We have previously shown that both endogenous auxin and ethylenepromote adventitious root formation in the hypocotyls of derootedsunflower (Helianthus annuus) seedlings. Experiments here showedthat promotive effects on rooting of the ethylene precursor,1-aminocyclopropane-l-carboxylic acid (ACC) and the ethylene-releasingcompound, ethephon (2-chloro-ethylphosphonic acid), dependedon the existence of cotyledons and apical bud (major sourcesof auxin) or the presence of exogenously applied indole-3-aceticacid (IAA). Ethephon, ACC, aminoethoxyvinylglycine (an inhibitorof ethylene biosynthesis), and silver thiosulphate (STS, aninhibitor of ethylene action), applied for a length of timethat significantly influenced adventitious rooting, showed noinhibitory effect on the basipetal transport of [3H]IAA. Theseregulators also had no effect on the metabolism of [3H]IAA andendogenous IAA levels measured by gas chromatography-mass spectrometry.ACC enhanced the rooting response of hypocotyls to exogenousIAA and decreased the inhibition of rooting by IAA transportinhibitor, N-1-naphthylphthalamic acid (NPA). STS reduced therooting response of hypocotyls to exogenous IAA and increasedthe inhibition of rooting by NPA. Exogenous auxins promotedethylene production in the rooting zone of the hypocotyls. Decapitationof the cuttings or application of NPA to the hypocotyl belowthe cotyledons did not alter ethylene production in the rootingzone, but greatly reduced the number of root primordia. We concludethat auxin is a primary controller of adventitious root formationin sunflower hypocotyls, while the effect of ethylene is mediatedby auxin. The effect of glucose on flower bud formation was studied inthin-layer tissue cultures of epidermal strips from flower stalksof Nicotiana tabacum L. cv. Samsun. A minimum concentration of 30 mol m–3 glucose in the MS-mediumcontaining 1.0 mmol m–3 of both NAA and BA was necessaryfor flower bud formation. With 150 mol m–3 glucose a minimumstay of 10 d was required for optimal flower bud formation. Withholding glucose for a limited period at different time intervalsafter the onset of culture caused a delay in flower bud formationand did not affect previous development on glucose. The resultsindicated that competence for flower bud initiation is not restrictedto the early stage of culture. The process may start at anytime later at the appropriate glucose concentration. However,for both optimal initiation and further development of flowerbuds the presence of a metabolizable sugar is required. Peanut (Arachis hypogaea L. ) seed powder accumulated ATP fromAMP and phosphoenolpyruvate (PEP) at a rate of approx. 100 pmolmin–1mg powder at 35° C. When peanut seed powderwas incubated with various substrates, which may result in PEPor AMP (ADP) synthesis, then ATP accumulated. The best substratecombinations examined so far were AMP + succinate, NADH2, andAMP + malate + NAD, with activities of 33, 12 and 12 pmol minmg–1powder,respectively; AMP + malate showed very low activity. Some combinationsexhibited linear activities with time, while others had an exponential-typeprofile. The temperature dependence of the ATP accumulationdemonstrated by the Ahrrenius plot had a double phase with atransition point at 25° C. The Ea values between 15°C and 25° C were 25 000–50 000 cal/mol, while above25° C the Ea values fluctuated between 6000 and 8000 cal/mol(depending on the substrate). The AMP + PEP combination exhibiteda single-phase profile between 15° C and 40° C, withan Ea value of 22 000 cal/mol. In the presence of some substrates,ethephon (ethylene) had a stimulatory effect and caused an increasein the Ea values at the high temperature phase. A comparisonof seed powder from dormant seeds with that from non-dormantseeds revealed that some substrate combinations accumulate ATPfaster in non-dormant seeds and others do so in dormant seeds. The results of phosphorus fractionation in the sediments from a contaminated river containing different aquatic plants, analyzed by solution 31P-NMR for Organic Phosphorus, showed that the concentration of Inorganic Phosphorus dominated in all species and Organic Phosphorus accounted for over 20% of Total Phosphorus. In general, orthophosphate was dominant in all the sampling sites. The proportion of Organic Phosphorus accounting for the Total Phosphorus in the sediments with different plant decreased in the following order: Paspalum distichum > Typha orientalis > Hydrilla verticillata. Phosphorus-accumulation ability of Paspalum distichum was obviously stronger than Typha orientalis and Hydrilla verticillata. The Organic Phosphorus was in aquatic plants dominated by humic-associated P (Hu-P), which converted to Inorganic Ohosphorus more significantly in submerged plants than in emerged plants. The sediment dominated by Paspalum distichum abundantly accumulated Organic Phosphorus in the orthophosphate monoester fraction. The degradation and mineralization of orthophosphate monoester was the important source of high Inorganic Phosphorus concentration and net primary productivity in Suoxu River. The Organic Phosphorus derived from Typha orientalis and Hydrilla verticillata was dramatically converted to Inorganic Phosphorus when the environmental factors varied.  相似文献

In vitro culture of Picea sitchensis (Bong.) Carr. needle explantson a number of basal culture media indicated that a complexmixture of organic additives was neither essential nor stimulatory.Adventitious bud production occurred at inorganic nitrogen levelsof 15–60 x 10–3M and 7.5–30 x 10–3 Min the adventitious bud induction and elongation media, respectively,when a well-balanced ratio of NH4+:NO3 was maintained.However, necrosis was increased at the highest level of inorganicnitrogen. Organogenesis was more sensitive to changes in theratio of NH4+:NO3. Complete replacement of NH4+ withNO3 during the adventitious bud induction passage severelyinhibited organogenesis, indicating that a reduced form of nitrogenmay be essential for adventitious bud differentiation. Conversely,a high proportion of NH4+ in either the adventitious bud inductionor elongation medium increased tissue necrosis and inhibitedbud induction, reflecting the potential toxicity of this ion.Explants from different individual trees were found to varyconsiderably in their morphogenetic responses, but NH4+:NO3ratios of 1:5 or 1:2 were o ptimal for all individual treeswhich developed adventitious buds from needle cultures. Application of ethephon to field-grown plants of both bush andtrailing forms of Cucurbita maxima and C. pepo caused leaf epinasty,suppression of male flowers and earlier production and increasein numbers of female flowers. This gave rise to an increasein the ratio of female to male flowers per plant and a decreasein the total number of flowers. Observations of C. pepo showed that even at the two true leafstage there are several nodes present in the unexpanded shoot.Each node has one main and several secondary buds. The sex ofthe main bud at the first five to six nodes is usually determinedat this stage but the secondary buds still have bisexual potential.The change in sex expression was brought about by all male flowerbuds that had formed by the spraying time aborting, and allbuds that developed (both main and secondary) for at least 7days after spraying became female flowers. Thus, nodes fiveand six had male flowers in the controls, whereas in ethephon-sprayedplants the presumptive male flowers aborted at the bud stageat these nodes and secondary primordia developed into functionalfemale flowers. Cucumber seedlings, when cultured in vitro, showed differencesin cotyledon etiolation rates among cultivars with differentgenetic backgrounds for sex expression. The chlorophyll contentin gynoecious cultivars (acrF/acrF) decreased rapidly whilethat in monoecious ones (acr+/acr+) decreased more slowly, andthat in mono-gynoecious ones (acr1/acr1) decreased at an intermediaterate. Etiolation was suppressed even in early-etiolating cultivarswhen the flask remained unsealed or endogenously evolved ethylenewas removed. Cotyledon etiolation was enhanced even in late-etiolatingcultivars when ethephon was added to the flask. The rate ofetiolation corresponded to the ethylene concentration in theflask; much more ethylene was detected in early-etiolating cultivarsthan in late-etiolating ones. Ethylene accumulation is one of the important factors involvedin the cotyledon etiolation observed in in vitro cultures. Thedifference in etiolation rates among seedlings with differentgenetic backgrounds for sex expression corresponds to theirability for ethylene evolution, in the order of acrF>acr1>acr+. Abscisic acid (ABA) at low concentrations brings about the formationof turions (dormant fronds) in Lemna polyrhiza within 3 to 5days after application. The incorporation of 3H-thymidine intoDNA, separated by polyacrylamide-gel electrophoresis, is inhibitedby 80 to 90 per cent within 1 to 3 h of ABA application. Theincorporation of 14C-orotate and 32P into RNA is not inhibiteduntil 3 to 9 h after ABA application, but 70 per cent inhibitionis reached after 24 h on 10–5 M ABA. There is little inhibitionof 14C-leucine incorporation into protein until 2 to 3 daysafter application of ABA. The capacity of nitrate to inducenitrate reductase in cultures previously grown on nitrate-freemedium is not affected by ABA even up to 3 days after application.The results are discussed in relation to the mode of actionof ABA.  相似文献

This study was designed to examine possible links between polyaminebiosynthesis and effects of growth regulatory compounds. Anauxin (IAA), a cytokinin [benzyladenine; benzylaminopurine (BAP)],an ethylene source (ethephon) and abscisic acid (ABA) were individuallyapplied to terminal buds of excised etiolated or red light (R)-exposedpea epicotyls. Effects were noted on bud fresh weight and onthe two main enzymes of putrescine biosynthesis, arginine decarboxylase(ADC; EC 4.1.1.19 [EC] ) and ornithine decarboxylase (ODC; EC 4.1.1.17 [EC] ).As previously reported [Dai and Galston (1981) Plant Physiol.67: 266], both bud growth and ADC activity are increased byR light. In such buds, ADC is raised further by 1–10 µMBAP or ABA and inhibited by 1–10 µM IAA or ethylene(50 mg/liter or more of ethephon). In all cases, effects ofR-irradiation plus 1 mM growth regulators on ODC activity wasthe inverse of their effects on ADC, indicating independentcontrol of these pathways. The long-term effect of Hydrilla verticillata to different dredged sediments was investigated in laboratory aquatic microcosms. Two kinds of lake sediments, i.e., fertile sludge and brown clay, which were acquired by deep or slight dredging and differed in their nutrients levels and particle sizes, were used as growth substrate. One year after H. verticillata seedlings were transplanted into the microcosms, growth and physio-biochemical characters of the plants were estimated. In our study, sludge sediment eventually promoted the increase of the biomass, shoot length, bifurcation, nitrogen, phosphorus, soluble protein content of H. verticillata, but not root number and root mass ratio. Additionally, the soluble sugar content was significantly lower in summer (August) in the sludge sediment. Sediment nutrients also increased photosynthetic pigment content, while POD and SOD activities were not affected. The fertile sludge and brown clay formed by different dredging are quite different, but both sediments could meet the requirement of H. verticillata during early growth stage. The fertile sediment after moderate dredge-up fit the growth of H. verticillata. The brown clay formed after excessive dredging showed a lack of nutrients and therefore led to the weak growth of the aquatic plants. It is concluded that moderate dredging was suitable for H. verticillata growth and for absorbing nitrogen and phosphorus from eutrophic fresh water. The experiment also suggested that submersed aquatic plant restoration in freshwater can be promoted by human assistance.  相似文献

By manipulation of various growth regulators and physical conditions,plants have been regenerated from excised roots, stem segments,cotyledons, leaves, and callus cultures of red cabbage (Brassicaoleracea var. capitata) grown under in vitro conditions. Shootbuds were induced on isolated root segments (1 cm long) culturedon Murashige and Skoog's medium and the frequency of bud formationwas greatly enhanced by the addition of kinetin (0.5 part 10–6).Callus obtained from the seeds, cotyledons, and hypocotyl segmentscultured on a medium fortified with 2,4-D (1 part 10–6),kinetin (0.1 part 10–6), and coconut milk (10%, v/v) hasbeen repeatedly subcultured. The callus is slow growing, andon transference to a kinetin (2 parts 10–6) and IAA (2parts 10–6) medium underwent morphogenesis to give riseto plants. 为了筛选出适宜修复白洋淀镉(Cd)污染水体的沉水植物,该研究通过室内模拟试验,分析了四种沉水植物黑藻、狐尾藻、金鱼藻和菹草对Cd的耐受性及对底泥Cd的富集和迁移能力。结果表明:(1)通过毒性测试研究,Cd对黑藻、狐尾藻、金鱼藻及菹草的4 d-EC50(半数抑制浓度)分别为0.51、0.81、0.03、0.12 mg·L-1,狐尾藻对Cd的耐性最强,黑藻次之,金鱼藻对Cd的耐性最低; 四种沉水植物对Cd的最大富集量分别为27.89、15.28、22.54、32.74 g·kg-1,菹草对Cd的富集能力最强,黑藻次之,狐尾藻对Cd的富集能力最低。(2)通过Cd污染底泥修复研究,黑藻、狐尾藻和菹草体内Cd富集量整体表现为根>叶片和茎(P<0.05); 地上部、根对Cd的富集能力分别表现为黑藻>菹草>狐尾藻,菹草>黑藻>狐尾藻; 三种沉水植物对Cd的迁移能力则表现为黑藻>狐尾藻>菹草。总之,黑藻对底泥中Cd富集和迁移能力均较强,且耐性较高,是最适合修复白洋淀Cd污染水体的沉水植物。  相似文献

Imbibition of seeds of oil seed rape (Brassica napus cv Jetneuf)in 10–3 M aminoethoxyvinylglycine (AVG) or 10–2silver thiosulphate (Ag+) had no effect on germination nor onthe emergence of seedlings from uncompacted or lightly compressedsoil, but significantly reduced emergence from moderately compressedsoil of 68.4 or 143.3 N cm–2 impedance. Exertion of force by emerging control seedlings against a staticcantilever bar fitted with strain gauges reached a maximum (Fmax)of 6 g over 10 h. Higher axial forces were achieved when theseedlings were emerging from compressed soil, without any changein the time required to reach Fmax, so that the build-up offorce was considerably (1.8 fold) faster than in uncompressedsoil. This adaptive response to soil impedance was modified by theseed pretreatments employed. Seedlings from AVG or Ag+pretreatedseeds produced lower axial forces than controls, and neitherFmax nor the rate at which force developed showed any responseto soil compression. After pretreatment in 10–3 ethephon or 10–4 naphthaleneacetic acid (NAA) the seedlings achieved similar Fmax to controlseedlings, but responded more rapidly to soil compression sothat the rate of build up of emergence force was 2.3 fold (NAA)or 2.8 fold (ethephon) faster in compressed than in uncompressedsoil. The results suggest that the exertion of force by a seedlingagainst a barrier involves a dynamic response to impedance onthe part of the seedling. This response can be either enhancedor suppressed by substances which affect ethylene productionor ethylene action. Such compounds may have promise for modifyingseedling emergence from impeding soils. Malformin stimulated ethylene production of Phaseolus vulgarisL. seedlings and explants. However, when malformin was vacuum-infiltratedinto apical bud sections, the production of ethylene was inhibited,ethane production was stimulated and the sections became softand pliable; in pure oxygen, ethylene production was- stimulatedand the sections remained firm. Prolonged stimulation of ethaneproduction by malformin-treated sections required oxygen. The data of chlorophyll amounts in diverse bio-communities arecompiled and discussed with reference to matter production. The chlorophyll amount in euphotic zone of lakes and oceanswas less than 1 g/m2, mostly less than 0.1 g/m2. In phytoplanktonblooms it was ca. 0.1–1 g/m2. Large values of 5–20g/m2 were obtained in the outdoor mass cultures of Chlorella,in which the high population density and chlorophyll contentof the alga were observed. In terrestrial higher plant communitiesthe chlorophyll amount (ca. 1–10 g/m2) was usually higherthan in aquatic phytoplankton communities. The largest (13.3g/m2) was obtained with an evergreen gallery forest in Thailand.The chlorophyll amount of desert bio-community could be expectedto be as high as that in water blooms. Apical dominance phenomena have been studied in seedlings ofVicia faba particularly in relation to the movement about theplant of uracil-2-14C applied to the cotyledons. Decapitationjust below the second node releases the growth of the lowermostlateral bud and inhibition is completely reimposed by applicationof indole-3-acetic acid (IAA) to the cut surface. Uracil-2-14Capplied in solution to the cotyledons is distributed in thestems of all experimental seedlings with no consistent differencesdue to decapitation or IAA application. On the other hand, decapitationresults in a rapid increase in uracil-2-14C content in the lateralbuds which far exceeds their promoted growth. This uptake iscompletely suppressed by IAA application. A ring of tri-iodobenzoicacid (TIBA)-lanolin paste around the stem above the bud suppressesIAA action both in bud growth and on uracil-2-14C uptake, andalso on the movement of IAA-1-14C down the stem. TIBA-application to the base of the bud does not prevent IAAaction on bud growth, but also does not prevent the movementof IAA-1-14C (or a water soluble product of its metabolism)into the bud. Direct application of kinetin to the lateral bud of intact plantscauses a short-lived release of growth. Gibberellic acid producesa smaller and scarcely significant increase which is additiveto the kinetin effect. Neither has any effect on uracil-2-14Cmovement into the bud. The implications of these findings are discussed in relationto various existing theories of the mode of auxin action inapical dominance and it is concluded that their strongest supportis for a mechanism involving the suppression of phloem differentiationin the vascular supply to the bud.  相似文献

The composition of the lipophilic extract from Hydrilla verticillata, the common aquatic weed collected from Sasthamkotta Lake, the largest freshwater lake in Kerala, south of the west coast of India, was investigated. The lake is a designated wetland of international importance under the Ramsar Convention since 2002. GC-MS study of the unsaponifiable lipophilic extract of H. verticillata confirmed the presence of 3,5,11,15-tetramethyl 1- hexadecen-3-ol (C20H40O) and phytol (C20H40O) as major components, and their structures were elucidated. Phytol and 3,5,11,15-tetramethyl 1- hexadecen3-ol are the two isomers of the diterpeneol (C20H40O) found in the unsaponifible lipophilic extract of H. verticillata and are formed by the hydrolysis of the alcohol moiety of chlorophyll. On quantification, an appreciable concentration of phytol (6.39 g?Kg?1) was estimated. The feasibility to utilize H. verticillata to produce phytol is to be addressed by further studies since H. verticillata is considered as one of the world’s fast widely spread aquatic weeds on account of its numerous mechanisms of vegetative reproductions.  相似文献

Explants of taro cultivars belonging to Colocasia esculentavar. esculenta have been nearly impossible to culture untilrecently. Here, we describe a method which induces callus formationfrom bud explants of Colocasia esculenta var. esculenta cv.Akalomamale, brings about shoot and root production, and leadsto plantlet regeneration. The medium used is half-strength Murashige-Skoog(HMS) solution containing 25 ml taro tuber extract (TE), 2 mg2, 4, 5-trichlorophenoxyacetic acid and 200 mg glutamine 1–1.TE is an important requirement for bud explants and callus tissues.Root induction on callus-derived shoots (i.e. plantlet formation)occurs on HMS containing only 25 ml TE and 100 ml coconut water1–1. The possibility has been investigated that abscisic acid (ABA)might act as a correlative inhibitor of lateral bud growth inPisum sativum and Phaseolus vulgaris. Application of ABA insmall quantities (2µg) to axillary buds on decapitatedplants of P. sativum caused appreciable inhibition of theirgrowth, and induced a compensatory growth of the bud on an adjacentnode. Application of this same quantity of ABA to axillary budson decapitated plants of Phaseolus vulgaris was without effect,but a high concentration in lanolin (1 mg g–1) did substantiallyreduce bud outgrowth. Endogenous ABA-like substances in Phaseolusvulgaris, detected by bioassay and electron capture g.l.c.,were present in similar concentrations in shoot tips, lateralbuds on intact plants and lateral buds on plants decapitated24 h earlier. The effects of applied ABA suggested that it might be involvedin the mechanism of correlative inhibition in Pisum sativum,but it was not possible to test this hypothesis by determiningendogenous ABA levels in axillary buds because of their smallsize. The evidence presented here suggests that ABA is not acorrelative inhibitor in Phaseolus vulgaris even though at highconcentration it can inhibit the growth of axillary buds.  相似文献
