The effect of orciprenaline sulphate on the in vitro production of PGs E and F by guinea pig uterine tissue homogenates has been investigated. Estimations of PGs were carried out using sensitive radioimmunoassay techniques. The results indicate that orciprenaline can effectively reduce the in vitro yield of PGs E and F from guinea pig uterine tissue. This could in turn be due to an inhibitory effect of orciprenaline on PG formation or due to stimulation of PG breakdown by the drug. The implications of these results in relation to premature labour are discussed.  相似文献

The possibility of a direct effect of prostaglandins of the E, A, and F series upon renal electrolyte and water transport was assessed using in vitro preparations of rabbit cortical and medullary tubular suspensions as well as cortical renal slices from rat and guinea pig and medullary renal slices from rabbit. Net fluxes of Na, K, Cl and H2O between the intracellular compartment and the extracellular fluid were measured in the presence of PGE1, PGE2, PGA1, PGA2 and PGF in concentrations ranging from 1 × 10?5 to 1 × 10?10M. No inhibitory action was observed with any of these prostaglandins and in fact a slight stimulation of Na transport was seen under some circumstances. We conclude that the natriuresis which follows in vivo administration of some prostaglandins is not the result of a direct inhibition of Na reabsorption at the contraluminal pump site and is most likely secondary to renal vasodilation.We also studied net and isotopic Na fluxes in human erythrocytes. Na transport was not affected by prostaglandins of the E, A or F series using both normal and high sodium erythrocytes. Our results emphasize the need for caution in extrapolating the effects of prostaglandins upon Na transport from one tissue to another since their actions appear to be tissue-specific.  相似文献

Apocynin is a constituent of root extracts of the medicinal herb Picrorhiza kurroa and has been shown to possess anti-inflammatory properties. We investigated the effects of apocynin on the production of arachidonic acid-derived inflammatory mediators by guinea pig pulmonary macrophages. Apocynin concentration-dependently inhibited the formation of thromboxane A2, whereas the release of prostaglandins E2 and F2 alpha was stimulated. Apocynin potently inhibited arachidonic acid-induced aggregation of bovine platelets, possibly through inhibition of thromboxane formation. The present results suggest that apocynin might, beside its therapeutic effects in inflammatory conditions when given in a root extract of P. kurroa, also be a valuable tool in the development of new anti-inflammatory or anti-thrombic drugs.  相似文献

Thromboxane B2 was formed from endogenous precursors during short incubations of guinea pig and rat cerebral cortex. The amount formed by guinea pig brain tissue was 5–6 times the formation of prostaglandin F and E2. Noradrenalin stimulated and indomethacin and mercaptoethanol inhibited thromboxane B2 formation. The mass spectrum of the brain compound was identical to thromboxane B2 formed from arachidonic acid by guinea pig lung and human platelets.  相似文献

Recent reports suggest that prostaglandins, rather than cAMP, play a major role in mediating cholera toxin-induced water and electrolyte secretion from rabbit intestinal loops. We examined the role of prostaglandins in mediating toxin-induced pancreatic and gastric exocrine secretion. In these tissues, indomethacin, a potent inhibitor of prostaglandin synthesis, did not alter the stimulatory effects of cholera toxin on increases in cellular cAMP or enzyme secretion. Moreover, the addition of cholera toxin did not alter prostaglandin E2 release from either tissue. In contrast to their effects in rabbit intestinal loops, prostaglandins do not regulate cholera toxin-induced enzyme secretion from the guinea pig pancreas or stomach.  相似文献

In vitro synthesis of progesterone and estradiol-17 beta from endogenous precursors was studied in the placenta from women in early stage of gestation (less than 7 weeks). Radioimmunoassay techniques were used to measure progesterone and estradiol-17 beta. It was shown that placental tissue from as early as six weeks of gestation can synthesize both progesterone and estradiol-17 beta in vitro. Prostaglandins F2 alpha and E2 in concentration of 100 micrograms/ml of the incubation media did not have any significant effect on the in vitro synthesis of progesterone and estradiol-17 beta in the placental tissue. It seems unlikely that the abortifacient effect of natural prostaglandins PGE2 and PGF2 alpha is due to their direct action on the synthesis of progesterone and estradiol-17 beta in the placenta.  相似文献

The ability of 8-ethoxycarbonyl-10, 11 dihydro-A-prostaglandin(HR 546) to antagonise smooth muscle contracting effect of prostaglandins E2 and F2alpha on isolated preparations of rat and hamster stomach fundus, guinea pig ileum and gerbil colon has been studied. HR 546 was found to be a potent, non-specific, probably competitive, prostaglandin antagonist on these four smooth muscle preparations.  相似文献

Bovine gastric mucosal and muscle microsomes synthesize prostaglandins and thromboxane b/ (TXB2) from aratchidonic acid (AA). TXB2 and 6-keto-prostaglandin F1 alpha (6-keto-PGF1 alpha) were the majro products synthesized by pylorus, body, and cardiac region of the gastric mucosa. Gastric muscle mainly synthesized 6-keto-PGF1 alpha. TXB2 and 6-keto-PGF1 alpha synthesis occurs at an appreciable rate from endogenous precursors but more rapidly with added arachidonate. Prostaglandins E2, F2 alpha and D2 were synthesized in smaller amounts under the conditions studied.  相似文献

The effect of photoperiod and reproductive cycle on the fluctuations in basal serum cortisol (F) levels were determined in adult female guinea pigs bled by intracardiac puncture under light methoxyflurane anesthesia. Blood samples were collected at regular intervals over a 24 hour period in order to assess the effects of the lighting regime (14 hr light/day: lights on 0600 hr) on basal levels of F. Peak F levels were observed at the beginning of the light cycle and lowest levels observed just prior to the onset of darkness in random, cyclic female guinea pigs. When analyzed for the influence of reproductive state, basal F levels were found to be elevated during proestrus, Day 2 (Day 0 = estrus) and the mid-luteal (Day 6-7) period of the estrus cycle. In ovariectomized animals a 10 microgram dose of estradiol (E) induced a dramatic rise (P less than 0.001) in adrenal weight and the basal levels of serum F as compared with oil-treated controls. Lower doses of E (1 microgram) and combined progesterone (2 mg)-E (1 microgram) treatment induced an elevation in adrenal weight and/or basal levels of serum F. Progesterone administration alone had no effect on either of these adrenal indices as compared with controls. These data indicate that both photoperiod and ovarian steroid hormone milieu affect adrenal function in the female guinea pig.  相似文献

The effect of exogenous prostaglandins E1, E2 and F2alpha (PGE1, PGE2 and PGF2alpha) on 3H-leucine, 3H-uridine, 3H-thymidine and 3H-proline incorporation in experimental cutaneous wounds has been studied in rats. Prostaglandins E1 and E2 markedly stimulate the incorporation of these tritiated precursors, into protein, RNA, DNA and collagen synthesis, whereas F2 inhibits it. All tested prostaglandins exhibit their maximum effect within the first hours following administration. Most active is PGE1. These observations indicate that application of prostaglandins significantly stimulate incorporation with protein, RNA, DNA and collagen synthesis in the skin of wounded rats and thus, may play a role in epidermal cell growth and division as well as in scar-forming tissue.  相似文献

Slow reacting substance (SRS) injected into the pulmonary artery released prostaglandins E (PGE) and F2alpha (PGF2alpha) and the 15-keto-13, 14-dihydro PG metabolites from non-sensitized and ovalbumin sensitized, isolated, perfused guinea pig lungs. PGs were also released from lungs incubated with SRS. Sensitized lungs released more PGs in both types of preparations. Indomethacin inhibited the effect of SRS. Passively sensitized human lung fragments, in parallel to guinea pig lung, released PGE, PGF2alpha and the metabolites when incubated with SRS or antigen. In in vivo experiments, SRS and arachidonic acid given intravenously increased the airway insufflation pressure in anesthetized quinea pigs. These effects, but not the action of injected PGF2alpha and histamine, were abolished by indomethacin. The results indicate that one of the modes of SRS action is by release of PGs, and are consistent with the hypothesis that PGs are predominantly "secondary" mediators (in the temporal sense) of the antigen-antibody reaction.  相似文献

In vitro antigen challenge has multiple effects on the excitability of guinea pig bronchial parasympathetic ganglion neurons, including depolarization, causing phasic neurons to fire with a repetitive action potential pattern and potentiating synaptic transmission. In the present study, guinea pigs were passively sensitized to the antigen ovalbumin. After sensitization, the bronchi were prepared for in vitro electrophysiological intracellular recording of parasympathetic ganglia neurons to investigate the contribution of cyclooxygenase activation and prostanoids on parasympathetic nerve activity. Cyclooxygenase inhibition with either indomethacin or piroxicam before in vitro antigen challenge blocked the change in accommodation. These cyclooxygenase inhibitors also blocked the release of prostaglandin D(2) (PGD(2)) from bronchial tissue during antigen challenge. We also determined that PGE(2) and PGD(2) decreased the duration of the action potential after hyperpolarization, whereas PGF(2alpha) potentiated synaptic transmission. Thus prostaglandins released during antigen challenge have multiple effects on the excitability of guinea pig bronchial parasympathetic ganglia neurons, which may consequently affect the output from these neurons and thereby alter parasympathetic tone in the lower airways.  相似文献

Auditory function and cochlear morphology have previously been described in the postnatal German waltzing guinea pig, a strain with recessive deafness. In the present study, cochlear histopathology was further investigated in the inner ear of the developing German waltzing guinea pig (gw/gw). The lumen of the cochlear duct diminished progressively from embryonic day (E) 35 to E45 and was absent at E50 because of the complete collapse of Reissner's membrane onto the hearing organ. The embryonic stria vascularis, consisting of a simple epithelium, failed to transform into the complex trilaminar tissue seen in normal animals and displayed signs of degeneration. Subsequent degeneration of the sensory epithelium was observed from E50 and onwards. Defective and insufficient numbers of melanocytes were observed in the developing gw/gw stria vascularis. A gene involved in cochlear melanocyte development, Pax3, was markedly reduced in lateral wall tissue of the cochlea of both E40 and adult gw/gw individuals, whereas its expression was normal in the skin and diaphragm muscle of adult gw/gw animals. The Pax3 gene may thus be involved in the pathological process but is unlikely to be the primary mutated gene in the German waltzing guinea pig. TUNEL assay showed no signs of apoptotic cell death in the developing stria vascularis of this type of guinea pig. Thus, malformation of the stria vascularis appears to be the primary defect in the inner ear of the German waltzing guinea pig. Defective and insufficient numbers of melanocytes might migrate to the developing stria vascularis but fail to provide the proper support for the subsequent development of marginal and basal cells, thereby leading to stria vascularis malformation and dysfunction in the inner ear of the German waltzing guinea pig.  相似文献

Various species have been used as models to study the effects of adenosine (ADO) on atrial and ventricular myocardium, but few direct tissue comparisons between species have been made. This study further characterizes adenosine A(1) receptor binding, adenylate cyclase activity and direct and indirect A(1) receptor-mediated functional activity in atrial and ventricular tissue from Sprague-Dawley rats and Hartley guinea pigs. Rat right atria (RA) were found to be significantly more sensitive to cyclopentyladenosine (CPA), while guinea pig left atria (LA) were more sensitive to CPA. After the addition of isoproterenol (ISO), the reduction of CPA response in rat RA was significantly greater than in guinea pig; however, after ISO treatment, the guinea pig LA was more sensitive to CPA than the rat. Adenylate cyclase inhibition by CPA was significantly greater in atria and ventricles obtained from guinea pig than rat. In competition binding experiments, guinea pig RA had significantly more high affinity sites than rat, but the K(i)s were not significantly different. There were no significant differences between guinea pig LA and rat LA. Guinea pig ventricular tissue had fewer high affinity sites than rat without any differences in their K(i) values. In antagonist saturation experiments, the density and affinity of A(1) receptors in guinea pig cardiac membranes were significantly greater than in rat. Our results indicate definite species differences as well as tissue differences between rat and guinea pig. These differences must be considered when interpreting studies using rat and guinea pig tissue as models for cardiac function.  相似文献

Prostaglandin synthesis in vitro and its inhibition by dibutyryl cyclic AMP and forskolin were examined in postpartum follicles of the guppy. The follicles were cultured for up to 72h under chemically-defined conditions, and the synthesis of prostaglandins was determined by radioimmunoassay. The results show unequivocally that the ovarian follicles devoid of embryos can synthesize PGE and PGF from both endogenous precursors as well as added arachidonic acid. That this could be inhibited by indomethacin is indicative of active synthesis rather than release of preformed prostaglandins. Dibutyryl cAMP and forskolin also inhibited prostaglandin synthesis in a dose-dependent manner, as did 3-isobutyl-l-methylxanthine. A similar effect was also evident in vitellogenic oocytes surrounded by follicle layers at the yolk-globule stage of development. While the physiological significance of this action of the cyclic nucleotide remains uncertain at this stage, it is hypothesized that it could explain the non-occurrence of ovulation in this viviparous fish soon after final oocyte maturation.  相似文献

The aim of the present study was to investigate and compare specific in vitro pharmacological actions of human alpha- and beta-CGRP applied as single concentrations to prostaglandin F2alpha precontracted segments of guinea pig basilar artery. To support the suggestion of a possible link between the pharmacological actions of alpha- and beta-CGRP and a specific receptor, we wished to determine whether mRNAs required for the expression of calcitonin receptor-like receptor (CRLR) derived CGRP receptors were present in the guinea pig basilar artery. In the pharmacological experiments we demonstrated an increase in the cAMP content by 2.5-fold and a concomitant significant vasorelaxation of the precontracted basilar artery segments following 1 min of stimulation by 10(-7) M alpha- or beta-CGRP. In another set of experiments, the time course of alpha- and beta-CGRP induced vasodilatation was investigated and concentration dependent responses of the two peptides were demonstrated. No significant differences between the actions of alpha- and beta-CGRP regarding induction of cAMP formation, amount of vasodilatation, time course of vasodilatation and mode of inhibition by the CGRP receptor antagonist, human alpha-CGRP(8-37), could be detected. The presence of mRNA encoding the guinea pig CRLR and the guinea pig CGRP receptor component protein (RCP) in the guinea pig basilar artery was demonstrated by RT-PCR methods. Furthermore, a partial sequence of mRNA encoding the guinea pig CRLR was determined. The expression in this tissue of a CRLR derived CGRP receptor and a functional RCP is therefore likely, and the equipotent pharmacological actions of alpha- and beta-CGRP might be mediated via CRLR derived CGRP receptors.  相似文献

The results presented in this paper indicate that: 1. The prostaglandin synthesis inhibitor, indomethacin, increases noradrenaline turnover in a variety of rat organs. This observation increases the probability that prostaglandins are involved in the control of adrenergic neurotransmission in vivo. 2. Administration of endoperoxides inhibits the release of noradrenaline from adrenergic nerve terminals. The effect can be explained, however, at least in part, by formation of degradation products, presumably mainly prostaglandin E2. 3. Prostaglandin F2 alpha enhances smooth muscle responses to adrenergic nerve stimulation in rabbit heart and guinea pig vas deferens. These actions must be considered prostjunctional, since the release of noradrenaline is unchanged or depressed.  相似文献

Although numerous data exist concerning tritium kinetic isotope effect in enzymic reactions, little is related to the metabolism of tritiated prostaglandins. The present study reports an evaluation of the kinetic isotope effect which occurs during the oxidation of 15-hydroxyl group of tritium-labeled prostaglandins E2 and F2 alpha by the 15-hydroxyprostaglandin dehydrogenase and during the oxidation of 9-hydroxyl group of tritium-labeled prostaglandin F2 alpha by the 9-hydroxyprostaglandin dehydrogenase. The large kinetic isotope effect tends to limit the validity of the dehydrogenase assay using tritium-labeled prostaglandins as substrate. However these assays can be considered to be an indication of relative enzyme activity.  相似文献

Freshly isolated neonatal porcine aortic tissue (smooth muscle with or without endothelium present) produced approximately 30 ng/mg wet tissue of 6-oxo-prostaglandin F1 alpha (the stable hydrolysis product from prostacyclin) and approximately 15 ng/mg of prostaglandin E2, as measured by radioimmunoassay after 24 h incubation in culture medium. Primary cultures of porcine endothelial and smooth muscle cells (isolated by enzymic digestion of aortic tissue) exhibited the same pattern of prostaglandin production, but absolute values were greater than for fresh tissue, particularly in the case of endothelium. Subcultures of endothelium produced smaller amounts of prostaglandins, although the pattern remained similar. In contrast, subcultures of smooth muscle cells produced a greater total amount of prostaglandins than did primary cultures, and the main product was prostaglandin E2. Experiments with [14C] prostaglandin H2 or [14C]arachidonic acid confirmed that aortic tissue, cultured endothelium, and primary cultures or aortic smooth muscle cells synthesized prostacyclin, and demonstrated that subcultured smooth muscle cells enzymically isomerised prostaglandin H2 to prostaglandin E2. Kinetic studies showed that prostaglandin production by cultured vascular cells was transiently increased by subculture or changing the growth medium, and that production per cell declined with increasing cell density. The change in pattern of prostaglandin production during culture was shown to be due to a rapid decline in the rate of prostacyclin production (which apparently began immediately after tissue isolation), together with a more gradual rise in prostaglandin E2 production. These results indicate that the amounts and ratios of prostaglandins produced by vascular endothelial and smooth muscle cells are greatly affected by the conditions used to isolate and culture the cells; vascular cells in vivo may similarly alter their pattern of prostaglandin production in response to local changes in their environment.  相似文献

Synthetic analogues of prostaglandins E2 or F2a (monocyclic bisenoic prostaglandins), like the endogenous prostaglandin endoperoxides (prostaglandins G2 and H2) from platelets, and like synthetic analogues of prostaglandin H2 (bicyclic bisenoic prostaglandins), can induce aggregation of human platelets, although prostaglandins E2 and F2a themselves are inactive. 2. All the prostanoid compounds that induce platelet aggregation release 5-hydroxytryptamine from platelet dense bodies, but do not release beta-N-acetylglucosaminidase from lysosomal granules. Arachidonic acid evokes a similar response. 3. All endoperoxide analogues tested (bicyclic compounds) were powerful platelet stimulants, and all active compounds (whether mono- or bi-cyclid) apparently acted via the same receptor as the endogenous prostaglandin endoperoxides. 4. The nature and stereospecificity of substituents at positions 11 and 15 (or 16) on prostaglandin E2 are critical determinants for platelet-stimulating activity: deoxy substitution at position 11 plus methylation at position 15 (or 16) produces a potent stimulant, particularly if the groups around C-15 are in the S configuration. 5. The effects of these structural modifications are apparently due to, at least in part, a change in side-chain conformation.  相似文献
