The dosages of aflatoxin B1 (AFB1) required to produce significant changes in concentrations of B vitamins in plasma and bile and of amino acids in plasma of rabbits were determined. Folate increased by 98% in plasma, whereas concentration of thiamine, vitamin B6, and biotin decreased by more than 50%. In bile, choline and biotin increased 14- and 18-fold, respectively, whereas folate and niacin decreased by more than 50%. All amino acids in plasma increased between 76 and 155%. The dosages of AFB1 required to induce these changes were usually between 12.5 and 37.5 microgram/kg of body weight per day. Except for changes in biliary concentrations of pantothenic acid, folic acid, and biotin, lower threshold dosages of aflatoxin were required to produce weight loss and anorexia (5.0 and 8.5 microgram of AFB1/kg per day, respectively) than for changes in vitamins and amino acids (approximately 25 to 50 microgram of AFB1/kg per day). The data indicated that AFB1 interfered with the metabolism of B vitamins and amino acids in rabbits.  相似文献

In the respiro-fermentative region of aerobic chemostat cultures at steady state, Saccharomyces cerevisiae CBS 8066 produced high concentrations of ethanol with concomitant low levels of residual glucose which followed Monod kinetics. By contrast, very high residual glucose concentrations were observed in cultures of S. cerevisiae strains ATCC 4126 and NRRL Y132 at dilution rates above 60% of the washout dilution rate, resulting in much lower ethanol concentrations, even though clearly glucose-limited at lower dilution rates in the respiratory region. The addition of a vitamin mixture resulted in decreased residual glucose concentrations in respiro-fermentative cultures of all three strains, but the effect was much more pronounced with strains ATCC 4126 and NRRL Y132. Meso-inositol was mainly responsible for this effect, although with strain ATCC 4126 other vitamins as well as an amino acid mixture were also required to minimise the steady-state residual glucose levels. The residual glucose concentration in continuous culture was, therefore, greatly dependent on the growth factor requirements of the particular yeast strain, which apparently increased on increasing the dilution rate into the respiro-fermentative region. The strain differences with respect to growth factor requirements at high dilution rates, which were not evident at low dilution rates, had a profound effect on the kinetics of glucose assimilation in aerobic chemostat culture.  相似文献

Established mammalian cells segregate variants, named autotrophs, able to proliferate in the absence of hormones and proteins of any kind. We provided complete formulas for autotrophic media composed of amino acids, vitamins, glucose, and inorganic salts only and showed that such media are sufficient to sustain continuous propagation of Chinese hamster fibroblasts and human keratinocytes. The cells grew on bare polystyrene and divided every 32 to 44 hours reaching approximately 8 x 10(5) cells/cm2. The results showed that the autotrophs may replace conventional cultures in most experimental and industrial cell production systems.  相似文献

A balanced supplementation method was applied to develop a serum and protein- free medium supporting hybridoma cell batch culture. The aim was to improve systematically the initial formulation of the medium to prevent limitations due to unbalanced concentrations of vitamins and amino acids. In a first step, supplementation of the basal formulation with 13 amino acids, led to an increase of the specific IgA production rate from 0.60 to 1.07 pg cell−1 h−1. The specific growth rate remained unchanged, but the supplementation enabled maintenance of high cell viability during the stationary phase of batch cultures for some 70 h. Since IgA production was not growth- related, this resulted in an approximately4-fold increase in the final IgA concentration, from 26.6 to 100.2 mgl−1. In a second step, the liposoluble vitamins E and K3 were added to the medium formulation. Although this induced a slightly higher maximal cell concentration, it was followed by a sharp decline phase with the specific IgA production rate falling to 0.47 pg cell−1 h−1. However, by applying a second cycle of balanced supplementation with amino acids this decline phase could be reduced and a high cell viability maintained for over 300 h of culture. In this vitamin- and amino acid- supplemented medium, the specific IgA production rate reached a value of 1.10 pg cell−1h−1 with a final IgA concentration of 129.8 mgl−1. The latter represents an increase of approximately5-fold compared to the non- supplemented basal medium. Rhizobia require the availability of 20 amino acids for the establishment of effective symbiosis with legumes. Some of these amino acids are synthesized by rhizobium, whereas the remaining are supplied by the host plant. The supply from plant appears to be plant-type specific. Alfalfa provides arginine, cysteine, isoleucine, valine and tryptophan, and cowpea and soybean provide histidine. The production of ornithine and anthranilic acid, the intermediates in the biosynthetic pathways of arginine and tryptophan, respectively, seems to be essential for effective symbiosis of Sinorhizobium meliloti with alfalfa. The expression of ilvC gene of S. meliloti is required for induction of nodules on the roots of alfalfa plants. An undiminished metabolic flow through the rhizobial pathways for the synthesis of purines and pyrimidines and the synthesis of biotin, nicotinic acid, riboflavin and thiamine by rhizobium appear to be requirements for normal symbiosis. To the best of our knowledge, this is the first review article on the role of rhizobial biosynthetic pathways of amino acids, nucleotide bases and vitamins in rhizobium-legume symbiosis. The scientific developments of about 35 years in this field have been reviewed.  相似文献

The effect of vitamins (B1, B2, B3, B6, B12, H, PP and folic acid) and amino acids (glutamic and aspartic acids) on glucose isomerase biosynthesis was studied in Streptomyces albogriseolus. These compounds were added either alone or in combinations to different growth media (synthetic and complex). The results were processed using mathematical methods, and the following mixture of vitamins and amino acids was proposed to be added to the complex fermentation medium: B2, 10 micrograms/L; B6, 10 micrograms/L; H, 1 microgram/L; aspartic acid, 0.01 microM. The production of glucose isomerase rose more than 1.5 times after such additions.  相似文献

SUMMARY. 1. Heterotrophic assimilation rates and concentrations of dissolved free amino acids (DFAA) were followed during diel studies in the eutrophic Lake Mossø, Lake Esrom and Lake øm in spring and summer in 1982. In all three lakes, three to four fold diel variations in concentrations and assimilation rates were measured. These fluctuations appeared to be iindependent of phytoplankton and bacteria production. Pools of DFAA varied from 380 nM (Lake Mossø) to 2430 nM (Lake ørn), with serine, glycine, alanine and ornithine as dominant free amino acids.
2. When similar water samples were incubated in a natural light-dark cycle or in total darkness, different pools of DFAA were measured in light and dark.
3. Decomposition of organic matter or zooplankton activity (rather than e.g. phytoplankton exudates) appear to be responsible for the concentration changes.
4. Observed discrepancies between simultaneous concentration changes and assimilation rates are discussed in relation to the applied tracer procedure and the concentration measurements.
5. Assimilation of DFAA sustained from 6% to 25% of the bacterial carbon requirement, corresponding to 2–12% of the phytoplankton production in the lakes.  相似文献

Graded levels of aflatoxin fed to broiler chickens for 3 weeks decreased the levels of most B vitamins in plasma, bile, and liver and decreased all free and hydrolyzed amino acids from peptides in plasma. The levels of thiamine, riboflavin, vitamin B6, pantothenic acid, and choline decreased by more than 60% in bile; vitamin B6, pantothenic acid, and choline decreased by more than 49% in plasma; thiamine, vitamin B6, pantothenic acid, choline, folate, and niacin decreased by more than 19% in liver; and only bile and plasma levels of folate increased (78 and 12%, respectively) with a dietary level of 5 microgram of aflatoxin per g of feed. Plasma levels of cystine and cysteine, methionine, threonine, serine, and aspartic acid decreased by more than 37%, whereas valine, lysine, leucine, histidine, hydroxyproline, and alanine decreased by more than 9% with 2.5 microgram of aflatoxin per g of feed. The data suggest that aflatoxin interferes with the metabolism of B vitamins and amino acids in chicks. However, these changes may be measuring the effects of aflatoxin-induced hepatic necrosis.  相似文献

It has been observed that beta-hydroxy-alpha-amino acids are transformed into other amino acids, when heated in dilute solutions with phosphorous acid, phosphoric acid or their ammonium salts. It has been shown that as in the case of previously reported glycine-aldehyde reactions, glycine also reacts with acetone to give beta-hydroxyvaline under prebiologically feasible conditions. It is suggested, therefore, that the formation of beta-hydroxy-alpha-amino acids and their transformation to other amino acids may have been a pathway for the synthesis of amino acids under primitive earth conditions.  相似文献
