A rapidly frozen vitrified aqueous suspension of influenza A virus was observed by high resolution electron cryomicroscopy. The influenza particles were grouped into small (diameter < 150 nm) spherical particles with well organized interiors, large spherical ones with less internal organization, and filamentous ones. Envelopes of most of the large virus particles were phospholipid bilayers, and the chromatography fraction containing these large particles was largely devoid of viral activity. The envelopes of most of the filamentous and small spherical virus particles, on the other hand, gave a strange contrast which could be ascribed to a combination of a thin outer lipid monolayer and a 7.2 nm thick protein-containing inner layer. These latter particles represented most of the viral activity in the preparation. Densitometric traces of the near in-focus images confirmed these structural differences. Some viral envelope structures apparently intermediate between these two distinct types of membrane were also detected. A structural model of intact biologically active influenza virus particles was formulated from these results, together with computer simulations.  相似文献

Borna disease virus (BDV), a negative nonsegmented single-stranded RNA virus, has not been fully characterized morphologically. Here we present what is to our knowledge the first data on the fine ultrastructure and morphogenesis of BDV. The supernatant of MDCK cells persistently infected with BDV treated with n-butyrate contained many virus-like particles and more BDV-specific RNA than that of untreated samples. The particles were spherical, enveloped, and approximately 130 nm in diameter; had spikes 7 nm in length; and reacted with BDV p40 antibody. A thin nucleocapsid, 4 nm in width, was present peripherally in contrast to the thick nucleocapsid of hemagglutinating virus of Japan. The causal agent of Chloris striate mosaic disease appears to be a virus with polyhedral particles 18 nm in diameter usually occurring as paired structures about 18 times 30 nm in negatively stained preparations. These particles were detected in the nuclei of infected plants forming characteristic inclusions in all cells except those of the epidermis. Such particles were not detected in thin sections of viruliferous leaf hopper vectors (Nesoclutha pallida). Purified virus preparations were shown to be highly infective when assayed by feeding vector leaf hoppers through membranes and confining them on indicator plants. In particle morphology, chloris striate mosaic virus (CSMV) differs from other viruses of Gramineae in Australia but resembles maize streak virus isolated in Africa, which however is serologically unrelated.  相似文献

A new virus was found to be associated with acute gastroenteritis in children. In duodenal biopsies, it was observed infecting only intestinal epithelial cells, and it resembled orbiviruses in its morphogenesis. For diagnsotic purposes the virus was readily demonstrated by negative staining of fecal extracts. Two forms of particles were seen: double-sheeled particles (70 to 75 nm in diameter) resembling those of reovirus with a sharper outline, and single-shelled particles (60 nm in diameter) with obvious capsomer structure and resembling those of orbiviruses. The morphological resemblance of this human virus to the viruses of "Nebraska" calf scours and epizootic diarrhoea of infant mice is emphasized.  相似文献

在病鱼肾组织中发现有病毒颗粒,在头肾组织中也看到类似的颗粒。病毒呈球形或六角形,直径为60—78毫微米,平均为69毫微米;颗粒中央有一电子密度较高的核心,其直径平均为32毫微米,核心周围包有外膜,宽20毫微米左右。此外,还有一种无外膜的、电子密度均匀的病毒颗粒,直径为46—60毫微米,平均为52毫微米,这种颗粒出现在细胞核和胞质包涵体内。所观察到的病毒颗粒均出现在肾的造血组织内,但红血球和颗粒白血球中没有发现。肾小管上皮细胞正常,也未观察到病毒颗粒。在正常鱼的肾组织中没有发现与病鱼标本中类似的病毒颗粒。因此可以认为,我们人工感染致病的草鱼肾组织中所观察到的病毒颗粒,是草鱼出血病的病原,暂名为草鱼疱疹病毒。    相似文献

SARS患者病理尸检肺组织样品分离病毒出现细胞病变的Hep2 培养细胞,按常规制作超薄切片,透射电镜下观察。电镜下,检出在感染细胞内复制、组装的呼肠孤病毒及其包涵体。病毒粒子衣壳立体对称、无包膜、直径在60~80nm。成熟病毒粒子核心致密常排列呈晶格状,不成熟病毒粒子核心空亮。数目不等的上述两种病毒粒子、长短不等的微管样结构和病毒浆常在核旁胞质内组成大小不等、无定形的病毒包涵体。此发现进一步提供了呼肠孤病毒感染有可能与SARS相关的形态学依据。  相似文献

Hepatitis A antigen (HAAG) was localized in liver tissue from marmosets inoculated with human hepatitis A virus (HAV) by light and electron microscopy by using a peroxidase-conjugated antibody method. The fine granular peroxidase staining was scattered throughout the cytoplasm of liver cells when viewed with the light microscope. The distribution of HAAg-positive cells was focal. Virus-like particles, 24 to 27 nm in diameter, were observed in the cytoplasm of hepatocytes and smaller cells, resembling Kupffer cells, by standard thin-section electron microscopy (thin section EM). By immunoperoxidase electron microscopy (immunoperoxidase EM), HAAg was detected on the particles, which were aggregated within cytoplasmic vesicles of the hepatocyte. The surrounding membrane of the vesicles was also HAAg- positive. Similar HAAg particles were observed in the cytoplasm of smaller cells adjacent to hepatocytes as well. Thus, immunoperoxidase EM revealed that the 24- to 27-nm virus-like particles in the cytoplasm of liver cells obtained from marmosets were infected with HAV contained HAAg.  相似文献

In the present work, we demonstrate virus-like particles (VLPs) with various morphological variations in Trichomonas vaginalis. The VLPs were distinct based on size, shape and electron density, with VLPs being either electron-dense or electron-lucent. We used electron microscopy thin sections of several T. vaginalis strains virus-infected, and also negative staining of fractions obtained after purification by CsCl buoyant density gradient centrifugation. The particles observed in fractions are identical to those previously described, but by thin sections, we found new forms. The shapes found were icosahedral, spherical and oblong, and the sizes varied from 33 to 120nm in diameter with the most common VLP being spherical and having a size range from 83 to 104nm. The VLPs were found in the cytoplasm closely associated with the Golgi complex, with some VLPs budding from the Golgi, and other VLPs were detected adjacent to the plasma membrane. Unidentified cytoplasmic inclusions were observed in the region close to the VLPs and Golgi. Clusters of the already described icosahedral virus were also observed in the cytoplasm, although less frequently. These results indicate that T. vaginalis organisms may be infected with different dsRNA viruses simultaneously.  相似文献

将感染病毒的小麦全蚀菌山东烟台株培养 20天的菌体细胞,进行超微结构的研究。于电镜下观察到球状病毒颗粒,平均直径23—30nm,多是无规则松散的分布于胞质中;或紧密聚集于液泡、线粒体周围;或排列成线状;或7—8个颗粒排列成环状。病毒仅分布于细胞质中,细胞核、脂肪体内均未见病毒颗粒。病毒浓度在较老的菌体内有增加的趋势。全蚀菌的菌丝细胞壁有三层,外层电子致密内含纤维状物,内层电子较为透明,中层为一电子致密度很深的狭窄夹层。壁的厚度不均,外缘不规则;在菌丝体产生隔膜的早期阶段,于隔膜附近有1—3个外被膜结构的沃罗宁体 Woronin body,隔膜形成的后期,见电子致密物质沉积在核膜孔上,形成中的隔膜顶端为尖状突起向基部逐渐增宽略成金字塔形。  相似文献

Lesions were found in the eyes of cultured shrimp Penaeus monodon that displayed non-specific signs of disease, including lethargy, dark pigmentation, brown gills, empty midgut, anorexia, white tail muscle, necrosis of uropods and fouled cuticle. Eye lesions were associated with sexual development in moribund shrimp in at least 1 disease event. Suppurative inflammation, granuloma and malacia were observed in histological examination of the eye and the causative agents of lesions appear to be Vibrio spp. and a rod-shaped virus (similar to Lymphoid Organ Virus, Gill-Associated Virus [GAV] and Yellow-Head Virus). Suppurative inflammation was characterised by edema, infiltration of haemocytes and local sites of abscesses. Eyes with granuloma usually appeared white in pond-side examinations, and histology showed that fibrous tissue replaced ommatidia, ganglia and internal structures of the eye. Malacia of the eye was characterised by necrosis of nervous tissue, vacuolation and vascular proliferation in the medulla ganglia. Levels of presumptive Vibrionaceace were high in moribund specimens and Gram-negative rods were observed in some specimens as free particles in the interstitial fluid and haemolymph in the eye. Transmission electron microscopy showed that nerve cells in the fasciculated zone (near the basement membrane) contained cytoplasmic vesicles (1 to 3 microm in diameter) with particles (15 to 26 nm in diameter) and rod-shaped nucleocapsids. The rods were similar to those of GAV and were 130 to 260 nm long, 10 to 16 nm in diameter and had helical symmetry with a screw-like thread (2.4 to 3.5 nm pitch). Also, unidentified enveloped virions, averaging 74 nm in diameter, were observed in cytoplasmic vesicles in the fasciculated zone. In conclusion, it is suggested that bacterial and viral infections of the eye could result in impaired neuroendocrine functions, which may cause a range of clinical signs of disease.  相似文献

Sequential effects of rubella virus infection in BHK-21 cells were studied by electron microscopy of thin sections of control and infected cells, 2 to 7 days after infection. Vacuolization of cytoplasm in Golgi areas apparently preceded budding of virions from vacuole membranes and involvement of the endoplasmic reticulum. Newly formed endoplasmic reticulum cisternae encircled and segregated virionforming vacuoles together with other cellular elements. Large vacuolar complexes with numerous virus particles developed, and virus release from these areas occurred with disruption at the cell periphery. The viral particles, with a mean diameter of about 56 nm, consisted of an electron-dense core surrounded by a less dense capsid, enveloped by a typical unit membrane derived from the vacuole membrane.  相似文献

Lymphocytic choriomeningitis (LCM) virus was observed by electron microscopy in thin sections of infected tissue cultures. The particles were pleomorphic and varied greatly in size. The smaller particles (50 to 200 nm) appeared to be spherical, whereas the largest (over 200 nm) were often cup-shaped. All particles contained one to eight or more electron-dense granules which were removed by ribonuclease. The particles were formed by budding from the plasma membrane and appeared to have spikes. The morphological evidence suggests that LCM should be considered as belonging to the presently unclassified group of lipoprotein-enveloped ribonucleic acid viruses.  相似文献

The temporal sequence of development of lymphocystis disease virus (LDV) was studied by electron microscopy of thin sections of infected tissue-culture monolayers. Neither the typical cytoplasmic inclusion nor virus was detected at 4 days postinfection (PI). Inclusions, but no viruses, were detected at 8 days PI. Inclusions and associated virions were detected at 15 days PI, and by 28 days PI the undisrupted cells were filled with the typical virions. No release mechanism was detected, and severe clumping of particles was noted. Negatively stained preparations revealed particles 200 nm in diameter with no capsomere structure and apparent spikes associated with the particle. The relationship of LDV to the well-defined deoxyribonucleic acid virus groups is discussed.  相似文献

Nonoccluded baculovirus-and filamentous virus-like particles were found in nuclei of hemocytes or midgut cells of field-collected spotted cucumber beetles. Each type of particle was associated with a different type of virogenic stroma containing various viral components similar to those referred to as capsid, nucleocapsid, viroplasm, and viral envelope in other known baculovirus infections. Nucleocapsids of the virus which occured only in hemocytes were rod-shaped particles approximately 230 nm long and 52 nm wide and were enveloped singly by a trilaminar unit membrane. Enveloped and partly enveloped particles appeared to be released from the nucleus to the cytoplasm by budding through the nuclear envelope acquiring additional membranes. The nucleocapsids of the virus which occurred only in nuclei of midgut cells were filamentous particles with an average diameter of 25 nm and variable length up to 2 μm. Some extremely long particles were bent almost 360° near the middle, resulting in a hairpin-like configuration. The particles were always enveloped singly. No particles budding through the nuclear envelope were observed.  相似文献

The structural organization of Ectromelia virus DNA in infected mouse liver cells has been studied by using thin sections stained with the Feulgen-like osmium-ammine reaction. We found that in the cytoplasmic factories, free viral DNA was structured into completely extended filaments 2-3 nm thick. Viral DNA in immature virions, however, appeared to have a structural organization that superimposed that of eukaryotic chromatin. This was constituted by roundish subunits, with a diameter of 11-13 nm, composed of a DNA ring encircling an unstained inner core. The mature virion was composed of the same type of subunits, which were arranged in threads twisted into a figure 8 configuration. The distribution of basic proteins was also investigated with the acrolein silver-methenamine technique. In the viral particles only nucleoids were stained; a uniformly distributed positive reaction was observed in the cytoplasmic factories.  相似文献

我国香菇人工栽培历史悠久;在今天纯种人工栽培同样是食用菌生产的主要方向。经查上海栽培香菇“菌种”内生长不正常的菌丝体制剂中有直径为20nm,长度多为100—200nm的类似棒状病毒颗粒。在接种后14—20天,生长缓慢的菌丝体中有直径为28、36、40nm三种不同大小的类似球状病毒颗粒,未见棒状颗粒。在香菇子实体的制剂中同样见到与菌丝体中一样的球状颗粒以及大小为15—16nm×100—300nm的较细的棒状颗粒。病香菇菌褶超薄切片中显示棒状颗粒结晶及球状颗粒的聚集;在液泡中有直径为15—16nm的棒状颗粒。  相似文献

Summary The three-dimensional structure of synaptic ribbons in photoreceptor cells of the frog retina was studied with freeze-etching and freeze-substitution methods, combined with a rapid-freezing technique. Although the synaptic ribbon consisted of two electron-dense plaques bisected by an electron-lucent layer in conventional thin sections, such lamellar nature was not so evident in freeze-etched replicas. The cytoplasmic surfaces of the synaptic ribbon presented an extremely regular arrangements of small particles 4–6 nm in diameter. Fine filaments 8–10 nm in diameter and 30–50 nm in length connected synaptic vesicles and the ribbon surface. These connections were mediated by large particles on both ends of the filaments. Approximately 3–5 filaments attached to one synaptic vesicle. Synaptic ribbons were anchored to a characteristic meshwork underlying the presynaptic membrane via another group of similar fine filaments. The meshwork seemed to be an etched replicated image of the presynaptic archiform density observed in thin sections.  相似文献

A rhabdovirus was found to be associated with a lethal hemorrhagic disease in the cultured Chinese sucker Myxocyprinus asiaticus Bleeker. The rhabdovirus was amplified and isolated from the infected GCO (grass carp ovary) cells. In ultrathin sections of liver cells from the diseased fish, the virus particles exhibited the characteristic bacilliform morphology, and budded through vesicle membranes of the infected cells. The isolated rhabdovirus particles were found to have a bacilliform morphology with 2 rounded ends rather than a typical flat base. The virus particles were measured and ranged in size from 150 to 200 nm in length and 50 to 60 nm in diameter. Most other characteristics, including their size, extensive virus infectivity to fish cell lines, strong cytopathogenic effects, stability at high temperatures, vesicle formation in infected cells, structure protein electrophoretic patterns and the presence of an RNA genome, very closely resembled those of other fish rhabdoviruses. At present it is not known if this is a novel virus species or if it is an isolate of a known fish rhabdovirus. Until a confirmed identification can be made, we will temporarily refer to this virus as Chinese sucker rhabdovirus (CSRV).  相似文献
