The pseudobranchs of bass, Dicentrarchus labrax L. , and grey mullet, Liza ramada (Risso) ( Mugil capito ), were examined in the scanning electron microscope (S.E.M.). Both pseudobranchs appeared gill-like in situ but the S.E.M. revealed their gross morphology to be different. The bass pseudobranch was a'free' pseudobranch, having separate lamellae along the filaments, with areas of fusion on the leading edges of some lamellae, whilst the mullet pseudobranch was 'semi-free' since the secondary lamellae were fused over a large area and only free along their trailing (opercular) edge where the chloride cells were situated. The surface of the epithelial cells presented different patterns of microridges and microvilli depending on their position in the pseudobranch. Three types of cell opening were found at the cell surface, belonging to chloride cells, mucous cells, and possibly rodlet cells. The S.E.M. observations were correlated with those of transmission electron microscope (T.E.M.) studies.  相似文献

The structure of the glandular pseudobranch of the air-breathing fish Anabas testudineus is described on the basis of light and electron microscopy. It is shown that the pseudobranch has the same basic structure as a typical gill, although in this case it is not freely exposed to the water.
Individual secondary lamellae can be recognized in which well-defined, but narrow, blood channels are present between typical pillar cells. The epithelial layer is mainly represented by enlarged mitochondria-rich cells which constitute a large proportion of the whole organ. Mitochondria-rich cells contain an abundant endoplasmic reticulum which is in close contact with the mitochondria and becomes concentrated near the vascular borders of the cell and opens directly into the basement lamina.
The presence of numerous pinocytotic vesicles in the enlarged pillar cell flanges may transfer material to the blood channels. The precise nature and role of any materials released in this way remains to be investigated.  相似文献

Rodlet cells in various stages of development were found in large numbers in the bass gill and pseudobranch. In the gill, rodlet cells were found in the epithelium at the base of the secondary lamellae and on the filament between adjacent lamellae, whilst in the pseudobranch they were found over the whole area of the secondary lamellae as well as in the filament epithelium.
During early development, rodlet cells are characterised by their amorphous cell inclusions, prominent supranuclear Golgi complex and network of granular endoplasmic reticulum. Later, with formation of a fibrous border the arrangement of the cell organelles undergoes reorganisation; the endoplasmic reticulum becomes distended, numerous vesicles appear and the mitochondria aggregate in the apical region of the cell. One of the most striking features is the development of club-shaped sacs containing electron dense cores, which are orientated towards the open apex of the cell.
Various staining properties of rodlet cells for light and electron microscopy were compared with those of mucous cells found in the same tissues. Possible functions of the cell are discussed.  相似文献

An elaborate and apparently unique specialization of the endoplasmic reticulum having the form of tubules and a precise orientation with respect to the mitochondria has been described for the specific cell of the pseudobranch gland. The tubules also are concentrated near the vascular border of the cell where they show continuity with the plasma membrane and open directly against the basement membrane. On the other side of the basement membrane, the endothelial cells of the sinusoid show openings or discontinuities characteristically associated with secretory cells. The pseudobranch gland is presumed to have carbonic anhydrase as one of its primary products, if not its only one, and the elaborate ultrastructure is thought to be associated with the special problems of secreting this enzyme.  相似文献

The ultrastructure of the primary epithelium, and the efferent half of the subepithelium, of the primary gill lamellae of slimy mackerel ( Scomber australasicus ) is described. The following cells are identified and described: light nucleated epithelial cells (surface and basal), dark nucleated cells, mucous cells, acidophilic cells, type 1 cells, type 2 cells, type 3 cells and chloride cells in the epithelial region, and subepithelial cells A and B, fibroblasts, chondrocytes, cells of the wall of efferent blood vessel and some blood cells in the subepithelium.  相似文献

Atlantic cod bearing pseudobranchial tumours were investigated from both North Sea and Baltic Sea areas. Light and electron microscopic studies revealed a tumorous structure previously described in cod from Atlantic and Pacific waters. Additionally, cysts, multinucleated cells and early stages of mitosis are described. These findings support the hypothesis that pseudobranchial tumours are instigated by a protozoan infection causing xenotumours associated with the pseudobranch organ. The cells contained in these lesions showed similarities to amoebae.  相似文献

Luminal surface epithelial cells, excluding a few endocrine cells of the normal human rectum, were studied electron microscopically and 5 types of cells were recognized with special reference to some structure containing mucous substances. Principal-1 cells showing few tiny vesicles and Principal-2 cells containing some tiny vesicles seemed to belong to the absorptive cell group. Vesicle cells having numerous tiny vesicles, and Columnar mucous cells accompanied by numerous tiny vesicles and some round or oval mucous vacuoles, seemed to be labelled as of the secretory cell group. The common features of the epithelial columnar cells, except for the Goblet cell, were columnar shape, microvilli whose length and density had considerable variation, glycocalyceal bodies around the microvilli, and thick surface coat. Goblet cells were characterized by a goblet shape which was expanded by numerous mucous droplets. It is of special interest that 4 different types of columnar epithelial cells are recognized on the luminal surface of the normal human rectum, and that Vesicle cells and Columnar mucous cells are first observed on the luminal surface of the large intestine. Similar epithelial cells have only been reported in the crypt of the large intestine and not on the luminal surface.  相似文献

Summary Cellular differentiation and migration in the fundic glands of adult and larval Xenopus laevis have been examined using bromodeoxyuridine-immunohistochemistry. In the adult fundic gland, cumulative labeling with bromodeoxyuridine revealed a proliferative cell zone between the surface mucous cells and mucous neck cells, in what is referred to as the neck portion of the gland. The labeling-index of mucous neck cells had rapidly increased by week-5. The labeling-index of oxynticopeptic cells showed a more delayed increase until week-7, coincident with the decrease in the labeling of mucous neck cells. In the immature fundic glands of larvae, the labeled proliferating cells were randomly distributed throughout the developing gastric mucosa. During metamorphosis, the labeling-index of immature epithelial cells was highest at stage 63. Following administration of bromodeoxyurdine at this, stage, there was no significant loss of labeled epithelial cells during the metamorphosing period. Furthermore, there was no significant difference in the labeling-indices among the epithelial cells, such as surface mucous cells/generative cells, mucous neck cells, and oxynticopeptic cells, 7 days after administration. Cellular differentiation and migration pathways of epithelial cells in the fundic gland of adult X. laevis and its larvae are discussed.  相似文献

The zinc iodide-osmium tetroxide (ZIO) fixative-staining method was used along with topographical ultrastructure to investigate cholecystectomized human gallbladders under light and electron microscopic techniques. This method delineated neuronal structures which may be involved in controlling the functions of the gallbladder epithelium. Three epithelial cell types were described in the surface epithelium: 1) Columnar clear cells; 2) dark, tuft osmiophilic cells; and 3) basal clear cells with electron-dense granules and showing intense ZIOphilic staining properties. While mucous granules were delineated in the first two cell types, the columnar epithelial clear cells are of uncertain function(s) and content but are probably absorptive cells. The small basal clear cells displaying intense ZIOphilia are associated with intraepithelial nerve endings. These nerve structures may have a sensory and/or motor function(s); they were detected throughout the gallbladder epithelial lining and mucosa.  相似文献

Dorsal and ventral epithelium of the terrestrial slug, Incilaria fruhstorferi, is simple and consists of five cell types: microvillous, ciliated, round mucous, tubular mucous and channel. Microvillous cells were similar to human intestinal epithelial cells morphologically and functionally. At the base of microvilli, pinocytic vesicles which ultimately fused to form larger vacuoles, or multivesicular bodies were present. At the edge of tail or mouth, ciliated epithelial cells possessed the typical axonemes (9 plus 2 arrangement of microtubles). Mucous secretory cells were either tubular or round and their granules were membrane-bound and secreted by exocytosis. Granules of round mucous cells were proteinaceous but those of tubular cells were acidic mucopolysaccharides. Channel cells were elongate U-shaped and the central lumen was filled with a large amount of fluid (hemolymph). The function of channel cells is thought to remove hemolymph accumulated during hyperhydration. Our experiments of some markers-injection revealed that the fluid containing large molecules passed transcellularly from the hemolymph, across the basal or side region of the cell and into the central lumen. These results suggest that channel cell of the slug skin and vertebrate nephron showed some parallels in structure and function.  相似文献

Exposure of pulmonary airways to environmental toxins and allergens may cause proliferation of airway epithelial cells and mucous cell metaplasia (MCM); however, it is unclear to what extent proliferating cells differentiate into mucus-storing cells and contribute to MCM. Our previous studies demonstrated that Bcl-2, an inhibitor of apoptosis with cell cycle regulatory functions, is expressed in metaplastic mucous cells. The purpose of the present study was to investigate the number of metaplastic mucous cells that are derived from proliferating epithelial cells and whether Bcl-2 has a role in cell cycle entry in these cells. Rats were intratracheally instilled with 100 microg of LPS from Pseudomonas aeruginosa in 500 microl of saline, and proliferating airway cells were labeled with bromodeoxyuridine (BrdU) by implanting a subcutaneous osmotic pump 24 h before instillation. The volume of stored mucosubstance and the number of mucous cells were increased 10- and 3-fold, respectively, from 24-48 h after instillation. The number of total epithelial cells per millimeter of basal lamina increased, and the number of serous cells per millimeter of basal lamina decreased during this time. Approximately 50% of Alcian blue-periodic acid Schiff-stained mucous cells were labeled with BrdU at 48 h after instillation, suggesting that one-half of the secretory cells were derived from proliferating cells. Furthermore, 50% of the Bcl-2-positive mucous cells were BrdU negative and therefore derived from nonproliferating, preexisting cells. Our findings demonstrate that preexisting and proliferating cells differentiate into mucous cells and compose LPS-induced metaplasia and that Bcl-2 does not have cell cycle regulatory function in these cells.  相似文献

Structural organization of the epithelium of the lips and associated structures of the Indian major carp,Catla catla, is described. The upper lip is thin and is associated on its dorsal side with a membranous fold of skin and the rostral cap. In contrast, the lower lip is thick and very conspicuous. It is associated on its ventral side with a fold of skin between it and the ventral head skin. The lower lip is divided into a non-projectile portion, a projectile portion and an intermediate groove region. The projectile portion remains folded covering a part of the ventral head skin when the mouth is closed. Their role in relation to the formation of the characteristic feeding tube is discussed. The epithelium of the lips and associated structures is stratified in nature and is composed of the epithelial cells, mucous cells, club cells, lymphocytes and the taste buds. The mucous cells are small, few or even absent and do not appear to secrete profusely at the surfaces of the upper and the lower lips. This suggests that the lips inCatla catla, which feeds on micro-organisms, do not need extra lubrication for protection against abrasion during feeding. In the epithelium at the folds of skin, the voluminous mucous cells secrete profusely and provide extra lubrication to their surface. This reduces the resistance to surface drag during stretching and enables the jaws to protrude with increasing efficiency and swiftness. The club cells are developed additionally to complement the mucous cells in the rostral cap and the upper lip epithelium. Their primary function appears protective in some way, which needs further confirmation. The taste buds, though few in the lower lip, are located in a good number in the upper lip on the characteristic epithelial papillae-like projections, and are projected at the surface. These have been associated with the acute gustatory sense of the fish. The taste buds are absent on the folds of skin where they may not be of much significance.  相似文献

The anatomy and enzyme histœhemistry of the œsophagus of a freshwater teleost perch ( Perca fluviatilis L.) has been studied. The mucosa is composed of an undifferentiated layer of basal epithelial cells, mucous cells and surface epithelial cells. The submucosa is compact and contains blood vessels, nerves and bundles of striated longitudinal muscles. The outer circular muscular layer contains both striated and smooth muscle fibres. Electron microscopy shows that the surface epithelial cells are degenerative and that they surround and support pores of the mucous cells. Undifferentiated epithelial cells undergo cytoplasmic changes and eventually become surface epithelial cells. Mucous cells arise from the undifferentiated epithelial cells and their droplets contain acid and neutral mucosubstances. Alkaline phosphatase, non-specific carboxylic esterases and aminopeptidase activity is absent in the mucosa. However, acid phosphatase activity is localized in the surface epithelial cells.  相似文献

Summary The free or non-glandular pseudobranch of the flounder Paralichthys lethostigma was studied with the electron microscope. Cells typical of glandular type pseudobranchs are found. This indicates that, at least in the flounder, the free pseudobranch should be called glandular. In addition, the chloride-type cells, commonly found in the gill, buccal epithelium, and surface epithelium of other fish, have been found in the pseudobranch, where they have not been described previously. The fine structure of both the chloride-type and the pseudobranch-type cell has been characterized and contrasted. 200 gall-bladders of goat were studied for histology and histochemistry. The lining epithelium is tall columnar with striated border. The glands are mucous and serous, and the secretion of both surface and glands is a polyssaccharide-protein complex. The regional differences at neck, body and fundus of the gall-bladder are described. Few enterochromaffin cells with granules, which are PAS-positive and contain 5-hydroxytryptamine, are encountered at the wedge between the granular epithelial cells and the cells lining the crypts. The relationship between these cells and lymphocytic infiltration is discussed. The thick circular muscle layer at the neck region is considered a sphincter of the gall-bladder.  相似文献

The gastric surface epithelium is situated at an air-liquid interface because the luminal surface of the alimentary tract is in continuity with the air phase. However, the effects of this microenvironment on the gastric epithelium remain unclear. The aim of this study was to clarify the effects of an air-liquid interface on gastric epithelial cell biology. Gastric surface mucous cells (GSM06) were cultured at an air-liquid interface. Cultured cells were examined by histology, histochemistry, and transmission electron microscopy. When the cells were cultured at an air-liquid interface, the surface cells on the collagen gel became tall columnar and secreted periodic acid-Shiff-positive substances at the apical surface. These cells indicated many mucous granules in the apical cytoplasm and organized the basal lamina at the contact side with the gel. In contrast, under immersed condition, the surface cells showed immature features. This is the first report of an air-liquid interface promoting the differentiation of gastric surface mucous cells in a reconstruction culture of the gastric surface epithelial layer, suggesting that an air-liquid interface may function as a crucial luminal factor to maintain the homeostasis of gastric mucosa.  相似文献

The gill epithelium of the airdwelling fish Periophthalmus vulgaris has been studied with the electron microscope. The following celltypes can be distinguished: flat covering epithelial cells, chloride cells, mucous cells, basal cells, various leucocytes as well as a specific granule containing cell which is possibly an epithelial cell. The covering epithelial cells exhibit a relatively smooth apical surface and contain in their apical half densely packed microfilaments, pinocytotic vesicles are rare. These characteristics are not to be found in water dwelling fish and possibly represent adaptations to the air containing surroundings. In the chloride cells are numerous, especially in the basal halves of the secondary lamellae. The distal parts of the secondary lamellae the barrier for the respiratory gases measures about 0,9 micrometer. The basal cells are ribosome rich replacement cells. Two types of mucous cells occur. Individual intraepithelial nerve fibres have been observed.  相似文献

The seminal vesicle epithelium of the mouse and golden hamster was examined by light microscopy and by transmission and scanning electron microscopy. By transmission electron microscopy, in the seminal vesicle epithelium of both animals secretory epithelial cells which consisted of mostly light and a few dark cells were observed. The epithelial cells possessed secretory granules which contained a densely stained core. The secretory granules in the mouse epithelium reacted weakly with periodic acid-Schiff (PAS) stain and were slightly stained with alcian blue (AB), and those in the golden hamster exhibited strongly positive reactions with PAS and AB. The nuclei in the mouse tissue were spherical or ovoid, and those in the golden hamster tissue had a few lobes. By scanning electron microscopy, the apical surfaces of most of the epithelial cells were commonly flat or domed, and those of some epithelial cells protruded into the lumen as apocrine-like processes, or possessed small and large orifices. Besides the epithelial cells, there were cells characterized by pseudopodium-like cytoplasmic projections, a few membranous structures, an irregular nucleus, and cytoplasm containing a few dense bodies, in the basal portions of the epithelial cells, or between the basal lamina and the epithelial cells. Summary A method is described for the maceration (dissociation) of hydra tissue into single cells. The cells have characteristic morphology such that all basic types — epithelial, gland, mucous, interstitial, nematoblast, and nerve — can be distinguished. Criteria are given for identifying each cell type by phase contrast microscopy. It is shown that maceration quantitatively recovers cells from hydra tissue.  相似文献

The structure and function of the pseudobranch has long interested scientists, but its overall role has remained a mystery. Previous studies have attributed respiratory, endocrine, osmoregulatory and sensory roles to the pseudobranch, and the present review concentrates on new findings. Perfusion experiments on the pseudobranch of the rainbow trout (Oncorhynchus mykiss) using both erythrocyte suspensions and Ringer solution have shown that this organ is able to generate values for the respiratory quotient (RQ) greater than 1.0. The release of carbon dioxide into the perfusate was found to be largely independent of flow between perfusion rates of 120-190 microl/min and could be inhibited by acetazolamide (10(-5) M), indicating a role for carbonic anhydrase. Noradrenaline (10(-5) M) had no effect on oxygen consumption or carbon dioxide release of the pseudobranch. The rate of carbon dioxide release was also dependent on the pH of the pre-pseudobranch perfusate, carbon dioxide release being reduced at lower perfusate pH values. Based on the glucose balance of the isolated saline-perfused rainbow trout pseudobranch and on the enzyme profiles for the rainbow trout, cod, swordfish and deep-water grenadier pseudobranch, it is suggested that the pentose phosphate shunt might be a source of carbon dioxide, yielding the high RQ values found for this organ. Most evidence now available indicates that the pseudobranch is integrally linked with the choroid rete and the supply of oxygen to the retina of the fish eye.  相似文献
