Kinetic studies on ammonia and methane oxidation by Nitrosococcus oceanus   总被引:2,自引:0,他引:2  
The kinetics of ammonia oxidation and the ability of a marine ammonia-oxidizing bacterium, Nitrosococcus oceanus, to metabolize methane were investigated in semicontinuous batch culture. The effects of inhibitors (acetylene and nitrapyrin) and coreactants were determined in order to elucidate the behavior of the ammonia oxygenase enzyme in N. oceanus. Acetylene and nitrapyrin were potent inhibitors and their effects were not mitigated by increased ammonia concentrations. Oxygen concentration had the effect of a mixed-type inhibitor; reduced oxygen inhibited the rate or ammonia oxidation at high substrate concentration but may enhance the rate at low substrate concentrations. Substrate affinity in terms of NH 4 + increased (K m decreased) with increasing pH. Optimal pH was about 8. Methane inhibited ammonia oxidation; the interaction was not simple competitive inhibition and the presence of multiple active sites on the enzyme was indicated by the behaviour of the inhibited treatments. Half-saturation constants for methane (K i=6.6 M) and ammonia (K m=8.1 M) were similar. N. oceanus oxidized methanol and methane linearly over time, with CO2 and cell material being produced at approximately equal rates.  相似文献

The growth of six rapid-cycling lines of Brassica species, B. napus, B. campestris, B. nigra, B. juncea, B. oleracea and B. carinata was inhibited by seawater salinity. Based on the change in dry matter reduction relative to the control at varying concentrations of salts (4, 8 and 12 dS m-1), the relative salt tolerance of these species was evaluated. B. napus and B. carinata were the most tolerant and most sensitive species, respectively, while the other four species were moderately tolerant. The influence of seawater on the concentrations of 12 elements including macronutrients and micronutrients in the shoots of these Brassica plants was characterized to determine the relationship between nutritional disturbance and relative salt tolerance. It was found that seawater salinity had a significant effect on the concentrations of Ca, Mg, K, Cl, Na and total N in the shoots of these plants but only the change in Ca concentration was significantly related to the relative salt tolerance of these six rapid-cycling Brassica species according to a rank analysis of the data. This finding indicates that Ca may play a regulatory role in the responses of Brassica species to saline conditions.  相似文献

This study focuses on the inhibitory effect of salinity on the leaf extension of three different grass species: Hordeum jubatum L., Hordeum vulgare L. and Zea mays L. Leaf elongation rates (LER) were measured on the third leaf of the plants. NaCl was added to the hydroponic solution (0, 40, 80 and 120 mM) and changes in LER were measured over time with a displacement transducer. Salinity inhibited LER immediately in all three species, and a new, but lower steady-state LER was reached within 5 h. The decrease in LER was proportional to the salinity level. Differences in salt tolerance (% of control LER) were evident between genotypes within 5 h after salinization, but the relative salt tolerance of the plant at this stage was not necessarily indicative of the long-term salt tolerance of the species. In general, H. jubatum was more tolerant than maize, which was more tolerant than barley to these short-term salinity stresses. In contrast, barley is more salt tolerant than maize over the long term. The mechanisms of inhibition of LER by salinity, as tested by the applied-tension technique, varied with the species examined, affecting either the apparent yield threshold, the hydraulic conductance of the whole plant or both. The cell wall extensibility was not significantly affected by salinity in the three species tested in this study.  相似文献

During a survey fungal diversity of xylariaceous fungi in Thailand, a new Nemania species, N. plumbea, was identified. Nemania plumbea is characterized by soft-textured grey stromata on a persistent mat of white hyphae, pale brown ascospores with a short germ-slit on the more convex side. It also produces a Geniculosporium-like anamorph in culture. In order to evaluate its phylogenetic relationships among related species and genera, ITS-5.8S rDNA and RPB2 were analysed separately and simultaneously. Results from the phylogenetic analyses indicate that there is close phylogenetic association between N. plumbea and N. aenea. A preliminary account into the natural grouping of Xylariaceae based on ITS-5.8S rDNA and RPB2 sequences is also discussed.  相似文献

Pitfall traps were exposed between February 1993 andSeptember 1994 every 4th to 6th week near the highwater mark in a Ria Formosa salt marsh (Portugal).With only a few exceptions, peracarid crustaceansaccounted for more than 95% of the individualscaught. Three amphipod species (Orchestiagammarellus(Pallas), Orchestia mediterraneaCosta, Talorchestia deshayesii(Audouin)) and3 isopod species (Tylos ponticus(Arcangeli),Halophiloscia couchi(Kinahan) and Porcellio lamellatus(Uljanin) Budde-Lund) wereidentified. In general, these species were most activefrom spring to autumn.Data collected included information on growth rate,life expectancy and timing of reproduction. Theamphipods displayed more opportunistic life-historypatterns with high growth rates, reproductive activityduring most of the year, early sexual maturity andrelatively short life expectancies. The isopods weremore persistent with slower growth rates, morerestricted reproductive periods and longer lifeexpectancies.There is no indication in the literature that most ofthese species are particularly common in salt marshes.In addition, peracarid crustaceans are not mentionedas dominant taxa in most studies of salt marsh fauna.We suggest that the peracarids play an important rolein the degradation of organic matter in salt marshesand hypothesize that the high numbers of peracaridsfound in Ria Formosa is due to the high contributionof leaf shedding plants.  相似文献

Nitraria retusa and Atriplex halimus (xero-halophytes) plants were grown in the range 0–800 mM NaCl while Medicago arborea (glycophyte) in 0–300 mM NaCl. Salt stress caused a marked decrease in osmotic potential and a significant accumulation of Na+ and Cl in leaves of both species. Moderate salinity had a stimulating effect on growth rate, net CO2 assimilation, transpiration and stomatal conductance for the xero-halophytic species. At higher salinities, these physiological parameters decreased significantly, and their percentages of reduction were higher in A. halimus than in N. retusa whereas, in M. arborea they decreased linearly with salinity. Nitraria retusa PSII photochemistry and carotenoid content were unaffected by salinity, but a reduction in chlorophyll content was observed at 800 mM NaCl. Similar results were found in A. halimus, but with a decrease in the efficiency of PSII (F′v/F′m) occurred at 800 mM. Conversely, in M. arborea plants we observed a significant reduction in pigment concentrations and chlorophyll fluorescence parameters. The marked toxic effect of Na+ and/or Cl observed in M. arborea indicates that salt damage effect could be attributed to ions’ toxicity, and that the reduction in photosynthesis is most probably due to damages in the photosynthetic apparatus rather than factors affecting stomatal closure. For the two halophyte species, it appears that there is occurrence of co-limitation of photosynthesis by stomatal and non-stomatal factors. Our results suggest that both N. retusa and A. halimus show high tolerance to both high salinity and photoinhibition while M. arborea was considered as a slightly salt tolerant species.  相似文献

Contrary to the opinion ofBateman and most subsequent authors, we believe that the available evidence indicates that some populations ofN. triandrus andN. fernandesii, at least, are tristylous; other populations ofN. triandrus are distylous.Hugonia cf.penicillanthemum (Linaceae) from new Caledonia is distylous, but it remains possible that other species ofHugonia are tristylous. The disputed occurrence of heterostyly in S. African species ofBauhinia (Leguminosae),Cleome (Capparaceae) andAneilema (Commelinaceae), and inAgelaea (Connaraceae) is discussed.  相似文献

High levels of naturally occurring selenium (Se) are often found in conjunction with different forms of salinity in central California. Plants considered for use in phytoremediation of high Se levels must therefore be salt tolerant. Selenium accumulation was evaluated for the following species under increasing salt (NaCl and CaCl) conditions:Brassica napus L. (canola),Hibiscus cannibinus L. (kenaf),Festuca arundinacea L. (tall fescue), andLotus tenuis L. (birdsfoot trefoil). The experimental design was a complete randomized block with four salt treatments of <1, 5, 10, and 20 dS m-1, four plant species, three blocks, and six replicates per treatment. Ninety days after growing in the respective salt treated soil with a Se concentration of 2 mg Se kg-1 soil, added as Na2SeO4, all plant species were completely harvested. Among the species tested, shoot and root dry matter yield of kenaf was most significantly (p<0.001) affected by the highest salt treatment and tall fescue and canola were the least affected species. Generally there was a decrease in tissue accumulation of Se with increasing salt levels, except that low levels of salinity stimulated Se accumulation in canola. Canola leaf and root tissue accumulated the highest concentrations of Se (315 and 80 mg Se kg-1 DM) and tall fescue the least (35 and 7 mg Se kg-1 DM). Total soil Se concentrations all harvest were significantly (p<0.05) lower for all species at all salt treatments. Removal of Se from soil was greatest by canola followed by birdsfoot trefoil, kenaf and tall fescue. Among the four species, canola was the best candidate for removing Se under the tested salinity conditions. Kenaf may be effective because of its large biomass production, while tall fescue and birdsfoot trefoil may be effective because they can be repeatedly clipped as perennial crops.  相似文献

A method is evaluated that employs variation in stable C and N isotopes from fractionations in C and N acquisition and growth to predict root biomasses of three plant species in mixtures. Celtis laevigata Willd. (C3), Prosopis glandulosa Torr. (C3, legume) and Schizachyrium scoparium (Michx.) Nash (C4), or Gossypium hirsutum L. (C3), Glycine max (L.) Merr. (C3 legume), and Sorghum bicolor (L.) Moench (C4) were grown together in separate, three-species combinations. Surface roots (0–10 cm depth) of each species from each of the two combinations were mixed in various proportions, and the relative abundances of 15N and 14N and 13C and 12C in prepared mixtures, surface roots of single species, and roots extracted from the 80-cm soil profile in which each species combination was grown were analyzed by mass spectrometry. An algebraic determination which employed the δ 13C, % 15N, and C and N concentrations of root subsamples of individual species accounted for more than 95% of the variance in biomass of each species in prepared mixtures with G. max, G. hirsutum, and S. bicolor. A similar analysis demonstrated species-specific differences in rooting patterns. Root biomasses of the C4 monocots in each combination, S. scoparium and S. bicolor, were concentrated in the upper 20 cm of soil, while those of G. hirsutum and the woody P. glandulosa were largest in lower soil strata. Analyses of stable C and N isotopes can effectively be used to distinguish roots of species which differ in ratios of 15N to 14N and 13C to 12C and thus to study belowground competition between or rooting patterns of associated species with different C and N isotope signatures. The method evaluated can be extended to quantify aboveground and belowground biomasses of component species in mixtures with isotopes of other elements or element concentrations that differ consistently among plants of interest.  相似文献

Among the isolates from soil of corn and sugar cane plantations in São Paulo State, Brazil, two new species ofNeosartorya, N. botucatensis andN. paulistensis, are described and illustrated.Neosartorya botucatensis differs from the other known species of the genus in having ascospores with long spines on the convex walls.Neosartorya paulistensis is characterized by its ascospore walls with spinose and verruculose ornamentation. Six non-native invasive fish species from the Slovak part of the Tisa River basin, namely Carassius gibelio, Pseudorasbora parva, Ameiurus melas, A. nebulosus, Lepomis gibbosus, Perccottus glenii, and three gobiid species from the Danube River, namely Neogobius melanostomus, N. fluviatilis and N. kessleri, were investigated for endohelminth parasites. The expanding Asian cestode Nippotaenia mogurndae (syn. Amurotaenia perccotti) (Nippotaeniidea) has been introduced to Europe with its invasive host P. glenii. Pumpkinseed, Lepomis gibbosus, is a new definitive host of Proteocephalus percae and it is reported as the second intermediate host of the bothriocephalidean cestode Triaenophorus nodulosus in Slovakia.  相似文献

Fourteen species of Characeae were analyzed for their free amino acids. This study, in combination with previous results (Sakanoet al., 1984) showed that all the species that belong to the generaChara (6 species),Lamprothamnium (1 species),Nitellopsis (1 species),Lychnothammus (1 species) and 5Nitella species of the members of the subsectionAnarthrodactylae contained a large amount of isoasparagine. In contrast, no isoasparagine was found in the species belonging toTolypella (3 species) and otherNitella (7 species). Presence of isoasparagine in some species of Characeae (N. flexillis andNitellopsis obtusa) was found to be independent of their localities (Japan, Canada and England). Species lacking isoasparagine (N. oligogyra and N. axilliformis) did not produce isoasparagine even under the condition that induced a great increase of this amino acid in the species that contained it (C. corallina andN. flexilis). These results indicate that isoasparagine is a distinct taxonomic marker and suggest that theNitella species of the subsectionAnarthrodactylae are the most primitive group inNitelleae in that they share synthesis and accumulation of isoasparagine withChareae and, hence, support the view (Kasaki, 1964) that the subsection may be treated as an independent genus.  相似文献

Field plots were used to assess the restoration potential of three salt marsh species, Juncus maritimus, Leptocarpus similis and Schoenoplectus pungens, within an established salt marsh near Christchurch, New Zealand. A split-block design was used to asses the effects of soil type, (soil from a proposed marsh construction site or estuarine mud from a natural salt marsh), species and plant source (commercial nursery stocks or natural marsh stock). In December, after 9 months, there was no mortality in L. similis and a significant increase in the biomass compared with J. maritimus. S. pungens failed to regenerate following seasonal die-back. Plant biomass was unaffected by soil type, despite occasional higher salinities recorded in the reclamation soil. Natural stocks had significantly higher biomass than nursery stocks (p < 0.01). These results suggest that both L. similis and J. maritimus are appropriate species for transplanting in the Canterbury region and that the soil from the proposed area is suitable for the restoration and construction of tidal wetlands. The success of the transplants could be enhanced by sourcing hardy stock and management regimes may be necessary to reduce salinity extremes and herbivory by rabbits.  相似文献

A pot experiment was conducted in a greenhouse using the 15N isotope dilution method and two reference plants, Parkia biglobosa and Tamarindus indica to estimate nitrogen fixed in four Acacia species: A raddiana, A. senegal, A. seyal and Faidherbia albida (synonym Acacia albida). For the reference plants, the 15N enrichments in leaves, stems and roots were similar. With the fixing plants, leaves and stems had similar 15N enrichments; they were higher than the 15N enrichment of roots. The amounts of nitrogen fixed at 5 months after planting were similar using either reference plant. Estimates of the percentage of N derived from fixation (%Ndfa) for the above ground parts, in contrast to %Ndfa in roots, were similar to those for the whole plant. However, none of the individual plant parts estimated accurately total N fixed in the whole plant, and excluding the roots resulted in at least 30% underestimation of the amounts of N fixed. Between species, differences in N2 fixation were observed, both for %Ndfa and total N fixed. For %Ndfa, the best were A. seyal (average, 63%) and A. raddiana (average, 62%), being at least twice the %Ndfa in A. senegal and F. albida. Because of its very high N content, A. seyal was clearly the best in total N fixed, fixing 1.62 g N plant–1 compared to an average of 0.48 g N plant–1 for the other Acacia species. Our results show the wide variability existing between Acacia species in terms of both %Ndfa and total N fixed: A. seyal was classified as having a high N2 fixing potential (NFP) while the other Acacia species had a low NFP.  相似文献

Three new species of Neosartorya and one new Aspergillus of section Fumigati are proposed using a polyphasic approach based on morphology, extrolite production and partial β-tubulin, calmodulin, and actin gene sequences. The phylogenetic analyses using the three genes clearly show that the taxa grouped separately from the known species and confirmed the phenotypic differences. Neosartorya denticulata is characterized by its unique denticulate ascospores with a prominent equatorial furrow; N. assulata by well developed flaps on the convex surface of the ascospores which in addition have two distinct equatorial crests and N. galapagensis by a funiculose colony morphology, short and narrow conidiophores and ascospores with two wide equatorial crests with a microtuberculate convex surface. Aspergillus turcosus can be distinguished by velvety, gray turquoise colonies and short, loosely columnar conidial heads. The four new taxa also have unique extrolite profiles, which contain the mycotoxins gliotoxin and viriditoxin in N. denticulate; apolar compounds provisionally named NEPS in N. assulata and gregatins in N. galapagensis. A. turcosus produced kotanins. N. denticulata sp. nov., N. assulata sp. nov., N. galapagensis sp. nov., and A. turcosus sp. nov. are described and illustrated. Plant invasions of coastal wetlands are rapidly changing the structure and function of these systems globally. Alteration of litter dynamics represents one of the fundamental impacts of an invasive plant on salt marsh ecosystems. Tamarisk species (Tamarix spp.), which extensively invade terrestrial and riparian habitats, have been demonstrated to enter food webs in these ecosystems. However, the trophic impacts of the relatively new invasion of tamarisk into marine ecosystem have not been assessed. We evaluated the trophic consequences of invasion by tamarisk for detrital food chains in the Tijuana River National Estuarine Research Reserve salt marsh using litter dynamics techniques and stable isotope enrichment experiments. The observations of a short residence time for tamarisk combined with relatively low C:N values indicate that tamarisk is a relatively available and labile food source. With an isotopic (15N) enrichment of tamarisk, we demonstrated that numerous macroinvertebrate taxonomic and trophic groups, both within and on the sediment, utilized 15N derived from labeled tamarisk detritus. Infaunal invertebrate species that took up no or limited 15N from labeled tamarisk (A. californica, enchytraeid oligochaetes, coleoptera larvae) occurred in lower abundance in the tamarisk-invaded environment. In contrast, species that utilized significant 15N from the labeled tamarisk, such as psychodid insects, an exotic amphipod, and an oniscid isopod, either did not change or occurred in higher abundance. Our research supports the hypothesis that invasive species can alter the trophic structure of an environment through addition of detritus and can also potentially impact higher trophic levels by shifting dominance within the invertebrate community to species not widely consumed. Synopsis The spatial distribution, seasonal abundance and diel activity of four sticklebacks,Gasterosteus aculeatus, G. wheatlandi, Pungitius pungitius, andApeltes quadracus. coexisting in a St. Lawrence salt marsh were examined to see how these closely related species share their habitat. While all four species breed in the Riviè des Vases, a tidal creek, only three species are found in the adjacent salt marsh pools,A. quadracus being absent. Results are interpreted in terms of avoidance of interspecific competition for space during the relatively short breeding season at this latitude.  相似文献
