分子标记技术近年来发展很快,目前,常用的分子标记主要可以分为基于杂交的分子标记、基于PCR的分子标记、基于限制性酶切和PCR结合的分子标记以及新一代分子标记.本文对这几类分子标记中常用类型的基本原理进行介绍,并从分子标记在作物种质遗传多样性、基因定位和基因克隆以及分子标记辅助选择育种和分子设计育种中应用进行了阐述. 相似文献

为筛选出适用于重庆加工型辣椒疫病抗性鉴定的分子标记,以63份重庆加工型辣椒种质资源为材料,研究了12个辣椒疫病抗性相关分子标记的筛选效率。结果表明,7个分子标记在种质间无差异条带,2个分子标记的扩增结果不稳定,只有ZL6203、ZL6726和E73等3个分子标记在种质间能扩增出差异条带。ZL6203筛选高抗、抗性和中抗材料的效率分别为87.50%、77.78%和63.64%;ZL6726筛选抗性和感病材料的效率分别为100.00%和66.67%;E73筛选抗性和高抗材料的效率分别为77.78%和62.50%。因此,同一辣椒抗疫病分子标记对不同抗性材料筛选效率存在较大差异,ZL6203、ZL6726和E73适用于重庆地区加工型辣椒抗疫病材料的鉴定。 相似文献

利用分子标记和数量性状基因型值构建作物核心种质库的研究 总被引:7,自引:0,他引:7
提出了一种基于分子标记数据及数量性状基因型值构建作物种质资源核心种质库的方法.采用包括基因型与环境互作的遗传模型及相应的混合线性模型统计分析方法,无偏预测各材料的基因型值,分别用基因型值和分子标记数据计算个体间的相似系数,加权得到最终的相似距离.采用不加权类平均法(UPGMA)进行系统聚类,用多次聚类随机取样法构建核心种质库.以水稻DH群体111个基因型8个农艺性状、175个分子标记位点的数据为实例,按四种抽样比率(25%,20%,15%,10%)构建了四个核心种质库,比较了核心种质库与整个群体的分子标记多样性及数量性状的遗传变异,评价了所用方法的有效性。 相似文献

初步确定了2个新的猪产仔数分子标记,雌激素受体基因ESR的第8外显子处的ESRB位点、催乳素受体基因的第7外显子的FSHRB位点。通过比较和分析多个产仔数的效应,初步确定了4个有利于产仔数提高的分子标记基因型,基因位点ESR、FSHRB的基因型BB的产仔数显著地高于AB、AA型;位点ESRB、PRLR的基因型AA的产仔数显著地高于AB、BB型;4个基因位点多态性与仔猪生长性能、母猪乳头数不存在显著的影响。 相似文献

A genetic map of potato (Solanum tuberosum L.) integrating molecular markers with morphological and isozyme markers was constructed using a backcross population of 67 diploid potato plants. A general method for map construction is described that differs from previous methods employed in potato and other outbreeding plants. First, separate maps for the female and male parents were constructed. The female map contained 132 markers, whereas the male map contained 138 markers. Second, on the basis of the markers in common the two integrated parental maps were combined into one with the computer programme JoinMap. This combined map consisted of 175 molecular markers, 10 morphological markers and 8 isozyme markers. Ninety-two of the molecular markers were derived from DNA sequences flanking either T-DNA inserts in potato or reintegrated maize transposable elements originating from these T-DNA constructs. Clusters of distorted segregation were found on chromosomes 1,2,8 and 11 for the male parent and chromosome 5 for both parents. The total length of the combined map is 1120 cM. 相似文献

The informativeness and inheritance of randomly amplified polymorphic DNA (RAPD) markers were investigated in an intraspecific F1 progeny derived from two heterozygous parents. The analysis confirmed the utility of RAPD markers for comparing candidate parents for the development of a molecular genetic map, and provided numerous markers for linkage analysis in a crop with a very limited history of classical or molecular genetic studies. Six potential parental lines (themselves F1 hybrid clones) showed between 1.82 and 0.62 segregating bands per primer in three hybrid families. Forty-three percent (309) of 722 primers produced polymorphic products in the most informative of these three crosses, revealing 328 single-dose (SD) markers segregating 1:1 for presence/absence in a progeny of 90 individuals. A second class of informative markers were those present in both parents but segregating in the progeny. Fifty-seven or 67% of the monomorphic but segregating markers exhibited the 3:1 ratio expected for SD dominant markers in a cross between heterozygotes. Linkage groups were constructed from the segregation of SD RAPD markers originating in the female (TMS 30572) and the male (CM2177-2) parent. Adult plant resistance (APR) is considered potentially more durable for controlling barley leaf rust than seedling Rph (Resistance to Puccinia hordei) genes. A major gene for adult plant resistance to barley leaf rust has been mapped to the telomere region of chromosome
5HS. PCR-based molecular markers were developed for saturation of this region based on previously mapped simple sequence repeat,
restriction fragment length polymorphism and Diversity Arrays Technology markers. In addition, defence gene homologue (DGH)
and wheat expressed sequence tags mapped in specific bins were used to develop new PCR markers. Seventeen PCR-based markers
were mapped to the short arm of chromosome 5H in 292 doubled haploid lines from a cross of Pompadour × Stirling, in which
seven markers were mapped within 5 cM of the APR gene. The closest linked marker was about 0.7 cM from the APR gene. The wheat
deletion bin map together with defence gene homologues was demonstrated to be an efficient tool for development of new molecular
markers associated with the disease resistance gene. Four DGH markers were associated with the APR gene. The new molecular
markers are a useful tool for marker-assisted selection of the APR gene and provided a better understanding of the molecular
