The exine of birch pollen was examined by scanning and transmission electron microscopy in the native state and after fixation in different aqueous fixatives: glutaraldehyde + OsO4; glutaraldehyde + cetylpyridinium chloride (CPC) + OsO4; glutaraldehyde + cuprolinic blue (CB); and periodate + lysine + paraformaldehyde (PLP). The native pollen exine showed a thin (3-5-nm) border of electron-dense material lining the tectum and electron-dense material within microchannels and bacula cavities. Fixation with the addition of CPC resulted in a voluminous surface coat surrounding the pollen grain, but empty microchannels and bacula cavities. After fixation with the addition of CB, there was a thin surface coat, whereas microchannels and bacula cavities were partially filled with electron-dense material. The other fixatives led to empty microchannels and bacula cavities. There was no surface coat on the pollen grain. However, after all fixation procedures, a thin electron-dense border of the tectum remained visible. Concerning the electron-dense material filling microchannels and bacula cavities in the native pollen grain, the results obtained in the present study suggest that it is either completely lost (after conventional and PLP fixation) or, after fixation with a precipitating additive, partially (CB) or completely (CPC) solubilized and precipitated on the surface of the pollen grain as a surface coat.  相似文献

Irradiation of light-curing dental filling materials in a single direction results in a temporary hardness gradient in the direction of the irradiation. The photoactivated polymerisation process begins at the site of the highest light intensity. In the simplest possible model, the polymerizing composites irradiated in a single direction shows three adjacent co-existing phases: an almost hardened, a gelled and a still plastic phase. As long as all three phases are present, any shrinking of the contracting phases can be compensated by the plastic phase. A knowledge of the distribution of these phases and their spatial and temporal modulation by the selection of suitable curing light parameters provides simple techniques for reducing shrinkage gaps around voluminous fillings in large dental cavities.  相似文献

The reproductive performance of Zebu cattle is generally low. However, blood concentrations of progesterone may be of practical value in assessing the reproductive status of the Zebu female. This paper reviews recent findings regarding plasma progesterone profiles during different phases of the reproductive cycle of some Nigerian Zebu cattle. The reproductive phases examined include the estrous cycle, early pregnancy and the postpartum periods. Our findings describe variations in plasma progesterone profiles observed under field conditions. The practical implications of these findings in assessing reproductive functions in indigenous Nigerian Zebu cattle are examined.  相似文献

The propagation of ordinary bulk modes coupled with extraordinary surface modes in coaxial metal waveguides completely filled with cold magnetoactive plasma is investigated theoretically. The interaction between modes propagating across the waveguide axis in the presence of the axial and azimuthal components of the external magnetic field is examined. The effect of the azimuthal magnetic field on the dispersion properties of these modes is thoroughly studied for the case of a uniform plasma.  相似文献

A general method is developed for a numerical analysis of the frequency spectra of internal, internal-surface, and surface slow waves in a waveguide with transverse plasma density variations. For waveguides with a piecewise constant plasma filling, the spectra of slow waves are thoroughly examined in the limits of an infinitely weak and an infinitely strong external magnetic field. For a smooth plasma density profile, the frequency spectrum of long-wavelength surface waves remains unchanged, but a slow damping rate appears that is caused by the conversion of the surface waves into internal plasma waves at the plasma resonance point. As for short-wavelength internal waves, they are strongly damped by this effect. It is pointed out that, for annular plasma geometry, which is of interest from the experimental point of view, the spectrum of the surface waves depends weakly on the magnetic field strength in the waveguide.  相似文献

Prostaglandin (PG) E2 inhibited both phases of contraction produced by electrical field stimulation of the guinea-pig vas deferens. PGF2alpha and PGD2 were without effect on this preparation. Carbacyclin (a PGI2) analogue inhibited the first phase of contraction at higher concentrations, whereas U46619 (a thromboxane mimetic) potentiated both phases of contraction. As exogenous arachidonic acid inhibits both phases of contraction of the electrically field-stimulated guinea-pig vas deferens, it is likely that the arachidonic acid is converted to PGE2 in the vas deferens. Indomethacin, a non-specific inhibitor of prostaglandin H synthase (PGHS), attenuated the inhibitory actions of exogenous arachidonic acid when examined on the first phase of contraction. NS-398, a relatively specific inhibitor of PGHS-2, also prevented the inhibitory action of exogenous arachidonic acid. However, NS-398 was much less effective than indomethacin in this respect even though NS-398 and indomethacin inhibit PGHS-2 with similar potencies. Consequently, the findings suggest that exogenous arachidonic acid is converted to PGE2 in the guinea-pig vas deferens by the actions of PGHS-1 and, to a lesser extent, by PGHS-2.  相似文献

In SPF female rats, the pineal gland is less voluminous and its cell-nuclei are smaller than those of conventionnal female rats. This confirms (with the voluminous adrenals previously reported) the stressing action of the absence of pathogenic germs on these animals. The anterior lobe of the hypophysis is slightly smaller and subfornical organ a little larger than in conventionnal female rats.  相似文献

The behavior of cholesteryl esters at the air-buffer interface was studied as a function of molecular area and the presence of noncholesterol-containing lipids (colipids). The data obtained indicate that cholesteryl esters with other than long, saturated acyl groups can be present in surface phases up to packing densities approximately those in natural membranes. Their apparent molecular areas in such phases, which are largely determined by colipid structure, suggest their orientation with the ester function toward the interface. The extent of miscibility in the surface phase is also a strong function of colipid structure. Reversibility of the monolayer to bulk phase transition is determined exclusively by the acyl structure of the cholesteryl ester. Of the esters examined, only those with cis unsaturation collapsed reversibly. Our data predict that cholesteryl esters should be present in small, but finite amounts on the surface of arterial lipid deposits and that a prerequisite for the removal of such deposits is that the bulk lipid phase be in a liquid or liquid crystalline state.  相似文献

The lamellar/nonlamellar phase preferences of lipid model membranes composed of mixtures of several cationic lipids with various zwitterionic and anionic phospholipids were examined by a combination of differential scanning calorimetry and (31)P NMR spectroscopy. All of the cationic lipids utilized in this study form only lamellar phases in isolation. Mixtures of these cationic lipids with zwitterionic strongly lamellar phase-preferring lipids such as phosphatidylcholine form only the lamellar liquid-crystalline phase even at high temperatures, as expected. Moreover, mixtures of these cationic lipids with strongly nonlamellar phase-preferring zwitterionic lipids such as phosphatidylethanolamine exhibit a markedly reduced propensity to form inverted nonlamellar phases, again as expected. However, when mixed with anionic lipids such as phosphatidylserine, phosphatidylglycerol, cardiolipin, or phosphatidic acid, a marked enhancement of nonlamellar phase-forming propensity occurs, despite the fact both components of the mixture are nominally lamellar phase-preferring. An examination of the lamellar/nonlamellar phase transition temperatures and the nature of the nonlamellar phases formed, as a function of temperature and of the composition of the mixture, indicates that the propensity to form inverted nonlamellar phases is maximal in mixtures where the mean surface charge of the membrane surface approaches neutrality and decreases markedly with increases in the density of positive or negative charge at the membrane surface. Moreover, the onset temperatures of the reversed hexagonal phase rise more steeply than do those of the inverted cubic phase as the ratio of cationic and anionic lipids is varied, suggesting that the formation of inverted hexagonal phases is more sensitive to this surface charge effect. These results indicate that surface charge per se is a significant and effective modulator of the lamellar/nonlamellar phase preferences of membrane lipids and that charged group interactions at membrane surfaces may have a major role in regulating this particular membrane property.  相似文献

From measurements of the equilibrium spreading pressure pie for dispersions of lecithin--dimyristoyl (DML) or dioleoyl (DOL)--and cholesterol (CHOL) in water, we have deduced the phase relations in both the aqueous dispersions and the equilibrium surface films. At 29.5 degrees C, when the mole fraction of cholesterol in the dispersion chi(CHOL) is 0 chi(CHOL) less than chi(CHOL) less than 0.33, pie is constant and equal to the value for pure lecithin (DOL or DML). The phase rule predicts than two bulk lipid phases coexist; these are pure lecithin and lecithin:cholesterol 2:1 complex. The equilibrium surface film contants only lecithin and therefore lecithin and 2:1 complex are immiscible in surface films. When 0.33 less than chi/CHOL) less than 1.0, pie is also contant with a value intermediate between that for pure lecithin and cholesterol. In this range of lipid composition two bulk lipid phases also coexist: lecithin:cholesterol 2:1 complex and pure cholesterol. However, the equilibrium surface film contains only the 2:1 complex and, therefore, 2:1 complex is also immiscible with cholesterol in surface films. When pi less than pie, as in the case of spread films, we deduce that two surface phases may coexist; the composition of the phases will depend on chi(CHOL). When 0 less than chi(CHOL) less than 0.33, both lecithin and 2:1 complex coexist, and when 0.33 less than chi(CHOL) less than 1.0, 2:1 complex and cholesterol coexist. The "condensing" effect of cholesterol in lecithin surface films is reexamined. The effect is attributed to formation of the lecithin:cholesterol 2:1 complex and nonequilibrium conditions in the two-phase surface film.  相似文献

The flow field less than one diameter downstream of the end of a collapsible tube executing self excited oscillations was examined using a two-component fiber-optic laser-Doppler anemometer. The time-averaged Reynolds number of the flow was 11,000. With the tube oscillating periodically, results obtained during many cycles of oscillation were combined to yield surface plots of the axial component over the cross section at 16 phases of the cycle. By combining measurements obtained with the laser probe in two different orientations, secondary flow vectors over the cross section were likewise constructed for 16 phases. The measurements showed strongly phasic turbulence intensity, with the phase of high intensity coinciding with the time of maximal tube collapse. Reverse flow occurred during much of the cycle, at places in the cross section that agree with our previous observations of laminar and turbulent steady flow through a rigid simulated collapsed tube.  相似文献

Recent experimental results show that the spreading coefficient frequently becomes positive when Candida lipolytica is cultivated on n-hexadecane. The effects of oil spreading at the surface of air bubbles in an airlift fermentor are examined using a mathematical model. The distribution of the oil phase with position and among the phases is determined using computer simulation. The simulation results qualitatively explain some of the experimental observations which have been previously reported.  相似文献

Electron microscopic study of the microbial cells of the I, II phases and the R-form was carried out. Intact cells were examined by negative contrasting, and morphological differences of various bacterial phases were shown: cells of the I phase had a relatively smooth surface, bacteria of the II phase had a smooth surface, but many cell wall fragments were split from them; the surface of the R-form cells was coarse, folded, and cell wall fragments were split from the majority of bacteria. Antigenic determinants responsible for phasic specificity in bacteria of the I and II phases were located at some distance from the external membrane of the cell wall; as to the R-form cells--they were localized on the wall.  相似文献

Aims

In this study, a chicken manure biochar (CM biochar) and a paper sludge biochar (PS biochar), prepared under similar treatment conditions, were amended into ferrosol as part of an agronomic field trial. The aim of this study is to investigate interactions between these biochars and the soil after a 3 month trial.

Methods

Soil samples following field trials were taken and biochar was separated from the soil, and studied for both surface oxidation and the degree of interaction with surrounding soil by X-ray photoelectron spectroscopy (XPS), SEM and TEM equipped with EDS for elemental analysis.

Results

Following incubation in field soil, both biochars showed that soil mineral incorporation on to their surfaces occurred within the first year, although the attachment was localized at specific sites on the surface. A relatively high concentration of Al was found at the interface between the biochar and mineral phases in both aged biochars, indicating a binding role of Al. For the CM biochar, a soil-iron redox reaction may be associated with the formation of biochar-mineral complexes due to the relatively higher labile carbon content and higher pH value of this biochar.

Conclusions

Soil mineral attachment may occur directly on to the biochar surface because of the formation of carboxylic and phenolic functional groups on the aged CM biochar surface by an oxidation reaction. For the PS biochar, adsorption of organic matter from the soil facilitated interactions between the biochar and mineral phases in the soil. Calcium is believed to be important in this process.  相似文献

Summary Membrane-bound Ca2+-ATPase activity was localized cytochemically in the blood vessels of the spinal cord of rat embryos to obtain a better understanding of the membrane activities of vascular cells.The cytochemical method revealed a growth of the parenchymal vasculature. In the parenchyma, reaction product was dense over the entire plasma membrane of voluminous endothelial cells provided with large nuclei and enriched cytoplasmic organelles, suggesting that the endothelial cells may be of a vascular sprout. The parenchymal vessels with a wide lumen were frequently associated with pericytes, and the Ca2+-ATPase activity was diminished in intensity on the luminal surface of the flattened endothelial cells. On the other hand, the endothelium of extraparenchymal capillaries exhibited Ca2+-ATPase activity primarily on the luminal surface of the plasma membrane. Quercetin, a Ca2+-transporting ATPase inhibitor, considerably decreased the abluminal activity in the voluminous endothelial cells with slit-like vascular lumen and the luminal activity of functioning capillary endothelium as well. Thus, a dual activity of Ca2+-ATPase, postulating for the activities of Ca2+-transporting ATPase and ecto-ATPase, was closely correlated with the maturation processes of the capillary endothelium.  相似文献

The sea urchin sperm nucleus rapidly loses its conoid morphology and becomes more voluminous and spherical upon its entry into the egg cytoplasm during fertilization. This investigation has attempted to determine what are the structural constraints placed upon the sperm nucleus, so that further investigations might determine the egg cytoplasmic factors that are responsible for modifying nuclear morphology. Isolated sperm nuclei were subjected to various extraction procedures in order to remove the majority of the proteins (histones) and also the DNA; subsequently, the residual structures were processed for and examined by electron microscopy. The data presented in this investigation demonstrate the removal of the sperm nuclear histones plus other nonhistone proteins has no effect on the conoid morphology of the sperm nucleus, yet this protein removal has a profound effect on the structure of the nuclear chromatin. It is also shown that removal of the majority of the nuclear DNA has no effect on the shape of the sperm nucleus. These results indicate that there are other components (possibly a nuclear matrix) associated with the sperm nucleus that are responsible for maintaining its conoid morphology.  相似文献

Factors affecting the ice nucleation temperature of plants and plant tissues were examined. The mass of a sample had a marked effect on ice nucleation temperature. Small tissue samples supercooled to −10°C and were not accurate predictors of the nucleation temperature of intact plants in either laboratory or field experiments. This effect was not unique to plant tissues and was observed in autoclaved and control soil samples. Ice nucleation temperatures of bean, corn, cotton, and soybean seedlings were influenced by the length of subzero exposure, presence of ice nucleation active bacteria, and leaf surface wetness. The number of factors influencing ice nucleation temperature suggested that predicting the freezing behavior of plants in the field will be complex.  相似文献

A single pair of protonephridia is the typical larval excretory organ of molluscs. Their presence in postlarval developmental stages was discovered only recently. We found that the protonephridia of the polyplacophoran mollusc, Lepidochitona corrugata, achieve their most elaborate differentiation and become largest during the postlarval period. This study describes the protonephridia of L. corrugata using light and electron microscopy and interactive three‐dimensional visualization. We focus on the postlarval developmental period, in which the protonephridia consist of three parts: the terminal part with the ultrafiltration sites at the distal end, the voluminous protonephridial kidney, and the efferent nephroduct leading to the nephropore. The ultrafiltration sites show filtration slits between regularly arranged thin pedicles. The ciliary flame originates from both the terminal cell and the duct cells of the terminal portion. The efferent duct also shows ciliation. The most conspicuous structures, the protonephridial kidneys, are voluminous swellings composed of reabsorptive cells (“nephrocytes”). These cells exhibit strong vacuolization and an infolding system increasing the basal surface. The protonephridial kidneys, previously not reported at such a level of organization in molluscs, strikingly resemble (metanephridial) kidneys of adult molluscan excretory systems. J. The solubilities of two fluorescent lipid amphiphiles with comparable apolar structures and different polar head groups, NBD-hexadecylamine and RG-tetradecylamine (or -octadecylamine), were compared in lipid bilayers at a molar ratio of 1/50 at 23 degrees C. Bilayers examined were in the solid, liquid-disordered, or liquid-ordered phases. While NBD-hexadecylamine was soluble in all the examined bilayer membrane phases, RG-tetradecylamine was stably soluble only in the liquid-disordered phase. RG-tetradecylamine insolubility in solid and liquid-ordered phases manifests itself as an aggregation of the amphiphile over a period of several days and the kinetics of aggregation were studied. Solubility of these amphiphiles in the different phases examined seems to be related to the dipole moment of the amphiphile (in particular, of the polar fluorophore) and its orientation relative to the dipolar potential of the membrane. We propose that amphiphilic molecules inserted into membranes (including lipid-attached proteins) partition into different coexisting membrane phases based upon: (1) nature of the apolar structure (chain length, degree of saturation, and chain branching as has been proposed in the literature); (2) magnitude and orientation of the dipole moment of the polar portion of the molecules relative to the membrane dipolar potential; and (3) hydration forces that are a consequence of ordering of water dipoles at the membrane surface.  相似文献

The effect of alpha-tocopherol on the structure and thermotropic phase behaviour of distearoylphosphatidylethanolamine was examined by using synchrotron X-ray diffraction methods. There was evidence that alpha-tocopherol does not distribute randomly in the dispersed phospholipid but instead phospholipid phases enriched in alpha-tocopherol are formed. Heating codispersions from lamellar gel phase induced formation of hexagonal-II phase at temperatures below the main transition of the pure phospholipid and which were enriched in alpha-tocopherol. Codispersions containing 5 or 10 mol% alpha-tocopherol were induced to form a cubic phase at temperatures above the lamellar to hexagonal-II phase transition. Such phases were not observed in codispersions containing 2.5 or 20 mol% alpha-tocopherol in which only lamellar and hexagonal-II phases were formed. The space group of the cubic phases were tentatively assigned as Pn3m. Equilibration of codispersions at 4 degrees C results in the formation of lamellar crystalline phases enriched in alpha-tocopherol and phase separated domains of pure phospholipid. Two lamellar crystalline phases were characterized on the basis of their particular wide-angle X-ray scattering patterns. The lamellar crystalline phases were also distinguished from other lamellar phases of the pure phospholipid by the lamellar repeat. Partitioning of alpha-tocopherol into phosphatidylethanolamine domains in membranes may introduce instability into the structure.  相似文献
