Seaweed extract, prepared by alkaline extraction of Ascophyllum nodosum (L.) Le Jol., applied either to the soil or to the foliage of tomato plants, produced leaves with higher chlorophyll levels than those of control plants. The effects on leaf chlorophyll content were investigated using a cucumber bioassay procedure devised for cytokinins. The seaweed extract was shown to increase the chlorophyll levels of the cucumber cotyledons, but ‘peaks’ of activity were obtained when widely different concentrations were used. The possibility that these effects were the result of betaines present in the extract was considered. Glycinebetaine, γ-aminobutyric acid betaine and δ-aminovaleric acid betaine all produced significantly enhanced chlorophyll concentrations in the cotyledons. ‘Peaks’ of activity were observed for each betaine: for glycinebetaine at 10−6 and between 10−4 and 101 mg 1−1, for γ-aminobutyric acid betaine at 10−6, between 10−4 and 10−1, and 101 mg 1−1, and for δ-aminovaleric acid betaine between 10−5 and 101 mg 1−1. It was concluded that the effects of enhancing chlorophyll levels produced by the seaweed extract were due, at least in part, to betaines.  相似文献

Enhanced leaf chlorophyll levels in plants treated with seaweed extract   总被引:3,自引:0,他引:3  
Application to the soil of an aqueous alkaline extract ofAscophyllum nodosum resulted in higher concentrations of chlorophyll in the leaves of treated plants in comparison to control plants treated with an equivalent volume of water. Positive results were obtained with all species tested (tomato, dwarf French bean, wheat, barley, maize). When the seaweed extract was applied as a foliar spray, similar effects on leaf chlorophyll contents were obtained, except in the case of dwarf French bean plants, for which no significant difference was recorded between test and control plants. When the betaines present in the seaweed extract were applied as a mixture in the same concentrations as those in the diluted seaweed extract (-aminobutyric acid betaine 0.96 mg L–1, -aminovaleric acid betaine 0.43 mg L–1, glycinebetaine 0.34 mg L–1), very similar leaf chlorophyll levels were recorded for the seaweed extract and betaine treated plants. This suggests strongly that the enhanced leaf chlorophyll content of plants treated with seaweed extract is dependent on the betaines present.  相似文献

Treatment of Arabidopsis thaliana plants with a commercially-available, alkaline extract of the marine brown alga, Ascophyllum nodosum, resulted in a significant decrease in the number of females of the root-knot nematode, Meloidogyne javanica, which developed in the roots compared to those of plants grown in a water control medium. Significant reductions in egg recovery were also achieved from plants treated with the seaweed extract. Similar effects were produced when betaine components of the seaweed extract (γ-aminobutyric acid betaine, δ-aminovaleric acid betaine and glycinebetaine) were used in quantities equivalent to those applied in the seaweed extract treatment. As the experiments were conducted under monoxenic conditions, it can be concluded that the results obtained with the application of either the seaweed extract or betaines are indicative of their effects on the plants and are not dependent on microorganisms associated with the rhizosphere. An off-line, overpressured layer chromatographic procedure has been developed for the evaluation of betaines in commercial seaweed extracts using Dragendorff's reagent for the detection of the compounds and densitometry for their quantitative estimation. Using continuous development and a low sample volume, the amounts of -aminobutyric acid betaine and -aminovaleric acid betaine can be estimated, but a larger sample volume is necessary for the estimation of glycinebetaine and minor betaines.The critical steps of this analytical method are the visualization and the quantitative evaluation of the spots produced. Temperature plays a major role in the resolution, sensitivity and precision after derivatization with Dragendorff's reagent; this has led to the adoption of standardized conditions.Author for correspondence  相似文献

Aqueous extracts of Ascophyllum nodosum and several other brown seaweeds are manufactured commercially and widely distributed for use on agricultural crops. The increasingly regulated international trade in such products requires that they be standardized and defined to a degree not previously required. We examined commercially available extracts using quantitative 1H NMR and principal components analysis (PCA) techniques. Extracts manufactured over a 4-year period using the same process exhibited characteristic profiles that, on PCA, clustered as a discrete group distinct from the other commercial products examined. In addition to recognizing extracts made from different seaweeds, analysis of the 1H spectra in the 0.35–4.70 ppm region allowed us to distinguish amongst extracts produced from the same algal species by different manufacturers. This result established that the process used to make an extract is an important variable in defining its composition. A comparison of the 1H NMR integrals for the regions 1.0–3.0 ppm and 3.0–4.38 ppm revealed small but significant changes in the A. nodosum spectra that we attribute to seasonal variation in gross composition of the harvested seaweed. Such changes are reflected in the PCA scores plots and contribute to the scatter observed within the data point cluster observed for Acadian soluble extracts when all data are pooled. Quantitative analysis using 1H NMR (qNMR) with a certified external standard (caffeine) showed a linear relationship with extract concentration over at least an order of magnitude (2.5–33 mg/mL; R 2 > 0.97) for both spectral regions integrated. We conclude that qNMR can be used to profile (or “fingerprint”) commercial seaweed extracts and to quantify the amount of extract present relative to a suitably chosen standard. Extracts of the brown seaweed Ascophyllum nodosum have been used as a biostimulant to promote growth and productivity in a number of agricultural production systems. Although the extracts have been shown to improve seedling emergence and vigor in a variety of plants, including barley, the mechanism(s) of this growth-promoting effect is(are) largely unknown. In our study, A. nodosum extract induced amylase activity in barley seed-halves; a significant difference in amylase activity was observed in seeds without an embryo. The addition of activated charcoal to the treatment media negated the bioactivity of the extracts suggesting the organic nature of bioactive compounds in A. nodosum extracts. The extracts induced amylase activity in a gibberellic acid (GA)-deficient barley mutant (grd2). LC-MS-MS analysis failed to detect the presence of GA3 in the extracts. ABA supplementation of the medium caused a significant reduction of amylase activity in GA-treated seeds compared with those treated with the A. nodosum extract. Taken together, our results suggest that the organic components of A. nodosum extract induce amylase activity independent of GA3 and might act in concert with GA-dependent amylase production leading to enhanced germination and seedling vigor in barley. Being derived from a renewable resource, the bioactive compounds from A. nodosum could be used to improve crop productivity in sustainable agricultural systems.  相似文献

The extracts obtained from 28 species of marine algae were evaluated for their antioxidant activity (AA) versus the positive controls butylated hydroxytoluene (BHT), gallic acid (GA), and ascorbic acid (AscA). Most of the tested samples displayed antioxidant activity to various degrees. Among them, the extract of Symphyocladia latiuscula exhibited the strongest AA, which was comparable to BHT, GA, and AscA in radical scavenging activity, as shown in the DPPH (α,α-diphenyl-β-picrylhydrazyl) assay, and higher than those of the positive controls in β-carotene-linoleate assay system. In addition, the ethyl acetate-soluble fraction isolated from the crude extract of S. latiuscula exhibited the highest antioxidant activity in both assay systems. This fraction was further fractionated into seven subfractions (F1-F7) by vacuum liquid chromatography (VLC). F1 and F4 were found to be the most effective subfractions in scavenging DPPH radical assay and in the β-carotene-linoleate assay, respectively. The total phenolic content (TPC) and reducing power (RP) for all of the extracts, fractions, and subfractions (F1–F7) were also determined. The TPC of the 28 extracts ranged from 0.10 to 8.00 gallic acid equivalents (mg/g seaweed dry weight) while the RP ranged from 0.07 to 11.60 ascorbic acid equivalents (mg·g−1 seaweed dry weight). Highly positive relationships between AA and TPC as well as between AA and RP were found for the extracts and fractions, while for the subfractions F1–F7 only weak or no such relations were found. The results obtained from this study indicate that further analysis is needed of those marine algal species that contain the most antioxidant activity in order to identify the active principles.  相似文献

The aims of this study were to characterise the composition of seaweed species and to evaluate the efficacy of aqueous extracts as plant biostimulants. Five species (Ascophyllum nodosum, Fucus serratus, Fucus vesiculosus, Laminaria hyperborea and Sargassum muticum) of seaweed were harvested from Strangford Lough, Northern Ireland for the evaluation of polysaccharides, indole-3-acetic acid (IAA), carbon, nitrogen, lipid, ash and mineral contents. The compositional analyses of the five species and their freeze-dried extracts were also carried out using thermogravimetric analysis, scanning electron microscopy and X-ray microanalysis. The concentration of IAA in the acid extracts of the five species ranged between 2.74 and 46.8?ng?g?1. The carbon, nitrogen, lipid and ash contents ranged between 25.0 and 38.6, 1.37%, and 3.16, 0.83%, and 3.98 and 18.10 and 47.68%, respectively. L. hyperborea and S. muticum contained the highest amounts of minerals. The biostimulant activities of acidic (pH?3.0), neutral (pH?6.5) and alkaline (pH?9.0) extracts were determined by mung bean bioassay. The alkaline extracts from F. vesiculosus and A. nodosum stimulated significantly (P?<?0.001) higher dry matter (DM, %) yield of the mung bean plants. The majority of the acidic extracts significantly (P?<?0.001) enhanced root formation on the mung bean stem cuttings compared to alkaline or neutral extracts. The acidic extracts of the five species, water control and a commercial product were evaluated as foliar feeds for pak choi plants using a hydroponic production system. The interaction of species, e.g. A. nodosum and F. vesiculosus and the two treatment dilutions on DM yield increases of pak choi were significant (P?<?0.05).  相似文献

Ascophyllum nodosum extract products are used commercially in the form of liquid concentrate and soluble powder. These formulations are manufactured from seaweeds that are harvested from natural habitats with inherent environmental variability. The seaweeds by themselves are at different stages of their development life-cycle. Owing to these differences, there could be variability in chemical composition that could in turn affect product consistency and performance. Here, we have tested the applicability of using Arabidopsis thaliana as a model to study the activity of two different extracts from A. nodosum. Three different bioassays: Arabidopsis root-tip elongation bioassay, Arabidopsis liquid growth bioassay and greenhouse growth bioassay were evaluated as growth assays. Our results indicate that both extracts promoted root and shoot growth in comparison to controls. Further, using Arabidopsis plants with a DR5:GUS reporter gene construct, we provide evidence that components of the commercial A. nodosum extracts modulates the concentration and localisation of auxins which could account, at least in part, for the enhanced plant growth. The results suggest that A. thaliana could be used effectively as a rapid means to test the bioactivity of seaweed extracts and fractions.  相似文献

Anti‐herbivory defenses support persistence of seaweeds. Little is known, however, about temporal dynamics in the induction of grazer‐deterrent seaweed traits. In two induction experiments, consumption rates of the periwinkle Littorina obtusata (L.) on the brown seaweed Ascophyllum nodosum (L.) Le Jolis were measured in 3‐d intervals. Changes in palatability of directly grazed A. nodosum were tested every 3 d with feeding assays using fresh and reconstituted seaweed pieces. Likewise, assays with fresh A. nodosum assessed changes in seaweed palatability in response to water‐borne cues from nearby grazed conspecifics. Consumption rates of L. obtusata varied significantly during the 27‐d induction phase of each experiment. Direct grazing by L. obtusata lowered palatability of fresh and reconstituted A. nodosum pieces to conspecific grazers after 15 d as well as after 6 and 12 d, respectively. After 12, 18, and 24 d, fresh A. nodosum located downstream of L. obtusata‐grazed conspecifics was significantly less palatable than A. nodosum located downstream of ungrazed conspecifics. Changes in L. obtusata consumption rates and A. nodosum palatability during both induction experiments suggest temporal variation of grazer‐deterrent responses, which may complicate experimental detection of inducible anti‐herbivory defenses.  相似文献

The characterization of T. vulgaris plant material for quality control purposes was performed by NMR-based methods. Direct extraction of 141 T. vulgaris samples with DMSO-d 6 enabled the obtainment of crude extracts with a representative composition in terms of both volatile and non-volatile constituents. The acquisition of 600 MHz 1H NMR spectra resulted in a dataset which was analyzed by a combination of metabolic profiling and target analysis approaches. Preliminary analysis of the 1H NMR spectra was performed by principal component analysis, which revealed sample discrimination on a chemotype basis (thymol, carvacrol and linalool chemotypes). Further minor discriminative constituents were identified as p-cymene, γ-terpinene, rosmarinic acid, and 3,4,3′,4′-tetrahydroxy-5,5′-diisopropyl-2,2′-dimethylbiphenyl. Metabolite identification was accomplished by 1D and 2D NMR techniques and supported by spiking experiments. Fast dereplication of constituents not available as reference compounds was performed by HPLC–SPE–NMR experiments. A targeted approach based on qHNMR was validated for quantification of the identified secondary metabolites. Validation was performed in terms of precision (intra-day RSD ≤ 4.51%, inter-day RSD ≤ 4.18%), repeatability (RSD ≤ 2.30%), accuracy (recovery rates within 93.4 and 103.4%), linearity (correlation coefficients ≥ 0.9990), robustness, and stability. The amount of the dominant monoterpene in thymol, carvacrol, and linalool chemotypes was respectively found to be within 0.4–2.6, 0.7–2.3, and 1.1–3.6% (w/w). Variable amounts of the precursors p-cymene and γ-terpinene were found in thymol and carvacrol chemotypes. The highest amount of rosmarinic acid and 3,4,3′,4′-tetrahydroxy-5,5′-diisopropyl-2,2′-dimethylbiphenyl in the analyzed samples was respectively 4.6 and 0.4% (w/w). Since quantification is performed on a weight basis, the essential oil content can be estimated based on the sum of the quantified monoterpenes. The NMR-based analysis of T. vulgaris represents a more comprehensive approach in comparison to traditional chromatographic methods such as GC and LC, respectively employed for the analysis of volatile and non-volatile constituents. Further advantages lie in the simple sample preparation, rapidity and reproducibility of the NMR analysis.  相似文献

With the increasing demand for seaweed resources worldwide, management must ensure that the harvest of wild seaweed stocks is sustainable. We evaluate the impact of over 25 years of commercial harvesting of Ascophyllum nodosum in eastern Canada by comparing the biomass and height of the seaweed in the late 1990s to the late 2010s over a broad spatial scale spanning the provinces of Nova Scotia and New Brunswick. There has been no significant decrease in the biomass of A. nodosum in either province, and biomass has increased in some regions of New Brunswick during that period. The average height of A. nodosum has decreased by 7.8 cm in Nova Scotia while it increased by 13.8 cm in New Brunswick. Biomass of A. nodosum in unharvested sectors was 7% higher than that in harvested sectors while height of A. nodosum in unharvested sectors in New Brunswick is similar to the values observed in harvested sectors. Over the same period, water temperature has increased in both provinces and, in recent years, has at times exceeded the optimal growing temperature for A. nodosum within bays in Nova Scotia. We conclude that the current management and harvest of A. nodosum in eastern Canada are sustainable and maintain the biomass and height of A. nodosum beds but that control sites are necessary to offer adequate comparisons as environmental conditions are changing.

The aims of this study were to characterise the composition of five seaweed species (Ascophyllum nodosum, Fucus serratus, Fucus vesiculosus, Laminaria hyperborea and Sargassum muticum), their extracts and commercial formulations, using thermogravimetry (TGA), energy dispersive X-ray microanalysis (EDX), Fourier-transform infrared spectroscopy (FTIR) and pyrolysis gas chromatography/mass spectrometry (Py-GC/MS). Analyses of the samples by TGA and EDX provided information on the proportions of algal cell wall, inorganic fractions and minerals. The main carbohydrate constituents of the five species and extracts were identified by their pyrolysis products, e.g. 1-(2-furanyl) ethanone, 5-methyl-2-furcarboxaldehyde, 2-hydroxy-3-methyl-2-cyclopenten-1-one, diannhydromannitol, 1,6-anhydromannopyranose and 1,6-anhydromannofuranose, using Py-GC/MS. The differences in relative intensities of the infrared bands of the five species were enhanced, especially after acid extraction compared with alkaline or neutral treatments, resulting in improved understanding of the compositional changes. In addition four commercial formulations and two acidic extracts of A. nodosum were evaluated for composition using the techniques. The dry matter, pH, electrical conductivity, ash, carbon and nitrogen content of the six preparations showed significant differences in composition. Variations in fatty acid, alginic acid, mannitol, laminarin and fucoidan content of the six formulations were reported. The results have shown that TGA, EDX, Py-GC/MS and FTIR are complementary techniques for rapid evaluation of seaweed materials and products.  相似文献

Spring barley (Hordeum vulgare cv. Triumph) was grown hydroponically over a 6-week period. Two treatments were incorporated either into the hydroponic solution or sprayed onto the plants at rates of 1 ml per 3 litres. The treatments applied were: (i) a seaweed concentrate prepared fromAscophyllum nodosum (L.) Le Jolis (marketed as Maxicrop Triple), (ii) a ‘Trace element’ treatment incorporating the micro and macro nutrients added to the seaweed extract base to produce the formulated product Maxicrop Triple and (iii) a control treatment. Irrespective of the mode of application, plants treated with Maxicrop Triple grew faster than plants under either of the two other treatments. Elevated growth rates were also found for the ‘Trace element’ treated plants when incorporated into the hydroponic solution. At the final harvest, plants with Maxicrop Triple incorporated into the hydroponic solution showed increases from 56–63% over the control treatment for the growth characteristics measured. ‘Trace element’-treated plants produced increases of between 25–45%. When the treatments were sprayed the effect was less pronounced. Maxicrop Triple increased growth characters by 35–38% and the ‘trace element’ treatment gave increases in the range of 2–13%.  相似文献

A locally isolated Gram-negative bacterium, Cupriavidus sp. USMAA2-4 was found capable of producing terpolymer poly(3-hydroxybutyrate-co-3-hydroxyvalerate-co-4-hydroxybutyrate) [P(3HB-co-3HV-co-4HB)] using γ-butyrolactone or 1,4-butanediol with either valeric acid or 1-pentanol as the carbon source. The present of 3HB, 3HV and 4HB monomers were confirmed by gas chromatography (GC) and nuclear magnetic resonance (NMR) analysis. PHA concentration of 1.9 g/l was the highest value obtained using the combination of 1,4-butanediol and 1-pentanol through one-step cultivation process. PHA concentration obtained through two-step cultivation process was higher for all the combinations and the highest value achieved was 2.5 g/l using γ-butyrolactone and 1-pentanol as carbon source. Various molar fractions of 4HB and 3HV ranging from 6 to 14 mol% and 39 to 87 mol%, respectively were produced through two-step cultivation process by manipulating the concentration of γ-butyrolactone. As the culture aeration was reduced, the molar fraction of 3HV and 4HB increased from 40 to 67 mol% and 10 to 24 mol%, respectively while the dry cell weight and PHA content decreased. The terpolymer produced was characterized using gel permeation chromatography (GPC) and differential scanning calorimetry (DSC). The number-average molecular weight (M n) and the melting temperature (T m)) of the terpolymer were in the range of 177–484 kDa and 160–164°C, respectively.  相似文献

The tetrasporophyte of Asparagopsis armata has been previously established as a novel seaweed biofilter for integrated land-based mariculture. The species growth and biofiltration rates were much higher than the values described in the literature for Ulva spp., the most common seaweed biofilter. However, a validation of the advantage of one species over the other requires a study of the performances of these two species in the same system at the same time. In this work, we compared the biofiltration performance and biomass yield of A. armata and Ulva rigida cultivated in the effluents of a fish farm in southern Portugal. Comparisons were performed at different water renewal rates and in two seasons of the year. The maximum total ammonia nitrogen (TAN) removal rates were similar for both species in December (2.7 and 2.8 g TAN m–2 day–1 for U. rigida and A. armata, respectively) and higher for A. armata (6.5 g TAN m–2 day–1) than for U. rigida (5.1 g TAN m–2 day–1) in May. Higher differences were observed when estimating the nitrogen biofiltration through the organic nitrogen yield (N yield) of the biomass produced, particularly in May. This estimate is directly related with the biomass yield and the N content in the tissue which were always higher for A. armata than for U. rigida. In December, the maximum biomass yields were 71 g dry weight (DW) m–2 day–1 for A. armata and 44 g DW m–2 day–1 for U. rigida, while in May, the yield of A. armata was 125 g DW m–2 day–1 and of U. rigida was 73 g DW m–2 day–1. This study confirmed that A. armata is indeed a more efficient biofilter than U. rigida. To the best of our knowledge, the production rates reported here are the highest ever reported for macroalgae cultivated in tanks.  相似文献

The production of methane biofuel from seaweeds is limited by the hydrolysis of polysaccharides. The rumen microbiota of seaweed‐eating North Ronaldsay sheep was studied for polysaccharidic bacterial isolates degrading brown‐seaweed polysaccharides. Only nine isolates out of 65 utilized > 90% of the polysaccharide they were isolated on. The nine isolates (eight Prevotella spp. and one Clostridium butyricum) utilized whole Laminaria hyperborea extract and a range of seaweed polysaccharides, including alginate (seven out of nine isolates), laminarin and carboxymethylcellulose (eight out of nine isolates); while two out of nine isolates additionally hydrolysed fucoidan to some extent. Crude enzyme extracts from three of the isolates studied further had diverse glycosidases and polysaccharidase activities; particularly against laminarin and alginate (two isolates were shown to have alginate lyase activity) and notably fucoidan and carageenan (one isolate). In serial culture rumen microbiota hydrolysed a range of seaweed polysaccharides (fucoidan to a notably lesser degree) and homogenates of L. hyperborea, mixed Fucus spp. and Ascophyllum nodosum to produce methane and acetate. The rumen microbiota and isolates represent potential adjunct organisms or enzymes which may improve hydrolysis of seaweed components and thus improve the efficiency of seaweed anaerobic digestion for methane biofuel production.  相似文献
