Fruit set of plants largely depends on the biosynthesis and crosstalk of phytohormones. To date the role of cytokinins (CKs) in the fruit development is less understood. Here, we showed that parthenocarpic fruit could be induced by 1-(2-chloro-4-pyridyl)-3-phenylurea (CPPU, an active CK) in tomato ( Solanum lycopersicum cv. Micro-Tom). The fresh weight of CPPU-induced parthenocarpic fruits was comparable with that induced by GA3. Importantly, CPPU-induced parthenocarpy was found to be compromised by simultaneous application of paclobutrazol (a GA biosynthesis inhibitor), and this effect could be restored by exogenous GA3. Like pollination, CPPU-induced fruit showed enhanced accumulation of GA1+3 and indole-3-acetic (IAA), which were accompanied by elevated expression of GA biosynthesis genes like SlGPS, SlGA20ox1, SlGA20ox2 and SlGA3ox1, and IAA biosynthesis gene ToFZY. Elevated GAs level in CPPU-induced fruits was also associated with down-regulation of GA inactivation genes, namely SlGA2ox1,2,3,4,5 in comparison with untreated control. These results suggested that CKs may induce parthenocarpy in tomato partially through modulation of GA and IAA metabolisms.  相似文献

Plant D-type cyclin genes (CYCDs) are important regulators of cell division. However, little is known on their participation during the early developmental stage of cucumber fruit. In this study, cucumber CYCD genes were identified and characterized. The expression levels of these genes during early fruit development were assessed from 0 to 8 days after anthesis (DAA). The results revealed the presence of 13 different CYCD genes, which were named according to identity percentages of the corresponding orthologs in Arabidopsis thaliana and poplar. The genomic organization of each subgroup CYCD was similar to their orthologs in A. thaliana and poplar. The expression levels of CsCYCD genes were analyzed in cucumber fruits under different treatments including natural parthenocarpic fruit, pollinated fruit, and N-(2-chloro-4-pyidyl)-N′-phenyurea (CPPU)-induced parthenocarpic fruit. The highest expression levels of most CsCYCDs genes were at four DAA in natural parthenocarpic and pollinated fruits. Interestingly, the expression patterns of 8 of 13 CsCYCD genes in natural parthenocarpic fruit were similar to those in pollinated fruit, but different from those in CPPU-induced parthenocarpic fruit. Collectively, the results of this study provide insights on the CYCDs involved in cucumber parthenocarpic fruit development.  相似文献

The effects of NAA (naphthaleneacetic acid), GA3 (gibberellic acid), CPPU (N-(2-chloro-4-pyridyl)-N'-phenylurea) and pollination on fruit set, cell division and enlargement were studied in Lagenaria leucantha, an important vegetable. NAA and GA3 were ineffective in inducing parthenocarpy, whereas CPPU induced parthenocarpic fruit significantly larger than fruit that resulted from pollination. Cell division, which occurred during the first 4 days after pollination was not reactivated by NAA or GA3, but was effectively reactivated by CPPU. The cell number of the total cross-section of CPPU-treated fruit was 117.4% of that of pollinated fruit and 154.4% of that of unpollinated at 12 DAA (days after anthesis) respectively. The CPPU-induced parthenocarpic fruit had the largest cell cross-sectional area followed, successively, by pollinated fruit, NAA-treated fruit, GA3-treated fruit and unpollinated fruit. These results indicate that CPPU induced parthenocarpic fruit growth by directly reactivating cell division and expansion.  相似文献

The denatured protein profiles of developing tomato ( Lycopersicon esculentum Mill.) fruits, from the anthesis stage up to fruits at 30% of their final diameter, were examined in a pai-2l pat-2 parthenocarpic line and in its near isogenic non-partheno-carpic line. At anthesis no differences were detected between the protein patterns of ovaries developed on parthenocarpic and non-parthenocarpic plants. In subsequent stages the seeded and seedless fruits differed in the pattern of manifestation of several abundant proteins, none of which seem to be included in seeds The most prominent difference was found in an insoluble protein of 62 kDa; in developing seeded fruits of either the parthenocarpic or the non-parthenocarpic line, its rate of decline was much faster than in seedless fruits. In seeded fruits larger than 4-6 mm in diameter it was scarcely detected, whereas in parthenocarpic seedless 8–10 mm fruits it was still abundant. This protein is fruit specific; it is also enhanced in chemically (n-n-tolyl phthalamic acid) – induced parthenocarpic fruits of the non-parthenocarpic line. The prolonged manifestation in the parthenocarpic fruits results from de novo synthesis of this protein. There are indications that it is not a stress-related protein. This is the first demonstration of an association between the pattern of modulation of a protein and the phenotypic expression of genetically controlled parthenocarpy.  相似文献

Fruit size within a tomato (Lycopersicon esculentum Mill.) truss depends on both fruit position in the truss and the time of pollination among fruits. In the natural pollination sequence a difference of 5 days in the pollination of proximal and distal flowers results in significant final size differences between proximal and distal fruits. These final size differences were eliminated when all flowers were pollinated simultaneously. At anthesis proximal ovaries have higher cell numbers than distal ovaries but the cell division activity and cell enlargement in both positions was similar in the first 10 days of fruit growth. Simultaneous pollination resulted in lower cell numbers in proximal but higher cell numbers in distal fruits compared to control fruits.Hormone levels in different sized fruits were measured using radioimmunoassays. Cytokinin concentration during the cell division period indicated a possible role in the regulation of cell division. With other hormones no obvious correlations were found. The results are discussed in relation to factors determining final fruit size in tomato.  相似文献

Although the hormones, gibberellin and auxin, are known to play a role in the initiation of fruits, no such function has yet been demonstrated for abscisic acid (ABA). However, ABA signaling and ABA responses are high in tomato (Solanum lycopersicum L.) ovaries before pollination and decrease thereafter (Vriezen et al. in New Phytol 177:60–76, 2008). As a first step to understanding the role of ABA in ovary development and fruit set in tomato, we analyzed ABA content and the expression of genes involved in its metabolism in relation to pollination. We show that ABA levels are relatively high in mature ovaries and decrease directly after pollination, while an increase in the ABA metabolite dihydrophaseic acid was measured. An important regulator of ABA biosynthesis in tomato is 9-cis-epoxy-carotenoid dioxygenase (LeNCED1), whose mRNA level in ovaries is reduced after pollination. The increased catabolism is likely caused by strong induction of one of four newly identified putative (+)ABA 8′-hydroxylase genes. This gene was named SlCYP707A1 and is expressed specifically in ovules and placenta. Transgenic plants, overexpressing SlCYP707A1, have reduced ABA levels and exhibit ABA-deficient phenotypes suggesting that this gene encodes a functional ABA 8′-hydroxylase. Gibberellin and auxin application have different effects on the LeNCED1 and SlCYP707A1 gene expression. The crosstalk between auxins, gibberellins and ABA during fruit set is discussed.  相似文献

Application of GA3, IAA or 4-CPA to tomato ovaries induced the development of parthenocarpic fruit, which showed different growth rates. In the pericarp cell division and cell enlargement was affected differentially. GA3-induced fruits had considerably less but larger cells than seeded control fruits, IAA treatment resulted in the same number of cells but these were smaller and 4-CPA treatment induced fruits with about 20% more cells. Reduction in cell number had a similar effect on final fruit size as diminution of cell size. A reduction in the number of cell division centres (area around vascular bundles) as well as changes in the degree of endoploidy are possible reasons for the observed reductions in cell numbers. Hormonal causes for the different number and size of pericarp cells after the various treatments are discussed.  相似文献

Fruit set and fruit development in tomato is largely affected by changes in environmental conditions, therefore autonomous fruit set independent of fertilization is a highly desirable trait in tomato. Here, we report the production and characterization of male‐sterile transgenic plants that produce parthenocarpic fruits in two tomato cultivars (Micro‐Tom and Moneymaker). We generated male‐sterility using the cytotoxic gene barnase targeted to the anthers with the PsEND1 anther‐specific promoter. The ovaries of these plants grew in the absence of fertilization producing seedless, parthenocarpic fruits. Early anther ablation is essential to trigger the developing of the transgenic ovaries into fruits, in the absence of the signals usually generated during pollination and fertilization. Ovaries are fully functional and can be manually pollinated to obtain seeds. The transgenic plants obtained in the commercial cultivar Moneymaker show that the parthenocarpic development of the fruit does not have negative consequences in fruit quality. Throughout metabolomic analyses of the tomato fruits, we have identified two elite lines which showed increased levels of several health promoting metabolites and volatile compounds. Thus, early anther ablation can be considered a useful tool to promote fruit set and to obtain seedless and good quality fruits in tomato plants. These plants are also useful parental lines to be used in hybrid breeding approaches.  相似文献

The changes taking place during fruit development in the concentration of the 3 polyamine fractions, i.e. free, perchloric acid-soluble conjugates and perchloric acid-insoluble bound polyamines, were analyzed in tomato fruits ( Lycopersicon esculentum Mill, cv. F121) induced to set by either pollination or auxin application. Before the onset of cell division, total polyamines were 50% higher in auxin-treated fruits than in pollinated ones, most of the polyamines being found as perchloric acid-soluble conjugates in both fruit set treatments. At the onset the level of polyamines in both fruit types was 100 times higher than during cell expansion or ripening. The highest polyamine found during cell division was perchloric acid-soluble conjugated spermidine in both fruit set treatments. After cell division, polyamine concentration was similar in both fruit set treatments. The concentration of polyamines in the jelly was similar in pollinated and auxin-induced fruits during cell expansion but not during ripening. At the onset of ripening there was an increase of one order of magnitude in the concentration of perchloric acid-insoluble bound putrescine in the jelly of pollinated fruits. Polyamines were more than 5-fold higher in unpollinated ovaries than in fruits induced to set by either pollination or auxins. It is suggested that pollinated and parthenocarpic fruits differ in their polyamine metabolism during the initial stages of development, but not after cell division. It is also suggested that polyamines undergo rapid turnover during cell division. Perchloric acid-insoluble bound putrescine might play a role in seed formation in tomatoes.  相似文献

Parthenocarpy and fruit malformations are common among independent transgenic tomato lines, expressing genes encoding different pathogenesis-related (PR) protein and antimicrobal peptides. Abnormal phenotype developed independently of the expression and type of target genes, but distinctive features during flower and fruit development were detected in each transgenic line. We analyzed the morphology, anatomy, and cytoembryology of abnormal flowers and fruits from these transgenic tomato lines and compared them with flowers and fruits of wild tomatoes, line YaLF used for transformation, and transgenic plants with normal phenotype. We confirmed that the main cause of abnormal flower and fruit development was the alterations of determinate growth of generative meristem. These alterations triggered different types of anomalous growth, affecting the number of growing ectopic shoots and formation of new flowers. Investigation of the ovule ontogenesis did not show anomalies in embryo sac development, but fertilization did not occur and embryo sac degenerated. Nevertheless, the ovule continued to differentiate due to proliferation of endothelium cells. The latter substituted embryo sac and formed pseudoembryonic tissue. This process imitated embryogenesis and stimulated ovary growth, leading to the development of parthenocarpic fruit. We demonstrated that failed fertilization occurred due to defective male gametophyte formation, which was manifested in blocked division of the nucleus in the microspore and arrest of vegetative and generative cell formation. Maturing pollen grains were overgrown microspores, not competent for fertilization but capable to induce proliferation of endothelium and development of parthenocarpic ovary. Thus, our study provided new data on the structural transformations of reproductive organs during development of parthenocarpic fruits in transgenic tomato.  相似文献

The actions of plant regulators in set and development of fruitsare well known. However, the presence and function of endogenoushormones in parthenocarpic fruits have still not been sufficientlyinvestigated. A comparison between seeded and seedless fruitsmakes it possible to obtain a more accurate understanding ofsome relationships between growth regulators and stages of fruitdevelopment. Endogenous auxin and gibberellin activity levelsand some growth parameters (fresh and dry weight, cell numberand cell volume, DNA content) have been determined in tomatofruits (Lycopersicon esculentum Mill.) of the cultivar Venturaand of its isogenic parthenocarpic mutant. In both genotypes,auxin and gibberellin are present in the first week after anthesis,though at different concentrations and with different patterns.These two activities are involved in fruit setting. The simultaneousoccurrence of maximum auxin concentration and of the beginningof cell enlargement, in both genotypes, shows that the activitypresent at this time starts fruit development and possibly determinesthe size of the fruits. High auxin activity is observed only in seeded fruits 20–40days after anthesis, and it is probably synthesized by seeds.Gibberellin activity is present, corresponding to the changein fruit development from the mature green to pink stages. We investigated the role of gibberellins (GAs) in the effect of pat-2, a recessive mutation that induces facultative parthenocarpic fruit development in tomato (Lycopersicon esculentum Mill.) using near-isogenic lines with two different genetic backgrounds. Unpollinated wild-type Madrigal (MA/wt) and Cuarenteno (CU/wt) ovaries degenerated, but GA(3) application induced parthenocarpic fruit growth. On the contrary, parthenocarpic growth of MA/pat-2 and CU/pat-2 fruits, which occurs in the absence of pollination and hormone application, was not affected by GA(3). Pollinated MA/wt and parthenocarpic MA/pat-2 ovary development was negated by paclobutrazol, and this inhibitory effect was counteracted by GA(3). The main GAs of the early-13-hydroxylation pathway (GA(1), GA(3), GA(8), GA(19), GA(20), GA(29), GA(44), GA(53), and, tentatively, GA(81)) and two GAs of the non-13-hydroxylation pathway (GA(9) and GA(34)) were identified in MA/wt ovaries by gas chromatography-selected ion monitoring. GAs were quantified in unpollinated ovaries at flower bud, pre-anthesis, and anthesis. In unpollinated MA/pat-2 and CU/pat-2 ovaries, the GA(20) content was much higher (up to 160 times higher) and the GA(19) content was lower than in the corresponding non-parthenocarpic ovaries. The application of an inhibitor of 2-oxoglutarate-dependent dioxygenases suggested that GA(20) is not active per se. The pat-2 mutation may increase GA 20-oxidase activity in unpollinated ovaries, leading to a higher synthesis of GA(20), the precursor of an active GA.  相似文献
