Spleen cells from LSH hamsters inoculated with xenogeneic, allogeneic, or syngeneic (PARA-7) tumor cells were assayed for their ability to mediate direct cell-mediated cytotoxicity (DCMC) and lectin-dependent cell-mediated cytotoxicity (LDCC) in a 4-hr chromium release assay. Spleen cells from animals immune to xenogeneic or allogeneic cells demonstrated specific DCMC against homologous target cells in the absence of Con A and nonspecific LDCC against both homologous and heterologous target cells in the presence of Con A. Spleen cells from animals bearing syngeneic PARA-7 tumors (TBA) failed to express DCMC against homologous or heterologous target cells; however, significant lysis of all target cells occurred in the presence of Con A. LDCC was not detectable when nonsensitized spleen cells from normal animals were employed. The LDCC reaction was dependent on the concentration of Con A and the number of effector cells present in the reaction. The development of LDCC effector cells in the TBA appeared to parallel the development of both DCMC and LDCC effector cells in immune animals.  相似文献

Mononuclear cells from human peripheral blood are able to destroy alloantibody-coated target cells of a human lymphoblastoid line. The effector cell is a nonadherent, non-phagocytic, medium sized lymphoid cell. The cell is not detectable in early human fetuses. Its activity is destroyed by x-ray treatment in vitro but is not inhibited by anti-immunoglobulin sera. Its efficiency in antibody-dependent cell-mediated cytotoxicity is rapidly lost in culture and this loss is prevented by stimulation with allogeneic cells.  相似文献

It was observed that when normal mouse spleen cells were cultured alone in vitro (precultured) for 3 to 7 days, these cells lost the ability to generate cell-mediated cytotoxicity (CML) during subsequent in vitro sensitization with allogeneic spleen cells, trinitrophenyl (TNP)-modified syngeneic spleen cells, or syngeneic tumor cells. These precultured cells, which were themselves unable to generate CML, were also shown in mixing experiments to suppress, actively, the generation of CML by freshly explanted spleen cells. Suppression occurred at the sensitization phase of CML, and not at the effector level; supernatants from suppressive precultured cells were not suppressive. Suppression was totally abrogated by the treatment of spleen cells with a T cell-specific rabbit anti-mouse brain serum and complement (RalphaMB+C) either before or after preculturing, suggesting that a T cell eas essential both to the generation of suppressor activity and to its expression. Suppressor activity was entirely absent in precultured nylon wool column-nonadherent spleen cells, a T cell-enriched population containing most of the RalphaMB+C-sensitive cells in the spleen. Precultured nylon column-adherent cells (T cell-depleted) did have suppressive activity, and a mixture of nylon-adherent and nylon-non-adherent cells was a suppressive after preculture as the precultured unseparated spleen. Moreover, the ability of nylon-adherent spleen cells to generate suppressive activity during preculturing was abrogated by treatment with RalphaMB+C. Thus, the "spontaneous" generation of CML-suppressive activity was dependent upon a limited subpopulation of splenic T cells isolated in the nylon column-adherent fraction. The relationship of these data to a previously described synergy between subpopulations of normal spleen in the generation of CML is discussed, and the findings related to other suppressor systems described in the literature.  相似文献

The nature of the cell population involved in lymphocyte mediated cytotoxicity to baby-hamster-kidney (BHK-21) target cells persistently infected with rubella virus was investigated by a 51Cr-release microassay. After depletion of the T cell population with an antiserum to human0thymus-lymphoid tissue antigen (HTLA), the purified B cell population showed a decrease in E-rosette formation (9.0 +/- 2.2% compared to 69.6 +/- 9.1% before treatment) and an insignificant degree of cytotoxic activity against rubella-infected target cells (specific immune release of 51Cr was 0.9 +/- 2.6% compared to 24.2 +/- 3.8 before treatment). A purified T cell population, prepared by depletion of B cells with an anti-human immunoglobulin serum and complement, was found to show no alteration in E-rosette formation (85.2 +/- 6.2%) or cytotoxicity (30.3 +/- 4.4% SIR) but showed decreased EA- and EAC-rosette formation (2.7 +/- 1.5% and 10.5 +/- 3.2%, respectively, compared to 19.4 +/- 2.9% and 28.0 +/- 4.1% before treatment). A monocyte-depleted population prepared by removal of the plastic adherent mononuclear cells showed no significant alteration of rosette formation or cytotoxicity. These experiments suggest that the predominant lymphoid population responsible for direct cell-mediated cytotoxicity to virus infected target cells in the 51Cr-release microassay appears to be effected by a thymus-dependent lymphocyte population.  相似文献

In this study we characterized the time when cytotoxic T lymphocytes (CTL) can be induced in the thymus and spleen from their immediate CTL precursors (CTL-P). In contrast to fetal or newborn thymus, the thymus of 1 to 2-day-old C57BL/6 mice contained cells that, after cultivation in vitro with allogeneic DBA/2 stimulating cells, exhibited high levels (as great or greater than that induced in adult thymocytes) of CTL activity as measured by the ability to lyse P815 (DBA/2) tumor target cells. However, CTL activity induced in spleen cells remained how during the first 5 days of life, increased sharply between 6 to 9 days, and reached adult levels at 11 to 20 days. Furthermore, early postnatal spleen cells did not suppress the adult splenic CTL response. These results suggest 1) that the full potential to generate CTL in response to an allogeneic stimulus commences in the thymus on the first day after birth and 2) a different temporal appearance of immediate CTL precursors in the thymus and spleen.  相似文献

Human peripheral Wood lymphocytes were depleted of natural killer cells cytotoxic against human fetal fibroblasts by allowing them to attack the fibroblast targets grown on plastic beads followed by gravity sedimentation under conditions in which single cells floated but the attacker cells sedimented with the carrier beads. The attacker cells could be released from the bead-grown targets and shown to be greatly enriched in natural cytotoxic activity. The effector cells depleted by fibroblast adsorption were also depleted of cytotoxic activity against other monolayer targets whereas suspension grown lymphoma and leukemia cells (MOLT-4, RAJI, and K-562) were killed as effectively as by non-depleted effector cells. In competition assays other monolayer cells inhibited the natural cytotoxicity against fetal fibroblasts but the suspension-grown cells were unable to compete. The results suggested that different effector cell populations were probably involved when monolayer vs suspension targets were used in assays for human natural cell-mediated cytotoxicity. The separation was not, however, functionally complete since in competition assays with suspension-grown target cells also monolayer cells were able to compete. Preliminary morphological characterization of the natural killer cells against fetal fibroblasts is also presented.  相似文献

The evaluative technique of lymphocyte cytolytic activity in human peripheral blood has been designed. The target cells, lectin (Con A) concentration and incubation time for measuring cytolytic activity of lymphocytes pre-purified from adherent cells have been selected. In order to separate and characterize cytotoxic effector cells in natural cell-mediated cytotoxicity (NCMC), human lymphocytes were fractionated by Percoll continuous density gradient centrifugation (Pharmacia, Piscataway, N.J.). Lymphocytes from normal donors were fractionated through a 35-ml gradient and 2- or 3-ml aliquots were collected, counted, and grouped into three or more fractions in order to obtain sufficient cells for testing. Fractions of cells were tested for cytotoxicity in a 4-hr chromium release test and/or a 40-hr [3H]proline assay. Cell markers were assessed by testing for cells forming E rosettes, EA rosettes, and for cells with surface membrane immunoglobulin (SMIg). The lightest fraction contained larger cells and also usually contained the highest concentrations of cells with receptors for the Fc portion of IgG (FcR + cells), although slight variations were seen among individual donors. Results of cytotoxicity tests showed that cells from the top portions of the Percoll gradient had consistently greater cytotoxic activity on a per cell basis than the denser cells sedimenting lower. Estimation of cytotoxic activity in lytic units showed that 54–75% of the activity was recovered in the top 26–29% of the cells. This approach to investigating cell-mediated cytotoxicity should yield useful information regarding cellular interaction in, and regulation of, cytotoxic activities.  相似文献

Lectin-dependent neutrophil cytotoxicity against autologous human red cells was studied using an 111In(indium)-release assay. Human red cells were not readily killed by neutrophils in the presence of phytohemagglutinin (PHA). However, removal of red cell membrane sialic acids (desialylation) markedly enhanced their susceptibility to PHA-dependent neutrophil cytotoxicity. This neutrophil cytotoxicity was dependent on the energy supplied by anaerobic glycolysis, but it was independent of erythrophagocytosis. Catalase, superoxide dismutase, KCN, and Na azide did not inhibit PHA-dependent neutrophil cytotoxicity. Neutrophils from a patient with chronic granulomatous disease, in the presence of PHA, also killed desialylated red cells normally. On the other hand, desialylation of neutrophils had no effect on the expression of their cytotoxic effect. The results suggest that desialylated red cells are much more susceptible to lectin-dependent neutrophil cytotoxicity than normal red cells, and that lectin-dependent neutrophil cytotoxicity is independent of reactive oxygen species.  相似文献

Cell-mediated cytotoxicity (CMC) by lymphocytes from patients with oral squamous cell carcinoma, as well as from nonmalignant control donors, was tested by a microcytotoxicity assay against a cultured cell line derived from an oral squamous cell carcinoma. In terms of the degree of CMC, stage-related cytotoxicity was observed. A further study of the effector cell analysis revealed that the cytotoxic effects of lymphocytes from both patients and control donors were largely attributable to non-T cells. However, the effectors were also stage related, and in early stage patients, T-cell-mediated cytotoxicity was super-imposed on non-T-cell-mediated cytotoxicity. This conclusion was further supported by the evidence of elevated cytotoxic activity of T cells in early stage patients, which was computed from simultaneous equations proposed in the present paper for computing the cytotoxic activity of T cells (CTAT) and non-T cells (CTAnon-T).  相似文献

Bone marrow cells, cultured in L-929 CSF, consist of cells of granulocyte and macrophage lineages. Cells of the granulocyte lineage are known to be cytotoxic for Candida albicans. In this paper we report that macrophage precursor cells also display strong cell-mediated cytotoxicity against the yeast form of the dimorphic fungus C. albicans. The macrophage precursors responsible for this activity are nylon wool-nonadherent, nonphagocytic cells and lack asialo GM1 surface antigen. A purified population of macrophage precursors (greater than 95%) was obtained by means of Percoll density centrifugation. The interaction of these purified effectors with the target yeast cells was analyzed at a single cell level, and their activity was compared with that displayed by cells of the granulocytic series derived from the same bone marrow culture. Macrophage precursor cells proved to be more effective in binding the target cells and showed the same killing ability as the granulocytes: macrophage precursors were not damaged by contact with the target, in contrast to that which happened with granulocytes. In a long-term colony-forming unit assay, in fact, granulocytic cells showed a decrease over time in their ability to inhibit the growth of C. albicans, probably due to cell damage and death after the interaction with the target. In contrast, no loss of activity was observed with the macrophage precursor fraction. The same macrophage precursor cells also proved able to exert good natural killer activity against YAC-1 lymphoma cells, but not against P815 mastocytoma cells, as reported previously. The macrophage precursor cells, when cultivated in vitro to mature macrophages, lost completely their natural cytotoxicity against C. albicans and YAC-1 cells. The implications of these findings, as well as the possible role in vivo of such a precursor cell population during an infection, are discussed.  相似文献

The specificity of T cells for syngeneic target cells is directed to both antigens and products of the major histocompatibility complex (MHC) on the target cell surface. This dual requirement is best accounted for by the altered-self hypothesis, which implies that the MHC products on a cell's surface are able to form complexes with many other proteins on the surface of the same cell. To account for the ability of MHC products to bind so many different cell surface antigens we propose that interactions in general among macromolecules on the surface of a membrane may be dramatically enhanced by a purely physical effect. This effect derives from the confinement of membrane macromolecules to an effective volume which is the product of membrane surface area times d, the distance over which the center of mass of the molecules can move in a vertical direction (perpendicular to the membrane surface). Because d is very small the effective concentrations of surface molecules are extremely high and their interactions are correspondingly enhanced.  相似文献

There has been considerable recent interest in the possible role of extracellular ATP in cell-mediated cytotoxicity. To investigate the similarities and differences between ATP-mediated lysis and CTL-mediated lysis, we studied in detail the ATP-mediated lysis of EL-4 cells, which are highly susceptible to lysis by extracellular ATP. ATP-mediated lysis was time and concentration dependent; most lysis occurred within 4 to 6 h of incubation. The kinetics of ATP- and cell-mediated lysis were similar. However, in contrast to CTL-mediated lysis, ATP-mediated lysis of EL-4 cells did not appear to be accompanied by characteristic chromosomal degradation (apoptosis). In order to compare these cytotoxic processes in greater detail, ATP-resistant clones were derived by growing EL-4 cells continuously in the presence of high concentrations of ATP. These cloned EL-4 lines showed marked resistance to ATP-mediated cytotoxicity across a wide range of concentrations but were as susceptible as the parent EL-4 cells to cell-mediated cytotoxicity by an alloreactive effector population from a MLC. Thus, there appear to be a number of differences between ATP-mediated and cell-mediated cytotoxicity in this system; most notable is the identification of cell lines that are resistant to ATP but susceptible to CTL-mediated lysis.  相似文献
