Ferritins from the liver and spleen of the cold-adapted Antarctic teleosts Trematomus bernacchii and Trematomus newnesi have been isolated and characterized. Interestingly, only H- and M-chains are expressed and no L-chains. The H-chains contain the conserved ferroxidase center residues while M-chains harbor both the ferroxidase center and the micelle nucleation site ligands. Ferritins have an organ-specific subunit composition, they are: M homopolymers in spleen and H/M heteropolymers in liver. The M-chain homopolymer mineralizes iron at higher rate with respect to the H/M heteropolymer, which however is endowed with a lower activation energy for the iron incorporation process, indicative of a higher local flexibility. These findings and available literature data on ferritin expression in fish point to the role of tissue-specific expression of different chains in modulating the iron oxidation/mineralization process.  相似文献

The physiological response and ultrastructure of the pigment cells of Trematomus bernacchii, an Antarctic teleost that lives under the sea ice north of the Ross Ice Shelf, were studied. In the integument, two types of epidermal chromatophores, melanophores and xanthophores, were found; in the dermis, typically three types of chromatophores--melanophores, xanthophores, and iridophores--were observed. The occurrence of epidermal xanthophore is reported for the first time in fish. Dermal melanophores and xanthophores have well-developed arrays of cytoplasmic microtubules. They responded rapidly to epinephrine and teleost melanin-concentrating hormone (MCH) with pigment aggregation and to theophylline with pigment dispersion. Total darkness elicited pigment aggregation in the majority of dermal xanthophores of isolated scales, whereas melanophores remained dispersed under both light and dark conditions. Pigment organelles of epidermal and dermal xanthophores that translocate during the pigmentary responses are carotenoid droplets of relatively large size. Dermal iridophores containing large reflecting platelets appeared to be immobile.  相似文献

The cathodic hemoglobin component of the Antarctic fish Trematomus newnesi (HbCTn) is a Root-effect protein. The interpretation of its functional properties in relation to its sequence is puzzling. Indeed, HbCTn sequence is characterized by an extremely low histidyl content, and in particular by the lack of His146beta and His69beta, which are believed to be important in Bohr and Root effects, respectively. Furthermore, previous analyses suggested that the local environment of Asp95alpha, Asp99beta, and Asp101beta should not be appropriate for the formation of Asp-Asp interactions, which are important for the Root effect. Here, we report the high-resolution crystal structure of the deoxy form of HbCTn. Our data provide a structural interpretation for the very low oxygen affinity of the protein and insights into the structural determinants of the Root effect protein. The structure demonstrates that the presence of Ile41alpha and Ser97alpha at the alpha1beta2 interface does not prevent the formation of the inter-Asp interactions in HbCTn, as previous studies had suggested. The present data indicate that the hydrogen bond formed between Asp95alpha and Asp101beta, which is stabilized by Asp99beta, is per se sufficient to generate the Root effect, and it is the minimal structural requirement needed for the design of Root-effect Hbs.  相似文献

Species that live in extreme conditions have specially adapted physiology and tissue/organ organisation. The adaptation of lymphoid organs to low temperatures in polar species could be an original field of study, indicating how the immune system works under extreme conditions. In fishes, the head kidney is a key organ for immunity and here the cytology of this organ is studied in two common Antarctic species: Trematomus bernacchii and Chionodraco hamatus. Ultrastructural analysis revealed heterogeneity of epithelial cells, with reticular cells, subcapsular- and perivascular-limiting cells. Differences in the size and morphology of epithelial cells were observed between the polar species and warm water species of fish. Intermingled with epithelial cell leucocytes, such as lymphocytes, thrombocytes and macrophages, had comparable morphology in both species, contrary to sharp differences observed in the morphology of erythrocytes and granulocytes. The functional adaptation of the head kidney to the low temperatures of polar water is discussed.  相似文献

The presence and localization of α- and β-spectrins and of the spectrin cross-linking protein actin were investigated, in previtellogenic oocytes of three species of Antarctic teleosts: the two red-blooded nototheniids, Trematomus bernacchii and Trematomus newnesi , and the channichthyid, the ice fish Chionodraco hamatus . Analyses by western blotting indicated that these species had an unusual abundance of spectrin isoforms and that they were characterized by rather low molecular masses. The immunocytochemistry in situ demonstrated that α- and β-spectrins showed a variable pattern of localization that clearly depended on both the species considered and the stage of oocyte differentiation. In particular, the two Trematomus spp. showed a distribution of spectrins absolutely comparable and rather different from that of C. hamatus . The evidences collected confirmed that channichthyids have isolated early from the group of red-blooded species and suggest that in notothenioids significant changes might have occurred in spectrin genes and in their protein products.  相似文献

The response properties of anterior lateral line afferent neurones in Trematomus bernacchii were recorded extracellularly while stimulating the fish with unidirectional water flows of varying velocity. Afferent neurone responses were either flow-sensitive or flow-insensitive. Flow-sensitive neurones showed linear increases in response magnitude with increasing flow rate and tonic non-adapting response properties. These findings indicate that flow-sensitive afferent neurones originate from lateral line receptors that detect absolute flow velocity. The likely explanation is that flow-sensitive afferent neurones innervate neuromasts located superficially on the skin and flow-insensitive neurones innervate neuromasts situated in sub-epidermal fluid-filled canals.  相似文献

The Root effect describes the drastic drop of oxygen affinity and loss of cooperativity at acidic pH expressed in the hemoglobins (Hb) of certain fish. The comparison between the deoxy structures of the Root effect Hb from the Antarctic fish Trematomus bernacchii (HbTb) at different pHs (pH = 6.2 and pH = 8.4) shows that the most significant differences are localized at the CDα region, where a salt bridge between Asp48 and His55 breaks during the low-to-high pH transition. In order to shed light on the relationship between pH, CDα loop structure and dynamics, and oxygen access to the active site in the alpha chain of HbTb, different computer simulation techniques were performed. Our results highlight the importance of the protonation of His55 in regulating oxygen access, underscoring its pivotal role in the structural and functional properties of HbTb. These data provide further support to the hypothesis that this residue might contribute to the release of Root protons in HbTb and underline the fact that an efficient transport of molecular oxygen in Hbs relies on a subtle balance of tertiary structure and protein conformational flexibility.  相似文献

Accessory corner cones (ACC) have recently been suggested to be UV-sensitive photoreceptor cells. With a view toward explaining prey detection, we examined the topography of retinal ganglion cells and ACCs in two Antarctic nototheniids occupying different ecological niches: the cryopelagic Pagothenia borchgrevinki and the benthic Trematomus bernacchii. Isodensity maps of retinal ganglion cells showed that the main visual axis, coincident with the feeding vector, was in a forward direction in both species. Visual acuity was determined as 3.64 and 4.77 cycles/degree for the respective species. In P. borchgrevinki the highest density of ACCs was associated with the eye's main visual axis. This suggested that this species uses UV-vision during forward-swims and probably in encounters with prey. On the other hand, T. bernacchii possessed two horizontal band-shaped high-density areas of ACCs, which stretched from temporal to nasal and ventral to peripheral retinal regions. Therefore, this species appears to use UV-vision to watch prey across the entire circumference of the lateral area and in the water column above its head.  相似文献

Six polymorphic microsatellites were isolated from the dusky notothen, Trematomus newnesi (Boulenger 1902), using a microsatellite enrichment protocol and selective hybridization with di‐ tetra‐ and penta‐repeat probes. The loci were screened in 48 individuals captured in the Southern Ocean (coastal zone of Terre‐Adélie), revealing eight to 22 alleles per locus and an observed heterozygosity ranging from 0.70 to 0.92. These microsatellite markers provide a tool to study the relationship between the various morphs observed in this species and can be used for population genetic analysis and biodiversity studies.  相似文献

Lymphomyeloid organs of the Antarctic fish Trematomus bernacchii were studied with the aim of analysing some morphological aspects related to adaptation to low environmental temperature. The thymus of T. bernacchii was flattened, incompletely lobated and scarcely regionalised. It was filled by lymphoid elements intermingled with stromal elements. The head kidney appeared highly vascularised and mainly lymphopoietic. The spleen appeared mainly erythropoietic, with scarcely developed areas of white pulp. In spite of the unique conditions they have to operate under, the pineal organs of Antarctic fishes have not previously been examined. We determined immunohistochemically that in the end-vesicles and the pineal stalks of Pagothenia borchgrevinki (a species found directly beneath the sea-ice) as well as Trematomus bernacchii (a species preferring somewhat deeper water than the former) at least two populations of physiologically-different cells occurred that displayed reactions indicative of typical vertebrate photoreceptors. Comparisons with immunocytochemically treated retinal sections from the eyes of the same two species showed that anti-opsin reactivity, characteristic of rods, was particularly strong in the lumina of the pineal stalks of both species. Anti-visinin reactions stained cones in the retinal sections of both fishes and occurred throughout the pineal organs, but in particular in the end vesicles of the pineals of both species. The difference in preferred habitat depth between the two species appears to have had very little influence on both retinal and pineal immunocytochemistry. It is concluded that the pineal organs of both species, at least during the austral summer, exhibit signs of being directly photo-sensitive.  相似文献

The Antarctic notothenioid Trematomus bernacchii (rock cod) lives at a constant mean temperature of -1.9 degrees C. Gastric digestion under these conditions relies on the proteolytic activity of aspartic proteases such as pepsin. To understand the molecular mechanisms of Antarctic fish pepsins, T. bernacchii pepsins A1 and A2 were cloned, overexpressed in Escherichia coli, purified and characterized with a number of biochemical and biophysical methods. The properties of these two Antarctic isoenzymes were compared to those of porcine pepsin and found to be unique in a number of ways. Fish pepsins were found to be more temperature sensitive, generally less active at lower pH and more sensitive to inhibition by pepstatin than their mesophilic counterparts. The specificity of Antarctic fish pepsins was similar but not identical to that of pig pepsin, probably owing to changes in the sequence of fish enzymes near the active site. Gene duplication of Antarctic rock cod pepsins is the likely mechanism for adaptation to the harsh temperature environment in which these enzymes must function.  相似文献

Trematomus newnesi is a common inshore species with a circum-Antarctic distribution. It provides the only known example of phenotypic plasticity in Antarctic notothenioid fish, existing as populations of typical, large mouth and intermediate morphs that can be difficult to identify. Using specimens from both Potter Cove, King George/25 de Mayo Island, and from McMurdo Sound, we found that the morphometric measurements gape width/head length (HL), upper jaw length/HL and, to a lesser extent, orbit diameter/HL reliably separated the morphs. For use in a key, we converted the ratios into the qualitative characters head shape, head width and upper jaw length relative to middle of the eye. To increase the reliability of the key, we also assessed intra-morph variability in these characters. The key is supplemented with colour photographs illustrating the distinctive features for separation of the morphs. We discovered that, in the case of the specimens from Potter Cove, each morph had a distinct pattern of colouration: typical—trunk blotched, with yellow or orange-brown predominating especially on pectoral and caudal fins; large mouth—trunk blotched, with green predominating especially in pectoral and opercular regions; and intermediate—trunk less blotched, with homogeneous dark brown-grey on trunk, pectoral and caudal fins. Na+-K+-Cl- cotransport and Na+/K+ATPase were studied by immunohistochemistry in the kidney and urinary bladder of Trematomus bernacchii and Chionodraco hamatus. The activity was correlated to the density of mitochondria. The first segment of the renal proximal tubule was more active than the second one. In T. bernacchii and the temperate marine teleost Pagellus bogaraveo, the immunoreactivity for the antibody to cotransporters and to the !-subunit of the sodium pump was stronger than in the icefish. This difference indicates in the kidney of the icefish, a weaker secretory activity, a consequent lower osmolarity in the lumen and lower water loss, which correlates well with the need for a greater blood volume in the icefish. The epithelium of the urinary bladder in T. bernacchii, where intense immunostaining was observed, was composed of columnar cells. In C. hamatus the columnar cells, where the immunostaining was weaker, lined only a portion of the urinary bladder, the other region being composed of cuboidal cells.  相似文献

Little information is available regarding fish diets in Antarctica and how they relate to prey availability. The primary objective of this work was to describe the diet of Trematomus bernacchii. The second objective was to compare prey taken with prey present in the benthos along a spatial gradient of prey abundance using Ivlev's Index of Electivity. All samples were collected from 4 different sites on the east side of McMurdo Sound, Antarctica. Fish were captured by divers at the same depth at each site and their stomachs were flushed for dietary analysis. The diet of T. bernacchii varied among sites, and prey selectivity varied inversely with prey abundance. Many of the prey taken by T. bernacchii were sedentary species suggesting that T. bernacchii is a hunt and peck predator.  相似文献

1. The effects of temperature on the catalytic and regulatory properties of pyruvate kinases from the temperate-zone rainbow trout and the Antarctic fish Trematomus bernacchii were examined. 2. The K(m) value of pyruvate kinase for one of its two substrates, phosphoenolpyruvate, is temperature-dependent, and is lowest at temperatures that closely coincide with the habitat temperatures of the two fishes. 3. Two regulatory functions of pyruvate kinase, feedforward activation by fructose diphosphate and feedback inhibition by ATP, are temperature-independent. Enzyme-ADP interaction is also temperature-independent. 4. It is concluded that enzyme-substrate and enzyme-modulator interactions are important factors in short-term and in evolutionary adaptations by poikilotherms to changes in temperature. Though the K(m) for substrate may vary in apparently adaptive manners, the regulatory functions of an enzyme appear to be unchanged over the range of temperatures experienced by the organism in Nature.  相似文献

We investigated the occurrence of antibodies against protein antigens of the nematode parasite Pseudoterranova decipiens in the plasma and bile of the Antarctic teleost Trematomus bernacchii. Three different P. decipiens protein solutions were prepared: excreted/secreted proteins from live larvae (ESP); surface-associated proteins obtained by mild extraction of larval bodies (SAP); and cuticular soluble proteins recovered by extraction in strong reducing conditions (CSP). Using different immunoassays, these 3 preparations were tested for their ability to bind fish antibody. As determined by ELISA, the specific antibody binding activity was higher in SAP than in CSP. As determined by dot-blot immunoassay, the specific antigen binding activity versus SAP was higher in bile than in plasma antibodies. A different number of antigenic components of SAP and ESP were identified by immunoblotting performed with plasma or bile antibodies. These results led to the conclusion that T. bernacchii parasitism by nematodes involves plasma and bile anti-parasite antibodies. Furthermore bile antibodies were found to be more reactive and more heterogeneous than plasma.  相似文献

The liganded hemoglobin (Hb) high-salt crystallization condition described by Max Perutz has generated three different crystals of human adult carbonmonoxy hemoglobin (COHbA). The first crystal is isomorphous with the "classical" liganded or R Hb structure. The second crystal reveals a new liganded Hb quaternary structure, RR2, that assumes an intermediate conformation between the R form and another liganded Hb quaternary structure, R2, which was discovered more than a decade ago. Like the R2 structure, the diagnostic R state hydrogen bond between beta2His97 and alpha1Thr38 is missing in the RR2 structure. The third crystal adopts a novel liganded Hb conformation, which we have termed R3, and it shows substantial quaternary structural differences from the R, RR2, and R2 structures. The quaternary structure differences between T and R3 are as large as those between T and R2; however, the T --> R3 and T --> R2 transitions are in different directions as defined by rigid-body screw rotation. Moreover, R3 represents an end state. Compared to all known liganded Hb structures, R3 shows remarkably reduced strain at the alpha-heme, reduced steric contact between the beta-heme ligand and the distal residues, smaller alpha- and beta-clefts, and reduced alpha1-alpha2 and beta1-beta2 iron-iron distances. Together, these unique structural features in R3 should make it the most relaxed and/or greatly enhance its affinity for oxygen compared to the other liganded Hbs. The current Hb structure-function relationships that are now based on T --> R, T -->R --> R2, or T --> R2 --> R transitions may have to be reexamined to take into account the RR2 and R3 liganded structures.  相似文献

Three-dimensional structural models of the hemoglobin (Hb 1) of the Antarctic fish Trematomus newnesi were built by homology modelling, using as template the X-ray crystallographic structures of Trematomus (previously named Pagothenia) bernacchii Hb 1, both in R and T state. The Hbs of these two fishes, although showing remarkably different oxygen binding properties, differ only by 4 residues in the alpha chain (142 aa) and 10 residues in the beta chain (146 aa). T. newnesi Hb1 R-state model, essentially performed as a quality control of the adopted modelling procedure, showed a good correspondence with the crystallographic one. Modelling of T. newnesi Hb1 in the T state was performed taking into account that the proton uptake by aspartate residues, proposed to be responsible for half of the Root effect in T. bernacchii Hb 1 (showing sharp pH dependent oxygen affinity and T-state overstabilization at low pH, i. e. Bohr and Root effect), does not occur in T. newnesi Hb1 (having nearly pH-independent lower oxygen affinity). Comparison with the template structure (submitted to the same minimization procedure) indicates that, in T. newnesi Hb1 T-state model, the substitution of Ile for Thr in 41 C6, in central position of the switch region, induces at the alpha(1)beta(2) interface structural modifications able to hamper the protonation. Similar modifications are also found in T. bernacchii Hb 1 modelled in the T state with the single substitution Thr-->Ile in 41alpha. These models also suggest that the lower oxygen affinity observed in T. newnesi Hb1 is related to structural differences at the alpha(1)beta(2) interface leading to a more stable low-affinity T state.
