Chemical and biological diversity of protein structures and functions can be widely expanded by position-specific incorporation of non-natural amino acids carrying a variety of specialty side groups. After the pioneering works of Schultz's group and Chamberlin's group in 1989, noticeable progress has been made in expanding types of amino acids, in finding novel methods of tRNA aminoacylation and in extending genetic codes for directing the positions. Aminoacylation of tRNA with non-natural amino acids has been achieved by directed evolution of aminoacyl-tRNA synthetases or some ribozymes. Codons have been extended to include four-base codons or non-natural base pairs. Multiple incorporation of different non-natural amino acids has been achieved by the use of a different four-base codon for each tRNA. The combination of these novel techniques has opened the possibility of synthesising non-natural mutant proteins in living cells.  相似文献

The synthesis of soluble liver proteins and amino acid incorporation into blood serum proteins were studied in guinea pigs which were daily administered ethanol and cholesterol during 3 months. 9 fractions obtained by electrophoresis in polyacrylamide gel were analyzed. It has been found that ethanol and cholesterol in the liver suppress the synthesis of 4 out of 9 recorded protein fractions. In the serum the ethanol effect against a background of the cholesterol administration is characterized by a sharp decrease of specific radioactivity of albumins while the quantity of the protein fraction is unchanged. These results together with the data available in literature on the ethanol suppression of proteolysis suppose that the joint effect of ethanol and cholesterol is accompanied by a decrease in the blood serum albumin decomposition.  相似文献

Nuclei isolated from the lower eukaryote, Euglena gracilis, incorporate labelled amino acids into hot-TCA-insoluble material under conditions of incubation similar to those used for higher eukaryotes. Optimal temperature for amino acid incorporation by Euglena nucleic is 25° to 30°C. Optimal pH is 5.0. ATP stimulates incorporation only to a small extent. Evidence is presented that the product synthesized is protein.  相似文献

Labelled amino acids, 14C-leucine, 3H-glycine, 14C-GABA, 14C-glutamic acid and 14C-aspartic acid were applied to the surface of the cat's auditory cortex in dried filter paper strips for 40 min. The distribution of the amino acids taken up by the cortex was examined by autoradiography.The right acoustic area was stimulated by 2 cps acoustic clicks, through an ear-phone. Leucine showed cellular localization which was considerably dispersed by cortical excitation. Glycine was concentrated in nerve cells, mainly in layers I-II, but excitation intensified and extended its incorporation. GABA was accumulated rather diffusely in layers I-II with scattered cellular labelling which was depressed by stimulation. Glutamic and aspartic acids did not show any characteristic distribution pattern. The authors emphasize that the incorporation of glycine seems to be a good indicator of neurons being in excitation. The localizations of GABA uptake shows close correlation with the site of its physiological action.  相似文献

Isolated livers from fed rats were perfused with a medium containing glucose labeled uniformly with 14C and specifically with 3H. There was considerable formation of glucose from endogenous sources but simultaneously uptake of about half of the 14C in glucose. After 2 hours the 3H14C ratios in perfusate glucose decreased by 55–60% with (2-3H, U-14C), 40–50% with (5-3H, U-14C), 25–30% with (3-3H or 4-3H, U-14C) and by 10–15% with (6-3H, U-14C) glucose. Qualitatively comparable patterns were obtained with rat hepatocytes. These results demonstrate recycling of carbon between glucose and pyruvate. Superimposed upon this there is an extensive futile cycle between glucose and glucose 6-P. There is also futile cycling between fructose 6-P and fructose 1,6 P2 and to a small extent between phosphoenol pyruvate and pyruvate.  相似文献

1. The rate of gluconeogenesis from alanine in the perfused rat liver is affected by the presence of other metabolizable substances, especially fatty acids, ornithine and ethanol. Gluconeogenesis is accelerated by oleate and by ornithine. When both oleate and ornithine were present the acceleration was greater than expected on the basis of mere additive effects. 2. Much NH(3) and some urea were formed from alanine when no ornithine was added. With ornithine almost all the nitrogen released from alanine appeared as urea. 3. Lactate was a major product of alanine metabolism. Addition of oleate, and especially of oleate plus ornithine, decreased lactate formation. 4. Ethanol had no major effect on gluconeogenesis from alanine when this was the sole added precursor. Gluconeogenesis was strongly inhibited (87%) when oleate was also added, but ethanol greatly accelerated gluconeogenesis when ornithine was added together with alanine. 5. In the absence of ethanol the alanine carbon and alanine nitrogen removed were essentially recovered in the form of glucose, lactate, pyruvate, NH(3) and urea. 6. In the presence of ethanol the balance of both alanine carbon and alanine nitrogen showed substantial deficits. These deficits were largely accounted for by the formation of aspartate and glutamine, the formation of which was increased two- to three-fold. 7. When alanine was replaced by lactate plus NH(4)Cl, ethanol also caused a major accumulation of amino acids, especially of aspartate and alanine. 8. Earlier apparently discrepant results on the effects of ethanol on gluconeogenesis from alanine are explained by the fact that under well defined conditions ethanol can inhibit, or accelerate, or be without major effect on the rate of gluconeogenesis. 9. It is pointed out that in the synthesis of urea through the ornithine cycle half of the nitrogen must be supplied in the form of asparate and half in the form of carbamoyl phosphate. The accumulation of aspartate and other amino acids suggests that ethanol interferes with the control mechanisms which regulate the stoicheiometric formation of aspartate and carbamoyl phosphate.  相似文献

Primary cultures of rat liver parenchymal cells maintained as a monolayer in serum-free culture medium were used to investigate the characteristics of zinc accumulation in vitro. Liver parenchymal cells accumulated zinc by a temperature-dependent, saturable process that was inhibited by cyanide, azide, oligomycin, N-ethylmaleimide and iodoacetamide. Cadmium reversibly inhibited zinc accumulation in both serum-free and serum-containing media. Gel filtration chromatographic studies showed that recently accumulated intracellular zinc was present as a low molecular weight complex smaller than metallothionein, the zinc storage protein, but larger than individual amino acids. The quantity of zinc accumulated was affected by preincubation of the cells with various hor?ONES. Dexamethasone, prednisone and prednisolone each increased zinc uptake by 40--50% when either insulin or glucagon was also present. Hydrocortisone, cortisone and sex steroids did not influence zinc accumulation. Removal of the polypeptide hormones from the medium abolished the stimulatory effect of the synthetic glucocorticoid steroid hormones on zinc accumulation.  相似文献

Chronic ethanol consumption reduces the liver retinoid store in man and rat. We have studied the effect of ethanol on some aspects of retinoid metabolism in parenchymal and nonparenchymal liver cells. Rats fed 36% of total energy intake as ethanol for 5-6 weeks had the liver retinoid concentration reduced to about one-third, as compared to pair-fed controls. The reduction in liver retinoid affected both the parenchymal and the nonparenchymal cell fractions. Plasma retinol level was normal. Liver uptake of injected chylomicron [3H]retinyl ester was similar in the experimental and control group. The transport of retinoid from the parenchymal to the nonparenchymal cells was not found to be significantly retarded in the ethanol-fed rats. Despite the reduction in total retinoid level in liver, the concentrations of unesterified retinol and retinyl oleate were increased in the ethanol fed rats. Hepatic retinol esterification was not significantly affected in the ethanol-fed rats. Since our study has demonstrated that liver uptake of chylomicron retinyl ester is not impaired in the ethanol-fed rat, we suggest that liver retinoid metabolism may be increased.  相似文献
