Postpartum ovarian activity, uterine involution and plasma concentrations of calcium and 15-keto-13, 14 dihydro-prostaglandin F2alpha (PGFM) were assessed in dairy cows with retained fetal membranes (n=10) and milk fever (n=10) at parturition. In addition, calcium and PGFM were evaluated in dairy cows affected with uterine prolapse (n=10) and pyometra (n=14). Cows with retained fetal membrane averaged 24.2+/-3.7 d until their first postpartum ovulation, while controls averaged 29.0+/-3.7 d (P>0.10). In cows with retained fetal membranes, the difference in follicular activity between the contralateral and ipsilateral ovaries in relation to the previously gravid uterine horn was appreciably greater post partum when compared with that of the controls. Cows with milk fever had an average of 30.8+/-3.1 d until their first postpartum ovulation, while control cows had an average of 20.4+/-3.3 d (P<0.05). The mean diameter of the uterine horns in cows with milk fever was greater (P<0.05) compared with that of the controls between Days 15-32 post partum. Concentrations of plasma calcium were lower in cows with retained fetal membranes within 24 h after parturition and during the first week post partum than in the controls (6.27+/-0.18 vs 7.40+/-0.18 mg/100ml, P<0.05). Concentration of calcium was lower (P<0.05) in cows with milk fever on Day 1 prior to treatment (4.68+/-0.40 < 5.8+/-0.45 mg/100ml) than in control cows; however, the calcium (Ca) level was not different during the subsequent 7 d post partum after treatment. Cows with uterine prolapse had lower concentrations of Ca during the first 7 d post partum than the controls (6.10+/-0.15 vs 7.33+/-0.12mg/100ml; P<0.01). Forty-eight cows Holstein Friesian x Dutch Friesian (HF x DF) were randomly assigned to 2 groups fed 1 of 2 diets (isocaloric and isonitrogenous but different in linoleic/linolenic acid ratio) from 4 wk before expected parturition until 7 d after calving. Effects of the diet on plasma linoleic/linolenic acid ratio, plasma PGFM levels and placental explusion rate were studied. Dietary treatment resulted in significant differences in linoleic/linolenic acid ratio in blood plasma (1.00 +/- .22 vs 4.41 +/- .53). The placental expulsion rate was not significantly different between the 2 treatment groups. Plasma PGFM levels, as analyzed for 28 cows from 30 d before parturition until 1.5 d after parturition, were similar for the diets. Cows with a longer placental expulsion rate had lower PGFM levels at parturition (for instance, placental expulsion rate shorter (n = 11) and longer (n = 17) than 6 h, 1248 vs 2965 pg/ml, residual standard deviation 1185 pg/ml, P < 0.01). The results show that the dietary linoleic/linolenic acid ratio can influence the plasma linoleic/linolenic acid ratio without affecting the placental expulsion rate or plasma PGFM levels around parturition.  相似文献

Maternal nutrition affects the development of the fetus and postnatal performance of the calf. Methionine may play a critical role in developmental programming and is likely deficient in beef cows fed low-quality forage. The objective of this study was to determine the effect of metabolizable methionine supply to lactating beef cows during the periconception period on performance of cows, calves, and subsequent offspring. This project involved two consecutive production cycles commencing at calving in which dietary treatments were fed to cows during the periconception period along with measurements on cows and initial calves in Production Cycle 1, and measurements on subsequent calves in Production Cycle 2. Brangus-Angus crossbred lactating beef cows (N = 108; age = 6.4 (2.8) year) were stratified by previous calving date and assigned to one of three supplements: (1) control, molasses plus urea at 2.72 kg/day as fed, (2) fishmeal, 2.27 kg/day molasses plus urea plus 0.33 kg/day as fed of fishmeal, and (3) methionine, 2.72 kg/day of molasses plus urea plus 9.5 g/day of 2-hydroxy-4-(methylthio)-butanoic acid. Cows were fed supplements and low-quality limpograss (Hemarthria altissima) hay while grazing dormant bahiagrass (Paspalum notatum Flüggé) pastures during the 115-day periconception period from December 2014 to April 2015 in Production Cycle 1 only. Body weight change and milk yield of cows were measured during the periconception period in Production Cycle 1. Body weight of calves was measured at birth and weaning in both production cycles. Following weaning in Production Cycle 2, eight subsequent steer calves per treatment were individually housed for a 42-day metabolism experiment. Treatment did not affect (P > 0.10) BW change of cows, but cows fed methionine tended (P = 0.09) to produce more energy-corrected milk than control and fishmeal. Treatment did not affect (P > 0.10) 205-day adjusted weaning weight of calves in either production cycle. During the metabolism experiment, subsequent calves from dams fed fishmeal and methionine gained faster (P < 0.05) and had greater (P < 0.05) gain:feed than control calves. Methionine calves tended (P = 0.06) to have greater apparent total tract NDF and ADF digestibility and lesser (P < 0.05) blood glucose concentration than control and fishmeal calves. These data indicate that maternal methionine supply during the periconception period plays an important role in programming future performance of the offspring.  相似文献

Two methods for synchronization of parturition in beef cattle were examined. In the first experiment, four groups of cows and heifers were used: untreated (C, n=9), 10 mg flumethasone on day 281 of gestation (F, n=9), 100 mg progesterone daily from days 276 through 283 and 50 mg progesterone on day 284 (P, n=6), and (P + F on day 284, n=7). Variances in gestation lengths (C, 26.01; F, 0.77; P, 11.97; P + F, 1.93) and proportions of cows and heifers with retained placentas (C, 0 9 ; F, 4 9 ; P, 1 6 ; P + E, 0 7 ) differed significantly among groups. Differences among groups in calving difficulty scores and proportions of dead calves were not significant. Four of the thirteen cows and heifers treated with progesterone required assistance in calving and all four delivered dead calves. Pulling of these calves was not accompanied by uterine contractions. Conversely, the difficult calvings in the control- and flumetha-sone-treated cows and heifers were accompanied by uterine contractions. In the second experiment, two groups of cows were used: 1) a single injection of 20 mg dexamethasone on either day 276, 277 or 278 followed by injections of saline every 12 h for 2.5 additional days (n=6), and 2) repeated injections of 20 mg dexamethasone every 12 h for 3 days beginning on day 276, 277 or 278 (n=8). The interval from time of first treatment to calving was not different between groups (43.6 and 43.0 h, respectively). Differences between calving difficulty scores, proportions of dead calves and incidence of retained placentas were not significant. Induction very close to the expected calving date could reduce the problems of retained placenta; however, methods must be identified to safely delay parturition.  相似文献

The aim of this study was to assess the predisposing factors leading to uterine infection in dairy cows and evaluate its impact on the cow's performance. Performance indicators included feed intake, milk fat and protein composition, milk yield, fertility and culling. Data were studied from 2914 completed lactations from the Holstein Friesian Langhill Dairy herd between January 1990 and August 2005. There were 402 cases of uterine infection (from 321 cows) in the Langhill herd, a level of 13.8% during the study period. Calving assistance was highly associated with the incidence of uterine infection (chi(2)=106.63, P<0.001). Failure to conceive (chi(2)=3.89, P<0.05; chi(2)=11.20, P<0.001) and culling (chi(2)=13.66, P<0.001, chi(2)=8.55, P<0.01) were also implicated with calving assistance and uterine infection, respectively. The main predisposing factors related to an incidence of uterine infection were calving assistance, twin births, malpresented calves, and retained placenta. First-time calvers and cows with a shorter gestation length were associated with more cases of uterine infections. Additionally, lower daily dry matter intakes and milk yields during the first 100 days in milk (DIM) were associated with higher incidence levels of uterine infection. Also, a higher incidence level of uterine infection was related to lower milk protein composition from 1 to 21 DIM and a poorer body condition from 101 to 300 DIM. Cows with poorer drying off body condition scores and longer lactation lengths were associated with a higher incidence level of uterine infection. Calving assistance was associated with male and twin births, first-time calvers and cows with oversized calves. Cows with an assisted calving were also associated with an increased number of days from calving to first service. The predisposing factors influencing the occurrence of calving assistance can be influenced by suitable management to reduce the chances of oversized calves, the detrimental effects of twin births (such as calving assistance and retained placentas) and prenatal knowledge of the calf's sex. The occurrence of uterine infection can be reduced by minimising the need for assistance at calving. Assistance should only be carried out when necessary and appropriate hygiene should be used to decrease the incidence of uterine infections from intervention and involuntary culling due to cows failing to conceive.  相似文献

Two experiments were designed to determine whether pretreatment with Opticortenol (OPT), a long-acting corticosteroid, prior to induction of parturition with 25 mg of dexamethasone (DEX) alone or in combination with 500 mug cloprostenol (CLO) would result in a reduced incidence of retained placenta. In Experiment 1, 70% of the cows pretreated with 25 mg OPT on Day 270 of gestation calved before or within 24 hours of the scheduled induction treatment on Day 277. Cows induced to calve with DEX plus CLO without OPT pretreatment had an increased rate of placental retention (P<0.05), whereas, cows that received OPT were not different from the controls. In Experiment 2, cows received either 1 mg/25 kg OPT (high dosage) or 1 mg/50 kg OPT (low dosage) on Day 270 of gestation and were induced with DEX plus CLO on either Day 274 (4 days) or Day 276 (6 days). Cows claved 29.0 to 31.8 hours after induction treatment with 95% beginning to calve between 0700 and 1900 hours. The interval from calving to placental release and the incidence of retained placenta was not different between the high dosage 6-day group (29.4+/-8.2 hours, 29%) and the non-induced control cows (16.1+/-10.7 hours, 5%). When three cows in the high dosage 6-day group that retained their placentas for 30 to 36 hours were considered as not retained, the incidence of placental retention for that group was reduced still further to 17%. First service conception rates and pregnancy rates were lower in cows with retained placentas. Differences were significant (P<0.01) in Experiment 1 but not in Experiment 2. It was concluded that pretreatment with 1 mg/25 kg OPT 6 days prior to induction of parturition with DEX plus CLO in combination results in a predictable calving time, high calf viability, and a low incidence of placental retention.  相似文献

Peripheral blood plasma concentrations of progesterone and the main metabolite of prostaglandin F, (15-keto-13,14-dihydro-PGF) PGFM, were determined in 10 Holstein cows with retained foetal membranes (RFM) and 12 Holstein cows without RFM (NRFM) during the peripartal period. The rate of uterine involution in the postpartum cows was monitored.There was no difference in the rate of uterine involution between cows with or without RFM. Cyclical ovarian activity was resumed within a month after parturition in both group. Increases in the mean peripheral plasma PGFM concentrations were evident in the RFM cows 6 days before parturition, compared to 48 h before parturition in the NRFM cows. A gradual decline in PGFM to prepartum concentrations occurred in both groups by Day 12 after parturition, although in the RFM cows, PGFM concentrations remained high until the placenta was shed.In both groups, the mean peripheral plasma concentrations of progesterone showed a marked decline beginning 48 h before partusition. The mean plasma progesterone concentrations were less than 1 ng/ml during the immediate postpartum period.  相似文献

Daily individual voluntary intakes of dry matter (DM % of body weight) and estimated net energy (ENE, Mcal/100 kg) by 89 Holstein cows were compared between day 220 of gestation and day 30 postpartum over a 21-month period. The purpose was to compare effects of diet and health status (control vs. abnormal) on intakes of DM and ENE. The cows were fed either chopped hay, hay crop silage (HCS) or corn silage (CS). Compared to controls, voluntary intakes of DM and ENE were decreased (most to least) in cows with fat cow syndrome (FCS), parturient paresis (PP), mastitis (MST), retained fetal membranes-metritis (RFM-M), and displaced abomasum (DA) prepartum and FCS, DA, PP, RFM-M and MST peripartum. Cows fed hay had a lower incidence of RFM-M (28%) than cows fed HCS (57%) and CS (47%) as well as superior reproductive efficiency subsequently (89% conceived vs. 72% and 78%, respectively).  相似文献

The effectiveness of dexamethasone and prostaglandin in combination for induction and synchronization of parturition in cattle was evaluated in 100 pregnant Angus, Hereford, Charolais and Simmental cows. Cows were distributed equally by breed, day of gestation and cow age to one of three treatments: 1) Control, 2) Dexamethasone (25 mg) plus prostaglandin F(2alpha) (25 mg) or 3) Dexamethasone (25 mg) plus fenprostalene (1 mg). Hormones were administered simultaneously from 275 to 283 d of gestation. Gestation length at calving for control cows differed significantly (P < 0.01) among breeds: Angus, 278.5 +/- 0.9; Hereford, 283.1 +/- 1.1; Charolais, 283.2 +/- 1.5; and Simmental, 285.4 +/- 1.2 d. For hormone-treated cows, 80% of the calves were born between 30 and 46 h after the hormone injections; overall mean was 37.6 +/- 1.1 h. Calving response did not differ (P >0.1) between cows treated with prostaglandin F(2alpha) versus fenprostalene (36.5 +/- 1.6 vs 38.6 +/- 1.6 h) or among cow age, day of gestation, or breed. Also, duration of labor, calving difficulty and calf viability did not differ between calves born at an induced or spontaneous parturition. The incidence of placenta retained for >24 h was higher for induced than spontaneous parturition (21.0 vs 0.0%), but it did not differ (P >0.1) between cows treated with prostaglandin F(2alpha) or fenprostalene (19.2 vs 22.6%). An acceptable degree of synchrony of parturition was attained by the administration of prostaglandin F(2alpha) or fenprostalene in combination with dexamethasone. The higher incidence of retained placenta in treated than control cows did not affect subsequent fertility. The longer biological half-life for fenprostalene than for prostaglandin F(2alpha) provided no improvement in increasing synchrony of parturition or decreasing frequency of retained placenta.  相似文献

Blood concentrations of progesterone, 13,14-dihydro-15-keto-prostaglandin F2alpha (PGFM) and endotoxin, and uterine fluid concentrations of prostaglandin E(2) (PGE(2)), PGFM and endotoxin were evaluated in 14 dairy cows with puerperal endometritis (mild (n=6) and heavy (n=8)). Endotoxin was measured using a quantitative kinetic assay. Cows with heavy endometritis had significantly higher concentrations of plasma PGFM (P<0.01) and uterine fluid PGE(2) and endotoxin (P<0.05) than cows with mild endometritis. Concentrations of PGFM in plasma and uterine fluid, of PGFM and PGE(2), and PGE(2) and endotoxin in uterine fluid were positively and significantly (P<0.05) correlated. The presence of endotoxin in plasma was detected in one out of six mild and in eight out of eight heavy endometritis cows. Peak plasma endotoxin concentrations (0.08-9.14 endotoxin units/ml (EU/ml) were observed between 1 and 12 days postpartum (pp) and thereafter amounts generally remained below 0.1 EU/ml (last day of detection: Day 27 pp). Abnormal ovarian function was observed in six cows (four with prolonged anoestrus and two with long luteal phase after the first postpartum ovulation). Plasma endotoxin concentrations were detected in the anoestric cows. The results suggest that: (i) concentrations of uterine fluid endotoxin and PGE(2) and of plasma PGFM are related to the degree of endometritis; (ii) absorption of endotoxin from the uterus to the bloodstream occurs, mainly in heavy endometritis cows; and (iii) there is a relationship between uterine infection, endotoxin production and resumption of pp ovarian activity.  相似文献

The hypothesis was that supplementation during the late prepartum period will differentially affect reproductive and productive variables according to parity. Primiparous (n=22) and multiparous (n=22) pregnant autumn calving Holstein cows were stratified in two groups according to parity (primiparous or multiparous) and within each group were randomly assigned to two treatments: (a) low supplemented (LS) or (b) high supplemented (HS) prepartum diet. The LS group was offered 5.2 kg/cow/day (DM basis) of wheat silage, and the HS group 4.7 kg cow/day (DM basis)/of corn silage and 3.6 kg (DM basis) of wheat bran+12 g of urea. Both groups grazed on natural pastures. After calving, all cows received the same diet. The experimental period was from 3 weeks before calving to 7 weeks postpartum (PP); body condition score (BCS) and blood samples for hormonal analyses were obtained weekly and ovarian ultrasonography was conducted three times per week. The loss in BCS around calving was less pronounced in HS cows, but only multiparous supplemented cows maintained BCS throughout the study. Non-esterified fatty acids (NEFA) increased during the prepartum period in the LS but not in the HS cows, with peak values occurring on day 14 PP in all groups. During the remainder of the experiment NEFA was greater in LS than in HS cows. Prepartum treatment did not affect the proportion of cows that had ovulations from the first dominant follicle postpartum, but decreased the interval to first ovulation in multiparous cows (22.9 compared with 38.2 days; P<0.05). This was associated with greater plasma IGF-I concentrations at the time the dominant follicle of the first follicular wave reached its maximum diameter (8.0 compared with 3.6 nmol/L; P<0.05). However, prepartum treatment had no effect on onset of ovarian activity in primiparous cows. Supplementation had no effect on milk production or milk protein percentage but increased milk fat percentage. We conclude that feeding a high-supplemented prepartum diet to multiparous cows allowed them to maintain BCS around calving, and this was associated with greater concentrations of IGF-I and an earlier onset of estrous cycles after calving.  相似文献

The objective of the experiment was to determine the effects of fat supplementation on cyclicity, progesterone concentration, follicular development, conception rate, embryo mortality, and plasma concentrations of prostalglandin F metabolite (PGFM) in cattle. The hypothesis of this experiment was that feeding flaxseed, which is a source rich in C18:3, would increase conception rate of dairy cows due to decreased plasma PGFM concentrations. A total of 138 lactating Holstein cows were allotted at calving to three groups of 46 cows, blocked for similar calving dates. Cows within each block were assigned to one of three isonitrogenous, isoenergetic, and isolipidic supplements based on either whole flaxseed (FLA), Megalac (MEG) or micronized soybeans (SOY). The diets were fed from calving to Day 50 of pregnancy for pregnant cows, or 120 day postpartum for those not diagnosed pregnant after AI. Detailed measurements of PGFM and follicle dynamics were only made on four cows for FLA and five cows for both MEG and SOY. The response in PGFM concentration following the oxytocin challenge administered around Week 11 of lactation was similar over time among treatments. Plasma progesterone concentrations from Days 17 to 21 of the estrous cycle starting around Week 9 of lactation and determined on a subsample of cows (n=for FLA and n=5 for both MEG and SOY) were higher for cows fed FLA than for those fed SOY (P=0.04) or MEG (P=0.06). Conception rates were similar among treatments. Total embryo mortality was lower (P=0.07) for cows fed FLA (0%) compared to those fed either MEG (15.4%) or SOY (8.0%). The mean size of the CL measured during a complete estrous cycle from Week 9 of lactation was smaller for cows fed SOY (16.3 mm) compared to those fed either FLA (19.1 mm) or MEG (18.3 mm). We inferred that pregnancy losses could be reduced by feeding whole flaxseed as a result of its effects on different factors such as modulation in concentration of progesterone and size of the CL.  相似文献

This study was carried out in central Tanzania on a group of 45 Zebu and 37 crossbred cows which were 4 to 10 years old. At calving time, the animals were allocated to one of the 4 treatment groups. In addition to free access to grazing for all cows in the study, in group H: AR (n = 18), cows were fed a high level of concentrate supplementation (4kg/day) and calves were artificially reared; in group H:RS (n = 24), cows were fed a high level of concentrate supplementation (4kg/day) and calves were only allowed restricted suckling up until the weaning age of 6 months. In group L:AR (n = 23) cows were fed a low level of concentrate supplementation (2kg/day) and calves were artificially reared; and in group L:RS (n = 17) cows were fed a low level of concentrate supplementation (2kg/day) and calves were only allowed restricted suckling up until the weaning age of 6 months. Milk progesterone was used as a means of determining the postpartum resumption interval (PRI) and the interval from parturition to conception (PCI). The overall PRI was 47.4 ± 0.4 days and was significantly affected by breed but not by calving season, with crossbred cows exhibiting a shorter PRI than Zebu cows. The effect of the treatments was significant, with cows in the group H:AR displaying a significantly shorter PRI than those in the other groups, while cows in group L:RS showed a significantly longer PRI than those in the other groups. The overall PCI was 149.5 ±3.7 days, and was not significantly affected by breed or calving season. The effect of the treatments was significant, with cows in the group H:AR having a significantly shorter PCI than cows in the other groups, while cows in group L:RS showed a significantly longer PCI than those in the other groups. Crossbred cows had higher live weights at calving (299.4 kg) than Zebu cows (272.6 kg), while all cows gained weight during the first 3 months after calving. The treatments had a significant effect on weight gain, with cows in the group H:AR gaining significantly more weight than those in the other groups. Cows which had high live weights at calving exhibited significantly shorter PRI and PCI than the lighter cows. Animals which had gained more than 5 kg during the first month after calving, or which had gained more than 8 kg during the first 3 months after calving, showed significantly shorter PRI and PCI than cows which had gained less weight. The results show that the calf rearing system and the level of feed supplementation interact with each other and can influence the postpartum anoestrous period in Zebu and Zebu crossbred cattle. Increasing the level of nutrition in restricted suckling cows tended to improve the postpartum anoestrous period, but the positive effects of supplementation could not completely compensate for the negative effects of suckling.  相似文献

A dairy herd experienced an increased incidence of retained placenta and postpartum metritis during late spring and summer of 1977 and 1978. The incidence of retained placenta and postpartum metritis was 24.05% during the period of May through September compared to 12.24% the rest of the year. The gestation period of cows having retained placenta and postpartum metritis was an average of 5.25 days shorter than that of cows not exhibiting these symptoms. The gestation period of cows calving during the warm season was an average of 2.82 days shorter than that of the cows calving during the cool season. Heat stress resulting in early parturition was the probable cause of this increased incidence of retained placenta and postpartum metritis. The number of days open was influenced both by abnormal postpartum period and season of calving. Cows having retained placenta and metritis were open an average of 31.69 days longer than cows with normal postpartum periods. Cows that calved during the warm season were open an average of 24.42 days longer than those calving during the cool season.  相似文献

Animal performances were monitored in 30 Friesean dry cows (18 multiparous, MP-cows and 12 primiparous, PP-cows) starting six weeks before calving to eight weeks after calving. The cows were kept indoors and fed individually with a prepartum diet containing either low, moderate or high energy (0.75, 1.00 or 1.25 of the calculated ME requirement) and supplemented with low (0.3 kg day−1) or high (1.5 kg day−1) rapeseed meal (RSM). The diets were consisted of 1.5 kg hay, 20–25 kg wilted grass silage and grain (barley and oat, 1 : 1 DM basis) with RSM. The average ME intake during six weeks of prepartum was 75, 97 and 123 MJ day−1 on three different energy levels. After calving the cows were fed grass silage ad lib, 6.5 kg (primiparous) or 8.5 kg (multiparous) grain and 1.5 kg RSM. Liver biopsy and blood samples were taken to determine differences in metabolites due to diet and parity. Reduction of prepartum energy allowance to 0.75 of the moderate level did not affect the calf's birth weight and colostrum composition. Cows fed prepartum low energy–high rapeseed meal diet (LEHR) showed a faster increase in feed intake and milk production after parturition. Continuous low feed intake and milk production were observed in cows fed a prepartum high energy–low rapeseed meal diet (HELR). Milk yield was constantly lower for cows fed prepartum high energy diet, the difference being significant (p<0.01 and p<0.05) at six and seven weeks of lactation. Overall milk yield also showed a linear decrease with energy feeding level and an increase (p<0.10) with RSM feeding levels. Prepartum high energy feeding increased (p<0.01) milk protein content. Neither energy nor RSM feeding level affected the liver fat infiltration of the experimental cows. Cow parity however showed a significant influence on liver fat content, β-hydroxybutyrate (BHBA) and plasma glucose. A week before calving, the plasma concentration of essential amino acids (EAA) was lower for cows fed prepartum low energy diet. During the first four weeks of lactation, the concentrations of plasma non-esterified fatty acids (NEFA) and insulin were similar for all the treatment group.  相似文献

Daily voluntary intakes of feed by each of 89 Holstein cows were compared between day 220 of gestation and day 30 postpartum over a 21-month period. Diets designed to meet NRC requirements and which contained either chopped hay (29 cows), hay crop silage (HCS; 30 cows) or corn silage (CS; 30 cows) were compared prepartum (27 to 0 days), peripartum (1 day before to 3 days after calving) and postpartum (days 4 to 30 postpartum). Mixed rations, fed during lactation, were 60% forage and 40% concentrate dry matter (DM). Cow management was similar to commercial operations. The experimental hypothesis was that pre-disposition for partum and postpartum disorders (abnormal) could be related either to voluntary intake of different diets or to physical traits. Intakes (DM or estimated net energy) across diets decreased 30% between days 7 and 1 prepartum and averaged 18% and 20% lower peripartum and postpartum, respectively, in abnormal cows than in control cows. Seasonal effects on intakes were significant. In general, changes in body weight and condition and differences in udder edema and milk yield reflected intakes. Peripheral serum concentrations of prostaglandin F(2)alpha metabolite (PGFM), cortisol, and progesterone (P(4)) were determined in cows that retained placentae (RP; n = 18) and cows that did not retain placentae after parturition (NRP; n = 28). Blood samples collected from Day 15 before until Day 15 after parturition were assayed. Significant increases (P < 0.05) in PGFM concentrations occurred in RP cows between Days 7 and 6 before parturition compared with NRP cows. There were no differences in the interval required to attain peak concentrations of PGFM and the interval required to reach basal concentrations after parturition in both groups. However, the amount of PGFM secreted during the peripartum period was significantly higher in RP cows (P < 0.01). Starting from Day 6 before parturition, serum cortisol concentrations in RP cows increased significantly to peak levels 3 d before parturition (P < 0.05). Cortisol levels decreased to basal concentrations in RP cows on Day 1 postpartum. The peripartal serum P(4) concentrations in RP and NRP cows did not differ. The results indicate that prepartal PGFM and cortisol increases may constitute reliable indicators of retained placenta in dairy cows.  相似文献

Plasma progesterone was determined twice weekly for approximately 100 days postpartum in suckled purebred (Hereford) and crossbred (beef x dairy) cows that calved in the spring or late summer/fall season. The progestèrone profiles and occurrence of estrus were used to determine ovulation times and to monitor ovarian function. Postpartum ovulations occurred significantly earlier in crossbred than in purebred cows (38.1 ± 18.5 vs. 58.1 ± 21.8, P<0.01) and in fall compared to spring calving cows (32 ± 13.9 vs. 59.1 ± 20.3, P<0.001). Rations providing either 70 or 100% of requirements for metabolizable energy were fed from 30 days prepartum until the end of the subsequent rebreeding period. Cows receiving the 70% energy ration ovulated slightly earlier but there was no effect of ration on days to pregnancy. The minimal effect of energy ration was not unexpected in this trial since many of the cows were overconditioned during late gestation.The correlation between calendar date of calving and interval to first ovulation was significant for spring (r = ?0.38, P<0.01) but not for fall calving animals. Since cows were confined and received a balanced ration of stored feeds throughout the year, photoperiod and/or temperature rather than nutritional factors would be the probable cause of delayed ovarian activity in spring calvers.Reproductive performance was assessed during the period when samples were collected and the response for purebred (n=105) and crossbreds (n=142), respectively were: ovulated by day 60 postpartum, 57 vs. 87%; mated by day 100 without conception, 17 vs. 26%; ovulated before day 100 without detection and mating, 10 vs. 4%; anovulatory to day 100, 7 vs. 0%; pregnant by day 100, 67 vs. 70%. Similar comparisons for spring (n=133) and summer/fall (n=144) calvers, respectively, were: ovulated by 60 days postpartum, 56 vs. 96%; mated by day 100 without conception, 27 vs. 17%; ovulated by day 100 without detection and mating, 8 vs. 5%; anovulatory to day 100, 5 vs. 0%; pregnant by day 100, 60 vs. 78%. The difference with the greatest practical significance is that a higher proportion of the late summer/fall calving animals were pregnant by day 100 postpartum (P<0.01) which indicates that reproductive performance is superior in fall calving beef cows.  相似文献

The plasma estrogen and progesterone concentrations of 19 pregnant cows (average duration of pregnancy 266.0 +/- 2.3 d at the start of the study) were determined daily from Day 6 pre partum to Day 1 post partum. Parturition was induced in all cows by administration of 10 mg i.m. flumethasone. Values were centered around the delivery date (Day 0) following either induced normal calving (n = 3) or surgical delivery (n = 16). In animals showing spontaneous expulsion of the fetal membranes (Group 1, n = 6) the average total estrogen concentration increased significantly from Day 6 until Day 1 before parturition (1329.2 +/- 317.9 to 3719.8 +/- 951.2 pg/ml in total estrogens). A marked decrease was observed on Day 1 post partum (459.4 +/- 344.2 pg/ml). In comparison with Group 1, animals showing either a delayed or partial expulsion of the fetal membranes, or in which the placenta could be withdrawn 16 h after calving (Group 2, n = 5), had consistently lower total estrogen concentrations between Day 6 (595.4 +/- 174.8 pg/ml) and Day 1 (1884.3 +/- 565.1 pg/ml) before parturition. The estrogen values of the cows with retained placenta (Group 3, n = 8) from Days 6 to 0 pre partum were significantly lower than those of Group 2. Total estrogen concentrations of the three groups 1 d post partum did not differ significantly. It is generally recognized that estrogens play an important role in the maturation process of the placentomes. Our investigation demonstrates that not only is the magnitude of the prepartum rise in estrogens of great influence of the maturation process but the duration of this rise is likewise important. These two factors are vital for a normal expulsion of the fetal membranes.  相似文献

In this study we characterized the peripheral plasma pregnancy-specific protein-B (PSPB) profile throughout gestation and examined the effect of stage of gestation, fetal mass and number on this profile in Holstein cows after non surgical embryo transfer. Cows (n = 12) were divided into 2 groups: Group 1 = single embryo recipient cows (n = 5), Group 2 = twin-embryo recipient cows (n = 7). Blood was collected approximately every third day from Day 0 (Day 0 = first day of standing estrus), then daily for the last 10 d of gestation, and sampling was stopped 1 d post partum. Two twin-embryo recipient cows had abnormal pregnancies; therefore, their data were excluded from the group. The time trend concentrations of plasma PSPB were significantly affected by the stage of gestation (P < 0.001) and fetal number (P < 0.001). In both groups PSPB increased gradually, with the mean levels being significantly higher (P < 0.01) in the twin-bearing group from Day 50 onwards (0.7 +/- 0.2 vs 9.2 +/- 4.5 ng/ml, singleton and twin-bearing cows, respectively) except for Day 10 pre-partum. By mid-gestation (Day 140), mean PSPB levels increased in the singleton (P < 0.001) cows by thirty-fold (21.2 +/- 3.2 ng/ml) as opposed to a ten-fold (98.4 +/- 13.2 ng/ml) increase in the twin-bearing (P < 0.001) group. The mean PSPB concentrations between Days 30 to 20 prepartum dramatically increased by about 700 to 200% in singleton (128.8 +/- 46.3 to 745.6 +/- 66.7 ng/ml) and twin-bearing cows (375.6 +/- 130.4 to 861.5 +/- 127.9 ng/ml), respectively. The PSPB levels between Day 10 prepartum to parturition were significantly higher (P < 0.001) in the twin-bearing group than in the singleton group (745.6 +/- 66.7 to 1627.4 +/- 238.9 ng/ml vs 861.5 +/- 127.9 to 3103.0 +/- 643.0 ng/ml in singleton and twin-bearing groups, respectively). Calf birthweight was correlated (P < 0.01) to peripheral PSPB concentration in singleton cows; however, this relationship decreased with the subsequent increase in fetal number. Cows giving birth prematurely to stillborn calves or to a schistosomus reflexus calf exhibited abnormal PSPB profiles. These results indicate that peripheral PSPB levels are correlated to the stage of gestation and fetal number. In addition, the peripheral pattern of PSPB is a valuable guage for predicting fetoplacental viability.  相似文献
