Percutaneous osseointegrated implant technology provides a potential alternative to current socket prosthetics for individuals with limb loss. However, similar to other percutaneous devices, there remain concerns of periprosthetic infection. To understand this process of infection, bacterial isolates were collected and characterized from a sheep model of osseointegration. CSA-13, a novel cationic steroid antimicrobial, was used at the skin/implant interface in an attempt to reduce the rate of infection. Results indicated that in this application, normal flora and environmental organisms continued to colonize the skin/implant interface as well as cause infection in the presence of CSA-13. Two factors are believed to have contributed to this outcome: the delivery of CSA-13 and the lack of a skin seal at the skin/implant interface, which would create a biological barrier to infection.  相似文献

In comparative studies on potyviruses from West African yams (Dioscorea spp.) the following isolates were used: Dioscorea greenbanding mosaic virus (DGMV) and a Nigerian yam virus (YV-N), both isolated from Dioscorea rotundata, and a beet mosaic virus isolate from D. alata (BtMV-Y) formerly designated Dioscorea alata ring mottle virus. Naturally infected D. alata containing very few particles of BtMV-Y, contained primarily particles of a second potyvirus (Dioscorea alata virus, DaV) which could not be transmitted but which was included in these studies wherever possible. The normal lengths of DGMV, YV-N, DaV, and BtMV-Y were 754, 772, 805, and 812 nm, respectively. All viruses induced cytplasmic inclusions of the pinwheel type and laminated aggregates. In addition, the nucleoli of BtMV-Y infected cells contained characteristic electron dense inclusions. The buoyant density of purified DGMV and BtMV-Y in CsCl was 1.336 g/cm3 and 1.321 g/cm3, respectively. The sedimention velocities (Srel) of DGMV, YV-N, and BtMV-Y were 156, 158, and 162 Srespectively. In SDS-polyacrylamide gel electrophoresis the coat protein of purified DGMV and YV-N all migrated as a single band with an apparent molecular weight of 36 kd. Coat protein of purified DaV showed up to 5 bands with molecular weights of 36 to, 32 kd. Polypeptides of purified BtMV-Y had an estimated molecular weight of 35 kd but those from infected plant extracts had a molecular weight of 36 kd. DGMV, YV-N, and BtMV-Y particles contained a single nucleic acid with an apparent molecular weightof 3.2, 3.2, and 3.1 Md, respectively. Using λ-DNA digested with Hind III as a marker, the molecular weight of DGMV and BtMV-Y nucleic acid was calculated to be 3.6 Md ± 10%. The nucleic acid was determined to be single-stranded RNA by enzymatic digestion and by staining with acridine orange. In serological studies using immunoelectron microscopy (IEM), electro-blot immunoassay (EBIA), and enzyme-linked immunosorbent assay (ELISA), DGMV and YV-N were closely related. Strong serological reactions were also obtained in IEM and EBIA when DGMV and YV-N were tested with antiserum to yam mosaic virus (YMV). Antisera against DGMV, YV-N, and YMV also reacted strongly with DaV antigen. Serological reactions between these viruses and BtMV-Y were usually not found or were weak. A very close serological relationship could be detected between BtMV-Y and beet mosaic virus isolated from beet (BtMV); both isolates were also very similar in host range, symptomatology, and cytopathology.  相似文献

A set of 300 bacterial strains isolated from various extreme environments were screened for the presence of cellulase activity on CMC agar plates. Phylogenetic analysis of the positive strain, based on 16S rRNA gene sequences indicated that the isolates were clustered within Firmicutes and Actinobacteria. A majority (17) of the isolates were identified as Bacillus, Paenibacillus, and Lysinibacillus sp., and the remaining three were identified as Arthobacter, Rhodococcus, and Bhargavaea cecembensis. Among the 20 positive isolates, 6 were evaluated for the production of cellulases on five different cellulosic substrates. Two isolates, B. cecembensis and Bacillus sp., based on maximum enzyme production on all cellulosic substrates, especially CMC and rice straw, were evaluated in terms of enzyme properties and kinetics. The enzymes of these two isolates are found to be active over broad range of pH and temperature. Such thermostable enzymes facilitate the development of efficient and cost-effective forms of the simultaneous saccharification and fermentation process converting lignocellulosic biomass into biofuels and value-added products.  相似文献

Seeds from cotton plants infected with Xanthomonas campestris pv. malvacearum were collected in different parts of Nicaragua in 1986. When the seeds were homogenized the pathogen could not always be identified by dilution plating. Therefore, an enrichment of bacteria on the natural host was induced before isolation. The cotton seeds were shaken with water and sand for 2 days and then sown in sand for germination. Rather often the developing cotyledons showed typical water-soaked spots, from which the pathogen could be isolated easily. This new method needed more time but made it possible to detect a low level of bacterial infestation. Altogether, 42 bacterial isolates were obtained. For inoculation experiments suspensions with 5x105 CFU - ml?1 were infiltrated into cotton leaves. Incubations of inoculated plants in growth chambers, but not in greenhouses, resulted in typical and uniform disease symptoms (water-soaked leaf spots). Nine of the ten cotton differentials tested were highly susceptible to all the 42 bacterial isolates. Since only line 101-102B proved to be resistant, the Nicaraguan isolates of bacterial blight of cotton were characterized as race 18, of the pathogen. The main cotton cultivars grown in Nicaragua (H-373 and G-286) were strongly affected by the isolated bacterial strains. In order to reduce the disease incidence in Nicaragua, the cultivation of resistant cotton varieties is suggested.  相似文献

Diversity of Bacterial Isolates from Commercial and Homemade Composts   总被引:1,自引:0,他引:1  
The diversity of heterotrophic bacterial isolates of three commercial and two homemade composts was studied. The commercial composts were produced from poultry litter (PC), sewage sludge (SC), municipal solid waste (MC), and homemade composts (thermal compost [DC] and vermicompost [VC]) from food wastes. The taxonomic and physiological diversity of the heterotrophic culturable bacteria was assessed using phenotypic and genotypic characterization and the analysis of the partial 16S rRNA gene sequence. Composts DC and SC presented the higher genotypic diversity, as could be inferred from the number of distinct genotypic patterns observed, 28 and 21, respectively. Gram-positive bacteria, mainly Firmicutes, were predominant in all the composts. Some organisms related with taxa rarely reported in composts, as Rhodanobacter spathiphylli, Moraxella osloensis, Lysobacter, Corynebacterium, Pigmentiphaga kullae, and new taxa were also isolated. The highest relative proportion of isolates able to degrade starch was found in compost SC (>70%), to degrade gelatine in compost DC (>70%), to degrade Tween 80 in compost PC (>90%), and to degrade poly-epsilon-caprolactones in compost DC (>80%). Compost MC presented the lowest relative proportions of isolates able to degrade starch (<25%), gelatine (<20%), and poly-epsilon-caprolactone (<40%). When compared with the others, the homemade composts presented higher relative proportions of Gram-positive isolates able to inhibit the target organisms Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, or Pseudomonas aeruginosa. In compost MC, none of the Gram-positive isolates was able to inhibit those targets.  相似文献

The characterization of 61 Streptococcus suis strains isolated from Chinese slaughter pigs was investigated. S. suis serotypes 1, 2, 3, 4, 7, 9, 10, 11, 12, 13, 14, 15, 16, 22, 23, 25, 26, 28, 29, and 1/2 were found in the isolates by serum agglutination. Of all the prevalent serotypes, S. suis serotype 7 is the most predominant circulating in Chinese slaughter pigs. The virulence-associated genes profile and multilocus sequence typing scheme of the isolates were analyzed. The mrp-/epf-/sly- virulence-associated genes type is the most prevalent in the isolates from slaughter pigs. It is the first time to find S. suis serotypes 7 and 9 isolates with epf. The serotypes 7 and 9 isolates with mrp and/or epf genes did not express MRP and/or EF in the present research. Thirteen new ST types were identified for the first time. ST1 complex and ST27 complex of S. suis are prevalent in China. This paper supplied information to understand the characteristics, such as capsular serotypes, virulence factors, and gene backgrounds of S. suis carried by slaughter pigs.  相似文献

Two hundred sixty-six strains of Klebsiella pneumoniae isolated from natural water sources in geographically diverse areas (Florida, Massachusetts, and Oregon) were analyzed to determine the serotype, biochemical, virulence, and antimicrobial susceptibility differences between these natural strains and human Klebsiella isolates. Sixty of 72 defined serotypes were found among 210 typable strains. Geographic patterns were present, but in general were not pronounced among serotypes. Reactions with 28 biochemical tests showed percentage responses which were very similar to the summaries of primarily human Klebsiella isolates (as reported by Edwards and Ewing, 1972) and that represented diverse geographic sampling. Virulence studies in representative strains showed no geographic variability and little difference from comparable hospital patient-obtained isolates. In contrast to human hospital isolates, strains demonstrated 90% or greater susceptibility to all antibiotics except ampicillin and carbenicillin; and in further contrast, there was little multiple antibiotic resistance beyond that with ampicillin and carbenicillin.  相似文献

A total of 286 isolates of Pseudomonas maltophilia was collected from sheep exhibiting brown or yellowish fleece rot and from fly-strike lesions. Enzyme activities for 10 of the isolates were examined by plate tests and with the API ZYM system and compared with the enzymatic profile of a human type strain of P. maltophilia. The fact that ovine isolates of P. maltophilia are biochemically similar to pathogenic human strains suggests there may be an association between this organism and the brown to yellow type of fleece rot.  相似文献

The adaptive radiations of cichlid fishes in East Africa have been associated with the acquisition of evolutionary novelties as well as the ecological opportunities existing in the East African Great lakes. Two remarkable evolutionary innovations are the pharyngeal jaw apparatus, found in all cichlid species, and the anal fin egg-spots of mouthbrooding cichlids. Based on their conserved functions during the development of both the jaw apparatus and pigmentation, the endothelin ligands and receptors form a putative link between these naturally and sexually selected traits. Here we study the evolutionary history of four members of two endothelin pathways (Edn1/EdnrAa and Edn3b/EdnrB1a) to elucidate their possible roles during the evolution and development of key innovations in East African cichlids species. The analyses performed on partial sequences (ca. 6,000 bp per taxon) show that all four endothelin family members evolved under purifying selection, although both ligands are characterized by an accelerated rate of protein evolution in comparison to the receptors. In accordance with earlier findings, we show that the mature protein sequence of Edn1 and Edn3 are highly conserved, also in cichlids, whereas the preproendothelin parts are variable indicating relaxed selective constraints. In the receptors, nonsynonymous substitutions were mainly found in the ligand-binding domains suggesting functional divergence. Gene expression assays with Real-Time PCR indeed reveal that the two studied endothelin pathways are expressed in the cichlid pharyngeal jaw and in the haplochromine egg-spot (among other pigment-cell containing tissues), suggesting their involvement during morphogenesis of naturally and sexually selected traits in cichlids.  相似文献

Manganese Oxidation by Bacterial Isolates from the Indian Ridge System   总被引:1,自引:0,他引:1  
The abundance and activity of culturable manganese-oxidizing bacteria were assessed from near-bottom water samples of the tectonically active Carlsberg Ridge. Retrievable counts as colony forming units (CFU) on dilute nutrient agar medium (dilNA = 2 gm l−1 nutrient broth+2% agar) and on dilNA supplemented with 1, 2 and 3 mM MnCl2·4H2O were in the order of 106 CFU l−1. Retrievability of heterotrophs ranged from non-detectable levels (ND) to 2.82 × 106 CFU l−1. The retrievable counts on Mn amended dilNA ranged from ND to 3.21× 106, 1.47 × 106 and 1.45 × 106 CFU l−1 on 1, 2 and 3 mM, respectively. About 87% of the Mn tolerant isolates (n = 39) showed taxonomic affinities to Pseudomonas I and II sp. Two representative strains CR35 and CR48 (CR–Carlsberg Ridge) isolated on manganese-supplemented media were tested for their ability to tolerate a range of Mn amendments from 1 nM to 100 mM in terms of growth and respiration. CR35 represents 66% of the total CFU (3.04 × 106 CFU l−1), while CR48 represented only 6% of the total CFU (1.05 × 106 CFU l−1). The colonies of these two isolates were dark brown in color suggesting precipitation of Mn as oxide. Tests for the effect on growth and respiration were conducted in media simulating heterotrophic (amended with 0.01% glucose) and lithotrophic (unamended) conditions. Maximum stimulation in growth and respiration of CR35 occurred at 100 μM Mn both in unamended and amended media. At levels of Mn greater than 100 μM the counts decreased steadily. Total respiring cells of CR48 were stimulated to a maximum at 1 μM Mn in unamended medium and 1 nM in amended medium. Total cells counts for the same decreased beyond 100 μM Mn in unamended and 1 nM in amended medium. The isolates were tested for their ability to oxidize Mn ammendments from 1 μM to 10 mM Mn. At the end of a 76-day incubation period, there was evidence of manganese oxide precipitation at high Mn concentrations (≥1 mM) as a dark brown coloration on the sides of culture tubes. Highest Mn oxidation rates were observed at 10 mM Mn(II) concentration with CR35 oxidizing 27 and 25 μM Mn day−1 in unamended and amended condition, respectively. CR48 oxidized Mn at the rate of 26 μM Mn day−1 in unamended medium and 35 μM Mn day−1 in amended medium. Scanning electron microscope (SEM) observations of both isolates revealed free-living cells in clustered matrices ≈2 μm diameter. Energy dispersive spectrum of the cell matrix of CR35 cultured in 1 mM Mn detected 30% Mn, while the cell aggregates of CR48 harbored 7–10% Mn. The relatively high specific activity of these mixotrophic bacteria under relatively oligotrophic conditions suggests that they may be responsible for scavenging dissolved Mn from the Carlsberg Ridge waters and could potentially participate in oxidation.  相似文献

SYNOPSIS. Haemogregarines are described from the peripheral blood and in some instances from internal organs of 4 species of frogs from East Africa. The parasites from Hyperolius puncticulatus and from Rana wittei have not been reported previously. The haemogregarine from R. mascareniensis is possibly a young stage of a form previously inadequately described. R. angolensis was parasitized by Haemogregarina theileri and also by an organism of uncertain affinity found mainly in the lung capillaries.  相似文献

In Africa, the crinivirus Sweet potato chlorotic stunt virus (SPCSV) exists in two serologically and genetically distinct strains, geographically distinguished as a West African (SPCSVWA) and an East African (SPCSVEA) strain. To obtain a better understanding of the genetic diversity among SPCSVEA isolates, the major coat protein (CP) and heat shock protein 70 homologue (Hsp70h) gene sequences of 24 further isolates of SPCSVEA were determined and compared. SPCSVEA diversity was also examined using available monoclonal antibodies (mAbs) to SPCSVEA but there was no apparent coincidence between CP and partial Hsp70h gene nucleotide sequences and the subdivision of SPCSVEA isolates by the mAbs into two serotypes, suggesting this latter may not be of great biological significance. The nucleotide (nt) sequences of isolates of SPCSVEA displayed a high degree of conservation and the only variation observed consisted of a few base exchanges. Pairwise alignments of CP nucleotide sequences revealed differences of <4% between SPCSVEA isolates. Comparisons with published SPCSV sequences confirmed a more distant relationship (up to 34.6% nt; 12% amino acid divergence) between the Hsp70h sequences of isolates of SPCSVEA and SPCSVWA and indicated that SPCSVEA in East and Southern Africa is the more homogeneous than SPCSVWA isolates from West Africa, North and South America, which were up to 12.4% nt divergent among themselves.  相似文献

Two isolates of Pratylenchus coffeae were collected from coffee roots (in Marília, São Paulo State, Brazil) and Aglaonema (in Rio de Janeiro City, Rio de Janeiro State, Brazil) and maintained in the laboratory on alfalfa callus. Twenty-four plants were tested in the greenhouse to characterize the host preference of these isolates. The host ranges of the isolates differed from each other and, interestingly, coffee, banana, and citrus were not among the better hosts of either isolate. Rather, sorghum, maize, rice, millet, okra, melon, eggplant, and lettuce were the best hosts of the Marília isolate. Poor hosts included French marigold, Rangpur lime, banana, sesame, peanut, sunflower, cotton, French bean, onion, and small onion. The best hosts of the Rio de Janeiro isolate were sesame, soybean, sorghum, castor oil plant, watermelon, squash, eggplant, and melon; the poorest hosts were French marigold, coffee, Rangpur lime, banana, sunflower, peanut, maize, millet, French bean, cotton, onion, sweet pepper, lettuce, okra, and small onion. These isolates have important molecular and morphological differences, suggesting host preference is linked to these characteristics.  相似文献
