Rhopalosiphum padi L. (Homoptera: Aphididae) is sensitive to loline alkaloids present in tall fescue, Festuca arundinacea Shreb., infected with the endophytic fungus, Acremonium coenophialum Morgan-Jones & Gams. Aphid survival was higher on endophyte-free plants regardless of plant age after germination or age of regrowth tissue after clipping. Survival of aphids on endophyte-infected grass was lower on young tissue but increased as plants aged, although it never reached the same level on endophyte-free plants. Both N-formyl and N-acetyl loline increased as uncut or regrowth tissue aged; however, this was influenced by the age of the plant at the initial cut and the clipping frequency. Although even small amounts of loline cause high aphid mortality, the aphids are able to survive on endophyte-infected plants if the tillers have senescing leaves which contain lower amounts of loline. Preference for senescing leaves may help R. padi avoid plant parts containing high amounts of toxic allelochemicals, thus contributing to higher numbers of aphids on older, endophyte-infected plants.  相似文献

Ergot alkaloids in endophyte-infected (Neotyphodium coenophialum) tall fescue (Lolium arundinaceum) have been shown to cause a reduction in blood flow to the rumen epithelium as well as a decrease in volatile fatty acids (VFA) absorption from the washed rumen of steers. Previous data also indicates that incubating an extract of endophyte-infected tall fescue seed causes an increase in the amount of VFA absorbed per unit of blood flow, which could result from an alteration in the absorptive or barrier function of the rumen epithelium. An experiment was conducted to determine the acute effects of an endophyte-infected tall fescue seed extract (EXT) on total, passive or facilitated acetate and butyrate flux across the isolated bovine rumen as well as the barrier function measured by inulin flux and tissue conductance (Gt). Flux of ergovaline across the rumen epithelium was also evaluated. Rumen tissue from the caudal dorsal sac of Holstein steers (n=6), fed a common diet, was collected and isolated shortly after slaughter and mounted between two halves of Ussing chambers. In vitro treatments included vehicle control (80% methanol, 0.5% of total volume), Low EXT (50 ng ergovaline/ml) and High EXT (250 ng ergovaline/ml). Results indicate that there is no effect of acute exposure to ergot alkaloids on total, passive or facilitated flux of acetate or butyrate across the isolate bovine rumen epithelium (P>0.51). Inulin flux (P=0.16) and Gt (P>0.17) were not affected by EXT treatment, indicating no alteration in barrier function due to acute ergot alkaloid exposure. Ergovaline was detected in the serosal buffer of the High EXT treatment indicating that the flux rate is ~0.25 to 0.44 ng/cm2 per hour. Data indicate that specific pathways for VFA absorption and barrier function of the rumen epithelium are not affected by acute exposure to ergot alkaloids from tall fescue at the concentrations tested. Ergovaline has the potential to be absorbed from the rumen of cattle that could contribute to reduced blood flow and motility and lead to reduced growth rates of cattle.  相似文献

Caesalpinia crista seed extracts were investigated in the laboratory against Helicoverpa armigera (Hubner). The extracts exhibited powerful antifeedant and growth disruption activity. The toxic symptoms of the poisoned larvae included reduction in weight gain and mortality of larvae and pupae, larval-pupal intermediates and malformed adults. Maximum anti-feedance is caused by methanol extract (AI50=0.0186%), followed by hexane extract (0.0212%), ethyl acetate extract (0.0416%), butanol extract (0.0767%) and aqueous extract (0.2199%). The larval mortality ranged from 10.00 to 70.00% in different extracts. The 50% larval growth inhibition recorded at 3DAT was statistically at par by methanol and hexane extract. The percent 150 values for inhibiting normal adult emergence were 0.0287, 0.0325, 0.0485, 0.0977 and 0.0547% for methanol, hexane, ethyl acetate, aqueous and butanol extract. The biosafety evaluation of these extracts carried out against the predator, Coccinella septumpunctata showed no mortality of the adults till nine days after treatment. Though the observation taken at 10 DAT, showed slight mortality of adults by methanol extract at both 5.0 and 1.0% concentration.  相似文献

Two experiments were conducted to assess preference by sheep for endophyte-infected tall fescue growing in monoculture at least 5 m away from alfalfa (fescue-middle (FM)) over endophyte-infected tall fescue growing adjacent (0.2 to 1 m; fescue–alfalfa (FA)) to alfalfa (FA), and the effect of legume scent on preference for endophyte-infected tall fescue. In Experiment 1, 10 six-month-old lambs were offered for 12 days a choice of freshly harvested FA and FM. On days 13 and 14, lambs were offered the same choice, except cages (to allow access only to scent) containing freshly harvested alfalfa were put in the feeders containing FA, whereas cages containing freshly harvested FM were included with the feeders containing FM. Forage intake was measured 1 h after feeding and at three consecutive 2-h intervals thereafter. FA contained greater (P<0.002) concentrations of the alkaloid ergovaline (360±27 ppm) and CP (8±0.4%) than FM (219±27 ppm and 6±0.4%, respectively). Lambs preferred (P<0.05) FA to FM during the 1st hour of feeding, but the differences became smaller and disappeared in later feeding periods (P<0.005). Lambs offered FA with alfalfa scent or FM with FM scent preferred (P<0.05) FA but only on the 2nd day. In Experiment 2, 10 six-month-old lambs were offered a choice of FM with cages (to allow access only to scent) containing freshly harvested alfalfa or FM for 8 days. During the following 4 days, FM in the cages was replaced with freshly harvested sainfoin. Preference was greater (P<0.05) for FM offered with alfalfa scent than for FM offered with FM scent only on days 4 and 8. When lambs were offered FM with alfalfa or sainfoin in cages, they preferred (P<0.05) tall fescue with sainfoin scent over fescue with alfalfa scent, but intake was variable across hours and days (P<0.001). It is concluded that (1) lambs adjusted their intake of and preference for FA and FM over successive feeding bouts within each day, likely owing to an attempt to balance intakes of nutrients and alkaloids and (2) olfactory cues influenced preference, but to a lesser extent than nutrients and alkaloids in endophyte-infected tall fescue.  相似文献

Acremonium coenophialum, a fungal endosymbiont in tall fescue, is responsible for the production of alkaloid toxins. Animals grazing endophyte-infected tall fescue often show toxicosis. In marginal environments, the endophyte is important for long-term survival of tall fescue. Few differences in endophyte isolates from individual tall fescue plants have been reported. To aid development of a toxicosis-free tall fescue, it is important to identify differences in endophyte isolates. This report describes variation in nitrogen utilization in a defined culture medium by endophyte isolates from Kentucky-31 tall fescue. Overall, the best nitrogen sources for dry weight production of mycelium were proline and potassium nitrate. Thirty-four isolates grown on agar-solidified defined media with single nitrogen sources showed variation in nitrogen utilization. Fifty percent of the isolates were unable to utilize two or more amino acids. Manipulation of endophyte variation could lead to development of a toxicosis-free tall fescue cultivar.  相似文献

1. Elevated CO2 can alter plant physiology and morphology, and these changes are expected to impact diet quality for insect herbivores. While the plastic responses of insect herbivores have been well studied, less is known about the propensity of insects to adapt to such changes. Genetic variation in insect responses to elevated CO2 and genetic interactions between insects and their host plants may exist and provide the necessary raw material for adaptation. 2. We used clonal lines of Rhopalosiphum padi (L.) aphids to examine genotype‐specific responses to elevated CO2. We used the host plant Schedonorus arundinaceus (tall fescue; Schreb), which is capable of asexual reproduction, to investigate host plant genotype‐specific effects and possible host plant‐by‐insect genotype interactions. The abundance and density of three R. padi genotypes on three tall fescue genotypes under three concentrations of CO2 (ambient, 700, and 1000 ppm) in a controlled greenhouse environment were examined. 3. Aphid abundance decreased in the 700 ppm CO2 concentration, but increased in the 1000 ppm concentration relative to ambient. The effect of CO2 on aphid density was dependent on host plant genotype; the density of aphids in high CO2 decreased for two plant genotypes but was unchanged in one. No interaction between aphid genotype and elevated CO2 was found, nor did we find significant genotype‐by‐genotype interactions. 4. This study suggests that the density of R. padi aphids feeding on tall fescue may decrease under elevated CO2 for some plant genotypes. The likely impact of genotype‐specific responses on future changes in the genetic structure of plant and insect populations is discussed.  相似文献

The influence of grazing endophyte-infected tall fescue on endometrial cup formation and function, progestogen production, and embryonic and fetal development were examined in pregnant mares between Day 21 and Day 300 of gestation. Total immunoreactive progestogens and equine chorionic gonadotropin (eCG) concentrations were compared between untreated controls (endophyte-free, n = 12) and treated mares (endophyte-infected, n = 12). There were no differences in endometrial cup formation or function, as determined by eCG concentrations at Days 45, 60, 75, 90 and 120 (P > 0.05) between mares grazing endophyte-infected and endophyte-free tall fescue. Mares grazing the endophyte-infected tall fescue had lower total progestogen concentrations (P < 0.01) from Days 90 to 120 than the mares grazing endophyte-free tall fescue. Embryonic development based on mean vesicle height was not affected by endophyte exposure. No pregnancies were lost by mares in either treatment group during the trial period. The results of this study indicate that grazing endophyte-infected tall fescue between Day 21 and Day 300 does not alter endometrial cup formation and function, or result in increased pregnancy losses during this period. Lower progestogen concentrations between Days 90 and 120 with exposure to endophyte-infected tall fescue could reflect decreased luteal progesterone production.  相似文献

Tall fescue (Festuca arundinacea Schreb.), a highly competitive European grass that invades US grasslands, is reportedly allelopathic to many agronomic plants, but its ability to inhibit the germination or growth of native grassland plants is unknown. In three factorial glasshouse experiments, we tested the potential allelopathic effects of endophyte-infected (E+) and uninfected (E−) tall fescue on native grasses and forbs from Midwestern tallgrass prairies. Relative to a water control, at least one extract made from ground seed, or ground whole plant tissue of E+ or E− tall fescue reduced the germination of 10 of 11 species in petri dishes. In addition, the emergence of two native grasses in potting soil was lower when sown with E+ and E− tall fescue seedlings than when sown with seeds of conspecifics or tall fescue. However, when seeds of 13 prairie species were sown in sterilized, field-collected soil and given water or one of the four tall fescue extracts daily, seedling emergence was lower in one extract relative to water for only one species, and subsequent height growth did not differ among treatments for any species. We conclude that if tall fescue is allelopathic, its inhibitory effects on the germination and seedling growth of native prairie plants are limited, irrespective of endophyte infection. On the other hand, the apparent inability of these plants to detect tall fescue in field soil could hinder prairie restoration efforts if germination near this strong competitor confers fitness consequences. We propose that lack of chemical recognition may be common among resident and recently introduced non-indigenous plants because of temporally limited ecological interactions, and offer a view that challenges the existing allelopathy paradigm. Lastly, we suggest that tall fescue removal will have immediate benefits to the establishment of native grassland plants.  相似文献

Neotyphodium endophytes are assumed to have mutualistic relationship with their grass hosts, mainly resulting from mycotoxin production increasing plant resistance to herbivores by the fungus that subsists on the plant. To study importance of often ignored environmental effects on these associations, we performed a greenhouse experiment to examine the significance of endophyte infection and nutrient availability for bird-cherry aphid (Rhopalosiphum padi) performance on meadow fescue (Lolium pratense). Naturally endophyte-infected (E+), uninfected (E–), or manipulatively endophyte-free (ME–) half-sib families of meadow fescue were grown on two soil nutrient levels. Endophyte infection reduced aphid performance in general. However, to our knowledge, this is the first study to demonstrate experimentally that herbivore performance decreases on E+ host plants with increasing availability of nutrients in soils. Potential improvement in herbivore performance in high nutrient soils and decreased plant performance in low nutrient soils in ME– plants, compared to E– and E+ plants, suggests that loss of endophyte infection after long coevolutionary relationship may be critical to plant fitness.  相似文献

Effects of methanol extracts of Xanthium strumarium on different cancer cell lines and on the mortality rates of Aedes caspius, Culex pipiens (Diptera: Culicidae) were investigated. Among the cell lines tested, the Jurkat cell line was the most sensitive to the methanol extract and ethyl acetate fraction, with reported LC50 values of 50.18 and 48.73 μg/ml respectively. Conversely, methanol extracts were not that toxic to the A549 cell line though the toxicity increased on further purification. The percentage of growth inhibition was dose dependent for the methanol extract and ethyl acetate fraction. The ethyl acetate fraction showed higher toxicity to all cell lines tested when compared to the methanol extract. The results showed that methanol extracts of plant seeds caused 100% mortality of mosquito larvae at a concentration of 1000 μg/ml after 24 h of treatment. The LC50 and LC90 values of X. strumarium were found to be 531.07 and 905.95 μg/ml against Ae. caspius and 502.32 and 867.63 μg/ml against Cx. Pipiens, respectively. From the investigations, it was concluded that the crude extract of X. strumarium showed a weak potential for controlling the larval instars of Ae. caspius and Cx. pipiens. However, on further purification the extract lost the larvicidal activity. The ethyl acetate fraction showed higher toxicity to all cell lines tested when compared to the methanol extract. The ethyl acetate fraction investigated in this study appears to have a weak larvicidal activity but a promising cytotoxic activity. Future studies will include purification and investigation in further detail of the action of X. strumarium on Cancer Cell Lines and mosquitoes.  相似文献

The symbiosis between grasses and endophytic fungi is a common phenomenon and can affect herbivore performance through acquired, chemical plant defence by fungal alkaloids. In laboratory experiments, two species of common grass aphids, Rhopalosiphum padi and Metopolophium dirhodum were tested, in a population experiment (on four plant cultivars) and individually (on one plant cultivar) for the effects of the endophyte, Neotyphodium lolii, that forms symbiotic associations with perennial ryegrass Lolium perenne. In the population experiment that lasted for four aphid generations both aphid species showed decreased population sizes when feeding on each of the four endophyte-infected cultivars. Individuals of R. padi tested individually showed reduced adult life span and fecundity when feeding on infected plants. Individuals of M. dirhodum showed no response in any of the traits measured. This suggests that R. padi individuals are more sensitive to endophyte infection than M. dirhodum individuals. However, all infected grass cultivars reduced population sizes of both aphid species over four generations. Therefore, fungal endophytes can reduce populations of aphid herbivores independent of plant cultivars.  相似文献

Fungal endophytes of grasses are often included in agricultural management and in ecological studies of natural grass populations. In European agriculture and ecological studies, however, grass endophytes are largely ignored. In this study, we determined endophyte infection frequencies of 13 European cultivars and 49 wild tall fescue (Schedonorus phoenix) populations in Northern Europe. We then examined seed production and seed predation of endophyte-infected (E+) and endophyte-free (E?) tall fescue (in wild grass populations and in a field experiment) and meadow fescue (Schedonorus pratensis; in a field experiment only). Endophytes were detected in only one of the 13 cultivars. In contrast, >90% of wild tall fescue plants harbored endophytes in 45 wild populations but were absent in three inland populations in Estonia. In three wild tall fescue study sites, 17%, 22%, and 56% of the seeds were preyed upon by the cocksfoot moth. Endophyte infection did not affect seed mass of tall fescue in the field experiment. However, seed predation was lower in E+ than E? grasses in the two tall fescue populations with higher predation rates. For meadow fescue, the mean number of seeds from E+ plants was higher than E? plants, but E? and E+ seeds had equal rates of predation by the moth. Our results suggest that the effects of grass endophytes on seed production and cocksfoot moth seed predation vary considerably among grass species, and the effects may depend on herbivore pressure and other environmental conditions.  相似文献

The presence of the endophytic fungusAcremonium coenophialum Morgan-Jones et Gams in tall fescue (Festuca arundinacea Schreb.) induces toxicity when this grass is grazed by cattle; however, there is evidence that removing the endophyte reduces the stand vigor and longevity of fescue. A field trial was conducted to determine the effects of water supply and the presence of the endophytic fungus on plant growth, drought tolerance, and soil nematode populations in Kentucky 31 tall fescue. The design included two factors, level of endophyte infection (0 and 75%) and irrigation regime (none, low, and high). Where water deficits occurred, herbage yield and leaf area were lower, and percentage dead tissue and canopy minus air temperature were greater in endophyte-free compared with endophyte-infected fescue. Soil populations ofPratylenchus scribneri andTylenchorhynchus acutus were substantially higher in the noninfected than in the endophyte-infected plots. The endophyte apparently confers drought tolerance to Kentucky 31 tall fescue, and this effect may be at least partially mediated through enhanced resistance to soil-borne nematodes.Published with the approval of the Director of the Ark. In folk medicine, Adhatoda vasica Ness. (Acanthaceae) is used to treat asthma and cough. The leaves of A.vasica were powdered and extracted with hexane, ethyl acetate and methanol. The hexane extract showed 97% reduction in colony-forming units (CFU) at 100 μg/ml. The hexane extract was subjected to column chromatography. Two natural compounds, vasicine acetate and 2-acetyl benzylamine, were isolated from it. They were bioassayed against Mycobacterium tuberculosis. The two compounds showed strong antimycobacterial activity. Vasicine acetate and 2-acetyl benzylamine isolated from hexane extract of A.vasica leaves, significantly inhibited M. tuberculosis and one multi-drug-resistant (MDR) strain and one sensitive strain at 200 and 50 μg/ml, respectively. Our study demonstrated that both the compounds, vasicine acetate and 2-acetyl benzylamine, could be evaluated further for developing a drug to control M. tuberculosis.  相似文献

Electrical penetration graph recordings using direct current (DC-EPGs) were used to analyze aspects of the probing behavior of cowpea aphid,Aphis craccivora Koch, on intact plants and on hexane, ethyl acetate, and methanol extracts of leaves of aphid-resistant (ICV-12) and aphid-susceptible (ICV-1) cultivars of cowpeaVigna unguiculata (L.) Walp. In one set of experiments, recordings were done on plants with or without parafilm wrapping, or on plants painted with raw leaf juice and extracts of the two cultivars. In another study, recordings were done on leaf extracts homogenized in water or in 0.5M sucrose solution and then placed in parafilm membrane sachets. Electrodes were inserted into soil mix for the experiments on potted plants or into extract fractions and raw juice for the membrane feeding experiments on leaf extracts in parafilm sachets. Waveform signals were recorded from resistance fluctuations from interactions between aphids and substrates, and electromotive forces generated within each preparation. ICV-12 plants with or without parafilm wrapping, and ethyl acetate extracts and raw juice of that cultivar significantly (P≤0.05) reduced stylet penetration behavior. Thus, antixenosis as manifested by disruption of aphid stylet activity on host substrates, appeared to be a governing modality of aphid resistance in ICV-12.  相似文献

1. Female Sprague-Dawley rats (Rattus norvegicus) were randomly assigned to various dietary treatments containing: (1) 100% Purina rodent chow, ad libitum; (2) same as 1, but restricted to daily intake of 7; (3) 50% rodent chow (w/w) and 50% endophyte-free tall fescue (Festuca arundinacea) seed; (4) same as 3, but restricted to intake of 5; (5) 50% rodent chow, 25% endophyte-free tall fescue seed and 25% endophyte-infected (Acremonium coenophialum) tall fescue seed; (6) 50% rodent chow, 12.5% endophyte-free and 37.5% endophyte-infected tall fescue seed; and (7) 50% rodent chow and 50% endophyte infected tall fescue seed. 2. Average daily feed intakes and average daily weight gains decreased with higher levels of endophyte infected seed. 3. Frequency of litter production was affected by all endophyte-infected containing diets. 4. Conception was reduced only in dietary treatment (7). 5. Litter weights, number of pups per litter and weight per pup were proportionally reduced as higher levels of infected seed were incorporated in the ingested diets.  相似文献

In the present study, in vitro anticariogenic potential of ethyl acetate, hexane and methanol and aqueous extracts of plant leaves of Eucalyptus globules Labill. were evaluated by using four cariogenic bacteria, Lactobacillus acidophilus, Lactobacillus casei, Staphylococcus aureus and Streptococcus mutans. Agar well diffusion method and minimum inhibitory concentration (MIC) were used for this purpose. The ethyl acetate extracted fraction of plant leaves showed good inhibitory effects against all selected bacteria. In Eucalyptus globules, hexane and ethyl acetate extracts found highly effective against, Lactobacillus acidophilus with MIC value of 0.031 and 0.062 mg/mL, respectively. Qualitative phytochemical investigation of above extracts showed the presence of alkaloids, phenolic compounds, steroids, cardiac glycosides and terpenes. Based on the MIC value and bioautography, ethyl acetate of plant leaf was selected for further study. Further investigation on the structure elucidation of the bioactive compound using IR, GC-MS and NMR techniques revealed the presence of alpha-farnesene, a sesquiterpene. Eucalyptus globules plant leaf extracts have great potential as anticariogenic agents that may be useful in the treatment of oral disease.  相似文献

Human alteration of symbiont genetics among aboveground endophytic Epichloë coenophiala strains within tall fescue (Schedonorus arundinaceus) has led to widespread deployment of novel grass-endophyte combinations, yet little is known about their ecological consequences. In this study, clone pairs (endophyte-infected, endophyte-free) of two tall fescue genotypes received factorial combinations of increased temperature (+3 °C) and precipitation (+30% long-term annual mean) for 2 yr. We measured root arbuscular mycorrhizal fungi (AMF), dark septate endophyte (DSE) colonization, and soil AMF extraradical hyphae (ERH) length. We hypothesized that genetically distinct grass-E. coenophiala associations would differentially affect belowground fungi, and that these relationships would be climate-sensitive. Tall fescue genotype, endophyte presence, and climate treatment interactions affected AMF arbuscules, vesicles, and ERH. DSE decreased with E. coenophiala presence but increased with warming. Genetically distinct tall fescue-E. coenophiala associations may have divergent long-term impacts on other host-symbiont interactions, potentially impacting ecosystem function and response to climate change.  相似文献

Detection of prey DNA-remains in arthropod predators by polymerase chain reaction (PCR) is useful when investigating food webs. In this study we estimated how long after a feeding event it was possible to detect mitochondrial COII DNA (331 bp) from an important aphid pest, Rhopalosiphum padi (Homoptera: Aphididae), in spiders of the genus Pardosa (Araneae: Lycosidae). Following laboratory evaluations we tested spiders collected in spring-sown cereals for aphid predation during two seasons. Aphids were digested rapidly in laboratory-fed predators and the time point when prey DNA could be amplified from 50% of the spiders was estimated to be 3.7 h. A total of 372 field- collected predators were analyzed by PCR and despite low aphid densities many spiders (26% in 2004 and 19% in 2005) tested positive for R. padi, indicating consumption of at least one aphid within a few hours before capture. The percentage of spiders that tested positive for R. padi DNA varied considerably between fields and logistic regression analysis revealed that the probability of detecting aphid DNA was significantly influenced by location and year. We conclude that Pardosa spiders, under certain conditions, frequently feed on R. padi and deserve special attention in conservation biological control.  相似文献

We have investigated community and ecosystem consequences of endophyte symbiosis with tall fescue over the past 13 y. Lolium arundinaceum is the most abundant plant in the eastern USA, and most is infected by the wild-type KY-31 endophyte Neotyphodium coenophialum. We established two large experimental grasslands (in 1994 and in 2000) with endophyte-infected or endophyte-free seed sown on recently plowed herbaceous vegetation. Other plant species established by seed or vegetative fragments. No other treatments were applied and plots were subject to natural biotic and abiotic variation. A third experiment examined ecological influences on endophyte infection dynamics starting from an intermediate infection frequency. Finally, we synthesized recent literature investigating the impacts of the tall fescue endophyte on the abundance of associated arthropod species. We found wide-ranging consequences of the endophyte from significant effects on soil feedback and decomposition rates, to plant-plant competition, diversity, productivity, invasibility and succession, to plant-herbivore interactions and energy flow through the food web. Further, we found that herbivore pressure caused rapid increases in infection frequency. Our results suggest that endophyte symbiosis in tall fescue can have a transforming effect on ecological systems.  相似文献
