达乌尔黄鼠冬眠期间体温的变化和冬眠模式   总被引:2,自引:0,他引:2  
用植入式半导体温度记录元件iButton 记录了达乌尔黄鼠冬眠季节及其前后的体温,分析了其冬眠模式和体温调节特点。结果显示:1)实验室条件下,达乌尔黄鼠冬眠季节长短的个体差异较大,可以分成深冬眠型、
少冬眠型和不冬眠型三种类型;2)达乌尔黄鼠在冬季表现出深冬眠阵(最低体温Tbm in <20℃ ,冬眠阵的持续时间BD >24 h)、短冬眠阵(Tbmin < 20℃ , BD≤24h)和日眠阵(Tbmin ≥20℃ , BD≤24 h)3 种类型,最低体温分别
为2.54℃ ± 0.35℃ 、10.05℃ ± 1.97℃ 和23.09℃ ± 0.40℃ ,彼此之间差异显著。日眠阵阵间产热阶段的最高体温为38.09℃ ±0.17℃ ,高于深冬眠阵(37.31℃ ±0.15℃ )和短冬眠阵(37.22℃ ±0.31℃ ); 3)深冬眠阵和日
眠阵中最低体温均与环境温度显著相关,冬眠过程中的最低体温为-2.43℃ ;4)深冬眠过程中,多数个体可以短时(≤3 h)耐受- 2℃ ~ 0℃ 的低温,激醒或继续维持深冬眠,无致死效应,但长时间(15 h)或过度低温
(- 5℃ 以下)的条件下,深冬眠的达乌尔黄鼠被激醒(70% )或死亡(30% ),不能持续冬眠; 5)入眠前10 d体温日波动幅度显著增加,高于出眠后的日体温波动,且多数个体入眠前出现体温的“试降”。表明,冬眠前

J. Neurochem. (2012) 122, 934-940. ABSTRACT: Hibernation is an adaptation to overcome periods of resource limitation often associated with extreme climatic conditions. The hibernation season consists of prolonged bouts of torpor that are interrupted by brief interbout arousals. Physiological mechanisms regulating spontaneous arousals are poorly understood, but may be related to a need for gluconeogenesis or elimination of metabolic wastes. Glutamate is derived from glutamine through the glutamate-glutamine cycle and from glucose via the pyruvate carboxylase pathway when nitrogen balance favors formation of glutamine. This study tests the hypothesis that activation of NMDA-type glutamate receptors (NMDAR) maintains torpor in arctic ground squirrel (arctic ground squirrel (AGS); Urocitellus parryii). Administration of NMDAR antagonists MK-801 (5?mg/kg, i.p.) that crosses the blood-brain barrier and AP5 (5?mg/kg, i.p.) that does not cross the blood-brain barrier induced arousal in AGS. Central administration of MK-801 (0.2, 2, 20 or 200?μg; icv) to hibernating AGS failed to induce arousal. Results suggest that activation of NMDAR at a peripheral or circumventricular site is necessary to maintain prolonged torpor and that a decrease in glutamate at these sites may contribute to spontaneous arousal in AGS.  相似文献

Abstract

Wheel‐running activity of forty antelope ground squirrels, Ammospermophilus leucurus, was monitored for several months in both an outdoor cage and in the laboratory. The squirrels demonstrated a highly diurnal pattern which persisted in “constant conditions.” After removal from the field the initial free‐running period was close to 24 hrs, but typically lengthened in a nearly linear fashion at least for the first few months. There was no evidence of any difference in this trend for squirrels, in D/D, L/L 100 lx, 250 lx or 1200 lx. Eventually, about 90 percent of the squirrels had periods longer than 24 hrs.

The synchronizing capacity of the natural photoperiod was used to “catch the free‐running rhythm” and thereby demonstrate a response curve. Synchronization occurred by a shortening of the period when the time of sunrise was between 125° and 0° (subjective night) and by a lengthening of the period when the time of sunrise was between 0° and 125° (subjective day).

To more thoroughly examine the underlying mechanisms of phase control, phase‐response curves based on sixty one light‐pulse experiments were constructed. Comparisons of curves based on 6‐hr and 15‐min pulses, showed that the integral action of light is important (i.e., the entire pulse is involved in phase shifting). It was found that light pulses not only affected the phase of the rhythm but also the phase. Large phase shifts were usually associated with decreases in free‐running period. Several hypotheses on the controlling mechanisms were advanced.  相似文献

贮脂类动物在冬眠前大量积累脂肪来准备冬眠,并在入眠时迅速降低体温和代谢率。为探究入眠准备期达乌尔黄鼠体温、代谢率、呼吸商及能量代谢的变化,将其入眠准备期分为育肥期、体重高峰期、育肥后期和冬眠前的试降期,使用植入式半导体温度记录元件iButton、开放式代谢仪和改进的代谢笼,监测其体温、代谢率及呼吸商和能量摄入的变化。结果显示:(1)达乌尔黄鼠体温在冬眠前13 - 34 d 开始下降,远早于冬眠但晚于体重高峰期;体重高峰期体温有降低的趋势,持续时间为1 - 3 d;育肥后期体温显著下降,体温日波动幅度增加。(2)体重高峰期的静止代谢率高于育肥期,育肥后期有降低的趋势,试降期最低。(3)呼吸商在体重高峰期先升高,之后迅速衰减;入眠准备期的能量摄入在体重达高峰期前达到最大值。结果表明,达乌尔黄鼠在入眠准备期,其体温和代谢率已开始降低,能源物质已开始转变;体重高峰期可能是达乌尔黄鼠入眠的一个转折点或启动入眠的开关。  相似文献

Circadian clocks are centrally involved in the regulation of daily behavioural and physiological processes. These clocks are synchronized to the 24 h day by external cues (Zeitgeber), the most important of which is the light–dark cycle. In polar environments, however, the strength of the Zeitgeber is greatly reduced around the summer and winter solstices (continuous daylight or continuous darkness). How animals time their behaviour under such conditions has rarely been studied in the wild. Using a radio-telemetry-based system, we investigated daily activity rhythms under continuous daylight in Barrow, Alaska, throughout the breeding season in four bird species that differ in mating system and parental behaviour. We found substantial diversity in daily activity rhythms depending on species, sex and breeding stage. Individuals exhibited either robust, entrained 24 h activity cycles, were continuously active (arrhythmic) or showed ‘free-running’ activity cycles. In semipalmated sandpipers, a shorebird with biparental incubation, we show that the free-running rhythm is synchronized between pair mates. The diversity of diel time-keeping under continuous daylight emphasizes the plasticity of the circadian system, and the importance of the social and life-history context. Our results support the idea that circadian behaviour can be adaptively modified to enable species-specific time-keeping under polar conditions.  相似文献

The aim of the present study was to investigate the impact of endogenous and exogenous factors for the expression of the daily rhythms of body temperature (BT), blood pressure (BP) and heart rate (HR). One hundred and seventy-three young adults (YA), 17–24 years old (y.o.), of both genders were studied under a modified constant-routine (CR) protocol for 26 h. Participants were assigned randomly to groups with different lighting regimens: CR-LD, n = 77, lights (>400 l×) on from 09:00 to 17:00 h and off (<10 l×) from 17:00 to 09:00 next morning; CR-LL, n = 81, lights on (>400 l×) during the whole experimental session; CR-DD, n = 15, constant dim light (<10 l×) during the whole experiment. Systolic (SBP) and diastolic (DBP) BP, HR and BT were measured every 2 h. For comparison, the results of the former studies performed under conditions of regular life with an activity period from 07:00 to 23:00 h and sleep from 23:00 till 07:00 h (Control) were reanalyzed. Seven-day Ambulatory Blood Pressure Monitoring (ABPM) records from 27 YA (16–38 y.o.) and BT self-measurement data from 70 YA (17–30 y.o.) taken on ≥ 3 successive days at 08:00, 11:00, 14:00, 17:00, 20:00, 23:00 and 03:00 were available.

The obtained daily patterns were different between Control and CR-DD groups, due to effects of activity, sleep and light. The comparison of Control and CR-LD groups allowed the effects of sleep and activity to be estimated since the lighting conditions were similar. The activity level substantially elevated SBP, but not DBP. Sleep, on the other hand, lowered the nighttime DBP, but has no effect on SBP. HR was affected both by activity and sleep. In accordance with previous studies, these results confirm that the steep BP increase in the morning is not driven by the circadian clock, but rather by sympathoadrenal factors related to awakening and corresponding anticipatory mechanisms. The effect on BT was not significant.

To investigate the impact of light during the former dark time and darkness during the former light time, the CR-LL and CR-DD groups were each compared with the CR-LD group. Light delayed the evening decrease of BT, most likely via a suppression of the melatonin rise. Besides, it had a prominent arousal effect on SBP both in the former light and dark phases, a moderate effect on DBP and no effect on HR. Darkness induced decline in BT. BP values were decreased during the former light time. No effects on HR were found. Altogether, the results of the present paper show that BT, BP and HR are affected by exogenous factors differently. Moreover, the effect was gender-specific. Especially, the response of BT and BP to ambient light was evident only in females.

We suppose that the distinct, gender-specific responses of SBP, DBP and HR to activity, sleep and ambient light do reflect fundamental differences in the circadian control of various cardiovascular functions. Furthermore, the presented data are important for the elaboration of updated reference standards, the interpretation of rhythm disorders and for personalized chronotherapeutic approaches to prevent adverse cardiovascular events more effectively.  相似文献

We report a progressive disruption of 24-h rhythms in fasting blood glucose (FBG), body temperature (BT) and heart rate (HR) associated with metabolic dysfunction and the development of prediabetes (PD) and type 2 diabetes mellitus (T2DM) in overweight middle-aged (40–69 years old) humans. Increasing BT and HR mean values and declining 24-h BT and HR amplitudes accompany adverse changes in metabolic state. Increased nocturnal BT and a phase delay of the 24-h BT rhythm, deviant 24-h HR profile and a phase advance of the 24-h HR and FBG rhythms are early signs of the PD metabolic state. In T2DM, the 24-h FBG rhythm is no longer detectable, and the 24-h amplitudes of BT and HR are greatly diminished. In addition, lepton and creatinine values were lowered in T2DM. Moreover, positive correlations between FBG and body mass index, BMI, and negative correlations between the 24-h amplitude of FBG and BMI indicate that overweight is an additional factor causing disruption of the circadian rhythms. Further studies on circadian disruption as a consequence of metabolic dysfunction are necessary. The quantitative analysis of changing circadian BT and HR rhythms may provide prognostic markers of T2DM and therapeutic targets for its prevention and correction.  相似文献

Studies under real life conditions become more and more relevant in chronobiological and chronomedical research. The present study aims to analyze one of the most prominent biological rhythms: the core body temperature (CBT) rhythm in the real world outside the laboratory. CBT was recorded continuously in 37 healthy women (age between 21 and 44 years, median 29 years) with a newly developed intravaginal temperature sensor for up to 102 days. Sleep logs were available from 23 participants. To quantify the daily dynamics of each individual CBT-curve, novel measurement parameters are introduced which permit the quantification of the phase and shape of the CBT rhythms as well as their relation to the sleep–wake cycle. In addition to the classical phase markers (i.e. nadir and peak), the daily curves were segmented into quartiles by introducing the t25/t50/t75-values which can be used as phase and shape markers. At variance to previous studies, a conspicuous day-to-day variation was shown not only for the time point of the peak, but also for the time point of the nadir. However, the t-values, particularly the t75-value were relatively closely locked to external time and thus represent more reliable phase markers than the nadir. The (variable) time point of the nadir determined the period length, phase and shape of the subsequent CBT cycle. If a nadir occurred close to the wake-up time, the following cycle was considerably shorter than 24 hours, while a nadir distant from the wake-up time was followed by a longer cycle. Thus, the period lengths of the daily CBT cycles of each individual were characterized by an “expand/contract” rhythm. The analyses of the novel phase markers (t25/t50/t75) of the CBT curves allowed to identify “early” and “late” participants who may differ in their phase-response curves with regard to the entraining effect of light. In addition, the novel phase markers mirrored the different social entrainment conditions on weekends and workdays.  相似文献

We studied daily rhythmicity of body temperature (T b) before and during hibernation in Anatolian ground squirrels (Spermophilus xanthoprymnus) under natural and laboratory conditions using surgically implanted temperature loggers. Under both conditions, robust daily T b rhythmicity with parameters comparable to those of other ground squirrel species was observed before but not during hibernation. Euthermic animals had robust daily T b rhythms with a mean of 37.0°C and a range of excursion of approximately 4°C. No T b rhythm was detected during torpor bouts, either because T b rhythmicity was absent or because the daily range of excursion was smaller than 0.2°C. The general patterns of hibernation that we observed in Anatolian ground squirrels were similar to those previously observed by other investigators in other species of ground squirrels.  相似文献

Surface temperatures (Ts) of eight 13-lined ground squirrels and seven yellow-bellied marmots were measured during arousal from hibernation using infrared thermography (IRT) and recorded on videotape. Animals aroused normally in 5 degrees C cold rooms. Body temperatures were recorded during arousal using both cheek pouch and interscapular temperature probes. Warming rate in arousal was exponential. Mean mass specific warming rates show the squirrels warm faster (69.76 degrees C/h/kg) than the marmots (4.49 degrees C/h/kg). Surface temperatures (Ts) for 11 regions were measured every few minutes during arousal. The smaller ground squirrel shows the ability to perfuse distal regions without compromising rise in deep body temperature (Tb). All squirrel Ts's remained low as Tb rose to 18 degrees C, at which point, eyes opened, squirrels became more active and all Ts's rose parallel to Tb. Marmot Ts remained low as Tb rose initially. Each marmot showed a plateau phase where Tb remained constant (mean Tb 20.3+/-1.0 degrees C, duration 9.4+/-4.1 min) during which time all Ts's rose, and then remained relatively constant as Tb again began to rise. An anterior to posterior Ts gradient was evident in the ground squirrel, both body and feet. This gradient was only evident in the feet of the marmots.  相似文献

Body temperature of five European hamsters exposed to semi-natural environmental conditions at 47° N in Southern Germany was recorded over a 1.5-year period using intraperitoneal temperature-sensitive radio transmitters. The animals showed pronounced seasonal changes in body weight and reproductive status. Euthermic body temperature changed significantly throughout the year reaching its maximum of 37.9±0.2°C in April and its minimum of 36.1±0.4°C in December. Between November and March the hamsters showed regular bouts of hibernation and a few bouts of shallow torpor. During hibernation body temperature correlated with ambient temperature. Monthly means of body temperature during hibernation were highest in November (7.9±0.8°C) and March (8.2±0.5°C) and lowest in January (4.4±0.7°C). Using periodogram analysis methods, a clear diurnal rhythm of euthermic body temperature could be detected between March and August, whereas no such rhythm could be found during fall and winter. During hibernation bouts, no circadian rhythmicity was evident for body temperature apart from body temperature following ambient temperature with a time lag of 3–5 h. On average, hibernation bouts lasted 104.2±23.8 h with body temperature falling to 6.0±1.7°C. When entering hibernation the animals cooled at a rate of -0.8±0.2°C·h-1; when arousing from hibernation they warmed at a rate of 9.9±2.4°C·h-1. Warming rates were significantly lower in November and December than in January and February, and correlated with ambient temperature (r=-0.46, P<0.01) and hibernating body temperature (r=-0.47, P<0.01). Entry into hibrnation occured mostly in the middle of the night (mean time of day 0148 hours ±3.4 h), while spontaneous arousals were widely scattered across day and night. For all animals regression analysis revealed free-running circadian rhythms for the timing of arousal. African mole-rats (family: Bathyergidae) are strictly subterranean mammals that reside in extensive networks of underground tunnels. They are rarely, if ever, exposed to light and experience muted temperature ranges. Despite these constant conditions, the presence of a functional circadian clock capable of entraining to external light cues has been reported for a number of species. In this study, we examine a social mole-rat species, Cryptomys hottentotus mahali, to determine if it possesses a functional circadian clock that is capable of perceiving light and ambient temperature cycles, and can integrate these cues into circadian rhythms of locomotor activity and core body temperature. Eight male and eight female, non-reproductive individuals were subjected to six cycles of varying light and temperature regimes. The majority of the individuals displayed daily rhythms of locomotor activity and body temperature that are synchronised to the external light and temperature cycles. Furthermore, endogenous rhythms of both locomotor activity and core body temperature were displayed under constant conditions. Thus, we can conclude that C. h. mahali possesses a functional circadian clock that can integrate external light and temperature cues into circadian rhythms of locomotor activity and core-body temperature.  相似文献

Two species of the genus Acomys coexist in arid zones of southern Israel. Acomys russatus is distributed in extremely arid areas, while A. cahirinus is common in both Mediterranean and arid regions. Individuals of both species from a rodent community in the Ein Gedi Nature Reserve were implanted with temperature-sensitive transmitters. Body temperature (T b) rhythms were recorded in free-ranging mice at four different seasons of the year. A. cahirinus (30–45 g) showed a nocturnal rhythm of T b throughout the year. In the activity phase during the night T b increased to 38.2°C. During the day T b decreased to 34°C. This species displayed this pattern in summer also when ambient temperatures rose above T b. The T b of A. russatus (45–65 g) varied between 34.8 and 41°C during the hot season, showing a bimodal temperature rhythm with maximal values in the morning and in the evening. Measurements of activity in this species showed inactivity during the hottest period of a summer day. In winter A. russatus showed no clearly detectable diurnal or ultradian rhythm in T b, which remained constant between narrow limits of 35.2 and 36.8°C. 目的:同步遥测棕色脂肪组织(BAT)产热与体核温度昼夜律变化的时间曲线,分析二者昼夜节律变化的时间关系。方法:实验用成年雄性SD大鼠,在22℃环境温度下,明暗时间各12h,昼光时间为06:00h-18:00h,同步无线遥测体核温度(TC)、BAT温度(T队T)、腋窝温度(Tax)和动物活动的昼夜节律变化。结果:①在昼光中,TBAT较TC低0.67%,而在暗光中二者则相似。大鼠从昼光进入暗光时,TBAT升高的速率较TC升高速率快,开始上升的时间较TC提前8min;而从暗光进入昼光时,TBAT开始下降的时间则较TC提前4min。②Tax的昼夜节律幅度与TC相似,但无论动物在明光期或暗光期中,Tax均低于同步测量的TC。③从昼光期转入暗光期时,动物的行为活动出现增加反应先于TBAT和TC开始上升的时间。结论:实验结果证明,在暗光期中大鼠TC升高与BAT产热增加有关,说明BAT昼夜节律性产热的变化在维持体温昼夜生理节律中有重要的作用。  相似文献

Phospholipids and cholesterol were assayed in homogenates and microsomal fractions from the cerebral cortex of summer-active, winter-torpid, and winter-active Yakutian ground squirrels (Citellus undulatus). Ultrastructural analysis of both microsomal fraction and intact neurons was performed by serial ultramicrotomy. The levels of sphingomyelin (SM), phosphatidylserine (PS), and phosphatidylethanolamine (PEA) were decreased in homogenates from the cerebral cortex of winter ground squirrels compared with the summer-active animals, while the levels of phosphatidylcholine (PC) and cardiolipin (CL) were increased. The level of cholesterol was decreased in the cerebral cortex of winter-torpid animals compared with both winter-active and summer-active animals, and the level of total phospholipids was decreased in comparison to the summer-active animals. Three-dimensional reconstruction of serial membrane profiles displayed the microsomal fraction to be an interconnected system of cisterns and vesicles, which corresponds to endoplasmic reticulum and dictyosomes (Golgi stacks) of intact neurons. In winter the content of PC was increased in the microsomal fraction, while the contents of lysophosphatidylcholine (LPC), PS, phosphatidylinositol (PI), and SM were decreased. In winter-torpid animals compared with the winter-active ones the contents of total phospholipids, PEA, LPC, and cholesterol were decreased. As for the winter-active ground squirrels, their lipid contents did not differ from those in the summer-active animals, but LPC content was decreased. The changes in microsomal lipid contents in intact pyramidal neurons throughout the hibernation were accompanied by disassembly of dictyosomes and endoplasmic reticulum (ER), including the decomposition of polyribosomes to monosomes. The ultrastructural analysis of nucleoli, ER, and dictyosomes of both winter-active and torpid ground squirrels showed a direct correlation between the increasing contents of both cholesterol and total phospholipids (mainly PEA and LPC) in microsomes and the structural recovery of endoplasmic reticulum, Golgi stacks, and nucleoli in intact pyramidal neurons. A role of seasonal variations in lipid contents of brain cells in their adaptation to low temperature is discussed. We also propose an involvement of cholesterol in the activation of protein-synthesizing function of endoplasmic reticulum and Golgi stacks in intact neurons.  相似文献

The High Arctic summer with its permanent sunlight provides a situation in which one of the natural synchronizers, the light-dark alternation, is minimal. During the summers of 1981 and 1982 three healthy right-handed geographers who were performing field studies in Svalbard as part of their own research volunteered to document, 4-6 times per 24 hr for respectively 63,141 and 147 days, a set of circadian rhythms: self-rated fatigue, oral temperature, grip strength of both hands, heart rate and times of awakening and retiring. Tests were performed before departure from France, in Svalbard (79°IN latitude) where their daily activities were often strenuous, and after returning to France. Time series were treated individually according to three methods: display of data as a function of time, cosinor analyses to quantify rhythm parameters, and spectral analyses to estimate component periods of rhythms. Circadian parameters such as period and acrophase of activity-rest, oral temperature and fatigue rhythms were not altered. On the other hand, the circadian rhythm in grip strength was altered: the period differed from 24 hr in one subject, while grip strength acrophase of the left, but not the right, hand of the other two subjects was phase shifted during the sojourn in Svalbard. A prominent circahemidian (about 12 hr) rhythm was observed in two subjects for their heart rate in Svalbard, while a prominent circadian rhythm (differing from exactly 24 hr) was observed in France associated with a small circahemidian component.  相似文献

Rodents inhabiting high latitudes, close to the Arctic Circle or above it, are exposed to near 24 h daylight during the summer season. An example to such rodent species is the Norwegian lemming Lemmus lemmus, which is distributed in northern Fennoscandia. We measured daily rhythms of heat production (VO2), body temperature (Tb), motor activity and melatonin secretion (measured from its metabolite 6-sulfatoximelatonin 6-SMT) in individuals exposed to natural day light, Oulu Finland, in August 1997 and at a controlled ambient temperature of 22 °C. Our results show a daily rhythm of VO2 with an acrophase at 04:00 h and minimal values measured at 16:00 h, with a significant difference (p<0.001) between mean day and night values. 6-SMT also shows a daily rhythm with maximal secretion during the dark phase 24:00 and 06:00 h in which total 6-SMT values were 2.92±1.1 and 3.87±1.2 ng, respectively. The lowest values were recorded at 12:00 h; 0.86±0.63 ng. These results suggest that lemmings show a VO2 and melatonin daily rhythms, which seem to correlate with each other and it appears that melatonin secretion increases heat production.  相似文献

The nitric oxide (NO) system is involved in the regulation of the cardiovascular system in controlling central and peripheral vascular tone and cardiac functions. It was the aim of this study to investigate in wild-type C57BL/6 and endothelial nitric oxide synthase (eNOS) knock-out mice (eNOS-/-) the contribution of NO on the circadian rhythms in heart rate (HR), motility (motor activity [MA]), and body temperature (BT) under various environmental conditions. Experiments were performed in 12:12 h of a light:dark cycle (LD), under free-run in total darkness (DD), and after a phase delay shift of the LD cycle by -6 h (i.e., under simulation of a westward time zone transition). All parameters were monitored by radiotelemetry in freely moving mice. In LD, no significant differences in the rhythms of HR and MA were observed between the two strains of mice. BT, however, was significantly lower during the light phase in eNOS-/- mice, resulting in a significantly greater amplitude. The period of the free-running rhythm in DD was slightly shorter for all variables, though not significant. In general, rhythmicity was greater in eNOS-/- than in C57 mice both in LD and DD. After a delay shift of the LD cycle, HR and BT were resynchronized to the new LD schedule within 5-6 days, and resynchronization of MA occurred within 2-3 days. The results in telemetrically instrumented mice show that complete knock-out of the endothelial NO system—though expressed in the suprachiasmatic nuclei and in peripheral tissues—did not affect the circadian organization of heart rate and motility. The circadian regulation of the body temperature was slightly affected in eNOS-/- mice.  相似文献
