graft versus host reaction, and reduced primary immune response to certain antigens. Correction of these deficiencies offers a means of evaluating various thymic extracts and grafts. Lymphocytosis-stimulating hormone (LSH) is known to maintain the peripheral lymphoid organs and cause lymphocytosis in the thymectomized animal. Diffusion chamber studies of thymic grafts also show restored lymphoid tissue by a cell-free factor (CIF). These two factors may be the same and probably represent the basis of the highly purified lymphocyte-stimulating proteins, LSHr and LSHh, which restore the L/P ratio in thymectomized animals and may stimulate lymphopoiesis in spleen and lymph nodes. LSHr, unlike LSHh, increases the total lymphocyte count. LSHr has been found to increase the humoral antibody response in neonatal mice both by the PFC technique and by direct hemolysis of sheep erythrocytes. Homeostatic thymic hormone (HTH) is a thymic extract of small molecular weight and contains nucleic acid. In the thymectomized guinea pig it has been found to maintain normal levels of lymphocytes in the blood, spleen and lymph nodes, to restore antibody titers to typhoid H antigen and to restore the toxic allergic reaction. Thymic humoral factor (THF) is of smaller molecular weight (less than 1,000) and probably is not a protein. It also enhances lymphoid proliferation in neonatally thymectomized mice. There is evidence that THF participates in humoral antibody formation because it stimulates PFC formation from neonatally thymectomized mice after inoculation with sheep erythrocytes. Its effects on cell-mediated immunity are seen from findings that injection of THF restores the ability of thymectomized mice to reject skin allografts. THF enables spleen cells from thymectomized or neonatal animals to mount the graft versus host reaction, and causes maturation of bone marrow cells and spleen or lymph node cells so that they can participate in the graft versus host reaction. It has been reported to stimulate lymphocytes to kill isogeneic tumor cells in vitro. Thymosin is protein extracted from the thymus. The influence of thymectomy on the appearance of idiopathic paraproteinemia (IP) during aging was investigated in mice of the C57BL/KaLwRij and the CBA/BrARij strains, which under normal conditions develop IP in high and low frequency, respectively. Compared with sham-thymectomized mice, C57BL mice thymectomized at a young adult age showed a markedly increased frequency and an earlier onset of IP during aging; this was even more pronounced in neonatally thymectomized mice. A similar effect of thymectomy was also observed in mice of the CBA strain. Restriction in the heterogeneity of the serum immunoglobulins and the appearance of transient homogeneous Ig components was another frequent finding and this often preceded the appearance of IP in mice of both strains. Thymectomy did not substantially influence either the incidence of paraproteinemias due to a B cell malignancy or the isotype distribution among the paraproteins produced. The results are compatible with the hypothesis that IP develops in three stages as a consequence of an age-related immunnodeficiency that primarily affects the T immune system.  相似文献

Human lymphatic endothelial cells (LECs) have isolated prevalently from human derma and tumors. As specialized lymphatic organs within the oropharynx, palatine tonsils are easily obtained and rich in lymphatic venules. Using a two-step purification method based on the sorting of endothelial cells with Ulex Europaeus Agglutinin 1 (UEA-1)-coated beads, followed by purification with monoclonal antibody D2-40, we successfully purified LECs from human palatine tonsils. The LECs were expanded on flasks coated with collagen type 1 and fibronectin for up to 8-10 passages and then analyzed for phenotypic and functional properties. Cultured cells retained the phenotypic pattern of the lymphatic endothelium of palatine tonsils and expressed functional VEGFR-3 molecules. In fact, stimulation with VEGFR-3 ligand, the vascular endothelium grow factor C, induced a marked increase in cell proliferation. Similarly to blood endothelial cells (BECs), LECs were able to form tube-like structure when seeded in Cultrex basement membrane extract. Comparative studies performed on LECs derived from palatine tonsils and iliac lymphatic vessels (ILVs), obtained with the same procedures, showed substantial discrepancies in the expression of various lymphatic markers. This points to the existence of micro- and macrovessel-derived LECs with different phenotypes, possibly involving different biological activities and functions. Palatine tonsil- and ILV-derived LECs may, therefore, represent new models for investigating function and biochemical properties of these lymphatic endothelia.  相似文献

Mouse bone marrow contains cells capable of responding in vitro to the T cell mitogens PHA and Con A. These cellular responses are not demonstrable in the marrow of athymic nude mice (when compared with heterozygous littermates) and are depressed 47% in the marrow of neonatally thymectomized LAF1 mice (when compared with sham-operated littermates). Therefore, the population of “bone marrow T cells” is thymus dependent.  相似文献

The incidence of translocation of viable indigenous bacteria from the gastrointestinal tract to the mesenteric lymph node, spleen, liver, and kidney was compared in neonatally thymectomized mice and sham-thymectomized specific pathogen-free mice. The immunologic responses of the thymectomized mice to sheep erythrocytes were decreased compared to the responses of sham-thymectomized mice. Strictly anaerobic bacteria were isolated from only 1.8% of the organs from thymectomized mice and from none of the organs of shamthymectomized mice. Aerobic or facultatively anaerobic bacteria were cultured from 27.4% of the organs of thymectomized mice. Of the thymectomized mice, 70.7% contained viable aerobic or facultatively anaerobic bacteria in one or more of their organs tested, compared with only 10% of the sham-thymectomized mice.Escherichia coli was the predominant bacterial species isolated from these organs, althoughStaphylococcus aureus, Streptococcus, andCorynebacterium also were present.Bacteroides were the only strictly anaerobic bacteria cultured. Neonatal thymectomy promotes the translocation of certain indigenous bacteria from the gastrointestinal tract to the mesenteric lymph node, spleen, liver, and kidney.  相似文献

Murine cytomegalovirus was utilized as a model for human cytomegalovirus, which had no experimental animal, to study immunoprophylaxis of the cytomegalovirus infections. (1) Murine cytomegalovirus (MCMV) serially propagated in mouse embryonic fibroblasts had lost pathogenicity for weanling mice including neonatally thymectomized mice. (2) The cell culture-adapted MCMV was effective as a "live, attenuated virus vaccine" against challenge by virulent, mouse-passaged MCMV. (3) The immunization via intraperitoneal route protected mice from every parameter of MCMV infection. These included clinical signs, virus replication, histopathology and mortality. (4) The protective immunity was active against the virulent MCMV which was not neutralized by the rabbit anti-attenuated MCMV serum.  相似文献

M-cells are believed to play a pivotal role in initiation of the immune response. These cells, located in the epithelia that overlie mucosal lymphoid follicles, are responsible for the active uptake of particulate antigens and for their translocation to the underlying lymphoid tissue. The identification of reliable markers for M-cells is therefore extremely important for the study of the initial steps that lead to the immune response. For this purpose, we studied cytokeratin 20 (CK20) expression in the epithelium of rabbit palatine tonsils by immunofluorescence, confocal microscopy, and Western blotting. CK20+ cells were observed in all rabbit palatine tonsils examined. By Western blotting, one CK20-immunoreactive band was identified at 46 kD on samples of proteins from the intermediate filament-enriched cytoskeletal fraction of tonsil epithelium. Double labeling of CK20+ cells with cell-specific markers confirmed that such cells were actually M-cells. Moreover, CK20+ M-cells displayed a mature phenotype (they formed pockets harboring lymphoid cells) and were functionally competent because they could take up particulate antigens from the pharyngeal lumen. We conclude that CK20 is an M-cell marker for rabbit palatine tonsils. Moreover, we can hypothesize the use of M-cells as a possible site for antigen delivery of particle-based vaccines.  相似文献

By 7 weeks post-grafting, the number of small lymphocytes in the thoracic duct lymph (TDL) and blood of the thymus-grafted neonatally thymectomized adult rats had increased to 60% of the number of cells in sham controls, or 2-1/2 times thymectomized control values. This increasing consisted almost exclusively of long-lived, recirculating small lymphocytes and corresponded to a 60% recovery of cellular immunocompetence as measured by the mixed lymphocyte reaction (MLR). Associated with the return of cellular immunocompetence was an increased incorporation of 3H-uridine by the small lymphocytes. Cells from thymectomized animals grafted with lymph node fragments demonstrated no significant increase in lymphocyte numbers nor was there a return of immunocompetence as compared to thymectomized controls.  相似文献

A total of 385 albino Swiss mice, of both sexes, between 24 and 30 hours of age are neonatally thymectomized, by means of a suction pump method. One batch is operated under ether anesthesia, another with sodium pentobarbital, and a third one with Cryoanesthesia. The last batch is subdivided into 2. In one of these the mother is anesthetized with sodium pentobarbital intraperitoneally, during the postoperative recovery of the litter, and in the other the mother is left without treatment. The mothers of the first two batches of thymectomized animals are anesthetized with the same products as their offspring. The percentage of survival of thymectomized breeding mice in greatest in the batch operated under Cryoanesthesia and whose mothers received the above mentioned treatment. Statistically the differences are very significant and they might be accounted to the fact that the mothers are anesthetized while their offspring normalize their physiological constants with better recovery in feeding, hydration, shelter and rest.  相似文献

Differentiation of epithelial and lymphoid tissues of the palatine tonsils was studied in human embryos at the age of 8-34 weeks of development by means of histochemical, immunomorphological and morphometric methods. The anlage of the palatine tonsils appears at the age of 9 weeks of fetal development. At the age of 13-14 weeks of fetal development the tonsil suspension contains 2 subpopulations of lymphocytes possessing properties of T-cells differing in the ability of their superficial receptors to interact with sheep erythrocyte antigens forming rosettes (RFC--rosette forming cells) and with antigens of their own erithrocytes (autoRFC). The number both increases sharply by the 16th week of gestation. Simultaneously, essential alterations are noted in epithelial and lymphoid tissues. In epithelium of crypts cornified cells appear; the amount of lymphoid tissue increases sharply, primary follicles without reactive centers appear, lymphocytic infiltration of epithelium occurs. The amount of RFC does not change considerably, and the amount of autoRFC has a tendency towards some increase. From the data obtained, it is possible to suggest that human palatine tonsils already at embryonic period participate in functioning of immunogenic organs and in maintaining of immunologic homeostasis of the fetal organism.  相似文献

A method for chemically fractionating lymphatic organs has been described. The method has been shown to be applicable to bovine palatine tonsils, sheep palatine tonsils, and bovine thymus. Approximately 50 per cent of the dry weight of tonsils and about 30 per cent of thymus has been found to be soluble in the 0.15 M NaCl extract. Four components have been isolated which together account for 65 per cent by weight of the material in the extracts. Four other components have been identified and partially defined by means of electrophoretic mobility, solubility, or some other chemical or physical property.  相似文献

Neonatally thymectomized and sham-operated rats were given courses of hind foot pad injections of tritiated thymidine after immune stimuli, and the popliteal lymph nodes were examined for the presence of persistently labeled lymphocytes 32 days later. A larger proportion of labeled small lymphocytes was found in the nodes of thymectomized rats than in the nodes of sham-operated rats, indicating that a significant portion of the long-lived nonmigrating cells are B cells. In sham-operated rats given a course of tritiated thymidine after an immune stimulus with a Salmonella flagellin, labeled blasts appeared 3 days after secondary challenges with both the same or a serologically distinct flagellin. In neonatally thymectomized rats, a blastogenic response occurred after challenge with the same flagellin but not after challenge with the non-specific flagellin. Thus, in normal rats there are also long-lived nonmigrating T cells in primed lymph nodes, and these are the cells responsible for the nonspecific blasto-genesis seen in earlier experiments.  相似文献

Tonsils comprise part of the mucosal immune system and contain lymphocytes, macrophages, and follicular dendritic cells (FDCs). FDCs are located in the B cell area of the follicles of secondary lymphoid organs, such as the spleen, tonsils, or lymph nodes, and they trap and retain immune complexes on their surfaces to regulate B cell activation and maturation. Stromal cells from the palatine tonsils are often used for FDC in vitro studies, and it has been reported that human palatine tonsils may be a good source of multipotent mesenchymal cells. Therefore, we assessed whether tonsil-derived mesenchymal stromal cells could differentiate into a FDC-like phenotype. We discovered that stromal cells isolated from human tonsils not only had the potential to differentiate into various cell types of mesenchymal origin, but they also could differentiate into FDC-like cells under cytokine stimulation in vitro.  相似文献

To study the lifespan of virgin T lymphocytes, we removed the thymus from adult female mice and then, at various times afterward, tested their ability to mount an immune response to a newly encountered Ag, the male Ag H-Y. We found that unprimed thymectomized mice were able to generate a primary response to H-Y for some time after thymectomy but lost this ability at approximately 6 mo. In contrast, mice that were primed to H-Y just after thymectomy continued to display immunological memory to H-Y for >1 year. These experiments show that primary immune responses disappear in the absence of a thymus.  相似文献

Groups of mice were neonatally thymectomized and treated with antithymocyte serum (ATS) prior to challenge infection with viable yeast phase (YP)Histoplasma capsulatum G-17M. Moderate leucocytosis and moderate lymphopenia were seen in immunodeficient animals after infection. Surviving immunodeficient mice exhibited low levels of migration, inhibition activity, while peritoneal exudate cells and spleen cells harvested from surviving infected and untreated normal mice showed significant migration inhibition in the presence of histoplasmin antigen.The LD50 values for YP cells ofH. capsulatum were 1.1×106 for normal untreated mice, 6.0×105 for thymectomized mice, and 6.3×105 for ATS-treated mice. Thymectomized mice that also received ATS treatment exhibited an LD50 of 1.7×105 and were 6.5 times more susceptible to infection then normal mice. Mice which were either thymectomized or treated with ATS were 1.7 times as susceptible as normal mice to infection withH. capsulatum. The criterion of susceptibility is a decrease in the LD50 value.  相似文献

B6-lpr/lpr mice develop massive T cell lymphoproliferation, as associated with autoimmune disease. We found a reduced NK activity in the spleen of B6-lpr/lpr mice. Neonatal thymectomy markedly retarded the development of lymphoproliferation and the development of autoantibodies in the B6-lpr/lpr mice. These animals had a higher level of NK activity in the spleen. When the neonatally thymectomized B6-lpr/lpr mice were given anti-asialo GM1 serum (30 microliter) four times at 6-day intervals, initiated at the 8th-10th postnatal week, these mice developed lymphoproliferative disorders and splenomegaly, concomitantly with depression of NK activity. It is therefore tempting to speculate that NK cells are involved in the regulation of the occurrence of lymphoproliferative disorders.  相似文献

The human palatine tonsils have surface and crypt stratified epithelium and may be initiated via the epithelium to mount immune responses to various presenting antigens. Here we investigated the expression and function of tight junctions in the epithelium of human palatine tonsils from patients with tonsillar hypertrophy or recurrent tonsillitis. Occludin, ZO-1, JAM-1, and claudin-1, -3, -4, -7, -8, and -14 mRNAs were detected in tonsillar hypertrophy. Occludin and claudin-14 were expressed in the uppermost layer of the tonsil surface epithelium, whereas ZO-1, JAM-1, and claudin-1, -4, and -7 were found throughout the epithelium. In the crypt epithelium, claudin-4 was preferentially expressed in the upper layers. In freeze-fracture replicas, short fragments of continuous tight junction strands were observed but never formed networks. In the crypt epithelium of recurrent tonsillitis, the tracer was leaked from the surface regions where occludin and claudin-4 disappeared. Occludin, ZO-1, JAM-1, and claudin-1, -3, -4, and -14, but not claudin-7, mRNAs were decreased in recurrent tonsillitis compared with those of tonsillar hypertrophy. These studies suggest unique expression of tight junctions in human palatine tonsillar epithelium, and the crypt epithelium may possess an epithelial barrier different from that of the surface epithelium.  相似文献
