目的:研究鼠李糖脂对微生物降解石油烃废水的影响.方法:通过测定生物量和观察菌株表面来研究鼠李糖脂对菌株的影响;通过正交实验设计,确定石油烃降解率影响因素.通过石油烃降解率的测定,探讨鼠李糖脂与H2O2深度氧化协同作用对微生物降解石油烃的影响.结果:菌株对石油烃的降解率达53%,在相同条件下,添加鼠李糖脂的石油烃降解率提高了12%-20%.添加鼠李糖脂后菌株的生物量明显增多,菌株细胞表面疏水.正交设计表明,影响石油烃降解的主导因子是培养温度,其次是培养时间和鼠李糖脂的添加量.正交设计得到最佳组合为A3B2C1,即培养时间为7d;温度为35℃,鼠李糖脂浓度为60mg/L.3个因素的最佳组合下,石油烃降解率为82%.加入200 mg/L的H2O2时,降解率从82%提高到97%.结论:鼠李糖脂能促进菌株的生长.鼠李糖脂与H2O2深度氧化协同作用有助于微生物对石油烃类污染物降解效率的提高.  相似文献

微生物对石油烃的降解在自然衰减去除土壤和地下水石油烃污染的过程中发挥了重要作用。微生物通过其产生的一系列酶来利用和降解这类有机污染物,其中,编码关键降解酶的基因称为功能基因。功能基因可作为生物标志物用于分析环境中石油烃降解基因的多样性。因此,研究石油降解功能基因是分析土著微生物群落多样性、评价自然衰减潜力与构建基因工程菌的重要基础。本文主要介绍了烷烃和芳香烃在有氧和无氧条件下的微生物降解途径,重点总结了烷烃和芳香烃降解的主要功能基因及其作用,包括参与羟化作用的单加氧酶和双加氧酶基因、延胡索酸加成反应的琥珀酸合酶基因以及中心中间产物的降解酶基因等。  相似文献

为了研究小黑麦对石油污染盐碱土壤中的细菌群落与石油烃降解率的影响,采用高通量测序技术,设置0 g/kg,1 g/kg和5 g/kg三个石油浓度,以未种植小黑麦的土壤作为对照,对6组不同处理的盐碱土壤样品的细菌群落结构及其多样性进行测定,并分析土壤中的石油烃降解率。结果表明:在土壤石油浓度为1 g/kg和5 g/kg时,小黑麦根际土壤中的石油烃降解率相较对照组分别提高了36.67%和33.20%。从6个土壤样品中分别获得21398—27899条测序序列。在石油污染土壤中,小黑麦根际土壤的细菌群落多样性和丰度均大于对照组的土壤。同时,在"门","纲","属"的分类水平上,小黑麦根际土壤细菌群落中的一些根际细菌的相对丰度增加了,主要包括变形菌门(Proteobacteria)、酸杆菌门(Acidobacteria)、γ-变形菌纲(Gammaproteobacteria)、烷烃降解菌科-未命名菌属(Alcanivoracaceae_norank)、黄单胞菌属(Xanthomonas)、亚硝化单胞菌-不可培养菌属(Nitrosomonadaceae_unculture)等。有一些相对丰度增加的根际细菌是以石油及石油分解物为碳源的微生物。本研究证明种植小黑麦改变了石油污染盐碱土壤根际土壤细菌群落结构组成和多样性,促进了降解石油微生物群落的构建,显著提高了盐碱土壤石油污染的降解效果。研究结果为石油污染盐碱土壤的植物修复奠定了理论基础。  相似文献

铁还原菌是指能够利用细胞外Fe(III)作为末端电子受体,通过氧化有机物将Fe(III)还原为Fe(II)微生物的总称。铁还原作用广泛存在于土壤、河流、海洋、地表含水层以及高温高压的地下深部油藏。在厌氧或兼性厌氧条件下,Fe(III)还原耦合有机物的降解,对铁、碳元素的生物地球化学循环具有重要意义。本文介绍了铁还原菌的多样性和铁还原作用机理,综述了铁还原菌在石油烃降解方面的研究进展。此外,还总结了铁还原菌在生物修复中的潜在作用,并对未来的研究方向进行了展望。  相似文献

为研究陇东黄土高原石油开采对土壤线虫影响的强度与范围,选择长庆油田不同开采年限(1、10a和20a)油井共6口,按照距井基不同距离(3、6、10、20m和50m)采集土壤样品90个,共鉴定出土壤线虫22科43属;其中,食细菌线虫26属、食真菌线虫2属、植物寄生线虫9属、杂食/捕食线虫6属,优势类群为小杆属Rhabditis和孔咽属Aporcelaimus。土壤线虫生活史策略以c-p 2类群占优势,不同距离采样点,土壤线虫c-p 2和c-p 5差异极显著(P0.01)。随着距井基距离增加(3、6、10、20m和50m)线虫总数显著增加;其中,植食类、捕食杂食类和食真菌类线虫数量增加显著,且线虫群落多样性指数(H')显著增加(P0.05)。但不同距离之间优势度指数(λ),均匀度指数(J)和瓦斯乐斯卡指数(WI)差异不显著。不同开采年限油井,以10a土壤线虫群落多样性指数(H')、成熟度指数(MI)和瓦斯乐斯卡指数(WI)最低,而1a油井和20a油井较高,且与10a油井差异显著(P0.05)。油井作业改变周围土壤性质,导致土壤含水率降低、总石油烃(TPH)含量增高,但土壤线虫分布与土壤TPH无明显相关。研究结果表明,油田开采影响土壤线虫的组成与群落结构,但影响范围一般局限在作业区范围,指数H'和MI能较好的指示油田开采对线虫群落的影响。  相似文献

采用盆栽实验研究了小叶白蜡(Fraxinus sogdiana)接种4种外生菌根真菌(E1-毛边滑锈伞(Hebeloma mesophaeusm)、E2-劣味乳菇(Lactarius insulsns)、E3-松塔牛肝菌(Stro-bilomyces floccopus)和E4-丝膜菌(Cortinarius russus)对沈抚灌区土壤石油烃的降解效果。结果表明:在白蜡不同组合双接种及混合接种中,以混合接种对土壤石油烃的降解效果最好,降解率比对照提高23.6%;其次为双接种中的E1E3和E2E4组合,降解率分别比对照提高21.0%和12.7%。接种外生菌根真菌可促进白蜡生长,尤其可明显提高其根生物量,增加侧根数,接种E1E3、E2E4和混合菌使白蜡侧根数分别增加了100%、67%和81%。相关分析表明,石油烃降解率与白蜡的侧根数呈显著相关,可能是其降解率提高的主要原因。  相似文献

餐厨垃圾厌氧消化处理主要过程的微生物群落结构分析   总被引:2,自引:0,他引:2  
【背景】厌氧消化是我国餐厨垃圾处理的主要方法,微生物在其处理过程中起到关键作用,但是目前对其不同工艺单元微生物群落结构的研究较少。【目的】通过分析各工艺单元的微生物多样性与群落结构,为改进餐厨垃圾资源化处理技术、提高资源利用效率提供科学依据。【方法】采集某餐厨垃圾处理厂油水分离、厌氧发酵、沼渣脱水等3个工艺单元产生的废液样品,采用16S rRNA基因高通量测序技术,研究其菌群组成、丰度、优势菌群及其与环境因子的相关性。【结果】初始油水分离样品中的微生物群落种类相对较少,而经厌氧发酵和沼渣脱水处理后样品中的微生物群落种类较丰富。在门水平上,厚壁菌门(Firmicutes)在各单元样品中所占平均比例最高,为81.1%,其次为拟杆菌门(Bacteroidetes)和绿弯菌门(Chloroflexi),分别占15.81%和4.59%;在属水平上,相对丰度较高的菌属为乳酸菌属(Lactobacillus)、互营单胞菌属(Syntrophomonas)等。餐厨垃圾处理过程中的部分菌属可能具有资源-环境双重属性,例如在沼渣脱水单元相对丰度高达32.67%的假单胞菌属(Pseudomonas),该菌属中既存在少部分致病菌或条件致病菌,也具有生产聚羟基脂肪酸酯的功能菌。影响各组样品微生物群落组成结构最显著的因子是p H值,其次是总糖的含量。【结论】研究明确了典型餐厨垃圾厌氧消化处理工艺单元的微生物群落结构和多样性,并提出了优化处理工艺、强化资源利用效率的建议。  相似文献

大庆油田石油开采对土壤线虫群落的影响   总被引:4,自引:0,他引:4  
选择大庆油田环境一致的6口油井,研究石油开采干扰对土壤线虫的影响。共鉴定出土壤线虫18科15属。食细菌线虫最多,共15属占总数的49.2%,其次为植物寄生线虫共9属,占30.6%,食真菌线虫4属,占17.6%,杂食/捕食线虫2属,占2.6%。其中优势类群为丝尾垫刃属Filenchus和头叶属Cephalobus,占总数的42.5%。常见类群包括13个属,占总数的51.7%。稀有类群15个属占总数的6.34%。土壤的线虫生活史策略以c-p 2所占比例最大,其次是c-p3和c-p 1类群,受油井开采作业等影响,线虫通过大量繁殖来应对环境的干扰与压力。不同油井线虫总数差异不显著,但所有油井线虫总数比对照明显减少(p<0.001)。不同油井线虫群落结构差异不大,各油井线虫富集指数和结构指数表明所有油井线虫受干扰程度较高,土壤食物网趋于退化。随着采样距离增加,线虫总量差异不显著,但线虫群落的物种数量增加,优势度指数和多样性指数逐渐增加。结果表明油井开采对土壤线虫群落有一定的影响,开采距离对线虫群落结构影响较大。  相似文献

考察了厌氧水稻土泥浆体系中高氯代多氯联苯混合物Aroclor1260的脱氯过程,并对体系中的微生物群落结构变化进行分析.结果表明: Aroclor1260可在厌氧水稻土泥浆体系中发生脱氯,经过128 d,总消减率达到55.5%,在泥浆体系中引入驯化的脱氯富集培养体反而使脱氯效果下降,消减率为46.9%.Aroclor1260的主要脱氯过程发生在五、六、七氯联苯,其中七氯联苯脱氯过程最显著,五氯联苯作为脱氯产物有一定累积.有机物厌氧发酵产生的H2会被脱氯过程所消耗,从而将体系中的氢分压维持在较低水平,抑制产甲烷过程而保证脱氯过程的持续进行.不同条件和培养方式驯化得到的微生物群落结构差异较大,富集培养体引入可能导致其与原体系中脱氯相关菌群竞争,从而改变体系原有菌群结构,这可能是导致其脱氯效率下降的原因.  相似文献

We studied the role of aerobic and anaerobic petroleum hydrocarbon degradation at a boreal, light-weight fuel and lubrication oil contaminated site undergoing natural attenuation. At the site, anoxic conditions prevailed with high concentrations of CH4 (up to 25% v/v) and CO2 (up to 18% v/v) in the soil gas throughout the year. Subsurface samples were obtained mainly from the anoxic parts of the site and they represented both the unsaturated and saturated zone. The samples were incubated in microcosms at near in situ conditions (i.e. in situ temperature 8 degrees C, aerobic and anaerobic conditions, no nutrient amendments) resulting in the removal of mineral oil (as determined by gas chromatography) aerobically as well as anaerobically. In the aerobic microcosms on average 31% and 27% of the initial mineral oil was removed during a 3- and 4-month incubation, respectively. In the anaerobic microcosms, on average 44% and 15% of the initial mineral oil was removed during a 12- and 10-month anaerobic incubation, respectively, and e.g. n-alkanes from C11 to C15 were removed. A methane production rate of up to 2.5 microg CH4 h(-1) g(-1) dwt was recorded in these microcosms. In the aerobic as well as anaerobic microcosms, typically 90% of the mineral oil degraded belonged to the mineral oil fraction that eluted from the gas chromatograph after C10 and before C15, while 10% belonged to the fraction that eluted after C15 and before C40. Our results suggest that anaerobic petroleum hydrocarbon degradation, including n-alkane degradation, under methanogenic conditions plays a significant role in the natural attenuation in boreal conditions.  相似文献

Studies were conducted using a 10-chamber Micro-Oxymax (Columbus, OH, USA) respirometer to determine the effect of bioaugmentation and biostimulation (by diverse ways of O2 supply) on enhancing biodegradation of oil hydrocarbons to reduce risk at a former military airport in Kluczewo, Poland. Indigenous or exogenous bacteria bioaugmentation was used to degrade hydrocarbons. Aerated water and/or aqueous solutions of H2O2 or KMnO4 were used to supply O2. The intrinsic and enhanced biodegradation was evaluated by the O2 uptake and CO2 production rates obtained using a linear regression of the cumulative O2 uptake and CO2 production curves. Generally, in all cases biodegradation rates enhanced by bioaugmentation were two to four times higher than the rates of intrinsic biodegradation. Moreover, application of indigenous bacteria was more efficient in comparison to the exogenous consortia. The highest CO2 production rates were achieved when aqueous solution of KMnO4 was applied, as the increase of CO2 production rates were about 71% to 97% higher compared to a control. The aqueous solution of H2O2 did not cause any significant improvement of the biodegradation rates. Compared to a control, the addition of aerated water resulted in a decrease of CO2 production rates. Most probably the excessive soil moisture could reduce the air-filled porosity and, consequently, the oxygen contents in soil.  相似文献

Anaerobic degradation of alkylbenzenes with side chains longer than that of toluene was studied in freshwater mud samples in the presence of nitrate. Two new denitrifying strains, EbN1 and PbN1, were isolated on ethylbenzene and n-propylbenzene, respectively. For comparison, two further denitrifying strains, ToN1 and mXyN1, were isolated from the same mud with toluene and m-xylene, respectively. Sequencing of 16SrDNA revealed a close relationship of the new isolates to Thauera selenatis. The strains exhibited different specific capacities for degradation of alkylbenzenes. In addition to ethylbenzene, strain EbN1 utilized toluence, but not propylbenzene. In contrast, propylbenzene-degrading strain PbN1 did not grow on toluene, but was able to utilize ethylbenzene. Strain ToN1 used toluene as the only hydrocarbon substrate, whereas strain mXyN1 utilized both toluene and m-xylene. Measurement of the degradation balance demonstrated complete oxidation of ethylbenzene to CO2 by strain EbN1. Further characteristic substrates of strains EbN1 and PbN1 were 1-phenylethanol and acetophenone. In contrast to the other isolates, strain mXyN1 did not grow on benzyl alcohol. Benzyl alcohol (also m-methylbenzyl alcohol) was even a specific inhibitor of toluene and m-xylene utilization by strain mXyN1. None of the strains was able to grow on any of the alkylbenzenes with oxygen as electron acceptor. However, polar aromatic compounds such as benzoate were utilized under both oxic and anoxic conditions. All four isolates grew anaerobically on crude oil. Gas chromatographic analysis of crude oil after growth of strain ToN1 revealed specific depletion of toluene.  相似文献

本研究采取辽河油田曙光采油厂、欢喜岭采油厂和锦州采油厂井场周边土壤,并以未污染稻田土壤作为对照,分析了各采样点的土壤理化性质、石油烃浓度组成及土壤微生物群落结构。结果表明: 1) 3个采油厂井场周边土壤均受到严重的石油烃污染,但其石油烃浓度及组成存在一定的差异,曙光和欢喜岭采油厂土壤石油烃平均浓度是锦州采油厂的2倍以上;曙光采油厂土壤中胶质沥青质含量最高,而欢喜岭和锦州采油厂土壤中烷烃含量最高,比例均在40%以上。2)与稻田土壤相比,锦州采油厂土壤微生物操作分类单元(OTU)、Chao1指数和Shannon指数升高,而其在曙光和欢喜岭采油厂土壤中降低;各采油厂土壤样品中存在相同的优势菌门及菌属,但丰度存在较大差异。锦州采油厂土壤中分枝杆菌属、假单胞菌属的丰度高,曙光采油厂土壤中鞘氨醇单胞菌属、类诺卡氏菌属、马赛菌属的丰度高,而欢喜岭采油厂土壤中溶杆菌属、硫杆菌属、假节杆菌属的丰度高。3)相关分析表明,鞘氨醇单胞菌属、类诺卡氏菌属、硫杆菌属、马赛菌属、假节杆菌属与总石油烃、总有机碳和胶质沥青质含量呈显著正相关,分枝杆菌属、溶杆菌属、假单胞菌属与总氮和总磷呈显著正相关。本研究系统分析了不同采油厂土壤中石油烃、土壤理化性质和微生物群落特征,揭示了辽河油田污染土壤中特定的优势菌属和群落结构,为辽河油田石油烃污染土壤修复功能微生物筛选及修复过程菌群构建提供理论依据,也为其他油田高效降解菌筛选提供了方法借鉴。  相似文献

【目的】研究降雨条件对浴场细菌总数和优势菌群组成的影响。【方法】2014年8月强降雨前后采集秦皇岛西浴场3个站位的海水样品,采用荧光显微镜计数法和平板计数法分别对细菌总数和可培养细菌总数进行计数;对群落结构组成进行分析,并对可培养细菌进行鉴定。【结果】雨前3个站位细菌总数和可培养细菌总数平均值分别为5.6×10~9 CFU/L和8.3×10~7 CFU/L,雨后分别为9.2×109 CFU/L和2.1×10~8 CFU/L。在可培养菌群中,变形菌门(Proteobacteria,雨前占80%,雨后占73%)是主要的微生物类群,其次为拟杆菌门(Bacteroides,雨前占12%,雨后占13%)、厚壁菌门(Firmicutes,雨前占7%,雨后占11%)等;肠杆菌属(Enterobacter spp.,21株)、海杆菌属(Marinobacter spp.,13株)、弓形菌属(Arcobacter spp.,13株)、假单胞菌属(Pseudomonas spp.,10株)、芽孢杆菌属(Bacillus spp.,10株)和弧菌属(Vibrio spp.,6株)为雨前可培养细菌优势属,而雨后可培养细菌优势属为肠杆菌属(22株)、海杆菌属(21株)、芽孢杆菌属(14株)、不动杆菌属(Acinetobacter spp.,11株)、假单胞菌属(9株)和弓形菌属(5株)等。【结论】降雨对细菌总数有显著的影响,同时降雨后浴场微生物群落结构发生了改变。  相似文献

目的:探讨金属桩与玻璃纤维桩对老年上颌前牙残根残冠修复的临床疗效。方法:收集我院口腔科收治的上颌前牙残根残冠修复患者40例,随机分为玻璃纤维桩组和金属桩组,每组各20例,玻璃纤维桩组患者给予玻璃纤维桩进行残根残冠修复,金属桩组给予金属桩进行残根残冠修复,治疗结束后1年随访,对所有患者的牙龈指数(GI)、修复齿出血指数(SBI)、不良反应发生率、患者修复效果以及患者疗效进行检测并比较。结果:与治疗前相比,两组患者的牙龈指数(GI)以及修复齿出血指数(SBI)均下降(P0.05);与金属桩组相比,玻璃纤维桩组患者的牙龈指数(GI)以及修复齿出血指数(SBI)较低(P0.05),不良反应发生率较低(P0.05),颜色匹配率以及边缘适合率较高(P0.05),两组患者修复体完整例数百分率比较无明显差异(P0.05),玻璃纤维桩组患者的治疗成功率较高(P0.05)。结论:玻璃纤维桩在老年上颌前牙残根残冠修复的应用与金属桩组相比临床效果较好。  相似文献

Background: The reduction of forest areas and the fragmentation of remaining forest is a major threat to biodiversity and is one of the primary causes of species extinction in the Atlantic forest biome.

Aims: To evaluate the effects of the fragmentation and loss of habitat (size, edge effect, isolation and type of matrix) the richness, abundance, diversity and species composition of ferns in Atlantic forest fragments.

Methods: For each of 11 fragments, plots of 10 × 20 m2 were compared in the interior and at the forest edge. Data were analysed by general linear models, or by the use of similarity indices for floristic composition.

Results: Eighty-eight species were recorded. Fragment size and type of environment (interior vs. edge) were related to the biological variables studied: large fragments and forest interior environments had larger populations of ferns, with a higher diversity. Floristic compositions were distinct for both the interior vs. edge environments and large vs. small fragments.

Conclusions: Although larger fragments present a higher number of species, the preservation of smaller fragments, which have a different suite of species from those found in the large fragments, is important to guarantee the maintenance of the highest number of fern species in the Atlantic forest. Small fragments are not merely subsets of the species occurring in the larger fragments. The turnover in the species composition between fragments of different size suggests high spatial variability in the study area.  相似文献
