Summary New water-soluble red pigments were produced byMonascus sp. in a chemically defined fermentation medium containing glutamate as nitrogen source. They were isolated and characterized as glutamate derivatives of the well-known orangeMonascus pigments (monascorubrin and rubropunctatin). The new pigments have several advantages over the known redMonascus pigments (rubropunctamine and monascorubramine) including very high water-solubility, higher absorption coefficient, and greater resistance to decoloration by light. Adding glutamate, glycine or leucine to a resting-cell system led to the formation of specific water-soluble red pigments corresponding to the exogenous amino acid. The water-soluble red pigments produced by resting-cells have retention times identical to those of the corresponding red derivatives made chemically from the orange pigments in methanol-phosphate buffer at pH 7. The hydrophobicities of the amino acid sources correspond to the HPLC retention times of the red pigments derived from them.  相似文献

The production of red pigments and citrinin by Monascus purpureus CCT3802 was investigated in submerged batch cultures performed in two phases: in the first phase, cells were grown on glucose, at pH 4.5, 5.5 or 6.5; after glucose depletion, pH was adjusted, when necessary, to 4.5, 5.5, 6.5, 7.0, 8.0 or 8.5, for a production phase. The highest total red pigments absorbance of 11.3 U was 16 times greater than the lowest absorbance and was achieved with growth at pH 5.5, followed by production at pH 8.5, which causes an immediate reduction of the intra cellular red pigments from 75% to 17% of the total absorbance. The lowest citrinin concentration, 5.5 mg L−1, was verified in the same culture while the highest concentration, 55 mg L−1, was verified in cultures entirely carried out at pH 5.5. An alkaline medium, besides promoting intra cellular red pigments excretion, strongly represses citrinin synthesis.  相似文献

For the purpose of mass producingMonascus red pigments optimum medium composition and environmental conditions were investigated in submerged flask cultures. The optimum carbon and nitrogen sources were determined to be 30 g/L of glucose and 1.5 g/L of monosodium glutamate (MSG). Of the three metals examined, Fe2+ showed the stronges stimulatory effect on pigment production and some stimulatory effect was also found in Mn2+. Optimum pH and agitation speed were determined to be 6.5 and 700 rpm, respectively. Under the optimum culture conditions batch fermentation showed that the maximum biomass yield and specific productivity of red pigments were 0.20 g DCW/g glucose and, 32.5 OD500 g DCW−1 h−1, respectively.  相似文献

Adventitious roots were induced from red beet (Beta vulgaris L. cv. Detroit dark red) by infecting the plant with a soil bacterium, Agrobacterium rhizogenes. Based on analysis of opines which are uniquely produced in transformed hairy roots, the established clone was proved to be a transformed hairy root. In a shake culture of the beet hairy root clone with a liquid medium, it was found that significant amounts of pigments, mainly betanin and vulgaxanthin-I, were released into the medium by the cessation of culture shaking (temporary limitation of oxygen supply). The hairy root cells were capable of propagation even after the cells were subjected to shaking cessation. Repeated-batch culture of the beet hairy root was performed with the cell growth phases for 9 or 10 d and with pigment leakage phases during shaking cessation for 2 d, and more than 20% of the total intracellular pigments were recovered from the culture broth at a culture time of 35 d. The released pigments were confirmed to be substantially identical to those extracted from the hairy root and original plant cells of red beet.  相似文献

In parallel with the studies reported in the preceding paper(I), the modes of production of characteristic red pigmentsby Chlorella protothecoides cells were investigated under variousculture conditions, (i) During the course of "acetate-bleaching"of algal cells, excretion of red pigments in the medium proceededwith simultaneous disappearance of chlorophyll from algal cells.The total amount (weight) of the red pigments excreted intomedium was slightly less than that of the chlorophyll lost.No red pigment was detectable within the bleaching algal cells.Carotenoids were found to increase or remain nearly constantin their quantities per culture during the process of bleaching,(ii) In a later phase of "glucose-bleaching" some red pigmentswere found to be present inside as well as outside the algalcells, and the excreted pigments underwent further changes turningcolourless, (iii) Both the production of red pigments and disappearanceof chlorophyll were suppressed by light and this light effectwas insensitive to CMU. (iv) During the process of "regreening"of "glucose-bleached" algal cells, no production of red pigmentswas observed either in or outside the algal cells. Based on these results we concluded that the red pigments areproduced from chlorophyll during the bleaching process of algalcells induced by an organic carbon source. Summary A resting cell system was developed for the biosynthesis of soluble red pigments byMonascus. The medium contains glucose, glycine, ZnSO4 and MnSO4 in pH 7.0 MOPS buffer containing cycloheximide to prevent protein synthesis. The linear production observed over a period of at least four h was due to de novo polyketide synthesis and biological methylation, as shown by inhibition with cerulenin, iodoacetamide and ethionine. Production was inhibited by carbonyl reagents and stimulated by pyridoxamine suggesting that the conversion of endogenous intracellular orange pigments to extracellular red pigments involves Schiff base intermediates and vitamin B6 a cofactor. The resting cell system was used to study the mode of action of nutritional effectors previously pinpointed by experiments with growing cells. The negative effects of high concentrations of phosphate and Mg++ are due to inhibition of pigment synthase action, not to repression or inactivation of these enzymes. The positive effects of trace metals, especially Zn++, are due to stimulation of growth and enzyme action, not to induction or stabilization of the synthases.  相似文献

Optimal C:N ratio for the production of red pigments by Monascus ruber   总被引:1,自引:0,他引:1  
The carbon-to-nitrogen (C:N) ratio in the biomass of microfungi tends to be quite different (e.g. 10–15) compared with the C:N ratio in the red pigments (e.g. >20) of the fungus Monascus ruber. Therefore, determining an optimal C:N ratio in the culture medium for maximizing the production of the pigments is important. A culture medium composition is established for maximizing the production of the red pigment by the fungus M. ruber ICMP 15220 in submerged culture. The highest volumetric productivity of the red pigment was 0.023 AU L?1 h?1 in a batch culture (30 °C, initial pH of 6.5) with a defined medium of the following composition (g L?1): glucose (10), monosodium glutamate (MSG) (10), MgSO4·7H2O (0.5), KH2PO4 (5), K2HPO4 (5), ZnSO4·7H2O (0.01), FeSO4·7H2O (0.01), CaCl2 (0.1), MnSO4·H2O (0.03). This medium formulation had a C:N mole ratio of 9:1. Under these conditions, the specific growth rate of the fungus was 0.043 h?1 and the peak biomass concentration was 6.7 g L?1 in a 7-day culture. The biomass specific productivity of the red pigment was 1.06 AU g?1 h?1. The best nitrogen source proved to be MSG although four other inorganic nitrogen sources were evaluated.  相似文献

To maximize and sustain the productivity ofMonascus pigments, various environmental and nutritional parameters, such as the initial moisture content, pH, inoculum size, sample size, and nutrient supplement, that influence pigment production were evaluated in solid-state cultures as follows: initial moisture content, 50%; pH, 6.0; inoculum size 1 x 104 spore cells (grams of dry solid substrate)−1; sample size, 300 g. All supplementary nutrients (carbon, nitrogen, and mineral sources) added has inhibitory effects on the cell growth and red pigment production. In open tray culture the maximum biomass yield and specific productivity of red pigments were 223 mg DCW (grams of initial dry substrate)−1 and, 47.6 OD500 (DCW grams)−1 h−1, respectively.  相似文献

Visual pigments, oil droplets and photoreceptor types in the retinas of four species of true chameleons have been examined by microspectrophotometry. The species occupy different photic environments: two species of Chamaeleo are from Madagascar and two species of Furcifer are from Africa and the Arabian Peninsula. In addition to double cones, four spectrally distinct classes of single cone were identified. No rod photoreceptors were observed. The visual pigments appear to be mixtures of rhodopsins and porphyropsins. Double cones contained a pale oil droplet in the principle member and both outer segments contained a long-wave-sensitive visual pigment with a spectral maximum between about 555 nm and 610 nm, depending on the rhodopsin/porphyropsin mixture. Long-wave-sensitive single cones contained a visual pigment spectrally identical to the double cones, but combined with a yellow oil droplet. The other three classes of single cone contained visual pigments with maxima at about 480–505, 440–450 and 375–385 nm, combined with yellow, clear and transparent oil droplets respectively. The latter two classes were sparsely distributed. The transmission of the lens and cornea of C. dilepis was measured and found to be transparent throughout the visible and near ultraviolet, with a cut off at about 350 nm.  相似文献

Summary A higher producer of ascospores and pigments, Monascus strain TTWMB 6042, was used to study regulation of pigment production by nutrients. An initial medium containing 4% glucose, 0.3% NH4NO3 (75 mm nitrogen) and inorganic salts was used. We found that the formation of red pigments in this strain, measured by optical density at 500 nm (OD500) was strongly stimulated by monosodium glutamate (MSG) as the sole nitrogen source. The choice of carbon source and an initial pH of pH 5.5 were also important. High concentrations of phosphate and MgSO4 were inhibitory to pigment production. A new chemically defined medium was devised containing 5% maltose, 75 mm MSG, phosphate and MgSO4 at lower concentrations plus other mineral salts, which yielded a tenfold increase in OD500 and a reversal of the pigment location from predominantly cell-bound, including both intracellular and surface-bound pigments, to mainly extracellular. The major chromophore of a mixture of fluorescent pigments produced by Pseudomonas aeruginosa ATCC 9027 had pH-dependent absorption, excitation, and emission spectra, such that two ionic forms existed in the ground state and three in the excited states. The pigments could complex with several metal ions to change fluorescence and absorption spectra. Although the pigments were separable into several components, spectra indicated that the same fluorescent chromophore was present in each component. Hydrolysis of the mixture of pigments gave amino acids which did not include alanine or lysine. These pigments must therefore differ from those described by other workers, even though similarities of the chromophores were evident from comparisons with data in the literature, and from comparisons of a hydrolytic product of the mixture of pigments, termed compound F, with the chromophore of the fluorescent pigment of Azotobacter vinelandii. Drastic hydrolysis of the latter chromophore also yielded compound F.  相似文献

The formation of soluble Monascus red pigments is strongly positively and negatively regulated by different amino acids. Leucine, valine, lysine, and methionine had strong negative effects on pigment formation. Leucine supported poor pigment formation when used as sole nitrogen source in fermentations, yet it neither repressed pigment synthase(s) nor inhibited its action. The new pigments derived from the hydrophobic leucine were more hydrophilic than the conventional red pigments (lacking an amino acid side-chain) and were extracellularly produced. Therefore, the low level of red pigments produced when leucine was the nitrogen source was not due to feed-back regulation by cell-bound leucine pigments. The negative effect of leucine was caused by enhanced decay of pigment synthase(s). In this study, the ability of safflower-isolated root cultures to produce yellow pigments was tested. Initially, the growth of isolated roots in static liquid medium was evaluated with different volumes of culture medium. A volume of 6 ml of medium per flask of 250 ml gave the best growth performance and, in this condition of culture, production of pigments from isolated roots treated or not by light has been determined by spectrophotometry (321 and 400 nm). Under these conditions, the production of yellow pigments amounted to 13.18 mg g−1 fresh weight and the light stimulated the synthesis of these pigments by isolated roots. Total yellow pigments of 24.12, 38.91 and 46.38 mg g−1 fresh weight was produced by the roots treated with 9, 13.5 and 18% (v/v) gas oil, respectively, representing high values of production. The pigments were released in large quantities in the medium. The increased synthesis of pigments as a result of gas oil treatment was accompanied by a reduction of the peroxidase activity of roots. Given the high production of yellow pigments, systems of isolated root culture could be considered for the study of a larger scale production of safflower pigments widely used for various industrial purposes.  相似文献

Dark-adapted retinal cones of goldfish were measured microspectrophotometrically. The three types of spectra so obtained were subjected to a new method of data analysis. In order of types blue (B), green (G), and red (R), the best estimates for lambdamax were 453, 533, and 620 nm; for main band half width, 6,700, 4,700, and 3,900 cm-1. The extinction spectra of 11-cis 3,4-dehydroretinal and those of the three goldfish pigments were progressively fitted with Gaussian curves starting at the low-energy end of their spectra. The sum of the oscillator strengths of the first three Gaussian components throughout the four spectra were found to have nearly equal magnitudes. Functional relationships that connect the Gaussian parameters were obtained by curve-fitting, enabling partial absorption spectra to be generated for any lambdamax. The generated curves predicted the half width and peak extinction of porphyropsin-type absorption spectra more accurately than previously existing nomograms or hypothesis. The epsilonmax values thus obtained were 28,500, 32,000, and 35,700 liter/mole cm for the B-, G-, and R-type goldfish pigments; these were found to be consistent with the experimental determinations of +/- 10% estimated accuracy.  相似文献

The mode of production of the brown pigments of Bacillus subtilis 168 L-4, pigments frequently used as phenotypic markers for sporulation in this organism, has been studied. A defined liquid medium which promoted maximal pigment formation was developed. Five brown components, which could be resolved by thin-layer chromatography, were produced in the culture broth. Removal of cells from the medium at the end of logarithmic growth did not alter the type or amount of the pigments formed, indicating that the cells excreted pigment precursors into the medium during growth. Pigment formation from the precursors was found to occur by an oxygen-requiring, base-dependent, Mn2+-requiring, nonenzymatic pathway. Pigment production was also stimulated by the presence of tyrosine and histidine in the medium. The increases in extracellular pH often associated with spore formation in B. subtilis might be the cause of the concomitant appearance of brown pigments.  相似文献

In many birds, red, orange and yellow feathers are coloured by carotenoid pigments, but parrots are an exception. For over a century, biochemists have known that parrots use an unusual set of pigments to produce their rainbow of plumage colours, but their biochemical identity has remained elusive until recently. Here, we use high-performance liquid chromatography to survey the pigments present in the red feathers of 44 species of parrots representing each of the three psittaciform families. We found that all species used the same suite of five polyenal lipochromes (or psittacofulvins) to colour their plumage red, indicating that this unique system of pigmentation is remarkably conserved evolutionarily in parrots. Species with redder feathers had higher concentrations of psittacofulvins in their plumage, but neither feather colouration nor historical relatedness predicted the ratios in which the different pigments appeared. These polyenes were absent from blood at the time when birds were replacing their colourful feathers, suggesting that parrots do not acquire red plumage pigments from the diet, but instead manufacture them endogenously at growing feathers.  相似文献

Visual pigments are G-protein-coupled receptors that provide a critical interface between organisms and their external environment. Natural selection has generated vertebrate pigments that absorb light from the far-UV (360 nm) to the deep red (630 nm) while using a single chromophore, in either the A1 (11- cis-retinal) or A2 (11- cis-3,4-dehydroretinal) form. The fact that a single chromophore can be manipulated to have an absorption maximum across such an extended spectral region is remarkable. The mechanisms of wavelength regulation remain to be fully revealed, and one of the least well-understood mechanisms is that associated with the deep red pigments. We investigate theoretically the hypothesis that deep red cone pigments select a 6- s- trans conformation of the retinal chromophore ring geometry. This conformation is in contrast to the 6- s- cis ring geometry observed in rhodopsin and, through model chromophore studies, the vast majority of visual pigments. Nomographic spectral analysis of 294 A1 and A2 cone pigment literature absorption maxima indicates that the selection of a 6- s- trans geometry red shifts M/LWS A1 pigments by approximately 1500 cm (-1) ( approximately 50 nm) and A2 pigments by approximately 2700 cm (-1) ( approximately 100 nm). The homology models of seven cone pigments indicate that the deep red cone pigments select 6- s- trans chromophore conformations primarily via electrostatic steering. Our results reveal that the generation of a 6- s- trans conformation not only achieves a significant red shift but also provides enhanced stability of the chromophore within the deep red cone pigment binding sites.  相似文献

Filamentous fungi have gained growing interest as sources of diverse pigmented secondary metabolites. Some specific polyketides from Ascomycetous species have demonstrated a wide range of industrial applications in food, cosmetic, textile, and in the design of pharmaceutical products. The formulation of recipes containing fungal polyketides has increased over recent years. Fusarium strains were proven useful to mankind in a variety of technologies. Nevertheless, there is still need of new isolates of Fusarium for use in emerging and already existing fields. In this article, we report the concomitant production of the bioactive red bikaverin along with two novel purple pigments by the phytopathogenic Fusarium oxysporum LCP531 strain isolated from soil. In literature, the production of purple pigment had only been described in cultures of Fusarium Fujikuroi, Fusarium verticillioides, and Fusarium graminearum. The production of these naphthoquinonic pigments, their distribution (either produced in mycelia or excreted in liquid medium) and their chemical profiles were investigated with respect to nutrient composition. The pigments were extracted by using a pressurized liquid extraction method, monitored by colorimetric analysis and characterized by HPLC-DAD chromatography. To our knowledge, this is the first report of these two novel wild-type purple naphtoquinones pigments along with bikaverin, where additionally, the culture conditions were put into perspective to optimize fermentation cultures and extraction process accordingly to the pigment/biomolecule desired. These colored naphthoquinones should be promising fungal functional compounds which could be expected to have a place of choice, along with other antibacterial, antifungal, antiviral, anticancer, and antineoplastic derivatives. © 2018 American Institute of Chemical Engineers Biotechnol. Summary Absorption spectra of acetone-soluble plant pigments were obtained from several sources, including the overlying waters, epibenthic layers and sediments of two New England estuaries, the remains of Zostera marina and its epiphytes, and pure cultures of marine diatoms. Comparisons of the absorption spectra and various absorbance ratios revealed no significant differences in the extracts of pigments sampled from the overlying waters and epibenthic layers; the extracts from both layers differed significantly, however, from the extracts of sedimentary pigments. Unacidified extracts of pigments from healthy diatom cultures and detritus rich in epiphytic diatoms resembled those of the overlying waters and epibenthic layers; acidified extracts from the above two sources and unacidified and acidified extract from non-living detritus resembled those of the sediments. Good correlations between field and laboratory studies are indicated, and the uses of absorbance ratios as ecological indicators are discussed.  相似文献

Cone visual pigments are visual opsins that are present in vertebrate cone photoreceptor cells and act as photoreceptor molecules responsible for photopic vision. Like the rod visual pigment rhodopsin, which is responsible for scotopic vision, cone visual pigments contain the chromophore 11-cis-retinal, which undergoes cis–trans isomerization resulting in the induction of conformational changes of the protein moiety to form a G protein-activating state. There are multiple types of cone visual pigments with different absorption maxima, which are the molecular basis of color discrimination in animals. Cone visual pigments form a phylogenetic sister group with non-visual opsin groups such as pinopsin, VA opsin, parapinopsin and parietopsin groups. Cone visual pigments diverged into four groups with different absorption maxima, and the rhodopsin group diverged from one of the four groups of cone visual pigments. The photochemical behavior of cone visual pigments is similar to that of pinopsin but considerably different from those of other non-visual opsins. G protein activation efficiency of cone visual pigments is also comparable to that of pinopsin but higher than that of the other non-visual opsins. Recent measurements with sufficient time-resolution demonstrated that G protein activation efficiency of cone visual pigments is lower than that of rhodopsin, which is one of the molecular bases for the lower amplification of cones compared to rods. In this review, the uniqueness of cone visual pigments is shown by comparison of their molecular properties with those of non-visual opsins and rhodopsin.
