Leukotrienes generated by 5-lipoxygenase (5-LOX)-catalyzed reaction are key regulators of inflammation. In ionophore-stimulated (A23187; 1-2.5 μM) human blood neutrophils or differentiated HL-60 cells, vitamin E forms differentially inhibited leukotriene B(4) (LTB(4)) with an IC(50) of 5-20 μM for γ-tocopherol, δ-tocopherol (δT), and γ-tocotrienol, but a much higher IC(50) for α-tocopherol. 13'-Carboxychromanol, a long-chain metabolite of δT, suppressed neutrophil- and HL-60 cell-generated LTB(4) with an IC(50) of 4-7 μM and potently inhibited human recombinant 5-LOX activity with an IC(50) of 0.5-1 μM. In contrast, vitamin E forms had no effect on human 5-LOX activity but impaired ionophore-induced intracellular calcium increase and calcium influx as well as the subsequent signaling including ERK1/2 phosphorylation and 5-LOX translocation from cytosol to the nucleus, a key event for 5-LOX activation. Further investigation showed that δT suppressed cytosolic Ca(2+) increase and/or LTB(4) formation triggered by ionophores, sphingosine 1-phosphate, and lysophosphatidic acid but not by fMLP or thapsigargin, whereas 13'-carboxychromanol decreased cellular production of LTB(4) regardless of different stimuli, consistent with its strong inhibition of the 5-LOX activity. These observations suggest that δT does not likely affect fMLP receptor-mediated signaling or store depletion-induced calcium entry. Instead, we found that δT prevented ionophore-caused cytoplasmic membrane disruption, which may account for its blocking of calcium influx. These activities by vitamin E forms and long-chain carboxychromanol provide potential molecular bases for the differential anti-inflammatory effects of vitamin E forms in vivo.  相似文献

Because of its potent antioxidant function and important role in clinical treatment, alpha-tocopherol (vitamin E) is a good starting point in the development of new synthetic antioxidants with improved properties. In this paper the first example of antioxidant-functionalized gold nanoparticles, Au@Trolox, was synthesized by self-assembly of thiol ligands derived from Trolox, a vitamin E analogue, on gold nanoparticles. DPPH* (2,2-diphenyl-1-picrylhydrazyl) radical scavenging experiments revealed that the rate constant for the reaction of Au@Trolox with DPPH* was about eight times greater than that for Trolox. The product analysis showed that both the quinonoid and the diepoxide forms were possible oxidized products of the chromanol group of Au@Trolox treated with DPPH* radical. No remarkable influence was found on the antioxidant activity of Au@Trolox when the coverage rate of the antioxidant group on the surface of the gold was varied. All our results proved that the assembly of chromanol groups on gold nanoparticles could efficiently enhance the activity of the vitamin E-derived antioxidant, which presents a potential new strategy for antioxidant design with novel perspectives in potential applications.  相似文献

Assessment of antioxidant activity by using different in vitro methods   总被引:13,自引:0,他引:13  
In this study, six common tests for measuring antioxidant activity were evaluated by comparing four antioxidants and applying them to beverages (tea and juices): Trolox equivalent antioxidant capacity assay (TEAC I-III assay), Total radical-trapping antioxidant parameter assay (TRAP assay), 2,2-diphenyl- l -picrylhydrazyl assay (DPPH assay), N , N -dimethyl- p -phenylendiamine assay (DMPD assay), Photochemiluminescence assay (PCL assay) and Ferric reducing ability of plasma assay (FRAP assay). The antioxidants included gallic acid representing the group of polyphenols, uric acid as the main antioxidant in human plasma, ascorbic acid as a vitamin widely spread in fruits and Trolox ® as water soluble vitamin E analogue. The six methods presented can be divided into two groups depending on the oxidising reagent. Five methods use organic radical producers (TEAC I-III, TRAP, DPPH, DMPD, PCL) and one method works with metal ions for oxidation (FRAP). Another difference between these tests is the reaction procedure. Three assays use the delay in oxidation and determine the lag phase as parameter for the antioxidant activity (TEAC I, TRAP, PCL). They determine the delay of radical generation as well as the ability to scavenge the radical. In contrast, the assays TEAC II and III, DPPH, DMPD and FRAP analyse the ability to reduce the radical cation (TEAC II and III, DPPH, DMPD) or the ferric ion (FRAP). The three tests acting by radical reduction use preformed radicals and determine the decrease in absorbance while the FRAP assay measures the formed ferrous ions by increased absorbance. Gallic acid was the strongest antioxidant in all tests with exception of the DMPD assay. In contrast, uric acid and ascorbic acid showed low activity in some assays. Most of the assays determine the antioxidant activity in the micromolar range needing minutes to hours. Only one assay (PCL) is able to analyse the antioxidant activity in the nanomolar range. Black currant juice showed highest antioxidant activity in all tests compared to tea, apple juice and tomato juice. Despite these differences, results of these in vitro assays give an idea of the protective efficacy of secondary plant products. It is strongly recommended to use at least two methods due to the differences between the test systems investigated.  相似文献

本文研究了芦笋提取液的抗氧化活性、对结肠癌细胞的细胞毒作用及对α-葡萄糖苷酶活性的抑制作用。通过研究其对DPPH自由基、羟基自由基及亚硝酸盐的清除能力来评价芦笋提取液的抗氧化活性,通过MTT法研究了芦笋提取液对结肠癌细胞株的细胞毒作用,通过α-葡萄糖苷酶体外抑制试验观察了芦笋提取液对大鼠小肠α-葡萄糖苷酶活性的影响。研究结果表明,芦笋提取液清除羟基自由基和亚硝酸盐的能力强于清除DPPH自由基的能力,一定浓度的芦笋提取液对结肠癌细胞株具有较强的细胞毒作用,并能够有效抑制大鼠小肠α-葡萄糖苷酶活性。  相似文献

水麻果多酚的提取纯化及其抗氧化、抗肿瘤活性作用   总被引:2,自引:0,他引:2  
为提取分离水麻果多酚,探索水麻果多酚的抗氧化及抗肿瘤能力。本研究通过单因素试验和正交试验优化水麻果多酚超声提取工艺,使用大孔树脂纯化水麻果多酚,通过测定水麻果多酚的总还原力以及清除·OH、DPPH·、ABTS·的能力来表征其抗氧化活性,以宫颈癌Hela细胞和肺癌A549细胞为抗肿瘤研究对象,测定水麻果多酚的抗肿瘤作用。在最优超声提取工艺条件下,即乙醇浓度为60%,料液比为1∶30,超声功率200 W,超声温度为70℃,提取时间为40 min,水麻果多酚得率为3. 29%,纯化产物总酚含量为40. 47 mg/100 mg,水麻果多酚的总还原力与维生素C相当,对·OH、DPPH·、ABTS·均具有显著清除作用,可抑制Hela细胞和A549细胞的生长增殖,并导致癌细胞产生大量活性氧,出现凋亡形态特征。该提取工艺简单、高效且多酚得率高,提取物经大孔树脂纯化后总多酚含量显著提高,且具有显著的抗氧化和抗肿瘤活性。  相似文献

以北方三省7种黑豆种子为原料,经过优化提取,对维生素E含量进行对比分析。结果显示:黑豆种子维生素E优化提取条件为;40mmol.L-1显色剂(α,α-联吡啶)、1mmol.L-1反应物(FeCl3)、6mmol.L-1中止反应物质(H3PO4),显色时间为6min;Ⅲ(襄垣)与Ⅳ(灵丘)维生素E含量较高,分别为95.7μg.g-1和70.2μg.g-1;Ⅰ(赤峰)与Ⅱ(齐齐哈尔)维生素E含量较少,分别为23.1μg.g-1和33.7μg.g-1。该优化提取方法简便、易于操作,且精确度高;山西省襄垣和灵丘两地黑豆VE含量较高。研究结果对该地黑豆产品深加工与利用有一定的指导作用。  相似文献

Dietary antioxidants interact in a dynamic fashion, including recycling and sparing one another, to decrease oxidative stress. Limited information is available regarding the interrelationships in vivo between quercetin and vitamin E. We investigated the antioxidant activity and metabolism of quercetin (Q) in 65 F-344 rats (n=13 per group) randomly assigned to the following vitamin E (VE)-replete and -deficient diets: (a) VE replete (30 mg alpha-tocopherol acetate/kg diet) control ad libitum (C-AL), (b) VE replete pair fed (C-PF), (c) VE replete+5.0 g Q/kg diet (R-VE+5Q), (d) VE deplete (<1 mg/kg total tocopherols)+5.0 g Q/kg diet (D-VE+5Q) and (e) D-VE. After 12 weeks, blood and tissue were collected for measurement of plasma vitamin E, quercetin and its metabolites, serum pyruvate kinase (PK), plasma protein carbonyls, malondialdehyde (MDA) and oxygen radical absorbance capacity. D-VE diets decreased serum alpha-tocopherol and increased PK activity in a time-dependent manner. The D-VE diet increased plasma protein carbonyls but did not affect MDA. Dietary quercetin supplementation increased quercetin and its metabolites in plasma and liver but did not affect D-VE-induced changes in plasma alpha-tocopherol, PK or protein carbonyls. Plasma isorhamnetin and its disposition in muscle were enhanced by the D-VE diet, as compared to the R-VE diet. Conversely, tamarixetin disposition in muscle was decreased by the D-VE diet. Thus, quercetin did not slow vitamin E decline in vivo; neither did it provide antioxidant activity in vitamin-E-depleted rats. However, vitamin E status appears to enhance the distribution of isorhamnetin into the circulation and its disposition in muscle.  相似文献

3-Aryl-4-formylsydnone 4'-phenylthiosemicarbazones (3a-d) and 3-aryl-4-formylsydnone thiosemicarbazones (3e-h), which are precursors of 3-aryl-4-heterocyclic sydnones, are prepared by the condensation of 3-aryl-4-formylsydnones (1a-d) with 4'-phenylthiosemicarbazide (2a) and thiosemicarbazide (2b), respectively. The thiosemicarbazones 3 reacted with cyclic reagents such as ethyl chloroacetate (4a), ethyl 2-chloroacetoacetate (4b) and 2-bromoacetophenone (4c) to produce heterocyclic substituted sydnone derivatives 5-7 that possess 4-oxo-thiazolidine and thiazoline groups. The antioxidant activity of synthesized compounds 5a-7h was evaluated. Among these compounds, 4-methyl-2-[(3-arylsydnon-4-yl-methylene)hydrazono]-2,3-dihydro-thiazole-5-carboxylic acid ethyl ester (6e-h) and 4-phenyl-2-[(3-arylsydnon-4-yl-methylene)hydrazono]-2,3-dihydro-thiazoles (7e-h) exhibit the potent DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity, comparable to that of vitamin E.  相似文献

Twenty kinds of cyclic dipeptides containing l-leucine were synthesized, and their antioxidant activity against ·OH and O(2)(·-) was investigated. Compounds possessing polar amino acid residues, such as Asp, Cys, Glu, Lys, Pro, Ser, and Trp, exhibited higher antioxidant activity against ·OH than vitamin E. Twenty samples of two caper species were collected from various natural habitats of Calabria (South Italy). A sample program was designed in order to cover all taxa and morphotypes present in the studied area, trying to correlate ecological conditions, phytochemical content, and biological activities. These species were characterized through the detection, isolation, and capillary GC-GC/MS quantitative evaluation of chemical markers (phytosterols and vitamin E). The antioxidant activity of MeOH caper extracts was assayed using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and β-carotene bleaching tests. The chemical investigation showed a different composition according to the species and the populations. The best free radical (DPPH) scavenging activity was exerted by Capparis orientalis sample C4 (collected near Copanello village, granodiorite sea cliff) and Capparis sicula ssp. sicula sample C5 (collected near Tarsia village, clay soil). Sample C2 (Galatrella Valley, clay soil) showed the highest inhibition of linoleic acid oxidation with more efficacy than propyl gallate, the reference drug.  相似文献

Antioxidant activity of d-α-, dl-β-, d-γ- and d-δ-tocopherol was investigated with fatty acid methylester of milk fat from which unsaponifiable matter had been removed. Autoxidation was carried out at 50°C and its degree was indicated by peroxide value, α- or β-Tocopherol was more effective at lower concentrations (0.003 and 0.01%) than at higher concentrations (0.05, 0.1 and 0.5%). The antioxidant activity of γ- and δ-tocopherol was increased with the increase of tocopherol concentration within the range of 0.001 to 0.5%. The order of antioxidant activity of these tocopherols, which was compared in terms of the time to reach 30 meq of peroxide value, varied with the concentration; γ > β > δ > α at 0.001%, α > γ > β > δ at 0.003%, γ > δ > β > α at 0.01%, and δ > γ > β > α at the concentrations more than 0.05%. α-Tocopherol at the concentration of 0.003%, which corresponded to the concentration in original milk fat, was more effective than other tocopherols at the same concentration and α-tocopherol at other concentrations. Synergism due to the combination of β-, γ-, or δ-tocopherol with 0.003% of α-tocopherol was not observed.  相似文献

Abstract

Objectives

The aim of this study was to determine the relationship between the antioxidant profile of anesthetics and its relation to total antioxidant capacity (TAC) of plasma in children who underwent tourniquet-induced ischemia-reperfusion (IR) injury during extremity operations.

Methods

Children were randomized into three groups: general inhalational anesthesia with sevoflurane (group S), total intravenous anesthesia (TIVA) with propofol (group T), and regional anesthesia (group R). Venous blood samples were obtained before peripheral nerve block and induction of general anesthesia (baseline), 1 minute before tourniquet release (BTR), and 5 and 20 minutes after tourniquet release (ATR). Plasma TAC as well as antioxidant potential of propofol, thiopental, and bupivacaine were measured using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay.

Results

Plasma TAC in group T was increased significantly at 20 minutes ATR in comparison with basal and BTR values, and also was significantly higher in comparison with plasma TAC in groups S and R measured at the same time point. The radical scavenging activity of anesthetics in vitro indicated that only propofol possessed a significant antioxidative activity in the reaction with DPPH radical in comparison with thiopental and bupivacaine.

Discussion

These data confirm that TIVA with propofol attenuates oxidative stress related to tourniquet-induced ischaemia-reperfusion injury in children.  相似文献

Approaching living systems, aqueous solutions are appropriate to characterize antioxidants, whereas the frequently used standard 1,1-diphenyl-2-picrylhydrazyl (DPPH) is insoluble in water. Therefore, mixed water-ethanol solvents were investigated using the electron paramagnetic resonance (EPR) spectroscopy. Two forms of DPPH were identified: at higher ethanol ratios a quintet spectrum characteristic of solutions, and at lower ratios, a singlet spectrum typical for solid DPPH, were found. Mixed solvents with 0-50% (v/v) water reproduced the same antioxidant equivalent points well and the reaction rate between DPPH and the antioxidant may increase considerably with increasing water ratios, as demonstrated using vitamin E as an antioxidant. But at still higher water ratios (70-90% (v/v)) the antioxidant activities dropped, since a part of the DPPH in the aggregated form does not react sufficiently with the antioxidants. Characteristics of the most common antioxidants were determined in ethanol or its 50% (v/v) aqueous solution.  相似文献

This study aimed to investigate whether treatments with vitamin E, L-carnitine and melatonin can protect against CCl4 and diabetes-induced hepatic oxidative stress. Hepatic oxidative stress was performed in rats through 50% v/v carbon tetrachloride (CCl4) (1 ml/kg/3days, i.p.), and through diabetes mellitus induced by streptozotocin (STZ) (40 mg/kg, i.p.). Vitamin E (100 mg/kg/day, i.p), L-carnitine (300 mg/kg/day, i.p.) and melatonin (10 mg/kg/day, i.p.) were injected for a period of 6 weeks. Thereafter, changes in serum glucose level, liver function tests, hepatic malondialdehyde (MDA) content, hepatic reduced glutathione (GSH) content, hepatic superoxide dismutase (SOD) activity, and serum total antioxidant capacity (TAC) level were evaluated. In CCl4-induced liver fibrosis, the efficacy order was melatonin > L-carnitine > vitamin E, while in STZ-induced diabetes, the efficacy order was vitamin E ≥ melatonin > L-carnitine. In conclusion, these data indicate that low dose of melatonin is more effective than high doses of vitamin E and L-carnitine in reducing hepatic oxidative stress induced by CCl4 and diabetes. Moreover, the potent effect of vitamin E in ameliorating diabetes can be linked not only to the antioxidant actions, but also to the superior effect in reducing diabetes-induced hyperglycaemia. Meanwhile, potency of L-carnitine was nearly the same in CCl4 and diabetes-induced liver damage.  相似文献

AimsBreast cancer is the most common malignancy among women, with an age-specific incidence profile. During the last years much evidence has accumulated demonstrating the anticancer activity of tocotrienols (T3), a subfamily of natural vitamin E (VE). In this study, mouse and human breast cancer cells (with or without HER-2/neu oncogene overexpression) were used to investigate the anticancer effect of α-, γ-, and δ-tocotrienols in comparison with α-tocopheryl succinate (α-TOS), a synthetic derivative with widely recognized anticancer properties.Main methodsHuman and mouse breast cancer cell lines were used. The effect of VE compounds on cell viability was investigated using Alamar Blue assay. Apoptosis was assessed by propidium iodide and JC-1 staining. Expression of senescence-associated markers was evaluated by RT-PCR and Western blot analysis was used to examine the changes in the expression levels of HER-2/neu.Key findingsγ- and δ-Τ3 reduced cell viability with IC50 values of less than half those of α-T3 and α-TOS. γ- and δ-Τ3, and α-TOS to a lesser extent, induced apoptosis possibly via the mitochondrial pathway, and the expression of senescent-like growth arrest markers as p53, p21, and p16. Both α-TOS and tocotrienols downregulated HER-2/neu in tumor cells overexpressing this oncogene, but this effect did not seem to be essential for the antitumor activity of these compounds.SignificanceWe demonstrate that in HER-2/neu breast cancer cells, the non-alpha form of T3 shows stronger anticancer activity than the synthetic VE-derivative α-TOS and this effect occurs independently from the inhibition of HER-2/neu oncogene expression.  相似文献

Lycopene is the major carotenoid in tomatoes. Tomatoes contain a matrix of many bioactive components, including vitamin C, vitamin E, other carotenoids (a-, beta-, gamma- carotene, lutein), and flavonoids. Their synergistic interactions, when used in combination, may be responsible for the observed beneficial effects of tomato-based products. This study investigated the synergistic antioxidant activity of lycopene in combination with beta-carotene, vitamin E, and lutein. A liposome system was used to test the synergistic antioxidant activity. The carotenoid mixtures were more efficient in protecting liposome from oxidation than the individual carotenoid. Mixtures of lycopene and vitamin E appear to have the greatest synergistic antioxidant activity.  相似文献

Ante- and postpartum antioxidative/oxidative profiles in blood plasma of cows with and without retained placental membranes (RFM) were investigated. Twenty-two healthy pregnant cows were included in the study. Seven animals out of 22 suffered from RFM. Blood samples were obtained at 4, 3, 2, 1 weeks and 5 days antepartum (a.p.), at parturition and 1, 3, and 5 weeks postpartum. The following antioxidative parameters were measured using spectrophotometric methods: total antioxidant activity (TAC), β-carotene, vitamin A, vitamin C, and ceruloplasmin. The oxidative profile was based on the content of intermediates and end products of lipid and protein peroxidative processes which were measured by spectrophotometric and spectrofluorimetric methods. The examined parameters revealed a dynamic profile within the experimental period. The highest antioxidant and oxidant activity was noted at 2 and 1 week a.p. with a drop towards parturition suggesting the presence of oxidative stress during this time period and an apparent appropriate metabolic response of the macroorganism. Except for TAC and vitamin A, the contents of oxidative and antioxidative blood constituents did not differ between cows with and without RFM. A TAC and vitamin A by time interaction indicated higher antepartal concentrations of TAC and vitamin A in cows without RFM than in cows with RFM suggesting a possible role of antioxidative/oxidative imbalances in the aetiology of RFM.  相似文献

以初始体质量(7.270.40) g的青鱼为研究对象, 采用维生素E(VE)有效含量分别为14.36(对照组)、25.14、37.66、62.97、113.92和210.45 mg/kg 6种等氮等能的实验饲料, 饲养青鱼幼鱼8周后, 根据生长情况选取对照组、62.97和210.45 mg/kg VE组进行24h氨氮胁迫(20 mg/L), 研究VE对青鱼幼鱼生长、免疫及抗氨氮胁迫能力的影响。结果表明: 以特定生长率为指标, 折线模型分析表明青鱼有效维生素E需要量为45.00 mg/kg。肌肉、肝脏和血清VE含量与饲料中VE含量呈明显正相关, 当饲料VE含量超过113.92 mg/kg时, 肌肉和肝脏VE含量均达到饱和。VE对鳃丝Na+/K+-ATP酶活性(NKA)和血清皮质醇(COR)无显著影响, 但随着饲料VE含量的升高, 过氧化氢酶(CAT)和总超氧化物歧化酶活性(T-SOD)呈上升趋势, 丙二醛含量(MDA)呈下降趋势。氨氮胁迫对各处理组肌肉VE含量和血清CAT活性无影响, 但肝脏VE含量均显著降低(P0.05), 且62.97和210.4 5 mg/kg VE组血清VE水平有所升高。在胁迫后, 对照组血清T-SOD、鳃丝NKA活性显著降低, 皮质醇含量显著增加(P0.05)。与对照组相比, 62.97和210.45 mg/kg VE组T-SOD、NKA活性和皮质醇含量在胁迫前后无显著变化。各处理组MDA含量在胁迫后虽均显著升高, 但210.45 mg/kg VE组在胁迫后MDA含量仍显著低于对照组(P0.05)。以上结果说明, 青鱼幼鱼获得最大生长的有效维生素E需求量为45.00 mg/kg, 且较高VE能有效提高青鱼机体免疫力, 缓解氨氮胁迫对青鱼机体的负面影响。    相似文献
