与水驱技术相比,向油藏中注入碱、表面活性剂和聚合物(简称三元复合驱,ASP)能大幅提高石油采收率,但这些驱油剂对油藏中微生物多样性与群落结构的影响尚亟待阐明,这对油田水质管理与腐蚀控制均具有的重要意义. 本研究采用高通量测序技术解析了大庆油田ASP油藏4口油井采出水中的微生物多样性与群落结构. 结果表明: ASP油藏采出水的pH高达9.65. 采出水中微生物Shannon多样性指数为2.00~3.56,采出井间菌群多样性存在差异. 在门、纲、属分类水平上,变形菌门(85.5%~98.3%)、γ-变形菌纲(83.7%~97.8%)、栖碱菌属(51.8%~82.5%)是绝对优势菌群. 共检测到12个属的潜在硫化氢产生菌,以硫磺单胞菌属丰度最高(0.4%~7.4%). 与已发表的水驱油藏研究结果相比,三元复合驱油藏采出水微生物群落组成独特,呈嗜/耐碱趋势,其多样性偏低,群落结构更单一.  相似文献

人工干预后稠油藏内部微生物群落会发生复杂变化,为进一步明确微生物活性和代谢多样性,选取辽河油田不同开发方式内8口采油井,利用佰珞生态(Biolog-ecology,Biolog-ECO)技术解析地层水微生物群落功能多样性特征.结果显示,地层水微生物群落对多聚物类和氨基酸类代谢最显著,碳水化合物和羧酸类次之,对胺类和酚类化合物代谢程度最低.微生物群落颜色平均变化率(AWCD)的趋势为蒸汽吞吐驱(SS)>聚合物驱(PF)>蒸汽驱(SF),香农-威纳多样性指数(H')、辛普森优势度指数(D)及皮卢均匀度指数(J)的趋势为PF>SS>SF.采样井由水驱(WF)开发转变为PF开发后,微生物群落在油水体系中发生较大变异,多样性指数高于其他开采方式,表现出较高代谢多样性.SS开发温度梯度范围是46~350℃比SF开发的37~290℃范围更宽泛,意味着微生物可选择适宜的环境生长繁殖,其碳源代谢活性强于其他开采方式.研究表明,SS和PF开发地层水微生物群落对碳源利用能力强,其结果为了解复杂稠油藏内部微生物资源和代谢特征以及后续内源激活、功能微生物定向筛培等应用提供参考.  相似文献

【目的】新疆油田六中区为典型水驱普通稠油油藏,水驱效果较差,油藏具有丰富的内源微生物,本研究通过分析内源微生物驱油对油藏微生物活动的影响,确定内源微生物驱油技术在该类油藏的应用潜力。【方法】采用高通量测序及分析化学技术,系统研究实施内源微生物驱油技术后油藏细菌群落结构组成、细菌总数和功能菌群的浓度以及采出液的流体性质,总结内源微生物驱油对油藏微生物活动的影响。【结果】现场试验注入激活剂和空气后,内源微生物被显著激活,细菌群落结构发生明显变化,细菌总数及功能菌群浓度普遍提高了2–3个数量级;各种内源微生物代谢活动显著增强,与地层流体相互作用后,原油明显被乳化,最终石油采收率提高5.2%。【结论】对于内源微生物较为丰富的水驱普通稠油油藏,内源微生物驱油技术对油藏微生物活动的影响显著,具有显著的技术优势和较大的应用潜力,微生物群落结构、功能菌群浓度及其相关代谢产物可以作为评价内源微生物驱油现场激活效果的重要指标,为其他内源微生物驱油现场试验提供技术参考。  相似文献

石油污染对土壤微生物群落多样性的影响   总被引:3,自引:0,他引:3  
土壤中的微生物主要有细菌、放线菌、真菌三大类群,微生物在石油污染的土壤中发挥着维持生态平衡和生物降解的功能。文中以四川省遂宁市射洪县某废弃油井周围不同程度石油污染土壤为供试土壤,首先对各组供试土壤的基本理化性质进行测定分析;然后采用平板菌落计数法测定了供试土壤中三大类微生物数量的变化,结果表明:相比未被污染的对照土壤,石油污染的土壤中细菌、放线菌、真菌数量均减少,并且土壤中可培养微生物的数量与土壤含水量呈正相关;再采用454焦磷酸测序技术对土壤中的细菌群落多样性及变化进行16S rRNA基因分析。在所有供试的4个土壤样品中,共鉴定出不少于23 982个有效读取序列和6 123种微生物,相比于未被污染的对照土壤,石油污染土壤中细菌的种类更加丰富,主要优势门类为酸杆菌门、放线菌门、拟杆菌门、绿弯菌门、浮霉菌门和变形菌门。但不同土壤样品中优势菌群的群落结构有所差异,石油污染的土壤中,酸杆菌门、放线菌门和变形菌门的数量最多,未被石油污染的土壤中,放线菌门、拟杆菌门和变形菌门的数量最多。  相似文献

大庆油田油藏采出水的细菌群落结构   总被引:1,自引:0,他引:1  
采用ARDRA (扩增性rDNA限制性酶切片段多态性分析)技术对大庆油田聚驱、水驱和过渡带3种油藏采出水中的细菌群落的基因组总DNA的16S rDNA克隆文库进行分析,研究了细菌群落结构.结果表明:随机挑取的596个阳性克隆可分为85个操作分类单元 (OTUs),其中聚驱、水驱和过渡带文库分别含有28、41和33个.通过对优势OTUs测序,并与GenBank进行序列比对,发现油藏采出水中的优势菌群为不动杆菌属、弓形杆菌属、厚壁菌门、假单胞菌属和硫磺单胞菌属.聚驱样品中细菌群落组成最简单,优势菌群为不动杆菌属,占库容的85%,假单胞菌属占7%;水驱样品中的优势菌群也是不动杆菌属,占库容的62%,假单胞菌属和硫磺单胞菌属各占20%和6%;过渡带文库的细菌群落的优势菌群为弓形杆菌属,占库容的50%,不动杆菌属和厚壁菌门各占19%和18%.  相似文献

Microbes commonly exist in milieus of varying complexity and diversity. Although cultivation-based techniques have been unable to accurately capture the true diversity within microbial communities, these deficiencies have been overcome by applying molecular approaches that target the universally conserved 16S ribosomal RNA gene. The recent application of 454 pyrosequencing to simultaneously sequence thousands of 16S rDNA sequences (pyrotags) has revolutionized the characterization of complex microbial communities. To date, studies based on 454 pyrotags have dominated the field, but sequencing platforms that generate many more sequence reads at much lower costs have been developed. Here, we use the Illumina sequencing platform to design a strategy for 16S amplicon analysis (iTags), and assess its generality, practicality and potential complications. We fabricated and sequenced paired-end libraries of amplified hyper-variable 16S rDNA fragments from sets of samples that varied in their contents, ranging from a single bacterium to highly complex communities. We adopted an approach that allowed us to evaluate several potential sources of errors, including sequencing artifacts, amplification biases, non-corresponding paired-end reads and mistakes in taxonomic classification. By considering each source of error, we delineate ways to make biologically relevant and robust conclusions from the millions of sequencing reads that can be readily generated by this technology.  相似文献

Maintenance of a high degree of biodiversity in homogeneous environments is poorly understood. A complex cheese starter culture with a long history of use was characterized as a model system to study simple microbial communities. Eight distinct genetic lineages were identified, encompassing two species: Lactococcus lactis and Leuconostoc mesenteroides. The genetic lineages were found to be collections of strains with variable plasmid content and phage sensitivities. Kill-the-winner hypothesis explaining the suppression of the fittest strains by density-dependent phage predation was operational at the strain level. This prevents the eradication of entire genetic lineages from the community during propagation regimes (back-slopping), stabilizing the genetic heterogeneity in the starter culture against environmental uncertainty.  相似文献

【目的】比较和分析从堆肥中富集的水稻秸秆降解菌系F1和F2的纤维素分解能力、微生物群落结构及其在秸秆降解过程中的演替,从而探究微生物群落结构与秸秆降解效率的相关性。【方法】采用DNS(3,5-二硝基水杨酸,3,5-dinitrosalicylic acid)定糖法测定发酵液中的外切纤维素酶活;采用范氏(Van Soest)洗涤纤维分析法测定发酵前与发酵后的秸秆纤维素、半纤维素、木质素的含量,并计算降解率;采用16S r RNA基因序列分析和实时荧光定量PCR(Quantitative real-time PCR,Q-PCR)对秸秆降解过程中的微生物物种组成及特定的功能微生物进行定性和定量分析。【结果】复合菌系F1的水稻秸秆总降解率、纤维素降解率、半纤维素降解率显著高于复合菌系F2;2种复合菌系的外切纤维素酶活性与cel48基因的拷贝数变化趋势一致;复合菌系F1的物种较丰富,优势物种是好氧细菌,复合菌系F2的物种组成较单一,培养后期具有较高比例的厌氧纤维素分解菌;培养前4天,复合菌系F1和F2的优势物种均为Unclassified Bacillales和Bacillus;第4天之后,不同复合菌系的优势物种及丰度出现差异,F1的优势物种主要属于Bacteroidetes,F2的优势物种主要属于Firmicutes;虽然Petrimonas和Pusillimonas是培养后期的共有优势物种,但是Petrimonas在复合菌系F2中的相对丰度(38.30%)显著高于F1(9.47%),且培养第8天的F2中的Clostridiales OPB54增加至14.85%。【结论】cel48基因拷贝数变化与秸秆纤维素的降解效率、外切纤维素酶活性变化具有一定的相关性,cel48基因可作为潜在的生物分子标记监测秸秆纤维素的降解过程;微生物群落结构对秸秆纤维素的降解效率具有显著影响,Unclassified Bacillales,Bacillus,Petrimonas,Pusillimonas是复合菌系F1和F2降解秸秆纤维素过程中的重要物种。  相似文献

Microbial souring (H2S production) in porous media was investigated in an anaerobic upflow porous media reactor at 60 degrees C using microbial consortia obtained from oil reservoirs. Multiple carbon sources (formate, acetate, propionate, iso- and n-butyrates) found in reservoir waters as well as sulfate as the electron acceptor was used. Kinetics and rates of souring in the reactor system were analyzed. Higher volumetric substrate consumption rates (organic acids and sulfate) and a higher volumetric H(2)S production rate were found at the from part of the reactor column after H(2)S production had stabilized. Concentration gradients for the substrates (organic acids and sulfate) and H(2)S were generated along the column. Biomass accumulation throughout the entire column was observed. The average specific sulfate reduction rate (H(2)S production rate) in the present reactor after H(2)S production had stabilized was calculated to be 11062 +/-2.22 mg sulfate-S/day g biomass. The effects of slurry application method and weather conditions after application on ammonia volatilisation are well documented, however, the effect on slurry N recovery in herbage is less evident due to large variability of results. The objective of this field experiment was to determine the recovery of cattle slurry NH4-N in herbage and soil in the year of application as affected by application method (trailing shoe versus broadcast) and season of application (spring versus summer), using 15N as a tracer. In 2007 and 2008, 15N enriched slurry was applied on grassland plots. N recovery in herbage and soil during the year of application was determined. Both spring and trailing shoe application resulted in significantly higher herbage DM yields, N uptake and an increased recovery of 15NH4-N in herbage. Additionally, the recovery of slurry 15NH4-N in the soil at the end of the growing season was increased. Spring and trailing shoe application reduced the losses of slurry 15NH4-N by on average 14 and 18 percentage points, respectively, which corresponded closely to ammonia volatilisation as predicted by the ALFAM model. It was concluded that slurry N recovery in temperate pasture systems can be increased by adjusting the slurry application method or timing.  相似文献

Streams and rivers form conspicuous networks on the Earth and are among nature''s most effective integrators. Their dendritic structure reaches into the terrestrial landscape and accumulates water and sediment en route from abundant headwater streams to a single river mouth. The prevailing view over the last decades has been that biological diversity also accumulates downstream. Here, we show that this pattern does not hold for fluvial biofilms, which are the dominant mode of microbial life in streams and rivers and which fulfil critical ecosystem functions therein. Using 454 pyrosequencing on benthic biofilms from 114 streams, we found that microbial diversity decreased from headwaters downstream and especially at confluences. We suggest that the local environment and biotic interactions may modify the influence of metacommunity connectivity on local biofilm biodiversity throughout the network. In addition, there was a high degree of variability in species composition among headwater streams that could not be explained by geographical distance between catchments. This suggests that the dendritic nature of fluvial networks constrains the distributional patterns of microbial diversity similar to that of animals. Our observations highlight the contributions that headwaters make in the maintenance of microbial biodiversity in fluvial networks.  相似文献

Aim: To evaluate the bioenergy generation and the microbial community structure from palm oil mill effluent using microbial fuel cell. Methods and Results: Microbial fuel cells enriched with palm oil mill effluent (POME) were employed to harvest bioenergy from both artificial wastewater containing acetate and complex POME. The microbial fuel cell (MFC) showed maximum power density of 3004 mW m?2 after continuous feeding with artificial wastewater containing acetate substrate. Subsequent replacement of the acetate substrate with complex substrate of POME recorded maximum power density of 622 mW m?2. Based on 16S rDNA analyses, relatively higher abundance of Deltaproteobacteria (88·5%) was detected in the MFCs fed with acetate artificial wastewater as compared to POME. Meanwhile, members of Gammaproteobacteria, Epsilonproteobacteria and Betaproteobacteria codominated the microbial consortium of the MFC fed with POME with 21, 20 and 18·5% abundances, respectively. Conclusions: Enriched electrochemically active bacteria originated from POME demonstrated potential to generate bioenergy from both acetate and complex POME substrates. Further improvements including the development of MFC systems that are able to utilize both fermentative and nonfermentative substrates in POME are needed to maximize the bioenergy generation. Significance and Impact of the Study: A better understanding of microbial structure is critical for bioenergy generation from POME using MFC. Data obtained in this study improve our understanding of microbial community structure in conversion of POME to electricity.  相似文献

我国大多数陆上主力油田已进入"双高"(高含水、高采出程度)开采阶段。为进一步提高聚合物驱后油藏的石油采收率,本研究发现了一种促进油藏内源微生物生长代谢的高效激活剂,且该激活剂已通过室内产气和物理模拟驱油实验及岩心电镜观察和454(Pyrosequencing)焦磷酸测序方法的评价。结果表明激活剂在高压容器中静置培养60 d后产气增压达到2 MPa,用量0.35 PV,浓度为1.8%(W/W)的激活剂溶液在聚合物驱后的天然岩心上石油采收率可再提高3%,并在电镜观察到岩心内部激活的内源微生物菌体及其代谢产物。现场于2011年12月在萨南南二区东块的1注4采井组开展了试验,监测到4口有效生产井的甲烷和二氧化碳气体δ13C(PDB)含量产生明显变化,与萨南南二区东块对照试验区产油量增幅35.9%,含水稳定在94%,在3年半期间阶段累计增油5 957 t,为同类油藏进一步提高石油采收率提供了一条有效途径。  相似文献

Oil refineries are known for the large volume of water used in their processes, as well as the amount of wastewater generated at the end of the production chain. Due to strict environmental regulations, the recycling of water has now become a viable alternative for refineries. Among the many methods available to treat wastewater for reuse, the use of membranes in reverse osmosis systems stands out due to several economic and environmental benefits. However, these systems are vulnerable to contamination and deposition of microorganisms, mainly because of the feedwater quality. In this study, the microbial diversity of feedwater and reverse osmosis membranes was investigated using a combination of culture-dependent and culture-independent methods in order to characterize the microorganisms colonizing and deteriorating the membranes. In total, 37 bacterial isolates, 17 filamentous fungi and approximately 400 clones were obtained and analyzed. Among the bacterial genera identified, the most represented were Sphingobium, Acidovorax, Microbacterium, Rhizobium and Shinella. The results revealed genera that acted as candidate key players in initial biofilm formation in membrane systems, and provided important information concerning the microbial ecology of oligotrophic aquatic systems.  相似文献

油藏微生物群落的解析和认知是开发和应用微生物采油技术的基础。利用各种提高油藏微生物可培养性的方法和非培养技术解析不同油藏微生物的群落结构、功能和多样性,对定向调控油藏微生物群落、开发和应用有效微生物驱油技术具有重要的指导意义。通过调查新近发展的提高微生物可培养性的方法和措施以及不依赖于培养的分子微生物生态学技术,总结了油藏微生物群落研究方法学的最新进展。提高微生物可培养性的方法和措施主要通过模拟微生物的生存环境,减少富营养的毒害作用、添加信号分子维持微生物细胞间的作用和提供新型电子供体和受体等手段采用稀释法、高通量培养法等方法得以实现;不依赖于培养的分子微生物生态学技术主要包括荧光原位杂交、末端限制性片断长度多态性分析、变性梯度凝胶电泳和构建克隆文库等技术。这些方法学的进展为更有效的获得各种油藏微生物资源、调控油藏微生物群落以提高石油采收率提供理论指导。  相似文献

2005年4月-2006年3月调查了杭州市郊下沙区一个沼泽性水域原生动物群落的变化规律。共观察到各类原生动物265种,其中鞭毛虫114种,占原生动物总数的43%,其中植物性鞭毛虫占91%,动物性鞭毛虫仅占9%;肉足虫34种,占原生动物总数的13%,纤毛虫117种,占原生动物总数的44%。原生动物在此水域1个周年内的分布特征表现为植物性鞭毛虫种类多于动物性鞭毛虫,纤毛虫种类多于肉足虫种类。原生动物群落的结构参数(种类组成、个体丰度和多样性指数)因水质的变化而变化。此水域原生动物的优势种为梨形扁裸藻(Phacus pyrum)、旋转囊裸藻(Trachelomonas volvocina)、绿色裸藻(Euglenaviridis)和刺鱼状裸藻(Euglena gasterosteus),常年居留种为裸藻(Euglenasp.)、奇形扁裸藻(Phacus anomalus)、钟形虫(Vorticellasp.)、尾草履虫(Paramecium caudatum)和多态喇叭虫(Stentor polymorphrus)。全年3个采样站的Margalef生物多样性指数为1.056~6.054。  相似文献

Two sequencing batch reactors (SBR) were constructed and filled with different inocula of activated sludge (AS) and mature fine tailings (MFT) to treat oil sands process-affected water (OSPW). The COD was reduced by 82% in the AS-SBR and 43% in the MFT-SBR during phase I using 10% OSPW and 90% synthetic wastewater as reactor feed. However, COD removal reached 12% and 20% in the AS-SBR and the MFT-SBR, respectively, when 100% raw OSPW was fed into the reactors. Maximum removal of acid-extractable organics (AEO) was 8.7% and 16.6% in the AS-SBR and the MFT-SBR, respectively with a hydraulic retention time of one day. Pyrosequencing analysis revealed that Proteobacteria was the dominant phylum and beta- and gamma-Proteobacteria were dominant classes in both reactors. Evidence of a microbial community change was observed when influent raw OSPW was switched from 50 to 100%. More significant changes in the AS-SBR community were detected.  相似文献
