We evaluated the differential nature of interactions between waterborne Ca and Cd transport in the gills of yellow perch (Perca flavescens) and rainbow trout (Oncorhynchus mykiss), two species with a more than 400-fold difference in acute waterborne Cd tolerance. The Jmax (maximum rate of uptake) and Km (inverse of affinity) for Ca uptake, in the absence of Cd, were significantly lower in yellow perch (120.48 nM g–1 wet wt h–1 and 92.17 M, respectively) relative to rainbow trout (188.68 nM g–1 wet wt h–1 and 243.90 M, respectively). Similarly, the Jmax for Cd uptake, at the lowest waterborne Ca level (100 M) tested, was significantly lower in yellow perch (0.27 nM g–1 wet wt h–1) relative to rainbow trout (0.40 nM g–1 wet wt h–1), but no significant difference was observed in the Km values between the two species (yellow perch: 32.47 nM; rainbow trout: 31.27 nM). Waterborne Cd (0–890 nM) as well as waterborne Ca (100–1,000 M) competitively inhibited branchial uptake of each other in both species. However, analyses of inhibitor constants for branchial Ca uptake by waterborne Cd ( ) revealed that the inhibition was about 1.8 times more potent in rainbow trout compared to yellow perch. In contrast, analyses of inhibitor constants for branchial Cd uptake by waterborne Ca ( ) indicated that the inhibition was more than three fold more potent in yellow perch than in rainbow trout. Higher branchial Ca uptake and more potent inhibition by Cd as well as higher branchial Cd uptake and less potent inhibition by Ca were also reflected in whole-body measurements of Ca and Cd influx in trout relative to perch. Overall, whole-body effects were in accord with the branchial kinetic analyses. These results further strengthen the conclusion that branchial influxes of Ca and Cd occur through common pathways. The biomagnification of methylmercury (MeHg) amongst trophic levels results in high levels of this compound in many freshwater fish species. The role of parasites in MeHg cycling and trophic transfer in freshwater systems is largely unknown. This study examined the potential for metacercariae of Apophallus brevis to accumulate and biomagnify MeHg from their second intermediate host, yellow perch, Perca flavescens. Contrary to our prediction that MeHg levels would be higher in parasites than in the host muscle tissue in which they are embedded, we found that concentrations were similar. The lack of increase in MeHg levels from host to parasite may be due to limited assimilation of host muscle tissue or, in part, to low parasite metabolism. Parasite load did not reduce fish growth and subsequently alter MeHg concentrations. This study suggests that relationships between larval parasites and their hosts do not conform to typical patterns of MeHg biomagnification seen in aquatic systems.  相似文献

We examined relationships among perch population variable parameters in two types of lakes, lakes with perch (P-lakes, n?=?15) and lakes with perch and roach (PR-lakes, n?=?10) using redundancy analysis (RDA) to relate population variables to environmental factors. Effects from environmental factors were tested for significance by means of permutation tests (Monte Carlo). Three factors, pH, altitude and fraction of roach (by number) in the gill net catches, explaining 47.9% of the variation, had significant effects on perch population variables. The significance of pH was improved by partialing out the effect of conductivity and roach. Similarly, the significance of altitude was improved by partialing out the effect of pH and roach, and the significance of roach was improved by partialing out the effect of pH and altitude. When the fraction of pike in the catch was included in the analysis, the effect of roach was not significant and vice-versa, as roach and pike fractions were correlated with each other. The effect of pike was significant when roach was not included, but the effect was not as strong as the effect of roach. A biplot was constructed by plotting population variables on the first and second RDA axis, with arrows showing five selected environmental factors. Growth of 3+ to 5+ perch was positively related to pH and altitude, perch catch per unit effort was negatively related to pH and altitude, and age specific perch weight was negatively related to fraction of roach. The relationship between growth of 2+ perch and pH was not as strong as the relationship between pH and the growth of older perch. Moreover, the growth of 2+ perch was negatively related to the fraction of roach, probably due to competition between young zooplankton feeding perch and roach.  相似文献

A hypothesis on change in prey size selectivity in relation to illumination level was tested on the basis of data on weight and size composition of the content of the digestive tract of larvae of perch Perca fluviatilis and on zooplankton in the layer 0–6 m (Wallersee Lake, Austria). Larvae foraging in the twilight-night period had almost two times more food in the intestine than those foraging in the daytime. The size composition of perch larvae and concentration and size composition of zooplankton hardly differed in the daytime and twilight-night samples. For the first time, it is shown on field material that more intensive feeding of larvae at twilight is related to selection of significantly larger prey than in daytime feeding. In the day-time, the larvae consumed more prey but their maximum size did not exceed 0.6 of the diameter of mouth opening of the fish; at twilight it was over 0.8. In case of feeding on so large prey, not only the weight of the consumed feed increases but the time used for capture and swallowing also considerably increases. The larvae which in the period of investigation did not yet form schools, which perform the principal defensive function, were especially vulnerable for predators feeding on relatively large prey. The decrease of the part of small-sized prey at twilight is not related to their lesser availability due to a low illumination. It is assumed that feeding on energetically more valuable but less available prey is shifted to the period of low illumination when the larvae are less exposed to predation risk. Two repeated DNA sequences isolated from a partial genomic DNA library of Helianthus annuus, p HaS13 and p HaS211, were shown to represent portions of the int gene of a Ty3 /gypsy retroelement and of the RNase-Hgene of a Ty1 /copia retroelement, respectively. Southern blotting patterns obtained by hybridizing the two probes to BglII- or DraI-digested genomic DNA from different Helianthus species showed p HaS13 and p HaS211 were parts of dispersed repeats at least 8 and 7 kb in length, respectively, that were conserved in all species studied. Comparable hybridization patterns were obtained in all species with p HaS13. By contrast, the patterns obtained by hybridizing p HaS211 clearly differentiated annual species from perennials. The frequencies of p HaS13- and p HaS211-related sequences in different species were 4.3x10(4)-1.3x10(5) copies and 9.9x10(2)-8.1x10(3) copies per picogram of DNA, respectively. The frequency of p HaS13-related sequences varied widely within annual species, while no significant difference was observed among perennial species. Conversely, the frequency variation of p HaS211-related sequences was as large within annual species as within perennials. Sequences of both families were found to be dispersed along the length of all chromosomes in all species studied. However, Ty3 /gypsy-like sequences were localized preferentially at the centromeric regions, whereas Ty1/ copia-like sequences were less represented or absent around the centromeres and plentiful at the chromosome ends. These findings suggest that the two sequence families played a role in Helianthusgenome evolution and species divergence, evolved independently in the same genomic backgrounds and in annual or perennial species, and acquired different possible functions in the host genomes.  相似文献

Body size of consumer species is a fundamental trait that influences the trophic ecology of individuals and their contribution to the functioning of freshwater ecosystems. However, the relationship between body size and trophic ecology can be highly variable both within and between closely-related and similarly-sized species. In this study we compared the intra- and interspecific relationship between body size and trophic position for North American Yellow Perch Perca flavescens and European Perch Perca fluviatilis, which share similarities in morphology, life history traits and trophic requirements. We used stable isotope ratios (δ15N and δ13C) to characterize differences in size-dependency of trophic position and to trace consumer foraging history of Yellow Perch in lakes in the Northwestern United States and European Perch in lakes in Germany. The trophic position and stable isotope ratios of Yellow Perch and European Perch steadily increased with total body length, but European Perch were consistently feeding at higher trophic positions than Yellow Perch at a given length. European Perch occupied considerably higher trophic positions (mean trophic position = 3.9) than Yellow Perch (mean trophic position = 2.8). Large European Perch were increasingly piscivorous, whereas large Yellow Perch were more opportunistic and omnivorous predators of invertebrate prey. Overall, the trophic position among individual Yellow Perch varied more strongly than in European Perch. We conclude that both species similarly increase in trophic position with size, but the specific size-dependency of both trophic position and resource use varies with taxonomy and local ecological conditions. Thus, body size as a sole measure of trophic position should be considered cautiously when generalizing across populations and species.  相似文献

New combinations are proposed in anticipation of the Polygonaceae treatment in the forthcoming volume of Intermountain Flora: Polygonum kelloggii var. esotericum, P. kelloggii var. watsonii , Rumex densiflorus var. pycnanthus , R. salicifolius var. utahensis, and R. occidentalis var. tomentellus. Studying Pneumocystis has proven to be a challenge from the perspective of propagating a significant amount of the pathogen in a facile manner. The study of several fungal pathogens has been aided by the use of invertebrate model hosts. Our efforts to infect the invertebrate larvae Galleria mellonella with Pneumocystis proved futile since P. murina neither caused disease nor was able to proliferate within G. mellonella. It did, however, show that the pathogen could be rapidly cleared from the host.  相似文献

A genetic transformation system has been developed for callus cells of Crataegus aronia using Agrobacterium tumefaciens. Callus culture was established from internodal stem segments incubated on Murashige and Skoog (MS) medium supplemented with 5 mg l−1 Indole-3-butyric acid (IBA) and 0.5 mg l−1 6-benzyladenine (BA). In order to optimize the callus culture system with respect to callus growth and coloration, different types and concentrations of plant growth regulators were tested. Results indicated that the best average fresh weight of red colored callus was obtained on MS medium supplemented with 2 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.5 mg l−1 kinetin (Kin) (callus maintenance medium). Callus cells were co-cultivated with Agrobacterium harboring the binary plasmid pCAMBIA1302 carrying the mgfp5 and hygromycin phosphotransferase (hptII) genes conferring green fluorescent protein (GFP) activity and hygromycin resistance, respectively. Putative transgenic calli were obtained 4 weeks after incubation of the co-cultivated explants onto maintenance medium supplemented with 50 mg l−1 hygromycin. Molecular analysis confirmed the integration of the transgenes in transformed callus. To our knowledge, this is the first time to report an Agrobacterium-mediated transformation system in Crataegus aronia.  相似文献

The maT clade of transposons is a group of transposable elements intermediate in sequence and predicted protein structure to mariner and Tc transposons, with a distribution thus far limited to a few invertebrate species. We present evidence, based on searches of publicly available databases, that the nematode Caenorhabditis briggsae has several maT-like transposons, which we have designated as CbmaT elements, dispersed throughout its genome. We also describe two additional transposon sequences that probably share their evolutionary history with the CbmaT transposons. One resembles a fold back variant of a CbmaT element, with long (380-bp) inverted terminal repeats (ITRs) that show a high degree (71%) of identity to CbmaT1. The other, which shares only the 26-bp ITR sequences with one of the CbmaT variants, is present in eight nearly identical copies, but does not have a transposase gene and may therefore be cross mobilised by a CbmaT transposase. Using PCR-based mobility assays, we show that CbmaT1 transposons are capable of excising from the C. briggsae genome. CbmaT1 excised approximately 500 times less frequently than Tcb1 in the reference strain AF16, but both CbmaT1 and Tcb1 excised at extremely high frequencies in the HK105 strain. The HK105 strain also exhibited a high frequency of spontaneous induction of unc-22 mutants, suggesting that it may be a mutator strain of C. briggsae.  相似文献

Quantitative composition of leukocytes and structure of spleen of the perch Perca fluviatilis L. from water reservoirs with different pH and color of water were studied. Changes of proportion of lymphocytes, macrophages, and band neutrophils are shown in fish from reservoirs with different pH level. Two types of small blood vessels are revealed and changes of their structure are described: thickening of the connective tissue layer, the appearance of fibroblast-like cells, an increase of folding of the basal part of the endotheliocyte membrane. Traditionally viewed as an Andean grain crop,Chenopodium quinoa Willd. includes domesticated populations that are not Andean, and Andean populations that are not domesticated. Comparative analysis of leaf morphology and allozyme frequencies have demonstrated that Andean populations, both domesticated(quinua) and free-living(ajara), represent an exceptionally homogeneous unit that is well differentiated from allied domesticates of coastal Chile(quingua) and freeliving populations of the Argentine lowlands(C. hircinum). This pattern of relationships indicates that Andean populations represent a monophyletic crop/weed system that has possibly developed through cyclic differentiation (natural vs. human selection) and introgressive hybridization. Relative levels of variation suggest that this complex originated in the southern Andes, possibly from wild types allied withC. hircinum, with subsequent dispersal north to Colombia and south to the Chilean coast. Coastal populations were apparently isolated from post-dispersal differentiation and homogenization that occurred in the Andes. Other data point toward a center of origin in the northern Andes with secondary centers of genetic diversity subsequently developing in the southern Andes and the plains of Argentina. Comparative linkage of South American taxa, all tetraploid, with North American tetraploids of the subsection will eventually clarify this problem. While the possibility of a direct phyletic connection betweenC. quinoa and the Mexican domesticate(C. berlandieri subsp. nuttalliae,) cannot be excluded, available evidence indicates that the latter represents an autonomous lineage that is associated with the basal tetraploid, C. b. subsp.berlandieri, through var.sinuatum, whereas South American taxa show possible affinities to either var. zschackei or var.berlandieri. An extinct domesticate of eastern North America,C. b. subsp.jonesianum, represents either another instance of independent domestication, possibly from subsp. b. var.zschackei, or a northeastern outlier of subsp.nuttalliae.  相似文献

We have investigated the floral ontogeny of Arillastrum, Allosyncarpia, Stockwellia and Eucalyptopsis (of the eucalypt group, Myrtaceae) using scanning electron microscopy and light microscopy. Several critical characters for establishing relationships between these genera and to the eucalypts have been determined. The absence of compound petaline primordia in Arillastrum, Allosyncarpia, Stockwellia and Eucalyptopsis excludes these taxa from the eucalypt clade. Post-anthesis circumscissile abscission of the hypanthium above the ovary in Stockwellia, Eucalyptopsis and Allosyncarpia is evidence that these three taxa form a monophyletic group; undifferentiated perianth parts and elongated fusiform buds are characters that unite Stockwellia and Eucalyptopsis as sister taxa. No floral characters clearly associate Arillastrum with either the eucalypt clade or the clade of Stockwellia, Eucalyptopsis and Allosyncarpia.We gratefully acknowledge Clyde Dunlop and Bob Harwood (Northern Territory Herbarium) for collecting specimens of Allosyncarpia, and Bruce Gray (Atherton) for collecting specimens of Stockwellia. It was investigated whether quorum sensing (QS) mediated by N-acylhomoserine lactones (AHLs) was important for heterotrophic bacteria from the littoral zone of the oligotrophic Lake Constance for growth with organic particles. More than 900 colonies from lake water microcosms with artificial organic aggregates consisting of autoclaved unicellular algae embedded in agarose beads were screened for AHL-production. AHL-producing bacteria of the genus Aeromonas enriched in the microcosms but AHLs could not be detected in any microcosm. To test for a potential function of AHL-mediated QS, growth experiments with the wild type and an AHL-deficient mutant of Aeromonas hydrophila in lake water microcosms were performed. Growth of both strains did not differ in single cultures and showed no mutual influence in co-cultures. In co-cultures with a competitor bacterium belonging to the CytophagaFlavobacterium group, growth of both A. hydrophila strains was reduced while growth of the competitor bacterium was not affected. Exogenous AHL-addition did not influence growth of the Aeromonas strains in any microcosm experiment. These results showed that AHL-mediated QS was not required for A. hydrophila during colonization and degradation of organic particles in lake water microcosms, suggesting that cell–cell signalling of heterotrophic bacteria in oligotrophic waters relies on novel signal molecules.  相似文献

We used data from parasites and stable isotopes of yellow perch, Perca flavescens, to determine trophic status in four small Canadian Shield lakes as parasites allow the identification of both prey and non-prey dietary components in the host's community. Stable C isotope ratios for all perch ranged from –34 to –19 while stable N isotope ratios ranged from 4.5 to 12.5. These ranges are larger than those observed in many other fish species. Perch age was the most significant predictor of stable C isotope ratio and perch parasite fauna was the most significant predictor of stable N ratios. In particular, parasite fauna indicative of zooplanktivorous or piscivorous perch were most accurate for predicting fish trophic position and thus stable isotope ratio. Fish length and age showed fewer significant relationships with isotope ratios than parasite infracommunity or diet and suggests that trophic category for perch cannot always be predicted based on size.  相似文献

Notocactus scopa cv. Soonjung was subjected to in planta Agrobacterium tumefaciens-mediated transformation with vacuum infiltration, pin-pricking, and a combination of the two methods. The pin-pricking combined with vacuum infiltration (20-30 cmHg for 15 min) resulted in a transformation efficiency of 67-100%, and the expression of the uidA and nptII genes was detected in transformed cactus. The established in planta transformation technique generated a transgenic cactus with higher transformation efficiency, shortened selection process, and stable gene expression via asexual reproduction. All of the results showed that the in planta transformation method utilized in the current study provided an efficient and time-saving procedure for the delivery of genes into the cactus genome, and that this technique can be applied to other asexually reproducing succulent plant species.  相似文献

Single (unpaired) vegetative cells of freshwater pennate diatom Neidium cf. ampliatum differentiated into gametangia and produced a single zygote (auxospore) via a pedogamic process.
