This study investigated cadmium (Cd) uptake in Elodea canadensis shoots under different photosynthetic conditions, and its effects on internal (cytosolic) and external pH. The plants were grown under photosynthetic (light) or non‐photosynthetic (dark or in the presence of a photosynthetic inhibitor) conditions in the presence or absence of CdCl2 (0.5 μm ) in a medium with a starting pH of 5.0. The pH‐sensitive dye BCECF‐AM was used to monitor cytosolic pH changes in the leaves. Cadmium uptake in protoplasts and leaves was detected with a Cd‐specific fluorescent dye, Leadmium Green AM, and with atomic absorption spectrophotometry. During cultivation for 3 days without Cd, shoots of E. canadensis increased the pH of the surrounding water, irrespective of the photosynthetic conditions. This medium alkalisation was higher in the presence of CdCl2. Moreover, the presence of Cd also increased the cation exchange capacity of the shoots. The total Cd uptake by E. canadensis shoots was independent of photosynthetic conditions. Protoplasts from plants exposed to 0.5 μm CdCl2 for 3 days did not exhibit significant change in cytosolic [Cd2+] or pH. However, exposure to CdCl2 for 7 days resulted in increased cytosolic [Cd2+] as well as pH. The results suggest that E. canadensis subjected to a low CdCl2 concentration initially sequesters Cd into the apoplasm, but under prolonged exposure, Cd is transported into the cytosol and subsequently alters cytosolic pH. In contrast, addition of 10–50 μm CdCl2 directly to protoplasts resulted in immediate uptake of Cd into the cytosol.  相似文献

Aims

There is a need to predict trace metal concentration in plant organs at given development stages. The aim of this work was to describe the Cd hyperaccumulation kinetics in the different plant organs, throughout the complete cultivation cycle, independently of a possible soil effect.

Methods

Plants of Noccaea caerulescens were exposed in aeroponics to three constantly low Cd concentrations and harvested at 6 to 11 dates, until siliquae formation.

Results

Dry matter allocation between roots and shoots was constant over time and exposure concentrations, as well as Cd allocation. However 86 % of the Cd taken up was allocated to the shoots. Senescent rosette leaves showed similar Cd concentrations to the living ones, suggesting no redistribution from old to young organs. The Cd root influx was proportional to the exposure concentration and constant over time, indicating that plant development had no effect on this. The bio-concentration factor (BCF), i.e. [Cd]plant/[Cd2+]solution for the whole plant, roots or shoots was independent of the exposure concentration and of the plant stage.

Conclusions

Cadmium uptake in a given plant part could therefore be predicted at any plant stage by multiplying the plant part dry matter by the corresponding BCF and the Cd2+ concentration in the exposure solution.  相似文献

Summary Cadmium uptake by cultures of transformed hairy-roots of Solanum nigrum was studied. The effect of pH, buffer type, temperature, exposure time and Cd2+ content ranging from 0.2 to 2000 ppm was measured. Cd2+ uptake was dependent on increasing metal concentration and it was time dependent. From the variety of buffers tested, MES buffer and borate ions were found to be beneficial for the Cd2+ uptake. The high effectivness of Cd2+ accumulation in the roots decreased significantly after increasing the Cd2+ content in the buffer over 2 ppm.  相似文献

Effect of Cl on Cd uptake by Swiss chard in nutrient solutions   总被引:6,自引:1,他引:5  
Swiss chard (Beta vulgaris L., cv. Fordhook Giant) was grown in nutrient solution with Cl concentrations varying between 0.01 mM and 120 mM. Solution Na concentration and ionic strength were maintained in all treatments by compensating with NaNO3. All solutions contained Cd (50 nM, spiked with 109Cd). Three different Cd2+ buffering systems were used. In one experiment, Cd2+ activity was unbuffered; its activity decreased with increased Cl concentration as a result of the formation of CdCln 2–n species. In the other experiments, Cd2+ activity was buffered by the chelator nitrilotriacetate (NTA, 50 M) and ethylene-bis-(oxyethylenenitrilo)-tetraacetate (EGTA, 50 M) at about 10–9 M and 10–11 M, respectively. Plant growth was generally unaffected by increasing Cl concentrations in the three experiments. In unbuffered solutions, Cd concentrations in plant tissue decreased significantly (p<0.01) (approximately 2.4-fold) as solution Cl concentration increased from 0.01 mM to 120 mM. However, this decrease was smaller in magnitude than the 4.7-fold decrease in Cd2+ activity as calculated by the GEOCHEM-PC program for the same range of Cl concentrations. In solutions where Cd2+ activity was buffered by NTA, Cd concentrations in plant tissue increased approximately 1.4-fold with increasing Cl concentration in solution, while the Cd2+ activity was calculated to decrease 1.3-fold. In solutions where Cd2+ activity was buffered by EGTA, Cd concentrations in the roots increased 1.3-fold with increasing Cl concentration in solution but there was no effect of Cl on shoot Cd concentrations. CO2 exchange, variable chlorophyll fluorescence, the intensity of lipid peroxidation (POL), and the activity of antioxidant enzymes in leaves of two-week-old pea seedlings (Pisum sativum L.) exposed to 0.01, 0.1, and 1 mM aqueous solutions of Cd(NO3)2 for 2 h were studied. Incubation with Cd2+ ions resulted in a reduction of the maximum quantum efficiency of photosynthesis, CO2 uptake rate, and photosystem II (PSII) activity, as assessed by F v/F 0 ratio. The intensity of POL in leaves of all treated seedlings was below or close to the control level. The activity of superoxide dismutase (SOD) and glutathione reductase (GR) increased in all treatments; the activity of ascorbate peroxidase (AP) exceeded the control level only in seedlings exposed to the high Cd2+ concentration (1 mM), and the activity of peroxidase increased at the low concentration (0.01 mM). We found that the reduction in the activity of the photosynthetic apparatus under conditions of 2-h-long Cd2+-induced stress was not related to an intensification of POL processes. Cadmium toxicity has been extensively studied in plants, however its biochemical mechanism of action has not yet been well established. To fulfil this objective, four-weeks-old soybean nodulated plants were treated with 200 μM Cd2+ for 48 h. δ-aminolevulinic acid dehydratase (ALA-D, E.C. 4.2.1.24) activity and protein expression, as well as δ-aminolevulinic acid (ALA) and porphobilinogen (PBG) concentrations were determined in nodules, roots and leaves. In vitro experiments carried out in leaves were performed using leaf discs to evaluate the oxidant and antioxidant properties of ALA and S-adenosyl-l-methinone (SAM), respectively. Oxidative stress parameters such as thiobarbituric acid reactive substances (TBARS) and GSH levels as well as superoxide dismutase (SOD, E.C. 1.15.1.1), and guaiacol peroxidase (GPOX, E.C. 1.11.1.7) were also determined. Cadmium treatment caused 100% inhibition of ALA-D activity in roots and leaves, and 72% inhibition in nodules whereas protein expression remained unaltered in the three studied tissues. Plants accumulated ALA in nodules (46%), roots (2.5-fold) and leaves (104%), respect to controls. From in vitro experiments using leaf discs, exposed to ALA or Cd2+, it was found that TBARS levels were enhanced, while GSH content and SOD and GPOX activities and expressions were diminished. The protective role of SAM against oxidative stress generated by Cd2+ and ALA was also demonstrated. Data presented in this paper let us to suggest that accumulation of ALA in nodules, roots and leaves of soybean plants due to treatment with Cd2+ is highly responsible for oxidative stress generation in these tissues.  相似文献

Enhanced phytoextraction uses soil chelators to increase the bioavailability of heavy metals. This study tested the effectiveness of ethylenediaminetetraacetic acid (EDTA) and citric acid in enhancing cadmium (Cd) phytoextraction and their effects on the growth, yield, and ionic uptake of maize (Zea mays). Maize seeds of two cultivars were sown in pots treated with 15 (Cd15) or 30 mg Cd kg?1 soil (Cd30). EDTA and citric acid at 0.5 g kg?1 each were applied 2 weeks after germination. Results demonstrated that the growth, yield per plant, and total grain weight were reduced by exposure to Cd. EDTA increased the uptake of Cd in shoots, roots, and grains of both maize varieties. Citric acid did not enhance the uptake of Cd, rather it ameliorated the toxicity of Cd, as shown by increased shoot and root length and biomass. Cadmium toxicity reduced the number of grains, rather than the grain size. The maize cultivar Sahiwal-2002 extracted 1.6% and 3.6% of Cd from soil in both Cd+ EDTA treatments. Hence, our study implies that maize can be used to successfully phytoremediate Cd from soil using EDTA, without reducing plant biomass or yield.  相似文献

Information on cadmium (Cd) uptake and transport is essential to understand better the physiology of Cd tolerance in plants. In this study, Cd uptake, translocation, and tolerance were investigated in AHA1 (Arabidopsis plasma membrane H+-ATPase gene) overexpressed plants. Exposed to 10 μM CdCl2, AHA1OX showed a higher root elongation, accumulated more Cd, and maintained better integrity of nucleus membrane of root tips in comparison to the control plant (WT), suggesting that AHA1OX was more Cd tolerant than WT. To investigate Cd tolerance mechanism of AHA1OX plants, we measured the activity of plasma membrane H+-ATPase and the secretion of citrate. Results indicated that treatment with 10 μM of Cd stimulated the activity of plasma membrane H+-ATPase and the secretion of citrate, while 30 μM of Cd inhibited them. AHA1OX had higher activity of H+-ATPase and secretion of citrate than WT. Addition of citrate enhanced root-to-shoot translocation of Cd significantly. A higher root-to-shoot Cd translocation was observed in AHA1OX than in WT plants. Treatment with low temperature or metabolic inhibitor (carbonyl cyanide m-chlorophenylhydrazone) inhibited Cd uptake and translocation. The study of Cd forms using sequential extraction indicated that Cd was mainly present as a protein-bound form, and AHA1OX had more water-soluble Cd than WT. Taken together, our results suggested that the Cd tolerance of AHA1OX was associated with its root-to-shoot Cd translocation and secretion of citrate, which converts Cd2+ into less toxic and more easily transportable forms in plant cells.  相似文献

Cadmium (Cd) originating from atmospheric deposits, from industrial residues and from the application of phosphate fertilizers may accumulate in high concentrations in soil, water and food, thus becoming highly toxic to plants, animals and human beings. Once accumulated in an organism, Cd discharges and sets off a sequence of biochemical reactions and morphophysiological changes which may cause cell death in several tissues and organs. In order to test the hypothesis that Cd interferes in the metabolism of G. americana, a greenhouse experiment was conducted to measure eventual morphophysiological responses and cell death induced by Cd in this species. The plants were exposed to Cd concentrations ranging from 0 to 16 mg l−1, in a nutritive solution. In TUNEL reaction, it was shown that Cd caused morphological changes in the cell nucleus of root tip and leaf tissues, which are typical for apoptosis. Cadmium induced anatomical changes in roots and leaves, such as the lignification of cell walls in root tissues and leaf main vein. In addition, the leaf mesophyll showed increase of the intercellular spaces. On the other hand, Cd caused reductions in the net photosynthetic rate, stomatal conductance and leaf transpiration, while the maximum potential quantum efficiency of PS2 (Fv/Fm) was unchanged. Cadmium accumulated in the root system in high concentrations, with low translocation for the shoot, and promoted an increase of Ca and Zn levels in the roots and a decrease of K level in the leaves. High concentrations of Cd promoted morphophysiological changes and caused cell death in roots and leaves tissues of G. americana.  相似文献

The effects of different CdCl2 concentrations on the growth and on certain biochemical parameters of almond seedlings (Prunus dulcis) were studied under controlled conditions in the nutrient solutions containing increasing CdCl2 concentrations ranging from 0 to 150 μM CdCl2. Under Cd stress conditions, damage was variable. Cadmium reduced dry matter production in leaves and roots. While chlorophyll content was severely decreased, that of leaf sugars appeared to be increased. Furthermore, leaf nutritional status seemed to be more altered than that of roots. Both in roots and leaves, there was an increase in MDA content as metal concentration increased. It may be suggested from the present study that toxic concentrations of Cd cause oxidative damage as shown by the increase of lipid peroxidation.  相似文献

The effect of increased Cd2+ concentrations in the watering solution on citrus physiology was studied by using two citrus genotypes, Cleopatra mandarin and Carrizo citrange. Cadmium content in roots and leaves was tested together with measurements of leaf damage, gas exchange parameters, and hormonal contents. Citrus roots efficiently retained Cd2+ avoiding its translocation to the shoots and Cleopatra mandarin translocated less Cd2+ than Carrizo. With increasing Cd2+ concentration all gas exchange parameters were decreased more in Carrizo than in Cleopatra mandarin. Cd-induced increases in abscisic acid and salicylic acid contents were observed in leaves but not in roots of both genotypes.  相似文献

Summary Cadmium and zinc uptake parameters were determined for intact corn (Zea mays L.) seedlings grown for 15 and 22 in nutrient solutions containing levels of Cd and Zn that were similar to those found in soil solutions. Uptake of both elements was assumed to follow Michaelis-Menten kinetics. Calculations were based on the concentrations of free ionic Cd (Cd2+) and Zn (Zn2+) rather than the total solution concentration. Rates of Zn uptake were measured by determining depletion of Zn for periods of up to 30 h from solutions containing initial concentrations of 1.5 and 10μmol Zn 1−1. Depletion curves suggested that Zn uptake characteristics were similar at both levels of Zn in solution. The Imax for Zn uptake decreased from 550 to 400 pmol m−2 root surface s−1 between 16 and 22 d of growth while Km decreased from 2.2 to 1.5 μmol Zn2+ 1−1. Cadmium uptake parameters were measured by controlling Cd2+ activities in nutrient solution betwen 6.3 to 164 nmol l−1 by continuous circulation of nutrient solution through a mixed-resin system. Imax for Cd uptake was 400 pmol m−2 root surface s−1 at 15 and 22 d of growth. The magnitude of Km increased from 30 to 100 nmol Cd2+ 1−1 during this time period. The Km value suggests that corn is efficient for Cd uptake. The results of these uptake studies are consistent with the observed uptake of Zn and Cd by corn seedlings in soils.  相似文献

Tobacco (Nicotiana tabacum L.) is a tolerant species that accumulates cadmium. We studied the effect of Cd (0, 10, 25, 50, 100 μM) on growth parameters, chlorophyll and proline contents, enzymatic antioxidative response and lipid peroxidation of tobacco plants grown in hydroponic culture for 11 days to clarify the strategy of plant response against oxidative stress caused by this heavy metal. Cadmium accumulated more in roots than in shoots. Plant growth was not significantly affected by the cadmium concentrations used. Young leaves were more affected, showing visible chlorosis and a significant decrease in chlorophyll content at high Cd concentrations. Dry weight of both leaves and roots increased indicating a lower capacity for roots to absorb water. An increase in malondialdehyde levels was observed, indicating that lipid peroxidation occurred as a result of ROS formation. The activity of guaiacol peroxidase in leaves increased, indicating that it was very important in the scavenging of H2O2, while superoxide dismutase activity only increased in old leaves. Ascorbate peroxidase showed constant activity levels in tobacco leaves, suggesting that the ascorbate–glutathione pathway was less important as a defense mechanism.  相似文献

Fe uptake in dicotyledonous plants is mediated by a root plasma membrane-bound ferric reductase that reduces extracellular Fe(III)-chelates, releasing Fe2+ ions, which are then absorbed via a metal ion transporter. We previously showed that Fe deficiency induces an increased capacity to absorb Fe and other micronutrient and heavy metals such as Zn2+ and Cd2+ into pea (Pisum sativum L.) roots [Cohen et al. (1998) Plant Physiol 116:1063–1072). To investigate the molecular basis for this phenomenon, an Fe-regulated transporter that is a homologue of the Arabidopsis IRT1 micronutrient transporter was isolated from pea seedlings. This cDNA clone, designated RIT1 for root iron transporter, encodes a 348 amino acid polypeptide with eight putative membrane-spanning domains that is induced under Fe deficiency and can functionally complement yeast mutants defective in high- and low-affinity Fe transport. Chelate buffer techniques were used to control Fe2+ in the uptake solution at nanomolar activities representative of those found in the rhizosphere, and radiotracer methodologies were employed to show that RIT1 is a very high-affinity 59Fe2+ uptake system (K m =54–93 nM). Additionally, radiotracer (65Zn, 109Cd) flux techniques were used to show that RIT can also mediate a lower affinity Zn and Cd influx (K m of 4 and 100 M, for Zn2+ and Cd2+, respectively). These findings suggest that, in typical agricultural soils, RIT1 functions primarily as a high-affinity Fe2+ transporter that mediates root Fe acquisition. This is consistent with recent findings with Arabidopsis IRT1 knockout mutants that strongly suggest that this transporter plays a key role in root Fe uptake and nutrition. However, the ability of RIT1 to facilitate Zn and Cd uptake when these metals are present at elevated concentrations suggests that RIT1 may be one pathway for the entry of toxic metals into the food chain. Furthermore, the finding that plant Fe deficiency status may promote heavy metal uptake via increased expression of this transporter could have implications both for human nutrition and also for phytoremediation, the use of terrestrial plants to sequester toxic metals from contaminated soil.  相似文献

Cysteine, γ-glutamylcysteine, and glutathione and the extractable activity of the enzymes of glutathione biosynthesis, γ-glutamylcysteine synthetase (EC 6.3.2.2) and glutathione synthetase (EC 6.3.2.3), were measured in roots and leaves of maize seedlings (Zea mays L. cv LG 9) exposed to CdCl2 concentrations up to 200 micromolar. At 50 micromolar Cd2+, γ-glutamylcysteine contents increased continuously during 4 days up to 21-fold and eightfold of the control in roots and leaves, respectively. Even at 0.5 micromolar Cd2+, the concentration of γ-glutamylcysteine in the roots was significantly higher than in the control. At 5 micromolar and higher Cd2+ concentrations, a significant increase in γ-glutamylcysteine synthetase activity was measured in the roots, whereas in the leaves this enzyme activity was enhanced only at 200 micromolar Cd2+. Labeling of isolated roots with [35S]sulfate showed that both sulfate assimilation and glutathione synthesis were increased by Cd. The accumulation of γ-glutamylcysteine in the roots did not affect the root exudation rate of this compound. Our results indicate that maize roots are at least in part autonomous in providing the additional thiols required for phytochelatin synthesis induced by Cd.  相似文献

The effect of calcium (Ca2+) on Trifolium repens L. seedlings subjected to cadmium (Cd2+) stress was studied by investigating plant growth and changes in activity of antioxidative enzymes. Physiological analysis was carried out on seedlings cultured for 2 weeks on half-strength Hoagland medium with Cd2+ concentrations of 0, 400 and 600 μM, and on corresponding medium supplied with CaCl2 (5 mM). Exposure to increasing Cd2+ reduced the fresh weight of the upper part (stems + leaves) of the seedlings more strongly than that of the root system. In both parts of T. repens seedlings H2O2 level and lipid peroxidation increased. In the upper part, Cd2+ exposure led to a significant decrease in the activity of superoxide dismutase, catalase and glutathione peroxidase and an increase in ascorbate peroxidase activity. In contrast, the roots showed an increase in the activity of antioxidative enzymes under Cd2+ stress. Ca2+ addition to medium reduced the Cd2+ accumulation, and considerably reversed the Cd2+-induced decrease in fresh mass as well as the changes in lipid peroxidation in the both parts of T. repens seedlings. Ca2+ application diminished the Cd2+ effect on the activity of antioxidative enzymes in the upper part, even though it did not significantly affect these enzymes in the roots. So the possible mechanisms for the action of Ca2+ in Cd2+ stress were considered to reduce Cd2+ accumulation, alleviate lipid peroxidation and promote activity of antioxidative enzymes.  相似文献

To characterize the dynamics of Cd2+ flux in the rhizosphere and to study cadmium (Cd) plant‐internal partitioning in roots, wood, bark and leaves in relation to energy metabolism, reactive oxygen species (ROS) formation and antioxidants, Populus × canescens plantlets were exposed to either 0 or 50 µM CdSO4 for up to 20 days in the nutrient solution. A strong net Cd2+ influx in root apex was observed after Cd exposure for 24 h, even if net Cd2+ influx decreased gradually in roots. A large amount of Cd was accumulated in roots. Cd ions were uploaded via the xylem to leaves and further transported to the phloem where significant accumulation was detected. Cd accumulation led to decreased photosynthetic carbon assimilation but not to the depletion in soluble carbohydrates. Increased levels of ROS were present in all tissues, except the bark of Cd‐exposed poplars. To combat Cd‐induced superoxide and hydrogen peroxide, P.×canescens appeared to rely mainly on the formation of soluble phenolics as these compounds showed the highest accumulation in the bark and the lowest in wood. Other potential radical scavengers such as proline, sugar alcohols and antioxidant enzymes showed tissue‐ and exposure time‐specific responses to Cd. These results indicate a complex pattern of internal Cd allocation in P.×canescens resulting in higher ROS stress in wood than in bark and intermediate responses in roots and leaves, probably because of differential capacities of these tissues for the production of protective phenolic compounds.  相似文献

Low molecular weight organic acids are important components of root exudates and therefore, knowledge regarding the mechanisms of cadmium (Cd) uptake and distribution within plants under the influence of organic acids, is necessary for a better understanding of Cd behavior in the plant–soil system. In this study, acetic and malic acids increased the uptake of Cd by maize (Zea mays L. cv. TY2) roots and enhanced Cd accumulation in shoots under hydroponic conditions. Concentration-dependent net Cd influx in the presence and absence of organic acids could be resolved into linear and saturable components. The saturable component followed Michaelis–Menten kinetics, which indicated that Cd uptake across the plasma membrane was transporter-mediated. While the K m values were similar, the V max values in the presence of acetic and malic acids were respectively 6.0 and 3.0 times that of the control. Zinc transporters were the most probable pathways for Cd accumulation. It was hypothesized that Cd(II)–organic acid complexes associated with the root zone, could decompose and liberate Cd2+ for subsequent absorption by maize roots; and that in the layer of the roots or within the root free space, depletion of Cd2+ was buffered by the presence of Cd(II)–organic acid complexes. Plant response to elevated Cd levels involved overproduction of organic acids in maize roots as a resistance mechanism to alleviate Cd toxicity.  相似文献
