Background

Residual alcohol effects on physiological and psychological symptoms are commonly experienced the morning after alcohol consumption. The purpose of this study was to assess the effects of L-ornithine on subjective feelings and salivary stress markers the morning after alcohol consumption and to investigate whether L-ornithine acutely accelerates ethanol metabolism.

Methods

This study had a randomized, placebo-controlled, double-masked crossover design. Subjects were all healthy Japanese adults with the ‘flusher’ phenotype for alcohol tolerance. In experiment 1, 11 subjects drank 0.4 g/kg body weight alcohol 1.5 h before their usual bedtime. Half an hour after drinking, they ingested either a placebo or 400 mg ornithine. The next morning on awakening, subjects completed a questionnaire containing a visual analog scale (VAS), the Oguri-Shirakawa-Azumi sleep inventory MA version (OSA-MA), and a profile of mood states (POMS) and collected a saliva sample for measurement of salivary stress markers (cortisol, secretory immunoglobulin A, and α-amylase). In experiment 2, placebo or 400 mg ornithine were administrated to 16 subjects both before and after drinking, and the feeling of drunkenness, breath ethanol concentration and one-leg standing time were repeatedly investigated until 180 min after alcohol consumption.

Results

There were significant decreases in “awareness”, “feeling of fatigue” and “lassitude” VAS scores and in “anger-hostility” and “confusion” POMS scores and a significant increase in “sleep length” in the OSA-MA test. Salivary cortisol concentrations on awakening were reduced after ornithine supplementation. There were no differences between ornithine and placebo in any of the subjective or physiological parameters of acute alcohol metabolism.

Conclusions

Taking 400 mg ornithine after alcohol consumption improved various negative feelings and decreased the salivary stress marker cortisol the next morning. These effects were not caused by an increase in acute alcohol metabolism.

α-Amylase, which plays an essential role in starch degradation, is expressed mainly in the pancreas and salivary glands. Human α-amylase is also detected in other tissues, but it is unclear whether the α-amylase is endogenously expressed in each tissue or mixed exogenously with one expressed by the pancreas or salivary glands. Furthermore, the biological significance of these α-amylases detected in tissues other than the pancreas and salivary glands has not been elucidated. We discovered that human α-amylase is expressed in intestinal epithelial cells and analyzed the effects of suppressing α-amylase expression. α-Amylase was found to be expressed at the second-highest messenger RNA level in the duodenum in human normal tissues after the pancreas. α-Amylase was detected in the cell extract of Caco-2 intestinal epithelial cells but not secreted into the culture medium. The amount of α-amylase expressed increased depending on the length of the culture of Caco-2 cells, suggesting that α-amylase is expressed in small intestine epithelial cells rather than the colon because the cells differentiate spontaneously upon reaching confluence in culture to exhibit the characteristics of small intestinal epithelial cells rather than colon cells. The α-amylase expressed in Caco-2 cells had enzymatic activity and was identified as AMY2B, one of the two isoforms of pancreatic α-amylase. The suppression of α-amylase expression by small interfering RNA inhibited cell differentiation and proliferation. These results demonstrate for the first time that α-amylase is expressed in human intestinal epithelial cells and affects cell proliferation and differentiation. This α-amylase may induce the proliferation and differentiation of small intestine epithelial cells, supporting a rapid turnover of cells to maintain a healthy intestinal lumen.  相似文献

The hypoglycaemic effects of two Ribes sp. i.e., anthocyanin-rich black currants (BC) were compared to green currants (GC), which are low in anthocyanins to establish which compounds are involved in the regulation of postprandial glycaemia. We determined the effect of the currants on inhibiting carbohydrate digestive enzymes (α-amylase, α-glucosidase), intestinal sugar absorption and transport across CaCo-2 cells. The digestion of these currants was modelled using in vitro gastrointestinal digestion (IVGD) to identify the metabolites present in the digested extracts by LC–MS/MS. Freeze-dried BC and IVDG extracts inhibited yeast α-glucosidase activity (P<.0001) at lower concentrations than acarbose, whereas GC and IVDG GC at the same concentrations showed no inhibition. BC and GC both showed significant inhibitory effects on salivary α-amylase (P<.0001), glucose uptake (P<.0001) and the mRNA expression of sugar transporters (P<.0001). Taken together this suggests that the anthocyanins which are high in BC have their greatest effect on postprandial hyperglycaemia by inhibiting α-glucosidase activity. Phytochemical analysis identified the phenolics in the currants and confirmed that freeze-dried BC contained higher concentrations of anthocyanins compared to GC (39.80 vs. 9.85 g/kg dry weight). Specific phenolics were also shown to inhibit salivary α-amylase, α-glucosidase, and glucose uptake. However, specific anthocyanins identified in BC which were low in GC were shown to inhibit α-glucosidase. In conclusion the anthocyanins in BC appear to regulate postprandial hyperglycaemia primarily but not solely by inhibiting α-glucosidase while other phenolics modulate salivary α-amylase, glucose uptake and sugar transporters which together could lower the associated risk of developing type-2 diabetes.  相似文献

Activity of α-amylase was revealed in the midgut and salivary glands of the wheat and barley pentatomid pest, A. acuminata. The activity was determined in salivary gland more than those in midgut. Optimal activity of the enzyme occurred at 40°C. Optimal pH activity in salivary gland (pH = 6) was more than those in the midgut (pH = 4.5). pH stability analysis of the enzyme showed that the enzyme is more stable at slightly acidic pHs than those at acidic and alkaline pHs. However, α-amylase is more stable at acidic pH in long period of time. Temperature stability analysis determined the enzyme was remarkably active over a broad range of temperature (5–40°C). α-Amylase activity was decreased after addition of MgCl2, Tris, Triton X-100, CuSO4, SDS, urea and CaCl2. The salts NaCl and KCl increased the enzyme activity from midgut and salivary glands. Zymogram analysis of midgut and salivary gland extract showed at least two bands of amylase activity in the midgut and salivary glands.  相似文献

Strategies to manage the symptoms of exercise-induced muscle damage (EIMD) are widespread, though are often based on anecdotal evidence. The aim of this study was to determine the efficacy of a combination of manual massage and compressive clothing and compressive clothing individually as recovery strategies after muscle damage. Thirty-two female volunteers completed 100 plyometric drop jumps and were randomly assigned to a passive recovery (n = 17), combined treatment (n = 7), or compression treatment group (n = 8). Indices of muscle damage (perceived soreness, creatine kinase activity, isokinetic muscle strength, squat jump, and countermovement jump performance) were assessed immediately before and after 1, 24, 48, 72, and 96 hours of plyometric exercise. The compression treatment group wore compressive tights for 12 hours after damage and the combined treatment group received a 30-minute massage immediately after damaging exercise and wore compression stockings for the following 11.5 hours. Plyometric exercise had a significant effect on all indices of muscle damage (p < 0.05). The treatments significantly reduced decrements in isokinetic muscle strength, squat jump performance, and countermovement jump performance and reduced the level of perceived soreness in comparison with the passive recovery group (p < 0.05). The addition of sports massage to compression after muscle damage did not improve performance recovery, with recovery trends being similar in both treatment groups. The treatment combination of massage and compression significantly moderated perceived soreness at 48 and 72 hours after plyometric exercise (p < 0.05) in comparison with the passive recovery or compression alone treatment. The results indicate that the use of lower limb compression and a combined treatment of manual massage with lower limb compression are effective recovery strategies following EIMD. Minimal performance differences between treatments were observed, although the combination treatment may be beneficial in controlling perceived soreness.  相似文献

The aim of this study was to investigate the differences in the length-dependent changes in quadriceps muscle torque during voluntary isometric and isokinetic contractions performed after severe muscle-damaging exercise. Thirteen physically active men (age = 23.8 ± 3.2 years, body weight = 77.2 ± 4.5 kg) performed stretch-shortening cycle (SSC) exercise comprising 100 drop jumps with 30-second intervals between each jump. Changes in the voluntary and electrically evoked torque in concentric and isometric conditions at different muscle lengths, muscle soreness, and plasma creatine kinase (CK) activity were assessed within 72 hours after SSC exercise. Isokinetic knee extension torque decreased significantly (p < 0.05) at all joint angles after SSC exercise. At 2 minutes and at 72 hours after SSC exercise, the changes in knee torque were significantly smaller at 80° (where 180° = full knee extension) than at 110-130°. At 2 minutes after SSC exercise, the optimal angle for isokinetic knee extension torque shifted by 9.5 ± 8.9° to a longer muscle length (p < 0.05). Electrically induced torque at low-frequency (20-Hz) stimulation decreased significantly more at a knee joint angle of 130° than at 90°. The subjects felt acute muscle pain and CK activity in the blood increased to 1,593.9 ± 536.2 IU·L?1 within 72 hours after SSC exercise (p < 0.05). This study demonstrates that the effect of muscle-damaging exercise on isokinetic torque is greatest for contractions at short muscle lengths. These findings have practical importance because the movements in most physical activities are dynamic in nature, and the decrease in torque at various points in the range of motion during exercise might affect overall performance.  相似文献

从野生苋属植物 (Amaranthuspaniculatus)籽实中分离纯化出α淀粉酶的一种新型蛋白质类抑制剂 .该抑制剂被命名为WAI 1 .MALDI TOF质谱测得其分子量为 986 5 ,是目前报道的α 淀粉酶的蛋白质类抑制剂中分子量最小的 .初步的组成和结构分析结果表明 ,WAI 1由 9个氨基酸残基组成 ,其N端为焦谷氨酸 .直接用RP HPLC纯化后 ,WAI 1能在弱酸性条件下 ,以非竞争性抑制作用方式有效抑制美洲蜚蠊消化道α淀粉酶的活性 ,最适抑制pH 6 0 ,但对人唾液淀粉酶活性无影响 .WAI 1在 37℃下与酶预温浴约 30min后显示最大抑制活性 .当α淀粉酶用量一定时 ,α淀粉酶活性的抑制率在约 5 0 %的范围内随抑制剂 酶比例的增大而呈线性增加 ,超过 5 0 %后 ,抑制率随抑制剂 酶比例的增大而缓慢上升 ,最终达到最大值 (约 6 5 % ) .  相似文献

Reduction of saliva immunoglobulin levels by swim training   总被引:3,自引:0,他引:3  
Saliva immunoglobulin A (IgA) and cortisol levels were measured in 21 male members of a major midwestern swim team. Saliva samples were collected before and after training sessions four times during the fall season; the training intensity was light, moderate, heavy and during the taper period before a major competitive meet. Saliva IgA levels were decreased after each training session, reaching statistical significance with the moderate training intensity. Over the 3-month training period the pre-session and post-session IgA levels both decreased significantly during the heavy and taper training intensities later in the fall season. Cortisol levels were significantly elevated only after the heavy-intensity training session. The Profile of Mood States (POMS) was used to assess the swimmers' overall mood on each test day. No significant correlations were found between the global POMS score and IgA or cortisol. Also, cortisol and IgA were not significantly correlated except after the light training session. Results from this study indicate that acute bouts of exercise can reduce salivary IgA levels and that chronic exercise of high intensity can reduce the resting levels of IgA. These changes may render the athletes more vulnerable to respiratory infections after exercise and even at rest during the later stages of the competitive season.  相似文献

A trypsin inhibitor, isolated from whole-wheat grain (Triticum aestivum L.) by the method of biospecific chromatography on trypsin-Sepharose, was potent in inhibiting human salivary α-amylase. The bifunctional α-amylase/trypsin inhibitor was characterized by a narrow specificity for other α-amylases and proteinases. The high thermostability of the inhibitor was lost in the presence of SH group-reducing agents. The inhibitor-trypsin complex retained its activity against α-amylase. The inhibitor—α-amylase complex was active against trypsin. Studies of the enzyme kinetics demonstrated that the inhibition of α-amylase and trypsin was noncompetitive. Our results suggest the existence of two independent active sites responsible for the interaction with the enzymes.  相似文献

分析不同基因型小麦品种逆境萌发过程中种子萌发相关酶活性及基因表达差异,明确在逆境条件下,种子活力与种子萌发相关酶活性及基因表达量的关系.通过标准发芽试验和逆境(冷浸、人工老化、干旱胁迫)发芽试验,测定4个小麦品种种子活力、萌发过程中可溶性总糖和可溶性蛋白含量、α-淀粉酶活性、半胱氨酸蛋白酶活性及相关基因表达量.结果表明:干旱、人工老化和冷浸胁迫3种逆境对种子活力都有一定影响.不同萌发条件下,可溶性总糖含量呈现先小幅度升高后小幅度降低再迅速升高的趋势;而可溶性蛋白含量随着萌发时间的延长呈现逐渐下降的趋势.α-淀粉酶活性整体呈现逐渐升高的趋势,但在冷浸胁迫处理后,豫农949和轮选061的α-淀粉酶活性在萌发60 h后出现下降.半胱氨酸蛋白酶活性整体呈先降低后升高的趋势,但在干旱胁迫条件下,豫农949、豫麦49-198和轮选061的半胱氨酸蛋白酶活性呈现先升高后降低再升高的趋势.不同逆境萌发条件下,α-AMY(α-淀粉酶基因)表达量整体呈先上升后下降的趋势.冷浸胁迫处理后,轮选061的α-AMY表达量高于对照,在其他逆境萌发条件下,4个品种的α-AMY表达量均低于对照;人工老化处理后,长4738的CP(半胱氨酸蛋白酶基因)表达量与对照差异不显著,在其他逆境萌发条件下,4个品种的CP表达量均高于对照.种子萌发期间,不同萌发条件下α-淀粉酶和半胱氨酸蛋白酶活性与其基因表达并没有直接关系,α-淀粉酶活性与可溶性总糖含量达到显著正相关,半胱氨酸蛋白酶活性与可溶性蛋白含量的相关性不显著.在标准发芽条件下,α-淀粉酶活性与活力指数呈显著正相关,而在逆境萌发过程中,其相关性不显著.冷浸胁迫处理后,半胱氨酸蛋白酶活性与活力指数呈显著正相关,但在标准发芽、干旱胁迫、人工老化处理后,其相关性不显著.  相似文献

研究了不同酶反应缓冲体系、pH值、氯离子浓度以及噁唑哒嗪对5龄2日粘虫 Pseudaletia separata Walker 中肠α-淀粉酶活性的影响。结果表明,乙酸-乙酸钠缓冲体系(pH 5.8)和磷酸氢二钠-磷酸二氢钠缓冲体系(pH 8.0)有利于增强α-淀粉酶活性,比活力最高分别达到4.49和4.97。在乙酸-乙酸钠缓冲体系(pH 5.8)中,5、10、20、40和80 mmol/L氯离子浓度引起α-淀粉酶活性呈现先减弱后增强的变化规律,而在磷酸氢二钠-磷酸二氢钠缓冲体系(pH 8.0)中仅呈现减弱的趋势。1.4 mmol/L噁唑哒嗪对α-淀粉酶活性的抑制率可达70%,但抑制程度随着反应体系中蛋白含量的增加而逐渐降低。  相似文献

The participation of various isoenzymes of α-amylase in the formation of falling number values of the commodity grain of wheat grown in the Republic of Kazakhstan was investigated. It was found that active isoenzymes α-AMY1 and α-AMY2 of the embryonic shield were present in the grain with an index over 200. A significant decrease in the falling number depended mainly on the synthesis of α-AMY1 and α-AMY2 isoenzymes in the aleurone layer. In the grain, isoenzymes with high isoelectric points (p1 ≥ 7.3) were found these isoenzymes belong to α-amylase or late maturing or α-amylase of practically mature grains. It was discovered that the exogenous hormone (gibberellic acid) induced synthesis of α-amylase isoenzymes of scutellum, whole caryopses, and aleurone. It was shown that the impact of exogenous gibberellic acid on the activity and structure of α-amylase is reduced in grain with a low falling number.  相似文献

We investigated seasonal changes in blood concentrations of interleukin-6 (IL-6), adrenocorticotrophic hormone (ACTH), metabolites of catecholamine (VMA, HVA, and 5-HIAA) and cortisol in humans. Eight volunteers were investigated at four times during the year (February, May, August and October) at latitude 35° N. The mean ambient temperature at the collection periods was higher in the order of summer > autumn ≈ spring > winter. Changes in mood were also monitored by a profile of mood states (POMS) questionnaire. The concentration of IL-6 was significantly higher in winter and summer than in spring and autumn. The concentrations of ACTH, HVA and VMA were significantly higher in summer. No seasonal variation was detected in cortisol. There were significant differences among the seasons in subscale tension and anger in the POMS questionnaire; the tension subscale showed significant differences between spring and autumn, with a higher score in spring. The results demonstrate that Il-6, ACTH, HVA and VMA exhibit statistically significant seasonal rhythms, which might have important diagnostic and therapeutic implications.  相似文献

The pistachio green stink bug, Brachynema germari, has 3–5 generations per year and causes severe damages to pistachio crops in Iran. Physiological digestive processes, such as digestive carbohydrases, can be used to design new strategies in IPM programs for controlling this pest. The enzyme α-amylase digests starch during the initial stage of digestion. Complete breakdown of carbohydrates takes place in the midgut where α- and β-glucosidic activities are highest. Alpha-amylase and α- and β-glucosidase activities were found in the midgut and salivary glands of pistachio green stink bug adults. Overall enzyme activities were significantly higher in the midgut than in salivary glands. The highest α-amylase and α- and β-glucosidase activities were in section v3, whereas the lowest activities were in section v4. Vmax was higher and Km was lower in the midgut than in the salivary glands for these enzymes. In the pistachio green stink bug, the optimal pH was pH 5–6.5 and the optimal temperature was 30 °C to 35 °C for these enzymes. Alpha-amylase activity in the midgut and salivary glands decreased as the concentrations of MgCl2, EDTA and SDS increased. Enzyme activities in both midgut and salivary glands increased in the presence of NaCl, CaCl2, and KCl. NaCl had a negative effect on alpha-amylase extracted from salivary glands.  相似文献

The purpose of this study was to determine the effects of environmental cooling on force production in the quadriceps and hamstring muscles. Ten men (mean +/- SD: age = 21.4 +/- 2.2 years, height = 168.5 +/- 35.9 cm, body mass = 78.0 +/- 6.4 kg) participated in this study. On March 11, 2011, the Pacific side of Japan's northeast was devastated by an earthquake and tsunami. For years, many researchers have been working on ways of examining the psychological effects of earthquakes on survivors in disaster areas who have experienced aftershocks, catastrophic fires, and other damage caused by the earthquake. The goal of this study is to examine scores on psychological measures and salivary cortisol level in these individuals both before and three months after the earthquake. The participants had been measured for these variables before the earthquake. After the earthquake, we carried out PTSD screening using CAPS for participants for another experiment, and then again conducted the aforementioned tests. We collected saliva samples from all survivors. Our results show that social relationship scores on the WHO-QOL26, negative mood scores of the WHO-SUBI, total GHQ score, POMS confusion scores, and CMI emotional status score after the earthquake showed scores indicating significantly decreased compared to before the earthquake. On the other hand, salivary cortisol levels after the earthquake was significantly increased compared to before the earthquake. Moreover, the result of a multiple regression analysis found that negative mood score on the WHO-SUBI and social relationship score on the WHO-QOL26 were significantly related to salivary cortisol levels. Our results thus demonstrate that several psychological stress induced by the earthquake was associated with an increase in salivary cortisol levels. These results show similar findings to previous study. We anticipate that this study will provide a better understanding of posttraumatic responses in the early stages of adaptation to the trauma and expand effective prevention strategies and countermeasures for PTSD.  相似文献

We have determined the nucleotide sequence of two different mouse α-amylase mRNAs, one found in the pancreas and the other in the salivary gland. The 1577 and 1659 nucleotide mRNAs from pancreas and salivary gland, respectively, are the major α-amylase species which accumulate in each tissue. Differences in mRNA length are primarily in the 5′ noncoding regions. Comparable portions of the mRNAs are 89% homologous. The mRNA sequences predict α-amylase precursor proteins of 508 and 511 amino acid residues, accounting for nearly the entire coding capacity of the mRNAs; differences in protein length occur as a result of a nine nucleotide segment present within the translated portion of salivary gland, but not pancreas, mRNA. The lengths and amino acid compositions of the predicted proteins concur with those determined empirically by others. These proteins differ 12% in amino acid sequence, explaining previously observed differences in net charge and antigenic properties. Finally, translation of the salivary gland α-amylase mRNA is not initiated at the AUG codon nearest the 5′ terminus since that codon is almost immediately followed by the termination triplet UAA. This observation may have implications for the mechanism of translation initiation in eucaroytes.  相似文献

Human salivary α-amylase (HSAmy) has three distinct functions relevant to oral health: (1) hydrolysis of starch, (2) binding to hydroxyapatite (HA), and (3) binding to bacteria (e.g., viridans streptococci). Although the active site of HSAmy for starch hydrolysis is well-characterized, the regions responsible for bacterial binding are yet to be defined. Since HSAmy possesses several secondary saccharide-binding sites in which aromatic residues are prominently located, we hypothesized that one or more of the secondary saccharide-binding sites harboring the aromatic residues may play an important role in bacterial binding. To test this hypothesis, the aromatic residues at five secondary binding sites were mutated to alanine to generate six mutants representing either single (W203A, Y276A, and W284A), double (Y276A/W284A and W316A/W388A), or multiple [W134A/W203A/Y276A/W284A/W316A/W388A; human salivary α-amylase aromatic residue multiple mutant (HSAmy-ar)] mutations. The crystal structure of HSAmy-ar as an acarbose complex was determined at a resolution of 1.5 Å and compared with the existing wild-type acarbose complex. The wild-type and the mutant enzymes were characterized for their abilities to exhibit enzyme activity, starch-binding activity, HA-binding activity, and bacterial binding activity. Our results clearly showed that (1) mutation of aromatic residues does not alter the overall conformation of the molecule; (2) single or double mutants showed either moderate or minimal changes in both starch-binding activity and bacterial binding activity, whereas HSAmy-ar showed significant reduction in these activities; (3) starch-hydrolytic activity was reduced by 10-fold in HSAmy-ar; (4) oligosaccharide-hydrolytic activity was reduced in all mutants, but the action pattern was similar to that of the wild-type enzyme; and (5) HA binding was unaffected in HSAmy-ar. These results clearly show that the aromatic residues at the secondary saccharide-binding sites in HSAmy play a critical role in bacterial binding and in starch-hydrolytic functions of HSAmy.  相似文献

The transglycosylation activity of barley α-amylase 1 (AMY1) and active site AMY1 subsite mutant enzymes was investigated. We report here the transferase ability of the V47A, V47F, V47D and S48Y single mutants and V47K/S48G and V47G/S48D double mutant AMY1 enzymes in which the replaced amino acids play important role in substrate binding at subsites at −3 through −5. Although mutation increases the transglycosylation activity of enzymes, in the presence of acceptors the difference between wild type and mutants is not so significant. Oligomer transfer reactions of AMY1 wild type and its mutants were studied using maltoheptaose and maltopentaose donors and different chromophore containing acceptors. The conditions for the chemoenzymatic synthesis of 4-methylumbelliferyl-α-d-maltooligosaccharides (MU-α-d-MOSs) were optimized using 4-methylumbelliferyl-β-d-glucoside as acceptor and maltoheptaose as donor. 4-Methylumbelliferyl-α-d-maltoside, -maltotrioside, -maltotetraoside and -maltopentaoside have been synthesized. Products were identified by MALDI-TOF MS. 1H and 13C NMR analyses showed that AMY1 V47F preserved the stereo- and regioselectivity. The produced MU-α-d-MOSs of degree of polymerization DP 2, DP 3 and DP 5 were successfully applied to detect activity of Bacillus stearothermophilus maltogenic α-amylase, human salivary α-amylase and Bacillus licheniformis α-amylase, respectively in a fast and simple fluorometric assay.  相似文献

Seven times over the 24-h period, developed torque and myoelectric activity were estimated during maximal isometric voluntary contractions using an isokinetic dynamometer: elbow flexion for EAS in standardised sleep, and knee extension for EPS in complete sleep deprivation. The results showed nycthemeral changes in torque in both conditions (p < 0.005), with maximal values recorded at the beginning of night. Although during sleep deprivation (EPS) the rhythm followed neurophysiological factors, during EAS, this rhythm was accounted for by the variations in the contractile state of muscle.  相似文献
