Methotrexate and folic acid analogs are polar molecules and attempts to obtain electron impact and chemical ionization mass spectra of these compounds by several laboratories have failed. We have found that methylation of this important class of compounds with diazomethane produces derivatives which are sufficiently volatile to allow their mass spectra to be recorded. The mass spectra of the methyl derivatives of five compounds, 4-amino-4-deoxy-N10-methylpteroylglutamic acid, 4-amimo-4-deoxypteroylglutamic acid, pteroylglutamic acid, 4-amino-4-deoxypteroic acid and N10-methylpteroylglutamic acid are presented, and the fragmentation pathways of these compounds discussed.  相似文献

Techniques for coupling liquid chromatography with mass spectrometry are reviewed and an interface is described for the analysis of lipids by mass spectrometry. The interface for coupling liquid chromatography with mass spectrometry for lipid analysis is based on the moving wire transport principle using an endless stainless steel belt of novel construction. After evaporation of the solvent, the solute remains as a residue on the belt which transports it into a reactor where it is volatilized by evaporation or conversion to hydrocarbons. The volatile compounds are then fed into the source of a chemical ionization mass spectrometer for mass analysis by total or single ion monitoring as well as for structural identification or compositional analysis. The sensitivity of the system was approx. 1 mg per component separated in the eluate of a high efficiency column. The capabilities of the interface were demonstrated by its application to reference compounds representative of triglycerides, sterols, steryl esters, glyceryl ethers, glyceryl ether diesters, glycerophosphatides, sphingolipids, prostaglandins and fatty acid methyl esters. It also was applied to the analysis of methyl ester ozonides to demonstrate the use of LC-MS for the localization of double bonds in unsaturated fatty acids.  相似文献

Determination of volatile fatty acids in plasma after ethanolic extraction   总被引:5,自引:1,他引:5  
1. A new rapid micro-method for measuring plasma volatile fatty acids is described. The volatile fatty acids are extracted from plasma with ethanol in the presence of a known quantity of internal standard (sodium isobutyrate). After evaporation of the ethanolic solution of the sodium salts, the residue is dissolved in a dilute solution of orthophosphoric acid to permit analysis by g.l.c. 2. A technique of g.l.c. analysis is described which permits the separation of all the volatile fatty acids from the other plasma constituents at temperatures below 100 degrees C in 5 min. 3. Steam-distillation techniques are unsatisfactory when the acetic acid concentrations in the plasma are below 0.2mm. Heating of a number of plasma constituents in acid conditions gives rise to acetic acid. 4. The binding of volatile fatty acids to plasma proteins was studied; this binding is negligible for acetic acid, but increases with the length of the fatty acid carbon chain. 5. The limits of use of the method and the physiological implications are discussed.  相似文献

This paper describes a gas recirculation system for the exhaust gases from the aerobic fermenters normally used in acetic fermentation processes. With the application of this procedure, it is possible to operate in a closed system, so preventing the large losses of fermentation yield due to evaporation which occur in open systems. In addition, this system reduces losses of volatile organoleptic compounds (ethanol, acetic acid and ethyl acetate, among others) so enabling the product to be incorporated into processes for the manufacture of high quality vinegars.  相似文献

麻黄(Ephedrae Herba)具有发汗散寒、宣肺平喘、利水消肿的功效。现代研究表明麻黄对治疗支气管、哮喘有疗效。藏麻黄(Ephedra saxatilis)为西藏特有的麻黄属植物,其根、茎具有特殊香气,且目前其挥发性成分未有报道。为评价挥发性成分对支气管炎作用,用气质联用仪测定分析藏麻黄根、茎挥发性成分,并结合网络药理学和分子对接方法分析挥发性成分治疗支气管炎的关键靶点。分析检测到67个挥发性化合物,茎中含有挥发性主要成分为2,3,5,6-四甲基吡嗪(10.92%)、Z-9-十五烯醇(9.15%)、二羟基苯乙酮(7.92%);根含有的挥发性主要成分为亚油酸(7.81%)、红没药醇(7.1%)、Z-9-十五烯醇(5.98%)。有34个挥发性成分能够作用32个支气管炎疾病靶点。蛋白互作网络分析预测IL6、TNF、PTGS2、CXCL8为藏麻黄挥发性成分治疗支气管可能的潜在靶点。这些靶点与挥发性成分丁子香酚、月桂酸、反式法尼醇、α-萜品醇、榄香醇和棕榈酸6种化合物靶点相互结合程度高,且分子对接效果好。本研究推测藏麻黄挥发性成分对支气管炎的治疗作用机制可能是通过抑菌作用参与炎症反应,并通过抑制TNF、IL-17、Toll样受体等炎症与免疫信号通路中IL6、TNF、PTGS2、CXCL8的表达,从而起到治疗支气管炎的作用。研究结果为挖掘藏麻黄挥发性成分中治疗支气管炎靶标分子及其产品开发提供重要参考依据。  相似文献

Many mosses of the family Splachnaceae are entomophilous and rely on flies for spore dispersal. Splachnum ampullaceum produces a yellow‐ or pink‐coloured hypophysis that releases volatile compounds, attracting flies to the mature moss. The biosynthetic sources of the visual and aromatic cues within the hypophysis have not been identified, and may be either symbiotic cyanobacteria or chromoplasts that break down lipids into volatile compounds. Here, we used transmission electron microscopy and gas chromatography‐mass spectrometry (GC‐MS) to investigate the sources of these attractants, focusing on different tissues and stages of maturation. Microscopy revealed an abundance of plastids within the hypophysis, while no symbiotic bacteria were observed. During plant maturation, plastids differentiated from amyloplasts with large starch granules to photosynthetic chloroplasts and finally to chromoplasts with lipid accumulations. We used GC‐MS to identify over 50 volatile organic compounds from mature sporophytes including short‐chain oxygenated compounds, unsaturated irregular terpenoids, fatty acid‐derived 6‐ and 8‐carbon alcohols and ketones, and the aromatic compounds acetophenone and p‐cresol. The hypophysis showed localised production of pungent volatiles, mainly short‐chain fermentation compounds and p‐cresol. Some of these volatiles have been shown to be produced from lipid oxidase degradation of linolenic acid within chromoplasts. However, other compounds (such as cyclohexanecarboxylic acid esters) may have a microbial origin. Further investigation is necessary to identify the origin of fly attractants in these mosses.  相似文献

At least two different signalling pathways have been identified that result in clearly distinguishable volatile profiles in response to pathogens and herbivores in the lima bean Phaseolus lunatus. Alamethicin, a voltage-gated ion-channel-forming peptide from Trichoderma viride, is a potent inducer of volatile biosynthesis in the lima bean. Unlike elicitation with cellulysin or herbivore damage, which act through the jasmonic acid pathway and result in a complex pattern of volatile compounds, the emitted blend comprises only the two homoterpens, 4,11-dimethylnona-1,3,7-triene and 4,8,12-trimethyltrideca-1,3,7,11-tetraene, and methyl salicylate. Both pathways, represented by jasmonic acid and alamethicin, depend on lipid-derived signalling compounds, set off by the activation of a phospholipase A and further processing by lipoxygenase activity. The alamethicin-induced signal-transduction pathway interferes with the octadecanoid cascade, probably due to increased salicylic acid levels, resulting in an inhibition of the typical jasmonic acid-induced volatile profile.  相似文献

The clearwing moth, Paranthrene diaphana Dalla Torre & Strand (Lep.: Sesiidae) is one of the most destructive pests of willow trees in Tehran province which leads to dieback. In this study, female volatile compounds were identified. For this purpose, female volatile substances were extracted using Solid Phase Micro Extraction and then were identified using Head Space Chromatography method. Female insects were assessed separately at ages of 2–4, 4–6 and 6–8 days of preparation. Among the various compounds identified by GC/MS in the volatile compounds of female insect, 27 compounds were introduced while seven combinations have also been produced and released at all ages of insect which include: Pentadecane, Heptadecane, Dodecane, Tetradecane, Hexadecane, Eicosane and Tridecane. The peak area of curve (log scale) and frequency of these compounds, demonstrate that five combinations including: Tetradecane, Hexadecane, Tridecane, Eicosane and Dodecane are more important. In addition, five compounds including: Octadecane, Palmitic acid (Hexadecanoic acid), 9-Octadecenoic acid, Tricosane and Hexacosane have been released only from fourth to the eighth day of adult life. Therefore, it seems that female sex pheromone is a combination of these 12 compounds.  相似文献

BACKGROUND AND AIMS: Among the subspecies of Petunia axillaris are various lines emitting sensorially different scents. Analysis of variations in floral scent among genetically close individuals is a powerful approach to understanding the mechanisms for generating scent diversity. METHODS: Emitted and endogenous components were analysed independently to gain information about evaporation and endogenous production in 13 wild lines of P. axillaris. A dynamic headspace method was used to collect emitted components. Endogenous components were extracted with solvent. Both of these sample types were subjected to quantitative and qualitative analysis by gas chromatography (GC)-flame ionization detector (FID) and GC-mass spectrometry (MS). KEY RESULTS AND CONCLUSIONS: Whereas the profiles of emitted compounds showed qualitative homogeneity, being mainly composed of methyl benzoate with quantitative variation, the profiles of endogenous compounds showed both qualitative and quantitative variation. A negative correlation was found between the evaporation ratio and boiling point of each compound examined. Lower boiling point compounds were strongly represented in the emitted component, resulting in the reduction of qualitative variation in floral scent. In conclusion, floral scent diversity results from variation in both the endogenous production and the evaporation rate of the individual volatile compounds.  相似文献

The chemical composition of the extracted fixed oil (total fatty acid composition) and volatile oil of Nigella sativa L. seeds grown in Iran were determined by GC and GC/MS. Eight fatty acids (99.5%) and thirty-two compounds (86.7%) have been identified in the fixed and volatile oils, respectively. The main fatty acids of the fixed oil were linoleic acid (55.6%), oleic acid (23.4%), and palmitic acid (12.5%). The major compounds of the volatile oil were trans-anethole (38.3%), p-cymene (14.8%), limonene (4.3%), and carvone (4.0%).  相似文献

从贵州省宽阔水自然保护区采集滇黔金腰、大叶金腰和锈毛金腰,采用水蒸汽蒸馏法提取挥发油,用气相色谱—质谱联用技术对挥发油化学成分进行分析,共鉴定出85种成分,主要为十六烷酸、烷烃类、肉豆蔻酸和叶绿醇等化合物。其中,在滇黔金腰中共鉴定出48种化学成分,主要有十六烷酸(10.29%)和月桂酸(7.54%)等;在大叶金腰中共鉴定出50种化学成分,主要有邻苯二甲酸二(2-乙基己)酯(10.91%)和二十七烷(7.29%)等;在锈毛金腰中共鉴定出58种化学成分,主要有十六烷酸(12.66%)和三十二烷(8.15%)等。不同种金腰属植物挥发油的主要成分种类比较接近,但各自挥发油化合物组成又都含有其特有化学成分。  相似文献

哺乳动物的包皮腺分泌物对个体间性引诱及繁殖行为的信息交流和种群调节有着重要的作用。本研究以四川短尾鼩(Anourosorex squamipes)为研究对象对劳亚食虫目动物包皮腺挥发性化学成分和化学通讯功能进行报道。采用顶空固相微萃取—气质联用(HS-SPME-GC/MS)的方法,分析四川短尾鼩雄体包皮腺中挥发性物质的化学组成。结果表明:(1)四川短尾鼩雄体包皮腺的挥发性化学成分主要含烷烃类、醇类、酮类、醛类、醚类、酯类、酸类、芳香烃类等45种化合物;(2)成年雄性四川短尾鼩的包皮腺中挥发性成分有39种,幼年雄性含有28种,说明四川短尾鼩成年雄体的包皮腺中挥发性化学成分多于幼年雄体;(3)成年雄体的包皮腺分泌物中含有4种特有的挥发性化学成分,幼年雄体中特有成分仅为1种,表明四川短尾鼩包皮腺中的挥发性化学成分存在年龄差异;(4)四川短尾鼩包皮腺分泌物中不同化学成分相对含量不同,同种化学成分在不同个体间的相对含量存在差异。四川短尾鼩雄性包皮腺挥发性化合物种类丰富,个体、年龄差异明显。推测挥发性化合物中丁酸(Butanoicacid)、乙酸乙酯(Ethyl acetate)、苯酚(Phenol)为四川短尾鼩的信息素。本研究为进一步验证该物种的信息素成分及其传递机制提供了基础数据。  相似文献

Rosemary is commonly used as a spice and a flavoring agent in food processing. Although the antioxidative properties of its extracts have been investigated, there have been few reports on the volatile components of rosemary. We designed a novel antioxidative system which can generate the volatile constituents in the gaseous phase from a rosemary extract and evaluated the gaseous antioxidative activities against both lipid peroxidation and cell death induced by nitrogen dioxide and ultraviolet radiation. The antioxidative effects of the major volatile components on the oxidation of linoleic acid induced by azo compounds were also investigated in a solution. The volatile components in the novel antioxidative system suppressed the Jurkat cell death induced by nitrogen dioxide and the intracellular formation of reactive oxygen species in fibroblast cells induced by ultraviolet radiation. 1,8-Cineole among the volatile components exerted an antioxidative effect against the oxidation of linoleic acid in a solution induced by azo compounds and ultraviolet radiation. These data suggest that the volatile constituents of a rosemary extract had antioxidative properties and that gaseous exposure antioxidant is a promising method for promoting health.  相似文献

Plants synthesize volatile compounds to attract pollinators. The volatiles emitted by flowers are often complex mixtures of organic compounds; pollinators are capable of distinctly recognizing different volatile compounds. Plants also produce volatile compounds to protect themselves against herbivores and pathogens. Some of the volatile compounds produced in floral and vegetative tissues are toxic to insects and microbes. To adapt changes in the environment, plants have evolved the ability to synthesize a unique set of volatiles. Intensive studies have identified and characterized the enzymes responsible for the formation of plant volatiles. In particular, many biosynthetic genes have been isolated and their enzymatic functions have been proposed. This review describes how plants have evolved the biosynthetic pathways leading to the formation of green leaf volatiles and phenylpropene volatiles.  相似文献

Oidium sp. has been recovered as an endophyte in Terminalia catappa (tropical chestnut) in Costa Rica. The volatile organic compounds (VOCs) of this organism uniquely and primarily consist of esters of propanoic acid, 2-methyl-, butanoic acid, 2-methyl-, and butanoic acid, 3-methyl-. The VOCs of Oidium sp. are slightly inhibitory to many plant pathogenic fungi. Previous work on the VOCs of Muscodor albus demonstrated that besides esters of small organic acids, a small organic acid and a naphthalene derivative were needed to obtain maximum antibiotic activity. Thus, the addition of exogenous volatile compounds such as isobutyric acid and naphthalene, 1,1'-oxybis caused a dramatic synergistic increase in the antibiotic activity of the VOCs of Oidium sp. against Pythium ultimum. In fact, at elevated concentrations, there was not only 100% inhibition of P. ultimum but killing as well. In addition, a coculture of Muscodor vitigenus (making only naphthalene) and Oidium sp. plus isobutyric acid produced an additive antibiosis effect against P. ultimum. The biological implications of multiple volatile compounds acting to bring about antibiosis in nature are discussed.  相似文献

The volatile compounds from cooked potato irradiated with the doses of about 10,000 and 100,000 rad were determined quantitatively immediately after irradiation and after the storage for fifty days following irradiation. In both cases, no significant differences were observed between the volatile compounds from 10,000 rad irradiated and non-irradiated potato. Irradiation of 100,000 rad resulted in increase of volatile compounds, especially that of carbonyl compounds.

The effects of gamma irradiation and storage on ascorbic acid content of potato were also studied. Ascorbic acid content of raw potato decreased approximately 10 % and 30 % than that of control by irradiation of 10,000 and 100,000 rad, respectively.  相似文献

Seleniferous water continues to be a serious problem to wildlife in the central valley of California. Water samples collected from Kesterson Reservoir, Peck Ranch, and Lost Hills evaporation pond facilities contained between 0.005 and 5 mg of Se per liter. The objective of this study was to isolate Se-methylating organisms in evaporation pond water and to assess, through enrichment and manipulation of their optimal growth parameters, the environmental factors which govern microbial Se methylation. Alternaria alternata was isolated as an active Se-methylating organism. The volatile product was identified as dimethylselenide. The effects of pH, temperature, Se substrates, and methyl donors on the ability of A. alternata to methylate Se were investigated in liquid medium containing 100 mg of Se per liter. The optimum pH and temperature for methylation were 6.5 and 30 degrees C, respectively. Selenate and selenite were methylated more rapidly than selenium sulfide and various organic Se compounds (6-selenoguanosine, 6-selenoinosine, seleno-dl-methionine, and 6-selenopurine). l-Methionine and methyl cobalamine (0.1 muM) stimulated dimethylselenide production. This study demonstrates that Se-methylating organisms are present in evaporation pond water and are capable of liberating substantial quantities of Se in the volatile dimethylselenide form. By determining the optimum environmental conditions which stimulate volatilization, it may be possible to design a way to remove Se from seleniferous water in situ.  相似文献

A device for mixing air with a pollutant (e.g., volatile organic compounds) is presented, which is based on principles of evaporation and diffusion. A wide-necked flask containing an aluminium cylindrical body, allows the evaporating area to be preset to 5.75 cm2 or 23.03 cm2. The distance from the evaporating surface to the region of passage of the air is adjustable between 0 and 8 cm, which allows to assay a wide range of evaporation flows (for toluene: 1.5 to 200 mg/minute).  相似文献

The water-soluble products of the UV-initiated autoxidation of linoleic and linolenic acids emulsified in water were separated into volatile and relatively involatile components, each of which reacted with both thiobarbituric acid (TBA) and peroxidase. The volatile TBA-reactive compound is probably malonaldehyde and the volatile peroxidase-reactive compound is hydrogen peroxide. Additional compounds which absorb UV light were present in the volatile fraction. After thin-layer chromatography of the involatile fraction, reactivity toward TBA and peroxidase was found in the same spot. Approximate molar yields of hydrogen peroxide, malonaldehyde, "hydroperoxides", and other TBA-reactive compounds were estimated. The ratio of "hydroperoxide" to TBA reactivity was lower for linoleic than for linolenic acid. The mass of relatively involatile compounds was about 20 times greater than that predicted from either peroxidase or TBA assays of water extracts of oxidized linolenic acid. The properties of the water extract were similar to those shown by others for the products of prolonged autoxidation (without UV-irradiation) of emulsified methyl linoleate.  相似文献
