An investigation was undertaken to obtain data on the occurrence of aflatoxins and the aflatoxin producing potential of Aspergillus flavus strains isolated from dry fruit slices of quinces produced in jammu and Kashmir, India. A total of 147 A. flavus isolates recovered from dr fruit slices were grown in liquid rice flour medium and screened for the production of various aflatoxins by thin layer chromatography. The results showed that 23.14% of the tested isolates were aflatoxigenic, producing aflatoxins B1and B2 in varying amounts. Aflatoxins G1 and G2 were not detected. All 25 of the investigated market samples were also found to be aflatoxin B1 positive and the level of contamination ranged from 96 to 8164 g/kg of the dry fruit which is quite high in comparison to the permissible level of 30 ppb. As per these results biochemical composition of dry fruit slices of quinces, along with climatic conditions seem to be very favourable for aflatoxin production by the toxigenic A. flavus strains. Steam distilled oil of quince fruit (Cydonia oblonga Mill.=C. vulgaris Pers., marmelo in Japanese) was analyzed by gas chromatography and gas chromatography-mass spectrometry. Sixty-two compounds, 2 hydrocarbons, 13 esters, 11 alcohols, 11 aldehydes, 11 ketones, 5 lactones and 9 miscellaneous compounds, were identified. Of them, the chemical structures of two new oxide compounds, trans- and cis-3-methyl-5-[(E)-3′-methyl-13′-butadien-1′-yl]tetrahydrofuran, were elucidated by instrumental analyses. 相似文献

Cold stress adversely affects growth and productivity, and triggers a series of morphological, physiological, biochemical and molecular changes in plants. Since sugars are present in all floral nectars in greater amounts than any other constituent, the aim of this study was to examine how frost exposure changes sugar metabolism and how it affects on the content of sugar components in the nectar of quince. Three quince cultivars (‘Vranjska’, ‘Triumph’ and ‘Leskova?ka’) were investigated in this study. The contents of sugars (glucose, fructose, sucrose, trehalose, maltose, isomaltose, rhamnose, arabinose, ribose, melezitose, raffinose, and panose) and sugar alcohols (sorbitol, erythritol, mannitol and galactitol) were analyzed by high performance anion exchange chromatography (HPAEC) with amperometric detection. The results showed that after late spring frosts and irreversible damage of flower parts, the nectar of the three quince cultivars contained elevated levels of fructose, trehalose, arabinose, ribose, rhamnose, raffinose, galactitol and mannitol, indicating an impairment of central carbohydrate metabolism. The ratios between individual sugars, such as the glucose/fructose ratio, were changed in the nectar of damaged flowers in all three quince cultivars. The examined cultivars showed similar sugar response to cold stress. The only exception was ‘Leskova?ka’ for the glucose and melezitose pathway, which means that composition of those two sugars changed significantly according to the genotype. The larger are the carbohydrates reserves in different parts of a fruit tree, the higher is the tolerance to any form of frost damage, the results of this study could help in the understanding of how different quince cultivars react to this kind of stress and how they modulate their sugar metabolism. 相似文献

Shoot-tip cultures of Quince C (Cydonia oblonga Mill.) initiated on Murashige & Skoog (MS) medium containing 5 M BA and 0.6% Phytagar showed both shoot-tip necrosis and severe vitrification. Culturing explants on medium containing 1.2% Phytagar and Ca levels of 3 mM (MS medium), 18 mM and 30 mM showed a decrease in growth with increasing medium Ca levels, being especially severe at 30 mM. The Ca content of the explants increased linearly with increasing medium Ca. Culturing explants on medium containing 3 mM, 9 mM, and 18 mM Ca at 0.6, 0.9, and 1.2% agar resulted in reduction in growth, shoot-tip necrosis, and vitrification when either factor was increased. The reduction in shoot-tip necrosis could be accounted for primarily by an increase in medium Ca levels but may also be affected by a change in explant growth. Increasing Ca concentration in the medium resulted in a linear increase in explant K, Ca, Mg, and B levels and a decrease in Mn and Na. Although increasing medium Ca or agar levels reduced vitrification, it is unclear whether they were the direct cause of the reduction in vitrification or whether this response was an effect of the reduction in culture fresh weight.Approved by publication by the Director, West Virginia Agriculture anf Forestry Experimental Station as Scientific Article No. 2199 相似文献

A glucoamylase gene has been cloned from a Rhizopus genomic DNA library using synthetic oligonucleotides corresponding to the amino acid sequence of the glucoamylase. Since this glucoamylase gene was not expressed in yeast cells, we have cloned a glucoamylase gene from a cDNA library prepared from Rhizopus mRNA. Sequence analysis of both glucoamylase genes revealed that the genomic gene contained 4 intervening sequences and the cDNA gene lacked 145 nucleotides corresponding to the N-terminal region. The glucoamylase consists of 604 amino acids including a putative signal peptide and its molecular weight was calculated to be 65,000. The glucoamylase gene to be expressed in yeast cells was constructed by recombination of both genes. The yeast cells containing this constructed glucoamylase gene secreted the glucoamylase into the culture fluid and grew at almost the normal rate on a medium containing starch as the sole carbon source. 相似文献

An enzymatic method for glycolaldehyde production from ethylene glycol was investigated using immobilized alcohol oxidase and catalase. Those enzymes were immobilized onto Chitopearl BCW 3501. When only alcohol oxidase was immobilized onto it, the apparent activity was 190 units/g in wet gel using methanol as the substrate. Tris-HCl buffer (1.5 M; pH 9.0) was selected based on a high stability of glycolaldehyde and a low production of glyoxal as a by-product. Under the optimum conditions, 0.97 M glycolaldehyde was formed from 1.0 M ethylene glycol and the ratio of glyoxal to glycolaldehyde was less than 1%. 相似文献

Fifty four samples including 5 of broken rice, 8 of corn grains, 8 of corn gluten feed, 13 of cottonseed cake and 4 each of rice polish, corn gluten, sesame oil cake, guar meal and wheat bran were screened for the presence of aflatoxins. Among all the samples, 14 were damaged and 40 apparently undamaged. The incidende of aflatoxins was found to be 60, 25, 25, and 23 per cent in broken rice, corn grains, corn gluten feed and cottonseed cake. Aflatoxins were not detected from rice polish, corn gluten, sesame oil cake, guar meal and wheat bran. Damaged sample revealed a much higher incidence i.e. 50 per cent as compared to undamaged ones i.e. 7.5 per cent. Mean concentration of aflatoxin B and G was found to be 15.5 and 12.2 ppb respectively.Cultural examination of aflatoxin positive feedstuffs yielded 39 isolates of different fungi including 21 of Aspergillus, 7 of Mucor, 6 of Rhizopus, 4 of Fusarium and one of Penicillium. These strains when tested for aflatoxin producing ability, revealed this property in only one isolate, identified as Aspergillus parasiticus. 相似文献

Naturalized plants play pivotal roles in local plant biodiversity and ecological functions; however, the drivers of naturalization remain poorly understood at a fine scale. Thus, understanding the processes of the development and dominance of alien plants in local natural habitats is of paramount importance. In the present study, we report for the first time the naturalization of Cydonia oblonga in China based on community structure and population dynamics at a fine scale. We conducted a comprehensive survey of the species through field community investigations, interviews, and a literature review. Cydonia oblonga is an ancient fruit tree with a long introduction history of over 4500 years worldwide and a cultivation history of over 2500 years in China. We analyzed C. oblonga community structure using the spatiotemporal substitution method and quantitatively analyzed population dynamics using a static life table, survivorship curve, and time series model to explore the naturalization processes. The following results were obtained. (i) The community comprised 31 coexisting vascular plant species (16 woody and 15 herbaceous species) belonging to 28 genera in 20 families. Rosaceae and Asteraceae were the two most dominant families. (ii) All individuals in the shrub layer as well as the C. oblonga population exhibited a roughly inverted J-shaped basal diameter distribution. A complete age structure was noted, and the survival curve was classified as Deevey type II. According to time series analysis, the population is estimated to increase in the future, specifically of medium and large individuals. (iii) Religious exchange, potent resource competitiveness, and similarity with the native habitat may be the major drivers of the introduction and successful naturalization of C. oblonga. These results suggest that alien species closely related to native ones are more likely to invade, naturalize, and dominate communities in local habitats. 相似文献

During an investigation of the mycoflora on oilseed rape, the predominant fungal species present in 20 samples collected from Catalonia (Spain) wereAlternaria alternata (Fries) Keissler,Penicillium spp. andAspergillus flavus. None of the 20 samples analyzed presented contamination byAlternaria mycotoxins (tenuazonic acid, alternariol, alternariol methyl ether, altertoxin I and altertoxin II). Only aflatoxin B1 was detected in 1 of the 20 samples analyzed, with a concentration of 0.25 ppb. In order to clearly establish the properties of the enzymes responsible for hexose phosphorylation we have undertaken the
separation and characterization of these enzymes present in tomato fruit (Martinez-Barajas and Randall 1996). This report
describes the partial purification and characterization of glucokinase (EC. 2.7.1.1) from young green tomato fruit. The procedure
yielded a 360-fold enrichment of glucokinase. Tomato fruit glucokinase is a monomer with a molecular mass of 53 kDa. Glucokinase
activity was optimal between pH 7.5 and 8.5, preferred ATP as the phosphate donor (K
m = 0.223 mM) and exhibited low activity with GTP or UTP. The tomato fruit glucokinase showed highest affinity for glucose
(K
m = 65 μM). Activity observed with glucose was 4-fold greater than with mannose and 50-fold greater than with fructose. The tomato
fruit glucokinase was sensitive to product inhibition by ADP (K
i = 36 μM). Little inhibition was observed with glucose 6-phosphate (up to 15 mM) at pH 8.0; however, at pH 7.0 glucokinase
activity was inhibited 30–50% by physiological concentrations of glucose 6-phosphate.

Five strains of Aspergillus flavus lacking the ability to produce aflatoxins were examined in greenhouse tests for the ability to prevent a toxigenic strain from contaminating developing cottonseed with aflatoxins. All atoxigenic strains reduced contamination when inoculated into developing bolls 24 h prior to the toxigenic strain. However, only one strain, AF36, was highly effective when inoculated simultaneously with the toxigenic strain. All five strains were able to inhibit aflatoxin production by the toxigenic strain in liquid fermentation. Thus, in vitro activity did not predict the ability of an atoxigenic strain to prevent contamination of developing bolls. Therefore, strain selection for competitive exclusion to prevent aflatoxin contamination should include evaluation of efficacy in developing crops prior to field release. Atoxigenic strains were also characterized by the ability to convert several aflatoxin precursors into aflatoxin B1. Four atoxigenic strains failed to convert any of the aflatoxin biosynthetic precursors to aflatoxins. However, the strain (AF36) most effective in preventing aflatoxin contamination in developing bolls converted all tested precursors into aflatoxin B1, indicating that this strain made enzymes in the aflatoxin biosynthetic pathway. 相似文献
