将6 - 羟基多巴胺(6 - OHDA) 分两点注入SD大鼠左侧中脑黑质内侧端,制成震颤麻痹模型,毁损后6 ~8 周,用胚龄14~16 天同种胚鼠中脑黑质细胞悬液植入模型鼠尾壳核。实验分正常对照组,模型对照组,单纯胚中脑黑质移植组(MS组) ,含层粘蛋白Laminin 胚中脑黑质移植组(LMS组) ,含雪旺氏细胞胚中脑黑质移植组(SMS组) 。于移植后2 ~4 个月分别测试旋转试验,MS组、LMS组及SMS组与模型组间均有显著差异(P< 0-01) 。动物经测试后处死。脑切片观察发现,损伤侧中脑黑质DA神经元95% 以上死亡,移植组织位于尾壳核背侧或突入侧脑室,LMS组移植区> SMS组移植区> MS组移植区。三个移植区均见TH、GABA 免疫阳性细胞,其形态及大小与正常对照侧中脑黑质所见相似。LMS组与SMS组多巴胺阳性细胞突起较MS组多且长。本实验证明中脑黑质移植物Laminin 与雪旺氏细胞均能促进DA能神经元突起生长。  相似文献

目的:检测NCAM.140在多巴胺能神经细胞损伤过程中中脑黑质部位的表达变化,探索其在多巴胺能神经细胞损伤中的可能作用.方法:采用MPTP腹腔注射建立小鼠中脑黑质多巴胺能神经细胞慢性损伤模型,采用免疫荧光染色和westernblotting检测中脑黑质部酪氨酸羟化酶(TH)和NCAM-140的表达情况.结果:在小鼠中脑黑质一直有NCAM-140阳性细胞存在,且与TH阳性细胞共表达;TH在给予MPTP后第三周表达开始降低,NCAM-140在给予MPTP后第一周至第三周持续高表达,并且从第四周开始,其表达量明显降低.结论:NCAM-140的表达与多巴胺能神经细胞损伤有关.  相似文献

目的观察6-羟多巴胺(6-OHDA)单侧注射制备的帕金森病(PD)大鼠多巴胺(DA)能神经元的超微结构改变。方法单侧微量注射6-OHDA制备PD大鼠模型,用免疫荧光组织化学方法观察正常侧与6-OHDA注射侧黑质酪氨酸羟化酶(TH)阳性神经细胞及神经纤维的变化;并利用免疫电镜技术观察大鼠正常侧与注射侧黑质致密部DA能神经元的超微结构。结果免疫荧光法显示注射侧黑质致密部TH阳性细胞数和网状部TH阳性纤维面积与正常侧的百分比平均值分别为21.83%,23.19%。免疫电镜显示:TH免疫反应阳性产物表达于PD大鼠正常侧DA能神经元的高尔基复合体质膜面及胞质内,电子密度较高,注射侧很少见或几乎未见,且注射侧线粒体嵴有不同程度的溶解,呈空泡样变或髓样变,粗面内质网脱颗粒。结论6-OHDA可引起DA能神经元发生凋亡的超微结构改变。  相似文献

采用免疫组织化学SABC法,研究白介素-1α、干扰素-γ、神经生长因子-β和肿瘤坏死因子-α在成体中华蟾蜍脑中的表达和分布特点。结果发现,白介素-1α阳性细胞数量很多,分布于脑的各个区域。白介素-1α多在细胞的胞体中,而原始海马锥体细胞,中脑的背前侧被盖核和腹后侧被盖核中的细胞可见阳性的突起。干扰素-γ阳性细胞数量较多,分布在端脑的原始海马和隔区,丘脑腹外侧核,下丘脑的视前区、视交叉上核和腹侧漏斗核,中脑被盖的背前侧被盖核、腹前侧被盖核、背后侧被盖核和腹后侧被盖核中,小脑的Purkinje细胞层和延髓的网状核,其中原始海马,背前侧被盖核和背后侧被盖核,视交叉上核,Purkinje细胞层和网状核中的细胞中可见阳性突起。神经生长因子-β阳性细胞数量较少,主要存在于下丘脑的视前区和视交叉上核,中脑被盖的腹前侧被盖核,小脑的Purkinje细胞层和延髓的网状核中,其中视前区、Purkinje细胞层和网状核中细胞可见阳性突起。肿瘤坏死因子-α阳性细胞数量最少,分布范围仅限于中脑被盖背前侧区和延髓的网状核及中缝核,但细胞具有阳性突起。因此,白介素-1α和干扰素-γ在成体动物脑中分布较为广泛,可能是神经细胞生命活动所必需的;而神经生长因子-β和肿瘤坏死因子-α在成体动物脑中分布范围狭窄,其作用可能仅限于脑中的某些特殊区域。  相似文献

目的:为了检测RET在多巴胺能神经细胞损伤过程中中脑黑质部位的表达变化,并探索其在多巴胺能神经细胞损伤中的可能作用.方法:本实验将C57BL/6 d、鼠分为三组:MPTP组、NS组和空白对照组,采用MPTP腹腔注射建立小鼠中脑黑质多巴胺能神经细胞慢性损伤模型,设立第一周到第六周六个时间点(1w-6w),检测小鼠行为学变化,并采用免疫荧光染色和western blotting等方法检测中脑黑质部酪氨酸羟化酶(TH)和PET的表达情况.结果:行为学结果显示,随给药次数及时间延长,悬挂实验中,MPTP组悬挂评分逐渐下降,MPTP组第三周给药以后与NS组比较差异有统计学意义(P<0.05);跳台实验中,小鼠受电击后跳上跳台的时间逐渐延长,错误次数逐渐增多.免疫荧光双标结果显示,在检测的各时间点,在小鼠中脑黑质一直有RET阳性细胞存在,且与TH阳性细胞共表达;western blotting结果显示,TH在给予MPTP后第三周表达开始降低,RET在给予MPTP后第一周和第二周持续高表达,并且也从第三周开始,其表达量明显降低.结论:这种表达变化提示RET的表达与多巴胺能神经细胞损伤有关.  相似文献

采用 HRP逆行追踪结合谷氨酸免疫组织化学方法观察大鼠外侧膝状体背侧核 (d L GN)中继神经元的化学递质。光镜下 HRP标记细胞与谷氨酸免疫阳性细胞清晰可辩。HRP单标记细胞位于外侧膝状体背侧核内 ,胞浆及树突基部充满棕色颗粒。免疫金银法 (IGSS)单标记的谷氨酸免疫阳性神经元分布于外侧膝状体背侧核与腹侧核 ,胞体内充满黑色银颗粒。在外侧膝状体背侧核内可见 HRP和谷氨酸双标记细胞 ,其数目占 HRP标记细胞总数的 70 .9± 6 .4%。本文提示 ,谷氨酸可能是外侧膝状体背侧核投射至视皮质的中继神经元的神经递质之一。  相似文献

用依赖还原型辅酶Ⅱ的黄酶组织化学方法,研究了环节动物门寡毛纲种类河北环毛蚓(Pheretima tschiliensis)神经系统k 一氧化氮合酶(NOS)阳性细胞及阳性纤维的分布,结果表明,河北环毛蚓神经系统中脑神经节背侧有大量细胞呈现NO强阳性反应,胞体和突起染色明显。咽下神经中偶尔能见少数染色较浅的神经元。在脑神经节腹内侧、围咽神经、 咽下神经节外侧部及腹神经链中都有一氧化氮合酶阳性纤维存在脸染色很深,实验结果表明,在环节动物中作为信息分子的一氧化氮已广泛存在于神经系统中。  相似文献

采用HE染色和Holmes银染法对蟾蜍中脑的显微结构进行了研究.中脑背侧,视叶可分为顶盖和被盖,顶盖从外侧到内侧依次分为:带状层、外灰质层、浅白质层、中灰质层、中白质层、深灰质层、深白质层和中央灰质.被盖前端分层与顶盖相同,后端分层不明显.中脑腹侧包括被盖和大脑脚,HE染色和Holmes银染法显示,大脑脚从外向内颜色由浅变深,存在大量纵向神经纤维束,两脚底分界处有横向交错的神经纤维.被盖外侧细胞不分层,聚集形成核团.被盖内侧,细胞和纤维以中脑水管为中心,呈同心圆环分8层.通过比较蟾蜍中脑背腹差异程度,了解背腹功能不同.同时对中华蟾蜍中脑同其他脊椎动物的进行了比较.  相似文献

目的 探索一种新的胚胎大鼠腹侧中脑黑质细胞的混合培养方法,以获得具有高比例多巴胺神经元的原代细胞体系,有利于在体外条件下进行帕金森病(Parkinson's disease,PD)的发病机制和防治的研究.方法 分离E15 (Embryonic Day 15)SD胎鼠中脑黑质区域组织,分散为单细胞后,分别用DMEM/F12+ 10%FBS含血清培养基和Neurobasal+ N1无血清培养基设置不同组别进行培养,通过免疫组织化学方法检测在不同培养条件下细胞的生长状态及多巴胺能神经元的比例.结果 DMEM/F12+ 10% FBS与Neurobasal+ N1先后一周交替换液的的培养体系中获得的TH阳性神经元占神经元的比例可达31%左右,明显高于单独使用DMEM/F12+ 10% FBS培养组(约10%).结论 DMEM/F12+ 10% FBS与Neurobasal +N1先后一周交替换液是一种黑质神经元和胶质细胞混合培养并能获得高比例多巴胺神经元的有效培养方法.  相似文献

The chemical differentiation featured by the appearance of tyrosine hydroxylase (TH) and the distribution pattern of the dopaminergic cells of rat substantia nigra (SN) were studied with combined immunocytochemical and electronmicroscopic techniques. Under the light microscope, the earliest TH-positive cells at embryonic day 13 are localized at the ventral part of rostral midbrain. Later appearing TH-positive cells join the earlier ones dorsally and caudally. As to the stain intensity and morphology of the labeled cells in the region of the SN, there exists a ventral to dorsal and lateral to medial spatiotemporal gradient, namely the cells in the ventral and lateral parts, compared with the dorsal and medial ones, have more intense staining, larger cell bodies with smaller nuclei and more and longer processes. The earliest nigrostriatal projection fibers stem from the most laterally located SN cells. Under electron microscope, rough endoplasmic reticula are always seen within the positively stained cells. With the progression of development, the cells show more intense staining and contain more rough endoplasmic reticula and other organelles. Together with the results reported on the neurogenesis and migration of the SN cells, the present study indicates that the chemical differentiation of SN cells, with a spatiotemporal gradient, starts after the completion of cell migration, a process paralleling to their morphological differentiation.  相似文献

The preoptic area/anterior hypothalamus (POA/AH) sits as a boundary region rostral to the classical diencephalic hypothalamus and ventral to the telencephalic septal region. Numerous studies have pointed to the region's importance for sex‐dependent functions. Previous studies suggested that migratory guidance cues within this region might be particularly unique in their diversity. To better understand the early development and differentiation of the POA/AH, cytoarchitectural, birthdate, immunocytochemical, and cell migration studies were conducted in vivo and in vitro using embryonic C57BL/6J mice. A medial preoptic nucleus became discernible using Nissl stain in males and females between embryonic days (E) E15 and E17. Cells containing immunoreactive estrogen receptor‐α were detected in the POA/AH by E13, and increased in number with age in both sexes. From E15 to E17, examination of the radial glial fiber pattern by immunocytochemistry confirmed the presence of dual orientations for migratory guidance ventral to the anterior commissure (medial‐lateral and dorsal‐ventral) and uniform orientation more caudally (medial‐lateral). Video microscopy studies followed the migration of DiI‐labeled cells in coronal 250‐μm brain slices from E15 mice maintained in serum‐free media for 1–3 days. Analyses showed significant migration along a dorsal‐ventral orientation in addition to medial‐lateral. The video analyses showed significantly more medial‐lateral migration in males than females in the caudal POA/AH. In vivo, changes in the distribution of cells labeled by the mitotic indicator bromodeoxyuridine (BrdU) suggested their progressive migration through the POA/AH. BrdU analyses also indicated significant movement from dorsal to ventral regions ventral to the anterior commissure. The significant dorsal‐ventral migration of cells in the POA/AH provides additional support for the notion that the region integrates developmental information from both telencephalic and diencephalic compartments. The sex difference in the orientation of migration of cells in the caudal POA/AH suggests one locus for the influence of gonadal steroids in the embryonic mouse forebrain. © 1999 John Wiley & Sons, Inc. Structural differentiation of the guard cells of Vigna sinensis results from the integration of the following interrelated processes: a) intense activity of ribosomes, dictyosomes, endoplasmic reticulum (ER) membranes and mitochondria and patterned organization of microtubules; b) unequal thickening and ordered micellation of their walls and opening of the stomatal pore; and c) the divergent differentiation of the plastids. In differentiating guard cells, microtubules appear anticlinally oriented and more or less evenly distributed along the unthickened part of the dorsal wall and in the middle part of the ventral wall where thickening of the future pore occurs. In periclinal walls, microtubules fan away from the margins of the increasing thickening of the ventral wall and, later, from the rims of the stomatal pore towards the dorsal walls, parallel to the depositing radial microfibrils. Microtubules may be the cytoplasmic elements underlying guard-cell morphogenesis. Although cell-plate organization in guard-cell mother cells does not seem to differ from that of other protodermal cells, the middle lamella of the ventral wall becomes electron-translucent. The stomatal pore develops schizogenously from the internal and/or external ends of the ventral wall and proceeds inwards, remaining incomplete in most of the stomata of plants grown for 30 days in darkness and in some malformed ones which were developed after a prolonged action of colchicine. The guard cell, when approaching maturity, loses its organelle complexity and plasmodesmata, but it keeps a significant portion of its cytoplasm and organelles. Perigenous stomata generally exceed the size of mesoperigenous and mesogenous ones, develop large vacuoles and appear able to induce oriented divisions in their vicinity.  相似文献

应用PAP-PAAP双重免疫组化染色程序在同一切片上进行两种肽能物质的定位,观察了中国树鼩下丘脑视上核和室旁核内VP能和OT能神经元的比较解剖学分布,发现:视上核被视束分成主部和交叉后部。在视上核主部,其头侧部几乎仅含OT能神经元胞体,中间部VP能胞体出现并逐渐增多,尾侧部VP能胞体数目明显超过OT能胞体。在明显含有两种胞体的中间部和尾侧部,OT能胞体多位于背内侧,VP能胞体多位于腹外侧;在视上核交叉后部,其头侧部以VP能胞体为主,且多位于背外侧,OT能胞体多位于腹内侧。中间部OT能胞体多位于内侧,VP能胞体多位于外侧。尾侧部OT能胞体多位于背、腹两侧,VP能胞体则多位于中间;在室旁核,其头侧部几乎全由OT能胞体构成。中间部,VP能胞体出现并逐渐增多,OT和VP能胞体分别主要位于内、外侧。尾侧部两种神经元胞体较明显地分为内、外两群,内侧群主要为OT能胞体,外侧群几乎全为VP能胞体,该群的头侧半又可分为背腹两个亚群,至尾侧半,此二亚群渐合并。本文讨论了OT和VP能神经元在中国树鼩和大鼠视上核和室旁核内的比较分布。  相似文献

Specific antibodies against different opioid peptides derived from proenkephalin were used in light and electron microscopic studies to locate septal enkephalin-containing cells. Immunoreactive neurons were demonstrated only after pretreatment of the animals with colchicine. They were found in all three subdivisions of the lateral septal nucleus and in the ventral limb of the nucleus of the diagonal band. The medial septal nucleus and the dorsal limb of the nucleus of the diagonal band were devoid of immunoreactive cells. Electron microscopy showed intracellular enkephalin-like immunoreactivity with all antisera used in this study. The reaction was found in the cytoplasm, sometimes associated with the rough endoplasmic reticulum. Numerous enkephalin-immunoreactive nerve terminals and fibres were detected in the lateral septal nucleus, but axon terminals making contacts with enkephalin-immunoreactive neurons did not contain enkephalin-like immunoreactivity. The results suggest that some septal neurons synthesise proenkephalin. These cells may be either local interneurons or output cells to areas which receive innervation from the septal complex.  相似文献

应用免疫细胞化学 ABC 技术观察了催产素、精氨酸加压素、促肾上腺皮质激素释放激素、神经肽 Y、神经降压肽、P 物质及亮氨酸脑啡肽免疫反应阳性纤维在大鼠结合臂旁核中的分布。催产素阳性纤维稀少,分布于腹外侧亚核、外外侧亚核、背外侧亚核及外外侧亚核与背外侧亚核之间的移行区。加压素阳性纤维亦甚为稀少,分布于腹外侧亚核、背外侧亚核、外外侧亚核与极外侧亚核.促肾上腺皮质激素释放激素阳性纤维分布于尾侧部臂旁核腰区、外外侧亚核腹内侧部、极外侧亚核、背外侧亚核、中央外侧亚核、内外侧亚核及臂旁内侧核。神经肽 Y 阳性纤维大多为串珠状的终末祥结构,分布于尾侧部腹外侧亚核、背外侧亚核、外外侧亚核背外侧部及外内侧亚核,在上外侧亚核、结合臂背内侧端背侧及臂旁内侧核腹侧部也有少量分布.神经降压肽阳性纤维分布于尾侧部臂旁核腰区、背外侧亚核、背外侧亚核与外外侧亚核之间的移行区、外外侧亚核、外内侧亚核及中吻部臂旁内侧核腹侧部。P 物质及亮氨酸脑啡肽阳性纤维分布于所有的臂旁外侧核诸亚核及外内侧亚核。  相似文献

为探讨魟类螫伤机理,用光镜和电镜观察了光魟的毒棘腹外侧纵沟内的毒腺的组织结构。结果表明:光魟的毒腺为复层上皮。从基底面到游离面依次为基底层、棘细胞层和嗜酸性细胞层。基底层细胞和棘层的细胞嗜碱性,棘细胞间有少量、散在分布的嗜酸性细胞;棘细胞的外层至腺上皮的游离面的嗜酸性细胞密集排列。电镜下可见基底细胞有丰富的核糖体。棘细胞内粗面内质网、高尔基复合体等较丰富。嗜酸性细胞内有电子密度低的膜包分泌颗粒;接近表面的细胞内颗粒部分融合;表层细胞核消失,胞质充满融合的颗粒,游离面侧的胞膜呈角质样增厚。腺上皮内还可见到黑色素细胞、郎格罕细胞和梅克尔细胞。提示毒腺组织内有两种类型的细胞,一类为毒液形成细胞,另一类为非毒液形成细胞。嗜酸性细胞内的电子密度低的膜包分泌颗粒成分可能是造成螫伤剧痛及全身症状的关键因素。  相似文献

This paper describes the morphology of the male cloacal protuberance (CP) of the polygynandrous Alpine Accentor Prunella collaris , with special reference to intense sperm competition in the mating system. The fully developed CP consisted of dorsal (DL) and ventral (VL) lobes. The DL was found to be a massive part formed by a highly convoluted extension of the distal vas deferens with a fibro-muscular sheath. Here, the vas deferens contained abundant spermatozoa, round cells of unknown origin, and secretory droplets in the tubular lumen. The epithelium of the vas deferens was lined with columnar cells that showed numerous microvilli and apocrine processes in their periluminal portion, frequent engulfing of spermatozoa, and many mitochondria and vesicular endoplasmic reticula in their cytoplasm. The VL was a smaller subdivision distal to the DL and further connected to a terminal papilla. The VL was organized similarly to the DL, but showed different features: it lay beneath the thick skin; the fibro-muscular sheath was more muscular; the epithelial cells were cuboid and showed poorly developed microvilli; and the figures of secretion and engulfed spermatozoa were infrequent. Thus, the CP of the male Alpine Accentor showed a clear regional differentiation, i.e. the DL for storing/maintaining spermatozoa and the VL for storing/ejaculating spermatozoa.  相似文献

The localization of nitric oxide synthase (NOS) in vascular endothelial cells of submucosal blood vessels from the guinea-pig ileum was examined using NADPH diaphorase histochemistry at the light microscopic level, and endothelial NOS immunohistochemistry at the light and electron microscopic level. The pattern of staining observed following NADPH diaphorase histochemistry and endothelial NOS immunohistochemistry was identical. Endothelial cells of the arterioles, capillaries and venules showed small patches of intense, perinuclear staining. Under the electron microscope, endothelial NOS immunoreactivity was found predominantly in association with the Golgi apparatus and with the membranes of some vesicles. Small regions of the plasma membrane and the rough endoplasmic reticulum also showed some immunoreactivity. The presence of NOS in the Golgi apparatus and in vesicles raises the possibility that NOS may be exteriorized by endothelial cells, and hence that nitric oxide is synthesized extracellularly.  相似文献

In order to understand better the organisation of the ventral lateral geniculate nucleus of the ventral thalamus, this paper has examined the patterns of connections that this nucleus has with various nuclei of the dorsal thalamus in rats. Injections of biotinylated dextran or cholera toxin subunit B were made into the parafascicular, central lateral, posterior thalamic, medial dorsal, lateral dorsal, lateral posterior, dorsal lateral geniculate, anterior, ventral lateral, ventrobasal and medial geniculate nuclei of Sprague-Dawley rats and their brains were processed using standard tracer detection methods. Three general patterns of ventral lateral geniculate connectivity were seen. First, the parafascicular, central lateral, medial dorsal, posterior thalamic and lateral dorsal nuclei had heavy connections with the parvocellular (internal) lamina of the ventral lateral geniculate nucleus. This geniculate lamina has been shown previously to receive heavy inputs from many functionally diverse brainstem nuclei. Second, the visually related dorsal lateral geniculate and lateral posterior nuclei had heavy connections with the magnocellular (external) lamina of the ventral lateral geniculate nucleus. This geniculate lamina has been shown by previous studies to receive heavy inputs from the visual cortex and the retina. Finally, the anterior, ventral lateral, ventrobasal and medial geniculate nuclei had very sparse, if any, connections with the ventral lateral geniculate nucleus. Overall, our results strengthen the notion that one can package the ventral lateral geniculate nucleus into distinct visual (magnocellular) and non-visual (parvocellular) components.  相似文献
