During the last 15 years, European stone fruit yellows (ESFY) has become a major concern in Austrian fruit production. Therefore, presence and temporal dynamics of its vector Cacopsylla pruni were investigated using a beating tray method and yellow sticky traps on Prunus armeniaca, Prunus domestica, Prunus spinosa and P. cerasifera nigra. Infection rates of C. pruni and Prunus spp. trees were assessed by direct, nested and real‐time PCR. Movement of remigrants in a model apricot orchard was tracked by aid of a mark, release and recapture study. Insects were marked by fluorescent dyes. Movement of the marked insects and presence of naturally occurring insects were monitored by yellow sticky traps. In 2011, remigration of C. pruni to Prunus spp. started in calendar week 10 (8th of March) and in 2012, in calendar week 12 (18th of March). Remigrants were observed until calendar week 20 (middle of May), significant numbers of the springtime generation adults were present until week 26 (end of June). The phytoplasma was ascertained in 0–11.5% of the remigrants and in 0–3.44% of the springtime generation insects. About 9.8–63.3% of the apricot samples, 20–40% of the plum samples and single blackthorn samples were infected. The mark, release and recapture study proved a fast and frequent tree‐to‐tree movement of remigrated C. pruni adults. Insects easily covered distances from row to row or even farther (ca. 13 m) within 24 h after release and were present in a large part of the model orchard after 8 days (up to 24 m from release point).  相似文献

Polyploid Prunus spinosa (2n?=?4?×) and P. domestica subsp. insititia (2n?=?6?×) represent enormous genetic potential in Central Europe, which can be exploited in breeding programs. In Hungary, 16 cultivar candidates and a recognized cultivar ‘Zempléni’ were selected from wild-growing populations including ten P. spinosa, four P. domestica subsp. insititia and three P. spinosa?×?P. domestica hybrids (2n?=?5?×) were also created. Genotyping in eleven simple sequence repeat (SSR) loci and the multiallelic S-locus was used to characterize genetic variability and achieve a reliable identification of tested accessions. Nine SSR loci proved to be polymorphic and eight of those were highly informative (PIC values ? 0.7). A total of 129 SSR alleles were identified, which means 14.3 average allele number per locus and all accessions but two clones could be discriminated based on unique SSR fingerprints. A total of 23 S-RNase alleles were identified and the complete and partial S-genotype was determined for 10 and 7 accessions, respectively. The DNA sequence was determined for a total of 17 fragments representing 11 S-RNase alleles. ‘Zempléni’ was confirmed to be self-compatible carrying at least one non-functional S-RNase allele (SJ). Our results indicate that the S-allele pools of wild-growing P. spinosa and P. domestica subsp. insititia are overlapping in Hungary. Phylogenetic and principal component analyses confirmed the high level of diversity and genetic differentiation present within the analysed accessions and indicated putative ancestor–descendant relationships. Our data confirm that S-locus genotyping is suitable for diversity studies in polyploid Prunus species but non-related accessions sharing common S-alleles may distort phylogenetic inferences.

2C DNA content and ploidy level variation of Prunus spinosa and closely related taxa together with Prunus domestica L. and Prunus insititia L. was studied in Slovakia. The aim of the study was to define genome sizes and find differences between closely related taxa within Prunus spinosa sensu lato mentioned in previous works. According to our results, investigated taxa can be divided into three groups according to ploidy level: Prunus spinosa, Prunus dasyphylla, Prunus ×fruticans, Prunus ×dominii and Prunus ×schurii are tetraploids, Prunus ×fechtneri is pentaploid, and P. domestica and P. insititia are hexaploids. Genome size differences within tetraploid taxa were relatively small (Prunus spinosa: 1.40?±?0.02, P. ×domini: 1.44?±?0.01, P. ×fruticans: 1.48?±?0.02, P. ×schurii: 1.44?±?0.02), but statistically significant. Although further research is needed, it seems that the concept of several taxa as product of hybridization between P. spinosa and cultivated plum species has been supported by our study.  相似文献

Polyploid Prunus spinosa (2n = 4×) and P. insititia (2n = 6×) represent enormous genetic potential in Central Europe, which can be exploited in breeding programmes. In Hungary, 17 cultivar candidates were selected from wild-growing populations including 10 P. spinosa, 4 P. insititia and three P. spinosa × P. domestica hybrids (2n = 5×). Their taxonomic classification was based on their phenotypic characteristics. Six simple sequence repeats (SSRs) and the multiallelic S-locus genotyping were used to characterize genetic variability and reliable identification of the tested accessions. A total of 98 SSR alleles were identified, which presents 19.5 average allele number per locus, and each of the 17 genotypes could be discriminated based on unique SSR fingerprints. A total of 23 S-RNase alleles were identified. The complete and partial S-genotype was determined for 8 and 9 accessions, respectively. The identification of a cross-incompatible pair of cultivar candidates and several semi-compatible combinations help maximize fruit set in commercial orchards. Our results indicate that the S-allele pools of wild-growing P. spinosa and P. insititia are overlapping in Hungary. A phylogenetic and principal component analysis confirmed the high level of diversity and genetic differentiation present within the analysed genotypes and helped clarify doubtful taxonomic identities. Our data confirm that S-locus genotyping is suitable for diversity studies in polyploid Prunus species. The analysed accessions represent huge genetic potential that can be exploited in commercial cultivation.  相似文献

Domestication is an excellent model to study diversification and this evolutionary process can be different in perennial plants, such as fruit trees, compared to annual crops. Here, we inferred the history of wild apricot species divergence and of apricot domestication history across Eurasia, with a special focus on Central and Eastern Asia, based on microsatellite markers and approximate Bayesian computation. We significantly extended our previous sampling of apricots in Europe and Central Asia towards Eastern Asia, resulting in a total sample of 271 cultivated samples and 306 wild apricots across Eurasia, mainly Prunus armeniaca and Prunus sibirica, with some Prunus mume and Prunus mandshurica. We recovered wild Chinese species as genetically differentiated clusters, with P. sibirica being divided into two clusters, one possibly resulting from hybridization with P. armeniaca. Central Asia also appeared as a diversification centre of wild apricots. We further revealed at least three domestication events, without bottlenecks, that gave rise to European, Southern Central Asian and Chinese cultivated apricots, with ancient gene flow among them. The domestication event in China possibly resulted from ancient hybridization between wild populations from Central and Eastern Asia. We also detected extensive footprints of recent admixture in all groups of cultivated apricots. Our results thus show that apricot is an excellent model for studying speciation and domestication in long‐lived perennial fruit trees.  相似文献

Multivariate analysis of both endocarp and leaf morphometrics is combined with cluster analysis and Bayesian inference of AFLP markers to assess the morphologic and genetic variation of five European members of Prunus section Prunus (P. cerasifera, P. domestica, P. insititia, P. spinosa, and P. × fruticans). Endocarp morphometrics separate most Prunus taxa studied, but overlap remains between P. domestica and P. cerasifera, and P. spinosa and P. × fruticans. Leaf morphometrics yield better separation of P. domestica and P. cerasifera, but do not allow distinction between P. spinosa and P. × fruticans. Both cluster analysis and PCoA of AFLP markers equally produce three distinct clusters. A first consists of all P. cerasifera samples and the sole P. cocomilia; a second cluster includes all individuals of P. domestica and P. insititia; and a third group comprises all P. spinosa and P. × fruticans samples.  相似文献

The efficacy and the instant effect of 13 insecticides and antifeedants towards Cacopsylla pruni, the vector of ‘Candidatus Phytoplasma prunorum’ were examined in cage studies (no choice experiments with 10 field‐collected overwintered adults per experiment) on potted apricot trees (budding trees under outdoor conditions in early spring and foliated seedlings kept at 21°C). Cypermethrin caused 100% insect mortality within 2–4 h, thiacloprid 90–100% mortality within 24 h both on foliated and on budding trees. On budding trees spinosad led to 70–90% mortality within 24 h, thixotropic white trunk paint to 90% mortality within 48 h. On foliated seedlings flonicamid gave 70–100% mortality within 1 day, abamectin, spinosad, acetamiprid and spirotetramat 70–100% within 72 h. Field studies monitoring the effects of thiacloprid on remigrants of C. pruni by yellow sticky traps were carried out in two apricot orchards. Additionally the influence of the insecticide on insect dispersal was examined by mark, release and recapture trials. As compared to the control thiacloprid significantly reduced the catches of naturally occurring and released insects, decreased the number of trees on which released insects were recaptured (by 25–100%) and shortened the migration distances of the released insects by more than half. Our results suggest that appropriate insecticide treatments both reduce C. pruni populations and have a direct effect on pathogen transmission. Application of Cypermethrin before bloom and thiacloprid after bloom seem best suited to achieve these objectives. Thixotropic white trunk paint could, a formulation for spray application provided, eventually be a sustainable alternative or complement for treatments before bloom.  相似文献

A survey was made to determine the incidence of phytoplasmas in 39 sweet and sour cherry, peach, nectarine, apricot and plum commercial and experimental orchards in seven growing regions of Poland. Nested polymerase chain reaction (PCR) using the phytoplasma‐universal primer pairs P1/P7 followed by R16F2n/R16R2 showed the presence of phytoplasmas in 29 of 435 tested stone fruit trees. The random fragment length polymorphism (RFLP) patterns obtained after digestion of the nested PCR products separately with RsaI, AluI and SspI endonucleases indicated that selected Prunus spp. trees were infected by phytoplasmas belonging to three different subgroups of the apple proliferation group (16SrX‐A, ‐B, ‐C). Nucleotide sequence analysis of 16S rDNA fragment amplified with primers R16F2n/R16R2 confirmed the PCR/Restriction Fragment Length Polymorphism (RFLP) results and revealed that phytoplasma infecting sweet cherry cv. Regina (Reg), sour cherry cv. Sokowka (Sok), apricots cv. Early Orange (EO) and AI/5, Japanese plum cv. Ozark Premier (OzPr) and peach cv. Redhaven (RedH) was closely related to isolate European stone fruit yellows‐G1 of the ‘Candidatus Phytoplasma prunorum’ (16SrX‐B). Sequence and phylogenetic analyses resulted in the highest similarity of the 16S rDNA fragment of phytoplasma from nectarine cv. Super Queen (SQ) with the parallel sequence of the strain AP15 of the ‘Candidatus Phytoplasma mali’ (16SrX‐A). The phytoplasma infecting sweet cherry cv. Kordia (Kord) was most similar to the PD1 strain of the ‘Candidatus Phytoplasma pyri’ (16SrX‐C). This is the first report of the occurrence of ‘Ca. P. prunorum’, ‘Ca. P. mali’ and ‘Ca. P. pyri’ in naturally infected stone fruit trees in Poland.  相似文献

Zonate leaf spots and severe defoliation were observed on Manchurian apricot (Prunus mandshurica) growing in a humid location in Korea from 2011 through 2013. The main symptoms included greyish green to brownish grey and zonate leaf spots without border lines, which mostly led to premature defoliation. The morphological characteristics of the causal agent were consistent with Hinomyces pruni. Identification was supported by analysing the sequence of the internal transcribed spacer region of ribosomal DNA from an isolate. The pathogenicity of the isolate was confirmed by artificial inoculation. This is the first report of zonate leaf spot caused by H. pruni on Manchurian apricot globally as well as in Korea.  相似文献

A quick, simple, and reliable method for the extraction of DNA from grapevine species, hybrids, andAmpelopsis brevipedunculata (Vitaceae) has been developed. This method, based on that of Doyle and Doyle (1990), is a CTBA-based extraction procedure modified by the use of NaCl to remove polysaccharides and PVP to eliminate polyphenols during DNA purification. The method has also been used successfully for extraction of total DNA from other fruit species such as apple (Malus domestica), apricot (Prunus armeniaca), cherry (Prunus avium), peach (Prunus persica), plum (Prunus domestica), and raspberry (Rubus idaeus). DNA yield from this procedure is high (up to 1 mg/g of leaf tissue). DNA is completely digestible with restriction endonucleases and amplifiable in the polymerase chain reaction (PCR), indicating freedom from common contaminating compounds.  相似文献

Plant remains from the 15th century drains at Paisley Abbey, Scotland include medicinal plants which may have grown in the abbey's physic garden. They are Chelidonium majus, Conium maculatum, Euphorbia lathyris, and Papaver somniferum. Plants with both medicinal and culinary uses are Rumex pseudoalpinus and cf Armoracia rusticana. Other vegetables are represented by Allium sp. and Brassica spp. Malus domestica and Prunus domestica ssp. insititia would have been grown in the abbey's orchard. Juglans regia, represented by nut and wood fragments, was either grown in the orchard or imported. Ficus carica was certainly imported as dried fruit from the Mediterranean region. Myristica fragrans as mace came from Indonesia. Locally grown plants are Avena strigosa, Hordeum, Triticum/Secale, Linum usitatissimum and the dye plant Reseda luteola. It is known that spices and other foodstuffs were purchased at fairs at Berry, Bruges and Antwerp and imported into Scotland at the end of the 15th century.  相似文献

Euseius stipulatus (Athias-Henriot) is a predatory mite widespread in the Mediterranean region considered to be important for the biological control of spider mites in citrus orchards. Development, survival and reproduction of this phytoseiid mite feeding on seven commercially obtained pollen were studied under constant laboratory conditions (20 ± 1°C, RH 65 ± 5%, photoperiod 16L: 8D h). Mites were kept individually at rearing units with ample quantity of almond (Prunus amygdalus Batch), apple (Malus domestica Borkh), apricot (Prunus armeniaca L.), cherry (Prunus avium L.), pear (Pyrus communis L.), plum (Prunus domestica L.) and walnut (Juglans regia L.) pollen as food source. Developmental time from egg to adult varied between the several pollen tested from 8.38 ± 0.08 to 9.58 ± 0.11 days for females and from 8.23 ± 0.12 and 9.07 ± 0.12 days for males. Female longevity varied from 11.53 ± 1.22 to 51.38 ± 2.45 days, while fecundity ranged from 22.84 ± 2.30 to 43.61 ± 3.78 eggs/female. The predator was unable to reproduce when feeding on walnut pollen. Data were submitted to life table analysis and values of the intrinsic rate of increase were derived, ranging from 0.079 to 0.146 (day−1). The cumulative Weibull function that was used to describe the age specific survival of females produced excellent fits to the survival data. Results show that almond, plum, cherry and apricot pollen possess higher nutritional value for E. stipulatus than pear and apple pollen and thus may contribute in sustaining and increasing the predator population in field conditions. Walnut pollen can be utilized by the predator only to survive during short periods of time when principal or alternative food sources are scarce.  相似文献

  • 1 We elucidated the life cycles of two jumping plant lice species (Hemiptera: Psyllidae): Cacopsylla picta, a vector of the apple proliferation phytoplasma (Candidatus Phytoplasma mali), and Cacopsylla melanoneura, a nonvectoring species in Germany and some neighbouring countries, which may transmit the phytoplasma in one region in Italy.
  • 2 The adults of C. picta reproduce exclusively on apple and migrate soon after emergence (emigrants) to conifers in mountainous regions, and return to apple plants in early spring (remigrants). Cacopsylla melanoneura also uses conifers as overwintering host plants but prefers to reproduce on hawthorn, despite its ability to reproduce on apple.
  • 3 Both psyllid species used chemical cues for the identification of their alternate host plants during migration. Remigrants of C. melanoneura preferred the odour of their main reproduction host plant hawthorn to apple but preferred the odour of apple when experienced by feeding and oviposition. Although emigrants of C. picta reportedly prefer the odour of apple trees infected by Ca. P. mali, the remigrants of both species did not distinguish between the odours of infected or uninfected apple plants.
  • 4 Investigating the distribution of Ca. P. mali in plant species involved in psyllid life cycle revealed that the phytoplasma is specialized on apple.
  • 5 Infection of apple by Ca. P. mali increased mortality and resulted in decreased body size of C. picta offspring.

  • 7 It is concluded that Ca. P. mali indirectly promotes its acquisition from infected plants and transmission to non‐infected plants by behavioural manipulation of its vector C. picta.

We studied the reproductive biology ofPrunus spinosa andPrunus mahaleb (Prunoideae, Rosaceae) in the northwest Iberian Peninsula. The two species flowered at the same time (peaking on March 9 and 11, respectively in 1990) but differ significantly in their fruit maturation times. Nectar volume peaked in the early morning in both species, and was ten times greater inP. spinosa than inP. mahaleb. Neither species shows apomixis, nor does fruit-set occur if pollinators are excluded. In both species self-pollination resulted in fewer fruits than open pollination. The principal pollinators belong to theApidae family (79% and 63% of visits toP. spinosa andP. mahaleb, respectively). Results are compared with those for other rosaceous plants with fleshy fruits.  相似文献

Nineteen cultivars of apricot (Prunus armeniaca) were distinguished using random amplified polymorphic DNA (RAPD) markers. One decamer out of 44 used was useful to differentiate cultivars of the Campania Region from those of Northern Italy, North America and Greece. A sequence characterized amplified region (SCAR) marker was obtained. The results provide a protocol to fingerprint DNA of apricots as an efficient way to quality control and fraud prevention.  相似文献

Aphidius transcaspicus Telenga (Hymenoptera: Braconidae: Aphidiinae) has been selected for use as a biological control agent against Hyalopterus pruni Geoffroy (Hemiptera: Aphididae) in prune orchards in California. In this study we focus on the host instar preference and functional response of A. transcaspicus. Host instar preference was determined using laboratory arenas by allowing female parasitoids to attack aphids on field-collected leaves of Prunus domestica L. supporting a natural range of H. pruni instars. A relative preference index indicated that A. transcaspicus was 1.4 times more likely to attack intermediate instars (2–4) than 1st or adult instars. The functional response of A. transcaspicus to H. pruni density was determined on enclosed P. domestica shoots in a prune orchard. The functional response to host density was linear (Type I) at the lowest aphid densities, and the asymptotic number of aphids attacked at higher aphid densities was 64.3?±?6.7 aphids day?1.  相似文献

We assessed host preference of adult plum curculio, Conotrachelus nenuphar (Herbst) (Coleoptera: Curculionidae), based on the total number of mark‐released and wild adults recovered and the total distance moved by mark‐released adults in an orchard whose layout was designed to specifically allow foraging plum curculios to choose among host tree species. Host trees included apple, Malus domestica Borkh.; pear, Pyrus communis (L.); peach, Prunus persica (L.) Batsch; apricot, Prunus armeniaca L.; tart cherry, Prunus cerasus L.; sweet cherry, Prunus avium (L.); European plum, Prunus domestica L.; and Japanese plum, Prunus salicina Lindl. (all Rosaceae). We released 2900 marked adults and recovered 17.7%. We used screen traps to provide a measure of the number of adults that arrived at and climbed up particular host trees and found that significantly greater numbers of marked adults and the greatest number of wild adults were recovered from screen traps attached to Japanese plum. We sampled host tree canopies by tapping limbs to provide a measure of the number of adults within a tree canopy at a particular moment. Again, significantly greater numbers of marked and wild adults were recovered from plum species, with no difference between Japanese and European plum cultivars for marked individuals, but with significantly greater numbers of wild individuals recovered from Japanese plum. The preference index (PI) for Japanese plum based on total distances moved by all marked adults recovered on Japanese plum divided by the total distance moved by marked adults recovered on other host trees indicated that Japanese plum was the most highly preferred host, followed by European plum, peach, sweet cherry, tart cherry, apricot, apple, and pear, respectively.  相似文献

Seventeen isolates of Entoloma clypeatum, a fungus associated with rosaceous woody plants and suspected to act as a root pathogen, were obtained from fruit-bodies collected at 7 localities in the Czech Republic. The fungus grew best on the medium supplied with cellobiose. The growth of E. clypeatum was inhibited by fungicide preparation Ridomil Gold MZ 68WP. A part of the LSU rRNA gene of 11 selected isolates was sequenced and two reverse primers recognizing the sequence motifs in the LSU rRNA gene of E. clypeatum and other related species associated with rosaceous woody plants were designed. One of these primers, NL4EC2, showed high specificity towards DNA of Entoloma spp. of interest and may be used for detection of these organisms in soil, in roots or in the substrate where the fruit trees are cultivated. The results of in vitro inoculation experiment suggest a resistance of roots of young Prunus domestica plants to colonization by Entoloma clypeatum, and the fungus could be detected only in the surrounding substrate. We were able to detect Entoloma spp. in soil and in roots of Prunus avium but only if the fructification of the fungus occurred at the locality. The method is robust towards the false positive detection caused by nonspecific amplification of DNA of other soil microorganisms.  相似文献

Epidemiology of European stone fruit yellows was studied by focussing on the life cycle and transmission characteristics of the vector Cacopsyllapruni. The proportion of both phytoplasma positive and inoculative insects was determined for the first C. pruni adults back colonising the stone fruit trees in spring and for the new generations of the vector, hatched at the beginning of summer. We showed that in spring, as soon as the insects moved to stone fruit trees from shelter plants, they were infective. After the vector fed on infected stone fruit trees, the proportion of phytoplasma positive insects increased. The new generation colonising Prunus species also acquired the phytoplasma from their hosts although several of these insects completed the latency period on secondary hosts. Results showed that the risk of natural transmission of European stone fruit yellows-phytoplasma by C. pruni within orchards is high when the vector is present. These results have implications for the control of European stone fruit yellows.  相似文献

During a survey in a limited area of the Shanxi province in China, phytoplasma symptoms were observed on woody plants such as Chinese scholar tree, apple, grapevine and apricot. The polymerase chain reaction/restriction fragment length polymorphism (PCR/RFLP) analyses on the phytoplasma 16S ribosomal gene confirmed that symptomatic samples from all these species were infected by phytoplasmas. The molecular characterization of the pathogen, performed also with sequencing of polymerase chain reaction amplified 16S rDNA, showed that the phytoplasmas detected in all plant species tested are closely related with stolbur, but two samples from a Chinese scholar tree were infected with phytoplasmas related to ‘Candidatus Phytoplasma japonicum’. The presence of RFLP polymorphism was found in the 16S rDNA amplicons with three of the six enzymes employed in the majority of phytoplasma strains studied.  相似文献
