The effects of light on the pH in the vacuole and the electricpotential difference across the plasmalemma and the tonoplastof Nitellopsis obtusa were investigated by means of conventionaland H+-specific glass or antimony microelectrodes. Illuminationis found to bring about a decrease in the pH of the vacuolarsap by 0.1–0.5 units concomitant with a depolarizationof the cell. The light-induced changes of the potential differenceand the vacuolar pH depend in different ways on the pH of theexternal medium (pHo). At pHo 9.0 cells exhibit great light-inducedpotential changes (up to 100 mV), but only small pH changesof the vacuolar sap. At neutral or slightly acidic pHo valuesthe amplitude of the light-induced pH changes in the vacuoleincreases up to 0.3–0.5 pH units, but the amplitudes ofthe potential changes at the plasmalemma are relatively small.At pHo 9.0 a transient acidification of the medium is observedupon illumination whereas at lower pH values light-induced alkalinizationwas only seen. Transfer of the cells from pHo 9.0 to pHo 7.5results in a cell hyperpolarization by 60–80 mV and adecrease of the vacuolar pH by 0.4–0.5 units under lightconditions but has no significant effect on the potential andthe vacuolar pH in the darkness. It is proposed that mechanismsof active H+ extrusion from the cytoplasm are located both inthe plasmalemma and the tonoplast. The observed acidificationin the vacuole appears to be determined by a light-induced increaseof the concentration of H+ in the cytoplasm. The H+ conductionof the plasmalemma seems to increase on illumination. The patternof the light-induced H+ fluxes across the tonoplast and theplasmalemma depends crucially on the extent of the light-inducedchanges in the H+ conductance and on the electrochemical gradientfor H+ at the plasmalemma.  相似文献

K+ efflux from tobacco (Nicotiana tabacum L, cv. Samsun NN)leaf discs into the external medium was increased and the membranepotential (Em) changed in the positive direction with a changein pH from 8.0 to 4.0. Em was affected by the external concentrationof KCl, greatly decreasing with a change in concentration from1 mM to 100 mM. The equilibrium potential of the membrane forK+ (Ek) was decreased in a Nernst fashion with increasing externalconcentrations of KCl. Ek is more positive than Em above ca.50 µM KCl. Most of the experiments were carried out underconditions in which the difference between the electrochemicalpotential for K+ on the inside to the outside of the cell (µkis positive. Thus, K+ may passively flow to the outside of thecells accompanied by the depolarization of the membrane. Abscisic acid (ABA) increased the K+ efflux under conditionsof passive transport. K+ efflux was accelerated with an increasingconcentration of ABA, being maximal at 10–4 M–10–3M. This acceleration was due to the enhancement of the potassiummotive force (µk/F) which is the force causing the netpassive transport of K+. The membrane potential was decreasedfrom –205 mV to –170 mV by 2 x 10–4 M ABAwithin 10 min. The depolarization was not transient, being lostfor at least 3 hr. Kennedy, C. D. and Gonsalves, F. A. N. 1988. H+ efflux and trans-rootpotential measured while increasing the temperature of solutionsbathing excised roots of Zea mays.—J. exp. Bot. 39: 37–49. Novel temperature-ramp procedures have been used to measureH+ efflux and trans-root potential of excised roots of Zea mays(var. Fronica). Two types of experiment were performed: (1),increasing temperature from 17°C, and (2), pre-cooling theroots to 1°C before starting the temperature ramp. The ratesof increase of temperature for H+ efflux and trans-root potentialexperiments were 0·5 and 2·1°C min–1respectively The H+ scans revealed strong sharp maxima at 30°C and 32°C,for non-pre-cooled and pre-cooled roots respectively, the latterbeing significantly smaller. The trans-root potential scansfor the pre-cooled roots showed a corresponding maximum at 30°C,which was inhibited by KCN (1-0 mmol dm–3) with or withoutSHAM (10 mmol dm–3), or Hg2+ (1, 10, 100 µmol dm–3)in the bathing solutions. Some of the evidence suggests thatthese maxima are associated with electrogenic H+ pumping, mediatedby a plasma membrane-bound ATPase. However, no correspondingmaximum was observed in the trans-root potential scans for non-pre-cooledroots, the potential remaining at about — 75 m V from20°C to 35°C. As there is a 7-fold increase in H+ effluxbetween 20°C and 30°C, the relationship between netH+ efflux and electrogenic proton pumping in these roots isby no means clear. Some possibilities are considered here. Pre-cooled and non-pre-cooled roots show clear maxima in thetrans-root potential scans at about 46°C, at which temperaturethere is a slight net H+ influx. Mechanism of Photosynthate Efflux from Vicia faba L. Seed Coats   总被引:2,自引:0,他引:2  
In order to develop a tentative model of the mechanism of photosynthateefflux from the vascular region of Vicia faba L. seed coats,wash-out experiments were performed after removal of the embryo. The sulphydryl group modifiers, pCMBS and NEM, reduced 14C-photosynthateefflux by 40% and 50%, respectively. Their inhibitory effectcould be prevented or reduced (in the latter case) by includingDTT in the bathing solution. Maltose competed with sucrose forefflux; a concentration of 300 mol m–3 inhibited 14C-photosynthaterelease by 35%. The cations K+ , Na+ Mg2+ and TPP+ enhancedefflux significantly, whereas the countenon Cl had noeffect. The presence of the protonophore CCCP (0·1 molm–3) led to a reduction of efflux by 50% net proton extrusiondropped by 34%. To a lesser extent, an efflux inhibition wasalso achieved by decreasing the cytoplasmic pH with the weakacid DM0. In contrast, alterations in the external pH causedonly a feeble response. The ATPase inhibitor, EB, decreasedphotosynthate efflux and H+ extrusion. DES reduced efflux slightly,presumably by affecting ATPase activity as well as energy metabolism. Based on these findings, it is proposed that a sucrose/protonantiport mechanism could be responsible for photosynthate effluxfrom Vicia faba seed coats. Internodal cells of Nitellopsis were made tonoplast-free byperfusion with a medium containing EGTA. Cytoplasmic concentrationsof solutes were controlled by a second perfusion with mediaof known composition. The electrogenic pump current (Ip), whichwas calculated from electrical data obtained from cells withand without ATP, was compared with the current carried by H+(IH+) across the plasma membrane. A close correlation betweenIp and IH+ was found under various internal and external conditions.(1) Ip and IH+ depended on the internal ATP and showed Michaelis-Mententype saturation curves. For Ip, Km was 120 µM and themaximum current Vmax was 15.1 mA m–2, while for IH+, Kmwas 160 µM and Vmax was 16.6 mA m–2. (2) Ip andIH+ showed almost the same IH2+ dependence. The Mg2+-dependentIp was 19.5 mA m–2, while the Mg2+-dependent IH2+ was17.7 mA m–2. (3) IH2+ was maximal at an external pH of8 and decreased both in acidic and alkaline pH ranges. Ip wasnearly equal to IH+ in the pH range between 8 and 5. (4) IH+became maximal at an internal pH of 7.3, which is nearly thesame as the pH for maximal electrogenecity found by Mimura andTazawa (1984). The functional properties of the Saccharomyces cerevisiae bicarbonate transporter homolog Bor1p (YNL275wp) were characterized by measuring boron (H3BO3), Na+, and Cl fluxes. Neither Na+ nor Cl appears to be a transported substrate for Bor1p. Uphill efflux of boron mediated by Bor1p was demonstrated directly by loading cells with boron and resuspending in a low-boron medium. Cells with intact BOR1, but not the deletant strain, transport boron outward until the intracellular concentration is sevenfold lower than that in the medium. Boron efflux through Bor1p is a saturable function of intracellular boron (apparent Km 1–2 mM). The extracellular pH dependences of boron distribution and efflux indicate that uphill efflux is driven by the inward H+ gradient. Addition of 30 mM HCO3 does not affect boron extrusion by Bor1p, indicating that HCO3 does not participate in Bor1p function. Functional Bor1p is present in cells grown in medium with no added boron, and overnight growth in 10 mM H3BO3 causes only a small increase in the levels of functional Bor1p and in BOR1 mRNA. The fact that Bor1p is expressed when there is no need for boron extrusion and is not strongly induced in the presence of growth-inhibitory boron concentrations is surprising if the main physiological function of yeast Bor1p is boron efflux. A possible role in vacuolar dynamics for Bor1p was recently reported by Decker and Wickner (10). Continuous measurements of cytoplasmic pH (pHc) in Sinapis roothairs have been carried out with double-barrelled pH-micro-electrodesin order to gain information on translocation of protons acrossthe plasmalemma and cytoplasmic pH control. (i) The cytoplasmicpH of Sinapis (7–33 ? 0–12, standard conditions)changes no more than 0.1 pHc, per pHo-unit, regardless of whethercyanide is present or not. (ii) Weak acids rapidly acidify pHcand hyperpolarize, while weak bases alkalize pHc and depolarizethe cells, (iii) 1.0 mol M,3 NaCN acidifies the cytoplasm by0.4 to 0.7 pH-units, but alkalizes the vacuole. (iv) 20 mmolm–3 CCCP has no significant effect on pHc, if added atpH 9.6 or 7.2, but acidifies pHc by 1.3 units at pH 4.3. Inthe presence of CCCP, cyanide acidifies the cytoplasm, (v) Chloridetransiently acidifies pHc, while K+, Na+, and have no significant effects, (vi) Cytoplasmic buffer capacityforms a bell-shaped curve versus pHc with an optimum of about50 mol m–3 H+pHc-unit. The modes of proton re-entry and the effects of active and passiveproton transport on cellular pH control are critically discussed.It is suggested that the proton leak, consisting of H+-cotransport(e.g. H+/Cl) rather than H+-uniport, is no threat topHc. The proton export pump, although itself reacting to changesin pHc, influences pHc only to a minor extent. It is concludedthat buffer capacity and membrane transport play moderate rolesin pHc control in Sinapis, while the interlocked H+-producingand -consuming reactions of cellular metabolism are the mainregulating factors. Cucumber (Cucumis sativus L.) seedlings were grafted onto cucumber-(CG) or figleaf gourd- (FG, Cucurbita ficifolia Bouché)seedlings in order to determine the effect of solution temperature(12, 22, and 32°C) on the mineral composition of xylem sapand the plasma membrane K+-Mg++-ATPase activities of the roots.Low solution temperature (12°C) lowered the concentrationof NO3 and H2PO4 in xylem sap of CG plants butnot of FG plants. Concentrations of K+, Ca++ and Mg++ in xylemsap were less affected than anions by solution temperature.The plasma membrane of FG plants grown in 12°C solutiontemperature showed the highest K+- Mg++-ATPase activity at allATP concentrations up to 3 mM and at low reaction temperatureup to 12°C, indicating resistance of figleaf gourd to lowroot temperature. Ion absorption by Chara corallina from solutions containingpredominantly KC1 or RbCl at up to 100 mol m–3 resultedin accumulation of salts and turgor regulation. Turgor regulationdid not occur in solutions containing Na+ or Li+salts. Duringion absorption from various salts of K+ and Rb+ vacuolar cationconcentration exceeded Cl concentration. This differencewas shown to be balanced by the synthesis and accumulation ofmalate. Vacuolar malate concentration reached 48 mol m3,with accumulation occurring at rates of up to 0.45 mol m–3h–1. Malate accumulation was inhibited by low externalpH and was dependent upon external HCO3 concentration.The synthesis of malic acid and its subsequent dissociationimposed a severe acid load on the cell. Biophysical regulationof cellular pH was achieved by a H+efflux at a rate of about40 nmol m–2 s–1from the cell. The results presentedargue against cytoplasmic Cl, HCO3 or pH regulatingmalate accumulation in Chara and it is suggested that malatetransport across the tonoplast may regulate malate accumulation. Cytoplasmic pH (pHc) in Chara corallina was measured (from [14C]stribution)as a function of external pH (pH0)and temperature. With pH0near 7, pHc at 25?C is 7.80; pHcincreases by 0.005 pH units?C–1 temperature decrease, i.e. pHc at 5 ?C is 7.90. WithpH? near 5.5, the increase in pHc with decreasing temperatureis 0.015 units ?C–1 between 25 and 15?C, but 0.005 units?C–1 between 15 and 5?C. This implies a more precise regulationof pHc with variations in pHo at 5 or 15 ?C compared with 25?C. The observed dp Hc/dT is generally smaller than the –0.017units ?C–1 needed to maintain a constant H+/OH–1,or a constant fractional ionization of histidine in protein,with variation in temperature. It is closer to that needed tomaintain the fractional ionization of phosphorylated compoundsor of CO2–HCO3 The value of dpHc/dT has importantimplications for several regulatory aspects of cell metabolism.These include (all as a function of temperature) the rates ofenzyme reactions, the H+ at the plasmalemma(and hence the energy available for cotransport processes),and the mechanism for pHc regulation by the control of bidirectionalH+ fluxes at the plasmalemma.  相似文献

The acidophilic alga Dunaliella acidophila exhibits optimalgrowth at pH 1. We have investigated the regulation of phosphateuptake by this alga using tracer techniques and by performingintracellular phosphate measurements under different growthconditions including phosphate limitation. In batch culturewith 2·2 mol m–3 phosphate in the medium the uptakeof phosphate at micromolar phosphate concentrations followeda linear time dependence in the range of minutes and rates werein the range of 1 µmol phosphate mg–1 chl h–1,only. However, under discontinuous phosphate-limited growthconditions, tracer influx revealed a biphasic pattern at micromolarphosphate concentrations: An initial burst phase resulted ina 104-fold internal phosphate accumulation and levelled offafter about 10 s. A double reciprocal plot of the initial influxrates obtained for phosphate-limited and unlimited algae exhibitedMichaelis-Menten kinetics. Phosphate limitation caused a significantactivation of the maximum velocity of uptake, yielding Vmaxup to 1 mmol mg–1 chl h–1 as compared to valuesin the order of 50 µmol phosphate mg–1 chl h–1for the second phase (this magnitude is also representativefor non-limited batch cultures). Concomitantly the Michaelisconstant was altered from 4 mmol m–3 to 0·7 mmolm–3. The rapid uptake of phosphate was inhibited by arsenateand FCCP and was not stimulated by Na+. The pH dependence oftracer accumulation and measurements of the intracellular phosphatepool under different growth conditions indicate that at lowpH and low external phosphate concentrations the high protongradient present under these conditions is utilized for a H3PO4uptake or a H+/H2PO4 cotransport. However, when the externalphosphate concentration was increased to levels sufficientlyhigh for transport to be driven by the positive membrane potential(10 mol m–3 phosphate), the pH dependence of phosphateuptake was more complex, but could be explained by the uptakeof H3PO4 or a H+/H2PO4-cotransport at low pH and a differenttype H2PO4-transport (with unknown type of ion coupling)at high pH-values. It is suggested that this flexible couplingof phosphate transport is of essential importance for the acidresistance of Dunaliella acidophila. Catharanthus roseus(L.) G. Don cells acidified Mura-shige-Skoogmedium rapidly. Upon transfer to fresh medium, the medium pH(initially5.3) dropped below 4 within 2 d. This acidificationwas reversed under hypoxic conditions. The cells induced a similaracidification in a simple medium consisting of CaCl2, KCl, andglucose: medium pH dropped below 4 within 6 h. The acidificationwas accompanied by an influx of K+ at a H+(efflux)/K+ ratioof ca 0.6 as well as by an expansion of endogenous organic acidpool, in which malic and citric acids were the major components.Anoxia reversed all these processes: the direction of both K+and H+ fluxes reversed with a H+/K+ ratio of 1.70. Anoxia induceda cytoplasmic acidification from pH 7.6 (aerobic) to 7.4 asmeasured by 31P-NMR, accompanied by a rapid, long-lasting lactateaccumulation at expense of malic and citric acids. Evidencesuggested that accumulation of lactic acid was not a cause ofcytoplasmic acidification under anoxia, but a result of pH regulationby the biochemical pH-stat [Davies (1973) Symp. Soc. Exp. In this report I discuss ventilatory and circulatory adjustmentsthat prov for increased O2transport associated with increasedbody temperature in the snake Coluber constrictor. Also includedis the effect of temperature upon acid-base status. Minute ventilationincreases with rising body temperature but does not keep pacewith the increment in resting O2 consumption. The decrease inair convection requirement (i.e., ventilation ÷ oxygenconsumption) causes lung pO2 and arterial oxygen contentto falland lung pCO2 to rise. With the rise in lung pCO2, systemicarterial pCO2 and H+; concentration increase while plasma bicarbonateconcentration does not change. The effect of temperature uponair convection requirement, arterial pCO2, and pH are most pronouncedat body temperatures above about 27°C whereColuber behavesapproximately as an alphastat pH regulator. Despite the inverserelationship between temperature and lung pO2, systemic arterialpO2 is about 80 torr lower at 15°C than at 35°C. Thisdecline in arterial pO2 as temperature falls is explained byleft shifting the oxygen dissociation curve in the presenceof aconstant right-to-left intracardiac shunt.  相似文献

Putative chemoreceptors in the solitary complex (SC) are sensitive to hypercapnia and oxidative stress. We tested the hypothesis that oxidative stress stimulates SC neurons by a mechanism independent of intracellular pH (pHi). pHi was measured by using ratiometric fluorescence imaging microscopy, utilizing either the pH-sensitive fluorescent dye BCECF or, during whole cell recordings, pyranine in SC neurons in brain stem slices from rat pups. Oxidative stress decreased pHi in 270 of 436 (62%) SC neurons tested. Chloramine-T (CT), N-chlorosuccinimide (NCS), dihydroxyfumaric acid, and H2O2 decreased pHi by 0.19 ± 0.007, 0.20 ± 0.015, 0.15 ± 0.013, and 0.08 ± 0.002 pH unit, respectively. Hypercapnia decreased pHi by 0.26 ± 0.006 pH unit (n = 95). The combination of hypercapnia and CT or NCS had an additive effect on pHi, causing a 0.42 ± 0.03 (n = 21) pH unit acidification. CT slowed pHi recovery mediated by Na+/H+ exchange (NHE) from NH4Cl-induced acidification by 53% (n = 20) in -buffered medium and by 58% (n = 10) in HEPES-buffered medium. CT increased firing rate in 14 of 16 SC neurons, and there was no difference in the firing rate response to CT with or without a corresponding change in pHi. These results indicate that oxidative stress 1) decreases pHi in some SC neurons, 2) together with hypercapnia has an additive effect on pHi, 3) partially inhibits NHE, and 4) directly affects excitability of CO2/H+-chemosensitive SC neurons independently of pHi changes. Acclimation of NO3 transport fluxes (influx, efflux)in roots of oilseed rape (Brassica napus L. cv. Bien venu) andtheir sensitivity to growth at low root temperature was studiedin relation to external NO3 supply, defined by constantconcentrations ranging from sub- to supra-optimal with respectto plant growth rate. Plants were grown from seed in flowingnutrient solutions containing 250 mmol m–3 NO3at 17°C for 20d, and solution temperature in half the cultureunits was then lowered decrementally over 3 d to 7°C. Threedays later plants were supplied with NO3 at 1, 10, 100or 1000 mmol m–3 maintained for 18 d. Dry matter productionwas decreased more by low root zone temperature than low [NO3]e. Root specific growth rates were inversely related to [NO3]eand shoot:root ratios increased with time at [NO3]e between10–1000 mmol m–3. Net uptake of NO3 at 17°Cwas twice that at 7°C, and at both temperatures it doubledwith increasing [NO3]e between 1–10 mmol m–3with further small increases at higher [NO3]e. Mean unitabsorption rates of NO3 between 0–6 d and 6–14d were linearly related (r2 of 0.79–0.99) to log10[NO].Steady-state Q10 (7–17°C) for uptake between 0–6d were 0.91, 1.62, 1.27, and 1.10, respectively, at [NO3]eof 1, 10, 100, and 1000 mmol m–3, compared with correspondingvalues of 0.98, 1.38, 1.68, and 1.89 between 6–14 d. Thedata indicated that net uptake rates at 7 and 17°C divergedover time at high [NO3]e. Short-term uptake rates from1 mol m–3 NO3 measured at 17°C were higherin plants grown with roots at 7°C than at 17°C; for7°C plants there was a strong inverse linear relationship(r2=0.94) between uptake rate and treatment log10 [NO3]ewhilst rates in 17°C plants were independent of prior [NO3]e. Rates of NO3 influx and efflux under different steady-stateconditions of NO3 supply and root temperature were calculatedfrom dilution of 15N added to culture solutions. Efflux wassubstantial relative to net uptake in all treatments, and wasinversely related to [NO3]e at 17°C but not at 7°C.Ratios of influx: efflux ranged from 1.6–2.9 at 17°Cand 1.3–1.8 at 7°C, indicating the proportionatelygreater impact of efflux at low root temperature. Ratios ofefflux: net uptake were 0.53–1.56 at 17°C and 1.21–3.58at 7°C. The apparent sensitivities of influx and effluxto steady-state root temperature varied with [NO3]e.Both fluxes were higher at 17°C than 7°C in the presenceof 100–1000 mmol m–3 NO3 but the trend wasreversed at 1–10 mmol m–3 NO. Concentrations oftotal N measured in xylem exudate were at least 2-fold higherat 7°C compared with 17°C, attributable mainly to higherconcentrations of NO3 glutamine and proline. The resultsare discussed in terms of acclimatory and other responses shownby the NO3 transport system under conditions of limitingNO3 supply and low root temperature. H+ translocation driven by NO3, NO2 and N2O reductionswith endogenous substrates in cells of Rhodopseudomonas sphaeroidesforma sp. denitrificans was investigated by the oxidant pulsemethod. Upon injection of nitrogenous oxides to anaerobic cellsin darkness, an alkaline transient in the external medium wasobserved, followed by acidification. The alkaline transientwas enhanced by carbonyl cyanide m-chlorophenylhydrazone. When a viologen dye was used as an electron donor in the presenceof 1 mM Af-ethylmaleimide and 0.1 mM 2-n-heptyl-4-hydroxyquinoline-N-oxideto preclude respiration-linked H+ extrusion, addition of KNO3,KNO2 and N2O caused only a rapid alkalinization. The H+ consumptionstoichiometries, H+/2e ratios for NO3 reductionto NO2, NO2 reduction to 1/2 N2O and N2O reductionto N2 were –1.90, –3.18 and –2.04, respectively.These values agreed well with the fact that all reductions ofnitrogenous oxides in denitrification occur on the periplasmicside of the cytoplasmic membrane. The effects of root temperature and form of N nutrition (NH4or NOJ) on the mineral composition of the plant, the balanceof inorganic cation-anion uptake and on the apparent net effluxof H +/OHions from the roots were studied with 49-d-oldoilseed rape (Brassica napusL. cv. Bien venu) in flowing solutionculture. Plants were pre-treated for 14 d at a root temperatureof 5 °C prior to constant root temperatures of 3, 7, 11or 17°C for 14 d, with a common shoot temperature of 20/15°Cday/night. Nitrogen was supplied as NH+44 or NO3 at 10mmol m3. Values of Q10 (7-17°C) for mean unit absorptionrates of all the major nutrient ions (K+ , Mg++ , NH+4, SO4,H2PO4, NO3), except Ca++, were > 2.0 over the first 5 d oftreatment but thereafter were < 1.5; the apparent effectof temperature on uptake rates diminished with time. Under NH+4nutrition, inorganic cation uptake (Mg+ + + K++Ca+ + +NH+4)exceeded inorganic anion uptake (SO44+ H2PO4) over 14d at all temperatures, with the proportion of cation uptakeas NH4 remaining constant (0.67-0-68) irrespective of root temperature.The net efflux of H + from the roots approximately balancedNH+4 uptake (1:1) over 14 d at each temperature and also balancedthe difference between the total uptake of inorganic cationsand inorganic anions. Under NO3 nutrition, the sum of the netefflux of OH and the change in the carboxylate contents of plantsover 14 d approximately balanced the sum of NO3 and SO4reduced in the plant. The majority of the negative charge associatedwith the reduction of NO3 and SO4 was apparently effluxedas OH, but this fraction was lower at low root temperatures.The results are discussed in terms of mechanisms that have beenproposed to regulate the internal pH of plants. The net efflux of H+ from lucerne (Medicago saliva L.), redclover (Trifolium pratense L.) and white clover (Trifolium repensL.) growing in flowing solution culture and dependent upon symbioticfixation of atmospheric N, was measured over a 75 d experimentalperiod. Considerable and rapid increases in acidity of the nutrientsolution of up to 1.45 pH units were recorded when the pH wasriot held constant over a 30 h period. There was little differencein H+ efflux when solution pH was held constant at 4.75, 5.75or 6.75, but there was an immediate cessation when it was adjustedto 3.75. Differences in the daily net efflux of H+ closely followedthe pattern of daily differences in incoming radiation, andthere was also evidence of a diurnal pattern of H+ efflux. Althoughthere were initially distinct differences between the speciesin the calculated rate of net H+ efflux (µg H+ g–1dry shoot d), by day 75 these had diminished. In allspecies, however, the maximum rate of efflux per unit of shootsoccurred during the earlier rapid phases of growth. The measuredefflux of H+ was well equated with the plant content of excesscations (as measured by ash alkalinity) and, on average, theratio of acidity produced to N assimilated (expressed as anequivalent) was 0-24. Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown for 71 d in flowing nutrient solutions containingN as 10 mmol m–3 NH4NO3, under artificial illumination,with shoots at 20/15°C day/night temperatures and root temperaturereduced decrementally from 20 to 5°C. Root temperatureswere then changed to 3, 7, 9, 11, 13, 17 or 25°C, and theacquisition of N by N2 fixation, NH4+ and NO3 uptakewas measured over 14 d. Shoot specific growth rates (d. wt)doubled with increasing temperature between 7 and 17°C,whilst root specific growth rates showed little response; shoot:root ratios increased with root temperature, and over time at11°C. Net uptake of total N per plant (N2 fixation + NH4++ NO3) over 14 d increased three-fold between 3 and 17°C.The proportion contributed by N2 fixation decreased with increasingtemperature from 51% at 5°C to 18% at 25°C. Uptake ofNH4+ as a proportion of NH4+ + NO3 uptake over 14 d variedlittle (55–62%) with root temperature between 3 and 25°C,although it increased with time at most temperatures. Mean ratesof total N uptake per unit shoot f. wt over 14 d changed littlebetween 9 and 25°C, but decreased progressively with temperaturebelow 9°C, due to the decline in the rates of NH4+ and NO3uptake, even though N2 fixation increased. The results suggestthat N2 fixation in the presence of sustained low concentrationsof NH4+ and NO4 is less sensitive to low root temperaturethan are either NH4+ or NO3 uptake systems. The herbicide D5 (N-n-pentyl-N-methyl-N'-(3, 4-dichlorophenyl)-ureacan uncouple oxidative phosphorylation in isolated plant mitochondria.This paper confirms that D5 is an uncoupler that catalyzes thecollapse of the transmembrane potential gradient by inducinga movement of protons across the membrane. However, D5 is notitself capable of transporting protons. D5 gives complete uncouplingat 40 µM, a lower concentration than that required foruncoupling by the n-butyl homologue ‘neburon’. Analysisof the shape of the state 4 stimulation curve suggests thatD5 might act as a dimer in the membrane. Attempts to demonstrate binding of D5 to a membrane target gaveambiguous results, binding is not evident at 10 °C and 25°C but might occur at 15 °C and 20 °C. The calculatedherbicide concentration in the membrane (40 µM of which4–0 µM is as the dimer) is high and similar to thatof the major phospholipids. The calculated partition coefficientbetween medium and membrane (3.8 x 103) is in agreement withthe lipophilicity of the substituted urea herbicides. In the presence of a substrate, D3 blocks both influx and effluxcalcium movement through the mitochondrial membrane but in theabsence of substrate, D5 induces binding of calcium. Bindingrequires Mg++ but not K+ or phosphate and leads to a releaseof H+. Ruthenium Red causes a partial inhibition of bindingbut no other reagent or ionophore tested had any effect. Sincebinding does not occur in turnip mitochondria which are unableto transport Ca++ it is concluded that the effect is not directlylinked to the uncoupling action. The mechanism of action of D5 is discussed and it is concludedthat D5 probably acts as a dimer and perturbs membrane structure.The site of action is probably the lipid components of the membrane.
