Digestive enzyme distribution and activity in the digestive tracts of the rabbitfish, Siganus canaliculatus and the sea bass, Lates calcarifer were studied. Quantitative determinations of digestive enzymes in the guts of both fishes showed that they were capable of digesting carbohydrates and proteins in their diet. The carbohydrases, amylase, laminarinase, maltase, sucrase and trehalase were detected in the rabbitfish; their activities being mainly in the stomach, intestine and pyloriccaeca. Amylase, maltase, trehalase and chitinase activities were recorded in the gut of the sea bass, primarily in the intestine and the pyloriccaeca. Their activities were significantly lower than those in the rabbitfish. Proteases (pepsin, chymotrypsin, elastase, leucine aminopeptidase and trypsin) were found in both the rabbitfish and the sea bass. Pepsin activity however, was higher in the sea bass; while trypsin and chymotrypsin activities were higher in the rabbitfish. The activities of the various digestive enzymes in both fishes are discussed in relation to their feeding habits.  相似文献

This study investigated the basis of pair formation in the abundant herbivorous rabbitfish Siganus doliatus on Orpheus Island, Great Barrier Reef. Pair formation was the most common social system in S. doliatus, with 67·4% of all individuals occurring in pairs. Pairs were stable (i.e. individuals remained with the same partner throughout the study) and pair members were found within 5 m of each other 82·9% of the time. Of the examined pairs, 25% were homosexual resulting in a proportion of heterosexual pairs (75%) that was significantly lower than expected if pairs were formed solely for reproductive reasons. Therefore, although reproduction appears to be the main driver of pair formation in S. doliatus, other factors are likely to influence this behaviour. The high density of individuals on the reef crest (5·7 ± 0·9 individuals 200 m?2; mean ± s.e .) and extensively overlapping home ranges of pairs indicated that the defence of territories plays no role in pair formation. Instead, it appears that pair formation in S. doliatus is driven, in part, by other, non‐reproductive, ecological factors. It is suggested that pair formation allows for increased vigilance against predation and enables S. doliatus to execute a novel feeding behaviour.  相似文献

The siganid production in the Philippines is continuously declining from 2007 up to present. In Palompon, Leyte, annual yield of siganids, particularly Siganus canaliculatus (Park, 1797) also showed a decreasing trend of total production. This is consistent that this stock is under heavy pressure, and when left unmanaged, this could lead to further overexploitation and the collapse of the stock in the long run. Regardless of this, detailed reproductive biology and potential of this species are very few in the Philippines. Therefore, this study was conducted to provide information on the reproductive biology and breeding cycle of S. canaliculatus in Palompon, Leyte where fishing pressure is high and sound management formulation is necessary. S. canaliculatus is a group‐synchronous, multiple spawners with skewed sex ratio where males predominates over females. Maturity stages based on macroscopic examinations identified four maturity stages (immature, developing, mature and spent) while six maturity stages (immature, developing, mature, spawning, spent and re‐developing) were identified through histological analysis. A single ovo‐testis was identified among the 669 specimens examined. Based on histological sections, the smallest size of mature male was 5.5 cm standard length, and the smallest mature female was 7.1 cm standard length. On the other hand, length at first maturity for mature fish were 8.1 cm standard length for males and 9.7 cm standard length for females. Two defined peaks of gonadosomatic index were identified in both sexes: a major peak from February to May and a minor protracted one from July to December. Mature and older ovaries and testes occurred throughout the year, an indication of your round breeding season of S. canaliculatus in Palompon. Implementing a fishing ban during the major and the minor breeding spawning peaks would allow the mature and older individuals to contribute first to the population; allowing the continuity of the stock.  相似文献

采用Winkler法研究了温度、盐度、pH和麻醉剂对长鳍篮子鱼幼鱼(体长(3.325±0.205) cm,体质量(0.706±0.155) g)耗氧率的影响。结果表明:长鳍篮子鱼幼鱼耗氧率随着温度升高而增加,其回归关系可用对数函数Y=1.3819lnX-3.55(r=0.9475,P<0.01)表示,其生长的适宜温度为24 ℃~32 ℃;幼鱼耗氧率随着盐度的降低而逐渐增加,其回归关系可用线性函数Y=-0.1802X+1.7551(r=0.9837,P<0.01)来表示;在不同的pH下耗氧率有显著性差异(P<0.01),pH值在6.0时耗氧率最低,9.0时耗氧率最高;耗氧率随着丁香酚、MS-222的浓度升高而逐渐降低,各实验组的耗氧率要显著低于对照组,丁香酚和MS-222可以作为长鳍篮子鱼幼鱼运输过程中的麻醉剂。  相似文献

In this experiment, golden rabbitfish (Siganus guttatus) were allocated between three treatment groups. The fish were injected with saline, human chorionic gonadotropin (hCG) and D-Ala6, Pro9-Net-mGnRH. After injection, in 6 hr intervals, blood plasma samples were collected for steroid hormone (testosterone [T] in males and estradiol-17β [E2] in females) using enzyme immune assay (EIA). In male fish, T levels significantly increased and reached 170 and 650 pg/ml for hCG and D-Ala6, Pro9-Net-mGnRH treatments, respectively. Then T levels slightly decreased until 24 hr post injection. There were no significant changes of T levels in saline treatment. In female fish, we found significant changes in E2 levels at 2,567 and 524 pg/ml at 12 hr post injection in hCG and D-Ala6, Pro9-Net-mGnRH treatments, respectively. No significant differences of E2 levels were observed in saline group. In the second experiment, we injected 100 golden rabbitfish with both hCG and D-Ala6, Pro9-Net-mGnRH. Fish spawned successfully when hCG and D-Ala6, Pro9-Net-mGnRH were given individually and in combination. Latency periods were between 46–64 hr with an average fertilization rate of 70%–90% and hatching rate of 56%–74%. The embryonic duration was 16–20 hr. The saline-injected group produced no spawning. Our findings contribute to further understanding of exogenous hormones impact on golden rabbitfish reproductive endocrinology, refining breeding protocol and implications for fish propagation.  相似文献

Herbivores are widely acknowledged as key elements maintaining the health and resilience of terrestrial and aquatic ecosystems. Understanding and quantifying the impact of herbivores in ecosystems are fundamental to our ability to manage these systems. The traditional method of quantifying the impact of herbivorous fishes on coral reefs has been to use transplanted pieces of seagrass or algae as “bioassays”. However, these experiments leave a key question unanswered, namely: Which species are responsible for the impact being quantified? This study revisits the use of bioassays and tested the assumption that the visual abundance of species reflects their role in the removal of assay material. Using remote video cameras to film removal of assay material on an inner-shelf reef of the Great Barrier Reef, the species responsible for assay-based herbivory were identified. The video footage revealed that Siganus canaliculatus, a species not previously recorded at the study site, was primarily responsible for removal of macroalgal biomass. The average percentage decrease in thallus length of whole plants of Sargassum at the reef crest was 54 ± 8.9% (mean ± SE), and 50.4 ± 9.8% for individually presented Sargassum strands (for a 4.5-h deployment). Of the 14,656 bites taken from Sargassum plants and strands across all reef zones, nearly half (6,784 bites or 46%) were taken by S. canaliculatus, with the majority of the remainder attributable to Siganus doliatus. However, multiple regression analysis demonstrated that only the bites of S. canaliculatus were removing macroalgal biomass. The results indicate that, even with detailed observations, the species of herbivore that may be responsible for maintaining benthic community structure can go unnoticed. Some of our fundamental ideas of the relative importance of individual species in ecosystem processes may be in need of re-evaluation.  相似文献

We determined the complete nucleotide sequence of the mitochondrial genome for the rabbitfish Siganus fuscescens (Perciformes, Siganidae). This mitochondrial genome, consisting of 16,491 base pairs (bp), included 13 protein-coding genes, 2 ribosomal RNAs, 22 transfer RNAs, and a noncoding control region similar those found in other vertebrates; the gene order was identical to that of typical vertebrates. Most of the genes of S. fuscescens were encoded on the H-strand, while the ND6 and eight tRNA (Gln, Ala, Asn, Cys, Tyr, Ser [UCN], Glu, and Pro) genes were encoded on the L-strand. The reading frames of ATPase 8 and 6 and those of ND4L and ND4 overlapped by ten and seven nucleotides, respectively. All mitochondrial protein-coding genes began with an ATG start codon, except for CO1, which started with GTG. Open reading frames of S. fuscescens ended with TAA (ND1, CO1, ATPase 8, ND4L, ND5 and ND6), and the remainder had incomplete stop codons, either TA (ATPase 6 and CO3) or T (ND2, CO2, ND3, ND4, and Cytb). The origin of L-strand replication in S. fuscescens was located in a cluster of five tRNA genes (WANCY) and was 34 nucleotides in length. A major noncoding region between the tRNA-Pro and tRNA-Phe genes (828 bp) was considered to be the control region (D-loop). Within this sequence, we identified a conserved sequence block characteristic of this region. The rabbitfish was grouped with Siganus canaliculatus in most parsimony analyses, which showed 100% bootstrap support for their divergence. These findings are useful for inferring phylogenetic relationships and identification within the suborder Acanthuroidei.  相似文献

The annual reproductive and the lunar-related spawning cycles of the pencil-streaked rabbitfish Siganus doliatus collected from Chuuk Lagoon, Federated States of Micronesia, were examined histologically. Monthly collection of this species revealed that ovaries with developing oocytes containing yolk were observed from February to May and in August and September. Weekly collection of the fish showed that ovarian development and spawning after the first quarter moon were repeated during the reproductive season, suggesting that S. doliatus is a restricted lunar-synchronized spawner and releases gametes around the time of the first quarter moon.  相似文献

The synthesis and secretion of many hormones such as growth hormone (GH), melatonin, and corticosterone, exhibit temporal variations over each day and night. Oral administration of several nutritional factors, including l-ornithine, modulates these hormonal secretions and induces an acute increase in plasma GH levels. However, the impact of l-ornithine on the diurnal rhythms of hormone secretion remains unclear. In this study, we evaluated whether the diurnal rhythms of plasma GH, melatonin, and corticosterone secretion were altered by the daily administration of l-ornithine as well as the timing of the administration, in CBA/N mice. Our results showed that the plasma GH levels that peaked at light phase were amplified by l-ornithine (500?mg/kg) administered at Zeitgeber time (ZT) 22, but not at ZT10. Additionally, l-ornithine (1000?mg/kg) administered at ZT22 advanced the onset of the nocturnal rise of melatonin, which resulted in the elongation of the melatonin peak. On the other hand, l-ornithine (500 and 1000?mg/kg) administered at ZT10, but not at ZT22, suppressed the diurnal rhythm peaks of plasma corticosterone. The effects of l-ornithine on plasma GH rhythms lasted for at least 2 days after cessation of the daily administration. Running wheel activity during the active phase was slightly elevated by l-ornithine administration at ZT22, but the overall patterns were only slightly affected. l-Ornithine levels in the plasma and hypophysis after a single administration of l-ornithine at ZT22 were lower than those after administration at ZT10, suggesting that the metabolic rate of l-ornithine differs between day and night. In conclusion, our data suggest that a daily administration of l-ornithine regulates the diurnal rhythms of GH, melatonin, and corticosterone in a manner dependent on administration time, which might be related to the diurnal rhythms of l-ornithine metabolism.  相似文献

Characteristics of the lunar reproductive cycle in the golden rabbitfish, Siganus guttatus, were determined by histological observations of ovarian development, and immunological measurements of plasma steroid hormones, estradiol-17beta (E2), testosterone (T), 17alpha,20beta-dihydroxy-4-pregnen-3-one (DHP) and 17alpha,20beta,21-trihydroxy-4-pregnen-3-one (20beta-S), and vitellogenin (VTG). Ovarian and plasma samples were collected every week according to the lunar phases from May to July. Weekly change of gonadosomatic index (GSI) showed two peaks at the first lunar quarter in June and July. Yolky oocytes were also observed around this time. Histological observations revealed that the vitellogenic oocytes appeared again 1 week after spawning and developed synchronously. These results suggest that this species is a multiple spawner and the oocyte development is in a group-synchronous manner. Plasma steroid hormones (E2, T, DHP and 20beta-S) and VTG levels changed in parallel with changes in GSI. The peak of plasma VTG level occurred prior to spawning. These cyclic changes of plasma steroid hormones and VTG support the hypothesis that lunar periodicity is the major factor in stimulating reproductive activity of S. guttatus.  相似文献

Effects of different fish-based diets (freshwater smelt, Baltic herring, marine herring/cod offal or their mixtures), gender, beta-glucan supplement, exogenous melatonin, and PCB exposure (Aroclor 1242((R)), 1 mg per animal per day in feed) on plasma immunoglobulin G (IgG) in the mink (Mustela vison) were studied. The aims of the study were to find out whether plasma IgG of the mink is affected by the subchronic PCB exposure, and whether biological, nutritional and hormonal effects are large enough to mask the possible IgG response. The concentration of IgG was determined using enzyme-linked immunosorbent assay (ELISA). Sexual dimorphism was detected, the males having higher levels of plasma IgG. In addition, melatonin tended to decrease IgG in females but not males. Diet also affected the humoral immune arm; the mixed-fish diets caused an unfavorable ratio of the oxidation products of lipids vs. vitamin E in liver, and resulted in low IgG concentration in plasma. In males fed Baltic herring, the beta-glucan supplement also lowered IgG levels. The PCBs failed to affect the plasma IgG of the smelt-fed female mink, and IgG concentration was not correlated with increased hepatic EROD activity or with the decreased total retinol in the liver of exposed mink. It is concluded that hormonal/seasonal and dietary factors affect the plasma IgG levels to such an extent that possible change in plasma IgG level due to PCBs in wild populations of mink is difficult to detect without a large amount of reference data.  相似文献

We examined melatonin profiles of ruin lizards in different seasons (spring, summer, and autumn) under light:dark (LD) and concomitant responses when transferred to continuous darkness (DD) to determine the degree to which previously reported seasonally dependent effects of pinealectomy on locomotor behavior are related to melatonin secretion. The amplitude of the melatonin rhythm and the amount of melatonin produced over 24 h varied with season. In spring, the amount of melatonin produced was greatest and the amplitude 4- 5 times that found in summer or autumn. The degree of self-sustainment of the melatonin rhythm when transferred to DD also varied with season. In DD, melatonin levels remained high but did not exhibit circadian variation in spring. In summer, the melatonin profile persisted virtually unchanged in DD, showing the existence of a circadian rhythm. Finally, in the fall there was no circadian variation in DD and levels remained low. These responses correspond closely to previously reported effects of pinealectomy on locomotor behavior where there is little or no effect of pinealectomy in spring or fall but a profound alteration of locomotor behavior in summer. These results suggest that the seasonally dependent effects of pinealectomy on locomotor behavior in ruin lizards are related to a seasonally mediated change in the degree of self-sustainment of some component of the circadian pace-making system of which melatonin plays some role.  相似文献

Rabbitfishes are known to spawn synchronously around the species-specific lunar phase. It is considered that they perceive and utilize cues from the moon in order to be synchronized gonadal development and spawning with the lunar cycle. Using the golden rabbitfish, Siganus guttatus, which spawns synchronously around the first quarter moon during the reproductive season, we measured the fluctuation of melatonin levels and examined the response of the fish to moonlight intensity. Daily fluctuation of melatonin concentration in the blood of golden rabbitfish showed low levels during daytime and high levels during night-time, suggesting that melatonin functions in the perception and utilization of photoperiod. Plasma melatonin concentration at the new moon was higher than that at the full moon. When the fish were exposed to moonlight at midnight of the both moon phases, the melatonin concentrations decreased to the control levels. These results show that the fish possibly perceive moonlight intensity and plasma melatonin fluctuates according to 'lightness' at a point of night. At the first spawning period (experiment was started one month before the spawning), the fish reared under natural conditions spawned at the expected spawning dates, whereas the fish reared under the constant darkness and lightness of night did not spawn. At the second spawning period (experiment was started 2 weeks before the spawning), the fish reared under the conditions of natural and constant darkness of night spawned but not that of constant lightness of night. It is possible that night conditions are related to synchronous gonadal development and spawning in the golden rabbitfish.  相似文献

Melatonin production by the pineal organ is influenced by light intensity, as has been described in most vertebrate species, in which melatonin is considered a synchronizer of circadian rhythms. In tench, strict nocturnal activity rhythms have been described, although the role of melatonin has not been clarified. In this study we investigated daily activity and melatonin rhythms under 12:12 light-dark (LD) conditions with two different light intensities (58.6 and 1091 microW/cm2), and the effect of I h broad spectrum white light pulses of different intensities (3.3, 5.3, 10.5, 1091.4 microW/cm2) applied at middarkness (MD) on nocturnal circulating melatonin. The results showed that plasma melatonin in tench under LD 12:12 and high light conditions displayed rhythmic variation, where values at MD (255.8 +/- 65.9 pg/ml) were higher than at midlight (ML) (70.7 +/- 31.9 pg/ml). Such a difference between MD and ML values was reduced in animals exposed to LD 12: 12 and low light intensity. The application of 1 h light pulses at MD lowered plasma melatonin to 111.6 +/- 3.2 pg/ml (in the 3.3-10.5 microW/cm2 range) and to 61.8 +/- 18.3 pg/ml (with the 1091.4 microW/cm2 light pulse) and totally suppressed nocturnal locomotor activity. These results show that melatonin rhythms persisted in tench exposed to low light intensity although the amplitude of the rhythm is affected. In addition, it was observed that light pulses applied at MD affected plasma melatonin content and locomotor activity. Such a low threshold suggests that the melatonin system is capable of transducing light even under dim conditions, which may be used by this nocturnal fish to synchronize to weak night light signals (e.g., moonlight cycles).  相似文献

Infection patterns of Mycobacterium marinum were studied over a period of 3 yr in wild rabbitfish Siganus nivulatus populations associated with commercial mariculture cages and inhabiting various sites along the Israeli Red Sea coastline. Mycobacteriosis was first recorded from the Red Sea in 1990 in farmed sea bass Dicentrarchus labrax and is absent from records of studies on parasites and diseases of wild rabbitfish carried out in the 1970s and 1980s. A sharp increase in the prevalence of the disease in cultured and wild fish in the region has occurred since. A total of 1142 rabbitfish were examined over a 3 yr period from inside mariculture net cages, from the cage surroundings and from several sites along the coast. Histological sections of spleens were examined for presence of granulomatous lesions. Overall prevalence levels of 50% were recorded in the rabbitfish sampled inside the net cages and 39% at the cages' close surroundings, 21% at a sandy beach site 1.2 km westwards, 35% at Eilat harbour 3 km to the south and 42% at a coral reef site about 10 km south of the cages. In addition, 147 fish belonging to 18 native Red Sea species were sampled from 2 sites, the net cage farm perimeter and the coral reef area, and examined for similar lesions. None of those from the coral reef were infected with Mycobacterium; however, 9 of 14 species collected from the cage surroundings were infected. An increase in prevalence of mycobacteriosis in the mariculture farm area was noted from 1995 to 1997. At the same time, a significant increase in prevalence was also apparent at the coral reef sampling site. Two M. marinum isolates from rabbitfish captured at Eilat harbour and the coral reef site were shown by 16S rDNA sequencing analysis to be identical to isolates from rabbitfish trapped inside the mariculture cages as well as isolates from locally cultured sea bass D. labrax. The implications of spreading of M. marinum infection in wild fish populations in the Gulf of Eilat are discussed.  相似文献

The detection of threshold concentrations and relative stimulatory effectiveness (RSE) for 19 amino acids were studied by olfactory bulbar electroencephalogram (EEG) in the algivorous rabbitfish. The threshold concentrations for 19 amino acids ranged from 10−10 to 10−5 M. l -Alanine was the most effective amino acid, and the threshold concentration was estimated to be from 10−10 to 10−9M. These results were compared with the olfactory response in herbivorous and carnivorous fishes previously reported. The comparison of RSE for rabbitfish with those for other fishes showed correlation coefficients ranging from r = 0.97 to r = 0.50, but high similarities did not correspond with difference in feeding habits for herbivores and carnivores. The role of amino acids in fish chemosensory behaviour and the olfactory response of the rabbitfish was compared with the gustatory response reported previously. It was found that l -serine was the most potent stimulant in both chemoreceptors, l -proline and l -glutaminic acid were the most potent gustatory stimulants, and l -alanine, l -glutamine, l -arginine and l -lysine were the most potent olfactory stimulants.  相似文献

We investigated circadian ocular rhythms in the Japanese quail, Coturnix coturnix japonica. The birds were placed under light-dark cycles (LD 1212), constant light (LL) and constant darkness (DD), and the retinas were dissected out at four-hour intervals throughout 24 h. Following measurements were performed. (1) Melatonin content in the retina was measured by radioimmunoassay. It was low in light and several folds higher in darkness under LD 1212. The rhythm continued in DD, but disappeared in LL. (2) Mitotic figures in the corneal epithelium were counted. Similar rhythms to the melatonin content were observed in the corneal mitotic rate with a slight phase delay. (3) The retinas were fixed at 4-h intervals and immunostained with anti-bovine rhodopsin serum and anti-chicken iodopsin monoclonal antibodies. The outer segments of photoreceptor cells were stained intensively throughout 24 h in LD 1212, LL and DD. In contrast, the stainability of the locus close to the outer limiting membrane where the Golgi apparatus exists changed diurnally. Scores showing the ratio of cells with positive staining indicated high values from 4 h after the onset of light to the beginning of dark phase under LD 1212. The values were high throughout 24 h in LL and intermediate or low in DD. (4) To investigate the effect of melatonin on the corneal mitotic rate and visual pigments at the Golgi region, melatonin was injected into one eye and saline into the contralateral eye. Melatonin induced a phase advance in the corneal mitotic rate under LD 1212, but did not induce a rhythm under LL. The age, growth, mortality, reproduction and resource status of Siganus canaliculatus in the southern Arabian Gulf were investigated using a combination of size frequency, biological and size‐at‐age data. Defined structural increments consisting of alternating translucent and opaque bands in transverse sections of sagittal otoliths were validated as annuli. The maximum absolute age estimate was 7.8 years. Parameter values of the von Bertalanffy growth function fit to size‐at‐age data (males and females combined) were: k = 1.0, L = 24.8 cm (LF), to = −0.1 years. Fish in spawning condition were only observed between April and July although patterns in gonadosomatic indices suggested a second but less well defined spawning event in November. The mean sizes and ages at first sexual maturity were 21.5 cm LF (1.9 years) for males and 25.7 cm LF (2.1 years) for females. Fish were fully recruited to the fishery at a size (L100 = 19.7 cm LF) that was smaller than the sizes at which sexual maturity was attained. The annual instantaneous rate of fishing mortality (F = 0.85 year−1) (0.26–1.44 year−1 95% CI) was considerably greater than the target (Fopt = 0.33 year−1) and limit (Flimit = 0.44 year−1) biological reference points, indicating that the stock is overexploited.  相似文献

The study was conducted to investigate diurnal changes in pinealocyte ultrastructure, pineal serotonin content and plasma melatonin concentration in the domestic pig. The immature pigs (n=24) were kept under a cycle of 12 h light : 12 h dark, with a photophase between 0800 and 2000. During the photophase the animals were exposed to direct sunlight. After four weeks the gilts were slaughtered at 0900, 1400, 2100 and 0200. The pineals were removed and divided into two parts - one for quantitative ultrastructural study (by a point count method) and one for serotonin assay. Simultaneously, blood samples were taken for melatonin assay. The relative volume of mitochondria in pinealocyte perikarya was significantly higher at 1400 than at 0200 and 0900 as well as at 2100 than at 0200. The relative volume of Golgi apparatus was higher at 0900 and 1400 than at 0200. The relative volume of dense bodies of the MBB-1 type in pinealocyte perikarya was significantly lower at 1400 and 2100 than at 0900. In contrast, the relative volume of MBB-2 was higher at 1400 than at 0900 and 0200. The numerical density of DCV in perikarya was significantly higher at 0200 than at 1400. No significant differences were found in rough endoplasmic reticulum, lysosomes and multivesicular bodies. The pineal serotonin content showed a prominent rhythm with the maximum at 1400. The plasma melatonin concentration was significantly higher at 0200 than at 0900, 1400 and 2100. The obtained results demonstrate that both pinealocyte ultrastructure and pineal biochemistry in the pig undergo significant changes in the course of the diurnal rhythm.  相似文献
