In perfused human umbilical arteries both angiotensin I and II induced vasoconstriction with a monophasic response. Angiotensin I and II induced vasoconstrictions at doses greater than or equal to 10(-8) M and 10(-9) M respectively. Captopril inhibited the angiotensin I response while the angiotensin II receptor blocker Sar1-Ala8 AII inhibited the effect of both angiotensins. PGI2 attenuated the angiotensin II response in a dose dependent pattern. PGE2 and PGF2 alpha in concentrations below the critical levels for creating pressure responses per se, also attenuated the angiotensin II response. The cyclooxygenase inhibitor indomethacin potentiated the angiotensin II response indicating that endogenous production of prostanoids is of importance in the modulation of angiotensin effects.  相似文献

The purpose of this study was to determine the effects of chronic administration of the thromboxane synthetase inhibitor, UK 38,485, on noradrenergic neurotransmission. Male Sprague Dawley rats (n=14) were treated once daily with either UK 38,485 (100 mg/kg; n=7) or the vehicle of UK 38,485 (olive oil; n=7) by gavage. The dose of UK 38,485 chosen was sufficient to inhibit ex vivo platelet TXB2 production by >90% for 24 hours. One week into the treatment animals were prepared for in situ perfusion of their mesenteric vascular beds. Vasoconstrictor responses to both exogenous norepinephrine and periarterial nerve stimulation were determined both before and during an infusion of angiotensin II (9ng/min) into the superior mesenteric artery. UK 38,485 significantly (P<0.02) attenuated the vascular response to periarterial nerve stimulation without altering the vascular response to either norepinephrine or angiotensin II. UK 38,485 did not influence the baseline perfusion pressure, the mean arterial blood pressure or the potentiation of neurotransmission by angiotensin II. These data indicate that in the in situ rat mesentery UK 38,485 attenuates the release of neurotransmitter from sympathetic nerve terminals.  相似文献

Angiotensins I, II, and III were separated by reversed-phase high-performance liquid chromatography on an octadecylsilyl column. The peptides were isocratically eluted with 50 mm NaH2PO4-25% (vv) acetonitrile, pH 6.0. The retention times were 3.3, 6.0, and 9.6 min for angiotensin II, III, and I, respectively. 125I-Angiotensins II, III, and I eluted with retention times of 5.4, 16.8, and 19.9 min, respectively, under the same chromatographic conditions used for the unlabeled angiotensins. The effect of iodination of the tyrosine residue on the retention time was also demonstrated by chromatographic comparison of tyrosine and diiodotyrosine. Saralasin (Sar1, Ala8-angiotensin II), a partial agonist of angiotensin II, and des-Asp1, Ile8-angiotensin II, an inhibitor of angiotensin III, eluted with retention times of 2.5 and 3.9 min, respectively.  相似文献

A rabbit lung preparation, perfused in vitro, was used to examine pulmonary metabolism of prostaglandin A1 (PGA1) and to compare the vasoconstrictor actions of PGA1, prostaglandin F (PGF) and angiotensin II. PGF caused significantly more, and angiotensin II significantly less, vasoconstriction than did an equimolar concentration of PGA1. Of three likely PGA1 metabolites only 15-keto-PGA1 had any significant vasoconstrictor action. Furosemide and aminophylline (10?3 M) reduced PGA1, PGF or angiotensin II-induced vasoconstruction. Diphloretin phosphate potentiated the vascular effect of angiotensin II. Furosemide (10?3 M) and DPP (9.5 × 10?6 M) significantly reduced pulmonary metabolism of PGA1 while aminophylline (10?3 M) had no effect on this process. Perfusion of the lungs with a hypoxic medium had no effect on PGA1 metabolism.  相似文献

Two nitroxide radicals (TEMPO, I; OXAN, II) and a spin labeled penicillin (III) were reduced by Staphylococcus aureus. A short induction period preceded zero order reduction of these substrates leading to a Km of 8 × 10?4M, 6.67 × 10?5M and 5.7 × 10?4M and Vmax of 106, 26 and 11 μ mole/min mg bacteria for I, II and III, respectively.  相似文献

The amino- and carboxy-terminal substituent tetrapeptides of angiotensin II, Asp-Arg-Val-Tyr and Ile-His-Pro-Phe, elicit substantial human mononuclear leukocyte chemotactic responses invitro that attain maximal levels at tetrapeptide concentrations of 3 × 10?8 M and 3 × 10?7 M, respectively. In contrast, the angiotensin II-derived tetrapeptides evoke only marginal human neutrophil chemotactic responses. Amino acid deletions or substitutions that alter the properties of the tetrapeptides, reduce their chemotactic potency and activity. Limited proteolytic cleavage of angiotensin II thus may convert a pathway with predominantly humoral effects to a source of mediators that regulate cellular immunity and chronic inflammatory responses.  相似文献

The cell-free preparations from autotrophieally grown Pseudomonas saccharophila catalyzed the process of electron transport from H2 or various other organic electron donors to either O2 or NO3? with concomitant ATP generation. The respective PO ratios with H2 and NADH were 0.63 and 0.73, the respective PNO3? ratios were 0.57 and 0.54. In contrast, the PO and PNO3? ratios with succinate were 0.18 and 0.11, respectively. ATP formation coupled to the oxidation of ascorbate, in the absence or presence of added N,N,N′,N′-tetramethyl-p-phenylenediamine or cytochrome c, could not be detected. Various uncouplers inhibited phosphorylation with either O2 or NO3? as terminal electron acceptors without affecting the oxidation of H2 or other substrates. The NADH oxidation at the expense of O2 or NO3? reduction as well as the associated phosphorylation were inhibited by rotenone and amytal. The aerobic and anaerobic H2 oxidation and coupled ATP synthesis, on the other hand, was unaffected by the flavoprotein inhibitors as well as by the NADH trapping system. The NADH, H2, and succinate-linked electron transport to O2 or NO3? and the associated phosphorylations were sensitive, however, to antimycin A or 2-n-nonyl-4-hydroxyquino-line-N-oxide, and cyanide or azide. The data indicated that although the phosphorylation sites 1 and II were associated with NADH oxidation by O2 or NO3?, the energy conservation coupled to H2 oxidation under aerobic or anaerobic conditions appeared to involve site II only.  相似文献

We have recently demonstrated that gonadotropin releasing hormone (GnRH) acts directly on ovarian granulosa cells to inhibit the follicle stimulating hormone (FSH)-induced increase in granulosa cell steroidogenesis invitro. A GnRH antagonist, [D-pGlu1, D-Phe2, D-Trp3,6] GnRH (A), which is known to antagonize GnRH-stimulated gonadotropin release by cultured pituitary cells, was tested in the granulosa cell system. GnRH (10?8M) inhibited estrogen and progesterone production by FSH-treated granulosa cells invitro, whereas the antagonist A (10?6M) did not affect FSH stimulation of steroidogenesis. Antagonist A, when added together with GnRH and FSH, blocked the GnRH inhibition of FSH-induced steroidogenesis. Estrogen and progesterone production by granulosa cells was increased by 50% at a molar ratio (IDR50) of 201and121 ([antagonist]/[GnRH]), respectively. At 10?6M, antagonist A completely prevented the GnRH (10?8M) inhibition. A similar effect of antagonist A was seen in FSH-induced increase of luteinizing hormone (LH) receptor content. FSH treatment for 2 days invitro induced an 8-fold increase in LH receptor content in cultured granulosa cells; concomitant treatment with 10?8M GnRH completely inhibited the FSH effect. Antagonist A (10?6M), by itself, had no effect on the FSH action. However, when added together with FSH and GnRH, antagonist A completely abolished the inhibitory effect of GnRH. These results demonstrate that the direct inhibitory effect of GnRH on granulosa cell function can be prevented by a GnRH antagonist and that the GnRH action at the ovarian level may require stringent stereospecific interactions of these peptides with putative GnRH recognition sites.  相似文献

The concurrent effect of indomethacin or aspirin on prostaglandins (PGs) biosynthesis and on cellular fatty acid efflux were compared. Studies with rabbit kidney medulla slices and with isolated perfused rabbit kidney showed a marked difference between the two non-steroidal anti-inflammatory drugs, with regard to their effects on fatty acid efflux from kidney tissue. While aspirin effect was limited to inhibition of PGs biosynthesis, indomethacin also reduced the release of free fatty acids. In medullary slices, indomethacin inhibited the Ca2+ stimulation of phospholipase A2 activity and the resulting release of arachidonic and linoleic fatty acids. In the isolated perfused rabbit kidney, indomethacin inhibited the basal efflux of all fatty acids as well as the angiotensin II — induced selective release of arachidonate. Indomethacin also blunted the angiotensin II — induced temporal changes in the efflux of all other fatty acids. Neither indomethacin nor aspirin affected significantly the uptake and incorporation of exogenous (14C)-arachidonic acid into kidney total lipid fraction.Our tentative conclusion is that indomethacin inhibits basal as well as Ca2+ or hormone stimulated activity of kidney lipolytic enzymes. This action of indomethacin reduces the pool size of free arachidonate available for conversion to oxygenated products (both prostaglandin and non-prostaglandin types). The non-steroidal anti-inflammatory drugs can therefore be divided into two groups: a) aspirin-type compounds which inhibit PGs formation only by interacting with the prostaglandin endoperoxide synthetase and b) indomethacin-type compounds which inhibit PG generation by both reduction in the amount of available arachidonate and direct interaction with the enzyme.  相似文献

We have used [125I] angiotensin II to investigate the presence of specific angiotensin II receptors in beef heart sarcolemmal membranes. The observed binding is saturable, reversible and specific. The apparent equilibrium dissociation constant is 2.23 ± 0.15 (x ± SEM) and the maximal number of binding sites per mg membrane protein is 32.8 ± 5.4 fmol (x ± SEM). The specific binding is 80–100% of the total [125I] angiotensin II bound and is directly proportional to membrane protein concentration over the range of 33–173 μg protein per ml. Angiotensin II and its antagonists competed for binding in a potency order of (agent, Ki): angiotensin II, 0.9nM > Sar1 Ala3, 7 nM > Sar1-Ile3, 51 nM > Sar1-Leu3, 427nM > angiotensin I, 1709 nM. The ability to characterize and quantify these receptors should now provide a method for investigating the mechanisms underlying the effects of angiotensin II on myocardial tissues.  相似文献

In human cervical (CaSki) cells, extracellular adenosine triphosphate (ATP) induces an acute decrease in the resistance of the lateral intercellular space (R LIS), phase I response, followed by an increase in tight junctional resistance (R TJ), phase II response. ATP also stimulates release of calcium from intracellular stores, followed by augmented calcium influx, and both effects have similar sensitivities to ATP (EC50 of 6 μM). The objective of the study was to determine the degree to which the changes in [Ca2+]i mediate the responses to ATP. 1,2-bis (2-aminophenoxy) ethane-N,N,N1,N1-tetraacetic acid (BAPTA) abrogated calcium mobilization and phase I response; in contrast, nifedipine and verapamil inhibited calcium influx and attenuated phase II response. Barium, La3+, and Mn2+ attenuated phase I response and attenuated and shortened the ionomycin-induced phase I-like decrease inR LIS, suggesting that store depletion-activated calcium entry was inhibited. Barium and La3+ also inhibited the ATP-induced phase II response, but Mn2+ had no effect on phase II response, and in the presence of low extracellular calcium it partly restored the increase inR TJ. KCl-induced membrane depolarization stimulated an acute decrease inR LIS and a late increase inR TJ similar to ATP, but only the latter was inhibited by nifedipine. KCl also induced a nifedipine-sensitive calcium influx, suggesting that acute increases in [Ca2+]i, regardless of mobilization or influx, mediate phase I response. Phase II-like increases inR TJ could be induced by treatment with diC8, and were not affected by nifedipine. Biphasic ATP-like changes inR TE could be induced by treating the cells with ionomycin plus diC8. We conclude that calcium mobilization mediates the early decrease inR LIS, and calcium influx via calcium channels activates protein kinase C and mediates the late increase inR TJ.  相似文献

The role of ACTH-(1–24) on angiotensin II receptors has been studied in bovine adrenal glomerulosa cells in primary culture. Angiotensin II receptors were measured in cells pretreated or not by ACTH-(1–24) on day 4 of culture. ACTH-(1–24) decreased angiotensin II binding sites in a time and a dose-dependent manner. After 24 hours of treatment the minimal effective dose of ACTH-(1–24) was 10?11M and the maximal effect was obtained with 10?8M. Moreover, ACTH-(1–24) 10?8M decreased significantly angiotensin II receptors after 6 hours of treatment. Scatchard plot analysis showed that ACTH-(1–24) treatment did not modify the affinity of angiotensin II receptors (Ka = 0.42 and 0.44 × 109M?1 in control and treated cells respectively) but reduced by about half the number of angiotensin II sites per cell. Like ACTH-(1–24), 8-Bromo-cAMP, forskolin and cholera toxin decreased angiotensin II receptors. Factors such as prolactin, somatostatin, ACTH-(11–24) and dopamine which are bound to adrenal membranes without increasing cAMP production had no effect. In conclusion, these studies in vitro demonstrate for the first time that ACTH decreases angiotensin II receptors by a direct mechanism acting on glomerulosa cells, and they also suggest that this effect could be mediated by cAMP.  相似文献

The ATPase enzyme system from two-spotted spider mites, Tetranychusurticae (Koch) was sensitive, in vitro, to four acaricides. Tricyclohexylhydroxytin (PlictranR) was an outstanding inhibitor of oligomycin-sensitive (mitochondrial) Mg2+ATPase from fish brain and spider mite homogenates. The I50 values were 6.6×10?11M and 6.2×10?10M, respectively. Less effective were chlorbenside, chlorfenethol and ovotran. Plictran at a higher concentration (2×10?7M) was also more effective on Na+-K+ATPase both in mites and fish brain homogenates as compared to chlorfenethol, chlorbenside and ovotran. Plictran inhibited oligomycin-insensitive Mg2+ATPase at concentrations of 10?8M but stimulated at high concentrations (5×10?6M and higher).  相似文献

In this study we examined the conformation and side chain environments of angiotensins I, II, III, and [Sar1-Ile5-Ala8]angiotensin II using laser Raman spectroscopy. The positions of the amide I bands for all four peptides were found between 1664 and 1673 cm?1. D2O exchange studies confirmed the positions of the amide I and amide III bands. The positions of the amide I bands for all the angiotensins were found at approximately 1665 cm?1 and the amide III bands were all located between 1265 and 1278 cm?1. From the positions and intensities of the amide I and III bands we concluded that all peptides share the same overall conformation consisting of β-turn structure. Spectral analysis indicated that although the spectra for all the peptides were qualitatively identical there was evidence that the angiotensin conformations were more flexible in the aqueous phase than the solid phase. Examination of the 850830 cm?1 tyrosine doublet suggested that the tyrosine residue in the peptides is exposed to the solvent environment and becomes more exposed as the peptide length is decreased. Therefore, there are some localized conformational differences among the angiotensins. The conformational data yielded by this study leads us to conclude that the various biological properties ascribed to the angiotensins are not due to different conformations of the peptides. The biological differences could perhaps be attributed to localized interactions of the individual amino acid residues with themselves and with the hormone receptors.  相似文献

Both isomers of [3.6-dioxa-(8-(2,4-dinitrophenoxy))octyl] methylphosphono-fluoridate (I) react rapidly with eel acetylcholinesterase. At pH 6.61 and 25°C, the reaction rate constants for the fast and slow isomers are 3 × 109 and 5 × 107M?1 min?1. Despite the very high rate of reaction with acetylcholinesterase, which for the fast isomer is approximately 40 times that of sarin, the toxicity of I is only 19 that of sarin. Enzyme inhibited by I is resistant to reactivation by TMB-4.  相似文献

A general method for the determination of compounds possessing either the primary amine structure, R-CH2-NH2 (I), or the α-amino acid structure, RCHNH2COOH (II), has been devised using gas chromatography and mass spectrometry. Trimethylsilyl derivatives of the biogenic amines (phenylethylamines, indoleethylamines, or Ω-amino acids) produce an intense ion at me 174 upon fragmentation; TMS derivatives of α-amino acids produce an ion at me 218. For maximum sensitivity, chromatograms were obtained with the mass spectrometer tuned to detect a single ion fragment characteristic of a group of structurally related compounds (i.e., functional group GC-MS). At me 174 up to 14 compounds of Type I, including glycine, γ-aminobutyric acid, dopamine, and 5-hydroxytryptamine, could be determined in a single analysis. Detection limits range from 10–100 femtomoles (10?15 moles). At me 218, eight compounds of Type II, including isoleucine, phenylalanine, tyrosine, and DOPA could be determined. This technique has been applied to the assay of these compounds in extracts containing 0.1 mg mouse brain or abdominal ganglia of the marine molluse, Aplysia californica.  相似文献

NH2OH-treated, non-water-splitting chloroplasts can oxidize H2O2 to O2 through Photosystem II at substantial rates (100–250 μequiv · h?1 · mg?1 chlorophyll with 5 mM H2O2) using 2,5-dimethyl-p-benzoquinone as an electron acceptor in the presence of the plastoquinone antagonist dibromothymoquinone. This H2O2 → Photosystem II → dimethylquinone reaction supports phosphorylation with a Pe2 ratio of 0.25–0.35 and proton uptake with H+e values of 0.67 (pH 8)–0.85 (pH 6). These are close to the Pe2 value of 0.3–0.38 and the H+e values of 0.7–0.93 found in parallel experiments for the H2O → Photosystem II → dimethylquinone reaction in untreated chloroplasts. Semi-quantitative data are also presented which show that the donor → Photosystem II → dibromothymoquinone (→O2) reaction can support phosphorylation when the donor used is a proton-releasing reductant (benzidine, catechol) but not when it is a non-proton carrier (I?, ferrocyanide).  相似文献

Secretory vesicles isolated from adrenal medulla were found to fuse in vitro in response to incubation with Ca2+. Intervesicular fusion was detected by electron microscopy and was indicated by the appearance of twinned vesicles in freeze-fractured suspensions of vesicles and in thin-sectioned pellet. Two types of fusion could be distinguished: Type I, occurring between 10?7 M and 10?4 M Ca2+, was specific for Ca2+, was inhibited by other divalent cations and was abolished by pretreatment of vesicles with glutaraldehyde, neuraminidase or trypsin. Fusion type I was linear with temperature. A second type of intervesicular fusion was elicited by Ca2+ in concentrations higher than 2.5 mM and was morphologically characterized by multiple fusions of secretory vesicles. This type of fusion was found to be similar to fusion of liposomes prepared from the membrane lipids of adrenal medullary secretory vesicles: Ca2+ could be replaced by other divalent cations, the effect of different divalent cations was additive and pretreatments attacking membrane proteins were ineffective. Fusion type II of intact secretory vesicles as well as liposome fusion was discontinuous with temperature. Liposome fusion could be detected within 35 ms and persisted for 180 min. Using liposomes containing defined Ca2+ concentrations we have not found a major influence of Ca2+ asymmetry on fusion. Incorporation of the ganglioside GM3, which is present in the membranes of intact adrenal medullary secretory vesicles did not change the properties of liposomes fusion. Using a Ca2+-selective electrode we have identified in secretory vesicle membranes both high affinity binding sites for Ca2+ (Kd = 1.6 · 10?6M) and low affinity sites (Kd = 1.2 · 10?4M).  相似文献

Exogenously added histamine in non-cytotoxic concentrations (10?5?10?3M) suppresses in vitro proliferation of lymphocytes induced by PHA or Concanavalin A. This suppressive effect was observed when histamine was present for as short as 12 hr in the beginning of the culture. Histamine, in concentrations as high as 10?3M, did not cause increased release of isotope from 51Cr-labeled lymphocytes following 4 hr of incubation. The histamine H2 receptor antagonist, metiamide, but not the H1 receptor antagonists diphenhydramine or chlorpheniramine, blocked the histamine suppressive effect. Some of the biological implications of these findings are discussed.  相似文献

Ring hydroxylation of 1-(2-chloroethyl)-3-cyclohexyl-1-nitrosourea was shown to occur in the presence of liver microsomes prepared from both normal and phenobarbital induced rats. The metabolite was identified by mass spectrometry after selective extraction and purification by liquid chromatography. The microsomal catalyzed reaction was oxygen and NADPH dependent, inhibited by carbon monoxide and induced 4–5 fold by in vivo phenobarbital pre-treatment. Phenobarbital induced microsomes hydroxylated the substrate at a rate of 17.6 nmoles/min/mg protein at 37°. A Type I difference spectrum was observed with phenobarbital induced microsomes that also displayed a substrate binding constant (Ks of 4 × 10?5 M.  相似文献
