The mature female conceptacle of Sargassum horneri (Turner) C. Agardh has an ostiole filled with a gelatinous plug. The oogonium in the conceptacle has cell walls that can be differentiated into a dense outer and a less dense inner microfibrillar layer. Just prior to egg release, stalk material is produced inside the outer layer and the inner layer disappears. At this stage the gelatinous plug is extruded and mucilage is released through the ostiole. The released eggs are retained on the receptacle by the stalk and are surrounded by a large amount of the mucilage. Three-celled germlings form a primary wall with a polylamellated structure of microfibril layers. In multicellular germlings that have differentiated into thallus and rhizoids, the peripheral thallus cells have an outer cell wall consisting of a microfibril layer under the primary wall, while the cell wall of the rhizoid tip has an amorphous structure. The germlings are released from the stalk and become attached to the substratum by an adhesive substance secreted from rhizoidal cells. 相似文献

Summary. The ultrastructure of mitosis and cytokinesis in Scytosiphon lomentaria (Lyngbye) Link zygotes was studied by freeze fixation and substitution. During mitosis, the nuclear envelope remained mostly
intact. Spindle microtubules (MTs) from the centrosome passed through the gaps of the nuclear envelope and entered the nucleoplasm.
In anaphase and telophase, two daughter chromosome masses were partially surrounded with endoplasmic reticulum. After telophase,
the nuclear envelope was reconstructed and two daughter nuclei formed. Then, several large vacuoles occupied the space between
the daughter nuclei. MTs from the centrosomes extended toward the mid-plane between two daughter nuclei, among the vacuoles.
At that time, Golgi bodies near the centrosome actively produced many vesicles. Midway between the daughter nuclei, small
globular vesicles and tubular cisternae accumulated. These vesicles derived from Golgi bodies were transported from the centrosome
to the future division plane. Cytokinesis then proceeded by fusion of these vesicles, but not by a furrowing of the plasma
membrane. After completion of the continuity with the plasma membrane, cell wall material was deposited between the plasma
membranes. The tubular cisternae were still observed at the periphery of the newly formed septum. Microfilaments could not
be observed by this procedure. We conclude that cytokinesis in the brown algae proceeds by fusion of Golgi vesicles and tubular
cisternae, not by a furrowing of the plasma membrane.

Sargassum vachellianum Greville is one of the most important members in the subtidal seaweed flora along the coasts of the Eastern China Sea. In order to understand the embryology of zygotes and development of germlings in S. vachellianum, we performed artificial ripening, systematically observed the morphological changes and development of the fertilized eggs and germlings, and explored the effects of culture conditions on the growth of the germlings. The receptacles matured, and eggs and sperm were released after 1–5 days of culture at 21–24 °C under 60–90 μmol photons m?2 s?1 (14L:10D). Once fertilized, zygotes immediately detached from the female receptacles and began to germinate. Eight nuclei in the released fresh zygote soon begun to fuse, forming a large central nucleus, and underwent two horizontal divisions to produce a small “original rhizoid cell,” which eventually formed rhizoids after several divisions, and three other cells which eventually formed landmine-like germlings after several continuous divisions. The germlings then formed rhizoids and attached onto the bottom of the flask within 12 h of culture. The growth and development of the germlings younger than 20 days were significantly influenced by the culture temperature and light, with the optimal conditions being 21 °C and 40 μmol photons m?2 s?1. However, for germlings older than 30 days, the optimal conditions were 24 °C and 60 μmol photons m?2 s?1. 相似文献

Summary Processes of fertilization and zygote development inFucus distichus were studied by indirect immunofluorescence microscopy using anti- tubulin antibody and electron microscopy. Just after plasmogamy, sperm aster formation occurs during migration of a sperm nucleus toward an egg nucleus at the center of cytoplasm. Only sparse microtubules (MTs) exist around the egg nucleus. The sperm aster can be observed till karyogamy, but afterwards vanishes. Accompanying sperm aster formation, cortical MTs which are reticulately arranged develop further in the zygotes. In 4 h-old zygotes, characteristic structures which are composed of fine granular masses and consist of intermixed dense and lighter staining areas appear around the nucleus. These structures cannot be detected with anti- tubulin immunofluorescence microscopy. The two centrioles derived from the sperm separate and migrate to both poles. In 4 h-and 8 h-old zygotes, there are no defined MT foci around the zygote nucleus and MTs radiate from the circumference of it. In 12 h-old zygotes, each centriole has migrated to the poles and derivative centrioles are generated. The fine granular masses also migrate to both poles and finally disappear accompanying the appearance of numerous MTs radiating from the poles. Therefore, two distinct MT foci appear from 12 h onwards. Progressive stages of nuclear division were also examined with electron and immunofluorescence microscopy in 16 h-old zygotes. The sperm chloroplast with an eyespot and the sperm mitochondria with an intercristal tubular structure, which are distinctive from those of egg, can be detected after plasmogamy and karyogamy. Sargassum polycystum C. Agardh is one of the most abundant marine brown algae and is distributed widely in warm and temperate waters, particularly in the Indo-west Pacific region and Australia. Although its commercial potential and ecological and evolutionary importance are recognized, many pivotal aspects of its biology remain unexplored. Current knowledge of the historical biogeographical affinities and patterns is limited, but some data are available about its genetics, the genetic variation among populations, and spatial patterns. This study aimed to analyze the genetic population structure and distribution patterns of S. polycystum populations in 13 different locations from Indonesia to Japan using the mitochondrial gene cox3. The seven haplotypes of cox3 identified in this study indicated a low level of genetic diversity. Homogeneity of this haplotype was observed particularly in the Gulf of Thailand, Cambodia, and Japan, whereas higher haplotype diversity was found in Phuket (Thailand), Bali (Indonesia), and Singapore. Those data suggest that S. polycystum is likely to have expanded from the south of Indonesia and the west of the Malay Peninsula towards the northeast of the region. Geological studies showed that Sundaland, now corresponding to the Gulf of Thailand, was submerged due to sea level rises after the last glacial period. Therefore, the decrease in the genetic diversity of S. polycystum populations is interpreted here as a population expansion after the rise in sea levels. 相似文献

In brown algae, membrane resources for the new cell partition during cytokinesis are mainly flat cisternae (FCs) and Golgi-derived vesicles. We used electron tomography coupled with rapid freezing/freeze substitution of zygotes to clarify the structure of transient membrane compartments during cytokinesis in Silvetia zygotes. After mitosis, an amorphous membranous structure, considered to be an FC intermediate was observed near endoplasmic reticulum clusters, lying between two daughter nuclei. FCs were arrayed at the cytokinetic plane, and a tubular membranous network was formed around them. This network might be formed by the consecutive fusion of spherical vesicles that are linked to the edges of FCs to form a membranous network (MN). At the initial stage of the formation of a membranous sac (MS) from the MN, the MS had flat and swollen parts, with the latter showing membranous tunnels. Coated pits were detected with high frequency at the swollen parts of the MS. This observation indicated that membranous tunnels disappeared by recycling of excess membrane via endocytosis, and the swollen part became flat. The MN appeared at the edges of the growing MS. MN and the MN-MS complex were observed along the cytokinetic plane in several spaces. The MS expanded by the incorporation of MN or other MS in its neighborhood. With the maturation of the new cell partition membrane, the thickness of the MS became constant and the membrane cavity disappeared. The changes in the surface area and volume of the transient membrane compartment during cytokinesis were analyzed from the tomographic data. 相似文献

During cytokinesis in brown algal cells, Golgi-derived vesicles (GVs) and flat cisternae (FCs) are involved in building the
new cell partition membrane. In this study, we followed the membrane fusion process in Silvetia babingtonii zygotes using electron microscopy together with rapid freezing and freeze substitution. After mitosis, many FCs were formed
around endoplasmic reticulum clusters and these then spread toward the future cytokinetic plane. Actin depolymerization using
latrunculin B prevented the appearance of the FCs. Fusion of GVs to FCs resulted in structures that were thicker and more
elongated (EFCs; expanded flat cisternae). Some complicated membranous structures (MN; membranous network) were formed by
interconnection of EFCs and following the arrival of additional GVs. The MN grew into membranous sacs (MSs) as gaps between
the MNs disappeared. The MSs were observed in patches along the cytokinetic plane. Neighboring MSs were united to form the
new cell partition membrane. An immunocytochemical analysis indicated that fucoidan was synthesized in Golgi bodies and transported
by vesicles to the future cytokinetic plane, where the vesicles fused with the FCs. Alginate was not detected until the MS
phase. Incubation of sections with cellulase-gold showed that the cellulose content of the new cross wall was not comparable

Habitat-forming macroalgae play a central role in the ecology of temperate reefs worldwide but there exists a critical lack
of knowledge about important processes such as dispersal and gene flow. Understanding dispersal and gene flow of habitat-forming
seaweeds is particularly pertinent given that loss of habitat-forming algae is an increasingly prevalent problem worldwide.
Here, we develop 10 microsatellite markers for the monotypic Phyllospora comosa, an important habitat-forming macroalga that has undergone massive declines on urbanised coastlines of Sydney, Australia.
We characterise population genetic diversity and structure and estimate levels of dispersal and gene flow between the geographically

Ten species of Sargassum (Sargassaceae, Phaeophyceae) were found along the Gulf of Thailand. Morphological characteristics of Sargassum baccularia (Mertens) C.A. Agardh, S. binderi Sonder, S. cinereum J.G. Agardh, S.crassifolium J.G. Agardh, S. longifructum Tseng et Lu, S. oligocystum Montagne, S. polycystum C.A. Agardh, S. siliquosum J.G. Agardh, S. swartzii (Turner) C.A. Agardh and one unidentified species were examined and are described in detail. The most common species were
S. polycystum distributed widely in almost all the study sites, S. crassifolium restricted to Prachuap Khirikhan Province, S. longifructum restricted to Chumphon Province, S. siliquosum restricted to Surat Thani Province and one unidentified species restricted to Songkhla Province. Three species (S. cinereum, S. longifructum and S. swartzii) are new records for the algal flora of Thailand. Flagellar development in the plurilocular zoidangia of sporophytes of the brown alga Ectocarpus siliculosus was analyzed in detail using transmission electron microscopy and electron tomography. A series of cell divisions in the plurilocular zoidangia produced the spore-mother cells. In these cells, the centrioles differentiated into flagellar basal bodies with basal plates at their distal ends and attached to the plasma membrane. The plasma membrane formed a depression (flagellar pocket) into where the flagella elongated and in which variously sized vesicles and cytoplasmic fragments accumulated. The anterior and posterior flagella started elongating simultaneously, and the vesicles and cytoplasmic fragments in the flagellar pocket fused to the flagellar membranes. The two flagella (anterior and posterior) could be clearly distinguished from each other at the initial stage of their development by differences in length, diameter and the appendage flagellar rootlets. Flagella continued to elongate in the flagellar pocket and maintained their mutually parallel arrangement as the flagellar pocket gradually changed position. In mature zoids, the basal part of the posterior flagellum (paraflagellar body) characteristically became swollen and faced the eyespot region. Electron dense materials accumulated between the axoneme and the flagellar membrane, and crystallized materials could also be observed in the swollen region. Before liberation of the zoospores from the plurilocular zoidangia, mastigoneme attachment was restricted to the distal region of the anterior flagellum. Structures just below the flagellar membrane that connected to the mastigonemes were clearly visible by electron tomography. 相似文献

Durvillaea (southern bull-kelp) is an economically and ecologically important brown algal genus that dominates many exposed, rocky coasts in the cold-temperate Southern Hemisphere. Of its five currently-recognized species, four are non-buoyant and restricted to the south-western Pacific, whereas one is both buoyant and widely distributed. Durvillaea has had an unsettled taxonomic history. Although its position within the brown algae (Phaeophyceae) has now been largely resolved through the use of molecular techniques, the taxonomic status of several Durvillaea species/morphotypes remains unresolved. Previous molecular phylogenetic studies of phaeophycean taxa have included few Durvillaea samples, and have consequently paid little or no attention to variation within this genus. The current study presents phylogenetic analyses of four genetic markers (mitchondrial: COI; chloroplast: rbcL; and nuclear: 18S and 28S) to resolve phylogenetic relationships within Durvillaea. Results support the monophyly of solid-bladed taxa D. willana, D. potatorum, and D. sp. A (an undescribed species from the Antipodes Islands), whereas the widespread, buoyant D. antarctica is paraphyletic, with solid-bladed D.chathamensis placed sister to a D. antarctica clade from northern NZ but within D. antarctica sensu lato. The phylogenetic and ecological diversity detected within D. antarctica indicate that it is a species complex of five deeply divergent clades. Under a phylogenetic species concept, Durvillaea can be interpreted as a complex of nine distinct evolutionary lineages, only one of which has an intercontinental distribution ('subantarctic'D. antarctica). 相似文献

Photoprotection mechanisms protect photosynthetic organisms, especially under stress conditions, against photodamage that may inhibit photosynthesis. We investigated the effects of short-term immersion in hypo- and hypersalinity sea water on the photosynthesis and xanthophyll cycle in Sargassum fusiforme (Harvey) Setchell. The results indicated that under moderate light [110 μmol(photon) m?2 s?1], the effective quantum yield of PSII was not reduced in S. fusiforme fronds after 1 h in hyposalinity conditions, even in fresh water, but it was significantly affected by extreme hypersalinity treatment (90‰ sea water). Under high light [HL, 800 μmol(photon) m?2 s?1], photoprotective mechanisms operated efficiently in fronds immersed in fresh water as indicated by high reversible nonphotochemical quenching of chlorophyll fluorescence (NPQ) and de-epoxidation state; the quantum yield of PSII recovered during the subsequent relaxation period. In contrast, fronds immersed in 90‰ sea water did not withstand HL, barely developed reversible NPQ, and accumulated little antheraxanthin and zeaxanthin during HL, while recovery of the quantum yield of PSII was severely inhibited during the subsequent relaxation period. The data provided concrete evidence supporting the short-term tolerance of S. fusiforme to immersion in fresh water compared to hypersalinity conditions. The potential practical implications of these results were also discussed. 相似文献

Sargassum hemiphyllum is commonly found in Japan and Korea, with a variety, var. chinense, that is found distributed in the southern Chinese coast. We previously reported distinct genetic differentiation between the two taxa based on the PCR‐RFLP data of plastid RubiscoL‐S spacer. The present study aims at elucidating the phylogeographic pattern of S. hemiphyllum based on more markers in the nuclear and extranuclear genomes, with a view to reveal the occurrence of hybridization. The two allopatrically distributed taxa were found to be genetically distinct in nuclear ITS2, plastidial Rubisco (Rbc) and mitochondrial TrnW_I (Trn) spacers. Their divergence was postulated to be attributable to the vicariant event which resulted from the isolation of the Sea of Japan during the late Miocene (6.58–11.25 Mya). Divergence within both S. hemiphyllum and the chinense variety was observed based on Trn spacer, while the divergence in S. hemiphyllum was further confirmed in Rbc spacer. This divergence appears to correspond to the separation of the Japanese populations between the Sea of Japan and the Pacific that occurred around 0.92–2.88 Mya (the early Pleistocene). The presence of an ITS2 clone resembling var. chinense sequences in a Japanese population of S. hemiphyllum (JpNS) raises the possibility of the introgression of var. chinense individuals into S. hemiphyllum population. Compared to that between S. hemiphyllum and the chinense variety, hybridization among the Japanese and Korean populations of S. hemiphyllum is highly probable as all these individuals share a pool of nuclear ITS2 sequences, possibly attributable to incomplete concerted evolution of ITS2. 相似文献

The seasonality of standing crop of a Sargassum bed was investigated by conducting monthly sampling from February 1988 to July 1989. Environmental parameters of water movement, salinity, number of daytime minus tides, and water temperature were also measured. An intra-annual pattern of variation in standing crop of Sargassum crassifolium, S. cristaefolium, S. oligocystum, and S. polycystum was observed. Standing crop was generally lowest in February, March, April, or May, and highest in November through January. Sargassum accounted for about 35 to 85% of the monthly algal standing crop of the bed, and the observed variation in overall standing crop of the bed generally reflected the standing crop of Sargassum. The seasonality of the standing crops of the associated algal divisions also followed an annual cycle, but their maximum and minimum standing crops did not coincide with those of Sargassum. Individually, as well as collectively, the standing crops of the Sargassum spp. were poorly correlated with the environmental factors observed. 相似文献

Two separate 1?m2 plots were cleared of algal cover every 3 or 4 wk for 1 yr and each time, one was filled with cleaned, sun-dried coralline rocks. These plots were closely monitored in subsequent months for any sign of Sargassum regrowth. Control plots were also monitored during the same period. New growth was first detected in December in cleared and control plots, 3 months after the start of the reproductive season for Sargassum spp. in the area. New growth was observed in the plots with coralline rocks in January, and only on rocks placed at least 3 months earlier. No new growth was observed in cleared and control plots after June, nor throughout the rest of the experimental period on rocks placed after October. Pre-conditioning of the coralline rocks appeared to be a prerequisite for settling of Sargassum germlings and the importance of timing in laying these rocks for successful colonization of Sargassum spp. is discussed. 相似文献

The reproductive phenology and epiphytic macroalgae of Sargassum muticum were studied through an annual cycle (September 1987 to November 1988) at two sites on the northwestern coast of Baja California, Mexico, which were subjected to different degrees of wave exposure. Sargassum muticum is a brown alga of Japanese origin, now considered a permanent member of the marine flora of Baja California. A similar reproductive development was observed at both sites, with a maximum percentage of reproductive plants from May to July (spring–summer) and minimum from December to March (winter). Reproductive plants were found throughout the year. A total of 48 species of epiphytes were identified and seasonal variation in their diversity was observed. Four species of brown seaweeds, namely Sargassum baccularia, Sargassum binderi, Sargassum siliquosum and Turbinaria conoides, harvested from Port Dickson, Negeri Sembilan, Malaysia were analysed for ash content, alginate yield and alginate properties.
Seaweeds calcined at 450°C were found to have low amount of non-combustible residue as these were not contaminated by calcareous
animals. Alginate was extracted from these seaweeds by two methods: hot and cold. In the hot method, the storing time was
3 h and the processing temperature was 50°C, whilst in the cold method, the sample was stored overnight at room temperature.
Higher yield of alginate was obtained by the hot method compared to the cold method, but alginate extracted by the cold method
gave higher molecular weight. In the hot method, 49.9% of alginate was extracted from S. siliquosum, followed by T. conoides (41.4%), S. binderi (38.9%) and S. baccularia (26.7%). Alginate extracted from T. conoides has an average molecular weight, M
w, of 8.06 × 105 g mol−1, whereas alginate from S. siliquosum was the lowest in M
w (4.81 × 105 g mol−1) when the extraction was done at room temperature. Alginate extracted from S. baccularia was found to be very heat-sensitive. Its M
w has dropped more than 83%, from 7.52 × 105 to 1.23 × 105 g mol−1, when the extraction temperature was raised. The effect of heat on the extent of depolymerisation of the alginate molecule
of the other three brown seaweed species was less significant, with decrease in molecular weight ranging between 13% and 16%. 相似文献
