The present study investigates the possible circadian dependence of leptin effects on food intake, locomotor activity, glycemia and plasma cortisol levels in goldfish (Carassius auratus). Fish were maintained under 12L:12D photoperiod and subjected to two different feeding schedules, one group fed during photophase (10:00) and the other one during scotophase (22:00). Leptin or saline were intraperitoneally injected at two different times (10:00 or 22:00), coincident or not with the meal time. To eliminate the entraining effect of the light/dark cycle, goldfish maintained under 24 h light (LL) were fed and leptin-injected at 10:00. A reduction in food intake and locomotor activity and an increase in glycemia were found in goldfish fed and leptin-injected at 10:00. No significant changes in circulating cortisol were observed. Those effects were not observed when leptin was administered during the scotophase, regardless the feeding schedule; neither in fish maintained under LL, suggesting that a day/night cycle would be necessary to observe the actions of leptin administered during the photophase. Changes in locomotor activity and glycemia were only observed in goldfish when leptin was injected at daytime, coincident with the feeding schedule, suggesting that these leptin actions could be dependent on the feeding time as zeitgeber. In view of these results it appears that the circadian dependence of leptin actions in goldfish can be determined by the combination of both zeitgebers, light/dark cycle and food. Our results point out the relevance of the administration time when investigating regulatory functions of hormones.  相似文献

Three groups of rainbow trout, Oncorhynchus mykiss , (initial weight 40 g) were fed one of three rations (low, medium or high) for 73 days. Consumption by individual fish within the three ration groups was measured on four occasions (days 27, 55, 64 and 72) using radiography. Food intake by individual fish varied between days and this variability was expressed using the coefficient of variation (CV). Dominant fish within each ration group were defined as individuals with the greater share of the group meal and these fish had low CVs for food intake indicating relatively little variation in daily consumption. By contrast, the fish which had consumed a low mean proportion of the group meal displayed high CVs for food intake, indicating considerable variability in the sizes of individual meals consumed by these fish. As group ration increased, the range of meal sizes and individual CVs in daily feeding decreased, suggesting that the strength of the feeding hierarchy and the variability in individual consumption decreased as food availability increased. It is suggested that radiography can be used to assess social relationships within groups of fish allowing the assessment of feeding hierarchies in larger groups of fish than would be possible by observational techniques.  相似文献

This study evaluated how water temperature (26, 28, and 30°C), number of meals per day (one or two meals), and protein percent in diet (20, 25 and 30%) impact growth performance, biometric indices, and feeding behavior of Nile tilapia, Oreochromis niloticus. Fish were randomly allocated into 18 equal replicate groups. Higher final body weight was observed in fish reared at 30°C and fed one meal per day containing 30% crude protein. Better weight gain, weight gain %, feed conversion ratio, specific growth rate, and condition factor were recorded in fish reared at 26°C and fed one meal per day containing 30% protein. The best length weight relationship was obtained in fish reared at 26°C and fed one meal per day containing 30% crude protein. Shorter feeding duration and duration of appetite inhibition latency were recorded in fish reared at 30°C, fed one meal per day, and given a diet containing 30% protein. The highest proactivity was recorded in fish reared at 30°C, received one meal per day, and with 25% crude protein in their diet. Conclusively, rearing Nile tilapia at 26–30°C with a lower feeding frequency (one meal/day) and a 30% crude protein diet achieved better performance and feeding behavior.  相似文献

The present study was carried out to investigate the number of daily feeding sessions that results in maximum growth of juvenile cobia under laboratory conditions. Groups of eight fish (110 g) were randomly distributed in twenty 500‐L tanks and hand‐fed a commercial diet for 60 days. The same amount of feed was offered daily, divided in 1, 2, 3, four or six meals. None of the parameters associated with growth performance (survival, weight gain, specific growth rate, feed intake, condition factor or size variation) showed any significant differences among treatments. Although under the present conditions feeding frequency had no effect on the growth performance of cobia larger than 110 g, in commercial farming operations where large numbers of fish are kept within a single rearing structure, fish may have aggressive interactions during feeding. Under these conditions, it is difficult to ensure that all cobia are fed to satiation and thus it is usual to provide two or more meals per day. The present results indicate that for an individual cobia the provision of more than one daily meal has no significant effect on growth performance.  相似文献

A 30‐day growth trial was conducted to investigate the individual variations in feeding rate, growth rate, spontaneous activity, and their possible interrelationships in hybrid tilapia (Oreochromis niloticus × O. aureus) reared individually at feeding frequencies of one and two meals daily. Results show that the feeding rate in energy (FRe), specific growth rate in energy (SGRe), and food conversion efficiency (FCE) of fish fed twice a day were significantly higher than that of fish fed only once. However, no significant differences in distance or in time spent swimming were observed between the groups. SGRe was positively correlated with FRe in fish fed only once daily, and SGRe was positively correlated with food conversion efficiency in energy (FCEe) between the two groups. SGRe was only inversely correlated with the distance that fish swam when fed once daily. The results suggest that the individual growth differences of hybrid tilapia could be attributed mainly to variations in FRe, and that the energy costs of spontaneous activity play an important role in the differences at feeding frequency of one meal a day. However, at feeding frequencies of two meals a day, individual growth differences in fish may be caused by variations in FCEe, and energy costs of spontaneous activity have only marginal effects on the differences.  相似文献

After a pre-prandial period of starvation or feeding with unlabelled food, 0+ salmon parr (0.8–11.7 g) were fed a test meal of iron particle labelled food and subsequently were again either starved or fed unlabelled food. The quantity of labelled food consumed and the evacuation rate was determined by serial radiographs. In fish of all sizes, pre-prandial starvation causes a larger test meal (as a percentage of body weight) to be consumed when compared to pre-prandially fed fish. In addition, pre-prandial starvation results in relatively larger meals as a percentage of body weight being taken by smaller compared to larger fish. This result was not evident for pre-prandially fed fish. Evacuation rate was unrelated to body size irrespective of feeding history. Post-prandial starvation decreased evacuation rate but this effect was inversely related to the quantity of food consumed. Larger meals were not evacuated differently from smaller meals if feeding occurred post-prandially, irrespective of pre-prandial starvation.  相似文献

Periodic food availability can act as a potent zeitgeber capable of synchronizing many biological rhythms in fishes, including locomotor activity rhythms. In the present paper we investigated entrainment of locomotor rhythms to scheduled feeding under different light and feeding regimes. In experiment 1, fish were exposed to a 12:12?h light/dark cycle and fed one single daily meal in the middle of the light phase. In experiment 2, we tested the effect of random versus scheduled feeding on the daily distribution of activity. During random feeding, meals were randomly scheduled with intervals ranging from 12 to 36?h, while scheduled feeding consisted of one single daily meal set in the middle of the light or dark phase. Finally, in experiment 3, we studied the synchronization of activity rhythms to feeding under constant darkness (DD) and after shifting the feeding cycle by either advancing or delaying the feeding cycle by 9?h. The results revealed that goldfish synchronized to feeding, overcame light entrainment and significantly changed their daily distribution of activity according to their feeding schedule. In addition, the daily activity pattern modulated by feeding differed between layers: a peak of activity being noticeable directly after feeding at the bottom, while an anticipatory behaviour was obvious at the surface of the tank. Under DD and no food, free-running rhythms averaging 25.5?± 1.9?h (mean?±?SD) were detected. In conclusion, some properties of feeding entrainment (e.g. anticipation of the feeding time, free-running rhythms following termination of periodic feeding, and the stability of ø after shifting the feeding cycle) suggested that goldfish have (a) separate but tightly coupled light- and food-entrainable oscillators, or (b) a single oscillator that is entrainable by both light and food (one synchronizer being eventually stronger than the other).  相似文献

Keeping and management of horses can induce changes to instinctive and innate behavioural patterns. We investigated the effect of five different management conditions in five groups of horses. All groups were housed in individual boxes under natural environmental and lighting conditions. They were fed three times a day (07:00, 12:30 and 20:00) and had free access to water. Group A was fed with 8 kg/capo/die of hay divided in the three meals. Group B was fed with 8 kg/capo/die of an unifeed divided in the three meals. Group C was fed with unifeed at 07:00 and 12:30 and with hay at 20:00. They were kept in wood-bedded boxes. Groups D and E were fed with unifeed at 07:00 and 12:30, respectively, and in the other, two meals received hay. They were kept in straw-bedded boxes. Our results showed a daily rhythm of total locomotor activity in all groups, influenced by management conditions. Group A engaged in meal patterns similar to those seen in grazing animals. Groups B and C showed the highest MESOR values due to a high searching behaviour. Group C showed a nocturnal acrophase contrary to the other groups. Groups D and E showed a total locomotor activity pattern similar to that observed in Group A probably due to an increase in straw-bedding consuming. The reduction of fibre in diet has an impact on physiology and behaviour of horses. The valuation of diet and in bedding provided to horses kept in box is useful to guarantee the maintenance of the physiological daily rhythm of total locomotor activity.  相似文献

The present study was conducted to investigate whether individual rainbow trout Oncorhynchus mykiss can simultaneously anticipate two daily meals, and to evaluate the influence of time-restricted feeding schedules and light-dark (L:D) cycles on the timing of self-feeding activity. Individual rainbow trout were capable of anticipating not only a single meal but also two daily meals scheduled at different times; the duration of anticipation was short and precise. This seems to be the first convincing evidence to show that a single fish can simultaneously anticipate two daily meals. Change in the onset and cessation of light did not affect the duration of food-anticipatory activity, although it led to shift the phases of the light-related self-feeding activity. Moreover, under constant light and ad lib feeding conditions, only one band of rhythmic activity was observed in individual rainbow trout that had previously developed food-anticipatory activity to each of the two daily meals.  相似文献

Diurnal rhythm in body temperature of pigeons subjected to different experimental conditions (pinealectomized, sham-operated, melatonin-implanted, cold-exposed) was studied under a 12 h light and 12 h dark regimen. The body temperature of pigeons during photophase was higher than that during scotophase in the normal as well as every treatment group studied. Pinealectomized pigeons showed higher body temperature in the photophase as well as scotophase, than that of the normal and sham-operated birds when examined 2 or 3 weeks following the post-surgical acclimatization to 25 or 3 degrees C. However, subcutaneous implantation of melatonin pellets into pinealectomized pigeons nullified or even reversed the hyperthermic effect of pinealectomy. Exposure of pigeons to--18 degrees C for 280 min during photophase as well as scotophase, produced a marked drop in body temperature in pinealectomized, sham-operated and normal pigeons. The pinealectomized pigeons exhibited a higher body temperature than that of the sham-operated and normal ones when exposed to--18 degrees C during the photophase, but not during the scotophase. It was concluded that while the pineal is not necessary for maintaining the daily thermal rhythm in the avian body, it has a thermoregulatory role, in that it prevents rise in body temperature in warm (25 degrees C) acclimatized and chronic cold (3 degrees C) exposed birds. In acute short-term cold (--18 degrees C) exposure however, the temperature regulatory role of the pineal was not effective during the scotophase.  相似文献

Abstract. The adipokinetic hormone (Grb-AKH) content in the corpora car-diaca of the house cricket, Acheta domesticus , varied during a day with two peaks in the scotophase and one peak in the photophase. There were two distinct peaks of total lipid concentration in the blood, one early in the photophase and the other early in the scotophase. Fat body sensitivity to adipokinetic hormone also varied in close synchrony with the lipid rhythm. It was not possible to attribute the rhythm of blood lipid titre unequivocally to either the rhythm of adipokinetic hormone content in the corpora cardiaca or to the rhythm of sensitivity of the fat body to the hormone. In adult crickets the blood carbohydrate titre had two peaks in adult females, one towards the end of scotophase and another in the late photophase (c. 12h apart), but a single peak at the end of scotophase was apparent in last instar larvae. The blood carbohydrate rhythm persists in DD and is therefore endogenous. Carbohydrates were not mobilized by the Grb-AKH, therefore could not be involved in the blood carbohydrate rhythm. Exposure to various day lengths caused shifts in the patterns of the carbohydrate rhythms, but imposition of a cyclic temperature regime had no effect on the rhythm.  相似文献

This study investigated the effects of the photophase light intensity on the scotophase melatonin response. Twelve, 8-month-old crossbred gilts were allocated to three groups of four and housed in temperature- and lighting-controlled climate rooms. The rooms had a light intensity of 40, 200 or 10,000 lx and a light-dark cycle of 12 L:12 D. The gilts were allowed to acclimatize to a new lighting regimen for 1 week before being sampled at 2h intervals for 24h. Following the sampling, pigs were transferred under a different light intensity, allowed to adjust for 1 week and sampled again. The procedure was repeated three times so that all the groups went through all three lighting regimens (light intensities). All the gilts exhibited a clear circadian serum melatonin rhythm under each lighting regimen with high melatonin concentrations occurring during the scotophase. There was no difference in the scotophase melatonin response in terms of mean concentrations or duration of increased melatonin levels within or between the groups under different lighting regimens. There was considerable inter-individual variation in the dark phase melatonin response but the individual profiles were consistent under the different lighting regimens. It is concluded that when a certain threshold light intensity (<40lx) is exceeded, the photophase light intensity has no effect on the scotophase melatonin response. These results imply that extremely high light intensities during the photophase would provide no additional benefits compared with normal comfortable light intensity, if artificial lighting programs were introduced to commercial piggeries in order to reduce seasonal effects on reproduction.  相似文献

The timing of meals has been suggested to play an important role in circadian regulation and metabolic health. Three meals a day is a well-established human feeding habit, which in today's lifestyle may or may not be followed. The aim of this study was to test whether the absence of breakfast or supper significantly affects the circadian system and physiological function. The authors developed a rat model for their daily three meals study, whereby animals were divided into three groups (three meals, TM; no first meal, NF; no last meal, NL) all fed with the same amount of food every day. Rats in the NF group displayed significantly decreased levels of plasma triglyceride (TG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), and glucose in the activity phase, accompanied by delayed circadian phases of hepatic peripheral clock and downstream metabolic genes. Rats in the NL group showed lower concentration of plasma TC, HDL-C, and glucose in the rest phase, plus reduced adipose tissue accumulation and body weight gain. Real-time polymerase chain reaction (PCR) analysis indicated an attenuated rhythm in the food-entraining pathway, including down-regulated expression of the clock genes Per2, Bmal1, and Rev-erbα, which may further contribute to the delayed and decreased expression of FAS in lipogenesis in this group. Our findings are consistent with the conclusion that the daily first meal determines the circadian phasing of peripheral clocks, such as in the liver, whereas the daily last meal tightly couples to lipid metabolism and adipose tissue accumulation, which suggests differential physiological effects and function of the respective meal timings.  相似文献

SUMMARY. 1. The dry weight of food remaining in the stomachs of piscivorous trout decreased exponentially with time. Gastric evacuation rates increased exponentially with increasing temperature but were unaffected by predator size, meal size or type of fish prey.
2. Mathematical models were developed to estimate both the rate and time for the gastric evacuation of different meal sizes (expressed as dry weight), and were applicable to piscivorous trout of different sizes (length range 10–32 cm) feeding on trout fry or sticklebacks at different temperatures (range 5–18°C).
3. The wet weight of food in the stomachs also decreased exponentially with time, but evacuation rates both increased with temperature and decreased with increasing meal size; the latter relationship occurred because relative rates of water loss from a meal also decreased with increasing meal size. Use of wet or dry weights can therefore lead to different conclusions about the effect of meal size on evacuation rates.
4. When piscivorous trout were fed three consecutive meals of varying size, the models predicted the total dry weight of food left in the stomach, but not the weight remaining for each individual meal. Interactions between meals led to an increase in evacuation rates for meals consumed early in the series and a decrease in evacuation rates for later meals.
5. Evacuation rates for piscivorous trout were compared with those for trout feeding on invertebrates in an earlier study, and were close to those for caddis larvae as prey, higher than those for mealworms and lower than those for a variety of invertebrate prey. Although a great deal is now known about the daily food intake and growth rates of trout feeding on invertebrates, there is little comparable information for piscivorous trout.  相似文献

Abstract.  The Indian meal moth Plodia interpunctella Hubner (Lepidoptera: Pyralidae) measures night length and enters diapause as a last-instar larva. To examine the role of photophase on dark-time measurement, the main LD 7 : 17 h photoperiod is disrupted by various lengths of darkness at 25 °C. When the light phase is not disrupted, the incidence of diapause is 76%. As the dark pulse disrupting a 7-h photophase becomes longer, the incidence of diapause decreases. To detect the dynamic kinetics of the time-measuring process, the main scotophase of 17 h is scanned by a 2-h light pulse. When the dark pulse in a 7-h photophase is fixed at 1 h after dawn and its duration is varied systematically from 1 to 3 h, or when the end of the dark pulse is fixed at 1 h before dusk, diapause is prevented completely by a 2-h light pulse inserted in the middle of 17-h darkness. These results are compared with those of a single night interruption of a 17-h scotophase with a 2-h light pulse but with an intact 7-h photophase. The disruption of a 7-h photophase by a dark pulse shifts the descending and ascending slopes of the response curve to some extent toward dawn and dusk, respectively, indicating that the dark pulse tends to shorten the critical length of dark time for diapause induction. When the main photophase (7 h) is interrupted by a 1-h dark pulse at 3–4 h after dawn, the 2-h scanning light pulse in the main scotophase (17 h) appears to act effectively as a dusk signal in the early scotophase. However, those in the mid- and late scotophase do not define the critical night length from dusk as sharply as for the critical night length from a 2-h light pulse to dawn. The results indicate the importance of photophase in the dark-time measurement.  相似文献

When exposed to a human host, Anopheles gambiae started probing 4 h post-eclosion, but 95% successfully blood-fed by 16-20 h with maximal blood volumes of 5- 10 microl per female. When fed sugar, the 95% feeding was not observed until 36-40 h post-eclosion; sugar meals appeared to interfere with blood meals. Similarly in An. atroparvus, maximum volumes were 10 microl when starved but only 6 microl when fed sugar. This species did not bite before 2 d, and 95% biting was by 4 d. Given single blood meals to water-kept An. gambiae, a threshold body size for oogenesis was detected. With wing lengths below 2.8 mm, eggs never matured, but when sugar-fed, females of all sizes matured eggs including the synthesis of maternal deposits. Although sugar feeding interfered with blood feeding, more lipid was transferred to the yolk. In water-kept An. atroparvus only 5% of the females produced eggs. When sugar-fed for 4 d, all females matured eggs, so in this species sugar feeding appeared to be essential for oogenesis. An. gambiae always took multiple blood meals, tested at any time after the first ones, leading to 120 mature eggs/female. Yolk composition was 3.9 mcal protein and 3.8 mcal lipid/oocyte when kept on water, but 2.8 meal protein and 4.3 mcal lipid/oocyte with intermittent sugar meals, thus marking a surprising flexibility in synthesis of yolk protein and lipid that strongly depends on additional carbohydrates sources. Only 80% of water-fed An. atroparvus re-fed 2 d after a first blood meal with small females taking three blood meals but they still showed reduced fecundity. Only the large water-fed females matured eggs, with blood volumes higher than 9-12 microl. When fed sugar, the blood meal input was reduced, but oogenesis was possible, whereas water-fed females required three blood meals to reach the caloric level comparable to pre-feeding sugar-fed females. Water-fedAn. gambiae could survive on daily blood meals alone, but survival was further extended by intermittent sugar meals. When offered a blood donor daily, there was a behavioral difference. Females maintained alone showed a more or less regular 3 d feeding and oviposition activity, while females kept in groups fed daily followed a daily oviposition pattern, suggesting gonotrophic discordance.  相似文献

The diapause-programming response to photoperiod in Plodia interpunctella was analyzed by exposing larvae to various 24-h and non-24-h regimes of light and darkness. The response to 24-h regimes indicated three photoperiodic parameters—a critical scotophase, a minimal photophase, and a minimal scotophase for a full expression of the response. The critical response was based on dark-time measurement, because disruption of the scotophase abolished the response and the diapause incidence varied as a function of scotophase in non-24-h regimes. The critical scotophase varied with the duration of the preceding photophase. Prevention of diapause by single or double-night interruptions of long scotophases could be explained by resetting of the dark-time measurement. The effect of a light pulse was modified by the quantitative interaction of light and dark reactions. The sensitivity to resetting by a light pulse seemed to be decreased in the early scotophase with an increasing duration of the preceding light period. Therefore, the significance of light in the photoperiodic response was something more than delimiting scotophase for the time measurement.  相似文献

Female fish were kept under 16L:8D/20 degrees C in November and April and the onset of light and/or feeding times were shifted by several hours in the experimental groups. Photoperiod and feeding entrained significant fluctuations in serum gonadotropin hormone (GTH) levels when the onset of light and the first daily feeding were 4 hr apart, but not when they were 10 hr apart. Fish were subjected to 16L:8D for 14-16 days in February, and to either a constant warm (20 degrees C) or a diurnal sinusoidal (12-20 degrees C) temperature regime, the warmth being imposed during photophase or scotophase. While relatively high, uniform serum GTH levels were found throughout the 24-hr period in fish subjected to constant warmth, warm temperature during the day promoted fluctuations in serum GTH levels, and warmth during night resulted in relatively low, uniform serum GTH levels.  相似文献

Abstract

Resonance experiments (Nanda‐Hamner protocol) conducted at two temperatures for diapause termination in Pimpla instigator (Hymenoptera: Ichneumonidae) do not support the view that the photoperiodic clock has an oscillatory component, but suggest the presence of a non‐rhythmic timer or hourglass mechanism. These results are best explained by a two hourglasses model, one of which starts at light‐on and measures the photophase and the other is initiated by light‐off and measures the scotophase. The most likely hypothesis is that the ratio of photophase to scotophase lengths is the determining element. Good agreement is obtained between results predicted by two hourglasses model and results observed in Pimpla. The diurnal hourglass continues to run for long time (several months) in constant condition (LL) and does not require to be ‘turned over’ by D/L transition, in contrary to the classical model of hourglass which executes a single act of time measurement in extented phase and then stops. The most simple explanation is that some essential factor of diapause termination is synthesized during photophase and degraded during scotophase. Therefore an independent photoperiodic counter (for sommation of daily informations) is not necessary. The two hourglasses system serves as photoperiodic clock and accumulation of product as counter.  相似文献

Circadian clocks in many brain regions and peripheral tissues are entrained by the daily rhythm of food intake. Clocks in one or more of these locations generate a daily rhythm of locomotor activity that anticipates a regular mealtime. Rats and mice can also anticipate two daily meals. Whether this involves 1 or 2 circadian clocks is unknown. To gain insight into how the circadian system adjusts to 2 daily mealtimes, male rats in a 12∶12 light-dark cycle were fed a 2 h meal either 4 h after lights-on or 4 h after lights-off, or a 1 h meal at both times. After 30 days, brain, blood, adrenal and stomach tissue were collected at 6 time points. Multiple clock genes from adrenals and stomachs were assayed by RT-PCR. Blood was assayed for corticosterone and ghrelin. Bmal1 expression was quantified in 14 brain regions by in situ hybridization. Clock gene rhythms in adrenal and stomach from day-fed rats oscillated in antiphase with the rhythms in night-fed rats, and at an intermediate phase in rats fed twice daily. Corticosterone and ghrelin in 1-meal rats peaked at or prior to the expected mealtime. In 2-meal rats, corticosterone peaked only prior the nighttime meal, while ghrelin peaked prior to the daytime meal and then remained elevated. The olfactory bulb, nucleus accumbens, dorsal striatum, cerebellum and arcuate nucleus exhibited significant daily rhythms of Bmal1 in the night-fed groups that were approximately in antiphase in the day-fed groups, and at intermediate levels (arrhythmic) in rats anticipating 2 daily meals. The dissociations between anticipatory activity and the peripheral clocks and hormones in rats anticipating 2 daily meals argue against a role for these signals in the timing of behavioral rhythms. The absence of rhythmicity at the tissue level in brain regions from rats anticipating 2 daily meals support behavioral evidence that circadian clock cells in these tissues may reorganize into two populations coupled to different meals.  相似文献
