Regeneration blastemas were exchanged between surgically constructed forelimbs comprised of symmetrical tissues (double-anterior and double-posterior) and normal, unoperated forelimbs. Normal blastemas grafted at the stage of medium bud (MB) onto double-half forelimb stumps regenerated normal skeletal patterns in nearly all cases. Double-half blastemas transplanted at the stage of MB onto normal forelimb stumps did not regenerate complete limb patterns. These results indicate that a double-half blastema cannot be “rescued” by transplantation to a normal stump and that a double-half limb stump does not interfere with the ability of a normal blastema to distally transform. The regeneration blastema possesses sufficient positional information at the stage of MB to permit it to develop autonomously. Supernumerary forelimbs resulted from several types of graft-stump combinations. The location and handedness of these supernumerary limbs are predicted by the rules of a recently presented model for pattern regulation in epimorphic fields [French, V., Bryant, P. Scanning electron microscopy was used to investigate the morphology of intact imaginal wing discs of third-instar larvae of Drosophila melanogaster. The disc stalk, nerve and tracheal entries and the surface ultrastructure of the columnar cells, the peripodial membrane cells, and the adepithelial cells are described. The behavior of various fragments of the wing disc during culture in vivo was also studied. After injuring a wing disc by cuts with a tungsten needle, during the first day of culture the epithelium curls and the wound surface contracts. Subsequent closure of the wound in 34 and 14 sectors, in fragments generated by straight cuts and in central squares, leads to the confrontations of cells from formerly separate positions, as was proposed in connection with the polar coordinate model of French, Bryant, and Bryant [(1976). Wound healing comprises three steps: (1) Cell debris is removed; (2) occasional cell processes span the wound; (3) all cells at the wound edge contact cells on the opposite side. After 2–3 days, a continuous epithelium is re-established. The tissue distortion may lead to transient contacts of the columnar epithelium with the peripodial membrane and with itself. The latter can explain the occasional duplications of structures which, according to the fate map, arise from near the wound edge, and which have been previously reported from cultured imaginal disc fragments. The tissue movements appear to be due to the contractile properties of individual cells.  相似文献

The relationship between limb development and limb regeneration is considered with regard to the mechanisms by which pattern is established during limb outgrowth. In a previous paper (Muneoka, K. and Bryant, S. V. 1982 Nature (London) 298, 369-371) the interaction between cells from the developing limb bud and the regenerating limb blastema was found to result in the production of organized supernumerary limb structures. In this paper the relative cellular contribution from developing and regenerating cells to supernumerary limbs resulting from contralateral grafts between limb buds and blastemas has been analyzed using the triploid cell marker in the axolotl. Results show that there is substantial participation from both developing and regenerating limb cells to all supernumerary limbs analyzed. These data lend further support to the hypothesis that developing and regenerating limbs utilize the same patterning mechanisms during limb outgrowth. This conclusion is discussed in terms of patterning models for developing and regenerating limbs and it is proposed that the rules of the polar coordinate model can best explain the behavior of cells during limb development as well as limb regeneration.  相似文献

The polar-coordinate model of French, Bryant & Bryant (1976) describes epimorphic regeneration in insects and amphibians, and correctly predicts some surprising phenomena. Their model rests on two main rules. It is shown here that the first of these, the Rule of Intercalation, expresses a requirement that the pattern of positional values of the cells shall be continuous, and that the other, the Rule of the Complete Circle, is not an independent hypothesis, but simply a consequence of the continuity requirement. The formulation of the rules in terms of continuity is coordinate-free and applicable in any number of dimensions: polar coordinates do not have the fundamental significance attached to them by French, Bryant & Bryant (1976), nor are there any grounds to think that the system of positional values in an amphibian limb is two-dimensional. Some simple topology helps to clarify the concepts and prove these points. The phenomena codified by French et al. are to be expected in any epimorphically regenerative organ whose pattern of growth and pattern of positional values are defined by a maintained system of diffusible chemical signals.  相似文献

The polar co-ordinate model for pattern regulation in epimorphic fields, as first proposed by French, Bryant & Bryant (1976), belongs to a general class of models employing two co-ordinates for describing the field value distribution over a monolayer of cells.I first analyse the constraints imposed by the use of polar co-ordinates for biological modelling. These constraints entail: distinction of the actual and derived spaces for the field with their specific and characteristic mapping modes; representation of the field value or a measure of the morphogenetic activity at various positions in the actual space by two polar co-ordinates—the phase or angle and the radius—in the derived space; and attribution of a singular property to the origin in the derived space, which ought to be mapped onto a putative field centre in the actual space.Upon reformulating the polar co-ordinate model while adhering to these constraints, one realizes that the choice of the shortest route between two points in the derived space applies only to the angular, and not to the radial, co-ordinate. Hence the shortest route between two points cannot be conceived as a straight line connecting them.If we assume that cells have certain limits in resolving differences in the field value, the field will virtually fall into territories of distinctive field values, each of which might encompass more than one cell; cells that lie within a single territory should be mutually interchangeable for prospective developmental pathways which may themselves diversify. The central postulate of the model is that disparate field values brought in apposition will induce a field value intercalation through cell proliferation to restore an unbroken sequence of field values for the newly emerging array of territories. This is assumed universally for the angular component of the field value but it need not apply to the radial component.It follows that angular territories meeting at the field centre will produce a central discontinuity of the field value, leading to a perpetual angular intercalation unless some theoretical provision is made. This may be resolved by any ad hoc hypotheses but also by introducing the concept of degeneracy towards the field centre of the angular co-ordinate down to three distinctive values at the innermost level of radial territories. One can then deduce and generalize various empirically recognized behaviours of epimorphic fields. These include the shortest intercalation rule (which may be modified into the more generally applicable, minor-in-major rule), and the distal transformation and so-called complete circle rules (which may now be unified into the more feasible distalization rule proposed by Bryant, French & Bryant, 1981).The models are assessed against various experimental observations mainly for theoretical consistency. A series of experimental tests for various theoretical alternatives are suggested. This work has simply explored the internal logic of the polar co-ordinate model and no suggestion is made, at this stage, to distinguish different families of alternative models for epimorphic systems on experimental grounds.  相似文献

Lobate ctenophores (tentaculates) generally exhibit a remarkable ability to regenerate missing structures as adults. On the other hand, their embryos exhibit a highly mosaic behavior when cut into halves or when specific cells are ablated. These deficient embryos do not exhibit embryonic regulation, and generate incomplete adult body plans. Under certain conditions, however, these deficient animals are subsequently able to replace the missing structures during the adult phase in a process referred to as "post-regeneration." We have determined that successful post-regeneration can be predicted on the basis of a modified polar coordinate model, and the rules of intercalary regeneration, as defined by French et al. (V. French, P. J. Bryant, and S. V. Bryant, 1976, Science 193, 969-981.) The model makes certain assumptions about the organization of the ctenophore body plan that fit well with what we have determined on the basis of cell lineage fates maps, and their twofold rotational ("biradial") symmetry. The results suggest that cells composing the ctenophore adult body plan possess positional information, which is utilized to reconstruct the adult body plan. More specifically, we have found that the progeny of three specific cell lineages are required to support post-regeneration of the comb rows (the e(1), e(2), and m(1) micromeres). Furthermore, post-regeneration of the comb rows involves a suite of cell-cell inductive interactions, which are similar to those that take place during their embryonic formation. The significance of these findings is discussed in terms of the organization of the ctenophore body plan, and the mechanisms involved in cell fate specification. This situation is also contrasted with that of the atentaculate ctenophores, which are unable to undergo post-regeneration.  相似文献

When an amphibian limb, cockroach leg or Drosophila imaginal disc is subjected to a surgical operation, it is capable of regenerating or duplicating certain parts. Although the structure of the regenerated tissue varies depending on the location and mass of the amputated or transplanted part, it can be predicted from a set of formal rules, called the polar coordinate model [French et al., (1976) Science 193, 969-983; Bryant et al., (1981) Science 212, 993-1002]. In the polar coordinate model, it is assumed (and experimentally proven) that the juxtaposition of normally non-adjacent cells stimulates cell proliferation locally, which implies that the underlying mechanism which gives positional values to each cell, is also responsible for the control of cell growth. Because locally activated proliferation alters the shape and size of the developmental field, the question of how to control the cell growth is the central problem in the regeneration of the limbs and imaginal discs. In this paper, I propose a possible underlying mechanism for the 'polar coordinate rules', and show how this mechanistic model explains the experimental results using computer simulation. The proposed mechanism is an extension of Turing's model (1952). In addition to the reaction-diffusion of the molecules, cell proliferation is taken into consideration. With appropriate initial conditions, the computer simulation shows that a small mass of cells grows up to form a mature limb, and that the mature limb is able to respond to surgical operations as predicted by the polar coordinate model.  相似文献

The validity of the “charge-relay” system in serine esterases was examined by use of the general base-catalyzed hydrolysis of ethyl chloroacetate (I) as a model system. The general base catalytic rate for 2-benzimidazoleacetic acid (II) exhibited an eightfold positive deviation from the Brønsted plot including benzimidazole, imidazole, N-methylimidazole (V), and acetate ion, though in nucleophilic catalysis of the hydrolysis of p-nitrophenyl acetate, the point for II conformed to the Brønsted relationship together with imidazole and benzimidazole derivatives. The positive deviation of II from the Brønsted plot for the general base-catalysis was attributed to the cooperativity of the carboxyl group of II, the imidazolyl group of II, and the hydroxyl group of water. The present result provides support for the “charge-relay” system. Furthermore, the (essentially) total loss of the enzymatic activity due to N-3 methylation of histidine-57 in α-chymotrypsin is discussed in comparison to the general base-catalysis of V in the hydrolysis of I, which is also favorable for the “charge-relay” system.  相似文献

Summary Contralateral grafts were performed on the larval limb buds of the anuranBufo bufo. The dorsoventral axis of 80 buds at stages IV or V was inverted. Ten tadpoles were used as controls. Fifty-two supernumerary structures developed, all of them in dorsal or ventral locations on the host stump. The majority (32 out of the 44 outgrowths with more than 3 toes) were normal limbs of stump handedness. However, the following abnormal structures were also observed: 2 double-posterior, 3 mixed-symmetric, and 7 undetermined cases. These results are in agreement with the predictions of a hierarchical polar coordinate model for epimorphic regeneration.  相似文献

The fate of an imaginal disc cell of Drosophila can be affected by the associations and interactions that it has with other cells in the disc. A fragment of an imaginal disc, not regenerating under conditions allowing a complementary fragment to do so, can be stimulated to regenerate by interactions with cells of the complementary fragment [Haynie, J. L., and Bryant, P. J. (1976) Nature (London)259, 659–662]. We report here that one nonregenerating fragment of an imaginal wing disc cannot be stimulated to regenerate by interactions with cells from other parts of the disc. This fragment, containing the anlagen of the distal wing, fails to regenerate proximally when combined with a proximal fragment even though this association stimulates some proximal fragments to regenerate distally. We suggest that this may be a phenomenon similar to that observed in cockroach legs by H. Bohn (1970, Wilhelm Roux Arch. Entwicklungsmech. Organismen165, 303–341), in which proximal regeneration from grafted distal leg segments proceeds only to a limited extent. We consider the possibility that there exist reiterated sets of positional information arranged concentrically in the wing disc.  相似文献

Objective: To review data implicating microbiota influences on Coronavirus Disease 2019 (COVID-19) in patients with diabetes.Methods: Primary literature review included topics: “COVID-19,” “SARS,” “MERS,” “gut micro-biota,” “probiotics,” “immune system,” “ACE2,” and “metformin.”Results: Diabetes was prevalent (~11%) among COVID-19 patients and associated with increased mortality (about 3-fold) compared to patients without diabetes. COVID-19 could be associated with worsening diabetes control and new diabetes diagnosis that could be linked to high expression of angiotensin-converting enzyme 2 (ACE2) receptors (coronavirus point of entry into the host) in the endocrine pancreas. A pre-existing gut microbiota imbalance (dysbiosis) could contribute to COVID-19–related complications in patients with diabetes. The COVID-19 virus was found in fecal samples (~55%), persisted for about 5 weeks, and could be associated with diarrhea, suggesting a role for gut dysbiosis. ACE2 expressed on enterocytes and colonocytes could serve as an alternative route for acquiring COVID-19. Experimental models proposed some probiotics, including Lactobacillus casei, L. plantarum, and L. salivarius, as vectors for delivering or enhancing efficacy of anti-coronavirus vaccines. These Lactobacillus probiotics were also beneficial for diabetes. The potential mechanisms for interconnections between coronavirus, diabetes, and gut microbiota could be related to the immune system, ACE2 pathway, and metformin treatment. There were suggestions but no proof supporting probiotics benefits for COVID-19 infection.Conclusion: The data suggested that the host environment including the gut microbiota could play a role for COVID-19 in patients with diabetes. It is a challenge to the scientific community to investigate the beneficial potential of the gut microbiota for strengthening host defense against coronavirus in patients with diabetes.  相似文献

A synchronous, concerted chemical process is rigorously divided by the reaction force F(R), the negative gradient of V(R), into “reactant” and “product” regions which are dominated by structural changes and an intervening “transition” region which is electronically intensive. The reaction force constant κ(R), the second derivative of V(R), is negative throughout the transition region, not just at the nominal transition state, at which κ(R) has a minimum. This is consistent with experimental evidence that there is a transition region, not simply a specific point. We show graphically that significant nonsynchronicity in the process is associated with the development of a maximum of κ(R) in the transition region, which increases as the process becomes more nonsynchronous. (We speculate that for a nonconcerted process this maximum is actually positive.) Thus, κ(R) can serve as an indicator of the level of nonsynchronicity.

Main conclusion

Three independent patterns of vein formation in Cyperus involucratus Rottb. were identified based on rare spontaneous interruptions of scape vein development. A number of developmental anomalies of vascular bundles in Cyperus involucratus Rottb. were identified and they include “turnabout”, “absent”, “twins”, “doublet”, amphivasal and various stages of “arrested”. These were used to develop a computer program to explain the three vasculature patterns of the scape of (a) ordered deployment of vascular bundles, (b) arrangement of tissues within vascular bundles and (c) orientation of vascular bundles with respect to stem edge. The computer model is a cell-by-cell determination of cell types and facet states.

In this paper I present a hypothetical step on the route to turmorigenesis which may be common to many of the tumorogenic events taking place in vivo. According to this hypothesis portions of DNA from normal nontransformed dying cells may escape degradation, “transfect” other cells and under appropriate conditions also induce transformation. When various high risk factors for carcinogenesis are examined this hypothesis may easily fit into each of them. In addition, recent reports of experimental “transfection” indirectly support this hypothesis. It could now be argued that aging, ionizing radiation, immunosuppressive drugs, genetic errors, defects in DNA repair, immunodeficiency states, chronic infections and chronic inflammatory diseases may all increase the availability of free DNA or the readiness of cells to accommodate “transfection” DNA, or both. Thus the risk of cancer associated with these situations may be explained by increased chance for “transfection” with DNA.It is suggested that the minimal requirements for cell transformation consist of a certain sequence of DNA which does not have to be integrated into the nucleus at once, it may instead accumulate piece by piece so that the first “transfection” of a cell may occur long before transformation takes place. If the “transfecting” DNA sequence does not fulfil the minimal requirements for maintaining a state of repetitive uncontrolled mitosis, then this cell may wait silently or remain a benign tumor until “boosted” with an ultimate complementary DNA sequence to develop into a fully fledged cancer.  相似文献

The reaction force F(ξ) is the negative gradient of the potential energy of a chemical process along the intrinsic reaction coordinate ξ. We extend the rigorous concept of F(ξ) to the “activation strain model” of Bickelhaupt et al., to formulate the “strain” force F str(ξ) that retards a reaction and the “interaction” force F int(ξ) that drives it. These are investigated for a group of Diels-Alder cycloadditions. The results fully support the interpretation of the minimum of F(ξ) as defining the beginning of the transition from deformed reactants to eventual products.  相似文献

Arguments supporting the “bad luck” hypothesis presented by C. Tomasetti and B. Vogelstein ((2015) Science, 347, 78–81) and A. V. Lichtenstein ((2017) Biochemistry (Moscow), 82, 75–80) are critically discussed. Objective: Graft-versus-host disease (GVHD) is an immune phenomenon that occurs in 30 to 70% of patients after allogeneic hematopoietic stem cell transplantation (HST). Chronic GVHD is a state of immune dysregulation wherein, depending on the severity and organ involved, patients may require prolonged treatment with additional or higher corticosteroids and other immunosuppressive agents. The objective of this study was to review the endocrine manifestations following HST that can arise as a consequence of the primary disease or its treatment, including chemotherapeutic agents, corticosteroids, radiation, or GVHD.Methods: We performed a narrative review of GVHD after HST. An English-language search for relevant studies was conducted on PubMed from inception to August 1, 2018, using the following search terms: “endocrine complications,” “bone marrow transplantation,” “graft-versus-host disease,” and “GVHD.” The reference lists of relevant studies were also reviewed.Results: Chronic GVHD may be associated with considerable pediatric growth impairment and may also contribute to thyroid gland dysfunction and thyroid cancer. These patients may also be at increased risk for low bone mineral density, reduced fertility, metabolic syndrome, and suppression of the pituitary-adrenal axis with adrenal insufficiency.Conclusion: This review indicates the importance of monitoring, diagnosing, and properly treating the endocrine complications in this population. Objective: Traumatic brain injury (TBI) is now recognized as a major public health concern in the United States and is associated with substantial morbidity and mortality in both children and adults. Several lines of evidence indicate that TBI-induced hypopituitarism is not infrequent in TBI survivors and may contribute to the burden of illness in this population. The goal of this article is to review the published data and propose an approach for the neuroendocrine evaluation and management of these patients.Methods: To identify pertinent articles, electronic literature searches were conducted using the following keywords: “traumatic brain injury,” “pituitary,” “hypopituitarism,” “growth hormone deficiency,” “hypogonadism,” “hypoadrenalism,” and “hypothyroidism.” Relevant articles were identified and considered for inclusion in the present article.Results: TBI-induced hypopituitarism appears to be more common in patients with severe TBI. However, patients with mild TBI or those with repeated, sports-, or blast-related TBI are also at risk for hypopituitarism. Deficiencies of growth hormone and gonadotropins appear to be most common and have been associated with increased morbidity in this population. A systematic approach is advised in order to establish the presence of pituitary hormone deficiencies and implement appropriate replacement therapies.Conclusion: The presence of traumatic hypopituitarism should be considered during the acute phase as well as during the rehabilitation phase of patients with TBI. All patients with moderate to severe TBI require evaluation of pituitary function.
