Ultrastructural investigation and quantitative analysis of endocrine cells of intestine were studied in mice. The results obtained allow the quantitative composition and cytotopography of endocrine cells in different sections of small and large intestine in mice. The combination of these methods obtained allow to describe 15 types of endocrinocytes, and to demonstrate these localizations and electron microscopy characteristics.  相似文献

5-20 oocytes or cleaving embryos of CBA mice were dissolves in 0.2-0.5 ml of 1% buffered sodium dodecylsulfate (SDS), and soluble proteins were separated using electrophoresis in capillaries filled with PAA-SDS-gel. The method enabled us to calculate the approximate molecular weight of proteins by relative mobilities and allowed to determine 1-5 mmcg protein in a single band. 7 groups of proteins were identified in mouse oocytes using 10% PAA-0.1% SDS-gel with the following molecular weights: 208 000, 206 000, 155 000, 112 000, 59 000, 40 000 and 30 000, resp. The same groups of protein were discovered in zygotes and cleaving mouse embryos. The molecular weight distribution of low-molecular weight basic proteins of CBA mice is species-specific and reveals a qualitative changes in the early embryogenesis.  相似文献

The simulation of human serum levels is essential in animal models to extrapolate the experimental results to clinical practice. Administration of a nephrotoxic drug such as cisplatin can be used to cause renal dysfunction as an approach to mimic human serum levels of renally excreted drugs. We aimed to determine the dose of cisplatin that did not affect the survival rate of mice and to achieve human-like serum concentrations of cefepime. Different doses of cisplatin (0, 10, 14, 18, 22 and 26 mg/kg) were given by intraperitoneal (i.p.) injection to mice three days prior to the i.p. administration of 80 mg/kg cefepime. With cisplatin doses of 18 and 22 mg/kg, the half-life of cefepime was significantly prolonged (P < 0.001) and all mice survived. The pretreatment with 26 mg/kg cisplatin significantly decreased survival (P = 0.001), but the half-life of cefepime was not significantly longer than of 18 mg/kg cisplatin. Serum levels of cefepime after the pretreatment with 18 mg/kg cisplatin were comparable to published human data. The administration of cisplatin appears to be a suitable method in mice for simulating human serum concentrations of renally excreted drugs.  相似文献

Examination of 339 children at the age of 2 to 12 years, living in the caesium contaminated region (from 2 to 5 Ci/km2) has revealed that total radioactivity of their urine is, on the average, twice as high as that of children living in "pure" regions. Quantitative and qualitative changes were observed in the erythroid series, neutrophilic leukocytes, eosinophils and B-lymphocytes of the peripheral blood of children subjected to long-term low-level irradiation. It should be noted that the character and direction of these changes were a function of the children's age and the level of total radioactivity of their urine.  相似文献

A complicated character of the cytogenetic injury dependence upon radiation dose was revealed after low-level gamma irradiation of Vicia faba seedlings and Chinese hamster fibroblasts. The dependence was linear with low-level secondary exposure to 70 GeV protons. The authors discuss a threshold nature of induction of the cytogenetic damage repair responsible for a high outcome of damages under the effect of low-level gamma radiation.  相似文献

The influence of cyclosporine which is an immunodepressant on morphology of the lymphoid organs was studied on CBA mice. The immunodepressant was administered intramuscularly in a single LD50 and in a dose of 7 mg/kg for the treatment course of 21 days. The examinations were performed in various periods within 28 days after discontinuation of the drug use. It was shown that cyclosporine induced cell depletion in the thymus cortical and medullar zones, inhibition of lymphocyte mitotic activity, alteration of the Hassall corpuscles and impairment of their formation. It also induced devastation of the thymus-dependent zones in the mesenteric lymph nodes and spleen. When cyclosporine was used in the course doses the morphological changes in all the lymphoid organs were more marked. The morphological changes were reversible.  相似文献

In the male rat, castration causes an increase of the LH level measured by radioimmunoassay. The application of a compound extracted from a impuber child urine and having anti-gonadotropic effects revealed biologically cannot stop this increase that is even put up by the treatment.  相似文献

Golgi preparations of cervical part of the spinal cord of 30-day kittens were used to study sparely and densely branching neurons of lamina VII, sparely and densely branching neurons of lamina VIII and big densely branching motor neurons (as classified by Leontovich) of medial and lateral regions of lamina IX. Qualitative morphological characteristics of geometry of each cell type were obtained by the method of computerized morphometry. The details of the structure of neurons belonging to different laminae of grey matter are discussed.  相似文献

Conditions have been established for the effective separation of Fc+ and Fc- cells, using sheep red blood cells monolayer, sensitized with the anti-SRBC antibodies. The method was modified by replacement of poly-L-lysine by protamine sulfate. The electrophoretic distribution profiles of Fc+ and Fc- mouse splenocytes were determined. Fc+ cells had mostly a slow electrophoretic mobility. The Fc- cells were more mobile. The Fc+ fraction contained mainly B-lymphocytes, and the Fc- fraction was enriched with T cells. There was an extensive overlap between the electrophoretic profiles of the Fc+ and Fc- splenocytes. After the splenocyte passage through the glass bead column the overlap became much less.  相似文献

Protective effect of cystamine (150 mg/kg) against genetic damages induced by gamma-irradiation in germ cells of male mice of CBA strain (at doses of 100, 300, 600 r) was studied. The application of cystamine decreased the frequency of dominant lethal mutations induced by radiation in sperms, spermatids and spermatocytes. The degree of the protective effect of cystamine depended on a radiation dose. The protective effect of cystamine was the highest at the radiation dose of 300 r. It was negligible at the radiation dose of 100 r and was completely absent at the dose of 600 r. Cystamine did not affect the rate of induced reciprocal translocations in the spermatogonia at all the radiation doses used.  相似文献

Limitation was induced by electrocoagulation of bulbar magnocellular nucleus and caudal reticular pontine nucleus performed at the 7th day. Computerized morphometry revealed that partial deafferentation affects the geometry in all studied cell types except densely branching neurons of lamina VIII. Adaptive nature of structural reorganization of spinal neurons and possible relationship between morphological properties and functional profile of cells are discussed.  相似文献

The present study aims to shed light on the origin of abnormal oocytes ovulated by aged females. In order to reach this goal, cellular and morphological traits of ovulated oocytes from hybrid (C57Bl/6JIco female x CBA/JIco male) female mice retrieved after exogenous ovarian stimulation at the age of 12, 40-42, 50-52, or 57-62 wk were analyzed. Aging of female mice was associated with 1) decreased number of ovulated oocytes; 2) increased percentage of cumulus-free oocytes; 3) raised percentage of oocytes with intracellular mitochondrial aggregates; 4) reduced percentage of oocytes displaying a normal distribution of chromosomes in the metaphase-II plate; 5) increased percentage of normal oocytes exhibiting a DNA-containing polar body (PB); 6) higher percentage of oocytes with chromosome scattering; 7) increased percentage of chromosome-scattered oocytes without a DNA-containing PB and with intracytoplasmic mitochondrial aggregates; 8) raised percentage of oocytes exhibiting chromosome decondensation; 9) lower percentage of chromosome-decondensed oocytes lacking both a DNA-containing PB and intracytoplasmic mitochondrial aggregates; 10) increased percentage of abnormal/degenerated oocytes; 11) reduced percentage of abnormal/degenerated oocytes displaying cellular fragmentation; and 12) higher percentage of abnormal/degenerated oocytes with mitochondrial aggregates exhibiting no nuclear/chromosomal DNA fluorescence, cellular fragmentation, milky or dark cytoplasm, or cellular remains enclosed by the zona pellucida. Although several studies suggest aging females may ovulate aged or overripened oocytes, these data support the hypothesis that old females ovulate an increased percentage of atretic/apoptotic oocytes coming from rescued follicles that would have become atretic earlier in life.  相似文献

After thawing cells, previously cryopreserved in the presence of dimethyl sulfoxide (DMSO), a decrease in their viability and increase in unscheduled DNA synthesis was observed. In 7 days, these parameters restored to the control level. Cryopreservation without DMSO resulted in the decrease in both cell viability and replicative and unscheduled DNA synthesis. In 14 days, these characteristics were seen to return to the normal level. Cryopreservation of cells without DMSO and their preservation in liquid nitrogen induced the frequency of chromosomal aberrations, mostly chromosomal breaks. The frequency of chromosomal aberrations increased with the duration of cell preservation in liquid nitrogen. The normal level was achieved following 7 days after cell thawing. Cells treated with DMSO only (without cryopreservation) display an increased number of chromosomal and chromatid breaks and translocations. Nonrandom distribution of chromosomal aberrations was observed, with particular chromosomes being involved in the appearance of dicentrics and translocations. The data obtained indicate that cryoprotective activity of DMSO is probably associated with the cell repair systems. The detected antimutagenic and mutagenic activity of DMSO may presumably reflect various conditions for its interaction with cells (with or without cryopreservation), as well as it may be specific for the muntjac cell line used in the present work.  相似文献
